We surveyed the total mercury (T-Hg) and methyl mercury (M-Hg) levels in red meat products (n = 160) from small cetacean species sold for human consumption in markets throughout Japan from 2000 to 2003. Genetic identification showed that the red meat products originated from nine species: false killer whale, bottlenose dolphin, short-finned pilot whale, striped dolphin, rough-toothed dolphin, Risso's dolphin, pantropical spotted dolphin, Baird's beaked whale, and Dall's porpoise. T-Hg and M-Hg concentrations in all red meat products exceeded the provisional permitted levels of T-Hg (0.4 microg/wet g) and M-Hg (0.3 microg/ wet g) in fish and shellfish set by the Japanese government, respectively. The average M-Hg level in the most contaminated species (false killer whale) was 11.5 microg/wet g, and that in the least contaminated species (Dall's porpoise) was about 1.0 microg/wet g, exceeding or equaling the Codex guideline of M-Hg in predatory fishes (1.0 microg/wet g). Contamination levels of T-Hg and M-Hg differed considerably among samples of the nine species and among individuals of a particular species. The highest M-Hg was about 26 microg/ wet g in a sample from a striped dolphin, 87-times higher than the permitted level. The consumption of only 4 g of this product would exceed the provisional tolerable weekly intake of M-Hg for someone of 60 kg body weight (1.6 microg/kg-bw/ week). Although a high correlation between T-Hg and selenium (Se) was observed in these products, the molar ratio of T-Hg to Se was substantially higher than 1. The consumption of red meat from small cetaceans, therefore, could pose a health problem for not only pregnant women but also for the general population.  相似文献

This paper presents the results of a study examining whether the flooding of pasture by rivers gives rise to higher PCDD/F and PCB concentrations in cows' milk. Over 180 milk, soil, and grass samples, taken from 38 farms across 3 different river systems (River Dee, Trent, and Doe Lea/Rother/Don) in the United Kingdom, were analyzed for PCDD/Fs and PCBs. The concentrations were compared between flood-prone farms, where the animals had access to pasture that is often flooded, and control farms where the land does not flood. The results indicated that concentrations of PCDD/Fs and PCBs in cows' milk were higher in samples taken from farms prone to flooding, but only from the river systems flowing through industrial and urban areas. Raised levels of PCDD/F and PCBs were also found in soil and grass from farms prone to flooding providing strong corroborative evidence that the higher concentrations in cows' milk from such areas is likely to be due to the ingestion of contaminated grass and soil. Overall, the results provide strong evidence that flooding of pastureland can indeed result in elevated concentrations of PCDD/Fs and PCBs in milk from the farms so affected.  相似文献

Exceeding 1.2 million acres (4856 km(2)) since the 1930s, coastal wetland loss has been the most threatening environmental problem in Louisiana, United States. This study utilized high-resolution LiDAR (Light Detection and Ranging) and DEM (Digital Elevation Model) data sets to assess the risk of potential wetland loss due to future sea level rises, their spatial distribution, and the associated loss of soil organic carbon (SOC) and organic nitrogen (SON) estimated from the State Soil Geographic (STATSGO) Database and National Wetlands Inventory (NWI) digital data. Potential inundation areas were divided into five elevation scales: < 0 cm, 0-50 cm, 50-100 cm, 100-150 cm, and 150-200 cm above mean sea level. The study found that southeastern Louisiana on the Mississippi River Delta, specifically the Pontchartrain and Barataria Basins, are most vulnerable to sea-level rise induced inundation. Accordingly, approximately 42,264,600 t of SOC and 2,817,640 t of SON would be inundated by 2050 using an average wetland SOC density (203 t per hectare) for the inundation areas between 0 and 50 cm. The estimated annual SOC and SON loss from Louisiana's coast is 17% of annual organic carbon and 6-8% of annual organic nitrogen inputs from the Mississippi River.  相似文献

Although the ubiquitous distribution of organophosphate esters (OPEs) in the environment has been documented very thoroughly, data on their occurrence in soil is so far sparse. In this study, an analytical method was developed to determine six OPEs in soil. The method consists of a combination of Twisselmann extraction and solid-phase microextraction (SPME), followed by gas chromatography-mass spectrometry (GC-MS). To develop the method, spiked soil was extracted using a Twisselmann extractor after freeze-drying. The extract was evaporated to dryness, redissolved, and filtered. A volume of 7 mL was then analyzed by SPME, followed by GC-MS. The effects of different parameters on analyte recoveries during sample preparation, e.g., solvent for Twisselmann extraction, solvent for redissolving the extract, addition of copper, and filtration of the extract, were systematically investigated. Under optimum conditions, 10 g soil samples were extracted using toluene, and the extract was redissolved in methanol/water (1:14) and filtered. It was not necessary to add copper. For TnBP, TBEP, TCPP, and TCEP, recoveries ranged from 77.0% to 89.6%. Those for TPP and TDCP were much lower, at 31.5% and 42.0%, respectively (addition level 22.9-45.8 ng g(-1)). The variability of recoveries under these conditions was between 0.3 and 16.2% (n = 3). Limits of detection (LOD) were 0.002-3 ng g(-1). When ultrasonication was used in place of Twisselmann extraction in the developed method, recoveries were three to four times lower (27.4% to 30.6%), but the variability of recoveries was below 3% (n = 3). The method was applied to quantify OPEs in soil collected from the university campus in Osnabrueck (Germany). Average concentrations (n = 6) in soil samples ranged from 1.23 ng g(-1) to 4.96 ng g(-1) (dry weight) for TCPP, TPP, and TCEP. The influence of strain directions on the mechanical properties of raw meat samples in a compression test was analysed in relation to the contracted or stretched state and discussed in terms of connective network deformation. When the lateral deformation of a sample is in the direction of muscle fibres, the more the meat has been stretched ante-rigor, the smaller the strain range in which muscle fibres can be solicited alone and the lower the critical compression ratio at which collagenous fibres come under tension. Post-rigor contraction occurring in stretched muscles modifies the relations between muscle fibres and collagenous fibres and increases the critical compression ratio. The greatest post-rigor contractions have been observed in stretched slices of Semitendinosus and Lattissimus dorsi which are high elastin content muscles. This suggests that elastin has a major role in post-rigor contraction. In greatly contracted post-rigor samples, as muscle fibres are wavy, they are not involved at low strains, likewise collagenous fibres around muscle fibres cannot be stressed until the waviness of muscle fibres has been removed. Therefore, in greatly contracted post-rigor samples, the mechanical properties observed at low strains might belong mainly to elastin fibres. When the lateral deformation is perpendicular to the direction of muscle fibres these fibres are not involved and the more the meat has been stretched ante-rigor the higher the critical compression ratio.  相似文献

A data set was generated aiming to predict the toxicity of PAHs to soil organisms. Toxicity data include the effects of 16 PAHs on the survival and reproduction of the soil-dwelling springtail Folsomia fimetaria. The results show that only PAHs with reported log Kow values < or = 5.2 (i.e., naphthalene, acenaphthene, acenaphthylene, anthracene, phenanthrene, fluorene, pyrene, and fluoranthene) significantly affected the survival or reproduction of the test organisms. Threshold values for the toxicity of the individual PAHs could be expressed as pore-water concentrations by the use of reported organic carbon-normalized soil-pore-water partitioning coefficients (Koc values). For the PAHs with a log Kow < or = 5.2, toxicity significantly increased with increasing lipophilicity of the substances (r2 = 0.67; p = 0.012; n = 8), suggesting a narcotic mode of toxic action for most substances. However, the position of anthracene in the regression plot indicated a more specific mode of toxic action than narcosis, and removing this data point yielded the following regression equation: log EC10 (micromol/L) = -0.97 log Kow + 4.0 (r2 = 0.80; p = 0.006; n = 7). Using this quantitative structure-activity relationship (QSAR) to calculate threshold values for the toxicity of the remaining nontoxic substances (benz[a]anthracene, chrysene, benzo[b]fluoranthene, benzo[k]fluoranthene, dibenz[a,h]anthracene, benzo[a]pyrene, perylene, and indeno[1,2,3-cd]pyrene), the absence of toxicity could, in most cases, be explained by a limited water solubility, indicating that these substances do act by narcosis as the mode of toxic action and that their toxicity is governed by concentrations in the pore-water. The results provide important input to future model predictions of the ecological risk posed by PAH contaminated sites.  相似文献

The distribution of organic phosphorus in relation to soil organic matter and the influence of parent material and vegetation of organic P level was studied in a number of soil profiles in the northern half of Nigeria. There was a close relationship between soil organic carbon and organic phosphorus contents of these soils. The ratios of these two ingredients were all less than 200:1. This fact suggests that organic P is likely to be mineralised. The effect of parent rock on organic P contents was slight, but there was an indication that soil profiles on metamorphic parent rocks contained more organic phosphorus than those on sedimentary parent material. Vegetation had a great influence on organic P as shown by the relatively higher organic phosphorus contents of profiles from southern Guinea savanna and the lower contents of the Sudan vegetation zone. The profiles from impeded drainage sites (fadamas) were generally richer in organic phosphorus than were those from comparable vegetation zones on well drained sites.  相似文献

Exposure of humans and wildlife to various inorganic and organometallic forms of mercury (Hg) may induce adverse health effects. While human populations in developed countries are mainly exposed to marine fish monomethylmercury (MMHg), this is not necessarily the case for developing countries and diverse indigenous people. Identification of Hg exposure sources from biomonitor media such as urine or hair would be useful in combating exposure. Here we report on the Hg stable isotope signatures and Hg speciation in human hair across different gold miner, indigenous and urban populations in Bolivia and France. We found evidence for both mass-dependent isotope fractionation (MDF) and mass-independent isotope fractionation (MIF) in all hair samples. Three limiting cases of dominant exposure to inorganic Hg (IHg), freshwater fish MMHg, and marine fish MMHg sources are used to define approximate Hg isotope source signatures. Knowing the source signatures, we then estimated Hg exposure sources for the Bolivian gold miner populations. Modeled IHg levels in hair correspond well to measured IHg concentrations (R = 0.9), demonstrating that IHg exposure sources to gold miners can be monitored in hair samples following either its chemical speciation or isotopic composition. Different MMHg and inorganic exposure levels among gold miners appear to correspond to living and working conditions, including proximity to small towns, and artisanal vs large scale mining activity.  相似文献

The variations of the total and individual catechin contents of tea were evaluated among three cultivars grown in 10 locations; the dynamic changes of chlorophyll and catechin contents in two cultivars during young leaf development were also analysed. The results showed the variations of catechin contents among locations were larger than those among cultivars. Correlation analysis indicated that (+)-catechin was the major catechin fraction significantly correlated with climate factors and chlorophyll contents. Moreover, chlorophyll contents were significantly influenced by climate factors, with the highest correlation coefficient for daily lowest temperature. Furthermore, the rise of chlorophyll a content during young leaf development was observed alongside the increase of (−)-epicatechin, (−)-epigallocatechin and the decline of (+)-catechin, suggesting chlorophyll plays a vital role in the regulation of individual catechins.  相似文献

There is a need to standardize the NDF procedure. Procedures have varied because of the use of different amylases in attempts to remove starch interference. The original Bacillus subtilis enzyme Type IIIA (XIA) no longer is available and has been replaced by a less effective enzyme. For fiber work, a new enzyme has received AOAC approval and is rapidly displacing other amylases in analytical work. This enzyme is available from Sigma (Number A3306; Sigma Chemical Co., St. Louis, MO). The original publications for NDF and ADF (43, 53) and the Agricultural Handbook 379 (14) are obsolete and of historical interest only. Up to date procedures should be followed. Triethylene glycol has replaced 2-ethoxyethanol because of reported toxicity. Considerable development in regard to fiber methods has occurred over the past 5 yr because of a redefinition of dietary fiber for man and monogastric animals that includes lignin and all polysaccharides resistant to mammalian digestive enzymes. In addition to NDF, new improved methods for total dietary fiber and nonstarch polysaccharides including pectin and beta-glucans now are available. The latter are also of interest in rumen fermentation. Unlike starch, their fermentations are like that of cellulose but faster and yield no lactic acid. Physical and biological properties of carbohydrate fractions are more important than their intrinsic composition.  相似文献

Fumonisin B1 (FB1) is an inhibitor of sphinganine N-acyltransferase and the increase in the sphinganine/sphingosine (Sa/So) ratio in urine or serum has been proposed as a biomarker to evaluate exposure to fumonisins. The objectives of this study were to (1) develop a liquid chromatographic method sufficiently sensitive to determine the low concentration of free Sa in male human urine, and (2) analyse So and Sa in human urine and monitor the Sa/So ratio in urine of humans exposed to FB1 in corn diets over 1 month. The liquid chromatographic method involved isolation from human urine of exfoliated cells followed by an extraction of free sphingoid bases and their separation and quantification by high performance liquid chromatography. The detection limits for So and Sa were 0.15ng/ml in female urine (2ml used) and 0.005ng/ml in male urine (60ml used). Twenty-eight healthy adult volunteers consumed for 1 month a normal diet containing their homegrown corn potentially contaminated with FB1. Immediately preceding the start of the test, morning urine samples for the determination of So and Sa were collected from each person, and the corn samples used in cooking were obtained from each family for the determination of FB1. At the end of the test period, morning-urine samples were collected from each person and analysed again. The daily FB1 intakes were estimated and used to assess the relationship between them and the urinary Sa/So ratios in humans exposed to dietary FB1 over 1 month. We measured the relative importance of water and food as sources of the trace elements As, Cd, Co, Cu, Pb, and Zn for larvae of the benthic insect Sialis velata. To achieve this end, we first exposed its prey, the midge Chironomus riparius, to trace elements in a contaminated lake located near a nonferrous metal smelter in northwestern Quebec. Concentrations of five of the six trace elements reached a steady state in prey within one week. When predators were exposed to these contaminated prey, their trace element concentrations increased markedly for all of the trace elements studied. In contrast, exposure of S. velata to contaminated lake water led to no significant increase in their concentrations of five of the six trace elements ([Pb] increased only slightly). Additional treatments in which the predator was exposed to these trace elements in both food and water or in a combination of food and water and sediment did not lead to further increases in its contaminant concentrations beyond those measured in the food-only treatment. We conclude that food (prey) is the almost exclusive source of As, Cd, Co, Cu, and Zn for S. velata and that it is the source of 94% of its Pb. Model curves for trace element accumulation over time by the predator fit our measured data well, and trace element concentrations in food alone are sufficient to model the exchange of these contaminants in this insect.  相似文献

The bio-nutritional potentials of three tropical vegetable leaf meals (Telfairia occidentalis leaf meal, TOLM; Talinum triangulare leaf meal, TTLM and Amaranthus cruentus leaf meal, ACLM) were investigated using albino rat as the test animal. Some protein quality evaluation indices were measured when the three vegetable leaf meals (VLMs) were used as sole protein sources in diets fed to the experimental animals and results were compared with data obtained for a basal nitrogen free diet (diet 1) and another reference diet (diet 2) in which the protein was solely supplied by nutritional casein (pure protein). The weight gain value recorded over a 10 day experimental period for the test animals on the reference (casein) diet 2 was consistently higher (P < 0.05) than the weight gain value obtained for the animals on the 3 VLMs diets (diets 3–5). Feed intake values recorded for the rats on the VLMs were similar (P > 0.05) and significantly higher (P < 0.05) than the value obtained for rats on the reference (casein) diet. Nitrogen excreted in faeces (feacal nitrogen) was lowest (P < 0.05) for the animals in reference diet 2. However, the nitrogen excreted in urine (urinary nitrogen) was highest (P < 0.05) for the animals on reference diet 2 and lowest for animals on diet 3 (TOLM diet) (P < 0.05). The nitrogen retention (NR) value obtained for the test animals on the reference diet 2 was similar (P > 0.05) to the value obtained for animals on diet 3 (TOLM). These values were significantly higher (P > 0.05) than NR values obtained for animals on diets 4 (TTLM) and 5 (ACLM). Expectedly, the apparent nitrogen digestibility (AND), protein efficiency ratio (PER), net protein ratio (NPR), true digestibility (TD), biological value (BV) and net protein utilization (NPU) all indicated higher and better values (P < 0.05) than the corresponding values obtained for the 3 VLMs diets (diets 3–5).  相似文献

The objectives of this work were to determine the effects of the concentrations of glycerol and sorbitol (as hydrophilic plasticizers), Tween 80 (as surfactant) and chitosan on the wettability of Cuban chitosan-based edible coatings in view of their application on tomato and carrot and to develop a model allowing the optimization of coating composition.The values of the polar and dispersive components of the superficial tension of the foods were determined to be 3.04 and 25.67 mN/m, respectively, for tomato, and 0.34 and 26.13 mN/m, respectively, for carrot, the sum of the two components being the superficial tensions of tomato and carrot (28.71 and 26.48 mN/m, respectively). The skins of both foods are therefore low-energy surfaces, meaning that the Zisman method for the determination of wettability could be applied.The best experimental values of wettability were obtained for the following coating composition: 1.5% (w/v) of chitosan and 0.1% (w/w) of Tween 80.The increase in the concentration of chitosan and glycerol or sorbitol as plasticizers decreased the values of wettability and adhesion coefficients.The results of wettability were adjusted to a polynomial model that describes the dependence of the adhesion coefficient (Wa), cohesion coefficient (Wc) and spreading coefficient (Ws) on chitosan and Tween 80 concentrations. The optima calculated by the model equations were in excellent agreement (relative error below 3%) with the experimental values.  相似文献

The analytical method for determining the concentration of mercury in fish by thermal decomposition, amalgamation/ atomic absorption spectrophotometry was thoroughly studied. Specific issues addressed were accurate modeling of instrumental response, the use of quartz and nickel boats, carryover effects, software limitations, and troubleshooting. The DMA-80 Direct Mercury Analyzer instrument was calibrated using a total of 22 points, and the resultant curves statistically analyzed. At minimum, second-order polynomials were required to adequately model the data. TORT-2 standard reference material was analyzed in both quartz and nickel boats and found to give equivalent performance in both types of vessels and well within the 95% confidence interval. DOLT-3 standard reference material also yielded values well within the 95% confidence interval, but the DORM-2 standard reference material did not. Carryover effects were found to be minimal with a new catalyst tube but increased with catalyst age. Blanks should be run after the analysis of high mercury content samples; however, when the catalyst has aged, two blanks are required to reduce apparent mercury signals to nominal blank values. Comparable results between thermal decomposition, amalgamation/atomic absorption spectrophotometry and cold-vapor atomic absorption spectrophotometry were demonstrated. The feasibility of using this instrument to analyze hair was also explored and found to be suitable. Software problems and limitations have been noted when attempting to implement a high-throughput methodology. Instrumental drift was found to be minimal when operated over long periods. Blank values can provide important diagnostic indicators.  相似文献

Six open reading frames (ORFs) from chromosome, VII, YGL131c, YGL129c, YGL128c, YGL125w, YGL124c and YGL121c, were disrupted by deletion cassettes with short flanking regions homologous to the target locus (SFH). YGL129c is necessary for growth in non-fermentable carbon sources, YGL128c for growth at low or high temperatures and YGL125w is implicated in the biosynthesis of methionine. With regard to the other ORFs, basic phenotypic analyses did not reveal any significant clues about their function.  相似文献

Twenty-three Cabernet Sauvignon wines from the Mudgee region and thirty-two Shiraz wines from the Hunter Valley region were analyzed for phenolic content and antioxidant activity. Concentrations of (+)-catechin, quercetin, and transresveratrol, total phenolic content, and DPPH antioxidant activity varied considerably, both within and between varieties. Individual phenols, total phenols, and antioxidant activity were correlated with price and vintage. Shiraz wines showed positive and significant correlations for catechin and quercetin concentrations with total phenols, antioxidant activity, and vintage; and for total phenols and antioxidant activity with vintage. Cabernet Sauvignon wines showed positive and significant correlations for quercetin concentration with total phenols and antioxidant activity. There was a negative and significant correlation found between price and antioxidant activity for Cabernet Sauvignon wines. Results are discussed in terms of the potential for wine to be considered a functional food. PRACTICAL APPLICATION: We report on potential health benefits (antioxidant activity) of 55 wines typical of 2 geographically close, but distinct, wine regions of Australia. Our results highlight the variability in functional components as an issue that needs further research and consideration in relation to wine as a functional food. The price of studied wines is not reflective of their health functionality, based on antioxidant activities.  相似文献

This study assessed rural mothers' knowledge of, attitudes toward, and use of soybeans as compared with cowpeas for feeding their young children. Mothers who had at least one child below the age of five years in 239 sampled households were interviewed. The results indicate that mothers were well informed about the value of giving both foods to their young children. A comparison of attitudes shows that the mothers had a more favorable opinion of cowpeas and that they used cowpeas more than soybeans. Mothers' reasons for using both foods include nutritive value, affordability, and palatability, in that order. Cowpeas were favored for ease of preparation. More mothers introduced cowpeas to their children before the age of six than soybeans.  相似文献

The influence of exopolysaccharides (EPS), pre-acidification and use of two fat replacers, FR1 and FR2, on the textural and functional characteristics of Mozzarella cheeses were studied. Control cheeses made with EPS producing Streptococcus thermophilus 285 had the lowest moisture (52.84%) and yield while hardness and melt properties of these cheeses improved with storage. Addition of FR1 or FR2 and pre-acidification of milk increased the moisture content in cheeses. All cheeses were softer than the control. Cheeses made with FR1 exhibited the greatest stretch and melt; those made with FR2 showed poor stretch and melt. Each cheese showed a unique microstructure prior to, and after the hydration of the protein matrix. The rate of α- and β-casein proteolysis was greater in cheeses made with pre-acidified milk and those having higher moisture content. Pizza bake performance was improved with FR1, but FR2 cheeses had the least favourable bake characteristics. Coating cheese shreds with oil was necessary for adequate browning, melt and flow.  相似文献
