Perfluorinated compounds (PFCs) can be released to the surrounding environment during manufacturing and usage of PFC containing products, which are considered as main direct sources of PFCs in the environment. This study evaluates the release of perfluorooctane sulfonate (PFOS) and other PFCs to the ambient environment around a manufacturing plant. Among the nine PFCs analyzed, only PFOS, perfluorooctanoic acid (PFOA), and perfluorohexane sulfonate (PFHxS) were found in dust, water, soil, and chicken eggs. Very high concentrations of PFOS and PFOA were found in dust from the production storage, raw material stock room, and sulfonation workshop in the manufacturing facility, with the highest value at 4962 μg/g (dry weight) for PFOS and 160 μg/g for PFOA. A decreasing trend of the three PFCs concentrations in soils, water, and chicken eggs with increasing distance from the plant was found, indicating the production site to be the primary source of PFCs in this region. Risk quotients (RQs) assessment for surface water >500 m away from the plant were less than unity. Risk assessment of PFOS using predicted no-effect concentration (PNEC, 3.23 ng/g on a logarithmic scale) indicated no immediate ecological risk of a reduction in offspring survival. PFOS concentrations in most egg samples did not exceed the benchmark concentration derived in setting a reference dose for noncancer health effects (0.025 μg/(kgxd)).  相似文献

In 2000, 3M Company, the primary global manufacturer, announced a phase-out of perfluorooctanesulfonyl fluoride (POSF, C8F17SO2F)-based materials after perfluorooctanesulfonate (PFOS, C8F17SO3-) was reported in human populations and wildlife. The purpose of this study was to determine whether PFOS and other polyfluoroalkyl concentrations in plasma samples, collected in 2006 from six American Red Cross adult blood donor centers, have declined compared to nonpaired serum samples from the same locations in 2000-2001. For each location, 100 samples were obtained evenly distributed by age (20-69 years) and sex. Analytes measured, using tandem mass spectrometry, were PFOS, perfluorooctanoate (PFOA), perfluorohexanesulfonate (PFHxS), perfluorobutanesulfonate (PFBS), N-methyl perfluorooctanesulfonamidoacetate (Me-PFOSA-AcOH), and N-ethyl perfluorooctanesulfonamidoacetate (Et-PFOSA-AcOH). The geometric mean plasma concentrations were for PFOS 14.5 ng/mL (95% CI 13.9-15.2), PFOA 3.4 ng/ mL (95% CI 3.3-3.6), and PFHxS 1.5 ng/mL (95% CI 1.4-1.6). The majority of PFBS, Me-PFOSA-AcOH, and Et-PFOSA-AcOH concentrations were less than the lower limit of quantitation. Age- and sex-adjusted geometric means were lower in 2006 (approximately 60% for PFOS, 25% for PFOA, and 30% for PFHxS) than those in 2000-2001. The declines for PFOS and PFHxS are consistent with their serum elimination half-lives and the time since the phase-out of POSF-based materials. The shorter serum elimination half-life for PFOA and its smaller percentage decline than PFOS suggests PFOA concentrations measured in the general population are unlikely to be solely attributed to POSF-based materials. Direct and indirect exposure sources of PFOA could include historic and ongoing electrochemical cell fluorination (ECF) of PFOA, telomer production of PFOA, fluorotelomer-based precursors, and other fluoropoly-mer production.  相似文献

Concentration of serum growth hormone was increased and feed intake was reduced in lactating dairy cows fed an 84% concentrate diet supplemented with either 100.8 g MgO or 121.2 g of Mg(OH)2. Analysis of serum growth hormone, feed intake, and serum Mg showed an interaction of serum Mg and feed intake, which suggests low concentrations of serum Mg lessen the increase of concentration of serum growth hormone caused by reduced feed intake. Also, compared with control, treatments with Mg supplementation and reduced feed intake increased growth hormone concentration 12 d into the subsequent treatment.  相似文献

Perfluorochemicals (PFCs) are the subject of increasingly intense environmental research. Despite their detection both in biota and in aqueous systems, little attention has been paid to the possible presence of this class of compounds in solid environmental matrixes. The limited available data indicate that some PFCs such as perfluorooctane sulfonate (PFOS) may strongly sorb to solids, and sewage sludge is widely suspected as a major sink of PFCs entering municipal waste streams. A quantitative analytical method was developed that consists of liquid solvent extraction of the analytes from sediments and sludge, cleanup via solid-phase extraction, and injection of the extracts with internal standards into a high-performance liquid chromatography (HPLC) system coupled to a tandem mass spectrometer (LC/MS/MS). The limits of detections of the method were analyte and matrix dependent, but ranged from 0.7 to 2.2 ng/g and 0.041 to 0.246 ng/g (dry weight) for sludge and sediment, respectively. A demonstration of the method was performed by conducting a limited survey of domestic sludge and sediments. The concentration of PFCs in domestic sludge ranged from 5 to 152 ng/g for total perfluorocarboxylates and 55 to 3370 ng/g for total perfluoroalkyl sulfonyl-based chemicals. Data from a survey of San Francisco Bay Area sediments suggest widespread occurrence of PFCs in sediments at the low ng/g to sub-ng/g level. Furthermore, substances that may be transformed to PFOS, such as 2-(N-ethylperfluorooctanesulfonamido) acetic acid (N-EtFOSAA) and 2-(N-methylperfluorooctanesulfonamido) acetic acid (N-MeFOSAA), are present in both sediments and sludge at levels often exceeding PFOS.  相似文献

Evidence has suggested that the current requirement for vitamin A tabulated by the NRC [(approximately 3800 IU of vitamin A/kg of dry matter (DM)] for dairy calves fed liquid diets is too low. The objective of this study was to assess the effects of vitamin A content in milk replacers on serum and liver vitamin A concentrations, growth, and development of clinical signs of vitamin A deficiency in calves. Male Holstein calves were separated from their dams at birth and given standardized feedings of colostrum and milk replacer for 3 d. On d 4, calves were assigned to five groups and fed milk replacer containing 2300, 6200, 9000, 18,300, or 44,000 IU of vitamin A/kg of DM. Liver biopsies and serum samples were taken on d 4, 9, 15, 21, and 28 to monitor vitamin A concentrations. Weekly physical and neurological examinations were performed to monitor the development of deficiency signs. Fecal scores, body temperature, and the presence of nasal and ocular discharge were recorded daily. Liver vitamin A concentrations in calves allotted to diets with 2300 and 6200 IU of vitamin A/kg decreased from d 4 to 28. Calves fed 9000 IU of vitamin A/kg maintained liver stores, while those fed 18,300 and 44,000 IU of vitamin A/kg had significant increases in hepatic vitamin A. A strong negative association existed between incidence of hyperthermic temperatures and vitamin A concentration in the diet; calves fed 2300 IU of vitamin A/kg had approximately three times more hyperthermic readings than did calves fed other treatments. A strong negative association also existed between fecal score and concentration of vitamin A in the diet; calves fed diets containing low vitamin A concentration had a higher incidence of high fecal scores (more watery) than did calves fed diets with higher vitamin A concentrations. Although slight differences were detected in serum retinol concentration, growth performance and incidence of ocular and nasal discharges were not different among treatment groups. Our data indicate that vitamin A concentrations of less than 9000 IU/kg of DM in milk replacers result in declining liver vitamin A stores in preruminant calves. Using the human Dietary Reference Intakes as a model for calculating the requirement, we recommend that the vitamin A requirement for preruminant calves should be increased to 11,000 IU of vitamin A/kg of DM.  相似文献

Angus crossbred steers (n = 48) were either early-weaned (EW) at 137 days or weaned at a traditional age (TW) of 199 days to determine effects of weaning age and dietary vitamin A on serum and liver retinol, carcass traits, and lipid composition. Steers from both weaning ages were allotted to receive either 42,180 IU vitamin A/day (HA) or no supplemental vitamin A (NA). Early-weaned and TW steers consumed vitamin A treatments for 235 and 175 days, respectively. Serum and liver retinol of HA steers were dramatically higher (P < 0.01) than those of NA steers at the end of finishing. Steers were harvested in two groups 35 days apart at an average ultrasound 12th rib fat thickness of 1.0 cm. Live and HCW were similar (P > 0.10) between NA and HA steers, but HA steers had numerically greater (P  0.10) fat thickness (1.05 vs. 0.87 cm). Marbling score and %IMF fat were numerically (P > 0.10) higher for EWNA than EWHA steers. Ratio of marbling score/12th rib fat thickness was greater (P = 0.08), and ratios of either marbling or %IMF per unit of 12th rib fat thickness, days on finishing diet, unit of HCW, and tenth of yield grade consistently favored steers fed NA, particularly EW steers. Proportions of serum fatty acids changed (P < 0.05) during finishing; proportions of individual fatty acids of the longissimus muscle did not change. Restricting vitamin A during finishing has potential to increase carcass marbling and to decrease waste fat, particularly for EW.  相似文献

The disaccharide lactulose (beta-D-galactopyranosyl-(1-4)-D-fructofuranose) is thought to be poorly hydrolized by intestinal enzymes but is broken down by microorganisms in the colon. Thus lactulose shares with pectin similar characteristics. Lactulose is used in the food industry and in the treatment of constipation. There is preliminary evidence that dietary lactulose (5%, w/w) causes increased concentrations of serum and liver cholesterol in rats when compared with pectin. This paper describes dose-response relationships between dietary lactulose and cholesterol in either serum or liver.  相似文献

To investigate the relationships between helminth infections and iron status among school-aged children, 1,115 Tanzanian children in grades 2 through 5 were randomly assigned to treatment or control groups. The children in the treatment group were screened for infection with Schistosoma haematobium and hookworm at baseline, 3 months, and 15 months; infected children were given albendazole against hookworm and praziquantel against schistosomiasis. The control group received a placebo and did not undergo parasitological screening until 15 months after the baseline. Hematological variables were compared between the treatment and control groups. The main results were, first, that the hemoglobin concentration significantly improved after treatment for hookworm (p < .001) by 9.3 g/L in children treated for hookworm only and by 8.8 g/L in children treated for hookworm and schistosomiasis. The ferritin concentration also improved in children treated for schistosomiasis (p = .001) or hookworm (p = .019). Second, a longitudinal analysis of the data from the children in the control group showed that hookworm and schistosomiasis loads were negatively associated with hemoglobin and ferritin concentrations. Moreover, ferritin concentrations increased as C-reactive protein levels increased. Overall, the results showed that anthelmintic treatment is a useful tool for reducing anemia in areas with high hookworm and schistosomiasis endemicity. The empirical relationship between ferritin and C-reactive protein indicated that simple procedures for adjusting cutoff points for the use of ferritin as an indicator of low iron stores were unlikely to be useful in this population.  相似文献

Enhancing micronutrient (i.e., mineral and vitamin) concentrations within milk and serum from dairy cows is important for both the health of the cow and the nutritive value of the milk for human consumption. However, a good understanding of the genetics underlying the micronutrient content in dairy cattle is needed to facilitate such enhancements through feeding or breeding practices. In this study, milk (n = 950) and serum (n = 766) samples were collected from Holstein-Friesian dairy cows (n = 479) on 19 occasions over a 59-mo period and analyzed for concentrations of important elements. Additionally, a subset of 256 milk samples was analyzed for concentrations of vitamin B12. Cows belonged to 2 genetic lines (average and highest genetic merit for milk fat plus protein yield) and were assigned to 1 of 2 diets based on either a by-product or homegrown ration. Univariate models accounting for repeated records were used to analyze element and vitamin B12 data and investigate the effect of genotype and feeding system as well as derive estimates of variance components and genetic parameters. Bivariate models were used to study correlations both within and between milk and serum. Only concentrations of Hg in milk were seen to be affected by genotype, with higher concentrations in cows with high genetic merit. In contrast, element concentrations were influenced by feeding system such that cows fed the homegrown diet had increased milk concentrations of Ca, Cu, I, Mn, Mo, P, and K and increased serum concentrations of Cd, Cu, Fe, Mo, and V. Cows on the by-product diet had increased milk concentrations of Mg, Se, and Na and increased serum concentrations of P and Se. Heritability (h2) estimates were obtained for 6 milk and 4 serum elements, including Mg (h2milk = 0.30), K (h2serum = 0.18), Ca (h2milk = 0.20; h2serum = 0.12), Mn (h2milk = 0.14), Cu (h2serum = 0.22), Zn (h2milk = 0.24), Se (h2milk = 0.15; h2serum = 0.10), and Mo (h2milk = 0.19). Significant estimates of repeatability were observed in all milk and serum quantity elements (Na, Mg, P, K, and Ca) as well as 5 milk and 7 serum trace elements. Only K in milk and serum was found to have a significant positive genetic and phenotypic correlation (0.52 and 0.22, respectively). Significant phenotypic associations were noted between milk and serum Ca (0.17), Mo (0.19), and Na (−0.79). Additional multivariate analyses between measures within sample type (i.e., milk or serum) revealed significant positive associations, both phenotypic and genetic, between some of the elements. In milk, Se was genetically correlated with Ca (0.63), Mg (0.59), Mn (0.40), P (0.53), and Zn (0.52), whereas in serum, V showed strong genetic associations with Cd (0.71), Ca (0.53), Mn (0.63), Mo (0.57), P (0.42), K (0.45), and Hg (−0.44). These results provide evidence that element concentrations in milk and blood of dairy cows are significantly influenced by both diet and genetics and demonstrate the potential for genetic selection and dietary manipulation to alter nutrient concentration to improve both cow health and the healthfulness of milk for human consumption.  相似文献

The recent implementation of soil and drinking water screening guidance values for two perfluorochemicals (PFCs), perfluorooctanoate (PFOA) and perfluorooctane sulfonate (PFOS) by the U.S. Environmental Protection Agency (EPA), reflects the growing concerns regarding the presence of these persistent and bioaccumulative chemicals in the natural environment. Previous work has established the potential risk to the environment from the land application of industrially contaminated biosolids, but studies focusing on environmental risk from land application of typical municipal biosolids are lacking. Thus, the present study investigated the occurrence and fate of PFCs from land-applied municipal biosolids by evaluating the levels, mass balance, desorption, and transport of PFCs in soils receiving application of municipal biosolids at various loading rates. This study is the first to report levels of PFCs in agricultural soils amended with typical municipal biosolids. PFOS was the dominant PFC in both biosolids (80-219 ng/g) and biosolids-amended soil (2-483 ng/g). Concentrations of all PFCs in soil increased linearly with increasing biosolids loading rate. These data were used to develop a model for predicting PFC soil concentrations in soils amended with typical municipal biosolids using cumulative biosolids loading rates. Mass balance calculations comparing PFCs applied vs those recovered in the surface soil interval indicated the potential transformation of PFC precursors. Laboratory desorption experiments indicated that the leaching potential of PFCs decreases with increasing chain length and that previously derived organic-carbon normalized partition coefficients may not be accurate predictors of the desorption of long-chain PFCs from biosolids-amended soils. Trace levels of PFCs were also detected in soil cores from biosolids-amended soils to depths of 120 cm, suggesting potential movement of these compounds within the soil profile over time and confirming the higher transport potential for short-chain PFCs in soils amended with municipal biosolids.  相似文献

目的 建立三类动物性膳食中典型全氟有机化合物的超高效液相色谱-串联质谱( UPLC-MS/MS)测定方法.方法 实验优化了样品提取方法和液相色谱质谱条件.样品经冷冻干燥后用甲醇提取,弱阴离子交换固相萃取柱净化;使用C18色谱柱,甲醇和2 mmol/L醋酸铵水溶液为流动相进行分离;采用ESI负离子源并在多反应监测模式(MRM)下进行测定.结果 目标化合物的回收率为82%~115%,检测限分别为76~129 pg/g干重和88~319 pg/g干重.运用该方法分析了9份膳食样品,三类样品中均检出目标化合物.结论 本方法简便快速、准确可靠,灵敏度足以满足动物性膳食样品的检测.  相似文献

Neutral, volatile polyfluorinated alkyl substances (PFAS) were measured in environmental air samples at two different sites in Northern Germany in spring 2005. The sampling locations were chosen to cover a metropolitan and a rural site, the Hamburg city center, and Waldhof, a background monitoring site. An optimized and validated analytical protocol was used to analyze two sets of parallel high-volume air samples. For both sampling locations as well as for individual samples, field blanks were taken to monitor possible background contamination. Gas chromatography coupled to mass spectrometry using positive chemical ionization (GC/ PCI-MS) was used for quantitative analyses. This article describes the first air concentration data of volatile PFAS outside North America reported in the peer-reviewed literature. The wide distribution of fluorotelomer alcohols (FTOHs), fluorinated sulfonamides, and sulfonamidoethanols (FOSAs/FOSEs) in German environmental air is presented. Furthermore, two volatile PFAS, i.e., N-methyl fluorooctane sulfonamide (NMeFOSA) and 4:2 FTOH, were determined for the first time in environmental air. Minimum-maximum sigmaFTOH concentrations of 64-311 pg/m3 (remote) up to 150-546 pg/m3 (urban) and minimum-maximum sigmaFOSA + FOSE concentrations between 12 and 54 pg/m3 (remote) and 29 and 151 pg/m3 (urban) were determined. 8:2 FTOH and 6:2 FTOH were found to be the predominant POPs determined in Waldhof so far. Blank contamination was found to be negligible. A significant correlation was found with the ambient temperature for the partitioning of airborne FOSEs between the gaseous and particulate phase (R = 0.853), whereas FTOHs and FOSAs were almost exclusively found in the gaseous phase. Furthermore, highest airborne PFAS concentrations were determined at relatively high ambient temperatures. Correlation coefficients (R) for sigmaFTOH and sigmaFOSA + FOSE concentrations with temperature were 0.954 and 0.968, respectively. Finally, the PFAS concentrations determined in this study are set into context with levels of "classical" persistent organic pollutants (POPs) in the same region and PFAS data available for North America.  相似文献

Nine multiparous and 12 primiparous cows were fed diets containing an anionic salt supplement and moderate Ca (0.99%) or high Ca (1.50%) concentrations for 21 d prepartum to determine the effects of dietary Ca concentration on serum and urine electrolytes and on postpartum intake and milk yield. Blood samples were collected during 21 to 1 d prepartum, 0 to 2 d postpartum, and 3 to 21 d postpartum. Dietary cation-anion difference (DCAD) for prepartum diets was approximately −6 mEq/100 g of dry matter (Na + K − Cl − S). Immediately postpartum, cows were fed diets with positive DCAD with greater than 1.00% Ca concentration. Mean serum Ca concentrations 21 to 1 d prepartum, 0 to 2 d postpartum, and 3 to 21 d postpartum were 9.62, 8.41, and 9.38 mg/dL. There were no treatment effects on serum Ca concentration. Mean serum Ca concentration was higher for primiparous than multiparous cows (9.34 vs. 8.93 mg/dL) for the trial and at calving (8.77 vs. 8.13 mg/dL). Mean serum HCO3 and urinary pH, respectively, were 20.32 mEq/L and 5.67 prepartum, 25.82 mEq/L and 7.62 at calving, and 26.08 mEq/L and 8.25 postpartum. No differences due to treatment were observed for serum and urinary concentrations of HCO3, pH, Mg, Na, K, and Cl. Milk yield was similar for 0.99 and 1.50% Ca treatments (22.8 and 20.7 kg/d). Diets containing 0.99 or 1.5% Ca maintained serum Ca at adequate levels around parturition and resulted in similar dry matter intake and postpartum milk yield.  相似文献

Colostrum samples from 366 Charolais primiparous cows, as well as serum from their calves at 24 to 48 h of age, were collected to gain an overview of the situation regarding passive immune transfer in beef cattle, from both the phenotypic and genetic points of view. All samples were analyzed to quantify their G1 immunoglobulins by radial immunodiffusion (RID) and their IgG, IgA, and IgM using ELISA. The average concentrations obtained in colostrum were 84 mg/mL for RID-IgG1, and 158 mg/mL, 4.5 mg/mL and 10.8 mg/mL for ELISA-IgG, -IgA, and -IgM, respectively. The corresponding values in calf serum were 19.9, 30.6, 1.0, and 1.9 mg/mL. Apart from the general environmental effect (farm-year combination and laboratory conditions), the characteristics of the dams tested did not reveal any influence on colostrum immunoglobulin concentrations. Calving difficulty, as well as the birth weight and sex of calves, were found to be associated with serum concentrations in some cases. Heritability estimates were low to moderate, with the highest being for RID-IgG1 in colostrum (h2 = 0.28, standard error = 0.14) and serum (h2 = 0.36, standard error = 0.18). Phenotypic correlations among the different immunoglobulins were generally positive or null, and none of the genetic correlations were significant due to large standard errors. The phenotypic correlation between dam colostrum and calf serum values was 0.2 for RID-IgG1 and null for the 3 ELISA measurements. The correlation between RID-IgG1 and ELISA-IgG was, unexpectedly, null for colostrum and 0.4 for serum. Increased RID-IgG1 levels in calf serum were associated with improved survival, as well as better early growth and fewer health problems. These results thus showed that despite generally higher concentrations in beef than in dairy cattle, passive transfer was unsuccessful in a considerable number of calves. This should be brought to the attention of breeders to avoid negative effects on survival and subsequent performance. The heritability estimates were encouraging; however, obtaining phenotypes on a large scale constitutes a real limitation regarding these traits.  相似文献

Four multiparous lactating cows (175 to 220 d in milk) were used in a 4 x 4 Latin square design to assess the effects of four doses (0.0, 0.5, 1.0, 1.5 microg/kg of body weight) of lipopolysaccharide (LPS; Escherichia coli 0111:B4) on circulating concentrations of macrominerals and vitamin D metabolites. Treatments were dissolved in 100 ml of sterile saline and infused intravenously over a period of 100 min. Blood was sampled immediately before infusion (0 h), at 60-min intervals for 8 h, and at 24 and 48 h postinfusion. Vitamin D metabolites were analyzed in samples collected at 0, 2, 6, 24, and 48 h only. Serum Ca and P concentrations decreased after LPS infusion, but there was no effect on serum magnesium concentration. Plasma 25-OH vitamin D3 and 1,25-(OH)2 vitamin D3 were not affected by LPS infusion; however, when analyzed as 0 vs. all other doses of LPS combined, there was a tendency for plasma 1,25-(OH)2 vitamin D3 concentration to decrease when cows were infused with LPS. The inflammatory response elicited by LPS altered plasma macromineral concentrations, a result that may have important implications for calcium homeostasis and metabolic health of lactating dairy cows.  相似文献

Decomposition of perfluorooctanesulfonate (PFOS) and related chemicals in subcritical water was investigated. Although PFOS demonstrated little reactivity in pure subcritical water, addition of zerovalent metals to the reaction system enhanced the PFOS decomposition to form F-ions, with an increasing order of activity of no metal approximately equal Al < Cu < Zn < Fe. Use of iron led to the most efficient PFOS decomposition: When iron powder was added to an aqueous solution of PFOS (93-372 microM) and the mixture was heated at 350 degrees C for 6 h, PFOS concentration in the reaction solution fell below 2.2 microM (detection limit of HPLC with conductometric detection), with formation of F-ions with yields [i.e., (moles of F- formed)/(moles of fluorine content in initial PFOS) x 100] of 46.2-51.4% and without any formation of perfluorocarboxylic acids. A small amount of CHF3 was detected in the gas phase with a yield [i.e., (moles of CHF3)/(moles of carbon content in initial PFOS) x 100] of 0.7%, after the reaction of PFOS (372 microM) with iron at 350 degree C for 6 h. Spectroscopic measurements indicated that PFOS in water markedly adsorbed on the iron surface even at room temperature, and the adsorbed fluorinated species on the iron surface decomposed with rising temperature, with prominent release of F- ions to the solution phase above 250 degrees C. This method was also effective in decomposing other perfluoroalkylsulfonates bearing shorter chain (C2-C6) perfluoroalkyl groups and was successfully applied to the decomposition of PFOS contained in an antireflective coating agent used in semiconductor manufacturing.  相似文献

The present study investigated the potential consequences, positive or negative, that selection for favorable production-related traits may have on concentrations of vitamin B12 and key chemical elements in dairy cow milk and serum and the possible impact on milk healthiness, and associated benefits, for the dairy product consumer. Milk and serum samples (950 and 755, respectively) were collected from Holstein-Friesian dairy cows (n = 479) on 19 occasions over a 59-mo period, generating 34,258 individual records, and analyzed for concentrations of key trace and quantity elements, heavy metals, and milk vitamin B12. These data were then matched to economically important production data (milk, fat, and protein yield) and management data (dry matter intake, liveweight, and body condition score). Multivariate animal models, including full pedigree information, were used to analyze data and investigate relationships between traits of interest. Results highlighted negative genetic correlations between many quantity and trace elements in both milk and serum with production and management traits. Milk yield was strongly negatively correlated with the milk quantity elements Mg and Ca (genetic correlation between traits, ra = ?0.58 and ?0.63, respectively) as well as the trace elements Mn, Fe, Ni, Cu, Zn, and Mo (ra = ?0.32, ?0.58, ?0.52, ?0.40, ?0.34, and ?0.96, respectively); and in serum, Mg, Ca, Co, Fe, and Zn (ra = ?0.50, ?0.36, ?0.68, ?0.54, and ?0.90, respectively). Strong genetic correlations were noted between dry matter intake with V (ra = 0.97), Fe (ra = ?0.69), Ni (ra = ?0.81), and Zn (ra = ?0.75), and in serum, strong negative genetic correlations were observed between dry matter intake with Ca and Se (ra = ?0.95 and ?0.88, respectively). Body condition score was negatively correlated with serum P, Cu, Se, and Pb (ra = ?0.45, ?0.35, ?0.51, and ?0.64, respectively) and positively correlated with Mn, Fe, and Zn (ra = 0.40, 0.71, and 0.55, respectively). Our results suggest that breeding strategies aimed at improving economically important production-related traits would most likely result in a negative impact on levels of beneficial nutrients within milk for human consumption (such as Mg, Ca, Fe, Zn, and Se).  相似文献

The objective of the present study was to quantify the relationships between prepartum nonesterified fatty acid (NEFA) concentrations and the development of subsequent diseases or culling and to identify the optimal thresholds allowing identification of animals at high risk of developing postpartum diseases or being culled. A total of 1,299 Holstein cows from 50 commercial herds located around Saint-Hyacinthe (QC, Canada) were enrolled in this observational study. Blood samples were collected from enrolled cows between 1 and 14 d before calving for serum NEFA quantification. Data concerning postpartum diseases and culling were collected from computerized record systems. The association between prepartum NEFA concentrations and postpartum diseases and culling was quantified using generalized linear mixed models, accounting for parity, season, week of sampling, and herd. Optimal NEFA thresholds were evaluated with receiver operator characteristic curves analysis for all diseases and then confirmed with generalized linear mixed models, considering NEFA as a categorical variable (high or low). Prepartum serum NEFA concentrations were associated with diseases diagnosed during the first 30 d in milk (DIM) and culling within the first 50 DIM. The optimal NEFA threshold associated with diseases was ≥290 µmol/L for retained placenta, ≥300 µmol/L for metritis and abomasal displacement, and ≥280 µmol/L for clinical mastitis and hyperketonemia. The level associated with the occurrence of at least one of these diseases in the first 30 DIM was ≥280 µmol/L, but it was ≥260 µmol/L for culling in the first 50 DIM. No relationship was found between NEFA concentrations and reproductive tract diseases (purulent vaginal discharge or cytological endometritis) or subclinical intramammary infection. Despite the strong relationship between prepartum NEFA concentrations and many diseases, the NEFA optimal threshold accuracy found in our study was low. In conclusion, our results demonstrate a relationship between NEFA concentrations in the 14-d period before calving and the subsequent development of diseases and culling. Prepartum NEFA concentrations thresholds between ≥260 and 300 µmol/L appear to be a strategic choice. However, considering the low accuracy, their use at the cow level should be performed with caution.  相似文献

Follistatin has been isolated from human placenta and has been identified in human foetal membranes and fluids. Serum follistatin levels in women rise during pregnancy particularly near term. In this study, we examined the effect of induction and stage of labour on maternal plasma concentrations of follistatin. Women who gave birth after a normal pregnancy were retrospectively divided into three groups: those who went in labour spontaneously (n = 33), needed induction by amniotomy and IV oxytocin (n = 18) or underwent planned caesarean section (n = 10). Serum was collected at 38-40 weeks of gestation, periodically through labour with a vaginal examination and once within 36 h postpartum and assayed for oestradiol, progesterone, prolactin and C-reactive protein. Follistatin was measured using a rabbit antiserum (#204) raised against purified 35 kDa bovine follistatin. Human recombinant follistatin was used as both standard and tracer. Concentrations of follistatin at 38-40 weeks of gestation were significantly different between groups. Those who had a spontaneous labour had concentrations higher than those who were induced. Similarly, those who were induced had concentrations higher than those who underwent a caesarean. In the spontaneous group, follistatin rose during labour, peaking at 57.9 +/- 5.48 ng/ml at > 3 cm of cervical dilation, and after delivery follistatin decreased to 26.16 +/- 3.4 ng/ml at 24 h post-delivery. In induced patients follistatin continued increasing to peak following delivery at 26.9 +/- 3.0 ng/ml and decreased at > 3 h post-delivery. Follistatin concentrations in caesarean section patients at 24 h post-surgery (18.53 +/- 3.74 ng/ml) were not different from that before the surgery and were comparable with the other two groups. Follistatin is clearly implicated in the onset of labour; however, further studies with a larger cohort of women are necessary to determine the nature of its role.  相似文献
