Methanolic extracts from underripe, ripe, and ripe then dried pawpaw seeds (PPSE) were tested for antioxidative effectiveness. Underripe seeds contained more total phenolic compounds than ripe seeds, but ripe seeds showed the highest reducing potential. PPSE samples effectively scavenged DPPH (>86%). PPSE (0–26.8 μM gallic acid) effectively inhibited lipid oxidation biomarkers in 22:6 phosphatidylcholine (PC) liposome suspensions over 7 day’s incubation at room temperature. PPSE inhibited TBARS in 22:6 PC liposome suspensions induced by peroxyl radical generators, free radical generators that produce reactive oxygen, reactive nitrogen, and reactive sulfur species. After 120 min of storage TBARS inhibition by PPSE was in the order of H2O2 ? SIN-1 = DSSO = AAPH ? Sulfite > ONOO = MMb > iron/ascorbate. PPSE inhibited lipid oxidation biomarkers in skeletal muscle homogenates induced by iron/ascorbate. The development of value added products from pawpaw seeds may lead to successful commercialisation of this underutilised fruit.  相似文献

ABSTRACT: Amounts of total phenolics, anthocyanins, and ascorbic acid in 4 American cranberry varieties harvested at 4 stages of maturity were measured. The larger amount of phenolic compounds was found in berries of “Black Veil” cultivar (504 mg/100 g) at II stage of maturity. Significantly larger amounts of anthocyanins were determined in the overripe berries of the cultivars “Ben Lear” and “Black Veil.” The amount of ascorbic acid in berries increased during ripening from I to III stage, and slightly decreased in the overripe berries. The biggest quantities of ascorbic acid were found in the ripe berries of “Ben Lear” cultivar (15.8 mg/100 g). The distribution of anthocyanins pigments was determined by HPLC‐UV/MS in mature berries. The composition of individual anthocyanins in berries was quite similar in all the studied cranberry cultivars. While skins of cranberries are rich in anthocyanins and other phenolic compounds, the extracts of the by‐products of cranberries juice—berry cakes, were analyzed and obtained results were compared with the properties of extracts made from whole berries. The anthocyanins and total phenolics content, radical scavenging activity, antimicrobial activity of the whole berries, and their press cakes extracts were measured. All investigated extracts from berries and their press cakes showed good radical scavenging activity and revealed antimicrobial properties. It was found that Bacillus cereus (ATCC 10876) and Micrococcus luteus (ATCC 9341) were the most sensitive among 10 tested Gram‐negative and Gram‐positive bacteria.  相似文献

Total phenolics content, free radical scavenging activity, reducing power, and antioxidant activity of the pyroligenous acid from a mangrove plant, Rhizophora apiculata were evaluated. Dichloromethane extraction of the raw pyroligneous acid successfully yield 2 extracts, i.e. concentrated pyroligneous acid (CPA) and concentrated pyroligneous acid extract (CPAE). Phenolic contents in CPAE and CPA, expressed as (±)-catechin equivalents/g of the sample were 5465 ± 367 mg and 2502 ± 152 mg, and expressed as gallic acid equivalents/g of the sample were 2919 ± 209 mg and 1348 ± 90 mg, respectively. CPAE exhibited superior free radical scavenging activity with EC50 value = 0.1235 mg/ml, or 80.96% of free radical scavenging capability. The ferric reducing power of CPAE was approximately 3.7, 5.1, 6.1, and 21.3 times higher than that of ascorbic acid, BHA, BHT and alpha-tocopherol. In phosphomolybdenum assay, CPAE showed the greatest antioxidant efficacy (A695 = 1.278) compared to those of CPA and different standards. In addition, the free radical scavenging activity, ferric reducing power and total antioxidative activity of CPAE and CPA showed positive correlation with their total phenolic content with R2 values ranging from 0.9624 to 0.9979.  相似文献

The aim of this investigation was to compare the bioactive and nutrient compounds, fatty acids, and antioxidant and antiproliferative activities of Mon Thong durian at different stages of ripening. It was found that the total polyphenols, flavonoids, flavanols, ascorbic acid, tannins and the antioxidant activity determined by four assays (CUPRAC, DPPH, ABTS and FRAP) differed in immature, mature, ripe and overripe samples. The content of polyphenols and antioxidant activity were the highest in overripe durian, flavonoids were the highest in ripe durian, and flavanols and antiproliferative activity were the highest in mature durian (< 0.05). FTIR spectra of polyphenols, HPLC profiles of fatty acids, the antioxidant and antiproliferative activities can be used as indicators to characterise different stages of durian ripening.  相似文献

Three Lamiaceae often used in Portuguese folk medicine: Ground ivy (Glechoma hederaceae L.), oregano (Origanum vulgare subsp. virens (Hoffmanns. & Link) Ietswaart) and mastic thyme (Thymus mastichina L.), revealed to be good sources of powerful antioxidants such as vitamins (ascorbic acid and tocopherols) and phenolics. The HPLC methodology developed for tocopherols analysis proved to be, after validation assays, sensitive and accurate. Ground ivy showed the highest levels of vitamins: ascorbic acid (168 ± 2 μg/g dry weight) and tocopherols (3692 ± 57 μg/g dry weight). The phenolics extraction was optimized using mastic tyme, and different conditions (water and ethanol:water 50% v/v, 30 min at 25 °C and boiling temperature; ethanol and methanol, 24 h at 25 °C). The best methodology (25 °C, 50 ml of methanol, 24 h) was further applied to the other Lamiaceae. Oregano proved to have the highest radical scavenging and lipid peroxidation inhibition capacity. Particularly, the very low EC50 value (0.01 mg/ml) obtained for TBARS inhibition in brain homogenates is very promising, considering that brain is highly sensitive to oxidative damage. Significantly negative linear correlations were observed between phenolics, including flavonoids, and antioxidant activity EC50 values of the three Lamiaceae.  相似文献

The effects of salicylic acid (SA) on the quality and antioxidant activity of asparagus stored at 18 ± 2 °C were investigated by analyzing the color, chlorophyll, shear force, and the activity of antioxidant compounds such as ascorbic acid, phenolics, flavonoids, 1,1-Diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity ferric reducing antioxidant power (FRAP), and polyamines (PAs). The results showed that SA improved the color and maintained the chlorophyll, phenolic, flavonoid, and ascorbic acid content of asparagus. High concentrations of SA caused a deterioration in asparagus would harm to color and had no effect on shear force within 6 d. SA induced the maximum concentration of phenolics in postharvest asparagus, promoted the increase in total flavonoids before 6 to 9 d, affected the antioxidant activity positively as indicated by the resultant increase in FRAP concentration; however, SA was only active with regard to DPPH scavenging activity within 6 d of treatment. Spermidine (Spd) is the most common form of PA in asparagus, and free putrescine (Put) contents increased over the first 3 d following harvest and then decreased. Spd and Spm concentrations evolved in a similar way and decreased during storage. Higher Spd and Spm contents in the SA pre-treatment Put was inhabited and its peaks appeared later.  相似文献

Pineapple has several beneficial properties including antioxidant activity. The fruit of pineapple was extracted with ethyl acetate, methanol and water. The phenolic content of the extracts was determined by Folin-Ciocalteu method and antioxidant activity was assayed through some in vitro models such as antioxidant capacity by phosphomolybdenum, ??-carotene-linoleate, and radical scavenging activity using ??,??-diphenyl-??-picrylhydrazyl (DPPH) method. The phenolic contents of the extracts as caffeic acid equivalents were found to be highest in methanol (51.1%) followed by ethyl acetate (13.8%) and water extract (2.6%). Antioxidant capacity of the extracts as equivalent to ascorbic acid (??mol/g of the extract) was in the order of methanol extract > ethyl acetate extract > water extract. In comparison with butylated hydroxyanisole (BHA), at 100 ppm of concentration, the antioxidant and free radical scavenging activities of the extracts assayed through ??-carotene-linoleate and DPPH method were also found to be highest with methanol extract followed by ethyl acetate and water extracts. The results indicated that the extent of antioxidant activity of the extract is in accordance with the amount of phenolics present in that extract and the pineapple fruit being rich in phenolics may provide a good source of antioxidant.  相似文献

Red pepper has been recognised as an excellent source of antioxidants, being rich in ascorbic acid and other phytochemicals. Drying conditions, particularly temperature, leads to pepper modifications that can cause quality degradation. In this work, the effects of process temperatures between 50 and 90 °C on physico-chemical properties, rehydration, colour, texture, vitamin C, antioxidant capacity and total phenolics during the drying of red pepper were studied. The rehydration ratio decreased with temperature and the maximum water holding capacity was achieved at 50 °C. Both vitamin C content and the total phenolic content decreased as air-drying temperature decreased. The radical scavenging activity showed higher antioxidant activity at high temperatures (i.e. 80 and 90 °C) rather than at low temperatures (i.e. 50, 60 and 70 °C). Chromatic parameters (L*, a*, b*, C* and H°), non-enzymatic browning compounds and extractable colour were affected by drying temperature, which contributed to the discolouring of pepper during this process.  相似文献

The effect of steam treatment on free phenolic acids in Chaga mushrooms (Inonotus obliquus) was investigated. Untreated and steam-treated (120 °C, 3 h) samples of I. obliquus were extracted with organic solvents and free phenolic acid-containing fractions were isolated. Free phenolic acids were determined by LC/PDA (liquid chromatography/photodiode array), ESI LC/MS (electrospray ionisation liquid chromatography/mass spectrometry), and GC/MS (gas chromatography/mass spectrometry). After the steam treatment, the soluble phenolic content determined by modified Folin–Ciocalteu method was increased and antioxidant activity was enhanced, as confirmed by a DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity assay. The amounts of vanillic acid, protocatechuic acid, syringic acid, and 2,5-dihydroxyterephthalic acid were increased significantly as the result of the steam treatment, suggesting that the liberation of low molecular weight free phenolics was enhanced by the steaming process. Consequently, the radical scavenging activity was also significantly enhanced by free phenolics produced using this method.  相似文献

Major anthocyanins, ascorbic acid content, total phenolic content, and the radical scavenging activity against ABTS and DPPH radicals in petals of orange Nasturtium flowers (Tropaeolum majus), were investigated. Anthocyanin (ACN) content in the petals was 72 mg/100 g FW and pelargonidin 3-sophoroside represented 91% of the total ACN content. The ascorbic acid content was 71.5 mg/100 g and the total phenolic content as determined by the Folin–Ciocalteau method was 406 mg GAE/100 g FW. The radical scavenging activities against ABTS and DPPH radicals were 458 and 91.87 μm trolox eq/g FW, respectively. The excellent free radical scavenging activities along with high phenolic and ascorbic acid content of Nasturtium flowers suggest that they could be source of natural pigments and antioxidants for applications in functional foods.  相似文献

The antioxidant activities of different polarity solvents extracts from barley were evaluated by various antioxidant assays, including reducing power, free radical scavenging and lipid oxidation inhibition. Those various antioxidant activities were compared to standard antioxidants, butylated hydroxyluene (BHT) and ascorbic acid (Vit. C). The properties of the extracting solvents significantly affected the total phenolics, proanthocyanidins and antioxidant activities of barley extract. The highest contents of total phenolics and proanthocyanidins were obtained from extraction with 70% acetone. For three different solvent extracts, the antioxidant activities were in this order: 70% acetone extract > 70% ethanol extract ? 70% methanol extract. Reducing powers of three extracts and their scavenging effects on 2, 2-diphenyl-1-picrylhydrazyl radicals were effective at amount of 200 μg. The 70% acetone extract of barley exhibited high antioxidant activity in linoleic acid system, which was not significantly (P < 0.05) different from BHT during the incubation time.  相似文献

Four cultivars of Shanxi-grown pomegranate seeds were investigated for their phytochemical composition and antioxidant properties. Pomegranate seed oils were extracted and analysed for their fatty acid profiles, and tocopherol and carotenoid contents. The defatted flours were extracted with 50% acetone or 80% methanol, and evaluated for levels of phenolics, flavonoids and proanthocyanidins, and scavenging capacities against DPPH and ABTS+ and their ferric ion reducing abilities. Pomegranate seed oil was rich in punicic acid (73.5-78.8 g/100 g oil) and total tocopherols (5.3-12.0 μmol/g). The 50% acetone and 80% methanol were appropriate solvents for extraction of total phenolic and total flavonoid, respectively. The four cultivars were significantly different in levels of phenolics, flavonoids, proanthocyanidins and their antioxidant properties (P < 0.05), generally flowing an order: Suanshiliu > Tianhongdan > Sanbaitian ≈ Jingpitian. This study will assist in the cultivar selection of the above pomegranate seeds for commercial production to meet the market demand.  相似文献

Free-radical scavenging, reducing activity and some phytochemical content (total phenolic, anthocyanin and ferulic acid) of eighteen Mexican maize phenotypes were determined. Total phenolic contents ranged from 215.8 to 3400.1 mg gallic acid/100 g of whole grain flour and total anthocyanins ranged from 1.54 to 850.9 mg cyanidin-glucoside equivalents/100 g of whole grain flour. Most of the phenolics in grain were in the bound form (ca. 85%), while anthocyanins were the major free phenolic compounds. Among the different samples, bound phenolic extracts of corn appeared to have greater anti-radical and reducing activities than free phenolic extracts from the same grain samples when tested at a normalized phenolic concentration. The phenotypes Veracruz 42 and AREQ516540TL exhibited the greatest activities and these purple-colored strains were most enriched in anthocyanins. Extracts from a red-colored phenotype Pinto were also among the most effective at exhibiting anti-radical activities. Differences in free-radical scavenging and reducing activities appeared to be dependent on the unique profile of anthocyanins and other phenolics in each phenotype.  相似文献

Buddleia officinalis Maxim, commonly used as rice dye for festivals, was extracted with ethanol using microwave-assisted extraction and Soxhlet extraction. The antioxidant activities of microwave-assisted extract of B. officialis (MEB) and Soxhlet extract of B. officianils (SEB) at the optimum extraction conditions were evaluated and compared with synthetic antioxidant butylated hydroxytoluene (BHT) employing DPPH free radical assay, ABTS assay, total antioxidant activity and reducing power. MEB and SEB had stronger antioxidant activities than BHT in all assays except reducing power, and the effects decreased as follows: MEB > SEB > BHT. The total phenolic contents of MEB and SEB reached 113.56 mg/g and 100.94 mg/g dry weight of extract, respectively, expressed as pyrocatechol equivalents, while the total flavonoids contents were 75.33 mg/g and 62.56 mg/g dry weight of extract, respectively, expressed as catechin equivalents (P < 0.05). Higher phenolic and flavonoids compounds may be major contributors to their higher antioxidant activities. Following activity-oriented separation, luteolin was isolated as an active principle, which exhibited excellent free radical scavenging activities with DPPH IC50 3.09 μg/ml and ABTS IC50 2.20 μg/ml.  相似文献

Phenolic constituents of six barley varieties, namely Falcon, AC Metcalfe, Tyto, Tercel, Phoenix and Peregrine were separated into free, soluble conjugate and insoluble-bound fractions. Soluble conjugates and insoluble-bound phenolics were extracted into diethyl ether after consecutive alkaline hydrolysis for 4 h. Total phenolic content of each of the three fractions was determined by using Folin–Ciocalteau method. Total antioxidant capacity of the phenolic fractions was determined by trolox equivalent antioxidant capacity (TEAC) assay. The extracts were evaluated for their efficacy in scavenging of 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical. Oxygen radical scavenging capacity (ORACFL) of the extracts was determined using a fluorometric assay. Effectiveness of phenolic extracts in inhibiting oxidation of human LDL cholesterol and radical-induced supercoiled DNA breakage was also evaluated. The contribution of insoluble-bound phenolics toward total phenolic content was significantly (p < 0.05) higher than the soluble phenolics for all barley extracts tested. The ratio of soluble to insoluble phenolics ranged from 1:27 to 1:35. TEAC and ORAC values and DPPH radical scavenging capacity of the insoluble phenolic fraction were significantly (p < 0.05) higher than those of their insoluble counterparts. A similar trend was observed against inhibition of LDL cholesterol oxidation and radical-induced DNA breakage.  相似文献

Millets are generally dehulled and subjected to a hydrothermal treatment before consumption, thus the hulls can be used as a potential source of antioxidants. Several millet grains, namely kodo, finger (Ravi), finger (local), proso, foxtail, little and pearl millet were studied. Antioxidant activities of phenolic extracts obtained from whole grains, as well as their corresponding dehulled and cooked grains and hulls were studied for their total phenolic content (TPC), radical scavenging capacity, and antioxidant activity in a β-carotene/linoleate emulsion. The phenolics present in whole grains were identified and quantified using HPLC and HPLC/MS and results were expressed as total for each of the phenolic groups. The TPC ranged from 2 to 112 μmol ferulic acid equivalents/g defatted meal. All varieties exhibited effective inhibition of 2,2-diphenyl-1-picrylhydrazyl (DPPH), hydroxyl, peroxyl and superoxide radicals. Dehulling and cooking affected the TPC and radical scavenging and antioxidant activities of the grains, depending on the variety. In general, the antioxidant activity of phenolic extracts was in the order of hull > whole grain > dehulled grain > cooked dehulled grain With the exception of the two finger millet varieties, hulls of other millet grains had high TPC, thus demonstrating their superior antioxidant activity. Hydroxybenzoic acids, hydroxycinnamic acids and flavonoids in whole grains were identified as contributors to the observed effects. Therefore, dehulling of grain and hydrothermal treatments affect the phenolic content and antioxidant potential of millet grains.  相似文献

The antioxidant properties and total phenolic contents of methanol, acetone and water extracts of mulberry (Morus indica L.) leaves were examined. Various experimental models including iron (III) reducing capacity, total antioxidant capacity, DPPH radical scavenging activity and in vitro inhibition of ferrous sulphate-induced oxidation of lipid system were used for characterization of antioxidant activity of extracts. The three extracts showed varying degrees of efficacy in each assay in a dose-dependent manner. Methanolic extract with the highest amount of total phenolics, was the most potent antioxidant in all the assays used. In addition, the effect of temperature (50 °C and 100 °C), pH (3, 5, 7, 9 and 11) and storage (5 °C) on the antioxidant activity of methanolic extract was investigated. The antioxidant activity of the extract remained unchanged at 50 °C and was maximum at neutral pH. The extract stored at 5 °C in the dark was stable for 30 days after which the antioxidant activity decreased (p ? 0.05) gradually. On the basis of the results obtained, mulberry leaves were found to serve as a potential source of natural antioxidants due to their marked antioxidant activity.  相似文献

The extraction of antioxidant compounds from soybeans fermented with Aspergillusoryzae was optimised using a factorial design. A kinetic study of the total phenolic production and DPPH radical scavenging activity was first performed at the points selected in the factorial design. In both cases, the experimental profiles were fitted to a modified first-order kinetic model. To investigate the combined effects of temperature and solvent concentration on the extraction, the parameters obtained from the fitted kinetic models were used as response variables in a rotatable second-order design with quintuple replications in the centre of the experimental domain. The results obtained indicate that temperature had the most significant effect. The response surfaces show a maximum in the experimental domain studied. The optimum conditions for the extraction of total phenolic content were 65.3 °C and 73.1% ethanol, in which 56.2 mg of GAE/g were predicted. A scavenging activity of 81.6% DPPH radical was predicted at the optimum conditions of 61.6 °C and 60% ethanol.  相似文献

The nutritional and antioxidant properties of peels, pulp and seeds of kembayau (Dacryodes rostrata) fruits were evaluated. Kembayau seeds and pulp were rich in fat, while peels had the highest ash contents. Potassium was the most prevalent mineral in peels (380.72-1112.00 mg/100 g). In kembayau fruits, total flavonoid content (1012.74-28,022.28 mg rutin equivalent/100 g) was higher than total phenolic and total monomeric anthocyanin contents. Kembayau seeds exhibited high flavonoid and phenolic contents compared to the contents in peels and pulp. Antioxidant capacities were also higher in seeds as typified by trolox equivalent antioxidant capacity assay (51.39-74.59 mmol TE/100 g), ferric reducing antioxidant power assay (530.05-556.98 mmol Fe2+/100 g) and by 1,1-diphenyl-2-picryl hydrazyl radical scavenging activity (92.18-92.19%) when compared to peels and pulp. Pulp and peels of kembayau fruit may be an important source of energy and minerals for human consumption, while seeds have a good potential as antioxidants.  相似文献
