Background
β-alanine (βA) supplementation has been shown to increase intramuscular carnosine content and subsequent high-intensity performance in events lasting <?4?minutes (min), which may be dependent on total, as opposed to daily, dose. The ergogenic effect of βA has also been demonstrated for 2000-m rowing performance prompting interest in whether βA may be beneficial for sustained aerobic exercise. This study therefore investigated the effect of two βA dosing strategies on 30-min rowing and subsequent sprint performance.Methods
Following University Ethics approval, twenty-seven healthy, male rowers (age: 24?±?2?years; body-height: 1.81?±?0.02?m; body-mass: 82.3?±?2.5?kg; body-fat: 14.2?±?1.0%) were randomised in a double-blind manner to 4?weeks of: i) βA (2.4?g·d??1, βA1); ii) matched total βA (4.8?g on alternate days, βA2); or iii) cornflour placebo (2.4?g·d??1, PL). Participants completed a laboratory 30-min rowing time-trial, followed by 3x30-seconds (s) maximal sprint efforts at days 0, 14 and 28 (T1-T3). Total distance (m), average power (W), relative average power (W·kg??1), cardio-respiratory measures and perceived exertion were assessed for each 10-min split. Blood lactate ([La-]b mmol·L??1) was monitored pre-post time-trial and following maximal sprint efforts. A 3-way repeated measures ANOVA was employed for main analyses, with Bonferonni post-hoc assessment (P?≤?0.05).Results
Total 30-min time-trial distance significantly increased from T1-T3 within βA1 only (7397?±?195?m to 7580?±?171?m, P?=?0.002, ?p2?=?0.196), including absolute average power (194.8?±?18.3?W to 204.2?±?15.5?W, P?=?0.04, ?p2?=?0.115) and relative average power output (2.28?±?0.15?W·kg??1 to 2.41?±?0.12?W·kg??1, P?=?0.031, ?p2?=?0.122). These findings were potentially explained by within-group significance for the same variables for the first 10?min split (P?≤?0.01), and for distance covered (P?=?0.01) in the second 10-min split. However, no condition x time interactions were observed. No significant effects were found for sprint variables (P?>?0.05) with comparable values at T3 for mean distance (βA1: 163.9?±?3.8?m; βA2: 161.2?±?3.5?m; PL: 162.7?±?3.6?m), average power (βA1: 352.7?±?14.5?W; βA2: 342.2?±?13.5?W; PL: 348.2?±?13.9?W) and lactate (βA1: 10.0?±?0.9?mmol·L??1; βA2: 9.2?±?1.1?mmol·L??1; PL: 8.7?±?0.9?mmol·L??1).Conclusions
Whilst daily βA may confer individual benefits, these results demonstrate limited impact of βA (irrespective of dosing strategy) on 30-min rowing or subsequent sprint performance. Further investigation of βA dosage >?2.4?g·d??1 and/or chronic intervention periods (>?4–8?weeks) may be warranted based on within-group observations.3.

β-tricalcium phosphate (β-TCP) is an ideal biomaterial for the bone repair because of its biocompatibility and biodegradability. In this study, 0 mol%, 5 mol%, 15 mol% and 30%mol bivalent manganese ion (Mn2+) doped β-TCP (Mn-TCP) powders were synthesized by a sol-gel method. The amount of the dopants significantly influences the crystallinity and the parameters related with structure of β-TCP, such as the lattice parameters and crystallite dimensions. The particle size and the particle distribution of doped β-TCP powers were evaluated as well. Meanwhile, the as-synthesized powders were consolidated by sintering at 1000 °C in muffle furnace for 5 h to get Mn-TCP porous material and the degradation experiment was carried out in Simulated Body Fluid (SBF) solution for 28 days. Then, Mn-TCP porous material were characterized by Fourier-transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), and scanning electron microscope (SEM). Significantly, there were bone-like apatite materials deposited on the surface of bone-like porous materials. With the increasing doping amount of Mn2+, the newly formed apatite-like materials decreased, while the crystallinity increased significantly. The interaction between α‐tocopherol (500 mg/kg) and β‐carotene (10 mg/kg) during chlorophyll‐photosensitized oxidation of a sunflower oil emulsion was studied in the presence or absence of phosphatidylcholine (PC, 250 mg/kg) by determining peroxide (POV) and conjugated dienoic acid (CDA) values. Chlorophyll, α‐tocopherol, β‐carotene, and PC contents in the emulsion were also monitored. α‐Tocopherol and β‐carotene individually and interactively decreased the POV and CDA values of oil in the emulsion by singlet oxygen quenching. PC decreased the POV and CDA values of oil, however, the values of the emulsion with added α‐tocopherol, β‐carotene, and PC were not significantly different from those of the emulsion with added α‐tocopherol and β‐carotene without PC. Contents of α‐tocopherol did not change during 24‐h oxidation, whereas co‐present PC significantly caused α‐tocopherol and chlorophyll degradation. β‐Carotene and PC contents significantly decreased to 45.5 and 51.3 %, respectively, after 24 h, and α‐tocopherol protected β‐carotene from degradation. The results suggest that PC had no net effects on the interactive antioxidant activity of α‐tocopherol and β‐carotene during chlorophyll‐photosensitized oxidation of the emulsion through free radical generation, chlorophyll degradation, and lessening the potency of α‐tocopherol as a singlet oxygen quencher. 相似文献

To evaluate the antioxidant and immune effects of low molecular yeast β-glucan on mice, three sulfated glucans from Saccharomyces cerevisiae (sGSCs) with different molecular weight (MW) and degrees of sulfation (DS) were prepared. The structures of the sGSCs were analyzed through high performance liquid chromatography-gel permeation chromatography (HPLC-GPC) and Fourier transform infrared spectroscopy (FTIR). sGSC1, sGSC2, and sGSC3 had MW of 12.9, 16.5 and 19.2 kDa, respectively, and DS of 0.16, 0.24 and 0.27, respectively. In vitro and in vivo experiments were conducted to evaluate the antioxidant and immunological activities of the sGSCs. In vitro experiment, the reactive oxygen species (ROS) scavenging activities were determined. In vivo experiment, 50 male BALB/c mice were divided into five groups. The sGSC1, sGSC2 and sGSC3 treatment groups received the corresponding sGSCs at 50 mg/kg/day each. The GSC (glucans from Saccharomyces cerevisiae) treatment group received 50 mg/kg/day GSC. The normal control group received equal volume of physiological saline solution. All treatments were administered intragastrically for 14 day. Results showed that sGSC1, sGSC2 and sGSC3 can scavenge 1,1-diphenyl-2-picryl-hydrazyl (DPPH), superoxide, and hydroxyl radicals in vitro. The strength of the radical scavenging effects of the sGSCs was in the order of sGSC1 > sGSC2 > sGSC3. Oral administration of sGSC1 significantly improved serum catalase (CAT) and glutathione peroxidase (GSH-Px) activities and decreased malondialdehyde (MDA) level in mice. sGSC1 significantly improved the spleen and thymus indexes and the lymphocyte proliferation, effectively enhanced the percentage of CD4+ T cells, decreased the percentage of CD8+ T cells, and elevated the CD4+/CD8+ ratio. sGSC1 significantly promoted the secretion of IL-2 and IFN-γ. These results indicate that sGSC1 with low MW and DS has better antioxidant and immunological activities than the other sGSCs, and sGSC1 could be used as a new antioxidant and immune-enhancing agent. 相似文献

Background
Ageing is associated with a significant reduction in skeletal muscle carnosine which has been linked with a reduction in the buffering capacity of muscle and in theory, may increase the rate of fatigue during exercise. Supplementing beta-alanine has been shown to significantly increase skeletal muscle carnosine. The purpose of this study, therefore, was to examine the effects of ninety days of beta-alanine supplementation on the physical working capacity at the fatigue threshold (PWCFT) in elderly men and women.Methods
Using a double-blind placebo controlled design, twenty-six men (n = 9) and women (n = 17) (age ± SD = 72.8 ± 11.1 yrs) were randomly assigned to either beta-alanine (BA: 800 mg × 3 per day; n = 12; CarnoSyn?) or Placebo (PL; n = 14) group. Before (pre) and after (post) the supplementation period, participants performed a discontinuous cycle ergometry test to determine the PWCFT.Results
Significant increases in PWCFT (28.6%) from pre- to post-supplementation were found for the BA treatment group (p < 0.05), but no change was observed with PL treatment. These findings suggest that ninety days of BA supplementation may increase physical working capacity by delaying the onset of neuromuscular fatigue in elderly men and women.Conclusion
We suggest that BA supplementation, by improving intracellular pH control, improves muscle endurance in the elderly. This, we believe, could have importance in the prevention of falls, and the maintenance of health and independent living in elderly men and women. 相似文献9.

5–18. These pups were fed rat milk substitutes containing fats that provided 10% linoleic acid and 1% α-linolenic acid (%
fatty acids) and, using a 2×3 factorial design, one of two levels of DHA (0.5 and 2.5%), and one of three levels of GLA (0.5,
1.0, and 3.0%). A seventh artificially reared groups served as a reference group and was fed 0.5% DHA and 0.5% arachidonic
acid (AA, 20∶4n−6); these levels are within the range of those found in rat milk. The eighth group, the suckled control group,
was reared by nursing dams fed a standard American Institute of Nutrition 93M chow. The fatty acid composition of the phosphatidylethanolamine,
phosphatidyl-choline, and phosphatidylserine/phosphatidylinositol membrane fractions of the forebrain on day 18 reflected
the dietary composition in that high levels of dietary DHA resulted in increases in DHA but decreases in 22∶4n−6 and 22∶5n−6
in brain. High levels of GLA increased 22∶4n−6 but, in contrast to previous findings with high levels of AA, did not decrease
levels of DHA. These results suggest that dietary GLA, during development, differs from high dietary levels of AA in that

Purpose
This study determined the effects of 28 days of heavy resistance exercise combined with the nutritional supplement, NO-Shotgun®, on body composition, muscle strength and mass, markers of satellite cell activation, and clinical safety markers.Methods
Eighteen non-resistance-trained males participated in a resistance training program (3 × 10-RM) 4 times/wk for 28 days while also ingesting 27 g/day of placebo (PL) or NO-Shotgun® (NO) 30 min prior to exercise. Data were analyzed with separate 2 × 2 ANOVA and t-tests (p < 0.05).Results
Total body mass was increased in both groups (p = 0.001), but without any significant increases in total body water (p = 0.77). No significant changes occurred with fat mass (p = 0.62); however fat-free mass did increase with training (p = 0.001), and NO was significantly greater than PL (p = 0.001). Bench press strength for NO was significantly greater than PL (p = 0.003). Myofibrillar protein increased with training (p = 0.001), with NO being significantly greater than PL (p = 0.019). Serum IGF-1 (p = 0.046) and HGF (p = 0.06) were significantly increased with training and for NO HGF was greater than PL (p = 0.002). Muscle phosphorylated c-met was increased with training for both groups (p = 0.019). Total DNA was increased in both groups (p = 0.006), while NO was significantly greater than PL (p = 0.038). For DNA/protein, PL was decreased and NO was not changed (p = 0.014). All of the myogenic regulatory factors were increased with training; however, NO was shown to be significantly greater than PL for Myo-D (p = 0.008) and MRF-4 (p = 0.022). No significant differences were located for any of the whole blood and serum clinical chemistry markers (p > 0.05).Conclusion
When combined with heavy resistance training for 28 days, NO-Shotgun® is not associated with any negative side effects, nor does it abnormally impact any of the clinical chemistry markers. Rather, NO-Shotgun® effectively increases muscle strength and mass, myofibrillar protein content, and increases the content of markers indicative of satellite cell activation. 相似文献11.

The article deals with the effect of the conditions of modification of a natural aluminosilicate catalyst with a 10% HCl solution on the chemical and phase compositions, porous and crystal structures, and acidity of the material and on its catalytic properties in α-pinene isomerization. Treatment of the aluminosilicate with 10% HCl (25–250 mL per gram of solid) causes cation exchange, increases the concentration of protonic sites on the aluminosilicate surface, and removes impurity calcite and dolomite. The specific surface area of the aluminosilicate increases from 52 to 68–82 m2/g. Treatment of the aluminosilicate with 175.0 or 250.0 mL/g of HCl removes a considerable amount framework Al3+, Fe2+/3+, and Mg2+ cations, leading to a partial disruption of its structure and to a decrease in its acidity, specific surface area, and, as a consequence, catalytic activity relative to the same parameters of the samples treated with 50 or 100 mL/g of HCl. The highest catalytic activity is displayed by the aluminosilicate treated with 50 mL/g of HCl. The camphene and dipentene selectivity of the reaction (at 85% α-pinene conversion) with the original catalyst is 55 and 30%, respectively; modifying the aluminosilicate with HCl raises the camphene selectivity and reduces the dipentene selectivity by 5–6%. The lipids of the gram-negative bacterium Marinobacter hydrocarbonoclasticus grown in a synthetic seawater medium supplemented with various hydrocarbons as the sole carbon source were isolated, purified,
and their structures determined. The hydrocarbons were normal, iso, anteiso, and mid-chain branched alkanes, phenylalkanes, cyclohexylalkanes, and a terminal olefin. According to the sequential procedure
used for lipid extraction, three pools were isolated: unbound lipids extracted with organic solvents (corresponding to metabolic
lipids and to the main part of membrane lipids), OH− labile lipids [mainly ester-bound in the lipopolysaccharides (LPS)], and H+ labile lipids (mainly amide-bound in the LPS). Each pool contained FA, fatty alcohols, and β-hydroxy acids. The proportions
of these lipids in the unbound lipid pools were 84–98%, 1.1–11.6%, and 0.1–3.6% (w/w), respectively. The chemical structures
of the lipids were strongly correlated with those of the hydrocarbons fed; analytical data suggested a metabolism essentially
through oxidation into primary alcohol, then into FA and degradation via the β-oxidation pathway. Subterminal oxidation of the hydrocarbon chains, α-oxidation of FA or double-bond oxidation in the
case of the terminal olefin, were minor, although sometimes substantial, routes of hydrocarbon degradation. Cyclohexyldodecanedid
not support growth, likely because of the toxicity of cyclohexylacetic acid formed in the oxidation of the alkyl side chain.
In the OH− and H+ labile lipid pools, β-hydroxy acids, the lipophilic moiety of LPS, generally dominated (28–72% and 64–98%, w/w, respectively).
The most remarkable feature of these cultures on hydrocarbons was the incorporation in LPS of β-hydroxy acids with Codd, ω-unsaturated, iso, or anteiso alkyl chains in addition to the specific β-hydroxy acid of M. hydrocarbonoclasticus, 3-OH-n-12∶0. These β-hydroxy acids were tolerated insofar as their geometry and steric hindrance were close to those of the 3-OH-n-12∶0 acid. 相似文献

Background
Intra-articular hyaluronic acid represents a substantive addition to the therapeutic armamentarium in knee osteoarthritis. We examined the effect of dietary supplementation with a natural extract of chicken combs with a high content of hyaluronic acid (60%) (Hyal-Joint®) (active test product, AP) on pain and quality of life in subjects with osteoarthritis of the knee.Methods
Twenty subjects aged ≥40 years with knee osteoarthritis (pain for at least 15 days in the previous month, symptoms present for ≥6 months, Kellgren/Lawrence score ≥2) participated in a randomized double-blind controlled trial. Ten subjects received AP (80 mg/day) and 10 placebo for 8 weeks. The Western Ontario and McMaster Universities Osteoarthritis Index (WOMAC) and quality of life by the Short Form-36 (SF-36v2) were administered at baseline and after 4 and 8 weeks of treatment.Results
WOMAC pain (primary efficacy variable) was similar in both study groups (mean [SD]) with 6.6 (4.0) points in the AP group and 6.4 (2.7) in the placebo group (P = 0.943). As compared with baseline, subjects in both groups showed statistically significant improvements in WOMAC pain, stiffness, physical function subscales, and in the aggregate score, but the magnitude of changes was higher in the AP group for WOMAC physical function (-13.1 [12.0] vs. -10.1 [8.6], P = 0.575) and total symptoms (-18.6 [16.8] vs. -15.8 [11.4], P = 0.694). At 4 weeks, statistically significant mean changes compared with baseline were observed in the SF-36v2 scales of role-physical, bodily pain, social functioning and role-emotional among subjects in the AP group, and in physical functioning, bodily pain, and social functioning in the placebo group. At 8 weeks, changes were significant for role-physical, bodily pain, and physical component summary in the AP group, and for physical functioning and role-emotional in the placebo arm. Changes in bodily pain and social functioning were of greater magnitude in subjects given AP.Conclusion
This pilot clinical trial showed that daily supplementation with oral hyaluronic acid from a natural extract of chicken combs (Hyal-Joint®) was useful to enhance several markers of quality of life in adults with osteoarthritis of the knee. The results warrant further study in larger sample sizes.15.

Effects of dopants with different valences on the densification behavior and phase composition of a ZrO2–SiO2 nanocrystalline glass-ceramic (NCGC) during pressureless sintering were investigated in this study. The raw powder of Ca2+, La3+, Ce4+ and Ta5+ ions doped ZrO2–SiO2 (referred to as Ca-ZS, La-ZS, Ce-ZS, Ta-ZS, respectively) and pure ZrO2–SiO2 (PZS) sample were synthesized by sol-gel method, followed by pressureless sintering. Compared with the PZS sample, doping of Ca2+ and La3+ ions significantly promoted the densification of the NCGCs. The “densification promotion” effect was attributed to the formation of oxygen vacancies and the decrease of SiO2 viscosity due to doping of aliovalent cations. The dopants with various valences showed significant effects on the phase compositions of the NCGCs during sintering. Doping of Ca2+ ion accelerated the reaction kinetics between ZrO2 nanocrystallites and amorphous SiO2 to yield ZrSiO4. The La3+ ion acted as destabilizer of t-ZrO2, which resulted in a rapid tetragonal (t) to monoclinic (m) ZrO2 phase transformation during sintering, while in the Ta5+ and Ce4+ ions doped sample, the phase transformation occurred gradually. All the doping ions increased the lattice parameters and the volume of t-ZrO2 unit cell, while the effects of the doping ions on the lattice parameters of m-ZrO2 unit cell were more complex. 相似文献

The artificial rearing model was used to investigate the effects of short-term exposure to ethanol on growth and fatty acid
composition of forebrain (FB) and cerebellum (CB) during the brain growth spurt in either n−3 fatty acid-adequate (AD) or
n−3 deficient (DEF) rat pups. On postnatal day 5, offspring of female rats that had been fed AD or DEF diets from day 5 of
life were assigned to three groups: members of two groups were gastrostomized and artificially fed formulas appropriate for
their maternal history, and the third group (suckled control) was fostered to lactating dams of a similar dietary history.
Half of the artificially reared pups in each dietary condition were fed ethanol in their formula (7% vol/vol) in one-quarter
of their daily feedings, while the others received maltose-dextrin substituted isocalorically for ethanol. Blood alcohol concentrations
did not differ betwen the dietary groups. FB weight on postnatal day 9 was lower in ethanol-exposed offspring in both dietary
conditions. Brain fatty acid composition reflected dietary history in that, compared with AD pups, DEF pups had lower percentages
of docosahexaenoic acid, higher percentages of 22∶5n−6, and a higher n−6/n−3 fatty acid ratio. However, the effects of ethanol
exposure were inconsistent, lowering the n−6/n−3 ratio in the phosphatidylethanolamine (PF) fraction in FB but not in CB,
while increasing this ratio in the phosphatidylcholine (PC) fraction in FB of the DEF pups only. Thus, while ethanol had some
effects on lipid composition, there was no difference between the dietary groups in their vulnerability to the effects of
