Poly(lactic‐co‐glycolic acid) (PLGA) microspheres prepared using a traditional solvent evaporation or double emulsification method are usually polydisperse with an uncontrollable particle size distribution, which brings about poor application performance. In our research, monodisperse magnetic PLGA microspheres were prepared using a microchannel device based on a water‐in‐oil‐in‐water composite emulsion. The composite emulsion was formed by injecting a dichloromethane–gelatin water‐in‐oil emulsion into a microchannel together with an external water phase, i.e. poly(vinyl alcohol) (PVA) aqueous solution. Mean particle size control of the microspheres was executed using the osmotic pressure difference between internal and external aqueous phases caused by regulating NaCl concentration in PVA aqueous phase. It is found that monodisperse magnetic PLGA microspheres with high magnetic responsiveness can be successfully prepared combining the microchannel device with composite emulsion method. Mean particle size of the microspheres with coefficient of variation value below 4.72% is controllable from 123 to 203 µm depending on the osmotic pressure. The resulting samples have pyknotic and smooth surfaces, as well as spherical appearance. These monodisperse magnetic PLGA microspheres with good superparamagnetism and magnetic mobility have potential use as drug carriers for uniform release and magnetic targeting hyperthermia in biological fields. © 2015 Society of Chemical Industry  相似文献

BACKGROUND: Vitamin B12 is an essential vitamin required by all mammals. Absorption of vitamin B12 is facilitated by binding of intrinsic factor–vitamin B12 complex to specific receptors in the ileum. In humans a deficiency of this vitamin or a lack of intrinsic factor leads to pernicious anaemia. The major objective of the present study was to prepare intrinsic factor–vitamin B12 complex‐loaded poly[lactic‐co‐(glycolic acid)] (PLGA)‐based microparticles and to investigate their release kinetics. RESULTS: PLGA copolymer was synthesized by the ring‐opening polymerization method and characterized using gel permeation chromatography, Fourier transform infrared spectroscopy and 1H NMR. The glass transition temperature measurement showed a single Tg at 40 °C. The intrinsic factor–vitamin B12 complex‐loaded PLGA microspheres were prepared by a water‐in‐oil‐in‐water double emulsion solvent extraction/evaporation technique. An environmental scanning electron microscopy investigation demonstrated that the PLGA particles had a mean particle diameter of 38 µm. Interestingly, different drug release patterns (bi‐ and triphasic ones) were observed for vitamin B12‐loaded and intrinsic factor–vitamin B12 complex‐loaded microspheres. In contrast to the rapid release of vitamin B12 by itself, in vitro release tests showed that intrinsic factor and vitamin B12 in the complex were released from PLGA microspheres in a sustained manner over 15 days. One of the major problems of nanofiber scaffold or other devices like cardiovascular or blood‐contacting medical devices is their weak mechanical properties and the lack of hemocompatibility of their surfaces. In this study, halloysite nanotubes (HNTs) and carbon nanotubes (CNTs) were incorporated within poly(lactic‐co‐glycolic acid) (PLGA) nanofibers and the mechanical property and hemocompatibility of both types of composite nanofibers with different doping levels were thoroughly investigated. The morphology and internal distribution of the doped nanotubes within the nanofibers were characterized using scanning electron microscopy and transmission electron microscopy. Mechanical properties of the electrospun nanofibers were tested using a material testing machine. The hemocompatibility of the composite nanofibers was examined through hemolytic and anticoagulant assay, respectively. We show that the doped HNTs or CNTs are distributed in the nanofibers with a coaxial manner and the incorporation of HNTs or CNTs does not significantly change the morphology of the PLGA nanofibers. Importantly, the incorporation of HNTs or CNTs within PLGA nanofibers significantly improves the mechanical property of PLGA nanofibers, and PLGA nanofibers with or without doping of the HNTs and CNTs display good anticoagulant property and negligible hemolytic effect to human red blood cells. With the enhanced mechanical property, great hemocompatibility, and previously demonstrated biocompatibility of both HNTs‐ and CNTs‐doped composite PLGA nanofibers, these composite nanofibers may be used as therapeutic artificial tissue/organ substitutes for tissue engineering applications. © 2012 Wiley Periodicals, Inc. The objective of the present work was to study the release behavior of plain and blend microspheres (MS) of PLGA and Pluronic F68/127. In this study, a novel blend MS of poly(D ,L ‐lactic‐co‐glycolic acid) (PLGA) and Pluronic F68/127 (PLF68/127) were prepared by the emulsion–solvent evaporation method. Repaglinide, an antidiabetic drug with a very short half‐life, was successfully encapsulated into the blend MS. Various formulations were prepared by varying the ratio of PLGA and PLF68/127. Drug encapsulation up to 91% was achieved as measured by UV spectroscopy. Scanning electron microscopy showed that MS have smooth surfaces even after incorporation of PLF68/127. Particle size, as measured by using laser light scattering technique, gave an average size ranging from 12 to 47 μm. Differential scanning calorimetry (DSC) was performed to understand the crystalline nature of the drug after encapsulation into MS. DSC revealed the crystalline dispersion in the polymer matrix. In vitro release experiments performed in simulated intestinal fluid, indicated the dependence of release rate on the amount of PLF68/127 present in the MS; slow release was extended up to 153 h. Release data have been fitted to an empirical equation to compute the diffusional exponent (n), which indicated that the release mechanism to be non‐Fickian type. © 2009 Wiley Periodicals, Inc. Recently a method that uses water droplets at the air–solution interface as an ordered template was reported for the preparation of ordered micrometer‐size honeycomb structures. Here we show that the method can also be used for formation of honeycomb‐like porous films from random copolymers with certain hydrophilicity, besides those polymers with defined structures such as block copolymers, starlike homopolymers, amd amphiphilic polymers. This demonstrates that the stabilization of water droplets is the key factor for the regular structure. Also we indicate that size and structure of the films can be regulated by such variables as concentration and atmospheric humidity. © 2003 Wiley Periodicals, Inc. An operating window, which is bounded by two temperatures and draw ratios, defines the stable and defect‐free stretching region of a polymer film. Physical properties including the coefficient of thermal expansion (CTE), birefringence, and Young's modulus of a recyclable polyimide (PIR) film were measured under stretching conditions. While values of birefringence and Young's modulus increased with increasing stretching stress in the machine direction, the CTE was found to decrease. A semiempirical model for the prediction of birefringence and Young's modulus under stretching conditions was developed, from which the CTE could be estimated from the Young's modulus data. This study continues long‐standing efforts to develop protein delivery systems based on cyclodextrin‐conjugated polyester in our laboratory. The crude products of ethylenediamino bridged bis(β‐cyclodextrin)‐conjugated poly(DL ‐lactic‐co‐glycolic acid) were used in this study to make full use of unreacted reactant. With bovine serum albumin (BSA) as a model protein, the encapsulation effects (the encapsulation efficiency and particle size) of nanoparticles were similar to those of using pure conjugated products. Besides, a water‐in‐oil‐in‐water emulsification technique was conveniently modified. By adding polyvinyl alcohol (PVA) in the internal aqueous phase, a more stabilized emulsion was formed. Consequently, less PVA (~ 0.05%) was needed in the outer aqueous phase and less PVA (0.14 g/g nanoparticles) remained in the nanoparticles. This modification resulted in improved encapsulation efficiency (~ 89–94%) of BSA and an enlarged particle size (340–390 nm). Furthermore, the burst release of BSA at the 1st day was less pronounced (7–12% of the encapsulated amount) than that of nanoparticles with no PVA added in the internal aqueous phase. Degradation studies using transmission electron microscope and gel permeation chromatography suggested that the mechanism for protein release was mainly through nanoparticles erosion. © 2007 Wiley Periodicals, Inc. The ordered honeycomb structures of poly(L ‐lactic‐co‐glycolic acid) and poly(D ,L ‐lactic‐co‐glycolic acid) fabricated in a humid atmosphere were reported in this paper. It was found that surfactants were important in the formation of honeycomb films of hydrophobic polymer. Different strategies have been explored for the purpose of autologous or allogeneic dermal regeneration. We have developed a hybrid matrix by lyophilizing collagen within a poly[(lactic acid)‐co‐(glycolic acid)] (10:90, molar composition) knitted mesh, in order to assimilate the advantages of natural and synthetic materials. The porosity of the mesh was found to be almost 95 %, using Micro‐Computed Tomography Analysis, while the mechanical properties were comparable to native skin. In vitro biocompatibility was analyzed by culturing rat dermal fibroblasts in the matrices over 10 days. The cells were able to attach, proliferate and remain viable within the hybrid matrices. Subsequently, in vivo biocompatibility was analyzed by implanting the matrices subcutaneously in immunocompetent rats, for 2 weeks. Histological analysis showed that the poly[(lactic acid)‐co‐(glycolic acid)]–collagen hybrid matrices evoked minimal host tissue response in vivo. This study forms the basis of using poly[(lactic acid)‐co‐(glycolic acid)]–collagen hybrid matrices for our future work to develop a bioactive matrix for dermal regeneration. A systematic study of the effects of , flow rate, voltage, and composition on the morphology of electrospun PLGA nanofibers is reported. It is shown that changes of voltage and flow rate do not appreciably affect the morphology. However, the of PLGA predominantly determines the formation of bead structures. Uniform electrospun PLGA nanofibers with controllable diameters can be formed through optimization. Further, multi‐walled carbon nanotubes can be incorporated into the PLGA nanofibers, significantly enhancing their tensile strength and elasticity without compromising the uniform morphology. The variable size, porosity, and composition of the nanofibers are essential for their applications in regenerative medicine.

Monodispersed crosslinked cationic poly(4‐vinylpyridine‐co‐butyl acrylate) [P(4VP‐BA)] seed latexes were prepared by soapless emulsion polymerization, using 2,2′‐azobismethyl(propionamidine)dihydrochloride (V50) as an initiator and divinylbenzene (DVB) or ethylene glycol dimethacrylate (EGDMA) as a crosslinker. The optimum condition to obtain monodispersed stable latex was investigated. It was found that the colloidal stability of the P4VP latex can be improved by adding an adequate amount of BA (BA/4VP = 1/4, w/w), and adopting a semicontinuous monomer feed mode. Subsequently, poly(4‐vinylpyridine‐co‐butyl acrylate)/Poly(styrene‐co‐butyl acrylate) [P(4VP‐BA)/P(ST‐BA)] composite microspheres were synthesized by seeded polymerization, using the above latex as a seed and a mixture of ST and BA as the second‐stage monomers. The effects of the type of crosslinker, the degree of crosslinking, and the initiators (AIBN and V50) on the morphology of final composite particles are discussed in detail. It was found that P(4VP‐BA)/P(ST‐BA) composite microspheres were always surrounded by a PST‐rich shell when V50 was used as initiator, while sandwich‐like or popcorn‐like composite particles were produced when AIBN was employed. This is because the polarity of the polymer chains with AIBN fragments is lower than for the polymer with V50 fragments, hence leading to higher interfacial tension between the second‐stage PST‐rich polymer and the aqueous phase, and between PST‐rich polymer and P4VP‐rich seed polymer. As a result, the seed cannot be engulfed by the PST‐rich polymer. Furthermore, the decrease of Tg of the second‐stage polymer promoted phase separation between the seeds and the PST‐rich polymer: sandwich‐like particles formed more preferably than popcorn‐like particles. It is important knowledge that various morphologies different from PST‐rich core/P4VP‐rich shell morphology, can be obtained only by changing the initiator, considering P4VP is much more hydrophilic than PST. The zeta potential of composite particles initiated by AIBN in seeded polymerization shifted from a positive to a negative charge. © 2002 Wiley Periodicals, Inc. A comparison of the thermal properties of two classes of poly(D,L ‐lactic‐glycolic acid) multiblock copolymers is reported. In particular, the results of differential scanning calorimetry, and thermogravimetric analysis of copolymers containing poly(ethylene glycol) (PEG) or diol‐terminated poly(ϵ‐caprolactone) (PCDT) segments are described. The influence of the chemical structure and the length of PEG and PCDT on thermal stability, degree of crystallinity and glass transition temperature (Tg ) is discussed. Poly(L ‐lactic acid) (PLLA) was blended with poly(ethylene‐co‐vinyl alcohol) (EVOH) in the presence of an esterification catalyst to induce reaction between the hydroxyl groups of EVOH and the terminal carboxylic group of PLLA. Nascent low‐molecular‐weight PLLA, obtained from a direct condensation polymerization of L ‐lactic acid in bulk state, was used for the blending. Domain size of the PLLA phase in the graft copolymer was much smaller than that corresponding to a PLLA/EVOH simple blend. The mechanical properties of the graft copolymer were far superior to those of the simple blend, and the graft copolymer exhibited excellent mechanical properties even though the biodegradable fraction substantially exceeded the percolation level. The grafted PLLA reduced the crystallization rate of the EVOH moiety. Melting peak temperature (Tm) of the PLLA phase was not observed until the content of PLLA in the graft reaction medium went over 60 wt %. The modified Sturm test results demonstrated that biodegradation of EVOH‐g‐PLLA took place more slowly than that of an EVOH/PLLA simple blend, indicating that the chemically bound PLLA moiety was less susceptible to microbial attack than PLLA in the simple blend. © 2005 Wiley Periodicals, Inc. Poly(styrene‐co‐methacrylic acid) P(St‐co‐MAA) microspheres with a monodisperse size distribution were prepared by emulsifier‐free emulsion copolymerization of St and MAA. The effects of MAA content on the polymerization rate and the content of MAA in the copolymer were investigated by gravimetrical and IR methods, respectively. The results of XPS measurement indicated the presence of a carboxyl functional group. By chemical metal deposition, nickel or palladium particles were formed and deposited on the surface of P(St‐co‐MAA) microspheres to form P(St‐co‐MAA)Ni or P(St‐co‐MAA)Pd composite particles. XRD measurement and TEM observation confirmed that nickel and palladium metal particles in a small size (20–40 nm) were distributed on surface of the copolymer microspheres. © 2000 John Wiley & Sons, Inc. Differential scanning calorimetry (DSC), wide angle X‐ray diffraction (WAXD) and dynamic mechanical analysis (DMA) properties of poly(lactic acid)/ poly(butylene adipate‐co‐terephthalate) (PLA/PBAT) specimens suggest that only small amounts of poor PLA and/or PBAT crystals are present in their corresponding melt crystallized specimens. In fact, the percentage crystallinity, peak melting temperature and onset re‐crystallization temperature values of PLA/PBAT specimens reduce gradually as their PBAT contents increase. However, the glass transition temperatures of PLA molecules found by DSC and DMA analysis reduce to the minimum value as the PBAT contents of PLAxPBATy specimens reach 2.5 wt %. Further morphological and DMA analysis of PLA/PBAT specimens reveal that PBAT molecules are miscible with PLA molecules at PBAT contents equal to or less than 2.5 wt %, since no distinguished phase‐separated PBAT droplets and tan δ transitions were found on fracture surfaces and tan δ curves of PLA/PBAT specimens, respectively. In contrast to PLA, the PBAT specimen exhibits highly deformable properties. After blending proper amounts of PBAT in PLA, the inherent brittle deformation behavior of PLA was successfully improved. Possible reasons accounting for these interesting crystallization, compatible and tensile properties of PLA/PBAT specimens are proposed. © 2009 Wiley Periodicals, Inc. The hydrolytic degradation and corresponding content release of capsules made of poly(d,l ‐lactic‐co‐glycolic acid) (PLGA) strongly depends on the composition and material properties of the initially applied copolymer. Consecutive or simultaneous release from capsule batches of combinable material compositions, therefore, offers high control over the bioavailability of an encapsulated drug. The keynote of this study was the creation of a superordinated database that addressed the correlation between the release kinetics of filling agents with different molecular weights from PLGA capsules of alternating composition. Fluorescein isothiocyanate (FITC)–dextran (with molecular weights of 4, 40, and 2000 kDa) was chosen as a model analyte, whereas the copolymers were taken from various 50:50 PLGA, 75:25 PLGA, and polylactide blends. With reference to recent publications, the capsule properties, such as the size, morphology, and encapsulation efficiency, were further modified during production. Hence, uniform microdisperse and polydisperse submicrometer nanocapsules were prepared by two different water‐in‐oil‐in‐water emulsification techniques, and additional effects on the size and morphology were achieved by capsule solidification in two different sodium salt buffers. The qualitative and quantitative examination of the physical capsule properties was performed by confocal laser scanning microscopy, scanning electron microscopy, and Coulter counting techniques to evaluate the capsule size distribution and the morphological appearance of the different batches. The corresponding agent release was quantified by fluorescence measurement of the FITC–dextran in the incubation media and by the direct measurement of the capsule brightness via fluorescence microscopy. In summary, the observed agent release showed a highly controllable flexibility depending on the PLGA blends, preparation methods, and molecular weight of the used filling substances. © 2013 Wiley Periodicals, Inc. Poly(styrene‐co‐divinylbenzene) microspheres with size ranging from 1.6 to 1.8 μm were prepared in acetic acid by precipitation polymerization. The particle size and particle size distribution were determined by laser diffraction particle size analyzer, and the morphology of the particles was observed with scanning electron microscope. Besides, effects of various polymerization parameters such as initiator and total monomer concentration, divinylbenzene (DVB) content, polymerization time and polymerization temperature on the morphology and particle size were investigated in this article. In addition, the yield of microspheres increased with the increasing total monomer concentration, initiator loading, DVB concentration and polymerization time. In addition, the optimum polymerization conditions for synthesis of monodisperse crosslinked poly(styrene‐co‐divinylbenzene) microspheres by precipitation polymerization in acetic acid were obtained. © 2011 Wiley Periodicals, Inc. A new mathematical model for the prediction of the heterogeneous hydrolytic degradation of poly(D,L‐lactide‐co‐glycolide) (PLGA)‐based microspheres was developed. The model takes into account the autocatalytic effect of carboxylic groups and polymer composition on the degradation rate. It is based on mass balances for the different species, considering the kinetic and mass transport phenomena involved. The model estimates the evolution of average molecular weight, mass loss, and morphological change of the particles during degradation, and it was validated with novel experimental data. Theoretical predictions are in agreement with the hydrolysis data of PLGA microspheres (error values less than 5%). The model is able to predict the effect of particle size and molecular weight on the degradation of PLGA‐based microspheres and estimates the morphological changes of the particles due to the autocatalytic effect. © 2017 Wiley Periodicals, Inc. Poly(N‐isopropylacrylamide‐co‐acrylic acid) (poly(NIPAM‐co‐AA)) microgels with different copolymer compositions were prepared through soap‐free emulsion polymerization at 80°C, and 2, 2′‐azobisisobutyronitrile (AIBN) was used as initiator. Scanning electron microscope (SEM) characterization shows that the prepared microgels are regular and smooth and not easy to distort. Result of 1H‐NMR characterization shows that with increasing of the initial concentration of AA (AA in feed), the AA content in polymer chains increases. The thermal response of microgels latex was investigated by UV‐3010 spectrophometer through detecting the transmittance of the latex at different temperature in the range of 190–900 nm. The thermal response of the poly(NIPAM‐co‐AA) microgels was tested by dynamic light scattering (DLS). The results show that with the increase of AA content in polymer chains, the low critical solution temperature (LCST) of microgels latex first decreases and then increases. Still, with increasing of AA in poly(NIPAM‐co‐AA) microgels, the LCST of microgels first increases and then decreases. The basic reasons causing the changes of LCST of microgels latex and microgels are interpreted clearly in this article from the perspective of hydrogen bonding interaction. © 2011 Wiley Periodicals, Inc.
