Dietarytrans,trans-linoleate (trans 18∶2), when fed to rats in increasing amounts, caused a reduction in lung weights, particularly at very high dietary concentrations. The fatty acids of the phospholipids and triglycerides were altered. The percentage of oleic and arachidonic acid decreased as dietarytrans 18∶2 was increased. Eicosatrienoic acid (20∶3) appeared in the phospholipids of lungs from rats receiving 100% dietarytrans 18∶2, but its concentration was much lower than in lungs from rats on an essential fatty acid deficient diet, indicating thattrans 18∶2 inhibited the enzymes synthesizing 20∶3.  相似文献

Trans fatty acids may be involved in atherosclerotic vascular diseases. We investigated the incorporation of dietary trans fatty acids and oleic acid into the serum triglycerides (TG), cholesterol esters (CE), and phospholipids (PL). Fourteen healthy female volunteers, aged 23.2±3.1 yr (mean±SD), body mass index 20.8±2.1 kg/m2 participated in this study. All subjects consumed both a trans fatty acid-enriched diet (TRANS diet) and an oleic acid-enriched diet (OLEIC diet) for 4 wk according to a randomized crossover design. Both experimental diet periods were preceded by consumption of a baseline diet for 2 wk which supplied 37% of total energy (E%) as fat: 18 E% from saturated fatty acids (SFA), 12 E% from monounsaturated fatty acids, and 6 E% from polyunsaturated fatty acids. Five E% of the SFA in the baseline diet was replaced by trans fatty acids (18∶1 t and 18∶2 c,t+18∶2t,t, where c is cis and t is trans) in the TRANS diet and by oleic acid (18∶1n-9) in the OLEIC diet. After the TRANS diet, the proportions of 18∶1t and 18∶2t increased (P <0.001) in all serum lipid fractions analyzed. The increase of 18∶1 t in TG and PL (1.80±0.28 vs. 5.26±1.40; 1.07±0.34 vs. 3.39±0.76 mol% of total fatty acids, respectively) was markedly higher than that in CE (0.44±0.07 vs. 0.92±0.26), whereas that of 18∶2t was nearly the same in all three fractions. The proportions of palmitic, stearic, arachidonic, and eicosapentaenoic acids in TG, CE, and PL and that of oleic acid in TG and CE were decreased when compared with the baseline value. In contrast, the proportion of palmitoleic acid in TG and PL and that of linoleic acid in PL increased on the TRANS diet. After consumption of the OLEIC diet, the proportion of oleic acid increased in all three lipid fractions analyzed, and the percentage increase was nearly the same in all fractions. In contrast, the proportions of 18∶1 t in TG and PL and 18∶2 t in TG and CE decreased when compared with the baseline value. In conclusion, a moderate increase in dietary trans fatty acids resulted in a marked incorporation into serum lipids and decreased the conversion of linoleic acid to its more unsaturated long-chain metabolites. Analysis of 18∶1 t from serum TG and PL seems to reflect reliably the dietary intake of this fatty acid.  相似文献

trans Isometric fatty acids of partially hydrogenated fish oil (PHFO) consist oftrans 20∶1 andtrans 22∶1 in addition to thetrans isomers of 18∶1, which are abundant in hydrogenated vegetable oils, such as in partially hydrogenated soybean oil (PHSBO). The effects of dietarytrans fatty acids in PHFO and PHSBO on the fatty acid composition of milk were studied at 0 (colostrum) and 21 dayspostpartum in sows. The dietary fats were PHFO (28%trans), or PHSBO (36%trans) and lard. Sunflower seed oil (4%) was added to each diet. The fats were fed from three weeks of age throughout the lactation period of Experiment 1. In Experiment 2 PHFO or “fully” hydrogenated fish oil (HFO) (19%trans), in comparison with coconut oil (CF) (0%trans), was fed with two levels of dietary linoleic acid, 1 and 2.7% from conception throughout the lactation period. Feedingtrans-containing fats led to secretion oftrans fatty acids in the milk lipids. Levels oftrans 18∶1 andtrans 20∶1 in milk lipids, as percentages of totalcis+trans 18∶1 andcis+trans 20∶1, respectively, were about 60% of that of the dietary fats, with no significant differences between PHFO and PHSBO. The levels were similar for colostrum and milk. Feeding HFO gave relatively lesstrans 18∶1 andtrans 20∶1 fatty acids in milk lipids than did PHFO and PHSBO. Only low levels ofcis+trans 22∶1 were found in milk lipids. Feedingtrans-containing fat had no consistent effects on the level of polyenoic fatty acids but reduced the level of saturated fatty acids and increased the level ofcis+trans monoenoic fatty acids. Increasing the dietary level of linoleic acid had no effect on the secretion oftrans fatty acids but increased the level of linoleic acid in milk. The overall conclusion was that the effect of dietary fats containingtrans fatty acids on the fat content and the fatty acid composition of colostrum and milk in sows were moderate to minor.  相似文献

The effect of a diet containingtrans-fatty acids (tFA) on the fatty acid composition and fat accumulation in adipose tissue was investigated in mice. Male C57BI/6J mice were fed Control or Trans Diets that were similar, except that 50% of the 18∶1, which was allcis in the Control Diet, was replaced bytFA in the Trans Diet. At selected ages, body weight, epididymal fat pad weight, perirenal fat yield, adipose tissue cellularity and fatty acid composition were examined. Over the time period studied (2–24 mon), the proportion of 18∶0 and 16∶0 tended to decrease whilecis-18∶1 levels increased. Compared to the Control Diet, the Trans Diet resulted in adipose tissue lipids with higher percentages of 14∶0 and 18∶2n−6 and lower percentages ofcis-18∶1 and 20∶4n−6. In polar lipids,tFA replaced saturated fatty acids, whereastFA replacedcis-18∶1 in the nonpolar lipids. Body weights at 16 and 24 mon of age and epididymal fat pad weights at 8–24 mon of age were lower in mice fed the Trans Diet as compared to those fed the Control Diet. At the ages studied, the Trans Diet also resulted in lower values for perirenal fat weights, triacylglycerol to polar lipid ratios, and adipose cell size. The data suggest that chronic consumption oftFA affects lipid metabolism and results in decreased fat accumulation in murine adipose tissue.  相似文献

Cardiolipins (CL) have unique fatty acid profiles with generally high levels of polyunsaturated fatty acids, primarily 18∶2n−6, and low levels of saturated fatty acids. In order to study the effect of dietary fatty acid isomers on the fatty acid composition of cardiolipins, rats were fed partially hydrogenated marine oils (HMO), rich in 16∶1, 18∶1, 20∶1, and 22∶1 isomeric fatty acids, supplemented with linoleic acid at levels ranging from 1.9% to 14.5% of total fat. Although the dietary fats contained 33%trans fatty acids, the levels oftrans fatty acids in CL were below 2.5% in all organs. The fatty acid profiles of cardiolipins of liver, heart, kidney and testes showed different responses to dietary linoleic acid level. In liver, the contents of 18∶2 reflected the dietary levels. In heart and kidney, the levels of 18∶2 also parallelled increasing dietary levels, but in all groups fed HMO, levels of 18∶2 were considerably higher than in the reference group fed palm oil. In testes, the 18∶2 levels were unaffected by the dietary level of 18∶2 and HMO.  相似文献

Studies are reported on the effects of dietarytrans fatty acids on the 6- and 9-acyl desaturase activities in the liver microsomes of rats fed essential fatty acid (EFA)-deficient and non-FFA-deficient diets. In experiment I, weanling male rats were fed a semisynthetic diet with either 10% safflower oil (SAF) or 10% hydrogenated coconut oil (HCO). At the age of one year, half of the dietary fat was replaced by a supplement containing elaidate, linolelaidate andcis,trans-trans,cis-18∶2 (TRANS) for 12 weeks. In experiment II, male rats which were kept from weaning on a 10% SAF diet for one year received one of the following fat supplements for a 12-week period: 10% HCO, 9% HCO+1% TRANS, or 5% HCO+5% TRANS. Feeding TRANS depressed the 6-desaturase activity in the liver microsomes, especially in the EFA-deficient rats (HCO+TRANS group of experiment I). Unlike the 6-deaturase activity, the 9-desaturase activity was not inhibited by the dietarytrans fatty acids and was significantly stimulated in the non-EFA-deficient rats (SAF+TRANS group of experiment I and HCO+TRANS groups of experiment II). This was evidenced by incubation reactions and by comparisons of fatty acid consumptions and microsomal fatty acid levels, showing extra biosynthesis of 16∶1 and 18∶1 when TRANS was fed. The biosynthesis of essential (n−6) fatty acids was depressed by the TRANS supplement in EFA-deficient as well as in non-EFA-deficient animals.  相似文献

Male Sprague-Dawley rats were fed for 30 days a high-fat liquid ethanol diet with dihydroxyacetone, pyruvate and riboflavin added as supplements (AMA-). Plasma triglyceride (TG) levels were 6-fold greater in these rats than in those fed and alcohol with without the supplements (AA-). The liver TG content in rats fed the AMA-diet was similar to that of rats fed a control diet (CA-) in which alcohol was replaced with isocaloric amounts of dextrose. Livers of rats fed the AA- diet had 3 times more TG than controls. Alcohol ingestion also enhanced the hepatic content of cholesteryl esters (CE) and phospholipids (PL). These lipids were reduced to levels found in livers of rats fed the control diet (CA-) when dihydroxyacetone, pyruvate and riboflavin were included in the alcohol diet. The fatty acid compositions of TG, CE and PL from livers of rats fed the AMA-diet were similar to those of corresponding lipids from rats fed the control diet (CA-) but differed from compositions when fed the alcohol diet (AA-). Regardless of the diet fed, TG had the same fatty acid composition in plasma and liver. The same was true of PL fatty acid composition. However, the fatty acid composition of CE differed between liver and plasma. The major fatty acid in liver CE was 18∶1 whereas in plasma it was arachidonic acid (20∶4). Reduced fatty liver was observed in an earlier study when rats were fed ad libitum an ethanol diet containing 20∶4. In the present study, we pair-fed the same diet and fatty liver was not reduced. Dihydroxyacetone, pyruvate and riboflavin did not prevent alcohol-induced fatty liver when 20∶4 was included in the AMA-diet. Our results confirm that dietary dihydroxyacetone, pyruvate and riboflavin prevent alcohol-induced fatty liver, and show that this effect may result from increased mobilization of fat from liver.  相似文献

The influence of the linoleic acid levels of diets containing partially hydrogenated marine, oils (HMO) rich in isomeric 16∶1, 18∶1, 20∶1 and 22∶1 fatty acids on the fatty acid profiles of lipids from rat liver, heart and adipose tissue was examined. Five groups of rats were fed diets containing 20 wt% fat−16% HMO+4% vegetable oils. In these diets, the linoleic acid contents varied between 1.9% and 14.5% of the dietary fatty acids, whereas the contents oftrans fatty acids were 33% in all groups. A sixth group was fed a partially hydrogenated soybean oil (HSOY) diet containing 8% linoleic acid plus 32%trans fatty acids, mainly 18∶1, and a seventh group, 20% palm oil (PALM), with 10% linoleic acid and notrans fatty acids. As the level of linoleic acid in the HMO diets increased from 1.9% to 8.2%, the contents of (n−6) polyunsaturated fatty acids (PUFA) in the phospholipids increased correspondingly. At this dietary level of linoleic acid, a plateau in (n−6) PUFA was reached that was not affected by further increase in dietary 18∶2(n−6) up to 14.5%. Compared with the HSOY- or PALM-fed rats, the plateau value of 20∶4(n−6) were considerably lower and the contents of 18∶2(n−6) higher in liver phosphatidylcholines (PC) and heart PC. Heart phosphatidylethanolamines (PE) on the contrary, had elevated contents of 20∶4(n−6), but decreased 22∶5(n−6) compared with the PALM group. All groups fed HMO had similar contents oftrans fatty acids, mainly 16∶1 and 18∶1, in their phospholipids, irrespective of the dietary 18∶2 levels, and these contents were lower than in the HSOY group. High levels of linoleic acid consistently found in triglycerides of liver, heart and adipose tissue of rats fed HMO indicated that feeding HMO resulted in a reduction of the conversion of linoleic acid into long chain PUFA that could not be overcome by increasing the dietary level of linoleic acid.  相似文献

Female pigs were fed from three wk of age and up to two years a diet containing partially hydrogenated fish oil (PHFO, 28%trans monoenoic fatty acids), partially hydrogenated soybean oils (PHSBO, 36%trans fatty acids) or lard. No consistent differences were found between PHFO and PHSBO with regard to incorporation oftrans fatty acids in organ lipids, buttrans incorporations were highly organ-specific. Notrans fatty acids were detected in brain phosphatidylethanolamine (PE). The incorporation of monoenoictrans isomers, as a percentage of totalcis + trans, in other organs was highest in subcutaneous adipose tissue and liver mitochondria PE, followed by blood lipids with the lowest level in heart PE. The percentage oftrans isomers compared with that of dietary lipids was consistently lower for 20∶1, compared with 18∶1 in organs from PHFO-fed pigs. The only effect of dietarytrans fatty acids on the fatty acid pattern of brain PE was an increased level of 22∶5n−6. Heart PE and total serum lipids of pigs fed the hydrogenated fats contained higher levels of 18∶2n−6, and these lipids of the PHFO-fed group also contained slightly elevated amounts of 20∶3n−6, 18∶3n−3 and 20∶5n−3. Liver mitochondria PE of the PHFO group also contained higher levels of 20∶3n−6 and 22∶5n−6. Dietarytrans fatty acids caused a consistent decrease of saturated fatty acids compensated by increased levels of monoenes. Thus, it may be concluded that dietary long-chaintrans fatty acids in PHFO behaved similarly metabolically to 18∶1-trans in PHSBO in pigs, without noticeable influence on brain PE composition and with moderate to slight effects on the fatty acid profile of the other organs.  相似文献

The effects of dietarytrans fatty acids on the fatty acid composition of the brain in comparison with other organs were studied in 3-wk-old suckling piglets. In Experiment (Expt.) 1 the piglets were delivered from sows fed partially hydrogenated fish oil (PHFO) (28%trans), partially hydrogenated soybean oil (PHSBO) (36%trans) or lard (0%trans). In Expt. 2 the piglets were delivered from sows fed PHFO, hydrogenated fish oil (HFO) (19%trans) or coconut fat (CF) (0%trans) with two levels of dietary linoleic acid (1 and 2.7%) according to factorial design. In both experiments the mother's milk was the piglets' only food. The level of incorporation oftrans fatty acids in the organs was dependent on the levels in the diets and independent of fat source (i.e., PHSBO, PHFO or HFO). Incorporation oftrans fatty acids into brain PE (phosphatidylethanolamine) was non-detectable in Expt. 1. In Expt. 2, small amounts (less than 0.5%) of 18∶1trans isomers were found in the brain, the level being slightly more on the lower level of dietary linoleic acid compared to the higher. In the other organs the percentage of 18∶1trans increased in the following order: heart PE, liver mitochondria PE, plasma lipids and subcutaneous adipose tissue. Small amounts of 20∶1trans were found in adipose tissue and plasma lipids. Other very long-chain fatty acids from PHFO or HFO (i.e., 20∶1cis and 22∶1cis+trans) were found in all organ lipids except for brain PE. Dietarytrans fatty acids increased the percentage of 22∶5n−6 in brain PE. Except for the brain and the heart, dietarytrans fatty acids reduced the percentage of saturated fatty acids and increased the percentage of monoenoic acids (includingtrans). The overall conclusion was that dietarytrans fatty acids had no noticeable effect on the brain PE composition but slight to moderate effects on the fatty acid profile of other organs of suckling piglets.  相似文献

The fatty acid profile of cardiolipin (CL) from brain and cardiac mitochondria was measured to determine whether CL isolated from these two tissue sources responded similarly to alterations in dietary fat composition. Male Wistar rats were fed 20% (w/w) diets containing 2 to 12% (w/w) 18∶2n-6 for four weeks. Despite higher baseline levels of CL 18∶2n-6 in cardiac (54±1% of total fatty acids) compared to brain (13±1%) mitochondria, CL 18∶2n-6 levels increased in proportion to dietary 18∶2 levels. The degree of change in 18∶2n-6 was comparable with both tissues showing an approximate 1.5- to 2-fold increase. The time course of changes in CL fatty acid profile was examined in a subsequent experiment in which animals were fed 20% (w/w) fat diets containing either 3 or 15% α-linoleate. Changes in cardiac CL 18∶1, 18∶2n-6, and 22∶6n-3 levels were observed within one week of feeding. While statistically significant differences were not observed in brain CL until the second week of feeding, the time course did not differ substantively from that observed in heart. The results from this study suggest that while baseline fatty acid profile of cardiac and neural CL differ, mitochondria from both tissues show comparable sensitivity to changes in dietary fat composition. Furthermore, it would appear that the turnover rate of fatty acids in CL is similar in both tissues.  相似文献

Studies of the comparative effects of a semi-synthetic diet containing supplements of corn oil, no fat, linolenate ortrans,trans-linoleate on blood coagulation parameters are reported. In spite of large differences in fatty acid composition of the tissue lipids of the different groups, the only diets that appeared to produce abnormal hematologic and hemostatic properties were those containingtrans,trans-linoleate. These groups of animals showed significant differences from a control group of animals fed Purina rat chow in platelet and fibrinogen concentration, and values for hematocrit and prothrombin time. A positive fibrinolysin test was also obtained in about 50% of the animals fedtrans,trans-linoleate.  相似文献

Uncommoncis andtrans fatty acids can be desaturated and elongated to produce unusual C18 and C20 polyunsaturated fatty acids in animal tissues. In the present study we examined the formation of such metabolites derived fromcis andtrans isomers of oleic and linoleic acids of partially hydrogenated vegetable oil origin in rats. For two months, aduut male rats were fed a partially hydrogenated canola oil diet containing moderately high levels oftrans fatty acids (9.6 energy%) and an adequate level of linoleic acid (1.46 energy%). Analysis of the phospholipid (PL) fatty acids of liver, heart, serum and brain showed no new C18 polyunsaturated fatty acids, except for those uncommon 18∶2 isomers originating from the diet. However, minor levels (each <0.3% PL fatty acids) of six unusual C20 polyunsaturated fatty acids were detected in the tissues examined, except in brain PL. Identification of their structures indicated that the dietary 9c,13t−18∶2 isomer, which is the majortrans polyunsaturated fatty acid in partially hydrogenated vegetable oils, was desaturated and elongated to 5c,8c,11c,15t−20∶4, possibly by the same pathway that is operative for linoleic acid. Furthermore, dietary 12c−18∶1 was converted to 8c,14c−20∶2 and 5c,8c,14c−20∶3; dietary 9c,12t−18∶2 metabolized to 11c,14t−20∶2 and 5c,8c,11c14t−20∶4, and dietary 9t,12c to 11t,14c−20∶2. These results suggested that of all the possible isomers of oleic and linoleic acids in partially hydrogenated vegetable oils, 12c−18∶1, 9c,13t−18∶2, 9c,12t−18∶2 and 9t,12c−18∶2 are the preferred substrates for desaturation and elongation in rats. However, their conversions to C20 metabolites were not as efficient as that of oleic or linoleic acids.  相似文献

Milk samples were collected from 11 mothers who were at least 4 weeks postpartum. The amounts of fat and the fatty acid compositions of cholesteryl esters (CE) and triacylglycerols (TG) in the milk were determined. The mean concentration of total milk lipid was 3.01 gm/100 ml of milk±.42 SD. The major fatty acids esterified with CE and TG were 16∶0,cis 18∶1 and 18∶2. The patterns were similar except for a greater proportion ofcis 18∶1 in the CE. The majortrans fatty acid detected was the 18∶1 isomer which accounted for 4.48% of the TG fatty acids and 2.96% of the CE fatty acids. The effect of dietary α-linolenic acid (18∶3n−3) and its ratio to linoleic acid (18∶2n−6) on platelet and plasma phospholipid (PL) fatty acid patterns and prostanoid production were studied in normolipidemic men. The study consisted of two 42-d phases. Each was divided into a 6-d pre-experimental period, during which a mixed fat diet was fed, and two 18-d experimental periods, during which a mixture of sunflower and olive oil [low 18∶3n−3 content, high 18∶2/18∶3 ratio (LO-HI diet)], soybean oil (intermediate 18∶3n−3 content, intermediate 18∶2/18∶3 ratio), canola oil (intermediate 18∶3n−3 content, low 18∶2/18∶3 ratio) and a mixture of sunflower, olive and flax oil [high 18∶3n−3 content, low 18∶2/18∶3 ratio (HI-LO diet)] provided 77% of the fat (26% of the energy) in the diet. The 18∶3n−3 content and the 18∶2/18∶3 ratio of the experimental diets were: 0.8%, 27.4; 6.5%, 6.9; 6.6%, 3.0; and 13.4%, 2.7, respectively. There were appreciable differences in the fatty acid composition of platelet and plasma PLs. Nevertheless, 18∶1n−9, 18∶2n−6 and 18∶3n−3 levels in PL reflected the fatty acid composition of the diets, although very little 18∶3n−3 was incorporated into PL. Both the level of 18∶3n−3 in the diet and the 18∶2/18∶3 ratio were important in influencing the levels of longer chain n−3 fatty acid, especially 20∶5n−3, in platelet and plasma PL. Production of 6-keto-PGF was significantly (P<0.05) higher following the HI-LO diet than the LO-HI diet although dietary fat source had no effect on bleeding time or thromboxane B2 production. The present study showed that both the level of 18∶3n−3 in the diet and its ratio to 18∶2n−6 were important in influencing long-chain n−3 fatty acid levels in platelet and plasma PL and that prostanoid production coincided with the diet-induced differences in PL fatty acid patterns.  相似文献

The influence of dietary partially hydrogenated marine oils containing docosenoic acid on rat heart mitochondrial membrane phospholipid fatty acid composition was studied with particular reference to cardiolipin and oxidative phosphorylation. Five groups of male weanling rats were fed diets containing 20% (w/w) peanut oil (PO), partially hydrogenated peanut oil (HPO), partially hydrogenated Norwegian capelin oil (HCO), partially hydrogenated herring oil (HHO), and rapeseed oil (RSO) for 10 weeks. All the cardiac phospholipids investigated were influenced by the experimental diets. An increased amount of arachidonic acid observed in phosphatidylethanolamine (PE) after feeding partially hydrogenated oils suggests a changed regulation of the arachidonic acid metabolism in comparison with PO treatment. 22∶1 originating from the dietary oils was incorporated only to a small extent into phosphatidylcholine (PC) and PE. A selective incorporation of 18∶1 isomers into the 1- and 2-positions of PC and PE with respect to geometry and position of the double bond was observed. Large amounts of 18∶1trans were incorporated into the 1-position of PC and PE, irrespective of the amount of 18∶2 supplemented to the diets, replacing a considerable proportion of stearic acid in this position. After feeding HHO and RSO, the content of 22∶1 in mitochondrial cardiolipin of rat heart was found to be 3% (mainly cetoleic acid) and 10% (mainly erucic acid), respectively, indicating a high affinity forcis isomers of 22∶1, but also a considerable resistance against incorporation oftrans isomers was observed. The ability of rat cardiac mitochondria to oxidize palmitoylcarnitine and to synthesize ATP was depressed after feeding HHO and RSO. Dietarycis isomers of 22∶1 seem to have a specific ability to interfere with cardiac ATP synthesis and also to alter the fatty acid composition of cardiolipin of rat heart.  相似文献

The incorporation of dietary isomeric fatty acids into the membranes of liver mitochondria was investigated. Three groups of rats were fed diets containing 3% sunflower seed oil plus 15%, 20%, or 25% partially hydrogenated arachis oil. A fourth group was fed 25% partially hydrogenated arachis oil, but no sunflower seed oil. All diets were given for 3, 6, or 10 weeks. After 10 weeks, the content oftrans fatty acids in the lipids of the mitochondrial membranes was 15–19% of the total fatty acids. The composition of thetrans- and thecis-octadecenoic acids in the lipids of the mitochondrial membranes was similar for all groups supplemented with sunflower seed oil (SO), irrespective of time and dietary level of partially hydrogenated arachis oil (HAO). Thecis 18∶1 (n−8), which was a major isomer of the partially hydrogenated arachis oil, was almost excluded from the mitochondrial fatty acids. Likewise, the content oftrans 18∶1 (n−8) was considerably lower in the mitochondrial lipids than in the diet. On the contrary, the content oftrans 18∶1 (n−6) was higher in the mitochondrial lipids than in the diet. In the group fed without sunflower seed oil, isomers of linoleic acid and arachidonic acid were observed in the lipids of mitochondrial membranes. In order to determine the minimal amount of dietary 9-trans,12-trans-linoleate which can decrease endoperoxide metabolites synthesized and their precursor in rat platelets, graded amounts (0, 0.1, 0.5, 1.0, 2.5%) of thetrans-linoleate were fed to rats with a constant amount of all-cis-linoleate (2.5%) for 12 weeks. Arachidonic acid levels in platelet phospholipids of groups receiving thetrans-linoleate at 2.5 and 1.0% were significantly (p<0.01) lower than that of the control receiving notrans-linoleate. Concentrations of TXB2 and PGF in sera of the group receiving 2.5%trans-linoleate were significantly (p<0.05) lower than those of the control; however, there was no difference between the group receiving 1.0%trans-linoleate and the control. To determine whether the difference in serum concentrations of endoperoxide metabolites could be manifested if rats were fed for longer period of time, 2 groups of rats were again fed diets containing 0 and 1.0%trans-linoleate, respectively, for 16 weeks. Arachidonic acid in platelet phospholipids of the group receiving thetrans-linoleate was again significantly (p<0.01) lower than that of the control group. Concentrations of TXB2 and PGF, and 12-hydroxyeicosatetraenoic acid formed in platelets, were smaller in the group receivingtrans-linoleate than the control group; however, the difference was not statistically significant. These results indicated that all-trans-linoleate can reduce arachidonic acid metabolites formed in rat platelets when its dietary level is equal to or exceeds the level of all-cis-linoeate.  相似文献

Pigs were fed a commercial conjugated linoleic acid (CLA) mixture, prepared by alkali isomerization of sunflower oil, at 2% of the basal diet, from 61.5 to 106 kg live weight, and were compared to pigs fed the same basal diet with 2% added sunflower oil. The total lipids from liver, heart, inner back fat, and omental fat of pigs fed the CLA diet were analyzed for the incorporation of CLA isomers into all the tissue lipid classes. A total of 10 lipid classes were isolated by three-directional thin-layer chromatography and analyzed by gas chromatography (GC) on long capillary columns and by silver-ion high-performance liquid chromatography (Ag+-HPLC); cholesterol was determined spectrophotometrically. Only trace amounts (<0.1%; by GC) of the 9,11–18∶2 cis/trans and trans, trans isomers were observed in pigs fed the control diet. Ten and twelve CLA isomers in the diet and in pig tissue lipids were sepatated by GC and Ag+-HPLC, respectively. The relative concentration of all the CLA isomers in the different lipid classes ranged from 1 to 6% of the total fatty acids. The four major cis/trans isomers (18.9% 11 cis, 13 trans-18∶2; 26.3% 10 trans, 12 cis-18∶2; 20.4% 9 cis, 11 trans-18∶2; and 16.1% 8 trans, 10 cis-18∶2) constituted 82% of the total CLA isomers in the dietary CLA mixture, and smaller amounts of the corresponding cis,cis (7.4%) and trans,trans (10.1%) isomers were present. The distribution of CLA isomers in inner back fat and in omental fat of the pigs was similar to that found in the diet. The liver triacylglycerols (TAG), free fatty acids (FFA), and cholesteryl esters showed a similar patterns to that found in the diet. The major liver phospholipids showed a marked increase of 9 cis,11 trans-18∶2, ranging from 36 to 54%, compared to that present in the diet. However, liver diphosphatidylglycerol (DPG) showed a high incorporation of the 11 cis,13 trans-18∶2 isomer (43%). All heart lipid classes, except TAG, showed a high content of 11 cis,13 trans-18∶2, which was in marked contrast to results in the liver. The relative proportion of 11 cis,13 trans-18∶2 ranged from 30% in the FFA to 77% in DPG. The second major isomer in all heart lipids was 9 cis,11 trans-18∶2. In both liver and heart lipids the relative proportions of both 10 trans,12 cis-18∶2 and 8 trans,10 cis-18∶2 were significantly lower compared to that found in the diet. The FFA in liver and heart showed the highest content of trans,trans isomers (31 to 36%) among all the lipid classes. The preferential accumulation of the 11 cis,13 trans-18∶2 into cardiac lipids, and in particular the major phospholipid in the inner mitochondrial membrane, DPG, in both heart and liver, appears unique and may be of concern. The levels of 11 cis,13 trans-18∶2 naturally found in foods have not been established.  相似文献

Chicks were fed isocaloric and isonitrogenous diets containing 6% (w/w) added fat consisting of various proportions of animal tallow and flaxseed oil (FSO). No differences among treatments were seen in growth rate, muscular deposition of protein and lipids nor in the muscle phospholipid (PL) and triglyceride (TG) contents. Prostaglandin (PG)E2 synthesis in isolated skeletal muscle was depressed significantly by feeding FSO or by treatment with naproxen (6-methoxy-α-methyl-2-napthaleneacetic acid), an inhibitor of PG synthesis. The changes associated with diet may be related to differences in the fatty acid composition of muscle lipids. Levels of saturated fatty acids in muscle PL and TG were relatively insensitive to dietary treatments. Monounsaturated fatty acid levels were significantly lower in the FSO-fed groups. FSO diets caused significant depression in muscle PL 20∶4ω6 and almost completely inhibited 22∶5ω6 incorporation. FSO diets decreased ratios of ω6/ω3 fatty acids and increased the unsaturation index of muscle PL. Muscles of chicks fed FSO showed increased levels of 18∶3ω3, and of its derivatives 20∶4ω3 and 22∶5ω3. These results suggest that FSO inhibits PG synthesis and modifies the fatty acids of PL and TG of chick muscle. These changes may have implications for PG-dependent and/or membrane-dependent processes in muscle metabolism.  相似文献
