The collection of calf birth weight on US dairies is not a common practice. Calf birth weight was collected on 3 dairies (2 Holstein herds and 1 Jersey herd) over a 6-wk period. All calf birth weights were collected less than 2 h after birth. A total of 872 calves were weighed by a spring scale and their weight was also estimated using a hoof circumference tape, with both weights and sex recorded. The general linear models procedure (PROC GLM; SAS Institute Inc., Cary, NC) was used to estimate least squares means for spring scale birth weight along with sex of the calf and dairy. Calf spring scale versus hoof circumference-estimated birth weight was compared using the regression procedure (PROC REG; SAS Institute Inc.). Calf birth weight was also broken down into increments, and spring scale versus hoof circumference-estimated birth weight was compared by t-test. Bull calves had a heavier birth weight compared with heifer calves [36.7 ± 0.4 kg (n = 450) vs. 34.6 ± 0.4 kg (n = 422), respectively]. The dairy on which calves were born had a significant effect on calf birth weight [dairy 1: 36.8 ± 0.4 kg (n = 204); dairy 2: 39.5 ± 0.2 kg (n = 463); dairy 3: 25.9 ± 0.4 kg (n = 205)]. When the spring scale calf birth weight was linearly regressed by hoof circumference-estimated birth weight, a significant relationship was observed, with an R2 value of 0.91. For calves weighing less than 31.3 kg, the hoof circumference tape overestimated calf birth weight compared with the spring scale. However, for calves that weighed between 31.3 and 44.9 kg, no significant difference was observed between spring scale and hoof circumference tape-estimated birth weight. For calves weighing greater than 44.9 kg, the hoof circumference tape underestimated birth weight compared with the spring scale. Collection of calf BW by spring scale or estimated by hoof circumference tape appeared to be comparable for most calves because most calves weighed between 31 and 45 kg, but caution should be used for calves with a light or heavy birth weight.  相似文献

Serological testing in the early stages of Johne’s disease has been successful using specific antigens and in-house ELISA. However, the use of a commercial ELISA has not been evaluated shortly after Mycobacterium avium subspecies paratuberculosis (MAP) infection, nor has it been determined whether this serological response is age or dose dependent. Fifty-six calves were randomly allocated to challenge groups (5 per group) and a negative control group. Calves were inoculated orally on 2 consecutive days at 2 wk or at 3, 6, 9, or 12 mo. Within each age group, 5 calves received either a high or low dose of MAP. Using a commercial ELISA, antibody responses were detected in 42% of the inoculated calves and were present in all age and dose groups (except for the 6-mo low-dose group). Antibody response profiles differed among individual calves; persistent as well as peak and bimodal peak responses existed. Calves inoculated at 12 mo were ELISA positive within 4.5 mo after inoculation, whereas those inoculated at younger ages took longer to become ELISA positive. Furthermore, calves inoculated with a high dose of MAP more often became ELISA positive than low-dose calves when inoculated at a younger age. In conclusion, a dose-dependent antibody response was detected by ELISA in a larger proportion of calves than expected soon after inoculation.  相似文献

The objective of this study was to evaluate the possible transmission of Mycoplasma bovis from positive sand bedding to naïve dairy calves. Twelve preweaned Holstein bull calves were blocked in pairs and randomly assigned as unexposed controls (n = 6) bedded with control sand, or exposed calves (n = 6) bedded with sand previously positive for M. bovis at a dairy farm. Bedding sand was cultured weekly. Nasal and ear swabs and sera were collected weekly, tracheal swabs were collected monthly, and by the end of the 105-d study, all calves were euthanized (n = 10) or died (n = 2). Sera were tested for M. bovis-specific antibody. Mycoplasma spp. culture was performed on nasal and ear swabs; culture and a PCR differentiating multiple Mycoplasma spp. were performed on postmortem samples of lung, retropharyngeal lymph node, and trachea from each calf. A complete necropsy also was performed. During 6 wk, mycoplasma concentration in exposed group sand was between 200 and 32,000 cfu/g. All 166 tracheal swabs, nasal and ear swabs, and postmortem tests from all calves were negative for mycoplasma. All 94 sera were negative for M. bovis-specific antibody. No gross pathology suggestive of mycoplasma disease was detected. The probability of mycoplasma detection, if an exposed calf had become infected 4 wk after exposure, ranged between 97 and 99% depending on time of exposure for individual calves. There was no evidence that sand bedding contaminated with M. bovis might serve as a source of transmission to naïve dairy calves.  相似文献

Mycobacterium avium ssp. paratuberculosis (MAP) is the causative agent of Johne's disease (JD). One mode of transmission of MAP is through ingestion of contaminated milk and colostrum by susceptible calves. The objective of this study was to determine if the amount of MAP shed into the milk and colostrum of infected cows was affected by severity of infection as well as the number of days in milk (DIM). Milk was collected over the 305-d lactation period from naturally infected cows in the asymptomatic subclinical (n = 39) and symptomatic clinical (n = 29) stages of disease, as well as 8 noninfected control cows. All milk samples were assayed for MAP by culture on Herrold's egg yolk medium and either BACTEC 12B (Becton Dickinson, Franklin Lakes, NJ) or para-JEM (Thermo Fisher Scientific, Trek Diagnostic Systems Inc., Cleveland, OH) liquid medium, and by direct PCR for the IS900 target gene. Mycobacterium avium ssp. paratuberculosis was detected in 3.8, 4.1, and 12.6% of milk samples collected from cows with subclinical JD after culture in Herrold's egg yolk medium, liquid medium, and direct PCR, respectively. The frequency of MAP positivity increased to 12.9, 18.4, and 49.2% of milk samples collected from cows with clinical JD by these same methods, respectively. None of the milk samples collected from control cows was positive for MAP by any detection method. Viable MAP was primarily isolated from milk and colostrum of subclinically and clinically infected cows collected in early lactation (DIM 0–60), with negligible positive samples observed in mid (DIM 60–240) and late (DIM 240–305) lactation. This study demonstrates that shedding of MAP into milk is affected by infection status of the cow as well as stage of lactation, providing useful information to producers to help break the cycle of infection within a herd.  相似文献

Salmonella enterica ssp. enterica Dublin is the most common Salmonella serotype found in the dairy sector in Denmark. Salmonella antibody level in bulk-tank milk (BTM), indicative of Salmonella Dublin infection in the herd, has been recorded regularly in all Danish dairy herds through a surveillance program since 2002. The objective of this study was to investigate whether high BTM Salmonella antibody level was associated with high calf mortality at herd level. Other risk factors for high calf mortality were also investigated: breed, production type (organic vs. conventional), number of animals purchased, herd size, and number of neighbor herds within a 4.9-km radius. Data from the Danish Cattle Database including the Salmonella surveillance program from September 2007 through August 2008 were used. Dairy herds with more than 20 cows were included (n = 4,337). Because of a highly right-skewed distribution of calf mortality with many zero values, calf mortality had to be dichotomized for the analysis. Therefore, in this study, high calf mortality was defined as calf mortality of more than 6.5% for calves aged 1 to 180 d. A logistic analysis was performed to identify risk factors associated with the probability of a herd having high calf mortality. The following factors were significantly associated with high calf mortality: high BTM Salmonella antibody level, odds ratio (OR) = 2.0 (95% confidence interval = 1.6-2.4), organic production OR = 1.4 (95% confidence interval = 1.1-1.7) for organic versus conventional production, and breed. Purchase of 8 or more animals increased the OR of high calf mortality more than purchase of 1 to 7 animals, which again had a higher OR compared with purchase of 0 animals. Because only 14.3% of the population consisted of herds with high BTM Salmonella status, the estimated proportion of herds with high calf mortality could only be reduced from 38.7 to 36.5% by eradicating Salmonella from the Danish cattle population (i.e., a population attributable risk of 2.2%). This showed that although there is a strong association between BTM Salmonella status and calf mortality, the problem with high calf mortality will not be solved by eradicating Salmonella. All other things equal, a population with more Salmonella-infected herds would gain a larger reduction in calf mortality from a Salmonella control campaign. Nevertheless, individual herds with a high within-herd prevalence of Salmonella are likely to benefit, both economically and regarding animal welfare, from controlling pathogenic Salmonella types in cattle.  相似文献

Mycobacterium avium ssp. paratuberculosis (MAP) is shed into the milk of cattle affected by Johne’s disease and, therefore, is a route of transmission for infection in young stock in dairy herds. The objective of this study was to validate a decontamination and culture protocol for the recovery of MAP from individual bovine milk samples from known infected herds. Decontamination of milk samples (n = 17) with either 0.75% hexadecylpyridinium chloride for 5 h or N-acetyl-l-cysteine-1.5% sodium hydroxide (NALC-1.5% NaOH) for 15 min before culture in BACTEC 12B (Becton Dickinson, Franklin, NJ), para-JEM [Thermo Fisher Scientific (TREK Diagnostic Systems, Inc.), Cleveland, OH], and Herrold’s egg yolk (HEY; Becton Dickinson) media was compared. Treatment with NALC-NaOH resulted in a lower percentage (6%) of contaminated samples than did treatment with hexadecylpyridinium chloride (47%), regardless of culture medium. The decontamination protocol (NALC-1.5% NaOH) was then applied to milk samples (n = 144) collected from cows at 7 US dairies. Recovery of viable MAP from the milk samples was low, regardless of culture medium, with recovery from 2 samples cultured in BACTEC 12B medium, 1 sample cultured in para-JEM medium, and no viable MAP recovered on HEY medium. However, 32 cows were fecal culture positive and 13 milk samples were positive by direct PCR, suggesting that several cows were actively shedding MAP at the time of milk collection. Contamination rates were similar across media, with 39.6, 34.7, and 41.7% of samples contaminated after culture in BACTEC 12B, para-JEM, and HEY media, respectively. Herd-to-herd variation had a major effect on sample contamination, with the percentage of contaminated samples ranging from 4 to 83%. It was concluded that decontamination of milk with NALC-1.5% NaOH before culture in BACTEC 12B medium was the most efficacious method for the recovery of viable MAP from milk, although the ability to suppress the growth of contaminating microorganisms varied greatly between herds.  相似文献

Effects of growth rate on fat-soluble vitamin and macro- and micromineral concentrations in the circulation of preruminant dairy calves were evaluated. Dietary treatments were designed to achieve 3 targeted rates of gain [no growth (NG) = 0.0 kg/d; low growth (LG) = 0.55 kg/d; or high growth (HG) = 1.2 kg/d] over a 7-wk period. Milk replacer (MR) intakes necessary to achieve these growth rates were estimated using the National Research Council's Nutrient Requirements of Dairy Cattle calf model computer program. All of the calves were fed a 30% crude protein, 20% fat MR reconstituted to 14% dry matter. The diets were formulated to ensure that protein was not a limiting nutrient. No-growth and LG calves were supplemented additionally with vitamins A, D, and E to compensate for treatment differences in dry matter intake relative to the HG calves; however, no attempt was made to adjust mineral intake based on MR consumption. Growth rates for NG (0.11 kg/d), LG (0.58 kg/d), and HG (1.16 kg/d) calves differed during the study. Health was minimally affected by growth rate and this was reflected by comparable and relatively low serum haptoglobin concentrations in all calves during the 7-wk period. Concentrations of serum retinol, 25-(OH)-vitamin D3, and zinc were unaffected by growth rate. The HG calves had lower RRR-α-tocopherol concentrations than NG and LG calves at wk 7, suggesting that the increased growth rate of HG calves was associated with increased utilization of vitamin E. Serum concentrations of all vitamins increased with age. Copper, calcium, and phosphorous concentrations in HG calves exceeded those in LG and NG calves during the latter weeks of the study, likely because of increased MR intake by HG calves. Fat-soluble vitamin and mineral concentrations for all treatment groups remained within ranges considered normal for preruminant calves.  相似文献

Mycobacterium avium ssp. paratuberculosis (MAP) infections cause major losses to the dairy industry. Transmission of MAP occurs primarily via feces and in utero, but MAP can also be excreted in colostrum and milk. The objective of this study was to determine whether colostrum and milk fed to calves are important risk factors for infection with MAP. A questionnaire was sent to 1,050 farms participating in the Danish control program on paratuberculosis in early 2007. Details on practices regarding colostrum and milk feeding between 1999 and 2006 were obtained from 808 (77%) herds. Nine vaccinated herds were excluded. Information on MAP antibody-ELISA results, date of birth, and herd of birth of 93,994 animals was obtained from the Danish Cattle Database. A 2-level logistic regression model was fitted with a dichotomized ELISA response, with outcome, age, source of colostrum, and milk as fixed effects, and herd as a random effect. Animals fed colostrum from multiple cows had an odds ratio of 1.24 of being ELISA positive compared with animals fed colostrum from their own dam only. Calves suckling with foster cows had an odds ratio of 2.01 of being ELISA positive compared with calves fed milk replacer. Feeding bulk tank milk and pooled milk from cows with high somatic cell counts did not increase the risk of being ELISA positive. Overall, the results of the study suggested that source of milk was not of great importance for the transmission of MAP, but colostrum should be fed only from the dam of that calf.  相似文献

The objective of this study was to evaluate the feasibility of using the preruminant dairy calf as a model for evaluating effects of vitamin D status in the neonate. Because the newborn calf can be sustained during the first weeks of life solely on a fluid diet having a defined composition, has documented nutritional requirements, and is minimally affected by repeated samplings of peripheral blood, it has the potential to serve as a model for characterizing nutrient-specific effects on the growth and health of the neonate. Colostrum-fed Holstein bull calves (n = 13) entered the trial at approximately 4 d of age. All calves were fed a custom-formulated milk replacer devoid of vitamin D. Plasma 25-hydroxyvitamin D3 concentrations in all calves were determined on a regular basis beginning at d 0. Using this information, low- and high-status groups of calves were established by subcutaneous administration of 25-hydroxyvitamin D3. To maintain targeted plasma 25-hydroxyvitamin D3 concentrations in low (<30 ng/mL) and high (>60 ng/mL) vitamin D-status calves, low-status calves (n = 6) received a total of 8,600 IU (2,225 IU/wk) of vitamin D during the experimental period and high-status calves (n = 7) received 54,000 IU (13,500 IU/wk). Concentrations of 25-hydroxyvitamin D3 in low-status calves averaged 27 ng/mL, compared with 78 ng/mL in high-status calves, and were less at all sampling times from d 7 to d 28. Concentrations of 1,25-dihydroxyvitamin D3 and 25-hydroxyvitamin D3 were not correlated. Calcium, magnesium, and phosphorous concentrations were unaffected by 25-hydroxyvitamin D3 administration; however, plasma calcium and 1,25-dihydroxyvitamin D3 concentrations were correlated. Calcium and magnesium concentrations decreased with age but remained within normal ranges for dairy cattle. These results indicate that it is possible to predictably control vitamin D status over a 28-d period and suggest that the preruminant calf might be useful as a model for studying effects of vitamin D on growth, development, and immune function in the neonate.  相似文献

The objective of this study was to investigate the effect of artificial mothering, simulated by verbal and physical stimulation of the newborn, on passive transfer of IgG in the dairy calf. Newborn heifer calves born without dystocia were removed from the dam before suckling and randomly assigned to 1 of 2 treatment groups: no tactile or verbal stimulation other than that required for feeding (group 1; n = 20), or artificial mothering, consisting of 15 min of vigorous physical and verbal stimulation conducted within 1 to 2 h of birth at the time of colostrum feeding and repeated 1 to 2 h later (group 2; n = 21). All calves were fed 2.25 L (150 g of IgG) of a commercially available colostrum replacement using an esophageal tube feeder. Blood samples collected at 24 h of age showed that serum IgG levels and the apparent efficiency of absorption of the IgG were similar in both groups of calves. Artificial mothering by physical and verbal stimulation had no significant effect on IgG passive transfer in dairy heifers born without dystocia.  相似文献

Mycobacterium avium ssp. paratuberculosis (MAP) is the etiological agent of paratuberculosis, a chronic contagious bacterial disease primarily affecting dairy cattle. Paratuberculosis represents a dual problem for the milk production chain: in addition to economic losses to affected herds, MAP may have zoonotic potential. Infected herds must be identified in order to implement programs designed to reduce the incidence of disease within and between herds and to prevent MAP from entering the food chain. The objective of this study was to evaluate the sensitivity and specificity of a screening sampling plan (SSP) to detect MAP-positive dairy herds by repetitive analysis of bulk tank milk (BTM) samples by ELISA and in-line milk filter (ILMF) samples by PCR. Samples from BTM and ILMF were collected twice from 569 dairy herds in southern Italy. Additionally, 12,016 individual milk samples were collected: 9,509 from 102 SSP-positive herds (SSP MAP-positive) and 2,507 from 21 randomly selected SSP-negative herds (SSP MAP-negative). There was a total of 126 SSP MAP-positive herds (i.e., 21.3% SSP MAP-positive herds; 95% confidence interval = 18.0–24.9); the within-herd apparent prevalence (AP) ranged between 0.00 and 22.73% (mean 6.07%). A significant difference in within-herd AP was shown between SSP MAP-positive herds and SSP MAP-negative herds. A highly significant association was shown between the median AP herd status (>5%) and positivity to at least one ILMF or BTM sample. The SSP detected a minimum of 56.25% of low AP herds (AP ≤2.0%) up to a maximum of 100% of herds with a within-herd AP ≥8.0%. Overall, the SSP detected 85.57% of herds in which at least one individual milk sample was positive by ELISA. The proposed SSP was an inexpensive and useful tool to detect MAP-positive herds with a higher risk of infection diffusion and milk contamination. Although the SSP cannot be used for MAP-free certification of herds, it could be useful to prioritize appropriate control measures aimed at reducing the prevalence of infection in dairy herds and milk contamination.  相似文献

Developments in accelerometer technology offer new opportunities for automatic monitoring of animal behavior. Until now, commercially available accelerometers have been used to measure walking in adult cows but have failed to identify walking in calves. We described the pattern of acceleration associated with various gaits in calves and tested whether measures of acceleration could be used to count steps and distinguish among gait types. A triaxial accelerometer (sampling at 33 readings/s with maximum measurement at ±3.2 g) was attached to 1 hind leg of 7 dairy calves, and each calf was walked to a familiar large arena (29.1 × 4.8 m) and encouraged to walk and run for 8 to 10 min while being video recorded. The video recordings were watched in slow motion and a total of 54 recordings of 3 to 6 s duration of either galloping (n = 21), trotting (n = 13), or walking (n = 21) were identified and the number of steps were counted. Accelerometer data was then analyzed for each gait. Steps could be clearly identified by changes in the acceleration in the forward and vertical axes and vector sum, but less clearly in the lateral axis. The number of steps counted using the forward axis was highly correlated with the number observed from the video recordings. Galloping, trotting, and walking differed significantly in the median interpeak intervals in acceleration in the forward axis and in the vector sum of the acceleration in the 2 axes. Interpeak intervals could be used to discriminate among the 3 gaits, although walking was most clearly distinguished from galloping. Automated measures of acceleration of the leg in the forward and vertical dimensions can be used to count steps and classify gaits of calves.  相似文献

Cryptosporidium parvum is considered one of the most common enteropathogens, responsible for the high incidence of diarrhea and deleterious implications on immunity and health in neonatal calves. The pomegranate is well known for its health-promoting properties. Two experiments were designed to test the antiparasitical and antidiarrheal effects of concentrated pomegranate extract (CPE) supplement in milk in neonatal Holstein calves. Forty-one calves were randomly divided into control (n = 20) and treatment (n = 21) groups. For the first experiment, the treatment group was supplemented with 3.75% CPE in the daily milk ration, between 3 and 14 d of age, whereas the control group received only milk. Fecal samples were collected between d 5 and 13 to quantify Cryptosporidium oocysts, and the duration and intensity of diarrhea were evaluated. Reduced fecal oocyst count and diarrhea intensity and duration were revealed in the 3.75% CPE calves. No difference was noted in average daily gain between groups. In a second experiment, which was designed to test the effect of a lower CPE concentration (0.6% of daily milk allocation), no effects on fecal oocyst count and average daily gain were observed. However, compared with control, the lower CPE group was characterized by a shorter duration of diarrhea and higher weight gain among males at 14 d of age. These results suggest that the CPE supplement-to-calf milk ratio may potentially alleviate intestinal morbidity caused by Cryptosporidium.  相似文献

Johne's disease is a chronic enteritis caused by Mycobacterium avium ssp. paratuberculosis (MAP) that causes substantial financial losses for the cattle industry. Susceptibility to MAP infection is reported to be determined in part by genetic factors, so marker-assisted selection could help to obtain bovine populations that are increasingly resistant to MAP infection. Solute carrier family 11 member 1 (SLC11A1) was adjudged to be a potential candidate gene because of its role in innate immunity, its involvement in susceptibility to numerous intracellular infections, and its previous association with bovine MAP infection. The objectives of this study were to carry out an exhaustive process of discovery and compilation of polymorphisms in SLC11A1 gene, and to perform a population-based genetic association study to test its implication in susceptibility to MAP infection in cattle. In all, 57 single nucleotide polymorphisms (SNP) were detected, 25 of which are newly described in Bos taurus. Twenty-four SNP and two 3′-untranslated region polymorphisms, previously analyzed, were selected for a subsequent association study in 558 European Holstein-Friesian animals. The SNP c.1067C > G and c.1157-91A > T and a haplotype formed by these 2 SNP yielded significant association with susceptibility to MAP infection. The c.1067C > G is a nonsynonymous SNP that causes an amino acid change in codon 356 from proline to alanine (P356A) that could alter SLC11A1 protein function. This association study supports the involvement of SLC11A1 gene in susceptibility to MAP infection in cattle. Our results suggest that SNP c.1067C > G may be a potential causal variant, although functional studies are needed to assure this point.  相似文献

The objective of this study was to evaluate the effect of early-life feeding duration on growth and economics of group-fed organic dairy calves. Heifer calves born during the spring of 2011 (n = 67) and the spring of 2012 (n = 57) were used to evaluate the effect of weaning age, growth, and profitability of group-fed calves fed once per day in an organic dairy production system. Calves were assigned to replicate feeding groups of 10 in super hutches by birth order, and were born at the University of Minnesota West Central Research and Outreach Center, Morris organic dairy. Breed groups were Holsteins (n = 15) selected for high production, Holsteins (n = 23) maintained at 1964 breed-average level, crossbreds (n = 54) including combinations of Holstein, Montbéliarde, and Swedish Red, and crossbreds (n = 32) including combinations of Holstein, New Zealand Friesian, Jersey, and Swedish Red. Groups of calves were weaned at 30 (EW, early weaning), 60 (MW, mid weaning), or 90 (LW, late weaning) d of age, and groups were fed 1.5% of birth weight of 13% total solids organic whole milk once daily and weaned when the group of 10 calves consumed an average of 0.91 kg of organic calf starter per calf per day for 4 consecutive days. Body measurements were recorded at birth, weekly during the preweaning period, at weaning, and monthly thereafter. Profitability was estimated as a function of the total cost for organic milk and organic calf starter for weaning groups to weaning and to the first 90 d of age. Preweaning group performance was weaning age, EW: 47.6 d, MW: 64.5 d, LW: 93.7 d; weaning weight, EW: 61.8 kg, MW: 79.2 kg, LW: 108.1 kg; and gain per day, EW: 0.51 kg/d, MW: 0.63 kg/d, LW: 0.75 kg/d. Body weight (BW) did not differ among weaning groups at 90 d of age; however, MW calves had lower 120-d BW than did LW calves. The EW calves did not differ from either MW or LW calves for 120-d BW. Total feed costs to weaning for groups were $1,092.97 for EW calves, $1,871.24 for MW calves, and $2,956.64 for LW calves. The cost per kilogram of gain was higher for the EW ($5.54) group of calves than for the MW ($4.60) or LW ($4.14) groups during the preweaning period. Total costs and cost per kilogram of gain for the first 90 d of age were lowest for EW ($1,595.59, $3.02) calves and highest for LW ($2,956.64, $4.13) calves, respectively.  相似文献

This study was conducted to investigate hormonal imbalances preceding stillbirths and dystocia in primiparous heifers. The study was conducted between 2003 and 2004 on a German dairy farm, including 433 heifers. Starting 3 wk before calving, a weekly blood sample was collected. At calving, another blood sample was obtained, and the calving ease (grade 0 = unassisted to grade 2 = heavy pull with mechanical calf puller), sex, birth weight, as well as vitality status (stillborn, alive) of the calf were recorded. The blood serum was analyzed for estradiol-17β and progesterone concentration. At parturition, the measured estradiol-17β concentration was greater in heifers delivering bulls than in those with female calves and was increasing with greater birth weight of the calf and increasing calving difficulty score. Already 2 wk before calving, the serum estradiol-17β concentration was significantly smaller in heifers with stillborn than live calves. On the other hand, the progesterone concentration was greater 2 wk before calving in heifers with stillborn calves, but it was unaffected by the birth weight or sex of the calf or the calving difficulty score. Stillborn and live calves did not differ in birth weight or pregnancy duration. The smaller estradiol-17β concentrations of the heifers with stillborn calves could indicate an abnormality of the placenta or an abnormality of hormonal signals from the calf to the placenta in the weeks before the calving.  相似文献

Objectives of this study were to determine the physiological effects of psychological stress applied to dairy calves and to test if molasses consumption could be used to validate that a stressed condition was achieved. Twenty male calves (3 wk old) received jugular catheters and were randomly assigned to control (CTR; n = 4 pens of 1 calf per pen) or social stress treatments (STR; n = 4 pens of 4 calves per pen). The STR treatment included 5 cycles of 24-h isolation followed by regrouping with unfamiliar animals for 48 h (over 15 d). An ACTH challenge (0.1 IU/kg of body weight) was used to determine adrenal fatigue. Peak and total cortisol concentrations were greater for STR calves until the ACTH challenge. After the ACTH challenge, CTR calf cortisol increased and STR calf cortisol continued to decrease, suggesting adrenal fatigue. The number of calves that became positive for fecal shedding of Salmonella after the acute stress of being moved and the number of calves that were positive after the move decreased with each move. Fifty-six percent of STR calves changed from negative to positive for shedding after the first move compared with 18.75% of STR calves remaining negative after the third move. Difference in fecal shedding of Enterobacteriaceae from samples taken before and after moving calves on d 6 was less than that on d 2, 3, and 5. Leukocyte counts were not different, but trends for day effects were detected for neutrophil and monocyte percentages. Molasses consumption was greater for STR calves on d 2 and 11, as was total consumption. Latency to lie after eating also increased as the study progressed; STR calves required more time to lie after eating on d 12 than on d 3, and latency to lie was greater for STR than CTR on d 4, 8, 12, and 14. The STR calves also stood more than the CTR calves in the 4-h afternoon period on d 4, 5, 7, and 14. However, during the 4-h morning observations on d 14 (ACTH challenge), CTR calves stood more than STR calves. This model induced chronic stress, as characterized by adrenal fatigue, which was confirmed by molasses consumption and behavior changes. Therefore, molasses consumption could be used to confirm social stress in experimental models.  相似文献

The objectives of this study were to determine the effects of an increased jugular supply of l-Gln on the autophagy of weaned calves. At 35 d of age, 24 Holstein calves (initial body weight of 50 ± 0.5 kg; 35 ± 2 d of age) were randomly allocated to 4 treatments, with each treatment including 5 male calves and 1 female calf. Holstein calves were assigned to treatments of (1) intravenous infusion of 2 d of 0.85% NaCl (control group) (2) intravenous infusion of 8 g/d of l-Gln mixed with 2 d of 0.85% NaCl solution, (3) intravenous infusion of 16 g/d of l-Gln mixed with 2 d of 0.85% NaCl solution, and (4) intravenous infusion of 32 g/d of l-Gln mixed with 2 d of 0.85% NaCl. The infusion was administered 2 h/d for 7 consecutive days starting on d 1 after weaning. Feed and water were freely available to all calves. All calves were killed on d 7 postweaning to measure the autophagy of liver cells. The level of autophagy in liver cells was improved when the Gln infusion dose increased.  相似文献

This experiment tested the effects of pair versus single housing on the performance and behavior of dairy calves before and after weaning. Twenty-seven Holstein calves were separated from the dam within 6 h of birth, housed in individual pens for 4 d, and then assigned to either continued individual housing (n = 9 calves) or pair housing (n = 9 pairs). Calves had ad libitum access to starter, hay, and water via buckets. Pasteurized whole milk was fed via teat twice a day for 2 h at ad libitum volumes until d 36. During the milk-feeding period, paired calves showed higher intakes of starter than did the individually housed calves (averaging 93 vs. 59 ± 11 g/d per calf). Calves were weaned from milk from d 37 to 41 by progressive dilution of milk with water, and the teat was removed on d 49. Calves in both treatments vocalized in response to teat removal but this response was less in paired calves than in individually housed calves (84 vs. 194 ± 12 calls/2-h period per calf on d 49). On d 56, calves were moved to group pens, mixed with other calves, and observed for 15 d. Starter, water, and hay were available ad libitum via automatic feeders. Compared with calves previously housed in single pens, paired calves had a shorter latency to start feeding (9.1 ± 2.6 vs. 49.5 ± 4.1 h/calf), visited the starter feeder more frequently (41.6 ± 3.0 vs. 26.4 ± 3.3 visits/d per calf), spent more time at the feeder (87.8 ± 2.5 vs. 65.3 ± 2.9 min/d per calf), and consumed more starter (3.4 vs. 2.3 ± 0.2 kg/d per calf). Weight gains at mixing were higher for paired than for individually housed calves on d 2 and 3 after mixing (0.5 vs. −2.4 ± 0.3 kg/d per calf; and 0.8 vs. −0.9 ± 0.3 kg/d per calf, respectively). The results indicate that pair housing during the milk-feeding stage reduces calf responses to weaning and improves performance after weaning when calves are housed in groups.  相似文献

Pathogenic Yersinia enterocolitica is involved in yersiniosis through expression of chromosome-borne or plasmid-borne virulence factors. Yersinia enterocolitica is a cold-tolerant pathogen frequently isolated from refrigerated or frozen foods. However, little attention has been focused on the prevalence of pathogenic Y. enterocolitica in refrigerated or frozen dairy samples in China. In this study, we developed a new duplex PCR targeting the plasmid-borne virF gene and chromosome-borne ail gene for detection of pathogenic Y. enterocolitica isolates. We established a detection limit for the duplex PCR of 6.5 × 102 cfu/mL in artificially contaminated dairy samples. In addition, the duplex PCR could detect directly 4.5 to 5.7 cfu of Y. enterocolitica in 5 mL of brain heart infusion broth after 6 h of enrichment at 28°C. A newly developed dot hybridization assay further confirmed specificity of the duplex PCR for detection of virulent Y. enterocolitica. Furthermore, 13 Y. enterocolitica and 5 pathogenic strains, from 88 commercial frozen or refrigerated dairy products, were detected successfully by the China National Standard method (GB/T4789.8-2008) and the duplex PCR, respectively. Finally, biotypes and serotypes of pathogenic Y. enterocolitica strains were further characterized. The duplex PCR developed here is reliable for large-scale screening, routine monitoring, and risk assessment of pathogenic Y. enterocolitica in refrigerated or frozen dairy products.  相似文献
