以鲑鱼皮为原料,采用酸浸-水提法得到鲑鱼皮明胶,并通过酶解制备了鲑鱼皮胶原蛋白肽,其中超过91.5%的鲑鱼皮胶原蛋白肽分子量小于1 000 Da。通过葡聚糖凝胶G-15层析柱分离出5种组分,按其分子量的大小依次为:S1S2S3S4S5。体外试验结果表明,相对湿度81%的环境下,吸湿性顺序为:S5S4甘油鲑鱼皮胶原蛋白肽S3S2S1≈透明质酸鲑鱼皮明胶;保湿性顺序为:S5≈S4S3S2甘油鲑鱼皮胶原蛋白肽S1透明质酸鲑鱼皮明胶。相对湿度43%的环境下,吸湿性顺序为:S5甘油S4鲑鱼皮胶原蛋白肽≈S3透明质酸S2S1鲑鱼皮明胶;保湿性顺序为:S5S4甘油S3≈鲑鱼皮胶原蛋白肽透明质酸≈S2S1鲑鱼皮明胶。皮肤测试结果表明,涂抹含鲑鱼皮胶原蛋白肽的护肤品能够显著提高皮肤水分含量和降低经表皮失水率(TEWL),小分子鲑鱼皮胶原蛋白肽的保湿效果优于鲑鱼皮明胶。  相似文献

重组人源化Ⅲ型胶原蛋白(RHCⅢ)由人胶原原始基因序列经过优化重组表达所得,与人体天然胶原氨基酸序列高度一致。为探究重组人源化Ⅲ型胶原蛋白应用在化妆品中,对减缓皮肤皱纹的功效作用,通过测试其作用于人皮肤成纤维细胞和角质形成细胞,在皮肤功能性基因层面验证其抗皱功效。结果表明,5 mg/mL RHCⅢ作用于人皮肤成纤维细胞可显著促进胶原合成相关基因CollagenⅠ,CollagenⅢ和Smad3的表达;同时,在UVA刺激条件下可显著抑制胶原降解相关基因MMP-1,MMP-3,c-jun和c-fos的表达。作用于角质形成细胞可显著促进皮肤屏障修复和保湿相关基因LOR,TGM1,IVL,OCLN1,DSG1,FLG,Caspase14和K1的表达。综上所述,样品重组人源化Ⅲ型胶原蛋白在5 mg/mL的给药质量浓度下具有抗皱功效。  相似文献

采用高温抽提和分级醇沉,制备出了大、中、小不同重均分子量的YM-30、YM-50和YM-90甘露聚糖,测定其溶解度、pH、重均分子量及黏度,研究了它们的吸湿性、保湿性。结果表明:YM-30、YM-50和YM-90的重均分子量分别为1.24×10~5Da,4.47×10~4Da和2.16×10~4Da。YM-30不溶或微溶于水,YM-50和YM-90则易溶于水。YM-30溶液偏碱性,YM-50溶液为中性,YM-90溶液则偏酸性。YM-50和YM-90水溶液的黏度很低,几乎与水的黏度一样,且其黏度不受甘露聚糖质量浓度和温度的影响。红外光谱分析表明,3种甘露聚糖的糖链均为β-D-吡喃结构。在低相对湿度(43%)下,YM-90吸湿性最强;而在高相对湿度(81%)下,YM-30吸湿性最强。在相对湿度43%下,YM-30保湿性最优,其保湿率达到了98.4%,稍高于透明质酸。酵母甘露聚糖具有优良的吸湿、保湿性能。  相似文献

利用膦鞣革屑制备胶原蛋白类除醛剂   总被引:1,自引:0,他引:1  
以有机膦鞣革屑为原料,采用碱-酶两步法提取胶原蛋白,单因素实验结果表明:在m(氧化镁)∶m(革屑)=0.06∶1,反应温度70℃,反应时间2.5 h,m(碱性蛋白酶)∶m(革屑)=0.006∶1的条件下,提胶率达88%。采用多媒体显微镜、红外光谱仪以及凝胶渗透色谱仪对最优条件下提取的胶原蛋白进行了结构分析,并与市售的明胶水解产物做了分析对比。结果表明:提取的胶原蛋白与市售的明胶水解产物的谱图基本一致,市售的明胶水解产物的平均相对分子质量为1 346和564,而提取的胶原蛋白的平均相对分子质量较大,为1 928和755,说明提取的物质为胶原多肽和氨基酸的混合物。应用实验结果表明:当胶原多肽溶液的用量为80 mL/m3时,甲醛去除率可达81%。  相似文献

BACKGROUND: The large amount of limed bovine split wastes discharged by the leather industry has raised concerns regarding their environmental effect. The objective of this work was to perform pilot plant trials to extract high‐value native collagen from these wastes through improved pretreatment methods. RESULTS: EDTA‐ and HCl‐pretreatment gave similar removal percentages of inorganic substances. Owing to the open structure of fibers, the collagen yield of HCl‐pretreated splits (HPS) (41.31%) was higher than that of EDTA‐pretreated splits (EPS) (10.42%). Furthermore, HCl‐pretreated split collagen (HPC) had a more acidic isoelectric point, lower content of primary amino groups, larger Z‐average particle size and higher relative viscosity than EDTA‐pretreated split collagen (EPC). Electrophoretic analysis and circular dichroism spectra revealed the maintenance of polypeptide and triple helix conformation, respectively. In addition, the transition temperatures of EPC (34.7 °C) and HPC (34.6 °C) detected by differential scanning calorimetry (DSC) were close to that of commercial collagen from calfskin (CCC) (35.7 °C). CONCLUSION: A process of native collagen extraction from limed bovine split wastes was proposed. While both EPC and HPC represented similar physicochemical properties to those of CCC, the collagen yield of HPS was much higher than that of EPS. 通过干燥器控制湿度的方法对魔芋葡甘聚糖、羧甲基纤维素钠、壳聚糖、海藻酸钠等多糖与常用保湿剂甘油进行了吸湿和保湿性能测试.结果表明,在高相对湿度条件下,各试样的吸湿能力依次为:羧甲基纤维素钠>甘油>海藻酸钠>魔芋萄甘聚糖>壳聚糖;在低相对湿度条件下,各试样的吸湿能力依次为:甘油>羧甲基纤维素钠>海藻酸钠>魔芋葡甘聚糖>壳聚糖.各试样的保湿能力依次为:壳聚糖>魔芋葡甘聚糖>海藻酸钠>甘油>羧甲基纤维素钠.此外,采用热重分析法(TGA)对各试样的失重率进行了测试,其失重曲线与保湿曲线趋势一致.  相似文献

以海蜇、牛骨和鳕鱼皮胶原为原料,采用酶解制备了不同分子量(1 kDa和1~3 kDa)的6种胶原肽。利用透皮扩散试验仪,考察了时间、分子量及质量浓度对胶原肽透皮吸收性的影响,通过荧光显微镜观察了异硫氰酸荧光素(FITC)标记的胶原肽对小鼠皮肤的透皮吸收过程。结果表明,6种胶原肽的质量浓度均为10 g/L时,其在24 h时透皮吸收能力由大到小依次为:1 kDa鳕鱼皮胶原肽、1 kDa牛骨胶原肽、1 kDa海蜇胶原肽、1~3 kDa海蜇胶原肽、1~3kDa牛骨胶原肽和1~3 kDa鳕鱼皮胶原肽;1 kDa胶原肽透皮吸收能力优于1~3 kDa胶原肽。牛骨和海蜇胶原肽的质量浓度越大,透皮吸收性越好,而鳕鱼皮胶原肽则相反。3种来源胶原肽单位面积累积透过量均随时间延长而增加,分子量越大透皮吸收性能越差。  相似文献

Collagen solutions in the presence of 0.1–2.0M acetic acid (AA) were investigated to understand the interaction between collagen molecules and an acidic solvent. Fluorescence measurements of pyrene showed that the critical aggregation concentration (CAC) of collagen increased from 0.518 to 1.581 mg/mL for AA concentrations ranging from 0.1 to 2.0M, indicating that the aggregated state of collagen molecules was associated with AA concentration. The size of the collagen aggregates, determined by dynamic light scattering, demonstrated that their disaggregation was enhanced with increasing electrostatic repulsion between the collagen chains. The variations in the intrinsic viscosity and Huggins coefficient depended on the molecular interaction among the collagen molecules. Furthermore, the increased AA concentration has a different influence on the rigidity of collagen molecules above and below the CAC. This observation was manifested by the changes in the morphology of collagen molecules observed by atomic force microscopy. © 2017 Wiley Periodicals, Inc. J. Appl. Polym. 根据皮肤的组织结构与生理功能,研制了复合天然三维网络结构胶原组织工程支架材料,并以此为支架材料建立了3层结构的人工皮肤模型:上层是具有良好生物相容性且可降解的聚合物涂层,可以屏蔽细菌并保持人工皮肤良好的透水气性;中间是复合型天然三维网络结构胶原纤维支架材料层,为细胞的增殖提供三维空间和良好的生长环境,起到真皮基质的作用;下层是药物层,具有营养、抗菌消炎、刺激细胞生长的作用。  相似文献

分别采用酸法和酶法提取驴皮胶原蛋白,并测定了所提取胶原蛋白的性质。结果表明,两种方法提取的胶原蛋白紫外最大吸收峰都在234 nm处,在280 nm处吸收较小;两者红外吸收光谱相似,并且都具有三股螺旋结构;电泳图谱分析所得胶原蛋白的亚基组成形式为(α1)2α2,推测所提取的是Ⅰ型胶原蛋白;DSC(示差量热扫描法)测定发现,酶溶性胶原蛋白的热收缩温度(65.33℃)高于酸溶性胶原蛋白(50.90℃);羟脯氨酸测定发现,酶法提取驴皮胶原蛋白的纯度高于酸法提取驴皮胶原蛋白的纯度。  相似文献

以猪皮为原料,在柠檬酸和胃蛋白酶协同作用下提取胶原蛋白。采用旋转流变仪系统考察了浓度、pH、剪切速率、NaCl和CaCl2、乙醇和丙三醇对胶原蛋白粘度的影响。结果显示,胶原蛋白溶液粘度随浓度增大呈指数增加趋势;pH=3时粘度最大,pH=4和pH=6-9时粘度下降,pH=10时粘度又陡然回升;粘度随剪切速率的增大呈对数下降趋势;NaCl和CaCl2的添加都会使胶原蛋白溶液粘度下降;乙醇和丙三醇添加量在小于10%时,其添加浓度与粘度成正比例关系。通过对胶原蛋白粘度特性的研究,为制备新型胶原基医用粘合剂提供了理论基础。  相似文献

Collagen is a natural polymer much available as a waste material. However, a more extensive use of collagen as an engineering material is restricted due to its low mechanical strength. This work presents a collagen–microcapsules system and evaluates the hypothesis that a self‐healing ability can be imposed on a collagen matrix to improve its tensile strength. Collagen films containing various types of microcapsules were studied using tensile tests (TTs). To evaluate the self‐repairing capacity of the developed system, samples were submitted to a 20% strain (to develop micro‐cracks in the material that could lead to the rupture of microcapsules and release of diisocyanate) and then left to stand for 20 min. A new TT was performed, named paused tensile test (PTT). The samples of pure collagen underwent no alteration with respect to their mechanical properties comparing TT and PTT results. In contrast, some samples containing microcapsules presented an increase of around 70% in mechanical strength when compared with the pure collagen results, proving that the released diisocyanates were effective in reacting with collagen and improving its mechanical strength. This type of behavior and the observed enhanced property can extend the usage of collagen to a variety of engineering applications. © 2016 Society of Chemical Industry  相似文献

Gelatin films containing unsaturated lipid have been exposed to ultraviolet and visible irradiation. No sign of paramagnetism could be detected in the films, although the gelatin was undergoing cross-linking reactions. The addition of nitroxyl-forming radical scavengers decreased the rate of cross-linking, as did addition of ascorbic acid to the reacting mixture. Nitroxyls could not be detected in the gels, however. The conclusion is drawn that the main reaction in the cross-linking reaction of collagen is a condensation of amino groups and extrinsic or intrinsic carbonyl groups. The extrinsic aldehydes are formed in the autoxidation of unsaturated lipid.  相似文献
