Nine Holstein cows were injected bi-weekly with a prolonged-release formulation of N-methionyl bST, and 9 cows were injected with excipient. Intramuscular injections began at 60 +/- 3 d postpartum and continued at 14-d intervals for the full lactation. Administration of bST increased production of milk, total fat, and all milk fat components measured. Average fatty acid composition of milk fat was not influenced by bST treatment. Stage of lactation had a large influence on production and percentage of individual fatty acids in milk fat from both bST-treated and control cows. The stage of lactation impact on the fatty acid composition of milk fat reflected changes in the relative contributions of body fat mobilization and de novo synthesis of milk fat components in response to changes in energy balance. Initiation of bST treatment caused some transient changes in milk fatty acid composition that were related to energy balance. These changes were small compared with the normal changes because of stage of lactation in all cows. Phospholipid and cholesterol content of milk also changed with stage of lactation but were not influenced by bST treatment. Melting properties of milk fat were influenced greatly by stage of lactation. Bovine somatotropin did not cause any changes in composition or physical properties of milk fat that were outside the range of normal variation. 相似文献

Milk production, feed efficiency, health, and reproduction were evaluated in 46 Jersey cows that received either 500 mg of sometribove (n-methionyl bST) in a prolonged-release formulation or an equivalent volume of excipient bi-weekly beginning at 60 +/- 3 DIM. Cows were fed a TMR for ad libitum intake and were milked twice daily. Treatment with sometribove increased 3.5% FCM production 5.3 kg/d (31.4%) over controls. Milk composition was not changed, except that milk P content averaged 51 ppm higher in bST-treated cows. Net energy intake was 4.8 Mcal/d (22.9%) higher in the bST-treated cows than in the controls. General health of all cows was good throughout the study, but the cows treated with bST had more cases of mastitis than did the control cows. The bST treatment produced localized reactions at the site of injection in some cows, but these reactions did not affect milk production. Sometribove treatment had no effect on reproductive performance; 85% of the treated and 100% of the control cows calved successfully. Treatment with bST for a full lactation did not affect performance during the initial 60 d of the subsequent lactation. 相似文献

Eighty Holstein cows (first, second, or third lactation) were used to evaluate the efficacy of a prolonged-release formulation for sometribove (n-methionyl bovine somatotropin). Cows were fed ad libitum a complete mixed diet and milked twice daily. Cows were allocated randomly by parity to two treatment groups receiving 500 mg sometribove in a prolonged release formulation or excipient at 14-d intervals starting at 60 +/- 3 d postpartum and continuing for 36 wk. Treatment with sometribove increased FCM yield to a similar extent (11.4%, +3.1 kg/d) in primiparous and multiparous animals. Milk content of lactose, fat, ash, and Ca was not affected, but protein and phosphorous were slightly greater (less than 5%) in milk from sometribove-treated cows. Within a 14-d injection interval, animals treated with sometribove displayed a cyclic pattern in milk yield, but a similar pattern was not evident in feed intake. 相似文献

Effect of long-term bovine somatotropin (sometribove) treatment on nitrogen (protein) distribution in Jersey milk 总被引:1,自引:0,他引:1
Twenty-six Jersey cows were assigned randomly to one of two treatments. Twelve cows received biweekly subcutaneous injection of 500 mg of sometribove, USAN (recombinant methionyl bovine somatotropin), beginning 60 +/- 3 d postpartum and continuing throughout one lactation. Fourteen control animals received injections of placebo carrier. Milk samples were taken biweekly on weeks alternate to injection when differences in milk components were expected to be greatest compared with controls. The milk samples were analyzed for total nitrogen, noncasein nitrogen, and non-protein nitrogen. The average SCC for control and treatment groups was 44,000 +/- 47,000 and 56,000 +/- 65,000. Milk from sometribove-treated cows was significantly lower in total protein (3.92, 4.12%), true protein (3.74, 3.95%), and casein (3.11, 3.34%) than that from control cows on d 8 of the 14 d injection cycle. Casein as a percentage of true protein was lower (83.38, 84.52%), and non-protein nitrogen as a percentage of total nitrogen was higher (4.61, 4.26%) in milk from treated cows. The theoretical yield of Cheddar cheese was ca. .07% less for milk from treated cows than from control cows due to ca. 1% less casein as a percentage of true protein in the former. The differences in nitrogen distribution represent the response during the middle of the injection cycle when milk output was the highest and milk protein the lowest rather than the average response for the injection cycle. Milk production of dairy cows in 14 herds was increased by 3.8-32.1% by the administration of recombinant methionyl bovine somatotropin (bST) in a sustained-release vehicle at 14 d intervals at 40-94 d post partum. A greater response in multiparous than in primiparous cows was found in cows turned out to graze spring pasture. Administration of recombinant bST resulted in elevated plasma bST during the first 9 d after injection. Clinical characteristics such as respiration, heart rate and body temperature were unaffected by bST treatment, as were blood erythrocyte and leucocyte counts, haemoglobin concentrations and haematocrit values. Plasma levels of glucose, free fatty acids, urea and P, and serum alanine aminotransferase and aspartate aminotransferase were not affected by bST treatment, and acetone was not detected. No adverse effects of bST on general health, infection status of mammary glands, mastitis incidence and reproduction were found. 相似文献

The health of dairy cows given bovine somatotropin (bST) for one lactation was evaluated in 28 commercial herds located in four regions of the United States. At least six herds were in a region and at least one herd/region contained fewer than 60 cows. Cows (n = 1213) were assigned randomly to control or bST groups and were treated beginning in wk 9 to 10 of lactation and every 14 d until dry-off or d 400 of lactation. Management was according to site practices. Cows were observed for health-related signs by farm personnel daily and by the herd veterinarian biweekly. Average 305-d test-day milk yields were 932 kg greater for bST-treated cows. Pregnancy rates, days open, twinning, cystic ovaries, or abortions were unaffected by treatments. Supplementation of cows with bST had no effect on total mastitis cases, total days of mastitis, duration of mastitis, or the odds ratio of a cow to develop mastitis. Cows supplemented with bST used more medications for health events other than mastitis. This usage was associated primarily with treatments for disorders of the foot and hock. Supplemented cows had a slight increase in foot disorders. There was no effect of supplementation with bST on culling from the herd or removal from study. Overall, the results confirm that label directions for bST are adequate for safe use under field conditions. All clinical signs observed in this study occur normally in dairy herds and were managed in cows supplemented with bST. 相似文献

Three studies (84, 140, and 200 d) were performed to examine the effect of injecting dairy cows with various doses (0, 320, 640, or 960 mg/28 d; 0 or 640 mg/28 d; 0, 320 mg/14 d, or 320 or 640 mg/28 d) of bST on milk production, composition, and manufacturing properties. Mean bST response among studies on milk production varied from 0 (trial 1) to 7.3% (trial 2) and from 8.5 to 14.2% (trial 3) in relation to feeding conditions. Neither milk fat nor protein percentages in milk at time of maximum response were affected by the use of bST. Distribution of casein and protein in the whey was not affected by the treatments at any time. The nature of fatty acids varied more with time after injection than with bST doses. Neither coagulation time, standard curd firmness, nor soft or pressed cheese yields were affected by the treatments. 相似文献

Thirty-two Holstein cows were assigned to four treatment groups in a continuous lactation trial to evaluate effects of daily subcutaneous injections of 0 (A), 5.15 (B), 10.3 (C), and 20.6 mg (D) of recombinantly derived bovine somatotropin monomer. Responses were milk yield and composition, component yields, net energy intake, body weight, and somatic cell counts. Treatments began in fall, 28 to 35 d postpartum, and continued for 266 d. Base ration was corn silage and concentrate. Least squares means of milk and constituent yields and gross efficiency of milk production differed among treatments, but percentage fat and protein, energy intake, body weights, and somatic cell counts did not. Least squares means of 3.5% FCM were 21.3 (A), 25.0 (B), 26.2 (C), and 28.5 kg/d (D); fat and protein yields showed similar response to treatment. Milk yields adjusted for pretreatment yields were 21.1 (A), 25.5 (B), 26.5 (C), and 29.3 kg/d (D). Orthogonal contrasts showed lower milk and constituent yields and net energy intake for control (A) than for hormone-injected cows (B, C, D), but no differences were detected between treatments B, C, and D. The protective effect of bovine somatotropin (bST) during experimental Streptococcus uberis mastitis in cows was studied. The left quarters of 10 cows were infected with 500 cfu of S. uberis O140J. Five cows were subcutaneously treated with 500 mg of recombinant bST 7 d before and after infection, and 5 control cows received the excipient. In the treated cows, total milk production significantly increased after the first and second bST treatments. After infection, milk production decreased 24 and 40% in the infected quarters, 6 and 14% in the uninfected quarters, and 15 and 28% overall for treated and control cows, respectively. In the bST group, milk production was completely restored after 3 wk, but, in the control group, total production and the production of the infected quarters remained lower than preinfection production. The increase in somatic cell count occurred earlier and more rapidly in the control group, and the return to normal values was also more rapid in these cows. The amount of bacteria in milk was higher in the control cows. Changes in milk composition, such as lactose, protein, fat, Na+, K+, and Cl-, were significantly more pronounced in the control cows. Also, clinical symptoms were more prominent in the control cows. Somatotropin protected the mammary gland from excessive production losses and compositional changes during a subsequent episode of experimentally induced Streptococcus uberis mastitis and significantly improved the normalization of production and composition, which indicates a beneficial effect on the restoration of the integrity of the blood-milk barrier. 相似文献

Effects of daily sometribove administration on milk yield and composition, body condition score, BW, and SCC were evaluated in Holstein cows milked three times daily. Lactating cows (n =104) were assigned randomly to control or sometribove-treated (25 mg/d) groups. The experimental period was 16 wk, consisting of 2-wk pretreatment, 12-wk treatment, and 2-wk posttreatment periods. All cows were injected once daily starting at 53 to 180 d postpartum, housed in free stalls, and fed one of five total mixed rations according to milk production. Body weights were measured weekly, and body condition was scored biweekly. Milk yield was recorded daily, and weekly milk samples were analyzed for fat, protein, lactose, total solids, and SCC. Milk yield and milk protein were increased 18.8% (38.6 vs. 32.5 kg/d) and 3.3% (3.1 vs. 3.0%), respectively, whereas percentage of milk fat, lactose, SNF, SCC, and BW were unaffected by treatment. Overall average body condition scores were lower for the sometribove-treated group versus control (2.2 vs. 2.4). No apparent differences in the number of cows treated for mastitis, foot rot, displaced abomasum, or lameness were observed between treatment groups. Sometribove treatment significantly enhanced milk yield (6.1 kg/d) with no apparent negative effects on health in high producing cows milked three times per day. 相似文献

Forty-eight multiparous cows were blocked by calving date and milk production and assigned randomly to a TMR formulated to contain 68 or 55% of dietary CP as ruminally degradable CP. Diets contained corn silage, alfalfa haylage, and ground corn. Supplemental CP was soybean meal for the control diet or a combination of soybean meal, expeller-processed soybean meal, and fish meal for the low degradable protein diet. Two 10-wk phases began on d 31 +/- 3 (phase 1) and 110 +/- 7 postpartum [phase 2, all cows received subcutaneous implantations of pelleted (400 mg) bST (sometribove) every 14 d]. Dietary energy, CP, ruminally degradable CP, NDF, and ADF were similar between dietary treatments. Production of FCM increased in response to bST but was not affected by dietary treatment. Cows fed the expeller-processed soybean meal and fish meal TMR produced milk that contained less milk fat in phase 1 and less milk protein content in both phases. The DMI, BW, and body condition scores were not affected by diet. Hematocrit, plasma urea N, albumin, total protein, creatinine, glucose, and serum insulin were similar between dietary treatments. Replacing soybean meal with expeller processed soybean meal and fish meal did not affect ruminal degradation of protein or milk production but decreased milk fat and protein contents. 相似文献

Forty-eight multiparous Holstein cows were used in an experiment with a 2 x 2 factorial arrangement of treatments to study interactions of bST and nutrient density during wk 5 through 20 postpartum. Main effects were bST (0 or 10.3 mg/d) and diet (control or high protein and energy). The control diet was formulated to contain 16.3% CP and 1.65 Mcal of NEL/kg of DM; the high protein and energy diet was formulated to contain 18% CP and 1.74 Mcal of NEL/kg of DM with additional energy as fat from extruded soybeans and calcium salts of fatty acids. Diet ratios were 50:25:25 for concentrate:alfalfa hay:corn silage (DM basis). Bovine somatotropin increased milk production (40.9 vs. 37.9 kg/d) and production of 4% FCM (35.7 vs. 32.3 kg/d). The high protein and energy diet increased milk production (40.3 vs. 38.5 kg/d); 4% FCM production was higher for high protein and energy plus bST (36.0 kg) than without bST (33.6 kg). Percentage of milk fat was not affected by treatment. Percentage of milk protein was reduced with the high protein and energy diet, presumably because of the added fat, but this reduction was less with high protein and energy plus bST. Dry matter intake was not significantly elevated by bST (23.1 vs. 22.8 kg/d) but was lower for high protein and energy (22.0 vs. 23.8 kg/d). Dry periods of 40 to 60 d have been an industry standard because dry periods <40 d have resulted in reduced milk yields in the subsequent lactation by 10 to 30%. However, recent research has demonstrated no production losses for cows given a 30-d dry period. The current study evaluated milk production effects of shortened or omitted dry periods for cows at mature-equivalent production >12,000 kg of milk and treated with bovine somatotropin (bST). The study used 2 commercial dairies and one university dairy and included 4 treatments. Five multiparous and 5 primiparous cows from each farm were assigned to each treatment: 1) 60-d dry period, label use of bST (60DD); 2) 30-d dry period, label use of bST (30DD); 3) continuous milking, label use of bST (CMLST); and 4) continuous milking with continuous use of bST (CMCST). Per label, bST use started at 57 to 70 d in milk and ended 14 d before drying (60DD and 30DD) or expected calving date (CMLST). In primiparous cows, average milk yields during the first 17 wk of lactation were reduced for cows on treatments 30DD, CMLST, and CMCST vs. the 60DD treatment. (38.3, 35.1, and 37.5 vs. 44.1 +/- 1.3 kg/d, respectively). For multiparous cows, respective milk yields did not differ (46.6, 43.4, 46.5, and 47.7 +/- 2.1 kg/d). Shortened or omitted dry periods may impede mammary growth in primiparous cows, resulting in reduced milk yield in the subsequent lactation. In contrast, a shortened or omitted dry period with either bST protocol did not alter production in multiparous cows treated with bST. Quality aspects of prepartum milk and colostrum require additional characterization. For multiparous cows, milk income generated for short dry periods or for continuous milking might increase their profitability. At 17 wk of the subsequent lactation, estimates of the cumulative net margins of multiparous cows on the 30DD treatment and continuous milking treatments exceeded those of cows on the 60DD treatment by 40 dollars to 60 dollars per cow. 相似文献

Eighty-two lactating Holstein cows received either one, three, or five concurrent, intramuscular injections of a unit dose (.6 g) of zinc methionyl bST (some-tribove) or five doses of the vehicle. Injections were administered at 14-d intervals from 60 d postpartum until the end of lactation or necropsy. Thirty-eight cows continued on the same treatment for a 2nd yr. Blood bST antibodies developed within the first 7 wk of treatment, and the number of cows with anti-bST binding generally declined with time. Thirteen out of 59 cows receiving bST developed binding activity > 25% (positives) during the 1st yr. At the .6-g dose level, no binding was detected after wk 15. Seven of the 13 positive cows were among the group randomly selected to continue on study during yr 2. In the 2nd yr, only 2 out of 24 bST-treated cows were positive. Binding activity was associated with the IgG fraction in serum. Binding capacities of antibodies ranged from .625 to 3.04 mg of bST/L, and affinities ranged from 1.14 x 10(8) to 3.14 x 10(8) L/mol. Cows considered to be clinically positive had performance similar to those of their herdmates having binding < 25%. No evidence of a pathologic effect of antibodies existed in treated cows, their calves, or fetuses. The presence of anti-bST antibodies did not affect milk production of the cow or growth of the calves conceived during bST treatment. 相似文献

Bovine somatotropin (bST) and exposure to long daily photoperiod increase milk yield of dairy cattle. We tested the hypothesis that long daily photoperiod and bST treatment would increase milk yield in an additive manner in lactating cows. At winter solstice, 40 lactating cows were started on a 140-d experiment; cows were greater than 70 d in milk (DIM) and were balanced for uniformity of DIM and milk yield within parity. Cows were randomly assigned to one of four treatments (10/treatment): 1) natural photoperiod, 2) natural photoperiod + bST (14 mg/d Trobest i.m.), 3) long daily photoperiod (18 h of light and 6 h of darkness/d), or 4) long daily photoperiod + bST. Long daily photoperiod increased fat-corrected milk (FCM) yield 1.9 kg/d versus natural photoperiod. Treatment with bST increased FCM 5.7 kg/d versus natural photoperiod, and long daily photoperiod + bST increased FCM 5.8 kg/d versus long daily photoperiod. Long daily photoperiod + bST was additive, increasing FCM 7.7 kg/d versus natural photoperiod. Serum somatotropin increased with bST, but not photoperiod, and bST increased serum insulin-like growth factor-I. Long daily photoperiod tended to increase prolactin; bST had no effect. Long daily photoperiod + bST increased dry matter intake (DMI) relative to natural photoperiod and natural photoperiod + bST; long daily photoperiod increased DMI relative to natural photoperiod + bST. Photoperiod had no effect on net energy balance; however, bST decreased net energy balance. Generally, body weight and milk composition did not differ among treatments. In conclusion, combination of bST with long-daily photoperiod tended to amplify the increases in milk yield observed with either treatment individually. 相似文献

Twenty-eight multiparous Holstein cows were utilized to determine effects of long-term administration of recombinant bovine somatotropin on lactational performance, body condition, body composition, hormones, blood constituents, and physiological parameters. Treatments were 0 (control), 10.3, 20.6, and 41.2 mg recombinant bovine somatotropin administered daily as subcutaneous injections beginning 4 to 5 wk postpartum and continuing for 38 wk. A total mixed diet of 28% corn silage, 22% alfalfa hay cubes, and 50% corn-soybean meal-based grain mixture was fed. Fat-corrected milk yields were increased 12 to 25% for cows treated with somatotropin as compared with controls (29.9 kg/d). Milk composition was similar among treatments. Cows receiving somatotropin consumed 4 to 10% more feed and were 11 to 17% more efficient than controls for conversion of feed to milk. Body weight changes were not significantly different among treatments, but cows receiving somatotropin gained 4 to 10% less weight than controls. Body fat (kg) of cows receiving 20.6 and 41.2 mg/d was less than that of cows receiving 10.3 mg/d or no somatotropin. Estimated weights of body protein and mineral, most blood constituents, respiratory rates, and body temperature were not affected by somatotropin administration. Plasma fatty acids were elevated and hematocrit values were reduced. Plasma insulin, serum somatotropin, and heart rate increased concurrently with somatotropin administration. 相似文献

Primiparous (n = 105) and multiparous (n = 136) Holstein cows were used to evaluate efficacy of sometribove (n-methionyl bovine somatotropin, bST) in a dose titration study. Cows were fed TMR for ad libitum intake, were milked twice daily, and were allocated randomly within parity (1 vs. 2+) to treatments of 0, 250, 500, or 750 mg bST/14 d in a prolonged-release formulation. Subcutaneous injections commenced 60 +/- 3 d postpartum and continued throughout lactation. During a standardized treatment period of 252 d, treatment with increasing dosages of bST increased 3.5% FCM yield in a dose-dependent manner for both primiparous (2.5 kg/d, 10.2%; 3.5 kg/d, 14.3%; and 5.9 kg/d, 24.1%) and multiparous cows (3.1 kg/d, 12.1%; 3.9 kg/d, 15.2%; and 6.8 kg/d, 26.5%). Milk content of fat, protein, lactose, ash, Ca and P, and SCC were not affected by treatment. Over the 252-d treatment period, voluntary intake of energy was increased in bST-treated cows such that BW gain, body condition score, and net energy balance did not differ among treatment groups. Productive efficiency (milk per unit NEL intake corrected for BW change) over the treatment period was significantly increased in a dose-dependent manner for multiparous cows (4.1, 6.8, and 11.0%). Results demonstrated that bST administered in a prolonged-release formulation was efficacious in enhancing milk production and feed efficiency. 相似文献

Eight pregnant Holstein cows were given weekly injections of 15 g of recombinant bST over a 2-wk period for a total dose of 30 g to determine signs of acute toxicity. Cows were monitored intensively throughout the study, and samples were taken for analyses of hormones, metabolites, chemistries, hematology, and urine analytes. Animal health throughout the study was generally excellent. Mean rectal temperatures were significantly higher in treated cows (38.7 vs. 39.2 degrees C). Least squares means for 3.5% FCM production were 15.9 and 23.0 kg/d, and net energy intakes were 29.4 and 26.9 Mcal/d for control and treated cows, respectively. Somatotropin concentrations reached more than 250 ng/ml on d 10 and remained above 200 ng/ml. Insulin and glucose concentrations were also increased but returned toward baseline values. Free fatty acid concentrations were higher in treated cows, but beta-hydroxybutyrate was not affected. Most hematological measurements were unaffected except for a reduction in erythrocyte number in treated cows and decreases in hematocrit and hemoglobin, but values were within clinically normal ranges. The objective was to evaluate the effect of beta-lactoglobulin (beta-lg) polymorphism and seasonality on milk composition (fat, lactose, total solids, milk urea nitrogen, total protein, true protein, casein and somatic cell counts) of Holstein and Girolando cows. Milk and blood samples from 278 Holsteins cows and 156 Girolando cows were taken during two dry seasons and two rainy seasons, for milk composition analysis and to determine beta-lg genotypes, respectively. BB genotype was the most frequent for both breeds, followed by AA genotype for Holstein (BB>AA>AB) and by AB for Girolando cows (BB>AB>AA). No differences were found in milk compositional characteristics among genetic variants of beta-lg (AA, AB and BB) either between Holstein or Girolando cows. No association between milk composition and beta-lg genetic polymorphism was observed. During the dry season, independently of the breed considered, higher contents of lactose, true protein, casein and casein:true protein ratio were found. 相似文献

Eight lactating Holstein cows were used to study the effect of bovine somatotropin on mammary lipid metabolism, milk production, and milk composition. Eight cows were injected with either excipient (n = 4) or 50 IU somatotropin/d (n = 4). A 2-wk adjustment period preceded a 10-d period when treatments were administered. On d 9, serial blood samples were collected from 2 to 5 h post-injection to determine concentrations of somatotropin and metabolites in plasma. Mammary tissue biopsies were obtained 2 to 3 h after a.m. milking on d 10 to determine lipoprotein lipase activity and in vitro rates of acetate incorporation into fatty acids and acetate oxidation by mammary tissue slices. Activity of lipoprotein lipase in milk and mammary tissue and rate of acetate metabolism by mammary tissue were not affected by treatment. The following means for placebo and somatotropin, respectively, were: milk production (21.5 and 24.8 kg/d), milk fat (3.8 and 4.0%), and 4% FCM (20.7 and 25.1 kg/d). Plasma concentrations of somatotropin (3.0 and 43.4 ng/ml) and free fatty acids (105.6 and 219.5 mu eq/L) were significantly elevated with somatotropin. The injection of somatotropin into lactating dairy cows did not affect the mechanisms of fatty acid synthesis or metabolism evaluated in this study. 相似文献
