The purpose of this study was to optimize the conditions of a previously proposed enzymatic method used to estimate in situ undigested neutral detergent fiber (uNDF). We used a multi-step enzymatic approach, in which samples were first solubilized in NaOH solutions as a preincubation (PreInc) phase. After rinsing, samples were incubated (24 h at 39°C) in a buffered solution (pH 6) containing hemicellulase, cellulase, and Viscozyme L enzymes (Sigma-Aldrich s.r.l., Milan, Italy), followed by incubation (24 h at 39°C) in a buffered solution (pH 5) containing xylanase. Two sets of experiments were performed: a calibration trial (that tested different PreInc conditions on 9 selected forages) and a validation trial (that verified the results by testing multiple samples of 6 different forage types and a group of fibrous by-products). In the calibration trial, samples (300 mg in Ankom F57 filter bags; Ankom Technology Corp., Fairport, NY) were preincubated at 39°C in a 0.1 M NaOH solution for 90, 180, or 240 min, or in 0.2, 0.5, 1.0, or 2.0 M NaOH solution for 90 min. The results indicated that the best PreInc method, in terms of intra-laboratory repeatability and estimation of reference in situ values, was 90 min in a 0.2 M NaOH solution. Thus, we used this PreInc condition to determine enzymatic uNDF of 257 samples in the validation trial. Although the selected method generally had good accuracy in predicting in situ uNDF, inconsistencies were noted for certain forage types. Overall, when enzymatic uNDF was used to predict the in situ uNDF of all samples, the regression was satisfactory (intercept = 7.098, slope = 0.920, R2 = 0.73). The regression models developed for alfalfa hays, corn silages, and small grain silages had also acceptable regression performances and mean square error of prediction (MSEP) values, and the main sources of MSEP variation were error due to incomplete (co)variation and random error. Even when R2 values were >0.70, the MSEP value of the regression model for grass hays was 149.55, and that for nonforage fibrous feeds was 155.16. Although enzymatic uNDF partially overestimated the in situ uNDF, particularly in grass silages, the proposed procedure seems to be promising for accurately predicting in situ uNDF, because it generally had good repeatability and provided satisfactory estimates of in situ uNDF.  相似文献

The objective of this study was to assess the variability in amylase-treated neutral detergent fiber (aNDF) and crude protein (CP) analyses of feed and total mixed ration (TMR) samples among feed analysis laboratories. Two TMR were prepared that varied in the dry matter proportion of forage and concentrates: 45% forage (LF-TMR) versus 60% forage (HF-TMR). Replicated TMR and individual feed samples were dried, ground through a 4-mm screen, and sent to 10 commercial and 4 research or development laboratories for aNDF and CP analyses. Laboratories were asked to complete a detailed questionnaire regarding the aNDF procedure used. Variability in aNDF and CP analyses was assessed using univariate statistics and mixed modeling procedures. Significant variability in the aNDF analysis of individual feeds was found among the participating laboratories. The variability was particularly large for low-aNDF feeds such as distillers and barley grains. The variability among laboratories in the aNDF analysis of low-fiber TMR was greater than of high-fiber TMR, with the most likely reason being their greater proportions of grains and protein concentrates and the effect of variation in the aNDF protocols, particularly α-amylase use, on the analysis of these types of feeds. Variability due to the technique used for aNDF analysis was not statistically significant when outlier labs using the filter bag technique were removed; however, laboratories using the filter bag technique tended to produce more variable results than did laboratories using variation of the crucible technique (SE = 2.542 vs. 0.930, respectively). Calculated aNDF values for TMR, based on proportions and aNDF analysis of individual feeds, were slightly greater than analyzed aNDF values for TMR. Results from this ringtest emphasize the need for feed analysis laboratories to follow the official aNDF method exactly. Variation within and among laboratories can be reduced by replicating analysis and including reference materials in each analytical run. Results of CP analysis were more consistent among laboratories, and variability in CP analysis of individual feeds or TMR was marginally acceptable.  相似文献

In vitro and in situ procedures performed to estimate indigestible neutral detergent fiber (iNDF) in forage or fecal samples are time consuming, costly, and limited by intrinsic factors. In contrast, near infrared reflectance spectroscopy (NIRS) has become widely recognized as a valuable tool for accurately determining chemical composition and digestibility parameters of forages. The aim of this study was to build NIRS calibrations and equations for fecal iNDF. In total, 1,281 fecal samples were collected to build a calibration data set, but only 301 were used to develop equations. Once dried, samples were ground and chemically analyzed for crude protein, ash, amylase and sodium sulfite–treated NDF corrected for ash residue (aNDFom), acid detergent fiber, acid detergent lignin, and in vitro digestion at 240 h to estimate iNDF (uNDF240). Each fecal sample was scanned using a NIRSystem 6500 instrument (Perstorp Analytical Inc., Silver Spring, MD). Spectra selection was performed, resulting in 301 sample spectra used to develop regression equations with good accuracy and low standard error of prediction. The standard error of calibration (SEC), cross validation (SECV), and coefficients of determination for calibration (R2) and for cross validation (1 ? VR, where VR = variance ratio) were used to evaluate calibration and validation results. Moreover, the ratio performance deviation (RPD) and ratio of the range of the original data to SECV (range/SECV; range error ratio, RER) were also used to evaluate calibration and equation performance. Calibration data obtained on fiber fractions aNDFom (R2 = 0.92, 1 ? VR = 0.87, SEC = 1.48, SECV = 1.89, RPD = 2.80, and RER = 20.19), uNDF240 (R2 = 0.92, 1 ? VR = 0.86, SEC = 1.65, SECV = 2.24, RPD = 2.57, and RER = 14.30), and in vitro rumen aNDFom digestibility at 240 h (R2 = 0.90, 1 ? VR = 0.85, SEC = 2.68, SECV = 3.43, RPD = 2.53, and RER = 14.0) indicated the predictive equations had good predictive value.  相似文献

The effect of 3 fermentation methods, in situ (IS) in 4 lactating cows (average pH of 5.8), in vitro (IVn) with media pH of 6.8, or in vitro (IVa) with media pH adjusted to 6.0 using citric acid, on fiber degradation parameters was studied using feeds ground to different particle sizes. Corn silage (CS), grass silage (GS), barley grain (B), sugar beet pulp (BP), and rapeseed cake (RC) were ground using a shear mill. Silages were ground through 8-, 4-, 2-, or 1-mm screens, B and BP through 4-, 2-, or 1-mm screens, and RC through 2- or 1-mm screens. The amylase-treated NDF (aNDF) content of samples ground using a 1-mm screen was 399, 431, 197, 480, and 251 g/kg of DM for CS, GS, B, BP, and RC, respectively, but increased with increasing screen size. Materials were incubated for 0, 6, 12, 24, 48, and 96 h IS, IVn, or IVa. Inoculum for IVn and IVa was prepared as a composite from the cows used for IS. The potentially degradable aNDF (D0), indegradable aNDF (I), lag time (L), and fractional rate of degradation of potential degradable aNDF (kd) were estimated using PROC NLIN in SAS. Except for RC, fermentation methods affected most degradation parameters, especially kd and L. The IVn, IVa, and IS methods resulted in kd values of 0.291, 0.105, 0.080 h−1 and 0.262, 0.107, 0.103 h−1 for BP and RC, respectively, demonstrating a decreasing rate of degradation for these feeds when fermented under suboptimal pH. In CS, GS, and B, no difference was found in kd between the IVn and IVa methods, which suggests that differences in pH did not alter kd in vitro. The kd values obtained for CS, GS, and B were 0.058, 0.109, 0.168, and 0.028, 0.054, and 0.069 h−1 for the IVn and IS methods, respectively, indicating that the IS method using cows fed at production levels can underestimate the potential rate of NDF degradation. Using the IVa method, L was 12.1, 9.1, 7.8, and 2.5 h for CS, GS, BP, and RC, respectively, which was higher than L obtained from the IVn and IS methods for all feeds except B, where L in all methods were near the parameter boundary of zero hour in NLIN. It was concluded that fermentation methods were more important than grinding screen size on estimates for feed aNDF degradation and that the individual aNDF degradation parameters for the 5 feeds were affected differently by fermentation methods.  相似文献

Neutral detergent fiber (NDF) is the most commonly reported metric for fiber in dairy cattle nutrition. An empirical method, NDF is defined by the procedure used to measure it. The current definitive method for NDF treated with amylase (aNDF) is AOAC Official Method 2002.04 performed on dried samples ground through the 1-mm screen of a cutting mill with refluxing and then filtration through Gooch crucibles without (AOAC−; reference method) or with (AOAC+) a glass fiber filter filtration aid. Other methods in use include grinding materials through the 1-mm screen of an abrasion mill, using filtration through a Buchner funnel with a glass fiber filter (Buch), and use of the ANKOM system (ANKOM Technology, Macedon, NY) that simultaneously extracts and filters samples through filter bags with larger (F57) or smaller (F58) particle size retentions. Our objective was to compare the AOAC and alternative methods using samples ground through the 1-mm screens of cutting or abrasion mills. Materials analyzed were 2 alfalfa silages, 2 corn silages, dry ground and high-moisture corn grains, mixed grass hay, ryegrass silage, soybean hulls, calf starter, and sugar beet pulp. Samples were run in duplicate in replicate analytical runs performed on different days by experienced technicians. Compared with cutting mill–ground samples, the aNDF% of dry matter results from abrasion mill–ground samples were or tended to be lower for 8 of 11 samples. Method affected aNDF% results for all materials, with method × grind interactions for 6 of 11 samples. For ash-free aNDF% assessed with cutting mill–ground materials, a priori selected contrasts showed that the number of materials for which methods differed or tended to differ from the AOAC methods were 4 (Buch), 8 (F57), and 3 (F58); and 3 for AOAC– versus AOAC+. However, statistically different does not necessarily mean substantially different. For a given feed and grind, a positive value for the absolute difference between the AOAC– mean and an alternative method mean minus 2 times the standard deviation of AOAC− suggests that values for the alternative method fall outside of the range of results likely to be observed for the reference method. The number of observed positive values for materials processed with cutting and abrasion mills, respectively, were 0 and 2 (AOAC+); 2 and 2 (Buch); 8 and 10 (F57); 4 and 7 (F58); and 0 and 4 (AOAC−). With the materials tested, methods in order of agreement with the reference method were Buch, F58, and F57, which often gave lower values. The AOAC+ gave results similar to AOAC−, substantiating it as an allowed modification of AOAC−. Best agreement between the reference method and variant NDF methods was achieved with the 1-mm screen cutting mill grind. The 1-mm abrasion mill grind produced more aNDF% results that were lower than the reference method but with fewer differences when filter particle retention size was smaller. The use of filters that retain finer particles could be explored to improve comparability of variant NDF methods and grinds. Further evaluation with an expanded set of materials is warranted.  相似文献

This study evaluated the effects of feeding diets that were formulated to contain similar proportions of undigested neutral detergent fiber (uNDF) from forage, with wheat straw (WS) substituted for corn silage (CS), alfalfa hay (AH), or both. The diets were fed to lactating dairy cows and intake, digestibility, blood metabolites, and milk production were examined. Thirty-two multiparous Holstein cows (body weight = 642 ± 50 kg; days in milk = 78 ± 11 d; milk production = 56 ± 6 kg/d; mean ± standard deviation) were used in a randomized block design with 6-wk periods after a 10-d covariate period. Each period consisted of 14 d of adaptation followed by 28 d of data collection. The control diet contained CS and AH as forage sources (CSAH) with 17% of dietary dry matter as uNDF after 30 h of incubation (uNDF30). Wheat straw was substituted for AH (WSCS), CS (WSAH), or both (WSCSAH) on an uNDF30 basis, and beet pulp was used to obtain similar concentrations of NDF digestibility after 30 h of incubation (NDFD30 = 44.5% of NDF) across all diets. The 4 diets also contained similar concentrations of net energy for lactation and metabolizable protein. Dry matter intake was greatest for WSCS (27.8 kg/d), followed by CSAH (25.7 kg/d), WSCSAH (25.2 kg/d), and WSAH (24.2 kg/d). However, yields of milk, 3.5% fat-corrected milk (FCM), and energy-corrected milk did not differ, resulting in higher FCM efficiency (kg of FCM yield/kg of dry matter intake) for WSAH (1.83) and WSCSAH (1.79), followed by CSAH (1.69) and WSCS (1.64). Milk protein percentage was greater for CSAH (2.84%) and WSCS (2.83%) than for WSAH (2.78%), and WSCSAH (2.81%) was intermediate. The opposite trend was observed for milk urea nitrogen, which was lower for CSAH (15.8 mg/dL), WSCS (15.8 mg/dL), and WSCSAH (17.0 mg/dL) than for WSAH (20 mg/dL). Total-tract NDF digestibility and ruminal pH were greater for diets containing WS than the diet without WS (CSAH), but digestibility of other nutrients was not affected by dietary treatments. Cows fed WSAH had less body reserves (body weight change = −13.5 kg/period) than the cows fed the other diets, whereas energy balance was greatest for those fed WSCS. The results showed that feeding high-producing dairy cows diets containing different forage sources but formulated to supply similar concentrations of uNDF30 while maintaining NDFD30, net energy for lactation, and metabolizable protein constant did not influence milk production. However, a combination of WS and CS (WSCS diet) compared with a diet with CS and AH improved feed intake, ruminal pH, total-tract NDF digestibility, and energy balance of dairy cows.  相似文献

A portion of the forage cell wall, defined as neutral detergent fiber (NDF), is indigestible to anaerobic microbial digestion in ruminants. This fraction has been characterized by surface area relationships between acid detergent lignin, but recently, data have been published describing the dynamic nature of this relationship. In situ approaches have been described to estimate indigestible NDF, recovering the undigested NDF after long-term fermentations (uNDF). To be applicable to nutritionists and diet formulation, determining uNDF needs to be conducted in a commercial laboratory similar to other routine analyses of forage chemistry. A series of studies were conducted to evaluate an in vitro approach, to describe uNDF, which is repeatable and adaptable for routine feed evaluation. One hundred and two forages of several species were analyzed for NDF, acid detergent lignin, and uNDF. The uNDF was estimated by several approaches involving long-term fermentations and filtration steps to evaluate the length of time necessary to exhaust the digestible NDF and a filtration method necessary to maintain sample integrity by ensuring low sample loss and uniform recovery with residues from long-term in vitro fermentation. To determine uNDF, in vitro fermentations were conducted on 0.50 or 0.75 g of dry matter samples, in triplicate, at multiple time points up to 504 h and initially used Gooch crucibles with Celite (Thermo Fisher Scientific, Waltham, MA) as a filtering aid. The final method utilized a 1.5-µm pore size glass microfiber filter, which allowed for increased repeatability and improved sample recovery (lowest standard deviation). In this study, in vitro fermentations of 240 h were adequate to characterize and identify uNDF, which was repeatable among conventional forages provided the samples, after NDF analyses, were filtered through the same glass fiber filter. This approach could be adapted by commercial laboratories and would provide opportunities to develop near-infrared reflectance spectroscopy equations and calibrations.  相似文献

Long-term in vitro fermentation (240 h) evaluating amylase-treated, ash-corrected neutral detergent fiber, with addition of sodium sulfite (aNDFom) digestibility is required to quantify the indigestible fiber fraction. It is commonly accepted to inoculate rumen fluid more than one time during such fermentations, every 96 h or at 120 h. However, no studies have been conducted to verify if the reinoculation is actually required to properly carry out the fermentation process. The current study aims to evaluate the effects of these procedures on aNDFom digestibility at 240 h. The study was conducted on a total of 24 forage samples (8 alfalfa hays, 8 grass hays, and 8 corn silages). Samples were digested in triplicate at 240 h in vitro. Rumen fluid was added twice (at 96 and 192 h) in treatment 1, after 120 h in treatment 2, whereas no addition was made in treatment 3. At the end of the fermentations, residual aNDFom was quantified to calculate digestibility. Among treatments, no difference was found in digestibility of aNDFom. Moreover, treatment 1 resulted in higher variability compared with other treatments. Results obtained in the current study show that subsequent addition of rumen fluid is not necessary for a proper estimation of aNDFom digestibility, and can be avoided.  相似文献

The objective of this study was to investigate the effects of 2 alfalfa hays differing in undigested neutral detergent fiber content and digestibility used as the main forage source in diets fed to high producing cows for Parmigiano-Reggiano cheese production. Diets were designed to have 2 different amounts of undigestible NDF [high (Hu) and low (Lu)], as determined by 240-h in vitro analysis (uNDF240). Alfalfa hay in vitro digestibility [% of amylase- and sodium sulfite-treated NDF with ash correction (aNDFom)] at 24 and 240 h was 40.2 and 31.2% and 53.6 and 45.7% for low- (LD) and high-digestibility (HD) hays, respectively. The 4 experimental diets (Hu-HD, Lu-HD, Hu-LD, and Lu-LD) contained 46.8, 36.8, 38.8, and 30.1% of alfalfa hay, respectively, 8.6% wheat straw, and 35.3% corn (50% flake and 50% meal; DM basis). Soy hulls and soybean meal were used to replace hay to balance protein and energy among diets. Eight multiparous Holstein cows (average milk production = 46.0 ± 5.2 kg/d, 101 ± 38 d in milk, and 662 ± 42 kg of average body weight) were assigned to a 4 × 4 Latin square design, with 2 wk of adaptation and a 1-wk collection period. Dry matter and water intake, rumination time, ruminal pH, and milk production and composition were measured. Diets and feces were analyzed for NDF on an organic matter basis (aNDFom), acid detergent fiber, acid detergent lignin, and uNDF240 to estimate total-tract fiber digestibility. Dry matter intake and rumination times were higher in HD diets compared with LD diets, regardless of forage amount. Rumination time was constant per unit of dry matter intake but differed when expressed as a function of uNDF240, aNDFom, or physically effective NDF intake. No differences were found among treatments on average ruminal pH, but the amount of time with pH <5.8 was lower in Hu-HD diets. Milk production and components were not different among diets. Total-tract aNDFom and potentially digestible neutral detergent fiber fraction digestibility was higher for the LD diets (88.3 versus 85.8% aNDFom in HD), for which lower feed intakes were also observed. The Hu-HD diet allowed greater dry matter intake, longer rumination time, and higher ruminal pH, suggesting that the limiting factor for dry matter intake is neutral detergent fiber digestibility and its relative rumen retention time.  相似文献

Four lactating dairy cows equipped with ruminal and duodenal cannulas were used to determine the impact of different methods of treating soybean meal (SBM) on the ruminal degradability and intestinal digestibility of crude protein and AA. Solvent-extracted SBM (SE), expeller SBM (EP), lignosulfonate SBM (LS), and heat and soyhulls SBM (HS) were incubated in the rumen in nylon bags for 48, 24, 16, 8, 4, 2, and 0 h according to National Research Council (2001) guidelines. Additional samples of each SBM product were also incubated for 16 h in the rumen; the residues from these bags were transferred to mobile bags, soaked in pepsin HCl, and then used for determination of intestinal digestibility in situ or in vitro. Treatment of SBM (EP, LS, HS) protected the crude protein and AA from ruminal degradation, increasing rumen undegradable protein from 42% in SE to 68% in EP. Kinetic analysis of crude protein and AA degradation in the rumen revealed that, compared with LS and HS, EP exhibited slower rates of degradation but a shorter lag phase and a higher proportion of soluble protein. For all SBM products, the pattern of ruminal degradation, at 16 h of incubation, was characterized by extensive degradation of Lys and His, whereas Met and the branched-chain AA were degraded to the least extent. Estimates of intestinal digestibility of AA and crude protein were lower when measured in vitro than in situ; the magnitude of the difference between the 2 methods was greater (25%) with treated SBM products than with SE (10%). The availability of essential and nonessential AA was consistently greater (30%) with treated SBM than with SE. Among the treated SBM products, 4 essential AA (Ile, Leu, Phe, and Val) showed differences in availability, with values consistently lower for HS than for LS. This study showed that, based on in situ measures, heat and chemical treatment of SBM enhanced AA availability, and that compared with HS, EP and LS had a higher potential to enhance the AA supply to the small intestine of high-producing dairy cows.  相似文献

The effect of neutral detergent fiber (NDF) degradability of corn silage in diets containing lower and higher NDF concentrations on lactational performance, nutrient digestibility, and ruminal characteristics in lactating Holstein cows was measured. Eight ruminally cannulated Holstein cows averaging 91 ± 4 (standard error) days in milk were used in a replicated 4 × 4 Latin square design with 21-d periods (7-d collection periods). Dietary treatments were formulated to contain either conventional (CON; 48.6% 24-h NDF degradability; NDFD) or brown midrib-3 (BM3; 61.1% 24-h NDFD) corn silage and either lower NDF (LNDF) or higher NDF (HNDF) concentration (32.0 and 35.8% of ration dry matter, DM) by adjusting the dietary forage content (52 and 67% forage, DM basis). The dietary treatments were (1) CON-LNDF, (2) CON-HNDF, (3) BM3-LNDF, and (4) BM3-HNDF. Data were analyzed as a factorial arrangement of diets within a replicated Latin square design with the MIXED procedure of SAS (SAS Institute Inc., Cary, NC) with fixed effects of NDFD, NDF, NDFD × NDF, period(square), and square. Cow within square was the random effect. Time and its interactions with NDFD and NDF were included in the model when appropriate. An interaction between NDFD and NDF content resulted in the HNDF diet decreasing dry matter intake (DMI) with CON corn silage but not with BM3 silage. Cows fed the BM3 corn silage had higher DMI than cows fed the CON corn silage, whereas cows fed the HNDF diet consumed less DM than cows fed the LNDF diet. Cows fed the BM3 diets had greater energy-corrected milk yield, higher milk true protein content, and lower milk urea nitrogen concentration than cows fed CON diets. Additionally, cows fed the BM3 diets had greater total-tract digestibility of organic matter and NDF than cows fed the CON diets. Compared with CON diets, the BMR diets accelerated ruminal NDF turnover. When incorporated into higher NDF diets, corn silage with greater in vitro 24-h NDFD and lower undegradable NDF at 240 h of in vitro fermentation (uNDF240) allowed for greater DMI intake than CON. In contrast, for lower NDF diets, NDFD of corn silage did not affect DMI, which suggests that a threshold level of inclusion of higher NDFD corn silage is necessary to observe enhanced lactational performance. Results suggest that there is a maximum gut fill of dietary uNDF240 and that higher NDFD corn silage can be fed at greater dietary concentrations.  相似文献

Newborn Holstein bull calves (n = 32) were assigned to receive a colostrum supplement (CS) containing defibrinated bovine plasma or a colostrum replacer (CR) containing an immunoglobulin concentrate obtained by concentrating the immunoglobulin (Ig)G fraction of bovine plasma. The CS and CR contained 11.1 and 21.2% of dry matter as IgG, respectively. Each animal was fed two 454-g feedings at 1 and 8 h of age. The two feedings of CS and CR provided 95 and 187 g of IgG, respectively. Mean plasma IgG at 24 h of age was 8.0 and 13.6 g/L in calves fed CS and CR, respectively, indicating acceptable absorption of Ig from both sources. Mean apparent efficiency of IgG absorption in calves fed CS and CR were 33 and 30%, respectively, and did not differ between treatments. Mean plasma total protein at 24 h in calves fed CS and CR were 4.99 and 4.98 g/dl and did not differ between treatments. Increased plasma protein concentration from 0 to 24 h (4.5 g/L) was lower than the mean increase in plasma IgG concentration during the same period (10.3 g/L), indicating altered protein profile in the blood during the first 24 h of life. Correlation between plasma IgG and total protein at 24 h of age was significant within treatment, but the relationship between IgG and protein in plasma at 24 h varied between treatments. Predicted plasma total protein concentrations at 10 g of IgG/L of plasma at 24 h were 5.4 and 4.2 g/dl, in calves fed CS and CR, respectively. Prediction of plasma IgG concentration using total plasma protein may be inappropriate when calves are fed CS or CR.  相似文献

为有效延长即食小龙虾的货架期,探讨了4种不同复合包装袋(PA/CPP、PA/PE/RCP、PET/AL/NY/CPP、PET/AL/PA/CPP)对30℃贮藏温度下即食小龙虾(110℃反压杀菌20min)品质的影响。将即食小龙虾进行真空包装后,以感官特性、理化性能、微生物等作为评价指标,比较1个月内不同复合包装袋对即食小龙虾品质的影响。结果表明:在贮藏期间,PET/AL/NY/CPP复合袋的感官评分最优且始终大于5分;4种复合袋包装的即食小龙虾的硬度和咀嚼性值均随储存时间的延长而显著降低(p<0.05),但是弹性和回复性差异不显著(p>0.05);PET/AL/PA/CPP复合袋包装的即食小龙虾具有较低的TBA值,即脂肪氧化程度低于其他3种复合包装袋的;PA/CPP复合袋包装的即食小龙虾在贮藏后期感官评分低于不可接受值5分,其保鲜效果在4种复合包装袋中最差。  相似文献

At least 2 basic inputs are needed to formulate rations: the nutritional requirements of the animals to be fed and the nutritional composition of the feeds. David R. Mertens not only defined fiber requirements for dairy cattle but became a leading expert in the laboratory measurement of fiber in feeds, digesta, and feces. Fiber is a heterogeneous nutritional entity composed mainly of polysaccharides and polyphenolics. Because the method defines the fiber that is measured, methods must be described thoroughly and followed exactly to obtain results that are repeatable within a laboratory and reproducible among others. Filtration of neutral detergent fiber (NDF) can be difficult, and those who have worked in his laboratory can attest that Mertens rigorously studied this, along with other method details to improve NDF analysis from sample preparation to blank corrections. Mertens's procedure for amylase-treated NDF (aNDF), using α-amylase and sodium sulfite with crucibles, culminated in the Association of Official Analytical Chemists Official Method 2002.04 for aNDF, which was also accepted as International Standard ISO 16472:2006 and is used worldwide as a reference method for feed evaluation. Because aNDF digestibility is variable and a key factor in overall digestibility, Mertens also worked to improve in vitro ruminal digestibility and gas production procedures within and among laboratories, including procedures using flasks or filter bags. His in vitro gas production method is currently used by commercial laboratories that generate a significant share of the aNDF digestibility results reported worldwide. Outside of the laboratory, his extensive outreach to commercial and research laboratories has had a huge impact on fiber analysis, in vitro digestibility, and other laboratory procedures. While advising the National Forage Testing Association, Mertens provided program infrastructure that improved laboratory proficiency in more than 120 laboratories in the United States and around the world. Most importantly, thanks to his advances in fiber analysis and in vitro digestibility techniques, Mertens has enhanced the evaluation of feeds and the nutrition and health of dairy cows. These contributions have helped thousands of dairy farmers and nutritionists around the globe and continue to have a substantial impact on the industry.  相似文献

Limited information exists on postharvest preservation strategies of stored wheat in Ethiopia. The present study was conducted to evaluate the effectiveness of on-the shelf postharvest storage strategies of stored wheat in the country. The experiment consisted of eight treatments; (1) metal silos, (2) Purdue Improved Crop Storage (PICS) bags, (3) Super GrainPro bags, (4) industrial filter cake dust applied to wheat in polypropylene bag, (5) plastic drums, 6) Triplex applied to wheat in polypropylene bag, 7) Triplex applied to wheat in plastic drum, and 8) polypropylene bag as control. Measurements of oxygen and carbon dioxide levels, live adults of insects per kg, percentage seed damage, and percentage of weight loss, germination and seedling vigor were determined every two months for six months. Results indicated that storage strategies such as PICS and Super GrainPro bags, filter cake, Triplex, and plastic drums led to a significantly lower live insect density compared to the control. Besides, Triplex and filter cake dust or use of hermetic bags also resulted in a significantly lower rate of seed weight loss (%) compared to the control. After six months of storage, means ± SD germination of seed from the polypropylene bag (control) had decreased to 68.0 ± 6.1% while wheat in all other storage strategies exhibited means ± SD germination capacity ranging from 92.0 ± 3.6% to 98.0 ± 1.0%. The present results demonstrate the potential of preserving stored wheat without relying on synthetic insecticides by using hermetic bags, filter cake, and Triplex, with advantages over traditional strategies used by smallholder farmers in Ethiopia. We recommend that hermetic bags, filter cake dust, and Triplex powder should be promoted for use by farmers for the postharvest preservation of their stored wheat.  相似文献

Circulating levels of vitamin A (retinol) and lactoferrin (Lf) are low in calves at birth. Bovine colostrum contains relatively high amounts of vitamin A and Lf, and both substances are intestinally absorbed by neonatal calves. There is evidence that these compounds interact with insulin-like growth factor binding proteins and thus influence the status and effects of insulin-like growth factor. The hypothesis was therefore tested that vitamin A and Lf influence epithelial growth, development, and absorptive capacity of the small and large intestine and modulate intestinal immune tissues (Peyer's patches; PP). Four groups of calves (n = 7 per group) were fed a milk-based formula with or without vitamin A and (or) Lf. Group F received formula (F) only; group F(A) was fed F supplemented with vitamin A; group F(L) was fed F supplemented with Lf, and group F(AL) received F plus vitamin A plus Lf. An additional group of calves (group C; n = 7) served as positive control and was fed colostrum (C) from pooled milk obtained on d 1, 2, and 3 of lactation. Amounts of nutritive components in formula and colostrum were similar. Blood samples were taken to measure vitamin A and Lf, and plasma xylose (added on d 4 to feeds) was measured postprandially for 8 h as a marker of intestinal absorptive capacity. Plasma vitamin A was low at birth and further decreased in groups F and F(L), but increased in groups F(A), F(AL), and C. Plasma Lf was low at birth and transiently increased up to 4 h after the first meal in group C. Xylose absorption was higher in group C than in other groups. Incorporation of 5-bromo-2'-deoxyuridine into DNA (as a measure of cell proliferation rate) was enhanced in intestinal crypts in groups F and F(L) at all intestinal sites. Ileum villus heights of groups F and F(L) were smaller than of groups F(A) and F(AL). Villus height to crypt depth ratios were smaller in F-fed groups (especially in groups F and F(L)) than in C-fed calves in the duodenum and jejunum. Incorporation of 5-bromo-2'-deoxyuridine into colon crypt cells of group F was greater than in groups F(L) and F(A). Sizes of follicles of PP in the ileum were greater in group F(A) than in group F. In the ileum, vitamin A and Lf tended to interact with PP size. In conclusion, feed supplementation of vitamin A and Lf influenced growth of the ileum and colon. Interactions were observed between vitamin A and Lf on epithelial cell maturation, villus growth, and size of follicles in PP of neonatal calves.  相似文献

Provision of an adequate mass of IgG from maternal colostrum is essential to health and survival of neonatal calves. Colostrum supplements (CS) have been developed to provide supplemental immunoglobulin when maternal colostrum is of poor quality. However, colostrum replacers (CR) that provide > or = 100 g of IgG have not been formulated. Our objective was to determine the absorption of IgG in newborn calves fed CS derived from bovine serum or CR derived from bovine immunoglobulin concentrate. The CS were prepared by collecting, processing, and spray drying bovine serum and blending with other ingredients to provide 45 to 50 g of IgG per dose. The CR were prepared by further processing bovine serum to increase IgG concentration to > 50% IgG and blending with other ingredients to provide 100 to 122 g of IgG per dose. Holstein calves (n = 160) were fed 90 to 244 g of IgG from CS or CR in 1 or 2 feedings in two experiments. Blood was collected from each calf by jugular venipuncture at 0 and 24 h of age and plasma IgG was determined by turbidimetric immunoassay. Apparent efficiency of IgG absorption was calculated. Plasma IgG concentrations at 24 h of age were indicative of IgG intake and averaged 5.5 to 14.1 g/L in calves fed CS and CR. Mean apparent efficiency of IgG absorption in calves fed CS was 25 and 28% in experiments 1 and 2, respectively. Mean apparent efficiency of IgG absorption in calves fed CR ranged from 19 to 32% and were affected by method of processing and number of times fed. Treatment of plasma with polyethylene glycol reduced the efficiency of IgG absorption in experiment 1. The addition of animal fat to CR had no effect on IgG absorption. A second feeding of CR increased plasma IgG, but efficiency of absorption was reduced. Mean body weights at 60 d of age were not affected by treatment and ranged from 64.3 to 78.2 kg. Plasma IgG concentration in calves fed > or = 122 g of IgG from Ig concentrate approached (9.9 g/L) or exceeded 10 g/L, indicating successful transfer of passive immunity. Provision of IgG to prevent failure of passive transfer is possible with CR containing >20% IgG when fed at 454 g per dose.  相似文献

The objective of this study was to characterize absorption of colostrum replacer (CR) or supplement (CS) containing fractions of bovine plasma. Immunoglobulin concentrate (IGC) was prepared from bovine blood to a final purity of approximately 90% IgG. Blood was also processed to produce a fraction containing elevated concentrations of IGF-I and TGF-beta (GF). Both IGC and GF were spray-dried and blended with other ingredients to produce CR (30% IgG) or CS (15% IgG) containing 0 or 5% GF. Holstein bull calves were assigned to one of five treatments: 1) Pooled colostrum (MC)--1.9 L of pooled colostrum at 1 and 8 h of age to provide over 150 g of IgG; 2) Low supplement (LC)--1.9 L of CS at 1 and 8 h of age to provide 150 g of IgG; 3) Low supplement + GF (LG)--1.9 L of a CS with GF at 1 and 8 h of age to provide 150 g of IgG; 4) High supplement (HC)--1.9 L of CR at 1 h of age to provide 150 g of IgG and 1.9 L of a commercial milk replacer (MR) at 8 h of age; and 5) High supplement + GF (HG)--1.9 L of a CR with GF at 1 h of age to provide 150 g of IgG and 1.9 L of MR at 8 h of age. Apparent efficiency of IgG absorption was higher for calves fed HC and HG compared with those fed LC and LG and was lower for calves fed LG and HG compared with those fed LC and HC. IgG concentrations at 24 h were highest in calves fed MC compared with other calves and were higher in calves fed HC and HG compared to LC and LG. Calves fed LG and HG had lower IgG concentrations at 24 h of age compared with those fed LC and HC. These results indicate that 150 g of IgG provided in one dose soon after birth is superior to 150 g of IgG fed in two doses 7 h apart. Also, addition of a plasma fraction containing elevated concentrations of IGF-I and TGF-beta decreased IgG absorption in the neonatal calf.  相似文献
