采用单因素试验和正交试验优化核桃内种皮多酚的回流提取工艺条件,采用清除羟自由基(·OH)、超氧阴离子(·O2-)、过氧化氢(H2O2)评价核桃内种皮多酚的体外抗氧化活性。结果表明,核桃内种皮多酚提取工艺条件:乙醇体积分数45%、液固比60:1(mL∶g)、提取温度70℃、提取时间60 min。在此条件下,多酚得率为25.05%。核桃内种皮多酚具有显著的清除·OH、·O2-和H2O2的能力,其中,当质量浓度为160μg/mL时,对·OH的清除率达100%,优于维生素C。  相似文献

以茶多酚和VC为对照,采用Fenton反应法研究金针菇多糖对羟自由基·OH的清除作用;采用邻苯三酚自氧化法研究金针菇多糖对超氧阴离子自由基O2-·的清除作用。结果表明:在本试验含量范围内,金针菇多糖对·OH和O2-·具有一定的清除能力;在含量相同的情况下,金针菇多糖对·OH和O2-·的清除能力比茶多酚和VC强。此外,研究了金针菇多糖抗油脂自动氧化能力,结果表明:金针菇多糖具有一定的抗油脂氧化活性。  相似文献

研究小麦麸皮不同提取物对2,2-二苯基-1-苦肼基自由基(DPPH·),羟自由基(·OH)和超氧阴离子自由基(O2-·)的清除能力.利用3种不同溶剂(石油醚、乙醇、水)对小麦麸皮依次提取,并采用分光光度法测定各提取物对DPPH·、·OH和O2-·的清除效果.结果表明:乙醇提取物对DPPH·的清除能力(SC50为0.86g/L)最强;对·OH的清除能力,乙醇提取物(SC50为1.88 g/L)和石油醚提取物(SC50为1.94g/L)的清除效果较好,清除能力接近Vc;水提物对O2-·的清除效果(SC50为1.12g/L)强于其他两种提取物,清除能力与Vc相近.  相似文献

为筛选适宜的芒果核仁多酚提取方法,分别采用超声辅助提取法和热回流提取法制备芒果核仁多酚,在单因素试验的基础上,通过响应面法优化两种方法的工艺参数,探究提取方法对多酚得率的影响;利用清除1,1-二苯基-2-苦肼自由基(DPPH·)、2,2-联氨-双-3-乙基苯并噻唑啉-6-磺酸二铵盐阳离子自由基(ABTS+·)、超氧阴离子(O2-·)和羟自由基(·OH)的能力评价所得粗多酚的抗氧化活性。结果表明,超声辅助提取法的芒果核仁多酚提取得率较高,其最佳提取溶液为60%乙醇,其最佳工艺为超声时间30 min,超声功率200 W,超声温度79℃,液料比38∶1(mL/g),该条件下多酚提取得率为11.62%;体外抗氧化试验表明,超声辅助提取法所得芒果核仁多酚对DPPH·、ABTS+·、O2-·和·OH的半清除浓度(IC50)分别为0.02、0.24、592.98 mg/mL和38.76 mg/mL,清除能力均强于热回流提取法所得芒果核仁多酚;此外,超声辅助提取法具有省时和成本低的特点。  相似文献

研究海参脏器多糖HPS1、HPS2在体外对羟自由基(·OH)、超氧阴离子(O2-·)、以及1,1-二苯基苦基苯肼(DPPH·)自由基的清除能力.结果表明,HPS1、HPS2对·OH、O2-·和DPPH·自由基均有一定清除作用,且随着多糖质量浓度的增大,其抗氧化活性逐渐增加.HPS1对·OH、O2-·和DPPH.自由基清除能力IC50分别为0.89、0.98、0.31mg/mL;HPS2对·OH、O2-·和DPPH·自由基清除能力IC50分别为0.64、0.78、0.24mg/mL.2种多糖对DPPH.自由基的清除活性尤其明显.HPS2对于3种自由基的抗氧化活性均强于HPS1.  相似文献

文章用化学发光法研究蓝莓叶黄酮对超氧阴离子自由基(O2-)、羟基自由基(·OH)、过氧化氢(H2O2)的清除能力,并与芦丁进行比较.结果表明:蓝莓叶黄酮对O2-· ,·OH,H2O2等活性氧均具有清除和抑制作用且清除作用与浓度呈剂量关系,但蓝莓叶黄酮对O2-· ,·OH,H2O2的清除能力不如芦丁.蓝莓叶黄酮清除O2-·,·OH,H2O2的IC50分别为13.46 ,2.11,2.58 μg/mL,蓝莓叶黄酮对·OH的清除能力最强.  相似文献

介绍了苹果多酚的超声波提取方法,并通过DPPH法和2-脱氧-D-核糖法研究了苹果提取物的抗氧化性能。结果表明,苹果多酚清除DPPH自由基速度快,其抗氧化能力可与葡萄籽多酚相媲美。研究了其清除DPPH自由基的动力学性质,并与茶多酚、葡萄籽多酚、BHT比较其抗氧化能力。同时,通过2-脱氧-D-核糖法可以看出在一定浓度范围内,苹果提取物有很强的抗氧化能力,其最佳浓度范围为8~10mg/mL,此外,苹果提取物及葡萄籽提取物清除羟基自由基(·OH)的效果远远高于茶多酚。  相似文献

从清除超氧阴离子自由基、清除DPPH自由基、抑制羟基自由基的产生和还原力4个方面研究了云芝多糖的体外抗氧化效果,并同Vc进行比较.结果表明,云芝多糖对O2·清除作用明显,与Vc接近;还原力和清除DPPH·的能力比Vc差;对·OH的清除表现为在1g/L~5g/L浓度范围内,云芝多糖未达到EC50值.  相似文献

以金针菇为材料,采用微波辅助法提取多糖,并制备出乙酰化多糖衍生物,对其进行抗氧化性试验。多种体系的自由基清除实验结果显示:不同浓度的乙酰化金针菇多糖对·O2-、·OH、DPPH·都有一定的清除效果。与供试品Vc清除自由基能力相比,多糖对·O2-的清除率与Vc对·O2-的清除率相差不多;乙酰化金针菇多糖的还原力以及清除·OH和DPPH·的能力比Vc略弱。表明乙酰化金针菇多糖具较强的抗氧化活性。  相似文献

以大球盖菇为原料,通过正交试验L9(34)研究超声波提取多糖的工艺条件。结果表明,最佳提取工艺条件为提取温度65℃、提取时间1.0 h、超声波功率600 W、料液比1∶35(g/mL),此条件下,大球盖菇多糖得率为8.16%。提取效果影响大小的先后顺序为提取时间>提取温度>提取功率>料液比。应用化学发光法对大球盖菇粗多糖的清除·OH和·O2-自由基的能力进行了研究,结果表明:大球盖菇粗多糖对·OH和·O2-自由基均具有明显的清除能力,清除·O2-自由基的IC50为108.60μg/mL,清除·OH自由基的IC50为345.98μg/mL,大球盖菇粗多糖对·O2-自由基的清除能力是对·OH自由基的清除能力的3.2倍。  相似文献

Since grapevine ( Vitis spp .) rootstock material is being traded increasingly as disbudded woody material a lack of distinctive morphological features on such material necessitates an alternative and reliable means of identification. Methods described here were developed for rapid and efficient extraction of DNA from woody samples rich in phenolic compounds and polysaccharides, and for subsequent identification of varieties by RAPD PCR. Using these methods, and with the application of only one selected RAPD primer, we were able to differentiate sixteen rootstock varieties, including the seven varieties most commonly used in Germany. Problems commonly encountered with reproducibility of RAPD patterns were avoided by choosing primers with a dinucleotide sequence and a high G/C content that allowed a rather high annealing temperature of 45°C. Methods described here should also be useful for other horticultural crops, especially those with woody tissues rich in phenolic compounds and polysaccharides.  相似文献

The characterization of the aromatic profile of several apricot cultivars with molecular tracers in order to obtain objective data concerning the aromatic quality of this fruit was undertaken using headspace–solid phase microextraction (HS–SPME). Six apricot cultivars were selected according to their organoleptic characteristics: Iranien, Orangered, Goldrich, Hargrand, Rouge du Roussillon and A4025. The aromatic intensity of these varieties measured by HS–SPME–Olfactometry were defined and classified according to the presence and the intensity of grassy, fruity and apricot like notes. In the six varieties, 23 common volatile compounds were identified by HS–SPME–GC–MS. Finally, 10 compounds, ethyl acetate, hexyl acetate, limonene, β-cyclocitral, γ-decalactone, 6-methyl-5-hepten-2-one, linalool, β-ionone, menthone and (E)-hexen-2-al were recognized by HS–SPME–GC–O as responsible of the aromatic notes involved in apricot aroma and considered as molecular tracers of apricot aromatic quality which could be utilized to discriminate apricot varieties.  相似文献

The advent of the functional barrier concept in food packaging has brought with it a requirement for fast tests of permeation through potential barrier materials. In such tests it would be convenient for both foodstuffs and materials below the functional barrier (sub-barrier materials) to be represented by standard simulants. By means of inverse gas chromatography, liquid paraffin spiked with appropriate permeants was considered as a potential simulant of sub-barrier materials based on polypropylene (PP) or similar polyolefins. Experiments were performed to characterize the kinetics of the permeation of low molecular weight model permeants (octene, toluene and isopropanol) from liquid paraffin, through a surrogate potential functional barrier (25 μm-thick oriented PP) into the food simulants olive oil and 3% (w/v) acetic acid. These permeation results were interpreted in terms of three permeation kinetic models regarding the solubility of a particular model permeant in the post-barrier medium (i.e. the food simulant). The results obtained justify the development and evaluation of liquid sub-barrier simulants that would allow flexible yet rigorous testing of new laminated multilayer packaging materials.  相似文献

This paper describes the second part of a project undertaken to develop certified mussel reference materials for paralytic shellfish poisoning toxins. In the first part two interlaboratory studies were undertaken to investigate the performance of the analytical methodology for several PSP toxins, in particular saxitoxin and decarbamoyl-saxitoxin in lyophilized mussels, and to set criteria for the acceptance of results to be applied during the certification exercise. Fifteen laboratories participated in this certification study and were asked to measure saxitoxin and decarbamoyl-saxitoxin in rehydrated lyophilized mussel material and in a saxitoxin-enriched mussel material. The participants were allowed to use a method of their choice but with an extraction procedure to be strictly followed. The study included extra experiments to verify the detection limits for both saxitoxin and decarbamoyl-saxitoxin. Most participants (13 of 15) were able to meet all the criteria set for the certification study. Results for saxitoxin.2HCl yielded a certified mass fraction of <0.07 mg/kg in the rehydrated lyophilized mussels. Results obtained for decarbamoyl-saxitoxin.2HCl yielded a certified mass fraction of 1.59+/-0.20 mg/kg. The results for saxitoxin.2HCl in enriched blank mussel yielded a certified mass fraction of 0.48 +/- 0.06 mg/kg. These certified reference materials for paralytic shellfish poisoning toxins in lyophilized mussel material are the first available for laboratories to test their method for accuracy and performance.  相似文献
