Eight sighted male albino rats that had recovered spontaneous ingestive behavior after lesions of the lateral hypothalamus were challenged with acute injections of hypertonic NaCl administered at different times during the day-night cycle. Nine intact controls were also studied. Following these injections, drinking was observed only during the nighttime. After morning injections Ss frequently waited until nightfall before drinking, whereas Ss injected at night showed much shorter delays in the behavioral response; a similar nocturnal predominance of drinking was seen after food deprivation and in the ad-lib situation. Thirty subjects tests on the SAE for 8 hrs during morning and evening periods demonstrated significant ultradian rhythms in perception. The perceptual rhythms were nonstationary with respect to 1) the frequency of the rhythms reflected in some slowing of the rhythms during the course of the testing period, and in frequency differences between the morning and afternoon rhythms, and 2) the level of the illusion reflected in abrupt shifts in the magnitude of the SAE during the course of the testing period without subsequent recovery.  相似文献

The study investigates how the human body composition (BC) changes as a function of the day-night cycle. The BC was investigated using bioelectrical impedance analysis (BIA) of 10 clinically healthy subjects (CHS), monitored in supine position (readings at 2-h intervals), avoiding mealtimes, dietary abuses, and bladder and intestinal retention. Time series data were analyzed for their temporal characteristics and circadian rhythm (CR). All the variables of BC (lean body mass, fat body mass, body cell mass, total body water, intracellular and extracellular body water, sodium and potassium exchangeable pool) showed a within-day variability with nighttime crests. Such an oscillatory synchronism corroborates the hypothesis that the rest time plays a fundamental role, via its anabolic effects, in conferring the nocturnal phase to the CR of the human BC.  相似文献

Many retinal functions are circadian, but in most instances the location of the clock that drives the rhythm is not known. Cultured neural retinas of the golden hamster (Mesocricetus auratus) exhibited circadian rhythms of melatonin synthesis for at least 5 days at 27 degrees celsius. The rhythms were entrained by light cycles applied in vitro and were free-running in constant darkness. Retinas from hamsters homozygous for the circadian mutation tau, which shortens the free-running period of the circadian activity rhythm by 4 hours, showed a shortened free-running period of melatonin synthesis. The mammalian retina contains a genetically programmed circadian oscillator that regulates its synthesis of melatonin.  相似文献

We studied the time structure of pancreatic secretion in two experiments involving seven 6- to 7-wk-old intact male pigs, surgically fitted with a jugular vein catheter for blood sampling, pancreatic catheter, and a duodenal T-cannula for chronic pancreatic juice sampling for 72 h at 30- to 60-min intervals. Pigs were kept in metabolic cages in a regimen of 12 h of light alternating with 12 h of darkness and were fed at 0800, 1500, and 2200 daily a standard diet based on barley, soybean meal, and fish meal. Beginning 4 d after surgical recovery, three 24-h collections of pancreatic juice and blood sampling were begun either at 0700 or 0800 every 2nd d for 5 d. Pancreatic secretion exhibited a pattern characterized by distinct meal-related secretions of the first phase (postprandial), containing large amounts of protein and enzymes (trypsin and chymotrypsin), and by non-food-stimulated secretions of the second phase with less protein and enzymes. During the dark span, the first phase was practically absent; the response of the pancreatic secretion to the 2200 meal was not very pronounced. Apart from the anticipated circadian rhythm demonstrable by single cosinor analysis on a group basis, a prominent 8-h component was almost invariably statistically significant. Moreover, an approximately 3.43-h component was also prominent. These data indicate that pancreatic secretions are circadian periodic and that their response to a standard meal is also circadian-stage dependent. The circadian components may have been free-running because the pigs were adjusting themselves to the changing phase and that resulted in the period being different from exactly 24 h.  相似文献

Studied the development of drinking behavior in 3–20 day old Sprague-Dawley rats by tracing their ability to perform progressively more complex components of the drinking act. Even 3-day-old Ss responded to cellular dehydration by actively swallowing intraoral infusions of milk and water or by licking these fluids when they were spread in a thin film across the floor of the test container. Ss were not able to approach and maintain contact with a distant fluid source until 10–25 days of age. By 20 days of age, dehydration reduced the consumption of milk and other types of food (dehydration anorexia). Many mammalian species display seasonal breeding patterns correlated with annual cycles of change in the physiology and morphology of the reproductive system. Such annual reproductive cycles are often photoperiodically controlled (i.e, the annual change in day length determines when reproductive activity begins and when it ends within the annual cycle). Photoperiodic control of seasonally appropriate changes in reproductive activity is dependent on an endogenous time measuring process. Among mammals the physiological basis of photoperiodic time measurement has been studied most extensively in the golden hamster. Studies with this species indicate that photoperiodic time measurement is executed by the circadian system. The time measuring process depends on a circadian oscillation of responsiveness to light with properties similar to those of the hypothetical rhythm originally proposed by Bünning to explain photoperiodic phenomena in plants. The available evidence strongly suggests the participation of the pineal gland and the suprachiasmatic nucleus (SCN) in the photoperiodic regulation of mammalian reproductive cycles. However, little is known regarding concrete physiological mechanisms, and the extent to which the SCN and the pineal gland may participate in the time measuring process per se remains to be determined.  相似文献

The effect of cage size on spontaneous locomotor rhythms of laboratory mice was studied under simulated light-dark (12:12) cycles. On-line image analysis of bodily displacement yielded a locomotor signal over a period of 3 days. Continuous wavelet transform was applied to the signal, and ensemble averaging of eight mice revealed in the time-frequency plot bouts of increased motor activities. Notably, there were two bouts in the dark corresponding to ultradians of periods below 5 h: a first bout at the dark onset (at 0.6-1.0 cycle/h), and a second bout during the second half of the dark period (at 0.4-0.7 cycle/h). These increases of activity were more intense and distinct when the animals were kept inside the larger cage. Furthermore, the first bout disappeared when the animals were kept in the small cage for 3 days.  相似文献

The daily light-dark cycle synchronizes the internal circadian clock with the outside world. Blind organisms maintain this light-induced entrainment, suggesting the existence of a non-visual phototransduction pathway. Investigated circumstances under which a novel odor could elicit nipple attachment behavior in 3-day-old Sprague-Dawley rats. In Exp I, Ss suckled washed nipples scented with citral (a lemon odor) only if they either had received tactile stimulation (by stroking with a soft brush) or had been administered amphetamine in the presence of citral prior to the suckling test. Ss stimulated in citral's absence or only exposed to citral without stimulation failed to suckle such nipples. In Exp II, Ss stimulated in a benzaldehyde (an almond odor) ambience suckled washed nipples scented with benzaldehyde but not those with citral scent. The opposite held for those stimulated in a citral-rich environment. The stimulus conditions that supported this conditioning were investigated in Exp III. Simultaneously increasing citral concentration and raising ambient temperature markedly attenuated the phenomenon. Exp IV demonstrated that not all classes of stimulation produced conditioning. Caffeine, in a wide range of doses, did not allow citral to elicit suckling on washed nipples. Findings are discussed within a framework of higher-order conditioning and may provide a mechanism by which naturally occurring stimuli come to elicit the species- and age-typical behavior of suckling. Sleep-wake cycle disturbances suggest that circadian rhythms may be disrupted in patients with Alzheimer's disease (AD). In this study, we examined the circadian rhythms of core-body temperature and locomotor activity in 28 patients with probable AD and 10 healthy controls. AD patients had higher percent nocturnal activity than controls, corresponding to the clinical picture of fragmented sleep. The amplitude of the activity cycle in the AD patients was lower than that of controls and the acrophase of this cycle in AD patients was 4.5 h later. There was no difference in the amplitude of the core-body temperature circadian rhythm, but AD patients had delayed temperature acrophases. A subgroup of AD patients with large mean time differences between the acrophases of their activity and temperature cycles had lower temperature amplitudes and greater activity during the night. These findings suggest that a subgroup of AD patients with impaired endogenous pacemaker function may have a diminished capacity to synchronize the rhythm of core-body temperature with the circadian cycle of rest-activity. This circadian rhythm dysfunction may partly explain the fragmented nocturnal sleep exhibited by these patients.  相似文献

Examined the daily patterns of food intake and activity in normal and pontile-lesioned adult cats. Food intake in the dark and light phases of the light–dark (LD) cycles was determined separately by weighing the food, and a percentage nocturnal score was calculated. The measure of activity was infrared photobeam interruptions, with the photobeam placed in front of the cages, over the food bowl. No differences between normal and pontile-lesioned Ss were detected for any of the measures. Ss' food intake was influenced by simulated starlight and moonlight conditions and by the presence of humans. Ss in isolation from humans and human noises exhibited random patterns of activity in constant light and free-running circadian rhythms in constant dark. Idiosyncratic differences in entrainment to LD cycles were found among the Ss. The relevance of this variability is noted for studies of photoperiodic phenomena in this species. When the Japanese quail is held in constant darkness, retinal responses (ERG b-waves) increase during the animal's subjective night and decrease during its subjective day. Rod photoreceptors dominate the b-wave responses (lambdamax = 506 nm) to all stimulus intensities at night but only to those intensities below the cone threshold during the day. Above the cone threshold, cones dominate b-wave responses (lambdamax, approximately 550-600 nm) during the day regardless of the state of retinal adaptation. Apparently a circadian oscillator enables cone signals to block rod signals during the day but not at night. The ERG b-wave reflects the activity of bipolar cells that are postsynaptic to rods and cones. The ERG a-wave reflects the activity of both rods and cones. The amplitude of the isolated a-wave (PIII) changes with time of day, as does that of the b-wave, but its spectral sensitivity does not. The PIII responses are maximal at approximately 520 nm both day and night and may reflect multiple receptor mechanisms. The shift in rod-cone dominance detected with the ERG b-wave resembles the Purkinje shift of human vision but, unlike the Purkinje shift, does not require a change in ambient light intensity. The shift occurs in constant darkness, with a period of approximately 24 hr indicative of a circadian rhythm in the functional organization of the retina.  相似文献

The proximate determinants of fertility framework, developed in its current form by Bongaarts, has been used extensively by researchers for the past 20 years. Since the initial framework was developed, a wealth of new survey data on the proximate determinants has become available. This article reviews the new data and past experiences and suggests modifications to the framework that would take advantage of this experience. The major modifications suggested are (1) the use of sexual activity rather than marriage to indicate exposure to pregnancy; (2) a revision of the sterility index to measure infecundity from all causes; (3) a revised index of contraception that accounts for the fact that users of sterilization may become infecund before age 49; and (4) a revised definition and estimate of total fecundity.  相似文献

In 3 experiments with Long-Evans rats, the presence of an adult at a feeding site profoundly influenced a conspecific weanling's probability of eating there. Presence of an adult at a drinking site did not have a comparable effect. This indicates that the influence of adult presence at a feeding site on pup feeding site selection, reported in previous studies, is not simply an epiphenomenon reflecting a general affiliative tendency of rat pups. Male rats received subcutaneously silastic capsules, containing the aromatase inhibitor 1,4,6-androstatriene-3,17-dione (ATD), shortly after birth. Control males were given silastic capsules containing cholesterol. The capsules were removed at the age of 21 days. In adulthood, blood serum was collected early and late in the dark phase of the light/dark cycle (experiment I). Testosterone and luteinizing hormone and follicle stimulating hormone (FSH) fluctuated nocturnally, both in ATD and control males, with highest levels late in the dark phase. FSH levels were significantly higher in ATD males. Nocturnal levels of inhibin, a selective suppressor of pituitary FSH secretion, also fluctuated in both ATD and control males, with lowest levels late in the dark phase. In experiment II, ATD and control males were tested for partner preference behavior in a three-compartment box (choice: sexually active male vs. estrous female) early and late in the dark phase. When gonadally intact, ATD males, but not controls, showed a clear nocturnal rhythmicity in partner preference behavior and sexual behavior. Early in the dark phase, such ATD males preferred the vicinity of and interaction with a sexually active male. Late in the dark phase, this preference for the active male shifted to a preference for the estrous female. Control males preferred the estrous female. After castration and subsequent treatment with testosterone via silastic capsules, which ensured constant blood serum levels, ATD males continued to show their nocturnal rhythms in partner preference behavior and in sexual behavior. Thus, the underlying mechanism of the nocturnal rhythmicity phenomenon is an organizational effect of neonatal ATD treatment rather than an activational effect of fluctuating serum hormone levels.  相似文献

The circadian rhythms of the night shift worker show very little phase shift in response to the daytime sleep and night work schedule. One strategy for producing circadian adaptation may be to use appropriately timed exposure to high-intensity light. We attempted to shift the circadian temperature rhythms of seven normal subjects while they followed a sleep schedule that gradually delayed (2 h per day) until sleep occurred during the daytime, as is customary for workers during the night shift. After 5 days, the sleep schedule was gradually advanced back to baseline. High illuminance light (2 h per day) and the attenuation or avoidance of sunlight were timed to facilitate temperature rhythm phase shifts. In general, the temperature rhythm did not shift along with the sleep-wake schedule, but appeared either to free run or remain entrained to the natural 24-h zeitgebers. This study showed how difficult it can be to shift human circadian rhythms in the field, when subjects are exposed to competing 24-hr zeitgebers.  相似文献

To evaluate the lifetime carcinogenic hazards of exposure to ionizing radiation during development, 1,680 beagles received whole-body exposures to 60Co gamma rays or sham exposures. Eight groups of 120 dogs each received mean doses of 15.6-17.5 or 80.8-88.3 cGy in early, mid- or late gestation, at 8, 28 or 55 days postcoitus or at 2 days after birth. Another group of 120 dogs received a mean dose of 82.6 cGy as 70-day-old juveniles and one group of 240 dogs received a mean dose of 81.2 cGy as 365-day-old young adults. Sham irradiations were given to 360 controls. Sexes were equally represented. In 1,343 dogs allowed to live out their life span, neoplasia was a major disease, contributing to mortality in 40% of the dogs. There was a significant increase in benign and malignant neoplasms occurring in young dogs (<4 years old), including fatal malignancies, after irradiation in the perinatal (late fetal and neonatal) periods. The lifetime incidence of fatal neoplasms was also increased in dogs irradiated perinatally. Three malignancies-lymphomas, hemangiosarcomas and mammary carcinomas-accounted for 51% of all fatal tumors. There was an apparent lifetime increase and earlier onset of lymphomas in dogs exposed as fetuses. Fatal hemangiosarcomas were increased in dogs irradiated early and late in gestation. Fatal mammary carcinomas were not increased by irradiation, although non-fatal carcinomas were increased after perinatal exposure. Myeloproliferative disorders and central nervous system astrocytomas appeared to be increased in perinatally irradiated dogs. These data suggest that irradiation in both the fetal and neonatal periods is associated with increased early onset and lifetime cancer risk.  相似文献

The effect of water deprivation and water intake on experimental anxiety in rats was tested using burying behavior (BB) and social interaction (SI) anxiety paradigms. Two groups of animals were studied: a control group with free access to water, and a 72-h water-deprived experimental group. Anxiety was studied in a water-deprived group or following a 10-min period of ad lib water drinking. An increase in the mean time of defensive burying in animals deprived for 72 h was observed, whereas an important reduction occurred in the levels of burying behavior immediately after the animals were allowed to drink ad lib for 10 min. These results suggest that the observed increase in defensive burying in the water-deprived animals represents an anxiogenic effect, whereas the decrease in this behavior in water-satiated animals is considered an anxiolytic action. The temporal course of reduction in burying behavior, observed after water drinking, revealed that the anxiolytic action lasts 5 min, whereas 15-30 min after drinking, burying behavior levels were similar to those in the control group. In the social interaction experiment a partial anxiogenic/anxiolytic effect of water deprivation and water intake was observed. The adaptive meaning of anxiogenic and anxiolytic changes linked to consummatory behaviors in rats is discussed on the basis of behavioral and biochemical data.  相似文献
