Filter bags facilitate the measurement of amylase-treated neutral detergent fiber (aNDF) and in vitro (IV) undigested aNDF (uNDF) by eliminating the transfer of residues from beakers into filtration crucibles. The objectives of this study were to (1) determine effects of filter bags on recovery of aNDF and (2) evaluate effects of filter bags on IV uNDF. For study 1, 6 samples each of grass hay (GR), alfalfa (AL), and corn silage (CS) were selected. Large standard deviations (SD) of ash-free aNDF (aNDFom) for samples in each forage type indicated compositional diversity (15.1, 7.45, and 12.9% of DM for GR, AL, and CS, respectively), and starch SD for CS was 16.4% of DM. Samples were weighed into Berzelius beakers or filter bags [25-µm pores (F57) or 6-µm pores (F58); Ankom Technology, Macedon, NY] for measurement of aNDF and aNDFom. All samples were extracted with neutral detergent, thermostable α-amylase, and sodium sulfite, and then soaked in boiling water and then acetone. Residues from beakers were filtered through a sea sand–covered GF/D filter (Whatman, Marlborough, MA) in Gooch crucibles (CR). Filter bags were extracted in a pressurized chamber at 100°C. The aNDF values did not differ between F57 and CR, but F58 was greater than CR for CS and AL. For GR, F58 was greater than CR for aNDFom. For study 2, diverse samples with large SD of aNDFom (20.7, 7.45, and 12.9% of DM for GR, AL, and CS, respectively) were weighed as loose powder into medium bottles (LS) or F57 bags, which were weighted to prevent floating. Blended ruminal fluid from 3 steers fed a 30% aNDFom diet was used as inoculum. Three samples of 1 forage type were randomly assigned to 1 of 6 IV runs using both treatments (LS and F57), and 3 bottles of each sample–treatment combination were removed after 12 h and 2 were removed after 120 h to measure uNDF. For LS, residues were extracted as in study 1 for CR. For F57, bags were rinsed in cold water and extracted as described in study 1. After 12 h, uNDF of F57 was greater than LS in CS, AL, and overall types. Ash-free uNDF (uNDFom) after 12 h of F57 was greater than LS in CS and overall types. After 120 h, F57 was greater than LS for uNDF of CS, but no differences were detected for uNDFom. The SD of uNDFom, but not uNDF, was higher after 12 and 120 h for F57 compared with LS. From 6 to 96 h, overall gas production of F57 was less than LS, and F57 was less than LS for CS from 3 to 96 h. Overall, LS gave greater maximum and faster rates of gas production than F57, as did AL and CS, but lag did not vary. Results indicate that filter bags affected aNDF and aNDFom measurement and inhibited fermentation for some materials.  相似文献

Nine by‐products used in animal nutrition were examined for their nutritive value by determining the chemical composition (crude protein, crude fat (ether extract), neutral detergent fibre, acid detergent fibre, lignin), in vitro organic matter digestibility and rumen fermentation kinetics (from gas production curves measured in vitro). The by‐products studied were giant pumpkin, red pepper, stem broccoli, brewer's grain, fresh artichoke, scalded artichoke, lemon peel, orange peel and melon. The nutritive value was very variable, depending on the by‐product and on the process applied to the material during industrial processing. In vitro gas production was measured for 500 mg dry matter in quadruplicate at 39 °C after 6, 12, 24, 36, 48 and 72 h of incubation and fitted to single‐pool exponential equations. The fermentation kinetics indicated that brewer's grain was the only by‐product which fitted well with the p = a + b(1 ? e?ct) equation model; all other by‐products had a very fast degradation rate and their gas production fitted the equation p = b(1 ? e?c(t?L)), because in the other exponential model a was negative. © 2002 Society of Chemical Industry  相似文献

This study determined nitrogen degradability and digestibility of rumen undegradable nitrogen using mobile nylon bag (MNB) and pepsin/pancreatin in vitro technique (IV) of 40 browse species. Thirty Ethiopian highland sheep fitted with rumen cannulae were used in nitrogen (N) degradability studies. Six steers fitted with rumen cannulae were used in preparation of 16-h and 24-h ruminal undegraded residues and four steers fitted with distal abomasal cannulae were used in MNB technique. The browses varied widely in nitrogen solubility (15–468 g kg−1), potential degradability (223–976 g kg−1), rate of degradation (0·13–24% h−1) and effective degradability (135–821 g kg−1). The apparent N digestibility (ND) of the rumen undegraded residues differed significantly (P<0·05) among browse species. No significant difference (P>0·05) was observed in ND of 16-h and 24-h residues. The ND of the 16-h residue varied from -218 to 759 g kg−1 and 169 to 851 g kg−1 for MNB and IV methods, respectively. Browse species with high tannin contents such as Acacia hockii, A horrida, A melanoxylon, A persiciflora, A salicina, A saligna and Flemingia macrophylla had high rumen by-pass and a low ND, while Sesbania spp and A nilotica with low tannin contents underwent rapid and extensive dry matter and nitrogen degradation in the rumen. Acacia sieberiana, Chamaecytisus palmensis, Erythrina spp, Gliricidia sepium, Samanea saman and Enterolobium cyclocarpum had high proportions of protein escaping rumen degradation (BP ) and with a high proportion of the by-pass protein digested in the intestine, therefore these browses had a high potential as protein supplements. The ND measured with the MNB were significantly lower (P<0·001) than by the IV method. The correlation between MNB and IV was high and significant (R2=0·89, P<0·0001) as also indicated by the regression equation (SE in parentheses): MNB=-22·8 (4·55)+1·0 (0·08)IV (RSD=10·56, R2=0·79, n=40, P<0·001). The intercept of the linear relationship obtained was different from zero while the slope was not different from unity. Multiple regression analysis suggested that some of the unexplained variation could be accounted for by either nitrogen, acid detergent fibre, total phenolics or neutral detergent fibre bound tannin levels in browses. The IV method is accurate for estimating digestibility of ruminally undegradable N, and hence its use would considerably reduce the need for delicate surgery and the elaborate procedures involving the MNB technique. © 1998 SCI.  相似文献

Methane production from ruminant livestock varies with the diet as a result of factors such as dry matter intake, diet composition, and digestibility. To estimate the effect of dietary composition and feed additives, CH4 production can be measured in vitro as a first step because large numbers of samples can be incubated and analyzed at the same time. This study evaluated a recently developed in vitro method for prediction of in vivo CH4 production by examining the relationship between predicted and observed CH4 production values. A total of 49 different diets (observations), used in previous 13 in vivo studies, were selected to include diets varying in nutrient composition. Methane production was measured in all in vivo studies by respiration chambers or the GreenFeed system (C-Lock Inc., Rapid City, SD). Overall, the in vitro system predicted CH4 production well (R2 = 0.96), but the values obtained were slightly underestimated compared with observed in vivo values (mean 399 L/d compared with 418 L/d: root mean square prediction error = 51.6 L/d or 12.3% of observed mean). Further analysis of the effect on residuals showed no significant relationship between CH4 production and most factors known to affect CH4 production such as dry matter intake, digestibility, and dietary concentrations of fat and starch. However, some factors included in the model were not well predicted by the system, with residuals negatively related to neutral detergent fiber concentration and positively related to concentrate proportion. The in vitro system can thus be useful for screening diets and evaluation of feed additives as a first step that can be best interpreted when feeding cows at maintenance level.  相似文献

BACKGROUND: Current feed evaluation systems for dairy cattle assume that nutritive values of feed components can be added, but do not take into account possible interactions among feed components. The main objective of this study was to investigate associative effects on in vitro gas production from total mixed ration (TMR), based on grass silage and cereal concentrate, and separate TMR components. TMR was incubated with inocula of free rumen liquid (FRL) and particle‐associated rumen liquid (PAL) for 48 h and donor cows were fed TMRs with three different particle sizes to evaluate their influence on associative effects. RESULTS: Associative effects on gas production largely occurred at 2 and 4 h of incubation and dissipated with time of incubation. Incubation of TMR with PAL increased (P < 0.01) gas production compared to FRL at all incubation times; however, associative effects were not different between rumen inocula. Dietary particle size affected associative effects on gas production when TMR was incubated with FRL (P < 0.05) compared to PAL, particularly at early hours of incubation. CONCLUSIONS: Associative effects at early incubation hours emphasise that summing the nutritive values of feedstuffs may underestimate fermentation intensity of TMR which may have implications for prediction of ruminal fermentation yield. This study determined the chemical composition, in situ ruminal nutrient disappearance and in vitro gas production kinetics of three cactus varieties grown in northeastern Brazil. The varieties were Gigante, IPA‐20 and Miúda. Results of the chemical analysis showed no significant differences in ash, ether extract, crude protein (CP) and neutral detergent fibre (NDF) between the cactus varieties. However, acid detergent fibre was highest (P < 0.05) for IPA‐20, intermediate (P < 0.05) for Gigante, and lowest (P < 0.05) for Miúda. Fractionation of carbohydrate and true protein based on rates of ruminal degradation indicated that the main carbohydrate component was the rapidly degradable fraction, whereas the main true protein component was the intermediately degradable fraction. No differences in carbohydrate or protein fractions were observed between the cactus varieties. Results of the in situ experiment showed no differences in ruminal dry matter (DM, average 803 g kg?1 of DM), CP (900 g kg?1 of CP) and NDF (611 g kg?1 of NDF) disappearance between the three cactus varieties after 48 h of ruminal incubation. Potential gas production at the end of 48 h of incubation was higher (P < 0.05) for Gigante than for the Miúda or IPA‐20 variety. However, rate of gas production and lag time in gas production were similar for the three cactus varieties at 6.8% h?1 and 0.6 h respectively. The fermentation of the neutral detergent‐soluble (NDS) fraction of three forages (alfalfa hay, Italian ryegrass + oats hay and corn silage) was measured using a curve subtraction technique with in vitro gas production data from the whole forage (WF) and the isolated neutral detergent fibre (NDF). NDF disappearance and volatile fatty acid (VFA) production were determined. There was no significant difference between the VFA patterns from the whole forage and the isolated NDF. There were significant (P < 0.001) linear correlations between the volume of gas and moles of VFA produced and the mass of fibre digested in the NDF samples. Using the monophasic curve model, maximum gas production rates (RM) for the whole forages and the isolated NDF and NDS fractions can be calculated. For all three forages we obtained RM‐NDS > RM‐WF > RM‐NDF. Trends in the rates of gas and VFA production were similar in the whole forages. The NDF showed a high superimposition between substrate degradability and VFA production. Our experiment confirmed the curve subtraction technique as a simple method to obtain information on the size and digestion kinetics of the NDS fraction, which helps in understanding the nutritional significance of this important fraction of the forages studied. © 2001 Society of Chemical Industry  相似文献

BACKGROUND: Euphorbia esula L. (leafy spurge) is indigenous to Eurasia and has been known to cause grazing aversion in ruminant species. As a result, E. esula encroachment has negatively impacted rangelands in the Northern Great Plains and Intermountain West of the USA, as well as southern Canada. Our objectives were to evaluate the effect of increasing concentrations of E. esula on in vitro dry matter digestibility (DMD) and gas production. Two ruminally‐cannulated cows and ewes were used as rumen inoculum donors. To accomplish objectives, two studies were conducted. In study 1, animals were fed exclusively a barley hay (12% crude protein (CP), 55.4% neutral detergent fiber (NDF), DM basis) diet; whereas in study 2, animals were fed a diet of 15% E. esula (21.9% CP, 48% NDF, DM basis) and 85% barley hay based on previous day intake. RESULTS: The 24 and 48 h in vitro and 96 h gas production indicate that, regardless of inoculum source or substrate fermented, DMD was not influenced. Differences, however, were consistently observed across studies for NDF disappearance. CONCLUSION: Regardless of inoculum source NDF disappearance was greater when substrate being fermented contained 0%, 80%, or 100% E. esula. Published 2011 by John Wiley & Sons, Ltd.  相似文献

BACKGROUND: Cellulose and hemicellulose are quantitatively the most important structural carbohydrates present in ruminant diets. Rumen micro‐organisms produce enzymes that catalyse their hydrolysis, but the complex network formed by structural carbohydrates and lignin reduces their digestibility and restricts efficient utilisation of feeds by ruminants. This study aimed to produce two enzymatic extracts, apply them in ruminant diets to determine the best levels for ruminal digestibility and evaluate their effects on in vitro digestibility. RESULTS: In experiment 1 a two‐stage in vitro technique was used to examine the effects of different enzymatic levels of Aspergillus japonicus and Aspergillus terricola on tropical forages. Enzyme addition had minor effects on corn silage at the highest enzymatic level. In experiment 2 an in vitro gas production (GP) technique was applied to determine apparent in vitro organic matter digestibility and metabolisable energy. The addition of enzymes in GP showed interesting results. Good data were obtained using sugar cane and Tifton‐85 hay supplemented with extracts of A. japonicus and A. terricola respectively. CONCLUSION: Overall, the study suggests that addition of crude extracts containing exogenous fibrolytic enzymes to ruminant diets enhances the effective utilisation of ruminant feedstuffs such as forages. This study evaluated the fermentation characteristics and in vitro gas production of sudangrass (Sorghum sudanese) silage with tea leaf waste of green tea, oolong tea and black tea added. All types of tea leaf waste contain large amounts of nitrogen (N) and tannins, and a small amount of low‐water‐soluble carbohydrates. Sudangrass was preserved without (control) and with green tea waste (GTW), oolong tea waste (OTW) or black tea waste (BTW) at rates of 50, 100 and 200 g kg?1 fresh matter in laboratory silos. The pH and butyric acid levels were significantly lower in GTW at all rates and in OTW at the 200 g kg?1 rate, whereas those levels were high in the control and BTW‐treated silage. Lactic acid level was appreciably higher in GTW‐ and OTW‐treated silage than the control and BTW‐treated silage. The addition of all types of tea waste increased the total N and tannins of the silage, whereas the acid detergent insoluble N was increased in the OTW‐ and BTW‐treated silage. GTW treatment increased gas production within a 96‐h period compared with the control, whereas OTW and BTW additions at 200 g kg?1 rates caused lower gas production. It is concluded that the fermentation characteristics and feed value of the silage incorporating with tea leaf wastes are different for these types of tea leaf. This study compared measured gas production (GP) and computed CH4 production values provided by closed or vented bottles connected to gas collection bags. Two forages and 3 concentrates were incubated. Two incubations were conducted, where the 5 feeds were tested in 3 replicates in closed or vented bottles, plus 4 blanks, for a total of 64 bottles. Half of the bottles were not vented, and the others were vented at a fixed pressure (6.8 kPa) and gas was collected into one gas collection bag connected to each bottle. Each bottle (317 mL) was filled with 0.4000 ± 0.0010 g of feed sample and 60 mL of buffered rumen fluid (headspace volume = 257 mL) and incubated at 39.0°C for 24 h. At 24 h, gas samples were collected from the headspace of closed bottles or from headspace and bags of vented bottles and analyzed for CH4 concentration. Volumes of GP at 24 h were corrected for the gas dissolved in the fermentation fluid, according to Henry’s law of gas solubility. Methane concentration (mL/100 mL of GP) was measured and CH4 production (mL/g of incubated DM) was computed using corrected or uncorrected GP values. Data were analyzed for the effect of venting technique (T), feed (F), interaction between venting technique and feed (T × F), and incubation run as a random factor. Closed bottles provided lower uncorrected GP (−18%) compared with vented bottles, especially for concentrates. Correction for dissolved gas reduced but did not remove differences between techniques, and closed bottles (+25 mL of gas/g of incubated DM) had a greater magnitude of variation than did vented bottles (+1 mL of gas/g of incubated DM). Feeds differed in uncorrected and corrected GP, but the ranking was the same for the 2 techniques. The T × F interaction influenced uncorrected GP values, but this effect disappeared after correction. Closed bottles provided uncorrected CH4 concentrations 23% greater than that of vented bottles. Correction reduced but did not remove this difference. Methane concentration was influenced by feed but not by the T × F interaction. Corrected CH4 production was influenced by feed, but not by venting technique or the T × F interaction. Closed bottles provide good measurements of CH4 production but not of GP. Venting of bottles at low pressure permits a reliable evaluation of total GP and CH4 production.  相似文献

Sources of variation in rates of in vitro ruminal protein degradation   总被引:2,自引:0,他引:2  
Rates and extents of ruminal protein degradation for casein, solvent soybean meal (SSBM), expeller soybean meal (ESBM), and alfalfa hay were estimated from net appearance of NH3 and total amino acids in in vitro media containing 1 mM hydrazine and 30 mg/L of chloramphenicol. Protein was added at 0.13 mg of N/mL of medium, and incubations were conducted for 4 to 6 h, usually with hourly sampling. Inocula were obtained from ruminally cannulated donor cows fed diets of grass silage or alfalfa and corn silages plus concentrates. Preincubation or dialysis of inocula was used to suppress background NH3 and total amino acids; however, preincubation yielded more rapid degradation rates for casein and SSBM and was used in subsequent incubations. Preincubation with added vitamins, VFA, hemin, or N did not alter protein degradation. Protein degradation rates estimated for SSBM, ESBM, and alfalfa were not different when computed from total N release or N release in NH3 plus total amino acids, regardless of whether amino acids were quantified using ninhydrin colorimetry or o-phthalaldehyde fluorescence. Accounting for the release of peptide-N also did not affect estimated degradation. However, casein degradation rates were more rapid when using total N release or accounting for peptide-N, indicating significant accumulation of small peptides during its breakdown. Rates also were more rapid with inocula from lactating cows versus nonlactating cows with lower feed intake. Protein degradation rates were different due to time after feeding: casein rate was more rapid, but SSBM and ESBM rates were slower with inocula obtained after feeding. Several characteristics of ruminal inoculum that influenced breakdown of the rapidly degraded protein casein did not appear to have direct effects on degradation of protein in soybean meal.  相似文献

Sixty samples of ‘ForagePlus’ oat were selected from a previous plot study for analysis of in vitro gas production (IVGP) on the basis of 2 factors: (1) high (n = 29) or low (n = 31) neutral detergent fiber (NDF; 62.7 ± 2.61 and 45.1 ± 3.91%, respectively); and (2) the range of water-soluble carbohydrates (WSC) within the high- and low-NDF groups. For the WSC selection factor, concentrations ranged from 4.7 to 13.4% (mean = 7.9 ± 2.06%) and from 3.5 to 19.4% (mean = 9.7 ± 4.57%) within high- and low-NDF forages, respectively. Our objectives were to assess the relationships between IVGP and various agronomic or nutritional characteristics for high- and low-NDF fall-oat forages. Cumulative IVGP was fitted to a single-pool nonlinear regression model: Y = MAX × (1 – e [–K× (t – lag)]), where Y = cumulative gas produced (mL), MAX = maximum cumulative gas produced with infinite incubation time (mL), K = rate constant, t = incubation time (h), and lag = discrete lag time (h). Generally, cumulative IVGP after 12, 24, 36, or 48 h within high-NDF fall-oat forages was negatively correlated with NDF, hemicellulose, lignin, and ash, but positively correlated with WSC, nonfiber carbohydrate (NFC), and total digestible nutrients (TDN). For low-NDF fall-grown oat forages, IVGP was positively correlated with growth stage, canopy height, WSC, NFC, and TDN; negative correlations were observed with ash and crude protein (CP) but not generally with fiber components. These responses were also reflected in multiple regression analysis for high- and low-NDF forages. After 12, 24, or 36 h of incubation, cumulative IVGP within high-NDF fall-oat forages was explained by complex regression equations utilizing (lignin:NDF)2, lignin:NDF, hemicellulose, lignin, and TDN2 as independent variables (R2 ≥ 0.43). Within low-NDF fall-grown oat forages, cumulative IVGP at these incubation intervals was explained by positive linear relationships with NFC that also exhibited high coefficients of determination (R2 ≥ 0.75). Gas production was accelerated at early incubation times within low-NDF forages, specifically in response to large pools of WSC that were most likely to be present as forages approached boot stage by late-fall.  相似文献

探讨青贮玉米秸与豆秸组合对瘤胃发酵参数的影响,以确定适宜的饲料组合比例。本试验将青贮玉米秸与豆秸分别按0∶100、20∶80、40∶60、60∶40、80∶20、100∶0的比例进行组合,利用体外瘤胃发酵技术,分析不同比例组合对产气量、pH值、氨态氮、菌体蛋白和瘤胃干物质、中性洗涤纤维和酸性洗涤纤维的影响,并计算出各组合的单项组合效应值和综合组合效应值,进而筛选出两种饲料组合的适宜比例。结果表明:青贮玉米秸-豆秸不同比例组合后在产气量上差异显著(P〈0.05),以80∶20时最大;各比例对体外瘤胃液pH值影响差异不显著(P〉0.05);氨态氮质量浓度随青贮玉米秸比例的增加而增加,在40~50mg/dl变化;菌体蛋白质量浓度在两者比例为80∶20时最高;青贮玉米秸-豆秸不同比例组合的干物质、中性洗涤纤维和酸性洗涤纤维的瘤胃有效降解率均以80∶20时最大。在本试验条件下,以多项组合效应评定指数评定组合效应,青贮玉米秸与豆秸比例为80∶20时较为适宜,综合组合效应指数达到最大。  相似文献
