Controversy has existed as to whether monensin will provide equal or differential benefits in a higher-energy, lower-roughage close-up diet versus a higher-roughage, lower-energy diet. Our objective was to determine the rumen effects of a controlled-energy, high-fiber diet balanced to meet but not greatly exceed energy requirements during the dry period or a traditional 2-group approach of higher-energy close-up diet. The effects of added monensin in each diet type were determined. Multiparous Holstein cows (n = 17) were fitted surgically with ruminal cannulas. During the first 4 wk of the dry period, all cows were fed a controlled-energy, high-fiber diet (CE) as a total mixed ration for ad libitum intake. During the last 3 wk before calving, half of the cows were switched to a higher-energy, close-up diet until calving (CU), whereas the other half continued to receive the CE diet. Within each dietary group, half of the cows received monensin (MON) supplementation in the diet (24.2 g/t of total dry matter) and half did not (CON). After calving, all cows received the same lactation diet containing monensin (15.4 g/t of dietary dry matter). At 14 d prepartum, dry matter intake was not different across treatments. The weight of rumen contents was greater for cows fed CE. Rumen liquid dilution rate, solids passage rate, pH, total volatile fatty acid (VFA) concentrations, molar proportions of acetate and propionate, and papillae length did not differ among diets. Butyrate percentage tended to be greater for cows fed CE. Postpartum, dry matter intake, mass of rumen contents, solids passage rate, pH, total VFA concentration, molar percentages of propionate and butyrate, and papillae length did not differ among treatments. Liquid dilution rate (16.6, 10.7, 16.0, and 18.2%/h for CE + CON, CE + MON, CU + CON, and CU + MON, respectively) was affected by a diet × monensin interaction. Cows on the CE + CON diet had a greater ruminal proportion of acetate than did cows fed CU + CON, whereas cows fed monensin on either diet were intermediate (diet × monensin interaction). Addition of MON to the CU diet decreased the proportion of propionate (diet × monensin interaction). Cows fed CE had greater mass of rumen contents before parturtition but the high inclusion of wheat straw in the CE diet did not negatively affect rumen papillae length. Monensin inclusion differentially affected liquid passage rate and VFA concentrations.  相似文献

This experiment was conducted to compare conventional (CON; 21 d) and shortened (SH; 10 d) close-up period, and evaluate the effect of shortened close-up period combined with feeding different metabolizable protein (MP) levels on dry matter (DM) intake, metabolic status, and performance of dairy cows. Forty-eight multiparous Holstein cows with similar parity, body weight (BW), and previous lactation milk yield were divided into 2 groups. The first group (n = 24) received the far-off diet from ?60 to ?21 d (CON), and the second group (n = 24) received same far-off diet from ?60 to ?10 d (SH) relative to expected parturition. Cows were then moved to individual stalls and randomly allocated to 1 of 3 close-up diets: low MP diet (LMP; MP = 79 g/kg of DM), medium MP diet (MMP; MP = 101 g/kg of DM), or high MP diet (HMP; MP = 118 g/kg of DM). Treatments were used in a 2 × 3 factorial arrangement with 2 lengths of close-up period (CON and SH) and 3 levels of MP (LMP, MMP, and HMP). All diets were fed for ad libitum intake during the close-up period. After calving, all cows received the same fresh cow diet. We found no interaction between close-up period length and MP levels for traits, except for postpartum serum fatty acids and β-hydroxybutyrate (BHB). The concentrations of postpartum serum fatty acids and BHB were higher on LMP than MMP and HMP diets in SH group. The cows of the SH group tended to produce less colostrum in the first milking than cows in CON group. The length of close-up period did not affect pre- and postpartum DM intake or energy balance of cows during the last week of prepartum, but cows of the CON group had greater BW changes during the last 3 wk before parturition than cows in SH group. Cows fed MMP and HMP diets consumed 1.2 and 1 kg more DM than for those fed LMP prepartum, respectively. The concentrations of prepartum BHB and Ca were higher for SH cows than CON group cows. Except for blood urea N concentration, no other blood metabolite in prepartum was affected by dietary MP. We found no effects of close-up period length or MP levels in the close-up diet on urinary pH, purine derivative excretion, and microbial N flow. Postpartum, milk yield was not affected by close-up period length, but cows in CON group tended to have higher 4% fat-corrected milk yield, had higher milk fat content and yield, had greater BW and body condition score loss, and higher energy negative balance than cows in the SH group. Cows fed MMP diet ate 1.8 kg more DM and yielded 3.37 kg more milk than those fed the LMP diet. Milk fat, protein, and lactose content, milk urea N, and somatic cell count were not affected by MP levels, but the yield of milk protein and lactose were higher on MMP diet than on LMP diet. Concentrations of postpartum serum fatty acids and BHB were decreased by shortening the close-up period length, but glucose, cholesterol, and triglyceride were similar between close-up groups. During the postpartum period, serum fatty acids, BHB, aminotransferase, and Ca concentrations were decreased by increasing the MP levels in the close-up diet. It appears from this data set that multiparous cows will benefit from a shortened close-up period, and feeding a moderate MP diet could improve DM intake, milk yield, and metabolic status of periparturient dairy cows.  相似文献

Dairy cows commonly undergo negative Ca balance accompanied by hypocalcemia after parturition. A negative dietary cation-anion difference (DCAD) strategy has been used prepartum to improve periparturient Ca homeostasis. Our objective was to determine the influence of a negative DCAD diet with different amounts of dietary Ca on the blood acid-base balance, blood gases, and metabolic adaptation to lactation. Multiparous Holstein cows (n = 81) were blocked into 1 of 3 dietary treatments from 252 d of gestation until parturition: (1) positive DCAD diet and low Ca (CON; containing +6.0 mEq/100 g DM, 0.4% DM Ca); (2) negative DCAD diet and low Ca (ND; ?24.0 mEq/100 g DM, 0.4% DM Ca); or (3) negative DCAD diet plus high Ca supplementation (NDCA; ?24.1 mEq/100 g DM, 2.0% DM Ca). There were 28, 27, and 26 cows for CON, ND, and NDCA, respectively. Whole blood was sampled at 0, 24, 48, and 96 h after calving for immediate determination of blood acid-base status and blood gases. Serum samples collected at ?21, ?14, ?7, ?4, ?2, ?1, at calving, 1, 2, 4, 7, 14, 21, and 28 d relative to parturition were analyzed for metabolic components. Results indicated that cows fed ND or NDCA had lower blood pH at calving but greater pH at 24 h after calving compared with CON. Blood bicarbonate, base excess, and total CO2 (tCO2) concentrations of cows in ND and NDCA groups were less than those of cows in CON at calving but became greater from 24 to 96 h postpartum. The NDCA cows had lower blood bicarbonate, base excess, and tCO2 at 48 h and greater partial pressure of oxygen after calving compared with ND. Cows fed ND or NDCA diets had lower serum glucose concentrations than CON cows before calving but no differences were observed postpartum. Serum concentrations of total protein and albumin were greater prepartum for cows in ND and NDCA groups than for those in CON. Postpartum serum urea N and albumin concentrations tended to be higher for ND and NDCA cows. Cows fed ND or NDCA diets had elevated serum total cholesterol concentration prepartum. During the postpartum period, triglycerides and NEFA of cows fed ND or NDCA diets tended to be lower than those of CON. Cows fed the NDCA diet had greater postpartum total cholesterol in serum and lower NEFA concentration at calving than ND. In conclusion, feeding a prepartum negative DCAD diet altered blood acid-base balance and induced metabolic acidosis at calving, and improved protein and lipid metabolism. Supplementation of high Ca in the negative DCAD diet prepartum was more favorable to metabolic adaptation to lactation in dairy cows than the negative DCAD diet with low Ca.  相似文献

Our objective was to investigate the effects of prepartum metabolizable protein (MP) supply and management strategy on milk production and blood biomarkers in early lactation dairy cows. Ninety-six multigravida Holstein cows were used in a randomized complete block design study, blocked by calving date, and then assigned randomly to 1 of 3 treatments within block. Cows on the first treatment were fed a far-off lower MP diet [MP = 83 g/kg of dry matter (DM)] between ?55 and ?22 d before expected calving and then a close-up lower MP diet (MP = 83 g/kg of DM) until parturition (LPLP). Cows on the second treatment were fed the far-off lower MP diet between ?55 to ?22 d before expected parturition and then a prepartum higher MP diet (MP = 107 g/kg of DM) until calving (LPHP). Cows on the third treatment had a shortened 43-d dry period and were fed the prepartum higher MP diet from dry-off to parturition (SDHP). After calving, cows received the same fresh diet from d 0 to 14 and the same high diet from d 15 to 84. Data were analyzed separately for wk ?6 to ?1 and wk 1 to 12, relative to parturition. Dry matter intake from wk ?6 to ?1 was not different between LPHP and LPLP and increased for SDHP compared with LPLP. In contrast, dry matter intake for wk 1 to 12 postpartum did not change for LPHP versus LPLP or for SDHP versus LPLP. Compared with LPLP cows, LPHP cows had lower energy-corrected milk yield and tended to have decreased milk fat yield during wk 1 to 12 of lactation. Conversely, yields of energy-corrected milk and milk fat and protein were similar for SDHP compared with LPLP. Plasma urea N during wk ?3 to ?1 increased for LPHP versus LPLP and for SDHP versus LPLP; however, no differences in plasma urea N were observed postpartum. Elevated prepartum MP supply did not modify circulating total fatty acids, β-hydroxybutyrate, total protein, albumin, or aspartate aminotransferase during the prepartum and postpartum periods. Increased MP supply prepartum combined with a shorter dry period (SDHP vs. LPLP) tended to increase whole-blood β-hydroxybutyrate postpartum; however, other blood metabolites were not affected. Taken together, under the conditions of this study, elevated MP supply in close-up diets reduced milk production without affecting blood metabolites in multiparous dairy cows during early lactation. A combination of a shorter dry period and increased prepartum MP supply (i.e., SDHP vs. LPLP) improved prepartum dry matter intake without modifying energy-corrected milk yield and blood biomarkers in early lactation cows.  相似文献

The objective of this study was to evaluate the effects of supplementing a Saccharomyces cerevisiae fermentation product (SCFP; NutriTek, Diamond V, Cedar Rapids, IA) during the periparturient period (d ?28 ± 3 to 44 ± 3 relative to calving) on dry matter intake (DMI), milk production, apparent total-tract nutrient digestibility, and postpartum ovarian activity of dairy cows fed fresh diets varying in starch content. From d 28 ± 3 before the expected calving date until d 44 ± 3 after calving, 117 Holstein cows were fed diets with SCFP (SCFP; n = 59) or without (control, CON; n = 58). A common, basal, controlled-energy close-up diet (net energy for lactation: 1.43 Mcal/kg; 13.8% starch) was fed before calving. Cows within each treatment (CON or SCFP) were fed either a low- (LS; 22.1% starch) or high-starch (HS; 28.3% starch) diet from d 1 to 23 ± 3 after calving (fresh period), resulting in 4 treatment groups: LS-CON (n = 30), LS-SCFP (n = 29), HS-CON (n = 28), and HS-SCFP (n = 30). All cows were fed the HS diets from d 24 ± 3 to 44 ± 3 after calving (post-fresh period). Cows were assigned to treatment balanced for parity, body condition score, body weight, and expected calving date. Milk yield was higher for cows fed the LS diets compared with those fed the HS diets during the fresh period (34.1 vs. 32.1 kg/d), whereas DMI and 3.5% fat-corrected milk yield (FCM) were not affected by dietary starch content, and LS cows tended to lose more body condition than HS cows (?0.42 vs. ?0.35 per 21 d) during the fresh period. Overall DMI during the close-up and fresh periods did not differ between SCFP and CON cows. However, SCFP supplementation transiently increased DMI on d 1 (13.0 vs. 11.9 kg/d) and 5 (15.5 vs. 14.1 kg/d) after calving compared with CON. During the post-fresh period, SCFP cows tended to eat less than CON cows (19.8 vs. 20.6 kg/d) but had similar 3.5% FCM (44.9 vs. 43.6 kg/d), resulting in greater feed efficiency for SCFP cows (FCM/DMI; 2.27 vs. 2.13). Neither starch content of fresh diets nor SCFP supplementation affected the interval from calving to first ovulation or the incidence of double ovulation. These findings suggest that feeding low-starch diets during the fresh period can increase milk production of dairy cows during the fresh period, and that supplementation of SCFP may increase feed intake around calving and feed efficiency in the post-fresh period.  相似文献

Study objectives were to evaluate the effects of feeding rumen-protected Met (RPM) in pre- and postpartum total mixed rations (TMR) on health disorders and the interactions of health disorders with lactation and reproductive performance. Multiparous Holstein cows [470; 235 cows at University of Wisconsin (UW) and 235 cows at Cornell University (CU)] were enrolled at approximately 4 wk before parturition and housed in close-up dry cow (n = 6) and replicated lactation pens (n = 16). Pens were randomly assigned to treatment diets (pre- and postpartum, respectively): (1) control (CON): basal diet = 2.30% and 2.09% Met as % of metabolizable protein (MP) (UW) or 2.22% and 2.19% Met as % of MP (CU); (2) RPM: basal diet fed with RPM with 2.83% and 2.58% Met (Smartamine M, Adisseo Inc.; 12 g prepartum and 27 g postpartum), as % of MP (UW) or 2.85% and 2.65% Met (Smartamine M; 13 g prepartum and 28 g postpartum), as % of MP (CU). Total serum Ca was evaluated at the time of parturition and on d 3 ± 1 postpartum. Daily rumination was monitored from 7 d before parturition until 28 d postpartum. Health disorders were recorded during the experimental period until the time of first pregnancy diagnosis (32 d after timed artificial insemination; 112 ± 3 d in milk). Uterine health was evaluated on d 35 ± 3 postpartum. Time to pregnancy and herd exit were evaluated up to 350 d in milk. Treatment had no effect on the incidence of most health disorders and did not alter daily rumination. Cows fed RPM had reduced subclinical hypocalcemia (13.6 vs. 22%; UW only) on day of parturition relative to CON. Percentage of cows culled (13.1 vs. 19.3%) and hazard of herd exit due to culling [hazard ratio = 0.65, 95% confidence interval (CI): 0.42–1.02] tended to be reduced for cows fed RPM compared with CON. Moreover, cows fed RPM had greater milk protein concentration and protein yield overall, although retrospective analysis indicated that RPM only significantly increased protein yield in the group of cows with one or more health disorders (1.47 vs. 1.40 kg/d), not in cows without health disorders (1.49 vs. 1.46 kg/d) compared with CON. Overall, treatment had no effect on pregnancy per timed artificial insemination; however, among cows with health disorders, those fed RPM had reduced time to pregnancy compared with CON (hazard ratio = 0.71, 95% CI: 0.53–0.96). Thus, except for subclinical hypocalcemia on the day of parturition, feeding RPM in pre- and postpartum TMR did not reduce the incidence of health disorders, but our retrospective analysis indicated that it lessened the negative effects of health disorders on milk protein production and time to pregnancy.  相似文献

The objective of this study was to determine the effects of feeding synthetic zeolite A for 3 wk before expected calving on peripartal serum mineral concentrations, hypocalcemia, oxidant status, and performance. Holstein cows (n = 55) entering their second or greater lactations were assigned randomly to 1 of 2 dietary treatments starting 21 d before expected calving: control (CON: 40% corn silage, 33% wheat straw, and 27% concentrate; n = 29) or experimental [EXP: CON plus zeolite A (X-Zelit, Protekta Inc., Lucknow, ON, Canada/Vilofoss, Graasten, Denmark; n = 26) at an inclusion rate of 3.3% of dry matter, targeting 500 g/d as-fed]. Cows were fed the same postpartum diet and housed in individual tiestalls through 28 d in milk. Cows fed EXP had higher serum Ca concentrations as parturition approached and during the immediate postpartum period. Serum P concentrations were lower for the EXP-fed cows during the prepartum period and the first 2 d of lactation, whereas serum Mg concentrations were lower than those of the CON-fed cows only during the immediate periparturient period. Cows fed EXP had decreased prevalence of subclinical hypocalcemia (SCH) from d −1 through 3 relative to day of parturition, with the largest difference occurring within the first day postpartum. Prepartum dry matter intake tended to be decreased and rumination was decreased in cows fed EXP; however; postpartum dry matter intake, rumination, milk yield, milk component yield, and colostrum measurements did not differ between treatments. Cows fed EXP tended to have increased hazard of pregnancy by 150 d in milk when controlling for parity compared with CON-fed cows; potential reproductive benefits merit further study. This study demonstrated that zeolite A supplementation during the prepartum period results in markedly improved serum Ca concentrations around parturition and similar postpartum performance compared with controls and is effective at decreasing hypocalcemia in multiparous Holstein cows.  相似文献

Serum collected from multiparous Holsteins (n = 73) in a previous experiment was used to determine the effect of prepartum plane of energy intake on metabolites related to inflammation and liver function in periparturient cows. Prepartum diets were in a 3 × 2 factorial arrangement over the far-off (d ?65 to ?26 before expected parturition) and close-up (d ?25 relative to parturition until parturition) dry periods. During the far-off period, 2 diets were fed for ad libitum intake (ADLIB) to supply either 100% (100E) or 150% (150E) of National Research Council recommendations for net energy for lactation for mature cows in late gestation. For the third dietary far-off treatment, the 150E diet was fed at restricted intake (REST) to supply 80% (80E) of recommendations. During the close-up period, cows were fed a diet either at ADLIB or REST to supply 150% of net energy for lactation or 80% of net energy for lactation requirements, respectively. Beginning at parturition, all cows were fed a lactation diet through 56 d in milk. Cows fed 150E tended to accumulate more liver lipid postpartum; peak lipid accumulation occurred in all groups on d 14 postpartum. Cows fed 150E tended to have lower serum Ca on d 1 after calving, and cows fed REST had higher Ca than those fed ADLIB. Cows fed 150E tended to have higher serum bilirubin prepartum compared with other groups. Feeding REST in the close-up period resulted in higher bilirubin prepartum compared with ADLIB; bilirubin was positively associated (r = 0.34) with lipid accumulation postpartum. Feeding REST resulted in lower serum vitamin A (an indirect measure of retinol binding protein) prepartum compared with ADLIB, especially when coupled with 80E. Postpartum paraoxonase was negatively associated (r = ?0.27) and ceruloplasmin was positively associated (r = 0.21) with liver lipid accumulation postpartum. A larger spike in haptoglobin was observed on d 1 and 7 postpartum for cows fed 100E and 150E during the far-off period followed by REST during the close-up period. The ratio of albumin to globulin in serum was higher for cows fed 100E than for those fed 150E both prepartum and postpartum. Liver activity index indicated poorer liver function for cows fed 150E in the far-off period regardless of close-up diet. Negative liver function was also noted for cows fed 80E and REST. Cows in the 100E group fed ADLIB or REST had positive liver activity index postpartum. Avoidance of gross overconsumption of energy prepartum, particularly during the far-off period, appears best to reduce systemic inflammatory signals and improve liver function.  相似文献

The objective of this study was to evaluate the effects of the starch content of pre- and postpartum diets on productivity, plasma energy metabolites, and serum markers of inflammation of dairy cows during the calving transition period. Eighty-eight primiparous and multiparous cows were randomly assigned to pre- and postpartum dietary treatments balanced for parity and pretrial body condition score at d 28 ± 3 before expected calving date. Cows were fed either a control [Control; 14.0% starch, dry matter (DM) basis] or high-starch (High; 26.1% starch, DM basis) prepartum diet commencing 28 ± 3 d before expected calving date. Following calving, cows were fed either a high-fiber (HF; 33.8% neutral detergent fiber, 25.1% starch, DM basis) or high-starch (HS; 27.2% neutral detergent fiber, 32.8% starch, DM basis) postpartum diet for the first 20 ± 2 d following calving. Cows fed the High prepartum diet had greater DM intake (12.4 vs. 10.2 kg/d), plasma concentrations of insulin (1.72 vs. 14.2 ng/mL), glucose (68.1 vs. 65.0 mg/dL), and glucagon-like peptide-2 (0.41 vs. 0.32 ng/mL) before parturition, but increased plasma free fatty acid concentration (452 vs. 363 µEq/L) and milk fat yield (1.64 vs. 1.48 kg/d) after parturition. Cows fed the HS postpartum diet had lower plasma free fatty acid (372 vs. 442 µEq/L) and serum haptoglobin (0.46 vs. 0.70 mg/mL) concentrations over a 3-wk period after calving. In addition, there was a tendency for interaction between prepartum and postpartum diets for milk yield, where feeding the HS postpartum diet increased milk yield compared with the HF diet for cows fed the Control prepartum diet (40.8 vs. 37.9 kg/d) but not for cows fed the High prepartum diet. These results suggest that management efforts to minimize the change in diet fermentability during the calving transition by feeding the High prepartum diet, the HF postpartum diet, or both did not increase productivity of dairy cows but increased fat mobilization after calving. Our findings also suggest that feeding high-starch postpartum diets can decrease fat mobilization and serum indicators of systemic inflammation and increase milk production even with the transition from a low-starch prepartum diet.  相似文献

We used 48 multiparous Holstein cows to compare the response of dairy cows to a direct-fed mixture of cellulase and xylanase enzymes (1.25 L of enzyme concentrate/tonne of forage dry matter) applied to the forage portion (60% corn silage and 40% alfalfa hay) of a total mixed diet starting either in the close-up dry period, at calving, or at peak milk production. Cows were blocked by calving date and, within blocks, randomly assigned to one of four treatment diets. Treatments were: 1) an untreated control diet, 2) enzyme addition to the forage from wk 6 to 18 postpartum, 3) enzyme addition to the forage from calving to wk 18 postpartum, and 4) enzyme addition to the forage from wk 4 prepartum to wk 18 postpartum. Total mixed diets were 65% forage and 35% concentrate prepartum, and 50:50 forage:concentrate postpartum. The production of milk, solids-corrected milk, fat-corrected milk, and energy-corrected milk was higher for cows fed enzyme-treated diets than for cows fed control diet. Production was similar for cows in all enzyme-treated groups, although numerically highest for cows that started receiving enzyme-treated forages right after parturition and numerically lowest when started prepartum. Concentrations of fat, protein, and lactose in milk were similar for all treatments; yields of protein and fat were higher for cows fed enzyme-treated forages. Dry matter intake and body condition scores, both prepartum and postpartum, were similar for all diets. Eating rates, as determined in two 24-h studies, were similar for control and enzyme-treated diets. The feeding of enzyme-treated forages increased milk production. While the effect of when the feeding of enzyme-treated forages started was not statistically significant, we recommend starting soon after parturition because of the greatest total milk production when starting at that time.  相似文献

Negative energy balance during early postpartum is associated with reduced reproductive performance in dairy cows. A pooled statistical analysis of 7 studies completed in our group from 1993 to 2010 was conducted to investigate the association between prepartum energy feeding regimen and reproductive performance. The interval from calving to pregnancy (days to pregnancy, DTP) was the dependent variable to assess reproductive performance. Individual data for 408 cows (354 multiparous and 54 primiparous) were included in the analysis. The net energy for lactation (NEL) intake was determined from each cow’s average dry matter intake and calculated dietary NEL density. Treatments applied prepartum were classified as either controlled-energy (CE; limited NEL intake to ≤100% of requirement) or high-energy (HE; cows were allowed to consume >100%) diets fed during the far-off (FO) or close-up (CU) dry periods. Cow was the experimental unit. The Cox proportional hazard model revealed that days to pregnancy was shorter for CE (median = 157 d) than HE (median = 167 d) diets during the CU period [hazard ratio (HR) = 0.70]. Cows fed HE diets during the last 4 wk prepartum lost more body condition score in the first 6 wk postpartum than those fed CE diets (−0.43 and −0.30, respectively). Cows fed CE diets during the FO period had lower nonesterified fatty acids concentrations in wk 1, 2, and 3 of lactation than cows fed HE diets. Higher nonesterified fatty acids concentration in wk 1 postpartum was associated with a greater probability of disease (n = 251; odds ratio = 1.18). Cows on the CE regimen during the FO period had greater plasma glucose concentrations during wk 1 and 3 after calving than cows fed the HE regimen. Higher plasma glucose (HG) concentration compared with lower glucose (LG) in wk 3 (HG: n = 154; LG: n = 206) and wk 4 (HG: n = 71; LG: n = 254) after calving was associated with shorter days to pregnancy (wk 3: median = 151 and 171 d for HG and LG, respectively, and HR = 1.3; wk 4: median = 148 and 167 d, respectively, and HR = 1.4). In the first 2 wk after calving, cows that received HE diets in the FO period had higher concentrations of total lipids and triglyceride and greater ratio of triglyceride to glycogen in liver than cows fed CE diets. In conclusion, cows fed CE diets during the CU period had a shorter interval between parturition and conception, which may be explained by increased NEL intake during the first 4 wk postpartum and lower incidence of peripartal diseases. Lower body condition score loss during the first 6 wk postpartum and slightly higher glucose concentration at wk 3 likely contributed to improved reproductive performance.  相似文献

The objectives of this study were to determine the effect of decreasing dietary cation-anion difference [DCAD; (Na+ + K+) ? (Cl? + S2?)] of the prepartum diet on aspects of mineral metabolism, energy metabolism, and performance of peripartum dairy cows. Multiparous Holstein cows (n = 89) were enrolled between 38 and 31 d before expected parturition and randomized to treatments in a completely randomized design (restricted to balance for previous 305-d mature equivalent milk production, parity, and body condition score) at 24 d before expected parturition. Treatments consisted of a low-K ration without anion supplementation [CON; n = 30, DCAD = +18.3 mEq/100 g of dry matter (DM)]; partial anion supplementation to a low-K ration (MED; n = 30, DCAD = +5.9 mEq/100 g of DM); and anion supplementation to a low-K ration to reach a targeted average urine pH between 5.5 and 6.0 (LOW; n = 29, DCAD = ?7.4 mEq/100 g of DM). Cows were fed a common postpartum diet and data collected through 63 d in milk. Urine pH (CON = 8.22, MED = 7.89, and LOW = 5.96) was affected quadratically by decreasing prepartum DCAD. A linear relationship between urine pH and urine Ca:creatinine ratio was observed (r = ?0.81). Plasma Ca concentrations in the postpartum period (d 0 to 14; CON = 2.16, MED = 2.19, and LOW = 2.27 mmol/L) were increased linearly with decreasing prepartum DCAD. A treatment by parity (second vs. third and greater) interaction for postpartum plasma Ca concentration suggested that older cows had the greatest response to the low DCAD diet and older cows fed LOW had decreased prevalence of hypocalcemia after calving. A quadratic effect of decreasing DCAD on prepartum DMI was observed (CON = 13.6, MED = 14.0, and LOW = 13.2 kg/d). Milk production in the first 3 wk postpartum was increased linearly with decreasing DCAD (CON = 40.8, MED = 42.4, and LOW = 43.9 kg/d) and DMI in this period also tended to linearly increase (CON = 20.2, MED = 20.9, and LOW = 21.3 kg/d). Overall, effects on intake and milk yield analyzed over wk 1 to 9 postpartum were not significant. This study demonstrates that feeding lower DCAD diets prepartum improves plasma Ca status in the immediate postpartum period and results in increased DMI and milk production in the 3 wk after parturition. Compared with no anion supplementation or lower levels of anion supplementation, greater improvements were observed with the lower DCAD feeding strategy, in which an average urine pH of 5.5 to 6.0 was targeted.  相似文献

Eighty-five multiparous Holstein cows were used in a completely randomized design with restrictions to evaluate the effects of prepartum carbohydrate (CHO) source and monensin on periparturient dry matter intake (DMI), blood parameters, and lactation performance of dairy cows. Dietary treatments were arranged in a 2 × 2 factorial arrangement with a conventional (CONV) dry cow diet and a nonforage fiber source (NFFS) dry cow diet not supplemented (−) or supplemented (+) with 330 mg/cow per d of monensin as a top dressing. The CONV diet contained 70% forage and the NFFS diet contained nonforage fiber sources such that 28% of the forage was replaced with cottonseed hulls and soyhulls. The experimental diets (CONV and NFFS) were fed throughout the entire dry period (for 60 d before parturition). Monensin was top dressed once daily starting 28 d (27 ± 1.8 SD) before the expected calving date and continued until parturition. After parturition, all cows received the same lactating cow diet. During the last 28 d of gestation, cows receiving the NFFS diets prepartum had greater DMI (15.8 vs. 11.9 kg/d), DMI as a percentage of body weight (2.1 vs. 1.6% of body weight), plasma glucose (67.4 vs. 64.6 mg/dL), and serum insulin concentrations (0.59 vs. 0.45 ng/mL), and lower plasma nonesterified fatty acid concentrations (185 vs. 245 μEq/L) compared with cows receiving the CONV diets prepartum. Average milk production or composition during the first 56 d of lactation was not significantly affected by prepartum source of CHO, monensin, or their combination; however, there was a trend for the prepartum CHO source to affect milk production over time. Supplementation of monensin as a top dressing for 28 d prepartum had no effect on periparturient measurements. The prepartum diet did not affect postpartum DMI, blood glucose, nonesterified fatty acids, insulin concentrations, or liver triglyceride content. Results from this research demonstrated that partly replacing conventional dietary carbohydrate sources with NFFS, cottonseed hulls and soyhulls, in the dry cow diet improved or maintained the prepartum DMI and therefore enhanced the prepartum metabolic status, as indicated by key blood metabolite concentrations. Objectives were to evaluate the effect of prepartum energy intake on performance of dairy cows supplemented with or without ruminally protected choline (RPC; 0 or 17.3 g/d of choline chloride; 0 or 60 g/d of ReaShure, Balchem Corp., New Hampton, NY). At 47 ± 6 d before the expected calving date, 93 multiparous Holstein cows were assigned randomly to 1 of 4 dietary treatments in a 2 × 2 factorial arrangement. Cows were fed energy to excess [EXE; 1.63 Mcal of net energy for lactation/kg of dry matter (DM)] or to maintenance (MNE; 1.40 Mcal of net energy for lactation/kg of DM) in ad libitum amounts throughout the nonlactating period. The RPC was top-dressed for 17 ± 4.6 d prepartum through 21 d postpartum (PP). After calving, cows were fed the same methionine-balanced diet, apart from RPC supplementation, through 15 wk PP. Liver was biopsied at ?14, 7, 14, and 21 d relative to parturition. Cows fed EXE or MNE diets, respectively, consumed 40 or 10% more Mcal/d than required at 15 d before parturition. Cows fed the MNE compared with the EXE diet prepartum consumed 1.2 kg/d more DM postpartum but did not produce more milk (41.6 vs. 43.1 kg/d). Thus, PP cows fed the EXE diet prepartum were in greater mean negative energy balance, tended to have greater mean concentrations of circulating insulin, fatty acids, and β-hydroxybutyrate, and had greater triacylglycerol in liver tissue (8.3 vs. 10.7% of DM) compared with cows fed the MNE diet prepartum. Cows fed RPC in transition tended to produce more milk (43.5 vs. 41.3 kg/d) and energy-corrected milk (44.2 vs. 42.0 kg/d) without increasing DM intake (23.8 vs. 23.2 kg/d) during the first 15 wk PP, and tended to produce more milk over the first 40 wk PP (37.1 vs. 35.0 kg/d). Energy balance of cows fed RPC was more negative at wk 2, 3, and 6 PP, but mean circulating concentrations of fatty acids and β-hydroxybutyrate did not differ from those of cows not fed RPC. Despite differences in energy balance at 2 and 3 wk PP, mean concentration of hepatic triacylglycerol did not differ between RPC treatments. Feeding RPC reduced the daily prevalence of subclinical hypocalcemia from 25.5 to 10.5%, as defined by concentrations of total Ca of <8.0 mg/dL in serum in the first 7 d PP. Pregnancy at first artificial insemination tended to be greater for cows fed RPC (41.3 vs. 23.6%), but the proportion of pregnant cows did not differ by 40 wk PP. Heifers born from singleton calvings from cows fed RPC tended to experience greater daily gain between birth and 50 wk of age than heifers from cows not supplemented with RPC. Feeding RPC for approximately 38 d during the transition period tended to increase yield of milk for 40 wk regardless of amount of energy consumed during the pregnant, nonlactating period.  相似文献

The objective of this study was to determine the effect of dietary supplementation of n-3 polyunsaturated fatty acids (PUFA) and n-6 PUFA on dry matter intake (DMI), energy balance, oxidative stress, and performance of transition cows. Forty-five multiparous Holstein dairy cows with similar parity, body weight (BW), body condition score (BCS), and milk yield were used in a completely randomized design during a 56-d experimental period including 28 d prepartum and 28 d postpartum. At 240 d of pregnancy, cows were randomly assigned to one of the 3 isoenergetic and isoprotein dietary treatments, including a control ration containing 1% hydrogenated fatty acid (CON), a ration with 8% extruded soybean (HN6, high n-6 PUFA source), and a ration with 3.5% extruded flaxseed (HN3; high n-3 PUFA source). The HN6 and HN3 diets had an n-6/n-3 ratio of 3.05:1 and 0.64:1 in prepartum cows and 8.16:1 and 1.59:1 in postpartum cows, respectively. During the prepartum period (3, 2, and 1 wk before calving), DMI, DMI per unit of BW, total net energy intake, and net energy balance were higher in the HN3 than in the CON and NH6 groups. During the postpartum period (2, 3, and 4 wk after calving), cows fed HN3 and HN6 diets both showed increasing DMI, DMI as a percentage of BW, and total net energy intake compared with those fed the CON diet. The BW of calves in the HN3 group was 12.91% higher than those in the CON group. Yield and nutrient composition of colostrum (first milking after calving) were not affected by HN6 or HN3 but milk yield from 1 to 4 wk of milking was significantly improved compared with CON. During the transition period, BW, BCS, and BCS changes were not affected. Cows fed the HN6 diet had a higher plasma NEFA concentration compared with the CON cows during the prepartum period. Feeding HN3 reduced the proportion of de novo fatty acids and increased the proportion of preformed long-chain fatty acids in regular milk. In addition, the n-3 PUFA-enriched diet reduced the n-6/n-3 PUFA ratio in milk. In conclusion, increasing the n-3 fatty acids concentration in the diet increased both DMI during the transition period and milk production after calving, and supplementing n-3 fatty acids was more effective in mitigating the net energy balance after calving.  相似文献

Mechanisms regulating subcutaneous adipose tissue (SAT) insulin sensitivity and gene network expression during the peripartal period were evaluated in cows fed to meet or exceed prepartal energy requirements. Holstein cows were dried off at -50 d relative to expected parturition and fed a controlled-energy diet [CON; net energy for lactation=1.24 Mcal/kg of dry matter (DM); 36% of DM as wheat straw] until -21 d. Cows were then randomly assigned (n=7/diet) to either the same CON diet or a moderate-energy close-up diet (OVE; net energy for lactation=1.47 Mcal/kg of DM) until parturition. Biopsies of SAT were harvested at -10, 7, and 21 d for mRNA expression of 48 genes associated with insulin signaling, adipogenesis, and lipolysis. In vitro basal and insulin-stimulated insulin receptor substrate 1 tyrosine phosphorylation (IRS1-PY) was assessed at -10 and 7 d. The OVE led to more positive energy balance and greater serum insulin concentration prepartum. Compared with CON, OVE led to a more drastic increase in serum NEFA and also greater overall serum BHBA postcalving, both of which were associated with greater hepatic total lipid and triacylglycerol concentration. Close-up OVE did not improve any aspect of performance. In prepartal SAT, insulin-stimulated IRS1-PY was greater in OVE than in CON. However, IRS1-PY, serum insulin, and GLUT4 expression decreased postpartum regardless of prepartal treatment, suggesting a more severe state of insulin resistance. The expression of all genes encoding adipogenic regulators (PPARG and ZFP423), most lipogenic enzymes/inducers (FASN, SCD, DGAT2, and INSIG1), and basal-lipolysis regulators (ATGL and ABDH5) was greater at -10 d in OVE than in CON. Whereas adipogenic and basal lipolysis regulator expression remained greater in cows fed OVE by 7 d postpartum, expression of all lipogenic enzymes decreased regardless of diet. Despite those responses, the approximately 3-fold increase in expression of IRS1 and ZFP423 between 7 and 21 d suggested that insulin responsiveness and adipogenic capacity of SAT were partially restored. Expression of the preadipocyte marker DLK1, adiponutrin (PNPLA3), and fibroblast growth factor 21 (FGF21) was undetectable. Results suggested that close-up energy overfeeding did not exacerbate insulin resistance in SAT. Signs of restored insulin responsiveness (upregulation of IRS1, INSIG2, SREBF1, and ZFP423) were apparent as early as 3 wk postpartum. Thus, identifying specific nutrients capable of activating PPARγ after calving in AT might help accelerate its replenishment. A regulatory network encompassing the genes and physiological measurements obtained is proposed.  相似文献

We determined the effects of day relative to parturition and prepartum plane of nutrition on hepatic partitioning of palmitate metabolism to CO2, acid-soluble products (ASP), and esterified products (EP). Multiparous Holsteins (n = 74) were fed different amounts of nutrients during the dry period in a 3 (far-off period diet) × 2 (close-up period diet) factorial arrangement. During the far-off period (d −60 to −25) cows received a low-energy control diet fed ad libitum (100NRC) to meet National Research Council (NRC) requirements, a moderate-energy diet fed ad libitum to exceed NRC recommendations for net energy of lactation (NEL) by >50% (150NRC), or the same diet fed at restricted intake to provide 80% of NEL requirements (80NRC). During the close-up period (d −24 until parturition), cows were fed a diet for ad libitum intake to meet NRC recommendations or in restricted amounts to provide 80% of calculated NEL requirements. After parturition, all cows had ad libitum access to a lactation diet. Liver slices from biopsies on d −30, −14, 1, 14, and 28 relative to parturition were used to determine conversion of [1-14C] palmitate to CO2, ASP, and EP. Across diets, oxidation of palmitate to CO2 was decreased postpartum, whereas oxidation to ASP was increased at d 1 postpartum compared with other times. Conversion of palmitate to EP increased markedly postpartum, with the greatest rates at d 1 postpartum. Conversion of palmitate to CO2 and ASP on d 1 postpartum was lower and the proportion of palmitate metabolism as EP was greater for cows fed 150NRC than for those fed 100NRC or 80NRC. Hepatic triacylglycerol concentration at d 1 postpartum was greatest for cows fed 150NRC. Palmitate metabolism did not differ between close-up diets. Hepatic triacylglycerol was negatively correlated with tissue metabolism of palmitate to CO2 and ASP but positively correlated with metabolism to EP. Hepatic triacylglycerol was highly correlated with NEFA concentrations on the day of calving and d 1 postpartum but not with NEFA prepartum. In contrast, plasma BHBA postpartum was not correlated with hepatic palmitate metabolism by liver slices but was highly correlated with NEFA concentration prepartum. Excessive energy intake during the far-off dry period decreased hepatic palmitate oxidation and shifted palmitate metabolism toward greater esterification, consistent with greater hepatic triacylglycerol accumulation postpartum.  相似文献

Thirty-two multiparous Holstein cows were used to investigate the effects of chromium-l-methionine (Cr-Met) supplementation and dietary grain source on performance and lactation during the periparturient period. Cows were fed a total mixed ration consisting of either a barley-based diet (BBD) or a corn-based diet (CBD) from 21 d before anticipated calving through 28 d after calving. The Cr-Met was supplemented at dosages of 0 or 0.08 mg of Cr/kg of metabolic body weight. The study was designed as a randomized complete block design with 2 (Cr-Met levels) × 2 (grain sources) factorial arrangement. There was no Cr effect on prepartum dry matter intake (DMI) or postpartum DMI, body weight (BW), net energy balance, and whole tract apparent digestibility of nutrients. Prepartum DMI as a percentage of BW tended to increase with Cr-Met. Supplemental Cr-Met tended to increase milk yield whereas milk protein percentage decreased. Pre- and postpartum DMI, BW, net energy balance, milk yield, and milk composition were not affected by substituting ground barley with ground corn. The addition of Cr-Met increased prepartum DMI and tended to increase postpartum DMI of the BBD but not the CBD. The change in prepartum DMI was smaller when the BBD was supplemented with Cr-Met but remained unchanged when the CBD was supplemented with Cr-Met. Yields of crude protein and total solids in milk and prepartum digestibility of DM and organic matter tended to increase when Cr-Met was added to the BBD but remained unchanged when added to the CBD. Periparturient cows failed to respond to the grain source of the diet, whereas they showed greater response in milk yield to diets supplemented with Cr-Met. In conclusion, the present results demonstrate that the beneficial effect of Cr-Met supplementation during the periparturient period to improve feed intake may depend on the grain source of the diet.  相似文献

Holstein cows (n = 72) entering second or later lactation were used to determine whether productive performance and dry matter intake (DMI) are affected by carbohydrate source in the prepartum diet and chromium-L-methionine (Cr-Met) supplementation throughout the periparturient period. Cows were fed either a TMR with the concentrate portion based on starch-based cereals [high nonfiber carbohydrate (NFC); 1.59 Mcal/kg of net energy for lactation (NEL), 14.4% crude protein (CP), 40.3% NFC] or a TMR with the concentrate portion based on nonforage fiber sources (low NFC; 1.54 Mcal/kg NEL, 14.5% CP, 33.6% NFC) from 21 d before expected parturition until parturition. After parturition all cows were fed a lactation TMR (1.74 Mcal/kg NEL, 16.5% CP, 40.0% NFC). The Cr-Met was supplemented once daily via gelatin capsule at dosages of 0, 0.03, or 0.06 mg of Cr/kg of metabolic body weight. Thus, treatments were in a 2 (carbohydrate source) x 3 (Cr-Met) factorial arrangement. Neither prepartum nor postpartum DMI was affected by prepartum dietary carbohydrate source. Administering increasing amounts of Cr-Met linearly increased milk yield and, subsequently, postpartum DMI. Prepartum carbohydrate source did not affect postpartum milk yield; however, cows fed the low NFC diet tended to yield milk with a lower content of total solids. These data indicate that prepartum carbohydrate source has little influence on performance during the immediate peripartal period, and that increases in milk yield for cows supplemented with Cr-Met are independent of prepartum dietary carbohydrate source.  相似文献

An experiment was conducted to determine the effect of plane of energy intake prepartum on postpartum performance. Primiparous (n = 24) and multiparous (n = 23) Holsteins were randomly assigned by expected date of parturition to 1 of 3 prepartum energy intakes. A moderate energy diet [1.63 Mcal of net energy for lactation (NEL)/kg; 15% crude protein (CP)] was fed for either ad libitum intake (OVR) or restricted intake (RES) to supply 150 or 80% of National Research Council (2001) energy requirement, respectively, for dry cows in late gestation. To limit energy intake to 100% of NRC requirement at ad libitum dry matter intake (DMI), chopped wheat straw was included as 31.8% of dry matter (DM) in a control diet (CON; 1.21 Mcal of NEL/kg of DM; 14% CP). Multiparous and primiparous cows assigned to OVR gained body condition during the dry period [initial body condition score (BCS) = 3.3], but were not overconditioned by parturition (BCS = 3.5). Multiparous cows in the OVR group lost more BCS postpartum than multiparous RES or CON cows. Primiparous cows lost similar amounts of BCS among dietary treatment groups postpartum. Addition of chopped wheat straw to CON diets prevented a large decrease in DMI prepartum in both primiparous and multiparous cows. During the first 3 wk postpartum, DMI as a percentage of BW was lower for multiparous OVR cows than for multiparous RES cows. Prepartum diet effects did not carry over through the entire 8-wk lactation period. Because of greater mobilization of body stores, OVR cows had greater milk fat percentage and greater 3.5% fat-corrected milk yield during the first 3 wk postpartum. Multiparous cows assigned to OVR experienced a 55% decrease in energy balance and primiparous cows a 40% decrease in energy balance during the last 3 wk before parturition, compared with CON or RES cows that had little change. Multiparous cows fed OVR had a greater contribution of energy from body energy reserves to milk energy output than either CON or RES cows. Overfeeding energy prepartum resulted in large changes in periparturient energy balance. Even in the absence of overconditioning, a large change in DMI and energy balance prepartum influenced postpartum DMI and BCS loss, especially for multiparous cows. Chopped wheat straw was effective at controlling energy intake prepartum, although primiparous cows did not achieve predicted DMI. Even so, controlling or restricting energy intake in primiparous cows was not detrimental to lactational performance over the first 8 wk of lactation.  相似文献
