Rats (6 per group) were fed semipurified diets containing either particulate fibers (alfalfa, 10%; cellulose, 10%; bran, 10%), a soluble ionic fiber (pectin 5%), soluble, nonionic fibers (guar gum, 5%; Metamucil, 10%), a mixed fiber preparation (Fibyrax, 10%, or an insoluble, ionic bile acid-binding resin (cholestyramine, 2%). The control group was fed the unsupplemented diet. The feeding period, during which diet and water were provided ad libitum, was 28 days. Compared with the control group, serum total cholesterol levels were increased by more than 10% in rats fed alfalfa and decreased by more than 10% in rats fed cellulose, guar gum, Fibyrax and cholestyramine. There were no significant differences in percentage of plasma HDL cholesterol. Serum triglycerides were elevated in the groups fed alfalfa, pectin, guar gum or Fibyrax and reduced in the group fed Metamucil. Plasma phospholipids were elevated in rats fed alfalfa or bran, unaffected in rats fed pectin or Metamucil and reduced in the other groups. Liver total cholesterol was elevated in all groups but those fed wheat bran and cholestyramine. The percentage of liver cholesterol present as ester was elevated in every group except that fed cholestyramine. Liver triglycerides were reduced in rats fed guar gum or Metamucil and elevated in those fed alfalfa. Liver phospho-lipids were lowered in the group fed cellulose. Liver phospholipids were fractionated by thin layer chromatography to give phosphatidylcholine (PC), phosphatidylethanolamine (PE), sphingomyelin (Sph), lysophosphatidylcholine (LPC) and phosphatidylinositol plus phosphatidylserine (PI+PS). PC was elevated in all test groups (7–25%); PE levels ranged from 14% below to 0.3% above controls; Sph levels were sharply lower (20–53%) in all groups. LPC and PI+PS levels were close to the control value in all test groups. The results demonstrate that different dietary fibers can affect liver phospholipid composition. In view of the critical roles of phospholipids in many biological reactions, it will be interesting to survey the influence of dietary fiber on phospholipid spectra of other tissues. Different soluble dietary fibers known to alter cholesterol metabolism were fed to golden Syrian hamsters, and their specific impact on lipoproteins, biliary bile acid profile, and fecal sterol excretion was evaluated. Semipurified diets containing 20% fat; 0.12% cholesterol; and 8% of psyllium (PSY); high (hePE) and low (lePE) esterified pectin; or high (hvGG) and low (lvGG) viscous guar gum were fed for 5 wk. Compared to control, PSY caused a significant reduction in plasma cholesterol (2.9±0.5 vs. 5.5±0.5 mmol/L), whereas hePE, lePE, hvGG, or lvGG had no apparent effect on plasma lipids. Hepatic total and esterified cholesterol were substantially decreased with PSY, pectin and guar gum, whereby PSY produced the most pronounced effect. Distinctive changes existed in the bile acid profile related to the different fibers. In contrast to pectin and guar gum, PSY caused a significant increase in the cholate:chenodeoxycholate and the glycine:taurine conjugation ratio. Pectin and guar gum did not alter daily fecal neutral sterol excretion while PSY caused a 90% increase due to a higher fecal output. Daily fecal bile acid excretion and total fecal bile acid concentration were significantly increased by PSY, whereas hePE, lePE, hvGG, and lvGG revealed no or only minor effects. Taken together, the disparate hypocholesterolemic effects of PSY, pectin, and guar gum on cholesterol and bile acid metabolism in the hamster are possibly related to different physicochemical properties, e.g., viscosity and susceptibility to fermentation, affecting the fiber-mediated action in the intestine.  相似文献

Guar gum is an effective agent for use as a natural adhesive ingredient that can be used to replace the hazardous ingredients of spirit gum. This study describes the possibility of using guar gum as a renewable substituent for cosmetic adhesive. An adhesive base and cosmetic adhesive containing guar gum were prepared by a two-step process. The samples were tested for safety (cell toxicity test and patch test), stability (centrifugation, cycling and viscosity), and effectiveness (tensile strength). The results from the MTT assay show that the growth activity of human fibroblast skin cells was over 89% in all concentrations of cosmetic adhesive containing guar gum. In addition, no special skin reactions were reported in the patch test prepared adhesive containing guar gum. Moreover, the stability test demonstrated proper stability of all adhesive samples: the composition stability, heat stability, and viscosity of the adhesive samples maintained stable conditions. The efficacy test confirmed the superiority of the guar gum adhesive samples over spirit gum concerning the tensile strength. This study demonstrated that guar gum may be a viable replacement for synthetic rosins and also as a substituent in cosmetic.  相似文献

The adsorption of guar gum on illite and dolomite was studied as a function of polymer concentration and ionic strength. The adsorption results were correlated with colloid stability of aqueous suspensions of these two minerals. The adsorption density of guar gum on illite was not affected by the ionic strength of the system. Guar gum adsorption on dolomite was found to strongly decrease at higher electrolyte concentrations. Low concentrations of guar gum brought about flocculation of the minerals while higher dosages led to steric re‐dispersion. Results presented here are compared with previously published results for carboxymethyl cellulose (Pawlik et al., J. Colloid Interface Sci. 260 , 251‐258 (2003)).  相似文献

The effect of the type of dietary fiber on the bile acid and taurine metabolism was examined in rats. Diets containing 10% of various water-soluble fibers (citrus pectin, konjak mannan, guar gum) as compared to a fiber-free diet increased biliary excretion of total bile acids. In contrast, water-insoluble dietary fibers (cellulose, corn bran, chitin; 10% in the diets) as well as cholestyramine (5% in the diet) considerably, decreased bile acid excretion. Water-soluble dietary fibermediated increases in bile acid excretion were totally attributable to increases in glycine-conjugates. Thus, these fibers greatly increased by the bile acid glycine-to-taurine ratio (G/T). Excretio of glycine conjugates decreased more than that of taurine conjugates in rats fed various water-insoluble dietary fibers. As a results, G/T in rats fed water-insoluble fibers was significantly lowered as compared to G/T in animals fed a fiber-free diet. Cholestyramine did not affect the G/T ratio of bile acids. Fecal bile acid excretion and the activities of hepatic cholesterol 7α-hydroxylase (EC 1.14.13.17) in rats fed various water-soluble dietary fibers approximately doubled as compared to the respective values for rats fed a fiber-free diet. Whereas cholestyramine greatly increased these parameters, water-insoluble fibers did not significantly affect them. Various water-soluble fibers decreased hepatic concentration and urinary excretion of taurine as well as the activity of hepatic cysteine dioxygenase (EC 1.13.11.20). In contrast, water-insoluble fibers considerably increased hepatic taurine concentrations and enzyme activities. The parameters for taurine metabolism were unaffected by cholestyramine. It was suggested that the types of dietary fiber affected hepatic taurine synthesis and thus modified bile acid glycine/taurine ratios.  相似文献

This study investigated the potential effect of shear rheology and humic acids (HA) on the subsurface transport of polymeric fluids used for the remediation of contaminants. Polymeric fluids were prepared with guar, scleroglucan, and carboxymethyl cellulose (CMC). Guar fluids can be used to suspend reactive particles for contaminant degradation. Fluids prepared with 2.5 g/L of guar in water were viscous, and the crosslinker borax (1 g/L) made them viscoelastic. Microfluidics experiments showed that the increase in elasticity blocked the flow of guar in 350 μm channels. Guar, CMC, or scleroglucan fluids containing sodium thiosulfate can be used to trap toxic Cr(VI) in the subsurface and reduce it to harmless Cr(III). Trapping of Cr(VI) is achieved by the gelation of the fluids upon contact with chromium. Before mixing with chromium, HA did not affect the flow of CMC, guar, and scleroglucan in microfluidic channels. Quartz-crystal microbalance with dissipation monitoring experiments indicates that HA reduced sorption of guar onto silica, potentially promoting the transport of guar fluids in sandy aquifers. While HA slightly decreased the rate of gelation of CMC and scleroglucan upon contact with chromium, it did not affect the fast gelation rate of guar. © 2019 Wiley Periodicals, Inc. J. Appl. Polym. Guar galactomannan (referred to as guar gum) is a versatile polysaccharide, obtained from the seeds of the shrub Cyamopsis tetragonolobus, which finds several applications in either its native or chemically modified form. For textile printing, guar gum can also be partially depolymerised in order to promote dye penetration, improve swelling in water and achieve the desired rheological properties. Guar gum is obtained from guar seeds by a thermo‐mechanical process that leaves ca. 3% of largely insoluble proteins in the gum, originating from the endosperms aleurone layer. When printing silk fabrics with acid or premetallised dyes, guar endogenous insoluble proteins bind tightly to anionic dyes, causing deposition of coloured aggregates on the fabric. This causes imperfections on the printed fabric in the form of tiny, but visible, ‘dots’, which lowers the quality of the final articles. In order to eliminate ‘dotting’, a novel printing thickener composed of depolymerised guar gum mixed with a bioengineered subtilisin protease has been developed. Upon solubilisation of the gum, and during preparation of the printing paste mixture, the protease hydrolyses guar gum insoluble proteins, generating soluble peptides that are washed off by the post‐printing treatments of the fabric. This enzymatic application prevents ‘dotting’ and significantly improves the quality of the silk print, without any measurable tensile strength loss of the fabric.  相似文献

The alginate thickener is the thickener frequently used for reactive printing of textile. The thickener responds with reactive pigments and thus does not lead to the fabric composition becoming stiffer. In this study, we prepared oxidised natural guar gum with hydrogen peroxide, sodium hypochlorite and sodium hydroxide. All other polysaccharides comprise reactive hydroxyl units with a stronger reactivity that must be replaced if they are to be used in reactive printing. Guar derivatives were synthesised and verified using Fourier-transform infrared (FTIR) spectroscopy. Natural thickeners, synthetic guar gum derivatives, have been employed in textile printing technique. In comparison to other synthetic thickeners, modified environmental guar gum polymer has been shown to be an ecologically friendly and low-cost thickener. Cotton fabric printed with modified guar thickening with hydrogen peroxide has even stronger colour strength than fabric printed with sodium alginate thickener, which is highly favourable. Penetration properties, colour value, colour strength, colour fastness to washing, light and rubbing was compared with alginate thickener (readily available on the market). Guar gum thickeners showed enhanced features versus sodium alginate for reactive printing. Partially replaced guar gum is an appropriate option due to the colour and physical properties.  相似文献

Guar gum (GG) has been partially methyl-etherified under heterogeneous reaction conditions. The resulting products, (methyl ether guar: MG) with different degrees of substitution, have been characterized by means of viscosity, 1H NMR, and FTIR measurements. The introduction of methoxyl groups along the polysaccharidic chains reduces the hydrogen bonding sites on the guar backbone reducing primarely the extent of hydrogen bonding between guar macromolecules, hence their aggregation tendency. A comparative analysis of Mark–Houwink–Sakurada parameters and of the characteristic ratio (C) of GG and MG samples in aqueous solution has been carried out using the Burchard–Stockmayer–Fixman method for flexible and semiflexible chains. The MG chains exhibit more flexibility than those of native guar gum which is traceable to a disruption of intrachain hydrogen bonds.  相似文献

Cotton linter nano-fibers (CLNFs) were prepared from cotton linters by a refining process. The prepared CLNFs were characterized for morphology, crystallinity and degree of polymerization. CLNF was used as a reinforcing agent in guar gum to improve its performance properties. Guar gum/CLNF nanocomposite films were prepared by a solution-casting process. CLNF was added in concentrations of 0.1, 0.25, 0.5 and 1.0 % (w/w) in guar gum. The prepared guar gum/CLNF nano-composite films were characterized for mechanical, thermal, rheological, crystallinity, water vapor transmission rate (WVTR) and light transparency properties. The enthalpy of melting and melting temperature of guar gum increased with increased concentration of CLNF; but up to 0.25 % (w/w) concentration, above which they started decreasing. Tensile strength and Young’s modulus of guar gum increased by 32 and 35 %, respectively, by 0.25 % (w/w) addition of CLNF; however, it decreased on further increase in the concentration of CLNF. The percentage elongation at break and WVTR decreased by 58 and 57 % for 0.25 % (w/w) CLNF-added guar gum. The observed improvements in the properties were due to better interaction between CLNF and guar gum. CLNF was found to have uniformly dispersed in guar gum on addition up to 0.25 % concentration; however, it started forming aggregates at higher concentration, as evident from scanning electron microscopy. Viscosity increased, whereas transparency decreased with increased concentration of CLNF in guar gum.  相似文献

Guar gum is a galactomannan commonly used as a viscosity modifier in the food, pharmaceutical, and cosmetics industry. The aim of this study was to synthesize acryloyloxy guar gum via a Schotten‐Bauman reaction in aqueous media. The reaction products were characterized using FTIR, C13‐NMR, wide angle X‐ray diffraction techniques to ascertain the effect of acrylation on the structure of guar gum. The acrylation of guar gum was found to be limited to the primary hydroxyl groups on the guar gum molecule. The maximum degree of substitution (DS) was found to be 0.56, which was observed after 3 h of reaction. Since the reaction was carried out in an aqueous medium after 3 h of reaction the DS of the derivatised guar gum was found to decrease, because of hydrolysis of the formed ester linkages. The ester content and intrinsic viscosity of the derivatised guar gums were also evaluated. Thermal analysis showed that a higher DS resulted in products with lower thermal stability and there was no evidence of reaction via the acrylate groups on heating. © 2010 Wiley Periodicals, Inc. Guar gum was partially carboxymethylated by the reaction of the base polysaccharide with the sodium salt of monochloroacetic acid in presence of sodium hydroxide. The resulting products (carboxymethyl guar with different degrees of substitution) were characterized by a variety of material characterization techniques, such as intrinsic viscosity measurement, determination of molecular weight, elemental analysis, thermal analysis, 13C‐NMR spectra, and Fourier transform infrared analysis. The flow properties of mixed solution of guar gum and carboxymethyl guar gum, including shear-thinning and viscosity synergism, have been studied at different mixed ratios, shear rates and temperatures using rheometer. The results show that addition of carboxymethyl guar gum enhances shear-thinning behavior and increases viscosity in guar gum solution at a given range of mixed ratios. The temperature-dependence of the mixed solution is markedly different from a single solution. The activation energy of blends with 50 and 75% of guar gum were determined to be 11.02 and 10.39 kJ mol−1, respectively, lower than that of a single solution. These results indicate that the network structure of viscous solution has been changed because carboxymethyl groups branched in galactomannans had changed the interaction among macromolecular chains.  相似文献

A novel pretreatment paste containing fluoroacrylate copolymer and modified Guar Gum was carried out on the polyester fabric to provide partial water repellency and printability after inkjet printed with water‐based disperse dye inks. The stable fluorinated pastes were prepared via miniemulsion polymerization of fluoroalkyl acrylates, styrene, 2‐ethylhexyl acrylate, and modified guar gum in the presence of AIBN as initiator, CTAB and polyoxyethylene polyaryl ether as surfactants under suitable reaction conditions. In the analysis of monomer conversion and particle size, the highest polymerization rate with smallest particle size was obtained with increasing concentration of surfactants and initiator but the decreasing degree of substitution of guar gum to imply stronger interactions between fluoroacrylate monomers, hydroxyl groups of modified guar gum and EO units of nonionic surfactants. The 30% of stock fluorinated paste with urea concentration of 6% provided the prints with highest K/S value, satisfactory edge acuity and least color difference between front and back sides of the fabric to show more transparency. The rating 80 of water repellency tested on the best treated polyester fabric demonstrated stain proof properties after sprayed with water and dried. © 2009 Wiley Periodicals, Inc. Soybean protein and casein supplemented with 1% Arg were compared for their ability to prevent fatty livers caused by excess dietary Lys. The concentrations of serum lipids and lipoproteins of rats fed 5% Lys and having vatty livers were also compared with those of rats fed the identical diet but lacking fatty livers when killed. The total liver lipids, triglycerides and cholesterol of rats fed 15% casein +5% Lys were 3.9, 12.4 and 2 times control values, respectively. Rats fed 5% Lys +1% Arg or 5% Lys with 15% soybean protein had liver lipid concentrations similar to controls fed no supplemental Lys. Serum total lipids, triglycerides, cholesterol, phospholipids and free fatty acids also did not change, and serum ketone bodies were slightly elevated with Lys feeding whether the rats had fatty livers or not. The concentrations of circulating HDL were slightly depressed in all rats fed 5% Lys while LDL were significantly elevated, particularly in rats without fatty livers. Serum VLDL did not change with 5% dietary Lys. Overall, excessive dietary Lys caused fatty livers which were prevented by varying the diet or length of feeding. Excess Lys feeding altered lipoprotein metabolism shown by decreased serum HDL and a substantial elevation in LDL. The latter was more apparent when the fat accumulation in liver was less severe or absent. The data suggest that the fatty liver from Lys excess is probably unrelated to increased fat mobilization from storage, decreased fat oxidation or to a major block in the transport of triglycerides from the liver to the circulation.  相似文献

Mixed micelles were prepared containing combinations of either taurocholate or taurochenodeoxycholate, monoolein, oleic acid, dioleylphosphatidylcholine (lecithin) and cholesterol. These were incubated with commercial bile-acid-sequestering resins, cholestyramine and DEAE-Sephadex, or various dietary fibers and fiber components including wheat bran, cellulose, alfalfa, lignin and 2 viscosity grades of guar gum. Binding was determined as the difference between the radioactivity of each micellar component added and that recovered in the centrifugal supernatant after incubation. In general, the extent of bile salt sequestration was characteristic and reproducible for each bile salt, and was largely unaffected by the presence of one or more additional components of the micellar mixture, including the other bile salt. Cholestyramine bound 81–92% of the bile salts and 86–99% of the phospholipid and cholesterol present in micelles. DEAE-Sephadex sequestered only 49% of the taurocholate and 84% of the taurochenodeoxycholate, but completely removed all of the phospholipid and cholesterol from micelles containing either bile salt. Among the dietary fibers, guar gum of either viscosity bound between 20–38% of each micellar component, whereas lignin, alfalfa, wheat bran and cellulose were progressively less effective in sequestratin of individual components of mixed micelles. The extent of sequestration of micellar components by these resins and fibers is reasonably correlated with the effects of these same materials on lymphatic absorption of lipids and to their suggested hypocholesteremic properties.  相似文献

Rats were fed cholesterol, cacao butter, or olive oil diets to determine the effect of dietary lipids on the rate of drug biotransformation in the liver and duodenum. The cholesterol rich diet maintained the hepatic aryl hydrocarbon hydroxylase activity at the same level as did the standard diet. Rats fed olive oil and cacao butter diets showed lower hepatic aryl hydrocarbon hydrorylase activity. The p-nitroanisole 0-demethylase activity was doubled in hepatic microsomes of rats fed the high cholesterol diet when compared to rats fed the standard diet. The hepatic uridine diphosphate glucoronosyltransferase activity showed different patterns depending on the in vitro treatment of the microsomal membranes. If the enzyme activity was assayed from the native, untreated microsomes, an increase in the measurable uridine diphosphate glucuronosyl transferase activity was found in rats having cholesterol rich diet. After the in vitro activation of membrane-bound uridine diphosphate glucuronosyltransferase by trypsin, the increase in measurable activity was 10 fold in the group fed the standard diet, 6 fold in group fed cholesterol, 4 fold in group fed cacao butter, and 3 fold in group fed olive oil. Trypsin digestion of microsomes increased the measurable uridine diphosphate glucuronosyltransferase activity less in rats fed diets rich in neutral fats than those fed the standard diet. In the duodenal mucosa, lipid diets decreased the activities of drug hydroxylation and glucuronidation.  相似文献

This study investigated the effect of two partially hydrolyzed guar gums (PHGG) on fatty acid and sterol excretion. PHGG were obtained by chemical hydrolysis of guar gum (GG) with H2O:EtOH (1:1) at 100 °C for 1 h (PHGG1) or 2 h (PHGG2). The viscosity of the PHGG in a 1 % (w/v) aqueous solution corresponded to that of a pseudoplastic fluid and was higher for PHGG1 than PHGG2. Guinea pigs (n = 8 per group) were fed high fat diets (17/100 g) that contained 12/100 g of cellulose, PHGG1, or PHGG2 for 4 weeks. Despite the differences in viscosity, the two PHGG exerted similar physiological effects. Compared to the control cellulose group, the body weight gain was lower in animals fed PHGG, although no effect on food consumption was observed. PHGG increased the excretion of fatty acids and neutral sterols, but not bile acids. Consumption of PHGG did not alter the fecal fatty acid profile, while intestinal bioconversion of sterols tended to increase in response to PHGG2. A reduction in the viscosity within the range tested did not correlate with losses in the hypocholesterolemic capacity of PHGG as both were effective in reducing plasma cholesterol. Thus, we conclude that the chemical hydrolysis of guar gum renders the gum suitable for inclusion in food products without significantly altering its beneficial health effects.  相似文献
