Activity of the polyphenol oxidase (PPO) from eggplant fruit (Solanum melongena L.) on phenolic compounds of an extra virgin olive oil (EVOO) was studied. In standardized reaction solutions, the eggplant PPO, isolated in the laboratory, depleted completely chlorogenic and caffeic acids, oleuropein, and verbascoside, while the levels of hydroxytyrosol reduced by half. Conversely, no activity of the PPO was observed on the gallic and protocatechuic acids nor on mono‐phenols, such as tyrosol and the p‐coumaric, o‐coumaric, and ferulic acids. PPO activity on phenols extracted from eggplant fruit and EVOO confirmed the enzyme substrate specificity and caused a significant decrease in the measure of total phenols and o‐diphenols. Similarly, PPO crude extract caused a significant decrease of polyphenols directly in the EVOO. Moreover, maximum degradation of EVOO polyphenols was observed when olive oil was homogenized with eggplant fruit pulp to form a cream‐like purée. In fact, immediately after the preparation, total phenols and o‐diphenols of the olive oil recovered from the eggplant‐oil purée were decreased by ~80% and 100% compared to those of the initial EVOO. As a consequence, the oxidative stability of the recovered oil was ~60% lower than that of the initial EVOO. In conclusion, in the preparation of vegetable preserves, a residual activity of phenol oxidase may adversely affect the quality and shelf life of the extra virgin olive oil used as covering.  相似文献

This study was carried out to evaluate the influence of the lipase‐producing yeasts, present in the brine, on the quality of table olive oil. The oily fraction of olives processed microbiologically using the β‐glucosidase‐positive, lipase‐negative yeast Candida wickerhamii 1542 showed an excellent quality, such as the typical extra‐virgin olive oil. In contrast, the quality of the oily fraction of olives debittered microbiologically with the β‐glucosidase‐ and lipase‐producing yeast Williopsis californica 1639 worsened dramatically to such a point that the acidity of the oily fraction increased more than 90%. The microbiological analysis of brines showed that the lipase‐producing yeasts are part of the natural yeast population of the brine, and when their presence is high, it can damage significantly the quality of table olives. The lipase‐producing yeast W. californica 1639 in the brine showed the maximum lipolytic activity at pH 6, while the NaCl of the brine, at the maximum concentration of 12%, inhibited lipase activity by 70%. The concentration of the polyphenols present in the brine was not able to control the lipolytic activity of W. californica 1639 during the microbiological debittering process of table olives.  相似文献

Refined olive oil and olive‐pomace oil were enriched with olive leaf phenolic compounds in order to enhance its quality and bring it closer to virgin olive oil. The changes that occurred in the concentrations of pure oleuropein, oleuropein aglycone, hydroxytyrosol acetyl and α‐tocopherol at 400 µg/kg of oil during the storage of refined olive oil and olive‐pomace oil under accelerated conditions (50 °C) were investigated. In a period of 4 months, α‐tocopherol decomposed by 75% whereas less than 40% of the phenols were lost. During storage, enzymatic olive leaf extract hydrolysate that contains two major compounds, hydroxytyrosol and oleuropein aglycone showed the highest antioxidant activity and the lowest detected stability, followed by oleuropein. The oleuropein in olive leaf extracts exhibited similar degradation profiles, reducing by 60–50% and 80% for the olive oil and olive‐pomace oil in 6 months, respectively. The acetylated extract, however, displayed a loss of 10 and 5% in olive oil and olive‐pomace oil, respectively. In the fatty acid composition, an increase in oleic acid and a decrease in linoleic acid were observed. The antiradical activities of the olive oil and olive‐pomace oil enriched with olive leaf phenolic compounds at 400 ppm showed that enzymatic hydrolysate extract had the highest protective effect against oil oxidation. Based on the Rancimat method, the oils with added leaf enzymatic hydrolysate extract had the lowest peroxide value and the highest stability. After 6 months of storage and at 120 °C, the oxidative resistance of refined olive oil and olive‐pomace oil reached 0.71 and 0.89 h, respectively, whereas that of the non‐enriched samples fell to zero.  相似文献

Virgin olive oil has high levels of phenolic compounds that are highly bioavailable; these compounds are receiving considerable attention for their antioxidant activity, closely related to the prevention of non‐communicable chronic diseases. The aim of this work was to characterize the phenolic profile and antioxidant capacity of monovarietal olive oils cvs. Arauco, Arbequina, Farga and Empeltre produced in Argentina. This study focused on the relationship between the single molecules or classes of molecules quantified by SPE‐CZE, the corresponding Folin‐Ciocalteu results, and antioxidant capacity using three different tests. Fifteen compounds were simultaneously determined: tyrosol, vinylphenol, oleuropein, hydroxytyrosol, rutin, catechin, naringenin, cinnamic acid, chlorogenic acid, syringic acid, luteolin, apigenin, vanillin acid, quercetin, and caffeic acid. The phenolic contents of the monovarietal olive oils show significant differences between different varieties (p < 0.05), with positive and significant Pearson's correlation found between Folin–Ciocalteu and CZE. Besides, the correlation between the content of total polyphenols and antioxidant capacity was high for all the antioxidant assays performed. When analyzing the correlation coefficients of the different families of phenolic compounds studied, simple phenols and cinnamic acid derivatives show a higher correlation with antioxidant capacity. Thus, findings obtained in this study demonstrated that Arauco olive oil, autochthonous for Argentina, possesses the highest antioxidant/free‐radical scavenging properties, which are very likely due to the presence of high contents of phenolic compounds.  相似文献

The main objective of this study was to evaluate the effect of different deficit irrigation treatments (control, regulated deficit irrigation [RDI]‐1, RDI‐2, and RDI‐3) on the phenolic profile of the olive paste and oil content. Irrigation treatments with more stress water led to a considerable increase in the phenolic compounds of olive paste, especially in oleuropein (60.24%), hydroxytyrosol (82%), tyrosol (195%), and verbascoside (223%) compared to control. A significant increase in the content of total flavonoids and phenolic acids was also observed for these samples. In virgin olive oils (VOO) elaborated from the most stressed olive trees (RDI‐2 and RDI‐3), a noticeable increase in phenolic substances with antioxidant properties (oleuropein, hydroxytyrosol, tyrosol, secoiridoid derivatives, and o‐vanillin) was observed. Consequently, water stress conditions improved antioxidant activity of VOO.  相似文献

The phenolic composition and antioxidant activity of several monovarietal extra virgin olive oils used as blenders for the production of Collina di Brindisi protected designation of origin (PDO) oil, produced between December 2008 and January 2009 using two‐phases or three‐phases extraction system, were evaluated and compared with other manufacturer products designated as PDO. Oils were taken from the most representative ones industrial oil mills in the PDO geographical area. The parameters assessed were free acidity, peroxide value, K232 and K270 indices, organoleptic characteristics, total phenolic content (TPC), phenolic profile, and antioxidant activity coefficient (AAC). The phenolic contents and profiles of the monovarietal oils showed remarkable differences with respect to PDO oils. The variables that exerted a major influence on phenols concentration were the maturity degree of olives (December>January), followed by the extraction system (two‐phase>three‐phase), and place of growing. The Pearson r correlation index showed that AAC was positively correlated with TPC, p‐coumarate, and 3,4‐DHPEA‐EA, and negatively correlated with peroxide value. Practical applications: The results provide detailed information about: (i) the phenolic composition and the AAC of several monovarietal extra virgin olive oils used as blenders for the production of a PDO oil; (ii) the impact of genetic variability, place of growing, olive maturity degree, and extraction technology on oil phenol compounds; and (iii) the relationships among each phenolic compound and AAC, and their potential utilization as analytical index of antioxidant activity. It is important to study the phenolic compounds and antioxidant activity of monovarietal extra virgin olive oils used to produce PDO oil and to compare with the relative PDO samples in order to define a possible analytical tool able to verify what is stated in the label for consumer information and protection.  相似文献

The induction period (IP), determined using accelerated methods such as the Rancimat test, is a parameter that has been used to predict the shelf life of virgin olive oil. The oxygen radical absorbance capacity (ORAC) has recently been proposed as a quality index of virgin olive oil because it measures the efficiency of phenolic compounds in the protection against peroxyl radicals. This study aims to investigate relationships between the ORAC and IP values and proposes ORAC as a new parameter of virgin olive oil stability. The concentrations of phenolics, o-diphenols, tocopherol, β-carotene, lutein, and ORAC and IP values were determined in 33 virgin olive oils. Regression analyses showed that both ORAC and IP values correlate with total phenols and o-diphenols with highly significant indices, whereas the correlations of both ORAC and IP with tocopherols, β-carotene, and lutein were not significant. The ORAC values correlate with the IP values with low but significant indices (R=0.42; P<0.02). The results confirm the key role of phenolic compounds in accounting for the shelf life of virgin olive oil and suggest that the ORAC parameter may be used as a new index of quality and stability.  相似文献

Polar extracts of extra‐virgin olive oils (EVOO) contain a large number of phenolic compounds with antioxidant properties. The antioxidant capacity can be measured by different reaction mechanisms, as the single electron transfer (SET) or the hydrogen atom transfer (HAT). In this work, the total phenolic content (TPC) by the Folin‐Ciocalteu method and its correlation with four antioxidant capacity assays (FRAP, ABTS, DPPH? and ORAC) were evaluated for EVOO polar extracts. It was observed that the higher the total phenolic compounds in the EVOO extracts, the higher the antioxidant capacities, regardless of the method employed. The reaction mechanism observed for TPC by Folin‐Ciocalteu method and also for FRAP, ABTS and DPPH? antioxidant capacity assays is a single electron transfer, thus, a high correlation among their results is expected. However, the correlation between TPC and ORAC results was also high and significant, allowing to conclude that EVOO phenolic compounds are able to react by the hydrogen atom transfer mechanism, which indicates that they can act as effective radical chain‐breaking antioxidants. These results suggest that, for the EVOO polar extracts, TPC by Folin‐Ciocalteu and ORAC assays could be sufficient to evaluate their in vitro antioxidant capacity.  相似文献

The effect of extraction systems on the phenolic composition of virgin olive oils obtained from two different Italian cultivars (Coratina and Oliarola) was determined. The oils extracted using two-phase centrifugation showed in all cases higher phenolic concentration in comparison to oils obtained from three-phase centrifugation. In particular, the highest differences were observed for aglykone derivatives of oleuropein (3,4-DHPEA-EDA and 3,4-DHPEA-EA) that are the most concentrated antioxidant phenolic compounds of virgin olive oil. These results were confirmed by the autoxidation stability of the oils examined.  相似文献

We performed a survey on the yield, quality, and chemical characteristics of virgin olive oils from two olive varieties in Croatian Istria: Frantoio and Ascolana tenera, on Cherry leafroll virus‐infected and virus‐noninfected trees and on two harvest dates. Free acidity, peroxide value, specific spectrophotometric absorptions at 232 and 270 nm, fatty acid composition, total phenols, o‐diphenols, oil color, and pigments were determined. Infected olives had lower oil yield and maturity index versus healthy ones. Oils from infected fruits had significant lower value of K232 and K270 and very elevated total phenols content compared to those obtained from healthy olives. Infected Frantoio gave a lower content of o‐diphenols than the healthy ones, which is in contrast to infected Ascolana that had higher values. The aim of this study is to determine the chemical changes in virgin olive oils from healthy and infected trees related to virus influence. According to our knowledge, this is the first survey on the possible influence of viruses on olive fruits, oil yield, and virgin olive oil quality. Practical applications : There are only few papers which analyze the influence of viruses on crops (especially influence on wine quality) and their effects on yield or other agronomic parameters. This work evaluates for the first time the impact of Cherry leafroll virus on the quality of virgin olive oil obtained from Frantoio and A. tenera varieties in terms of basic parameters related to the hydrolitic and oxidative status, content in antioxidant compounds, and in pigments as well as in fatty acid composition.  相似文献

In this study, changes in an extra virgin olive oil treated with lycopene during storage were analyzed. Pure lycopene (0.5 and 1.0 mg) obtained from tomato was added to two separate bottles, each containing 100 mL of extra virgin olive oil, while another bottle containing the same oil was stored without any treatment. Samples enriched with pure lycopene showed PV remarkably lower than the sample without lycopene, and the total phenol contents were higher in treated oil samples than in the reference sample. In addition, a good correlation (r 2=0.969) between total phenol content and antioxidant power calculated as Trolox Equivalent Antioxidant Capacity (TEAC) was observed. The concentration of added lycopene decreased very slowly; after about 8 mon its residual value was over 60% with respect to the initial concentration.  相似文献

On the basis of the results obtained with pilot studies conducted in vitro on human low density lipoprotein (LDL) and on cell cultures (Caco-2), which had indicated the ability of certain molecules present in olive oil to inhibit prooxidative processes, an in vivo study was made of laboratory rabbits fed special diets. Three different diets were prepared: a standard diet for rabbits (diet A), a standard diet for rabbits modified by the addition of 10% (w/w) extra virgin olive oil (diet B), a modified standard diet for rabbits (diet C) differing from diet B only in the addition of 7 mg kg−1 of oleuropein. A series of biochemical parameters was therefore identified, both in the rabbit plasma and the related isolated LDL, before and after Cu-induced oxidation. The following, in particular, were selected: (i) biophenols, vitamins E and C, uric acid, and total, free, and ester cholesterol in the plasma; (ii) proteins, triglycerides, phospholipids, and total, free, and ester cholesterol in the native LDL (for the latter, the dimensions were also measured); (iii) lipid hydroperoxides, aldehydes, conjugated dienes, and relative electrophoretic mobility (REM) in the oxidized LDL (ox-LDL). In an attempt to summarize the results obtained, it can be said that this investigation has not only verified the antioxidant efficacy of extra virgin olive oil biophenols and, in particular, of oleuropein, but has also revealed a series of thus far unknown effects of the latter on the plasmatic lipid situation. In fact, the addition of oleuropein in diet C increased the ability of LDL to resist oxidation (less conjugated diene formation) and, at the same time, reduced the plasmatic levels of total, free, and ester cholesterol (−15, −12, and −17%, respectively), giving rise to a redistribution of the lipidic components of LDL (greater phospholipid and cholesterol amounts) with an indirect effect on their dimesions (bigger by about 12%).  相似文献

Olive fruits and virgin olive oils from a super‐high‐density olive orchard cv. Arbequina under different irrigation treatments (Control, RDI1, RDI2) were characterized by their pigment profile, CIElab colorimetric coordinates, oxidative stability and antioxidant capacity during the olive seasons 2011 and 2012. The control treatment were irrigated according to the total evapotranspiration of the olive grove (100% ETc), while RDI1 and RDI2 consisted of moderate and severe deficit irrigation, respectively. Olive fruits from severe water deficit irrigation treatment (RDI2) showed a significantly greater concentration of pigments compared to the others. In addition, the ratios a*/chlorophylls and chlorophylls/carotenoids indicated a delayed maturation in these olives. Irrigation treatments significantly affected the yellow component (b*) of the virgin olive oils contained. A reduction of 75% in the water applied (RDI2) produced a strong increase in lutein‐zeaxanthin content (38%) and chlorophyll a (50%), oxidative stability, that ranged from 8.37 h (Control) to 13.23 h (RDI2) and antioxidant activity compared to Control. However, the oil production from RDI2 decreased approximately a 49%.  相似文献

The aim of the present work was to investigate the influence of fruit ripening on oil quality and volatile compounds in an attempt to establish an optimum harvesting time for Oueslati olives, the minor olive variety cultivated in Tunisia. Our results showed that many analytical parameters, i.e., peroxide value, UV absorbance at 232–270 nm, chlorophyll pigments, carotenoids and oleic acid contents decreased during ripening, whilst linolenic acid increased. Free acidity remained practically stable with a very slight rise at the highest maturity index. The volatile compounds emitted by the Oueslati olive oil were characterized and quantified by HS‐SPME‐GC‐EIMS. Twenty‐three volatile compounds were identified, mainly aldehydes, sesquiterpenes and esters. The results show variations in the volatile fractions and quality parameters of Oueslati extra virgin olive oil obtained at different olive‐ripening stages. Fifteen sesquiterpenes were identified for the first time in this cultivar, mainly hydrocarbon derivatives, but also oxygenated ones. On the basis of the quality parameters and volatile fractions studied, the best stage of Oueslati olive fruits for oil processing seems to be at ripeness index about 3.0. Indeed, these results suggested the possibility of using sesquiterpenes for olive authenticity and traceability and demonstrated that the volatile fractions can be used as indicators of the degree of ripening of the olives used to obtain the corresponding virgin olive oils.  相似文献

Phenolic compounds have a high importance in olive oil because of their effect on shelf life and sensory properties. This study reports on the HPLC profiles of the phenolic compounds of virgin olive oils obtained from Arbequina olives from the harvesting in a super‐intensive orchard under a linear irrigation system. In addition, phenolic content, carotenoid and chlorophyllic pigments, and oxidative stability were analyzed. Total phenol content and 3,4‐DHPEA‐EDA increased up to a maximum throughout the ripening process. The simple phenols tyrosol and hydroxytyrosol acetate increased throughout the ripening process, however, there was not found a clear trend in hydroxytyrosol content. Minor constituents such as vanillic acid and p‐coumaric acid increased up to a maximum and then decreased, since vanillin decreased progressively throughout the time of harvest. 3,4‐DHPEA‐EDA and lignans were present in considerable amounts in the studied samples, while oleuropein aglycone was present in a low amount. Total phenol content and oil stability followed the same trend throughout the study, so a very good correlation was established between them. Total secoiridoids and, specifically, 3,4‐DHPEA‐EDA seemed to be responsible for oil stability. The pigment content decreased during ripening, and not a positive correlation was found between pigments and oil stability. Practical applications : The results can be used to determine the best time for harvesting in order to obtain olive oils with different phenols and pigment contents. This is important for sensory characteristics of the olive oils and also for olive oil stability.  相似文献

Fatty acid composition, peroxide value, acid value, iodine value, saponification number, unsaponifiable matter content, total tocopherols and phenolics contents, and wax content of Bene hull oil (BHO) were determined and compared to those of Bene kernel oil (BKO) and extra‐virgin olive oil (EVOLO). Considering the fatty acid composition and total tocopherols and phenolics contents, the resistance to the production of conjugated diene hydroperoxides and carbonyl compounds during the heating process at 170 °C for BHO was about 4.2 and 7.3 times and about 1.7 and 2.0 times those of BKO and EVOLO, respectively. The antioxidant activity of BHO was exactly the same as that of tert‐butylhydroquinone at low concentrations (100 ppm).  相似文献

General objective of the present work was to assess the isolation of polyphenols from olive leaves. The effects of extraction conditions on the total phenol content and antioxidant activity of olive leaf extract (OLE) were investigated. An extract with good antioxidant activity (7.52 mmol of Trolox equivalent antioxidant capacity (TEAC)/g olive leaf extract), and a high content of oleuropein (13.4%) and rutin (0.18%) could be obtained using 70% ethanol as extraction solvent. There was a good correlation between the antioxidant activity and the total phenol content. Furthermore, silk fibroin was used as a novel adsorbent to recover the polyphenols from the olive leaf extracts. The adsorbed amounts of rutin and oleuropein were 15 mg rutin/g silk fibroin and 96 mg oleuropein/g silk fibroin. Fraction consisting of mainly oleuropein and fraction rich in rutin, luteolin-7-glucoside, verbascoside, apigenin-7-glucoside were obtained by using silk fibroin filled column. Silk fibroin was found to be a promising adsorbent for the purification of oleuropein and rutin from olive leaf extracts.  相似文献

The use of the products derived from the olive tree on human health dates back centuries. In several civilizations, the olive tree had and still has a very strong cultural and religious symbolism. Notably, the official seal and emblem of the World Health Organization features the rod of Asclepius over a world map surrounded by olive tree branches, chosen as a symbol of peace and health. Recently, accumulating experimental, clinical and epidemiological data have provided support to the traditional beliefs of the beneficial effect provided by olive derivates. In particular, the polyphenols present in olive leaves, olives, virgin (unrefined) olive oil and olive mill waste are potent antioxidant and radical scavengers with anti-tumor and anti-inflammatory properties. Here, we review the positive impact on human health of oleuropein, the most prevalent polyphenol present in olives. In addition, we provide data collected in our laboratory on the role of oleuropein in counteracting lipid accumulation in a mouse model of non-alcoholic fatty liver disease.  相似文献

Commercially available sunflower oil was enriched in polyphenols by adding olive leaf extract. After extracting the dried and ground olive leaves with the assistance of homogenizer, total phenolic content (TPC) and oleuropein concentration of the extract were determined. The dried extract was partially dissolved into the sunflower oil to increase the quality and shelf-life of the oil enriched by the substances in the plants by means of solid-liquid extraction method. A face central composite design (FCCD) through response surface methodology (RSM) was used to investigate the effects of enrichment conditions (extract content, time and mixing speed) on the responses, TPC and oleuropein concentration of the enriched sunflower oil as well as to design of experiments, to model and to optimize the process. The enriched sunflower oil obtained at optimum conditions was evaluated in terms of its TPC, oleuropein, total carotenoid content (TCC), antioxidant activity (AA), peroxide value (PV) and induction time (IT), depending on those of the crude oil. Furthermore, artificial neural networks (ANN) were also employed to compare the predicted results of RSM.  相似文献

Phenolic compounds are useful markers to control olive oil technological processes, including the virgin olive oil (VOO)/water separation after olive oil extraction. In this investigation, VOO extracted from olives of cv. Coratina using a mild oil/water separator called the hydrocyclone sedimentation system (Hydroil) was compared with VOO obtained using a conventional vertical centrifuge separator (Cenoil), which is mostly used in the modern olive oil industry. Secoiridoid aglycones were selected, among phenolic compounds, as markers and analyzed using reversed‐phase liquid chromatography coupled to linear quadrupole ion‐trap mass spectrometry with electrospray ionization in the negative mode. VOO samples obtained using the Hydroil system were found to contain significantly higher levels of secoiridoid aglycones, compared to the Cenoyl‐type samples. In particular, the total content of the aglycones of decarboxymethyl oleuropein, decarboxymethyl ligstroside, ligstroside, and oleuropein, expressed in terms of oleuropein, was estimated as 35.40 ± 0.80 mg kg?1, compared to 8.06 ± 0.41 mg kg?1 in the Cenoil samples (n = 3). Since no significant difference in residual water (P < 0.05) was found between the two types of VOO samples, the higher amount of secoiridoids obtained for Hydroil‐type ones was explained by the lower extent of oxidation occurring during the mild oil/water separation achieved using the Hydroil system.  相似文献
