Comb‐type grafted hydrogels composed of polyacrylamide (PAAm) and chitosan (CT) were prepared and used for DNA adsorption. Instead of direct grafting of the acrylamide monomer onto the CT chain, semitelechelic PAAm with carboxylic acid end groups (PAAm–COOH) was synthesized by free‐radical polymerization with mercaptoacetic acid as the chain‐transfer agent, and it was grafted onto CT with amino groups. The synthesis of telechelic PAAm–COOH and the formation of comb‐type grafted hydrogels were confirmed by attenuated total reflectance/Fourier transform infrared spectroscopy and scanning electron microscopy measurements. The prepared comb‐type grafted hydrogels were used as sorbents in DNA adsorption experiments conducted at +4°C in a tris(hydroxymethyl)aminomethane/ethylenediaminetetraacetic acid solution of pH 7.4. DNA adsorption capacities as high as 2.0 × 103 μg of DNA/g of dry gel could be achieved by the comb‐type hydrogels with higher PAAm contents. This value was approximately 6 times higher than that of CT alone. In addition, the comb‐type hydrogels showed a high adsorption/desorption rate depending on the PAAm content in the hydrogel. As a result, these comb‐type hydrogels carrying higher amounts of DNA may be considered good candidates for achieving higher removal rates for anti‐DNA antibodies and for effective gene therapy systems. © 2008 Wiley Periodicals, Inc. The hydrogels composed of chitosan and eugenol were prepared to enhance and sustain antioxidant activities. The vinyl groups of eugenol monomer were directly grafted on the amino groups of chitosan, using ceric ammonium nitrate. The graft of eugenol onto chitosan was confirmed by using Fourier‐transform infrared and proton nuclear magnetic resonance spectroscopies. Results from the swelling behavior, thermal stability, and wide‐angle X‐ray diffraction revealed that the equilibrium water content decreased with increase of graft yields, because of the hydrophobicity of eugenol, although the introduction of eugenol as a side chain disturbed the ordered arrangement of chitosan's crystalline structure. The eugenol‐grafted chitosan hydrogels showed lower pH sensitivity in comparison with chitosan alone, because the amino groups, which were pH sensitive, of chitosan were grafted with eugenol. The scavenging activity of the tested hydrogels increased with graft yield of eugenol, because phenolic groups in the eugenol could play a major role as potent free‐radical terminators, in the results of improved antioxidant activity in eugenol‐grafted chitosan hydrogel in comparison with chitosan alone. © 2006 Wiley Periodicals, Inc. Graft copolymers of crosslinked poly(N‐isopropyl acrylamide) (PNIPAAm) and homopolyguluronic acid (GG) and homopolymannuronic acid (MM) fractions of alginic acid were synthesized. MM and GG block fractions were obtained by partial acid hydrolysis of the alkaline extract from the brown seaweed Macrocystis pyrifera. The conjugation of these block fractions with the synthetic polymer was achieved by amidation with crosslinked PNIPAAm functionalized with an amino group at the end of the polymer chain. The structure of conjugates was determined by Fourier transform infrared and NMR spectroscopy. Atomic force microscopy of the graft copolymer GG‐g‐PNIPAAm showed a regular porous pattern, whereas the MM‐g‐PNIPAAm graft copolymer showed a regular netlike structure. Aqueous solutions of the synthesized graft copolymers afforded hydrogels by stirring with 0.1M CaCl2. The hydrogels showed a well‐defined stimulus–response to temperature and pH. The swelling, thermal, and pH characterizations demonstrated the superior properties of the GG‐g‐PNIPAAm hydrogel over the MM‐g‐PNIPAAm hydrogel. © 2015 Wiley Periodicals, Inc. J. Appl. Polym. In this study, pH‐ and temperature‐responsive hydrogels based on linear sodium alginate (SA) and crosslinked poly(N‐isopropylacrylamide) (PNIPAAm) were prepared by semi‐interpenetrating network (semi‐IPN) technique. The dually responsive hydrogels were characterized by FTIR, DSC, and SEM, and their temperature‐ and pH‐responsive behaviors were investigated by measuring equilibrium swelling ratios and pulsatile swelling experiments. The results showed that these hydrogels underwent volume phase transition at around 33°C irrespective of the pH value of the medium, but their pH sensitivity was evident only below their volume phase transition temperature. Under basic conditions, the swelling ratios of SA/PNIPAAm semi‐IPN hydrogels were greater than that of pure PNIPAAm hydrogel and increased with increasing SA content incorporated into the hydrogels, but the case was inverse under acidic conditions. The pulsatile swelling experiments indicated that the higher the SA content in SA/PNIPAAm semi‐IPN hydrogels, the faster the response rate to both pH and temperature change. © 2005 Wiley Periodicals, Inc. Amino semitelechelic poly(N‐isopropylacrylamide) (PNIPAAm) was prepared by radical polymerization with aminoethanethiol hydrochloride as a chain‐transfer agent. Semi‐interpenetrating polymer network (semi‐IPN) hydrogels, composed of alginate and amine‐terminated PNIPAAm, were prepared by crosslinking with calcium chloride. From the swelling behaviors of semi‐IPNs at various pH's and Fourier transform infrared spectra at high temperatures, the formation of a polyelectrolyte complex was confirmed from the reaction between carboxyl groups in alginate and amino groups in modified PNIPAAm. Semi‐IPN hydrogels reached an equilibrium swelling state within 24 h. The water state in hydrogels, investigated by differential scanning calorimetry, showed that sample CAN55 [alginate/PNIPAAm (w/w) = 50/50] exhibited the lowest equilibrium water content and free water content among the hydrogels tested, which was attributed to its more compact structure compared to other samples and the high content of interchain bonding within the hydrogels. Alginate/PNIPAAm semi‐IPN hydrogels exhibited a reasonable sensitivity to the temperature, pH, and ionic strength of swelling medium. © 2002 Wiley Periodicals, Inc. Thermo‐ and pH‐sensitive polymers were prepared by graft polymerization or blending of chitosan and poly(N‐isopropylacrylamide) (PNIPAAm). The graft copolymer and blend were characterized by Fourier transform‐infrared, thermogravimetric analysis, X‐ray diffraction measurements, and solubility test. The maximum grafting (%) of chitosan‐g‐(N‐isopropylacrylamide) (NIPAAm) was obtained at the 0.5 M NIPAAm monomer concentration, 2 × 10−3 M of ceric ammonium nitrate initiator and 2 h of reaction time at 25°C. The percentage of grafting (%) and the efficiency of grafting (%) gradually increased with the concentration of NIPAAm up to 0.5 M, and then decreased at above 0.5 M NIPAAm concentration due to the increase in the homopolymerization of NIPAAm. Both crosslinked chitosan‐g‐NIPAAm and chitosan/PNIPAAm blend reached an equilibrium state within 30 min. The equilibrium water content of all IPN samples dropped sharply at pH > 6 and temperature > 30°C. In the buffer solutions of various pH and temperature, the chitosan/PNIPAAm blend IPN has a somewhat higher swelling than that of the chitosan‐g‐NIPAAm IPN. © 2000 John Wiley & Sons, Inc. In this study, poly(N,N‐dimethylacrylamide) grafted chitosan (PDMAAm‐g‐CT) hydrogels were prepared for deoxyribonucleic acid (DNA) adsorption. Instead of directly grafting the N,N‐dimethylacrylamide (DMAAm) monomer onto the chitosan (CT) chains, poly(N,N‐dimethylacrylamide) with carboxylic acid end group (PDMAAm‐COOH) was firstly synthesized by free‐radical polymerization using mercaptoacetic acid (MAAc) as the chain‐transfer agent and then grafted onto the CT having amino groups. The synthesis of PDMAAm‐COOH and its grafting onto the CT chains were confirmed by attenuated total reflectance Fourier transform infrared (ATR‐FTIR) spectroscopy. From gel permeation chromatography measurements, the number‐average molecular weight (M n) and polydispersity index of PDMAAm‐COOH were found as 2400 g/mol and 2.3, respectively. The PDMAAm‐g‐CT hydrogels were utilized as the adsorbents in DNA adsorption experiments conducted at +4°C in a trisEDTA solution of pH 7.4. The hydrogels produced with higher PDMAAm‐COOH content exhibited higher DNA adsorption capacity. The DNA adsorption capacity up to 4620 μg DNA/g dry gel could be achieved with the PDMAAm‐g‐CT hydrogels prepared in 80.0 wt % PDMAAm‐COOH feed concentration. Hydrogels have been widely used as mild biomaterials due to their bio‐affinity, high drug loading capability and controllable release profiles. However, hydrogel‐based carriers are greatly limited for the delivery of hydrophobic payloads due to the lack of hydrophobic binding sites. Herein, nano‐liposome micelles were embedded in semi‐interpenetrating poly[(N‐isopropylacrylamide)‐co‐chitosan] (PNIPAAm‐co‐CS) and poly[(N‐isopropylacrylamide)‐co‐(sodium alginate)] (PNIPAAm‐co‐SA) hydrogels which were responsive to both temperature and pH, thereby establishing tunable nanocomposite hydrogel delivery systems. Nano‐micelles formed via the self‐assembly of phospholipid could serve as the link between hydrophobic drug and hydrophilic hydrogel due to their special amphiphilic structure. The results of transmission and scanning electron microscopies and infrared spectroscopy showed that the porous hydrogels were successfully fabricated and the liposomes encapsulated with baicalein could be well contained in the network. In addition, the experimental results of response release in vitro revealed that the smart hydrogels showed different degree of sensitiveness under different pH and temperature stimuli. The results of the study demonstrate that combining PNIPAAm‐co‐SA and PNIPAAm‐co‐CS hydrogels with liposomes encapsulated with hydrophobic drugs is a feasible method for hydrophobic drug delivery and have potential application prospects in the medical field. © 2018 Society of Chemical Industry  相似文献

Novel poly(N‐isopropylacrylamide) (PNIPAAm)/chitosan (CS) semi‐interpenetrating polymer network hydrogel particles were prepared by inverse suspension polymerization. The prepared particles were sensitive to both temperature and pH, and they had good reversibility in solution at different temperatures and pH values. The swelling ratios of PNIPAAm/CS hydrogel particles decreased slightly with the addition of CS, which did not shift the lower critical solution temperature. The drug‐release behavior of the particles was investigated using cyclic adenosine 3′,5′‐monophosphate (cAMP) as a model drug. The release of cAMP from the hydrogel particles was affected by temperature, pH, and the CS content in the particles. These results showed that semi‐IPN hydrogel particles appeared to be of great promise in pH‐ and temperature‐sensitive oral drug release. © 2009 Wiley Periodicals, Inc. A series of pH‐responsive hydrogels were studied as potential drug carriers for the protection of insulin from the acidic environment of the stomach before releasing in the small intestine. Hydrogels based on poly(vinyl alcohol) networks grafted with acrylic acid or methacrylic acid were prepared by a two‐step process. Poly(vinyl alcohol) hydrogels were prepared by gamma ray irradiation (50 kGy) and then followed by grafting either acrylic acid or methacrylic acid onto these poly(vinyl alcohol) hydrogels with subsequent irradiation (5–20 kGy). These graft hydrogels showed pH‐sensitive swelling behavior and were used as carriers for the controlled release of insulin. The in vitro release of insulin was observed for the insulin‐loaded hydrogels in a simulated intestinal fluid (pH 6.8) but not in a simulated gastric fluid (pH 1.2). The release behavior of insulin in vivo in a rat model confirmed the effectiveness of the oral delivery of insulin to control the level of glucose. © 2003 Wiley Periodicals, Inc. A novel full‐polysaccharide hydrogel was prepared by crosslinking of chitosan with periodate‐oxidized sucrose. A tetraaldehyde molecule is synthesized via periodate oxidation of sucrose and then applied as a crosslinking agent to form a new hydrogel network. A mechanism for the superabsorbent hydrogel formation via reductive N‐alkylation was also suggested. The structure of the hydrogel was confirmed by FTIR spectroscopy, scanning electron microscopy (SEM), and thermogravimetric analysis (TGA). It is shown that crosslinking of chitosan can improve its thermal stability. The effects of crosslinker concentration, pH, and inorganic salt on the swelling behavior of the hydrogel were studied. The results indicate that the hydrogel has good pH sensitivity and pH reversible response. A straightforward strategy is described to synthesize poly(?‐caprolactone)‐graft‐poly(N‐isopropylacrylamide) (PCL‐g‐PNIPAAm) amphiphilic graft copolymers consisting of potentially biodegradable polyester backbones and thermoresponsive grafting chains. PCL with pendent chlorides was prepared by ring‐opening polymerization, followed by conversion of the pendent chlorides to azides. Alkyne‐terminated PNIPAAm was synthesized by atom transfer radial polymerization. Then, the alkyne end‐functionalized PNIPAAm was grafted onto the PCL backbone by a copper‐catalyzed azide–alkyne cycloaddition. PCL‐g‐PNIPAAm graft copolymers self‐assembled into spherical micelles comprised of PCL cores and PNIPAAm coronas. The critical micelle concentrations of the graft copolymers were in the range 7.8–18.2 mg L?1, depending on copolymer composition. Mean hydrodynamic diameters of micelles were in the range 65–135 nm, which increased as the length of grafting chains grew. A novel hydrolyzed collagen‐based hydrogel has been prepared by grafting the binary mixture of acrylamide and 2‐acrylamido‐2‐methylpropanesulfonic acid onto the collagen backbone in the presence of a crosslinking agent. Its physicochemical properties in aqueous solution were studied. The effect of reaction variables on both gel content and swelling capacity was investigated to achieve a hydrogel with improved absorbency and gel content. The absorbency under load of optimized hydrogel was also investigated by using an absorbency under load tester at various applied pressures. The swelling ratio in various salt solutions was also determined and additionally, the swelling of hydrogels was measured in solutions with pH ranged 1–13. The synthesized hydrogel exhibited a pH‐responsiveness character so that a swelling‐collapsing pulsatile behavior was recorded at pH 2 and 8. © 2007 Wiley Periodicals, Inc. Multifunctional hydrogels based on chitosan–quercetin (CHITQ) conjugate are prepared by a thermo‐induced radical procedure in the presence of N‐isopropylacrylamide (NIPAAm), acrylamide (AAm), and N,N′‐methylenebis(acrylamide) (MEBA). At first, quercetin (Q) is grafted onto chitosan backbone with a functionalization degree of 275 mg of Q per gram of conjugate, as calculated by 1H‐NMR analyses to impart antioxidant properties to the polysaccharide. Then, a pH and temperature sensitive hydrogel was obtained by involving CHITQ and NIPAAm in the polymerization reaction. The accessibility of phenolic moieties is modified in response to the hydrogel swelling/deswelling, as confirmed by antioxidant tests performed at different temperatures. Dual stimuli‐responsive hydrogels are proposed for the delivery of caffeine as model drug. The release profiles of caffeine depict a system particularly performing as on/off device at acidic pH with excellent applicability prospects.  相似文献

This research synthesized graft copolymers of chitosan‐g‐poly(N‐isopropyl acrylamide) (CS‐g‐PNIPAAm) by UV‐initiated free‐radical polymerization of NIPAAm monomer to CS selectively at the C‐6 position of pyranose ring. First, amino groups in CS were protected by reaction with phthalic anhydride (PA) to form PACS. The degree of phthaloylation was carefully controlled to ensure that most amino groups were protected, and only a very small amount of hydroxyl groups were reacted. In the second step, the vinyl functional group was introduced to the PACS by reaction with a vinyl compound containing an isocyanate group (3‐isopropenyl‐αα′‐dimethylbenzyl isocyanate), through the urethane linkage with hydroxyl groups at the C‐6 position. The phthaloyl groups were then removed by hydrazine to recover the amino groups in CS. Finally, PNIPAAm was grafted to the vinyl CS at the C‐6 position by UV‐initiated free‐radical polymerization. The synthesized CS‐g‐PNIPAAm copolymers were confirmed to have a structure of an AB‐crosslinked graft copolymer. Respectively, these copolymer hydrogels exhibited pH‐ and thermal‐responsive swelling properties in an aqueous solution due to their CS and PNIPAAm components. The test of cell viability with L929 fibroblast revealed that the CS‐g‐PNIPAAm copolymers having a grafting ratio lower than 1.7 had cellular compatibility as good as pure CS. © 2010 Wiley Periodicals, Inc. To reach sustained drug release, a new composite drug‐delivery system consisting of poly(d,l ‐lactide‐co‐glycolide) (PLGA) nanoparticles (NPs) embedded in thermosensitive poly(N‐isopropyl acrylamide) (PNIPAAm) hydrogels was developed. The PNIPAAm hydrogels were synthesized by free‐radical polymerization and were crosslinked with poly(ethylene glycol) diacrylate, and the PLGA NPs were prepared by a water‐in‐oil‐in‐water double‐emulsion solvent‐evaporation method. The release behavior of the composite hydrogels loaded with albumin–fluorescein isothiocyanate conjugate was studied and compared with that of the drug‐loaded neat hydrogel and PLGA NPs. The results indicate that we could best control the release rate of the drug by loading it to the PLGA NPs and then embedding the whole system in the PNIPAAm hydrogels. The developed composite hydrogel systems showed near zero‐order drug‐release kinetics along with a reduction or omission of initial burst release. The differential scanning calorimetry results reveal that the lower critical solution temperature of the developed composite systems remained almost unchanged (<1°C increase only). Such a characteristic indicated that the thermosensitivity of the PNIPAAm hydrogel was not distinctively affected by the addition of PLGA NPs. In conclusion, an approach was demonstrated for the successful preparation of a new hybrid hydrogel system having improved drug‐release behavior with retained thermosensitivity. Graft copolymerization of epoxy‐terminated poly(dimethylsiloxane) (PDMS) onto chitosan was reacted without using a catalyst. pH‐sensitive hydrogels were obtained that are based on two different components: a natural polymer and a synthetic polymer. These PDMS substitutents provide the basis for hydrophobic interactions that contribute to the formation of hydrogels. Various graft hydrogels were prepared from different weight ratios of chitosan and PDMS. Swelling behavior of these hydrogels was studied by immersion of the gels in various buffer solution. Photocrosslinked hydrogels exhibited a high equilibrium water content (EWC). Particularly, the sample CP31 of the highest chitosan–PDMS weight ratio showed the highest EWC in time‐dependent, temperature‐dependent, and pH‐dependent swelling behavior. © 2002 Wiley Periodicals, Inc. To improve the swelling and deswelling rate, comb-type macroporous hydrogels were prepared. The temperature-sensitive poly(N-isopropylacrylamide) (PNIPAAm) was grafted on the surface or bulk of the pH-responsive alginate. The larger surface area in pores and free chain mobility by comb-type graft copolymer reached its equilibrium swollen state within a minute. The swelling ratio of porous hydrogel in equilibrium state was over fifteen times greater than that of nonporous hydrogels. The increase of surface area by pores caused water molecules to transfer easily in and out of the matrix, resulting in a rapid deswelling. The degree of change in swelling ratio during deswelling might be affected by the phase transition behavior of PNIPAAm attached on only the surface of the pore rather than PNIPAAm grafted into mainchain of alginate. Surface-grafted alginate/PNIPAAm gels had a suitable mechanical strength without collapsing during the repeatable shrinkage and expansion due to swelling and deswelling, whereas bulk-grafted gels easily collapsed.  相似文献

A stimuli‐responsive porous hydrogel was synthesized from wheat straw hemicellulose using CaCO3 as the porogen, and its application for the removal of methylene blue was studied. The porous structure of the prepared hydrogel was confirmed by SEM analysis. The effects of pH and polyelectrolyte on the swelling of the hydrogels were discussed, and the porous hydrogels showed excellent sensitivity to pH and salt. The deswelling kinetic study indicated that the hydrogels exhibited rapid shrinking in NaCl aqueous solutions. The methylene blue adsorption on the hydrogels was investigated, and the obtained adsorption data was fitted to the pseudo‐first‐order, pseudo‐second‐order and intra‐particle diffusion kinetics models, and the pseudo‐first‐order kinetic model could describe the adsorption process, and the adsorption process of methylene blue on the hydrogels was controlled by external film diffusion. Graft copolymerization of D,L ‐lactic acid (LA) onto chitosan (CS) was attempted without using a catalyst. pH‐sensitive hydrogels were obtained which are based on two different components: a natural polymer and a synthetic polymer. These polyester substituents provide the basis for hydrophobic interactions that contribute to the formation of hydrogels. The swelling mechanisms in enzyme‐free simulated gastric fluid (SGF, pH 2.2) or simulated intestinal fluid (SIF, pH 7.4) at 37°C were investigated. Meanwhile, structural changes of the graft copolymers in the different pH buffers were studied by FTIR, and these are discussed together with the swelling mechanisms. The effect of pH on the water uptake of hydrogel was investigated by using McIlvaine buffer with the same ionic strength.
