BACKGROUND: Sugar content is one of the main characteristics related to the quality of fruit. Research confirms that nitric oxide (NO) involves a physiological process and prolongs the storage life of fruit. However, little attention has been paid to the effects of NO on sugar metabolism in fruit during storage. In this study, the effect of different concentrations (0, 10, 30 µmol L?1) of exogenous NO treatment on sugar content and related enzyme activities in ‘Feicheng’ peach fruit was investigated during storage (0–12 days after harvest) at room temperature (25 °C). RESULTS: Results showed that the decrease of firmness and accumulation of sugar and acid:sugar ratio in peach fruit during storage were significantly inhibited by treatment with 10 µmol L?1 NO. Treatment with 10 µmol L?1 NO could promote fructose and glucose metabolism during the first 4 days of storage, and increase the content of sucrose and the activities of sorbitol dehydrogenase, sorbitol oxidase and sucrose phosphate synthase in peach fruit during storage. However, acid invertase activity from 8 to 12 days of storage and neutral invertase activity during the first 4 days of storage were inhibited by treatment with 10 µmol L?1 NO. At the same time, treatment with 10 µmol L?1 NO inhibited sucrose synthase (SS) activity in decomposition during storage and SS activity in synthesis from 8 to 12 days of storage. CONCLUSION: Treatment with 10 µmol L?1 NO had a significant impact on content of soluble sugars and related enzyme activities in ‘Feicheng’ peach fruit during storage (0–12 days) at room temperature (25 °C). The effects of fumigating with 0, 10, 20, 30 μL L?1 nitric oxide (NO) gas and dipping in 0.5, 1.0 and 2.0 μmol L?1 NO aqueous solution on quality of kiwifruit (Actinidia Chinensis Planch cv Xuxiang) during storage at 20 °C were evaluated. It was found that fumigating with 20 μL L?1 NO gas and dipping in 1.0 μmol L?1 NO aqueous solution delayed firmness lost and increased SSC/TA ratios of kiwifruits. In comparison with the kiwifruits fumigated with 20 μL L?1 NO, the kiwifruits dipped in 1.0 μmol L?1 NO solution had slower ethylene production, lower contents of soluble solids and malondialdehyde (MDA), higher contents of vitamin C and E, but no significant difference existed in chlorophyll contents. The results suggested that dipping kiwifruits in 1.0 μmol L?1 NO aqueous solution was more effective in maintaining kiwifruits quality during 20 °C storage.  相似文献

BACKGROUND: The time course of polyphenol oxidase (PPO) activity in the leaves of two olive cultivars (Picual and FS‐17) irrigated with nutrient solutions differing in Mn concentration (0, 2 and 1280 µmol L?1) was studied under hydroponic conditions to determine whether PPO activity could be used as an early criterion of Mn status of olive plants, and to elucidate whether genotypic differences exist between the two olive cultivars studied, concerning the effect of Mn concentration on PPO activity. RESULTS: In all the Mn treatments, PPO activity was greater in Picual than in FS‐17. Under excess Mn (1280 µmol L?1), PPO activity gradually increased with time, starting from day 30 of the experiment in both cultivars, and this increase preceded the appearance of Mn toxicity symptoms. In contrast, in the other two Mn treatments (0 and 2 µmol L?1) PPO activity increased and afterwards decreased during the experiment, but the trend was not clear. In the 1280 µmol L?1 treatment, PPO activity linearly increased (R = 0.8836 for Picual and 0.943 for FS‐17) with the increase of Mn concentration in the leaves of both cultivars. In the 1280 µmol L?1 Mn treatment, PPO activity was negatively related with Fe and Zn concentrations in the leaves, and positively in the 0 and 2 µmol L?1 Mn treatments with the Ca, Mg and K concentrations. CONCLUSION: From the differential time course of PPO activity in the three Mn treatments (0, 2 and 1280 µmol L?1), it is concluded that periodic measurements of PPO activity in the leaves of the olive cultivars Picual and FS‐17 can be used for the early detection of Mn toxicity (before the appearance of symptoms). BACKGROUND: The effect of diet garlic supplementation on total antioxidant status (TAS), nitric oxide (NO) and routine biochemical and haematological parameters was investigated in blood of rats. A total of 30 male rats were divided equally into two groups. Each of 15 rats of treatment group was fed 600 mg kg?1 garlic solution in distilled water by gavage and controls only received distilled water. After garlic consumption for 1 month, blood serum total antioxidant, nitrate and some biochemical and haematological tests including serum lipids parameters, blood sugar, complete blood count (CBC), and haemoglobin were measured. RESULTS: The garlic treatment group showed significantly increase in the mean level of TAS from 0.77 ± 0.10 mol L?1 to 1.18 ± 0.11 mol L?1 (P < 0.01) and nitrate (a NO metabolite) from 0.78 ± 0.06 µmol L?1 to 1.44 ± 0.27 µmol L?1 (P < 0.05) in the blood sera of rats compared with the controls. There were no significant differences between the routine biochemical and haematological parameters. CONCLUSION: Garlic consumption should have antioxidant properties and may not affect the lipids profile and total blood cell counts. Copyright © 2009 Society of Chemical Industry  相似文献

BACKGROUND: Large amounts of citrus by‐products are released from juice‐processing plants every year. Most bioactive compounds are found in the peel and inner white pulp. Flavonoids are a widely distributed group of bioactive compounds. The methanolic extract of citrus peel powder has been shown to possess strong antioxidant activity. Therefore the aim of this study was to isolate the major antioxidant flavonoid compound from Citrus unshiu (satsuma) peel as citrus by‐product and evaluate its antioxidant activity. RESULTS: The major flavonoid isolated from C. unshiu peel was identified as quercetagetin. The structure of the compound was determined by tandem mass spectrometry and ultraviolet spectroscopy. Its antioxidant activity was assessed by assays of 2,2‐diphenyl‐1‐picrylhydrazyl (DPPH) radical, hydroxyl radical and intracellular reactive oxygen species (ROS) scavenging and DNA damage inhibition. Quercetagetin showed strong DPPH radical‐scavenging activity (IC507.89 µmol L?1) but much lower hydroxyl radical‐scavenging activity (IC50203.82 µmol L?1). Furthermore, it significantly reduced ROS in Vero cells and showed a strong protective effect against hydrogen peroxide‐induced DNA damage. This study evaluated the efficacy of ultrasound treatment and nano-zinc oxide (ZnO) coating individually and in combination in preserving the quality of fresh-cut kiwifruit. The nano-ZnO coating solution was prepared by mixing the ZnO nanoparticles in premixed chitosan–acetic acid solution. The fresh-cut kiwifruit were dipped in NaClO solution (50 μL?L?1 sodium, control), subjected to ultrasound treatment (40 KHz, 350 W, 10 min), or coated with nano-ZnO solution. The fresh-cut kiwifruit samples were also subjected to combined ultrasound treatment and nano-ZnO coating. All of these test samples were stored at 4 °C for 10 days. The effects of these treatments on the quality parameters such as the production of carbon dioxide and ethylene, mass loss, and flesh firmness were investigated. At the end of storage, the combination treatments with ultrasound treatment at 40 KHz with 1.2 g?L?1 nano-ZnO coating showed lower production of ethylene (1.86 μL?kg?1?h?1) and carbon dioxide (10.01 mg?kg?1?h?1), water loss (0.46 %), and texture (7.87 N). Hence, it was concluded that the combination of ultrasound treatment with nano-ZnO coating is a promising approach to extend the shelf-life of fresh-cut kiwifruit.  相似文献

BACKGROUND: Grapes and red wines are rich sources of phenolic compounds such as anthocyanins, catechins, flavonols and stilbenes, most of which are potent antioxidants showing cardioprotective properties. We first isolated scirpusin A, a hydroxystilbene dimer, from a wine grape of Xinjiang, and studied its antioxidant activity. RESULTS: Reactive oxygen species scavenging effects and the protection against reactive singlet oxygen‐induced DNA damage of scirpusin A have been investigated in our experiments. The concentration of scirpusin A required to inhibit 50% of 1O2 generation was 17 µmol L?1, while addition of scirpusin A at 140 µmol L?1 caused complete inhibition. Further kinetic study revealed that the reaction of Scirpusin A with singlet oxygen has an extremely high rate constant (ka = 4.68 × 109 L mol?1 s?1). Scirpusin A (140 µmol L?1) exhibited significant inhibition effects on pBR322 DNA breakage. However, scavenging effects of scirpusin A on superoxide anion O2?? and hydroxyl radical ·OH were not potent as the inhibitor rates at a concentration of 1400 µmol L?1 were 28.83% and 19.5%, respectively. CONCLUSION: The present study shows that scirpusin A is a selective quencher of singlet oxygen and a protector against reactive singlet oxygen‐induced pBR322 DNA damage at very low concentrations. BACKGROUND: A voltammetric study of vitamin E (DL‐ α‐tocopherol) detection using square wave stripping and cyclic voltammetry is discussed in this paper. The working sensor was made by mixing carbon nanotube powder with DNA (double‐stranded calf thymus DNA) and mineral oil. In this electrode, the anodic peak was obtained for ? 0.6 V in a 0.1 mol L?1 phosphate electrolyte solution. RESULTS: Under optimized stripping conditions, analytical linear working ranges of 0.5–4.0 µg L?1 and 40.0–160.0 µg L?1 were obtained. The RSD precision was pegged at 0.105% with seven points using an 80 µg L?1 spike. The detection limit (S/N) was found to be 0.056 µg L?1 (1.30 × 10?10 mol L?1). CONCLUSION: The developed method was found to be applicable to quality control analysis in the food, pharmaceutical and other manufacturing sectors. BACKGROUND: Horseradish plants (Armoracia rusticana) contain high concentrations of glucosinolates. Former studies have revealed that Armoracia plants cultivated in vitro have markedly lower glucosinolate concentrations than those grown in soils. Yet, these studies neglected that the sulfate concentration in the growth medium may have had a strong impact on glucosinolate metabolism. Accordingly, in this study horseradish in vitro plants were cultivated with differing sulfate concentrations and the glucosinolate concentrations were quantified by ion pair HPLC. RESULTS: Cultivation in 1.7 mmol L?1 sulfate (as used in the prior studies) resulted in the accumulation of 16.2 µmol g?1 DW glucosinolates, while the glucosinolate concentration increased to more than 23 µmol g?1 DW when 23.5 mmol L?1 sulfate was used in the medium. Correspondingly, the glucosinolate concentration decreased to 1.6 µmol g?1 DW when sulfate concentration was lowered to 0.2 mmol L?1. CONCLUSION: Since the glucosinolate accumulation in relation to the sulfate concentration follows a typical saturation curve, we deduce that the availability of sulfate determines the glucosinolate concentration in horseradish in vitro plants. © 2012 Society of Chemical Industry  相似文献

An inhibitory compound acting against rat platelet 12‐lipoxygenase was isolated from the peel of Lumie fruit (Citrus lumia) by activity‐guided separation. It was identified as eriocitrin (eriodictyol 7‐O‐rutinoside) by spectroscopic analyses. Eriocitrin inhibited 5‐lipoxygenase (IC5029.1 µmol L?1) from rat peritoneal polymorphonuclear leukocytes in addition to 12‐lipoxygenase (IC5022.3 µmol L?1). Its aglycone, eriodictyol (5,7,3′, 4′‐tetrahydroxyflavanone), was a much more potent inhibitor of both 12‐lipoxygenase (IC500.07 µmol L?1) and 5‐lipoxygenase (IC500.20 µmol L?1). It also inhibited the production of leukotriene B4 in intact peritoneal polymorphonuclear leukocytes stimulated with calcium ionophore A23187 (IC5012.7 µmol L?1). The distribution of eriocitrin in 39 citrus fruits was investigated by high‐performance liquid chromatography analysis. Lumie, eureka lemon (Citrus limon), Sambokan (Citrus sulcata), Sudachi (Citrus sudachi) and Koji (Citrus leiocarpa) fruits were found to contain high levels of eriocitrin in both peel and juice vesicles. BACKGROUND: Little is known about the relation between haemoglobin (Hb)‐mediated lipid and protein oxidation in muscle foods and how these two reactions can be inhibited by naturally occurring antioxidants. This study was aimed at evaluating (1) lipid oxidation and protein oxidation induced by 20 µmol L?1 Hb during chilled and frozen storage of washed cod mince and (2) the efficiency of 10–1000 ppm (0.063–6.3 mmol L?1) caffeic acid in preventing these reactions. RESULTS: Addition of 20 µmol L?1 Hb increased peroxide value (PV), rancid odour, protein carbonylation, protein insolubilisation, redness loss and α‐tocopherol loss in ice‐stored washed cod mince. Since both lipid and protein oxidation developed at the same time, it was not possible to conclude which reaction initiated the other. All studied reactions were efficiently inhibited by ≥ 50 ppm caffeic acid, which could be a result of α‐tocopherol regeneration, general radical scavenging, reduced formation of oxidised Hb forms and/or conformational changes in Hb structure. During frozen storage the only clear effect of Hb was increased PV, and here caffeic acid was less efficient as an antioxidant. CONCLUSION: Hb‐induced lipid and protein oxidation occurred quickly in ice‐stored washed cod mince, and the two reactions could not be separated in time. BACKGROUND: Higher plants possess several mechanisms of defense against plant pathogens. Proteins actively synthesized in response to those stresses are called defense‐related proteins which, among others, include certain protease inhibitors. It is of particular relevance to investigate plant natural defense mechanisms for pathogen control which include cystatins—specific inhibitors of cysteine proteases. RESULTS: In this study, a cysteine proteinase inhibitor (CPI), 11 kDa in size, was purified from green kiwifruit to homogeneity. Immuno‐tissue print results indicated that CPI is most abundant in the outer layer of pericarp, near the peel, and the inner most part of the pulp—sites where it could act as a natural barrier against pathogens entering the fruit. The purified protein (15 µmol L?1) showed antifungal activity against two phytopathogenic fungi (Alternaria radicina and Botrytis cinerea) by inhibiting fungal spore germination. In vivo, CPI (10 µmol L?1) was able to prevent artificial infection of apple and carrot with spore suspension of B. cinerea and A. radicina, respectively. It also exerted activity on both intracellular and fermentation fluid proteinases. CONCLUSION: Identification and characterization of plant defense molecules is the first step towards creation of improved methods for pathogen control based on naturally occurring molecules. BACKGROUND: In China alone, more than 400 million pigs are slaughtered each year to provide meat. Porcine blood is rich in proteins but is usually discarded, which can cause environmental contamination. Recovering porcine blood and converting it to high‐value products is therefore economically and environmentally desirable. However, very little information on antioxidant peptides from porcine blood by‐products is currently available. In this study the antioxidant properties of porcine plasma hydrolysates PPE and PPA prepared with pepsin and papain respectively were investigated. RESULTS: Both PPE and PPA showed excellent antioxidant activity in a linoleic acid system (AL) compared with α‐tocopherol (VE) at the same concentration (P < 0.01). Their activities were respectively 3.33 and 1.83 times stronger than that of VE at a concentration of 10 µg mL?1 and 5.4 and 5.6 times stronger at 100 µg mL?1. The 2,2‐diphenyl‐1‐picrylhydrazyl (DPPH) radical‐scavenging activity (DRSA) reached 48.4 and 43.1% for PPE and PPA respectively at 500 µg mL?1. The ferrous ion‐chelating power (FICP) of PPE at 100 µg mL?1 was about 1.5 times stronger than that of 10 µmol L?1 ethylene diamine tetraacetic acid (EDTA) in a 50 µmol L?1 Fe2+ system, whereas the FICP of PPA at 100 µg mL?1 was 61% that of 10 µmol L?1 EDTA. Furthermore, PPE was separated on Resource 15RPC and Superdex peptide 10/300GL columns, and the antioxidant activity of the peptides and its relationship to their polarity and molecular weight (MW) were analysed. The hydrolysate was divided into four groups (R1–R4) with hydrophobicities ranging from weak to strong by Resource 15RPC, while it was divided into three groups (S1, MW 7–12 kDa; S2, MW 3–7 kDa; S3, MW 1–3 kDa) by Superdex peptide 10/300GL. CONCLUSION: The results showed that AL was significantly and positively correlated with the relative amounts of R1, S2 and S3 and that DRSA was dependent on R3 and S1. BACKGOUND: Broccoli deteriorates very quickly after harvest at ambient temperature due to the loss of green colour and the consequent yellowing of florets. To search for an effective method to control quality deterioration, the effect of 1‐methylcyclopropene (1‐MCP) combined with 6‐benzylaminopurine (6‐BA) treatment on visual quality, antioxidant enzymes and bioactive compounds in broccoli florets were investigated. RESULTS: A combined treatment of 2.5 µL L?1 1‐MCP and 200 mg L?1 6‐BA significantly reduced the increase of lightness (L*) value, and retained a high level for the hue value (H) and chlorophyll content. Superoxide dismutase, ascobate peroxidase and catalase activities increased while the activity of peroxidase decreased during storage in treated samples in comparison with the controls. The combined treatment enhanced the biosynthesis of glucosinolate and the formation of the anticarcinogen sulforaphane, which improved the health benefit of broccoli. CONCLUSION: These results indicate that a combined treatment of 1‐MCP and 6‐BA could be a good candidate for maintaining the visual quality and enhancing the nutritional value in broccoli during storage at 15 °C. © 2012 Society of Chemical Industry  相似文献

BACKGROUND: ‘Hami’ melon is susceptible to softening during postharvest storage at ambient temperature, which enhances postharvest deterioration and limits transportation and storage. To look for a method of softening control, the effect of 1‐methylcyclopropene (1‐MCP) on regulating postharvest softening of ‘Hami’ melon fruit was investigated. RESULTS: 1‐MCP treatment at 1 µL L?1 significantly reduced ethylene production and maintained higher levels of fruit firmness. It also markedly inhibited the accumulation of 1‐aminocyclopropane‐1‐carboxylic acid (ACC) and maintained lower activities of ACC synthase and ACC oxidase. In addition, 1‐MCP treatment reduced the activities of fruit‐softening enzymes such as pectin methyl esterase, polygalacturonase, endo‐1,4‐β‐glucanase and β‐galactosidase. CONCLUSION: 1‐MCP was effective in suppressing ethylene production and delaying fruit softening in ‘Hami’ melon by decreasing the activities of ethylene biosynthesis enzymes and inhibiting fruit‐softening enzymes. BACKGROUND: In an effort to develop alternative therapeutic agents, strong inhibitory activity against α‐glucosidase and α‐amylase was detected in Eisenia bicyclis methanolic extract. RESULTS: In this study, two phlorotannins were isolated from E. bicyclis and characterised by chromatography and nuclear magnetic resonance. The active substances were identified as fucofuroeckol A (FF) and dioxinodehydroeckol (DD). To the authors' knowledge, this is the first report of the identification of these substances in E. bicyclis. However, to date, no antidiabetic activity of FF and DD has been reported. Both phlorotannins demonstrated significant inhibitory activity against α‐glucosidase and α‐amylase. FF showed potent antidiabetic activity, with IC50 values of 131.34 nmol L?1 against α‐glucosidase and 42.91 µmol L?1 against α‐amylase. The corresponding IC50 values of DD were 93.33 nmol L?1 and 472.7 µmol L?1. Furthermore, kinetic analysis revealed that FF and DD exhibited non‐competitive inhibitory activity against α‐glucosidase. CONCLUSION: These results suggest that FF and DD may be candidates for the development of an antidiabetic pharmaceutical agent or food additive. BACKGROUND: The presence of phenolics in fruit, red wine and vinegar has positive health effects due to their significant antioxidant activity. The aim of the study was to determine the effects of two different vinegar production methods on antioxidant activity and phenolic level of vinegars derived from Ulugbey Karasi grapes. Traditional surface and industrial submerge methods were used to make vinegar. Samples were taken from fresh red grape juice, maceration, wine, traditional vinegar and industrial vinegar. RESULTS: Total phenolic content of traditional and industrial vinegar samples were 2690 mg L?1 and 2461 mg L?1 GAE, respectively. ORAC values of traditional and industrial vinegar samples were 10.50 µmol mL?1and 8.84 µmol mL?1 TE, respectively. Antioxidant activity values of traditional and industrial vinegars were 13.50 mmol L?1 and 10.37 mmol L?1 TEAC, respectively. Gallic acid, catechin, epicatechin, chlorogenic acid, caffeic acid, syringic acid, p‐coumaric acid and ferulic acid were detected in grape juice, wine and vinegar samples. The content of catechin in industrial vinegar (27.50 mg L?1) was significantly higher than that of in traditional vinegar (13.76 mg L?1) (P < 0.05). Traditional vinegar had higher amounts of chlorogenic and syringic acids than the industrial vinegar (P > 0.05). BACKGROUND: Low‐temperature breakdown (LTB), a disorder inducing quality loss, during and after cold storage of ‘Hayward’ kiwifruit was investigated. Harvested kiwifruits during fruit maturation or after delayed storage (DS) at 20 °C for 0, 1, 2, 3 and 4 weeks and 1 µL L?1 ethylene treatment for 24 h were stored at ? 0.5 °C for 24 weeks and additional ripening at 20 °C for 5 days. Fruit quality indices and LTB incidence and severity were determined before and after treatments. RESULTS: Harvested fruits ripened during maturation, DS and after ethylene treatment. After storage and shelf life, fruits of all treatments were at complete ripening stage. LTB incidence of early harvested fruits was high, while that of fruits of the mid (third) and late harvests was low. Fruits of the third harvest date showed progressively increased LTB incidence with increasing duration of DS to as high as 95–100% after 4 weeks. Ethylene‐treated fruits showed a comparable increase in LTB to that corresponding to 2–3 weeks of DS. CONCLUSION: In contrast to fruit maturation, postharvest (after harvest and before storage) DS at non‐chilling temperature and ethylene treatment advanced the ripening of ‘Hayward’ kiwifruit and resulted in increased LTB incidence. BACKGROUND: Fungal contamination and Callosobruchus infestation results in qualitative and quantitative losses of chickpea seeds during storage. Most of the synthetic chemicals used as preservatives have adverse effects. Therefore, the antifungal and insecticidal potential of Mentha arvensis essential oil was evaluated to determine whether this could be an eco‐friendly substitute of synthetic preservatives. RESULTS: The stored chickpea seeds were dominated by Aspergillus flavus (46.1%) and 30% isolates among them were found toxigenic. The MIC of Mentha oil against A. flavus was recorded at 400 µL L?1 and it exhibited broad fungitoxic activity against 14 storage fungi. The oil was found superior to some prevalent synthetic fungicides. Mentha oil showed potent insecticidal activity against Callosobruchus chinensis at different concentrations and exposure times. The oviposition by C. chinensis was completely checked at 10 µL L?1 while F1 emergence was completely inhibited at 200 µL L?1. During in situ experiments, 94.05% protection of the chickpea from C. chinensis by Mentha oil showed superiority over the organophosphate insecticide malathion, where 90.75% protection was recorded. CONCLUSION: The Mentha EO showing potent fungitoxic and insecticidal efficacy and may be recommended as a plant‐based preservative in the management of fungal and insect infestation of chickpea and other pulses during storage. BACKGROUND: Pungency and perceived sweetness are important quality assurance parameters for onion producers; however, there is still a paucity of information describing the spatial changes within the bulb of some taste‐related compounds during storage. Accordingly, onion bulbs of cultivars (cvs.), SS1 (low pungency, low dry matter) and Renate (pungent, high dry matter) bulbs, were divided into 12 sections and both spatial and temporal variations of pyruvate and glucose were recorded during standard controlled atmosphere storage. RESULTS: Pyruvate within cv. SS1 bulbs reached 8.3 µmol g?1 fresh weight (FW) from pre‐storage levels of less than 3.4 µmol g?1 FW, whilst pyruvate changes in cv. Renate remained in the ‘pungent’ category, between 12 µmol g?1 FW and 15 µmol g?1 FW; thus having fewer implications for marketing and quality assurance. Glucose concentrations were significantly higher for both cvs. in the neck region as compared to basal tissue. Higher glucose levels were recorded in the core of the bulb than in outer scales of cv. Renate. The opposite was found for cv. SS1. There was an inverse correlation of ? 0.69 and ? 0.87 between glucose and pyruvate for cvs. SS1 and Renate, respectively. CONCLUSION: In order to measure overall bulb flavour, it is recommended that sampling procedures, particularly for routine pungency determination, be carried out on bulb quarters or wedges rather than equatorial sections.
