Poly[(l ‐lactide)‐co ‐(? ‐caprolactone)] (PLCL) and poly[(l ‐lactide)‐co ‐glycolide] (PLGA) copolymers are widely used in neural guide tissue regeneration. In this research, the surface modification of their hydrophilicity was achieved using plasma treatment. Attachment and proliferation of olfactory ensheathing cells on treated electrospun membranes increased by 26 and 32%, respectively, compared to the untreated PLCL and PLGA counterparts. Cells cultivated on both the PLCL and PLGA membranes showed high viability (>95%) and healthy morphologies with no evidence of cytotoxic effects. Cells grown on treated electrospun fibres displayed significant increases in mitochondrial activity and reductions in membrane leakage when compared to untreated samples. The results suggested that plasma treatment of the surface of the polymers enhanced both cell viability and growth without incurring any cytotoxic effects. © 2017 Society of Chemical Industry  相似文献

The electrospinning of polyglycolide (PGA), poly(L ‐lactide) (PLA), and poly(lactide‐co‐glycolide) (PLGA; L ‐lactide/glycolide = 50/50) was performed with chloroform or 1,1,1,3,3,3‐hexafluoro‐2‐propanol (HFIP) as a spinning solvent to fabricate their nanofiber matrices. The morphology of the electrospun PGA, PLA, and PLGA nanofibers was investigated with scanning electron microscopy (SEM). The PLGA nanofibers, electrospun with a nonpolar chloroform solvent, had a relatively large average diameter (760 nm), and it had a relatively broad distribution in the range of 200–1800 nm. On the other hand, the PGA and PLA fibers, electrospun with a polar HFIP solvent, had a small average diameter (~300 nm) with a narrow distribution. This difference in the fiber diameters may be associated with the polarity of the solvent. Also, the in vitro degradation of PGA, PLA, and PLGA nanofiber matrices was examined in phosphate buffer solutions (pH 7.4) at 37°C. The degradation rates of the nanofiber matrices were fast, in the order of PGA > PLGA ? PLA. Structural and morphological changes during in vitro degradation were investigated with differential scanning calorimetry and wide‐angle X‐ray diffraction. For the PGA matrix, a significant increase in the crystallinity during the early stage was detected, as well as a gradual decrease during the later period, and this indicated that preferential hydrolytic degradation in the amorphous regions occurred with cleavage‐induced crystallization, followed by further degradation in the crystalline region. © 2004 Wiley Periodicals, Inc. Mixtures of different bioerosionable polyesters were used to prepare microparticulated tamoxifen delivery systems to achieve anticancer effects in breast malignant cancer cells. Tamoxifen (TMX) was included into microspheres (MS) formulated via spray‐drying. Mixtures of poly(D ,L ‐lactide‐co‐glycolide) (PLGA) of different lactide/glycolide proportions (50 : 50 and 75 : 25) and poly(D ,L ‐lactic acid) (PLA) were used. The average diameter of the resultant TMX‐loaded microparticles was in the range 1.04 ± 0.51–1.55 ± 0.95 μm. The encapsulation efficiency of TMX was between 97.8% [48.9 ± 0.1 TMX (μg)/MS (mg)] and 69.6% [36.6 ± 0.1 TMX (μg)/MS (mg)] depending on the polymeric composition of the formulation. Drug burst effect was not observed. TMX was released from the polymeric matrices in a sustained release manner between 11 and 58 days depending on polymeric composition of microspheres. TMX‐loaded microspheres showed high efficacy in causing cell death in MCF7 breast malignant cancer cells. In this study, stereocomplexed poly(lactide) (PLA) was investigated by blending linear poly(l ‐lactide) (PLLA) and tri‐block copolymer poly(d ‐lactide) ? (polyethylene glycol) ? poly(d ‐lactide) (PDLA‐PEG‐PDLA). Synthesized PDLA‐PEG‐PDLA tri‐block copolymers with different PEG and PDLA segment lengths were studied and their influences on the degree of sterecomplexation and non‐isothermal crystallization behaviour of the PLLA/PDLA‐PEG‐PDLA blend were examined in detail by DSC, XRD and polarized optical microscopy. A full stereocomplexation between PLLA and PDLA‐PEG4k‐PDLA200 could be formed when the L/D ratio ranged from 7/3 to 5/5 without the presence of PLA homocrystals. The segmental mobility and length of both PEG and PDLA are the dominating factors in the critical D/L ratio to achieve full stereocomplexation and also for nucleation and spherulite growth during the non‐isothermal crystallization process. For fixed PEG segmental length, the stereocomplexed PLA formed showed first an increasing and then a decreasing melting temperature with increasing PDLA segments due to their intrinsic stiff mobility. Furthermore, the effect of PEG segmental mobility on PLA stereocomplexation was investigated. The results clearly showed that the crystallization temperature and melting temperature of stereocomplexed‐PLA kept increasing with increasing PEG segmental length, which was due to PEG soft mobility in the tri‐block copolymers. However, PEG was not favourable for nucleation but could facilitate the spherulite growth rate. Both the PDLA and PEG segmental lengths in the tri‐block copolymers affect the crystallinity of stereocomplexed‐PLA and the stereocomplexation formation process; they have a different influence on blends prepared by solution casting or the melting method. © 2015 Society of Chemical Industry  相似文献

The enzymatic degradation of poly(D ,L ‐lactide) (PLA) was investigated using two different lipases, Novozym 435 and Lipolase. The optimum temperature was 50°C for the enzymatic degradation of PLA. The effect of various solvents on the degradation of PLA was investigated at 50°C using Novozym 435, and toluene was found to be the best solvent among the solvents investigated. The enzymatic degradation of the blends of PLA and PVAc was investigated at 50°C in toluene. The enzymatic degradation of the blends of PLA and PVAc showed that there is an interaction between the polymers during degradation, which results in the reduction of degradation rate of both polymers in the blend. Surfactant‐free nanoparticles of poly(DL ‐lactide‐co‐glycolide) (PLGA) nanoparticles were prepared with or without poly(L ‐lactide)‐poly(ethylene oxide) (LE) diblock copolymer (abbreviated as PLGA/LE and PLGA nanoparticles) by dialysis method. LE diblock copolymer was used to make PLGA nanoparticles to alternate conventional surfactant. The size of PLGA and PLGA/LE nanoparticles was 295.3 ± 171.3 and 307.6 ± 27.2 nm, respectively, suggesting LE diblock copolymer might be coated onto the surface of nanoparticles. Observation of scanning electron microscope (SEM) showed that PLGA/LE nanoparticles have spherical shapes ranging ~ 200–500 nm. In 1H‐NMR study, characteristic peaks of the methyl protons of PLGA disappeared in D2O, whereas characteristic peaks of the methyl proton of both PEG and PLGA were shown in both CDCl3 and D2O, indicating that LE diblock copolymer coated on the surface of the PLGA nanoparticles. The higher the initial content of drug, the higher the drug contents and the lower the loading efficiency. PLGA/LE nanoparticles at higher drug contents resulted in slower adriamycin·HCl (ADR) release rate than that of lower drug contents. Also, slower release rate of ADR was achieved by entrapped into the PLGA/LE nanoparticles, whereas LE polymeric micelles showed rapid ADR release. © 2003 Wiley Periodicals, Inc. The quasiliving characteristics of the ring‐opening polymerization of ?‐caprolactone (CL) catalyzed by an organic amino calcium were demonstrated. Taking advantage of this feature, we synthesized a series of poly(?‐caprolactone) (PCL)–poly(L ‐lactide) (PLA) diblock copolymers with the sequential addition of the monomers CL and L ‐lactide. The block structure was confirmed by 1H‐NMR, 13C‐NMR, and gel permeation chromatography analysis. The crystalline structure of the copolymers was investigated by differential scanning calorimetry and wide‐angle X‐ray diffraction analysis. When the molecular weight of the PLA block was high enough, phase separation took place in the block copolymer to form PCL and PLA domains, respectively. © 2006 Wiley Periodicals, Inc. Ring‐opening polymerization of L ‐ or D ‐lactide was realized at 140 °C for a period of 7 days in the presence of dihydroxyl poly(ethylene glycol) (PEG), with M?n = 4000 g mol?1, using zinc lactate as initiator. The resulting poly(L ‐lactide)–PEG–poly(L ‐lactide) and poly(D ‐lactide)–PEG–poly(D ‐lactide) triblock copolymers are water soluble with polylactide (PLA) block length ranging from 11 to 17 units. Both the tube inverting method and rheological measurements were used to evaluate the gelation properties of aqueous solutions containing single copolymers or L /D copolymer pairs. Stereocomplexation between poly(L ‐lactide) and poly(D ‐lactide) blocks is observed for mixed solutions. Hydrogel formation is detected in the case of relatively long PLA blocks (DP PLA = 17), but not for copolymers with shorter PLA blocks (DP PLA = 11–13) due to partial racemization of L ‐lactyl units. Racemization is largely reduced when the reaction time is shortened to 1 day. Under these conditions, DP PLA of 8 is sufficient for the stereocomplexation of PLA–PEG block copolymers, and DP PLA above 10 leads to the formation of hydrogels of PLA–PEG block copolymers. On the other hand, racemization appears as a general phenomenon in the (co)polymerization of L ‐lactide with Zn(Lac)2 as initiator, although it is negligible or undetectable in the case of high molar mass polymers. Therefore, racemization is the limiting factor for the stereocomplexation‐induced gelation of water‐soluble PLA–PEG block copolymers where the PLA block length generally ranges from 10 to 30. Poly(D ,L ‐lactide‐co‐glycolide) (PLGA) and poly(ethylenimine) (PEI) were blended and found to form a homogeneous pH sensitive matrix for drug release. Differential scanning calorimetry (DSC) studies of the PLGA/PEI blends showed a single glass transition temperature at all compositions. Fourier transform infrared spectroscopy (FTIR) demonstrated that the PLGA carbonyl peak at 1760 cm?1 shifted to 1666 cm?1 as a result of amide bond formation between the two polymers. This was confirmed by 13C nuclear magnetic resonance studies. A PLGA/PEI matrix of 90/10 weight ratio was chosen for evaluation for controlled drug release. Both hydrophobic β‐lapachone and hydrophilic rhodamine B showed pH dependent release profiles with faster release kinetics at lower pH values. The observed pH sensitive drug release was mainly attributed to two factors, pH dependent swelling and protonation of the PEI‐PLGA matrix. Two series of biodegradable polymer blends were prepared from combinations of poly(L ‐lactide) (PLLA) with poly(?‐caprolactone) (PCL) and poly(butylene succinate‐co‐L ‐lactate) (PBSL) in proportions of 100/0, 90/10, 80/20, and 70/30 (based on the weight percentage). Their mechanical properties were investigated and related to their morphologies. The thermal properties, Fourier transform infrared spectroscopy, and melt flow index analysis of the binary blends and virgin polymers were then evaluated. The addition of PCL and PBSL to PLLA reduced the tensile strength and Young's modulus, whereas the elongation at break and melt flow index increased. The stress–strain curve showed that the blending of PLLA with ductile PCL and PBSL improved the toughness and increased the thermal stability of the blended polymers. A morphological analysis of the PLLA and the PLLA blends revealed that all the PLLA/PCL and PLLA/PBSL blends were immiscible with the PCL and PBSL phases finely dispersed in the PLLA‐rich phase. © 2009 Wiley Periodicals, Inc. We propose here, a novel technique to synthesize high molecular weight (MW) poly (L ‐lactic acid)‐clay nanocomposite (PLACN), via solid state polymerization (SSP). We synthesize prepolymer of PLACN (pre‐PLACN) from both, L ‐lactic acid and L ‐lactide, as starting materials. Synthesis of pre‐PLACN from L ‐lactic acid is carried out via in situ melt polycondensation (MP) of L ‐lactic acid oligomer, followed by SSP, to achieve high MW PLACN (Mw ∼ 138,000 Da). In case of L ‐lactide as the starting material, we prepare L ‐lactide–clay intercalated mixture which yields moderate MW pre‐PLACN during subsequent ring opening polymerization (ROP). Interestingly, ROP is performed by using hydroxyl functionalized ternary catalyst system (L ‐lactide–Sn(II) octoate–oligo (L‐lactic acid) complex), which provides the terminal hydroxyl end‐groups, required for step‐growth SSP. Pre‐PLACN MW is now increased to Mw ∼ 127,000 Da, by the subsequent SSP process. 1H NMR analyses confirm that these end‐groups, are indeed consumed during SSP. During SSP, the PLACN also achieves up to 90% crystallinity, which may be due to the synchronization of the slow step‐growth SSP of poly(L ‐lactic acid) (PLA) with the crystallization kinetics. Optical purity of PLACNs is similar to that of neat PLA, whereas the thermal stability of PLACNs is significantly superior. As evidenced by wide‐angle X‐ray scattering/small‐angle X‐ray scattering analyses and in line with the literature, both, intercalated and exfoliated PLACN morphologies, have been synthesized, by suitable selection of clays. We also verify the correlation between the PLA semicrystalline morphology and the PLACN morphology, which is consistent with those of PLACN synthesized by other techniques. POLYM. Supramolecular poly(?‐capolactone)/poly(lactide) alternating multiblock copolymers were prepared by UPy‐functionalized poly(lactide)‐b‐ poly(?‐capolactone)‐b‐ poly(lactide) copolymers. The prepared supramolecular polymers (SMPs) exhibit the characteristic properties of thermoplastic elastomers. The stereo multiblock SMPs (sc‐SMPs) were formed by blending UPy‐functionalized poly(l ‐lactide)‐b‐ PCL‐b‐ poly(l ‐lactide) (l ‐SMPs) and UPy‐functionalized poly(d ‐lactide)‐b‐ PCL‐b‐ poly(d ‐lactide) (d ‐SMPs) due to stereocomplexation of the PLLA and PDLA blocks. Sc‐SMPs with low content of d ‐SMPs (≤20%) are transparent, elastic solids, while those having high d ‐SMPs content are opaque, brittle solids. The effects of l ‐SMPs/d ‐SMPs mixing ratios on thermal, crystallization behaviors, crystal structure, mechanical and hydrophilic properties of sc‐SMPs were deeply investigated. The incorporation of UPy groups depresses the crystallization of polymer, and the stereocomplex formation accelerates the crystallization rate. The used initiator functionalized polyhedral oligomeric silsesquioxanes causes a different effect on the crystallization of PLA and PCL blocks. The tensile strength and elongation at break of l d /d d ‐SMPs (d represents the initiator diethylene glycol) are significantly larger than that of l p /d p ‐SMPs (p represents the initiator polyhedral oligomeric silsesquioxanes), and their heat resistance and hydrophilicity can be also modulated by the l ‐SMPs/d ‐SMPs mixing ratios and the different initiators. © 2017 Wiley Periodicals, Inc. Polymeric materials prepared from renewable natural resources are now being accepted as “bio‐based polymers”, because they are superior to the conventional petroleum‐based polymers in reducing the emission of carbon dioxide. Among them, poly(L ‐lactide) (PLLA) prepared by fermentation and polymerization is paid an immediate attention. Although PLLA exhibits a broad range of physico‐chemical properties, its thermal and mechanical properties are somewhat poorer for use as ordinary structural materials. For improving these inferior properties, a stereocomplex form consisting of PLLA and its enantiomer poly(D ‐lactide) (PDLA) has high potential because of showing high melting nature (230 °C). It can be formed by simple polymer blend of PLLA and PDLA or more easily with stereoblock polylactides (sb‐PLA) which are PLLA/PDLA block copolymers. These novel PLA polymers, named “Neo‐PLA”, can provide a wide range of properties that have never be attained with single PLLA. Neo‐PLA retains sustainability or bio‐based nature, because both monomers L ‐ and D ‐lactic acids are manufactured from starch by fermentation. Copyright © 2006 Society of Chemical Industry  相似文献

Random copolyester namely, poly(ethylene terephthalate‐co‐sebacate) (PETS), with relatively lower molecular weight was first synthesized, and then it was used as a macromonomer to initiate ring‐opening polymerization of l ‐lactide. 1H NMR quantified composition and structure of triblock copolyesters [poly(l ‐lactic acid)‐b‐poly(ethylene terephthalate‐co‐sebacate)‐b‐poly(l ‐lactic acid)] (PLLA‐PETS‐PLLA). Molecular weights of copolyesters were also estimated from NMR spectra, and confirmed by GPC. Copolyesters exhibited different solubilities according to the actual content of PLLA units in the main chain. Copolymerization effected melting behaviors significantly because of the incorporation of PETS and PLLA blocks. Crystalline morphology showed a special pattern for specimen with certain composition. It was obvious that copolyesters with more content of aromatic units of PET exhibited increased values in both of stress and modulus in tensile test. © 2007 Wiley Periodicals, Inc. The poly(2‐methacryloyloxyethyl phosphorylcholine)‐block‐poly(D ,L ‐lactide) (PMPC‐b‐PLA) was specially designed to develop biomimetic giant vesicles (GVs) and giant large compound vesicles via a simple spontaneous assemble in aqueous solution. The weight fraction of the hydrophilic PMPC block (fPC) was proved to play an important role in the size and morphology control of the self‐assembled aggregates. A biodegradable block copolymer, poly‐D ,L ‐lactide (PLA)‐co‐poly(ethylene glycol) (PEG), was prepared by the ring‐opening polymerization of lactide with stannous caprylate [Sn(Oct2)] as a catalyst; then, the PLA–PEG copolymer was made into nanoparticles by nanoprecipitation under different conditions. The average molecular weight and structure of PLA–PEG were detected by 1H‐NMR and gel permeation chromatography. The sizes and distributions of the nanoparticles were investigated with a laser particle‐size analyzer. The morphologies of the nanoparticles were examined by transmission electron microscopy. The effects of the solvent–nonsolvent system, operation conditions, and dosage of span‐80 on the sizes and distributions of the nanoparticles are discussed. The results show that acetone–water was a suitable solvent–nonsolvent system and the volume ratio of the nonsolvent phase to the solvent phase (O/W) (v/v), the concentration of PLA–PEG in the solvent phase, and the dosage of span‐80 had important effects on the particle sizes and distributions. © 2005 Wiley Periodicals, Inc. The degradation of polymers under ultraviolet (UV) irradiation has been a great concern for biomaterial and agricultural applications. The major objective of this research was to study the effect of UV irradiation on the representative bulk and surface properties of poly (L ‐lactide) (PLA) films. Two UV sources with different spectral outputs and intensities were chosen so that one of them could be used for surface modification and the other could be used for UV sterilization of the PLA films. The results established that the molecular weight of PLA decreased significantly during irradiation from the photografting lamp under atmospheric conditions. Irradiation through a Pyrex container was shown to minimize polymer degradation during UV exposure from the photografting lamp. The PLA films UV‐irradiated under the sterilization lamp for 12 h revealed a similar reduction in the molecular weight and no change in the surface hydrophilicity. Electrospinning of various polymers has been used to produce nanofibrous scaffolds that mimic the extracellular matrix and support cell attachment for the potential repair and engineering of nerve tissue. In the study reported here, an electrospun copolymer of l ‐lactide and ε‐caprolactone (67:33 mol%) resulted in a nanofibrous scaffold with average fibre diameter and pore size of 476 ± 88 and 253 ± 17 nm, respectively. Blending with low loadings of collagen (<2.5% w/w) significantly reduced the average diameter and pore size. The uniformity of fibre diameter distributions was supported with increasing collagen loadings. The nanofibrous scaffolds significantly promoted the attachment and proliferation of olfactory ensheathing cells compared to cells exhibiting asynchronous growth. Furthermore, analysis of cell health through mitochondrial activity, membrane leakage, cell cycle progression and apoptotic indices showed that the nanofibrous membranes promoted cell vigour, reducing necrosis. The study suggests that the use of more cost‐effective, low loadings of collagen supports morphological changes in electrospun poly[(l ‐lactide)‐co‐(ε‐caprolactone)] nanofibrous scaffolds, which also support attachment and proliferation of olfactory ensheathing cells while promoting cell health. The results here support further investigation of the electrospinning of these polymer blends as conduits for nerve repair. © 2013 Society of Chemical Industry  相似文献

A series of triblock co‐polymers, consisting of a poly(ethylene glycol) (PEG) central block joined to two blocks of random p‐dioxanone‐co‐L ‐lactide copolymers were synthesized by ring‐opening polymerization of p‐dioxanone (PDO) and L ‐lactide (LLA) initiated by PEG in the presence of stannous 2‐ethylhexanoate catalyst. The resulting copolymers were characterized by various techniques including 1H and 13C NMR and FTIR spectroscopies, gel permeation chromatography, inherent viscosity, wide‐angle X‐ray diffractometry (WAXD) and differential scanning calorimetry (DSC). The conversion of PDO and L ‐lactide into the polymer was studied various mole ratios and at different polymerization temperature from 1H NMR spectra. Results of WAXD and DSC showed that the crystallinity of PEG macroinitiator was greatly influenced by the composition of PDO and L ‐lactide in the copolymer. Temperature‐responsive polymers have become increasingly attractive as carrier for the injectable drug delivery systems. In the present work, we have studied the preparation of poly(N‐isopropylacrylamide‐acrylamide‐vinilpyrrolidone) (NIPAAm‐AAm‐VP terpolymer) nanoparticulated terpolymer and its blend with poly(lactide‐co‐glycolide, PLGA; molar ratio of lactide/glycolid 1/3). Thermosensitive terpolymer, poly(NIPAAm‐AAm‐VP) was prepared by free‐radical polymerization in aqueous solution. The nanoparticles of poly(NIPAAm‐AAm‐VP) and its blend with PLGA containing naltrexone were prepared using the evaporation and w/o emulsion‐solvent evaporation methods, respectively. Nanoparticles prepared from terpolymer‐PLGA blend at low polymer concentration (5%) shows larger particle size (>300 nm) and higher drug content%. Various types of nanoparticles showed a burst release of less than 10% after 24 h . The results suggest that by regulating different variables, desired release profiles of naltrexone can be achieved using a blend of PLGA‐poly(NIPAAm‐AAm‐VP) nanoparticulate system. © 2008 Wiley Periodicals, Inc.
