This multicenter, parallel group study determined plasma phospholipid and red blood cell (RBC) phosphatidylcholine and phosphatidylethanolamine fatty acids, plasma cholesterol, apo A-1 and B, growth and visual acuity (using the acuity card procedure) in term infants fed from birth to 90 d of age with formula containing palm-olein, high oleic sunflower, coconut and soy oil (22.2% 16∶0, 36.2% 18∶1, 18% 18∶2n−6, 1.9% 18∶3n−3) (n=59) or coconut and soy oil (10.3% 16∶0 18∶6% 18∶1, 34.2% 18∶2n−6, 4.7% 18∶3n−3) (n=57) or breast-fed (n=56) with no formula supplementation. Different centers in North America were included to overcome potential bias due to differences in n−6 or n−3 fatty acids at birth or in breast-fed infants that might occur in a single-site study. Plasma and RBC phospholipid docosahexaenoic acid (DHA, 22∶6n−3) and arachidonic acid (AA, 20∶4n−6), cholesterol and apo B were significantly lower in the formula- than breast-fed infants. There were no differences in looking acuity or growth among the breast-fed and formula-fed infants. No significant relations were found between DHA and looking acuity, or AA and growth within or among any of the infant groups. This study provides no evidence to suggest the formula provided inadequate n−6 or n−3 fatty acids for growth and looking acuity for the first 3 mon after birth.  相似文献

To test if linolenic acid (18∶3n−3) from vegetable oils would affect bleeding times and platelet counts in new-borns, piglets were used as a model fed milk replacer diets containing 25% (by wt) vegetable oils or oil mixtures for 28 d and compared to sow-reared piglets. The oils tested included soybean, canola, olive, high oleic sunflower (HOAS), a canola/coconut mixture and a mixture of oils mimicking canola in fatty acid composition. All piglets fed the milk replacer diets showed normal growth. Bleeding times increased after birth from 4–6 min to 7–10 min by week 4 (P<0.001), and were higher in pigs fed diets containing 18∶3n−3, as well as in sowreared piglets receiving n−3 polyunsaturated fatty acids (PUFA) in the milk, as compared to diets low in 18∶3n−3. Platelet numbers increased within the first week in newborn piglets from 300 to 550×109/L, and remained high thereafter. Milk replacer diets, containing vegetable oils, generally showed a transient delay in the rise of platelet numbers, which was partially associated with an increased platelet volume. The oils showed differences in the length of delay, but by the third week of age, all platelet counts were >500×109/L. The delay in rise in platelet counts appeared to be related to the fatty acid composition of the oil, as the effect was reproduced by a mixture of oils with a certain fatty acid profile, and disappeared upon the addition of saturated fatty acids to the vegetable oil. There were no alterations in the coagulation factors due to the dietary oils. Blood plasma, platelets and red blood cell membranes showed increased levels of 18∶3n−3 and long-chain n−3 PUFA in response to dietary 18∶3n−3. The level of saturated fatty acids in blood lipids was generally lower in canola and HOAS oil-fed piglets as compared to piglets fed soybean oil or reared with the sow. The results suggest that consumption of milk replacer diets containing vegetable oils rich in 18∶3n−3 does not represent a bleeding risk, and that the transient lower platelet count can be counterbalanced by the addition of saturated fatty acids to the vegetable oils.  相似文献

The seed oil from a genetically transformed canola (Brassica napus) containing 43% (w/w) of γ-linolenic acid (G, 18∶3n−6), 22% linoleic acid (L, 18∶2n−6), and 16% oleic acid (O, 18∶1n−9) was evaluated. In this high γ-linolenic acid canola oil (HGCO), the predominant 18∶3n−6-containing triacylglycerol (TG) molecular species were GGL (23%), GLO (20%), and GGG (11%). In the total TG, approximately 75% of the 18∶3n−6 was located at the sn-1,3 positions, while only 34% of linoleic acid was at the sn-1,3 positions. The GGL molecular species of HGCO contained approximately equal amounts of GLG and GGL positional isomers, while the GLO molecular species had 95% GOL and 5% GLO isomers. The general characteristics and the tocopherol and phytosterol contents were mostly similar between HGCO and nontransformed canola oil. No detectable amounts of amino acids and nucleotides were observed in the HGCO.  相似文献

Docosahexaenoic acid (22∶6n−3) can be synthesized in the liver and/or brain from α-linolenic acid (18∶3n−3) and is required in large amounts in structural membranes of developing brain and retina. The adequacy and efficacy of formulas containing 18∶3n−3 and/or fish oil in providing 22∶6n−3 for deposition was investigated in piglets fed formula from birth to 15 days. The test formulas contained high (HL) or low (LL) 18∶3n−3 (3.9 or 0.7% of the total formula fatty acids, respectively), or low 18∶3n−3 plus fish oil (LL+FO) to provide C20 and C22 n−3 polyunsaturated fatty acids (0.8% of total fatty acids). Fatty acid analyses of synaptic plasma membrane and retina ethanolamine phospholipids (EPL), which are especially enriched in 22∶6n−3, were compared to those of 15-day-old piglets fed sow milk (SM). Feeding LL resulted in lower 22∶6n−3 in synaptic plasma membrane. Fatty acid levels in HL and LL+FO piglets were equivalent to SM, with the exception of lower 22∶5n−3 in the synaptic plasma membrane of LL+FO and in the retina of HL and LL+FO-fed piglets. Levels of 22∶4n−6 were also lower in the retina of the LL+FO group. The results suggest formula 18∶3n−3 is at least 24% as effective as C20 and C22 n−3 fatty acids as a source of membrane 22∶6n−3. This study shows dietary 18∶3n−3, as the only n−3 fatty acid, can support deposition of comparable percentage of 22∶6n−3 to natural milk. Fish oil also supported tissue levels of 22∶6n−3 similar to natural milk; however, lower 22∶4n−6 may indicate possible inhibitory effects on n−6 metabolism. The effect of fish oil high in docosahexaenoic acid (22∶6n−3) and low in eicosapentaenoic acid (20∶5n−3) in formula on blood lipids and growth of full-term infants was studied. Infants were fed formula with about 15% oleic acid (18∶1), 32% linoleic acid (18∶2n−6), 4.9% linolenic acid (18∶3n−3) and 0, 0.10 or 0.22% 22∶6n−3, or 35% 18∶1, 20% 18∶2n−6, 2.1% 18∶3n−3 and 0, 0.11 or 0.24% 22∶6n−3 from 3 d to 16 wk of age (n=16, 18, 17, 21, 17, 16, respectively). The formulae had <0.1% 20∶5n−3 and no arachidonic acid (20∶4n−6). Breast-fed infants (n=26) were also studied. Plasma phospholipid and red blood cell (RBC) phosphatidylcholine (PC) and phosphatidylethanolamine (PE) fatty acids were determined at 3 d and 4, 8, and 16 wk of age. These longitudinal analyses showed differences in blood lipid 22∶6n−3 between breast-fed and formula-fed infants depending on the feeding duration. At 16 wk, infants fed formula with 0.10, 0.11% 22∶6n−3, or 0.22% 22∶6n−3 had similar 22∶6n−3 levels in the plasma phospholipid and RBC PC and PE compared with breast-fed infants and higher 22∶6n−3 than infants fed formula without 22∶6n−3. Formula with 0.24% 22∶6n−3, however, resulted in higher plasma phospholipid 22∶6n−3 than in breast-fed infants at 16, but not 4 or 8 wk of age. Plasma and RBC phospholipid 20∶4n−6 was lower in formula-fed than breast-fed infants, but no differences in growth were found. Higher blood lipid C20 and C22 n−6 and n−3 fatty acids in infants fed formula with 20% 18∶2n−6 and 2.4% 18∶3n−3 compared with 32% 18∶2n−6 and 4.9% 18∶3n−3 show the increase in blood lipid 22∶6n−3 in response to dietary 22∶6n−3 depending on other fatty acids in the formula.  相似文献

The sciatic nerve of rats fed sunflower oil (6 mg 18∶3n−3/100 g of diet) presented dramatic alterations in the long chain polyunsaturated fatty acids in comparison with those fed soy oil (130 mg 18∶3n−3/100 g of diet). In both 15-day-old and 60-day-old animals fed sunflower oil, 22∶6n−3 (cervonic acid) was fourfold less, 22∶5n−6 was 10-fold greater; adrenic acid (22∶4n−6) was slightly greater and arachidonic acid (20∶4n−6) was close to that in rats fed soy oil. The percentage distribution of total polyunsaturated fatty acids as well as the individual saturated and monounsaturated fatty acids were the same in both groups. When the sunflower oil-fed animals were switched to a soy oil-containing diet for either 15 or 60 days, the percentage distribution of 22∶6n−3 increased slowly to reach the control value 2.5 months later. Conversely 22∶5n−6 decreased slowly. The decay of 22∶5n−6 was more rapid than the increase of 22∶6n−3.  相似文献

Conifer (pine) seeds are a potential source of dietary oils, but their safety and nutritional properties are not well established. Conifer seed oils differ from common edible vegetable oils in having a series of unusual polyunsaturated fatty acids (PUFA) with a polymethylene-interrupted (PMI) double bond system and a double bond at the Δ5 position. A rat study was conducted to assess whether Δ5 PMI-PUFA of conifer seeds could alter the levels of n−6 and n−3 long-chain polyunsaturated fatty acids (LC-PUFA) in mothers' milk and the developing brain of fetuses and pups. Feeding maritime pine (Pinus pinaster) seed oil (MPO) diet with a Δ5 PMI-PUFA content of 1.4 g/100 g throughout pregnancy and lactation resulted in a large incorporation of Δ5 PMI-PUFA in mothers' milk (5.1±0.5% of total fatty acids). The fetus (17 d old) and pup (22 d) brains, however, accumulated very little (0.6 and 0.4% of total fatty acids, respectively) Δ5 PMI-PUFA. Mother's milk and pup's brain of the MPO group contained normal levels of 20∶4n−6, 22∶4n−6, and 20∶5n−3 compared to a reference group of rats fed a fat blend of sunflower, high-oleic sunflower, and canola oils. The level of 22∶6n−3, however, was slightly but significantly (P<0.05) higher in milk and pup brain of the MPO group. These results show that Δ5 PMI-PUFA of MPO exert no negative effect on the levels of n−6 and n−3 LC-PUFA in rat brain during its early development.  相似文献

The health benefits of long-chain n−3 PUFA (20∶5n−3 and 22∶6n−3) depend on the extent of incorporation of these FA into plasma and tissue lipids. This study aimed to investigate the effect of the background dietary fat (saturated, monounsaturated, or n−6 polyunsaturated) on the quantitative incorporation of dietary 18∶3n−3 and its elongated and desaturated products into the plasma and the liver lipids of rats. Female weanling Wistar rats (n=54) were randomly assigned to six diet groups (n=9). The fat added to the semipurified diets was tallow (SFA), tallow plus linseed oil (SFA-LNA), sunola oil (MUFA), sunola oil plus linseed oil (MUFA-LNA), sunflower oil (PUFA), or sunflower oil plus linseed oil (PUFA-LNA). At the completion of the 4-wk feeding period, quantitative FA analysis of the liver and plasma was undertaken by GC. The inclusion of linseed oil in the rat diets increased the level of 18∶3n−3, 20∶5n−3, and, to a smaller degree, 22∶6n−3 in plasma and liver lipids regardless of the background dietary fat. The extent of incorporation of 18∶3n−3, 20∶5n−3, and 22∶5n−3 followed the order SFA-LNA>MUFA-LNA>PUFA-LNA. Levels of 22∶6n−3 were increased to a similar extent regardless of the type of major fat in the rat diets. This indicates that the background diet affects the incorporation in liver and plasma FA pools of the n−3 PUFA with the exception of 22∶6n−3 and therefore the background diet has the potential to influence the already established health benefits of long-chain n−3 fatty acids.  相似文献

Increased mean platelet volume (MPV) has been suggested as an independent risk factor for acute myocardial infarction and the increased reactivity of large platelets. The aim of this study was to investigate the correlation between platelet phospholipid (PL) PUFA composition and MPV in 139 free-living healthy men ages 20–55 yr (vegans, n=18; ovolacto vegetarians, n=43; modrerate meat-eaters, n=60; and high meateaters, n=18). Each subject completed a semiquantitative Food Frequency Questionnaire and gave a blood sample. Platelet PL FA composition and MPV were determined by standard methods. MPV was significantly greater in the vegans than in the ovolacto vegetarian, moderate, or high meat-eater groups (P<0.01). Both vegan and ovolacto vegetarian groups had significantly higher platelet PL 18∶2n−6 and 22∶4n−6, and lower 20∶5n−3 and 22∶6n−3 compared with the moderate and high meat-eater groups. The vegans demonstrated a significant reduction in 20∶4n−6 and 22∶5n−3 compared with the ovolacto vegetarian, high meat-eater, and moderate meat-eater groups. Bivariate analysis results showed that MPV was significanly positively correlated with platelet PL 18∶2n−6 (P=0.048) and negatively correlated with 20∶3n−6 (P=0.02), 20∶5n−3 (P=0.005), and 22∶5n−3 (P<0.0001), respectively. In a multiple linear regression analysis, after controlling for potential confounding factors such as dietary group, age exercise, body mass index, and dietary polyunsaturated and saturated fat, cholesterol, carbohydrate, and fiber intake, the MPV was still strongly negatively correlated with platelet PL 20∶3n−6 (P=0.003) and 22∶5n−3 (P=0.001). The present data suggest that 20∶5n−3 and 20∶3n−6 may play a role in the structural function of the platelet membrane.  相似文献

Egg yolk was enriched with α-linolenic acid (18∶3n−3) by feeding laying hens diets containing flax, canola or soybean seeds. Fertilized eggs were incubated and the fatty acid composition of whole body, liver, plasma, brain and the cholesterol content of plasma and liver tissue of the hatched chicks were studied. Eggs enriched with 18∶2n−6 fatty acids by feeding hens diets containing sunflower seeds were used as the controls. Feeding flax enriched (P<0.05) egg yolk and the developing progeny with 18∶3n−3, 20∶5n−3, 22∶5n−3 and 22∶6n−3. Feeding sunflower seeds resulted in an increase (P<0.05) of 18∶2n−6, 20∶4n−6, 22∶4n−6 and 22∶5n−6. The predominant polyunsaturated fatty acid of the brain was docosahexaenoic acid (22∶6n−3) which was higher (P<0.05) in the flax and canola fed group. The cholesterol content of the liver tissue was lower (P<0.05) in chicks hatched from hens fed flax seeds. This study indicates that 18∶3n−3 and 18∶2n−6 in the maternal diet are potent modulators of long-chain polyunsaturated n−3 or n−6 fatty acid and of cholesterol content in the developing progeny.  相似文献

This study was undertaken to determine whether the neonate was more susceptible to the effects of dietary erucic acid (22∶1n−9) than the adult. Newborn piglets were used to assess the safety of different levels of 22∶1n−9 on lipid and histological changes in the heart. Newborn piglets showed no myocardial lipidosis as assessed by oil red 0 staining, but lipidosis appeared with consumption of sow milk and disappeared by seven days of age. Milk replacer diets containing soybean oil, or rapeseed oil mixtures with up to 5% 22∶1n−9 in the oil, or 1.25% in the diet, gave trace myocardial lipidosis. Rapeseed oil mixtures with 7 to 42.9% 22∶1n−9 showed definite myocardial lipidosis in newborn piglets, which correlated to dietary 22∶1n−9, showing a maximum after one week on diet. The severity of the lipidosis was greater than observed previously with weaned pigs. There were no significant differences among diets in cardiac lipid classes except for triacylglycerol (TAG), which increased in piglets fed a repeseed oil with 42.9% 22∶1n−9. TAG showed the highest incorporation of 22∶1n−9, the concentration of 22∶1n−9 in TAG was similar to that present in the dietary oil. Among the cardiac phospholipids, sphingomyelin and phosphatidylserine had the highest, and diphosphatidylglycerol (DPG) the lowest level of 22∶1n−9. The low content of 22∶1n−9 in DPG of newborn piglets is not observed in weaned pigs and rats fed high erucic acid rapeseed oil. The relative concentration of saturated fatty acids was lowered in all cardiac phospholipids of piglets fed rapeseed oils, possibly due to the low content of saturated fatty acids in rapeseed oils. The results suggest that piglets fed up to 750 mg 22∶1n−9/kg body weight/day showed no adverse nutritional or cardiac effects.  相似文献

We have utilized transgenic technology to develop a new source of γ-linolenic acid (GLA) using the canola plant as a host. The aim of the present study was to compare the growth and fatty acid metabolism in rats fed equal amounts of GLA obtained from the transgenic canola plant relative to GLA from the borage plant. Young male Sprague-Dawley rats (n=10/group) were randomized and fed a purified AIN93G diet (10% lipid by weight) containing either a mixture of high GLA canola oil (HGCO) and corn oil or a control diet containing borage oil (BO) for 6 wk. GLA accounted for 23% of the triglyceride fatty acids in both diets. Growth and diet consumption were monitored every 2–3 d throughout the study. At study termination, the fatty acid composition of the liver and plasma phospholipids was analyzed by gas chromatography. The growth and diet consumption of the HGCO group were similar to the BO group. There were no adverse effects of either diet on the general health or appearance of the rats, or on the morphology of the major organs. There was no significant difference between the diet groups for total percentage of n−6 polyunsaturated fatty acids present in either the total or individual phospholipid fractions of liver or plasma. The relative percentage of GLA and its main metabolite, arachidonic acid, in each phospholipid fraction of liver or plasma were also similar between groups. The percentage of 18∶2n−6 in liver phosphatidylethanolamine and phosphatidylinositol/serine was higher (P<0.05) and 22∶5n−6 was lower in the HGCO group than the BO group. This finding could be attributed to the higher 18∶3n−3 content in the HGCO diet than the BO diet. Results from this long-term feeding study of rats show for the first time that a diet containing transgenically modified canola oil was well-tolerated, and had similar biological effects, i.e., growth characteristics and hepatic metabolism of n−6 fatty acids, as a diet containing borage oil.  相似文献

The metabolism of α-linolenic acid from canola oil was studied in eight normolipidemic men. The 42-day study was divided into three periods: a 6-day pre-experimental and two 18-day experimental. Approximately 75% of the dietary fat (28% of total energy) was provided by a mixture of fats during the pre-experimental period and either canola oil (CO) or sunflower oil (SO) during the experimental periods. The CO and SO diets were fed in a cross-over design. The ratios of linoleic to linolenic acid were 2.6∶1 and 73.9∶1 in the CO and SO diets, respectively. Dietary fat source had an effect on plasma phospholipid fatty acids: 18∶1n−9, 18∶3n−3 and 20∶5n−3 were higher (p<0.05), and 18∶2n−6 was lower in the phosphatidylcholine fraction; 18∶1n−9 was higher and 20∶4n−6 lower in the phosphatidyl-ethanolamine fraction; and 18∶1n−9 and 20∶5n−3 were higher and 20∶4n−6 and 22∶6n−3 were lower in the alkenylacyl-ethanolamine phospholipid fraction on the CO diet as compared to the SO diet. Consumption of the canola oil diet resulted in higher n−3 fatty acid levels and lower n−6 fatty acid levels in plasma phospholipids than consumption of the sunflower oil diet.  相似文献

Low-fat diets and diets containing n−3 fatty acids (FA) slow the progression of renal injury in the male Han:Sprague-Dawley (SPRD)-cy rat model of polycystic kidney disease. To determine whether these dietary fat effects are similar in females and in another model of renal cystic disease, in this study we used both male and female pcy mice to examine the effects of fat level and type on disease progression. Adult pcy mice were fed 4, 10, or 20 g soybean oil/100 g diet for 130 d in study 1. In study 2, weanling pcy mice were fed high or low levels of fat rich in 18∶2n−6 (corn oil, CO) 18∶3n−3 (flaxseed oil/CO 4∶1 g/g, FO), or 22∶6n−3 (algal oil/CO 4∶1 g/g, DO) for 8 wk. In adult pcy mice, low-compared with high-fat diets lowered kidney weights (2.4±0.2 vs. 3.1±0.2 g/100 g body weight, P=0.006) and serum urea nitrogen (SUN) (9.6±0.6 vs. 11.9±0.6 mmol/L, P=0.009), whereas in young pcy mice it reduced renal fibrosis volumes (0.44±0.04 vs. 0.62±0.04 mL/kg body weight, P<0.0001). FO feeding in young pcy mice mitigated the detrimental effects of high fat on fibrosis while not altering kidney size, function, and oxidative damage when compared with the CO-fed mice. In contrast, DO-compared with CO-fed mice had higher kidney weights (2.64±0.07 vs. 2.24±0.08 g/100 g body weight, P=0.005), SUN (9.4±0.57 vs. 7.0±0.62 nmol/L, P<0.0001), and cyst volumes (7.9±0.28 vs. 6.2±0.30 mL/kg body weight, P<0.0001) and similar levels of oxidative damage and fibrosis. The FA compositions of the diets were reflected in the kidneys: 18∶2n−6, 18∶3n−3, and 22∶6n−3 were the highest in the CO, FO, and DO diets, respectively. Dietary effects on kidney disease progression were similar in males and females. A low-fat diet slows progression of renal injury in male and female pcy mice, consistent with findings in the male Han:SPRD-cy rat. Dietary fat type also influenced renal injury, with flaxseed oil diets rich in 18∶3n−3 slowing early fibrosis progression compared with diets rich in 18∶2n−6 or in 22∶6n−3.  相似文献

Cardiolipin (CL) is a unique mitochondrial phospholipid, containing up to 85 wt% 18∶2n−6 in mammals. The influence of maternal dietary fatty acids on the acyl composition of offspring CL has not been examined previously. Adult female mice were thus fed diets rich in 18∶1n−9 (olive oil), 18∶2n−6 (safflower oil), 18∶3n−3 (linseed oil) or 20∶5n−3 and 22∶6n−3 (fish oil/safflower, 9∶1, w/w), for a five month period, encompassing two breeding cycles. Offspring from the second breeding cycle were then fed these diets. The acyl composition of CL, phosphatidylcholine and phosphatidylethanolamine from liver and heart was evaluated from mice killed 3, 18 and 42 days after parturition. The primary nutrient sources at these three time points were transplacental nutrients, breast milk and the diet, respectively. Maternal diet was found to influence the acyl composition of CLvia both placental transfer of fatty acids and breast milk. Fish oil feeding resulted in replacement of a substantial portion of 18∶2n−6 with 22∶6n−3; after 42 days, the area% of 18∶2n−6 in heart CL was reduced from 62% in safflower oil fed mice to 12%. In comparison to fish oil feeding, linseed oil feeding resulted in a much lower accumulation of 22∶6n−3. Olive oil feeding resulted in substantial replacement of 18∶2n−6 with 18∶1n−9 (18∶2n−6 was reduced from 62% to 31%). Physiologically, these findings are relevant because changes in CL acyl composition may influence the activity of associated inner mitochondrial membrane enzymes. Diet and fatty acid metabolism interact in yet unknown ways to modulate membrane fatty acid composition and certain cellular functions. For example, dietary precursors or metabolic products of n-3 fatty acid metabolism differ in their ability to modify specific membrane components. In the present study, the effect of dietary 22∶6n−3 or its metabolic precursor, 18∶3n−3, on the selective accumulation of 22∶6n−3 by heart was investigated. The mass and fatty acid compositions of individual phospholipids (PL) in heart and liver were quantified in mice fed either 22∶6n−3 (from crocodile oil) or 18∶3n−3 (from soybean oil) for 13 wk. This study was conducted to determine if the selective accumulation of 22∶6n−3 in heart was due to the incorporation of 22∶6n−3 into cardiolipin (CL), a PL most prevalent in heart and known to accumulate 22∶6n−3. Although heart was significantly enriched with 22∶6n−3 relative to liver, the accumulation of 22∶6n−3 by CL in heart could not quantitatively account for this difference. CL from heart did accumulate 22∶6n−3, but only in mice fed preformed 22∶6n−3. Diets rich in non-22∶6n−3 fatty acids result in a fatty acid composition of phosphatidylcholine (PC) in heart that is unusually enriched with 22∶6n−3. In this study, the mass of PC in heart was positively correlated with the enrichment of 22∶6n−3 into PC. The increased mass of PC was coincident with a decrease in the mass of phosphatidylethanolamine, suggesting that 22∶6n−3 induced PC synthesis by increasing phosphatidylethanolamine-N-methyltransferase activity in the heart.  相似文献

The esterification, desaturation, and elongation of [1-14C]18∶3n−3, [1-14C]18∶2n−6, and [1-14C]20∶5n−3 at 5 and at 12°C were studied using cultivated hepatocytes from Atlantic salmon. The salmon were fed diets, in which 0, 50, or 100% of the supplementary fish oil had been replaced by soybean oil, for 950 day-degrees at 5 and 12°C. The endogenous percentage of 18∶2n−6 in hepatocyte lipids was 2% in cells from fish fed a diet with 100% of the supplemental lipid from fish oil, and it was slightly less than 25% in cells from fish fed the diet with 100% of the supplemental lipid from soybean oil. Furthermore, the percentages of 20∶3n−6 and 20∶4n−6 were significantly higher in hepatocytes from fish fed on soybean oil than they were in those of fish fed on fish oil. The percentages of 20∶5n−3 and 22∶6n−3, on the other hand, were lower. The endogenous levels of n−6 FA were not significantly correlated with the total amounts of radiolabeled FA esterified in hepatocyte lipids. The main radiolabeled products formed from 18∶2n−6 were 20∶2n−6 and 20∶3n−6. The level of the important eicosanoid precursor 20∶4n−6 was twice as high in hepatocyte phospholipids from fish fed the 100% soybean oil diet as it was in hepatocytes from fish fed the diet with 100% of supplemental lipid from fish oil. The main products formed from 18∶3n−3 were 20∶4n−3, 20∶5n−3, and 22∶6n−3. High levels of dietary 18∶2n−6 do allow, or even seem to increase, the production of 22∶6n−3 from 18∶3n−3 in hepatocytes. The main products formed from 20∶5n−3 were 22∶5n−3 and 22∶6n−3. The production of 22∶6n−3 from 20∶5n−3 was higher at 5°C than at 12°C. The percentage of 24∶5n−3 was higher at 5°C than it was at 12°C, as was the ratio of 24∶5 to 22∶5. These results suggest that the elongation rate of 22∶5n−3 to 24∶5n−3 is higher at the lower temperature.  相似文献

Canola oil is not presently permitted in infant formulations in the United States because of lack of information concenring the effects of feeding canola oil to the newborn. We have previously reported a transient decrease in platelet counts and an increase in platelet size in newborn piglets fed canola oil for 4 wk, and have confirmed this in the present study. In canola oil-fed piglets, changes in platelet size and number were overcome by adding either long-chain saturated fatty acids from cocoa butter (16:0 and 18:0), or shorter-chain saturates from coconut oil (12:0 and 14:0). Feeding a high erucic acid rapeseed (HEAR) oil, with 20% 22:1n−9, led to an even greater platelet reduction and increased platelet size throughout the 4-wk trial. Bleeding times were longer in piglets fed canola oil or HEAR oil compared to sow-reared and soybean oil-fed piglets. There were no other diet-related changes. Diet-induced platelet changes were not related to platelet lipid class composition, but there were fatty acid changes. The incorporation of 22:1n−9 into platelet phospholipids of piglets fed canola oil was low (0.2–1.2%), and even for the HEAR oil group ranged from only 0.2% in phosphatidylinositol to 2.4% in phosphatidylserine. A much greater change was observed in the concentration of 24:1n−9 and in the 24:1n−9/24:0 ratio in platelet sphingomyelin (SM). The 24:1n−9 increased to 49% in the HEAR oil group compared to about 12% in animals fed the control diets (sow-reared piglets and soybean oil-fed group), while the 24:1n−9/24:0 ratio increased from about 1 to 12. Even feeding canola oil, prepared to contain 2% 22:1n−9, led to a marked increase in 24:1n−9 to 29% and had a 24:1n−9/24:0 ratio of 5. The canola oil/cocoa butter group, which also contained 2% 22:1n−9, showed a lower level of 24:1n−9 (20%) and the 24:1n−9/24:0 ratio (3) compared to the canola oil group. The results suggest that the diet-related platelet changes in newborn piglets may be related to an increase in 24:1n−9 in platelet SM, resulting from chain elongation of 22:1n−9. The inclusion of canola oil as the sole source of fat in the milk-replacer diets of newborn piglets resulted in significant platelet and lipid changes.  相似文献

Recent studies have demonstrated that long-chain n−3 PUFA (LCn-3PUFA) are beneficial in reducing the risk of cardiac arrhythmias. This study was conducted to determine the extent of incorporation of LCn-3PUFA into human atrium following supplementation with a fish oil concentrate high in LCn-3PUFA. Volunteers preparing for coronary bypass surgery were randomized either to the treatment group (n=8), receiving 6 g/d of fish oil concentrate (4.4 g of LCn-3PUFA), or the placebo group (n=9), receiving 6 g/d of olive oil for a minimum period of 6 wk. Blood samples were collected prior to commencement of treatment, and preoperatively before bypass surgery. Atrial biopsies were obtained during surgery. The plasma and atrium samples were analyzed by GC following trans-methylation to determine FA profile. Post-supplementation, the treatment group had significantly higher plasma levels of 20∶5n−3, 22∶5n−3, and 22∶6n−3 than the placebo group. Analysis of the atrium total lipids revealed a significant increase in the proportion of 20∶5n−3 following fish oil supplementation. There was no significant difference in the concentration of 22∶5n−3 and 22∶6n−3 in the atrium total lipids; however, an upward trend was observed in subjects receiving fish oil supplementation. In the phospholipid fraction of the atrium, both 20∶5n−3 and 22∶6n−3 increased, whereas 20∶4n−6 levels decreased. This study demonstrates for the first time that short-term supplementation with fish oil concentrate results in significant incorporation of LNc-3PUFA with a concomitant depletion of the eicosanoid substrate (20∶4n−6) in the human atrium.  相似文献

Platelet lipid composition is important to normal platelet morphology and function, and is influenced by dietary fatty acids and cholesterol. The fatty acid composition and cholesterol content of infant formulas differs from those of human milk, but the possible effects on platelet lipids in young infants is not known. This was studied in piglets fed from birth to 18 d of age with one of eight formulas differing in saturated fatty acid chain length, or content of 18∶1, 20∶5n−3 plus 22∶6n−3, or cholesterol. A reference group of piglets fed sow milk was also studied. Sow milk has a fatty acid composition and cholesterol content similar to that of human milk. Piglets fed formulas high in 18∶1 (34.9–40.8% wt fatty acids) and low in 16.0 (≤6.5% wt fatty acids) had lower platelet counts and greater platelet size than piglets fed sow milk (40.4% 18∶1, 30.7% 16∶0). Piglets fed formulas high in 16∶0 (27–29.6%) and 18∶1 (40–40.6%), or low in both 16∶0 (5.9–6.1%) and 18∶1 (10.8–11.2%), had similar platelet counts and size to piglets fed sow milk. Platelet phospholipid % 20∶4n−6 was lower in all the groups of piglets fed formula than in the group fed sow milk. Addition of fish oil with 20∶5n−3 plus 22∶6n−3 to the formula further decreased platelet phospholipid 20∶4n−6. Addition of cholesterol to the formula increased the platelet phospholipid % 20∶4n−6 and platelet volume.  相似文献
