In Exp I, 16 New Zealand white rabbits were trained to perform an instrumental head-raising response for sucrose reward. A jaw-movement CR was established to a 2-sec CS by pairing it with sucrose; a control stimulus was unpaired with sucrose. Instrumental responding maintained by a VI 40-sec schedule was enhanced during 10-sec presentations of the paired, but not the unpaired, CS. Responding on a VR 15 schedule was unaffected except on trials on which the pre-CS baseline response rate was low; in such cases the paired CS caused a long-lasting acceleration of responding. Noncontingent presentation of the sucrose reinforcer itself briefly suppressed responding but had no long-term effect. In Exp II (6 Ss), a CS that had been conditioned at a 10-sec duration produced the same pattern of effects as in Exp I, indicating that facilitation resulted from CS presentation rather than from the frustrative effects of nonreinforcement of the CS. In Exp III (16 Ss), an inhibitory CS blocked facilitation by the excitatory CS but did not itself affect instrumental responding. In Exp I, virtually total depletion of cortical and hippocampal noradrenaline (NA) by stereotaxic injection of 6-hydroxydopamine into the fibers of the dorsal noradrenergic bundle produced no impairment in acquisition learning of a runway response for food reward by 10 male Wistar rats. Extinction of this response, once learned, was markedly slower in the treated group than in 10 controls, the treated animals perseverating in rapid running to the goal box even with no food present there. Similarly, in Exp II with the same Ss, no impairment was found on acquisition of a continuously reinforced leverpressing response for food. Extinction of this response, however, was again slower in the treated group. Exp III showed that the subsequent acquisition of a successive light-dark discrimination task was also slower in the same treated Ss, with these animals perseverating in responding to the negative stimulus. Although selective forebrain NA loss does not impair the acquisition of appetitive responses, the suppression of responses in the absence of reward is impeded. The comparator hypothesis is a response rule stating that responding to a Pavlovian conditioned stimulus/stimuli (CS) reflects the associative strength of the CS relative to that of other cues (comparator stimuli) that were present during CS training. Thus, modulation of the associative strength of a CS's comparator stimulus should alter responding to that CS. These studies examined the stimulus specificity of this effect using within-Ss designs. Rats were trained on 2 CSs, each with a unique comparator stimulus, to determine the degree to which posttraining extinction of the comparator stimulus for one CS influences responding to the other CS. Using negative contingency (Exps 1 and 2), overshadowing (Exp 3), and local context (Exp 4) preparations, stimulus specificity was observed. In each case, posttraining extinction of the comparator stimulus for one CS had greater impact on responding to that CS than on responding to the alternate CS. Tested 40 male Wistar rats in an open field apparatus to determine their responsiveness to exploratory incentives. The upper and lower quartiles of this group (n = 20) then had bipolar electrodes implanted into the lateral hypothalamic area of the brain and were subsequently tested for self-stimulation in a shuttlebox and Skinner box. Those Ss displaying the greatest amount of activity in the open field (high-exploratory) displayed a higher average rate of self-stimulation in both tests, but only at optimal current levels. These differences are thought to reflect differences in reward value of hypothalamic brain stimulation in the 2 experimental groups. The implications of these data for understanding the role of neural substrates of positive reinforcement in determining individual responses to the incentive value of a given reward are discussed. The effects of caudate-putamen lesions in the rat on conditioning were investigated in 3 experiments. In Exp I, rats with lesions were impaired on a spatial task that required learning to make the correct position response, but no deficit was obtained when the alternatives were differentiated by salient visual cues. Performance remained good even when the visual cues were removed. A classical conditioned suppression paradigm was used in Exp II, and caudate-putamen lesions were found not to impair acquisition of suppression or overshadowing. In Exp III poor instrumental learning in subjects with lesions as retarded acquisition of lever-press responding and depressed variable interval response rates were found. When a light stimulus was present together with reinforced responses, response rates were depressed in the control group (which suggested an overshadowing phenomenon) but were potentiated in the group with lesions. Results show that caudate-putamen lesions did not produce a general deficit in association formation. There was disruption of learning about responses but not stimuli and, moreover, it was found that redundant stimulus cues aided response learning. It appears that for normal subjects, the salience or associability of the response cues may largely-determine the influence of stimuli presented during instrumental conditioning. Four experiments with 202 rats investigated spontaneous configuring, using the conditioned flavor-aversion paradigm. In Exp I, extended training of a 2-flavor compound stimulus did not produce spontaneous differentiation of conditioned responding to that compound and its elements. In Exp II, it was found that extended nonreinforced exposure to a compound stimulus generated spontaneous element–compound differentiation when the elements were later conditioned. Ss that received extended preexposure to the compound showed less conditioned responding to the compound than to either of its elements. However, Ss that had not received preexposure to the compound showed greater conditioned responding to the compound than to either of its elements (summation). In Exp III, nonreinforced preexposure to a compound stimulus prior to minimal reinforced compound training produced spontaneous compound–element differentiation, but extended reinforced compound training eliminated that differentiation. In Exp IV, extended partial reinforcement training with a compound produced differentiation of the compound from its elements. Implications of these data for the mechanisms responsible for spontaneous configuring, and for the summation assumptions common to most learning theories, are discussed. Examined evidence of latent inhibition in a series of experiments with goldfish. In Exp I, 12 Ss were given nonreinforced preexposure to a color that subsequently predicted shock in an activity conditioning situation; their performance did not differ from that of 12 control Ss preexposed to a markedly different color. In Exp II, 12 Ss given nonreinforced preexposure to a tone and an unstimulated control group of 12 Ss were trained in an appetitive situation, with the tone serving either as a conditioned excitor or as a conditioned inhibitor. Preexposure had significant effect in the conditioned excitation training, but it reduced the level of responding both to the positive stimulus and to the negative compound in the conditioned inhibition training. In Exps III and IV, classical aversive conditioning was studied in the shuttle box. In Exp III, excitatory conditioning to a color was found to be impaired (relative to the performance of nonpreexposed control Ss) as much by nonreinforced preexposure to the training color as by nonreinforced preexposure to a markedly different color; substantial variation in amount of preexposure was without significant effect. In the conditioned inhibition training of Exp IV, 12 Ss with nonreinforced preexposure responded less than did 12 unstimulated control Ss, both to the positive stimulus and to the negative compound. Results demonstrate that the effect of preexposure on goldfish is their reduction of general responsiveness or level of arousal. Three experiments, with 32 male hooded Sprague-Dawley rats, examined whether long-term potentiation (LTP) could enhance the stimulus properties of electrical brain stimulation. In Exp I, a paradigm was developed in which single-pulse stimulation of the perforant path (PP) could acquire control over operant responses. Evoked potentials were recorded from the dentate gyrus (DG) to measure the postsynaptic consequences of the stimulus and to monitor synaptic efficacy in the PP–DG synapses. Exp II confirmed the relation between the amount of evoked activity and acquisition rate and also showed that transecting the PP impaired performance. In Exp III, high-frequency stimulation of the PP produced LTP and accelerated subsequent acquisition of behavioral responding to PP stimulation. Results document a link between increases in synaptic efficacy and changes in behavior and thereby demonstrate the ability of LTP to serve as at least 1 component of the neural bases of learning and memory. Circadian rhythms in rodents respond to arousing, nonphotic stimuli that contribute to daily patterns of entrainment. To examine whether the motivational significance of a stimulus is important for eliciting nonphotic circadian phase shirts in Syrian hamsters (Mesocricetus auratus), the authors compared responses to a highly rewarding stimulus (lateral hypothalamic brain stimulation reward [BSR]) and a highly aversive stimulus (footshock). Animals were housed on a 14:10-hr light-dark cycle until test day, when they were given a 1-hr BSR session (trained animals) or a 1-mA electric footshock at 1 of 8 circadian times, and were maintained in constant dark thereafter. Both BSR pulses and footshock produced nonphotic phase response curves. These results support the hypothesis that arousal resulting from the motivational significance of a stimulus is a major factor in nonphotic phase shifts. Systemic administration of lipopolysaccharide (LPS), a non-specific activator of proinflammatory cytokine release from macrophages, provokes sickness characterized by anorexia, soporific effects, and disturbances of locomotor activity and exploration. In addition, endotoxin treatment may provoke an anhedonic response. Assessment of anhedonia in appetitive paradigms, however, is compromised by the anorexia provoked by the treatment. The present investigation assessed the anhedonic effects of LPS on rewarding lateral hypothalamic brain stimulation. Using a simultaneous discrimination, current titration procedure in the assessment of intracranial self-stimulation (ICSS), it was found that acute systemic administration of LPS (50 microg, 100 microg or 200 microg) reduced ICSS during the ascending sequence of current presentations, but had little effect on responding to a series of descending currents. In a parallel experiment, peripheral administration of LPS (100 microg) increased in vivo dopamine (DA) efflux from the nucleus accumbens, a region thought to be involved in goal-directed responding to positively reinforcing stimuli. It is suggested that LPS alters ICSS in a manner different than that observed following stressor exposure or peripheral IL-2 treatment. Furthermore, LPS may engender an anhedonic effect (possibly secondary to sickness), and the decline of responding reflects the relation between the cost of responding given in the face of sickness and the reward received for responding.  相似文献

Thresholds determined from the frequency of pulses and the current for rewarding brain stimulation were obtained from rats with lateral hypothalamic electrodes. The threshold, defined as the frequency or current corresponding to one-half the maximum response rate, was interpolated from reward summation functions. Daily trials of both ascending and descending sequences of frequency and current yielded no significant difference between order of presentation. While there was more variability in the maximum response rates across the sessions, neither frequency- nor current-based threshold evaluations yielded significant rate effects. Findings suggest that the threshold procedure is generally not influenced by the sequence of delivery of stimulus values and, thus, may be regarded as a reliable measure of the reinforcing properties of brain-stimulation reward. In 3 experiments with 85 human 3-month-olds, the authors asked whether retroactive interference with their memory of the original training stimulus is temporary or permanent. Infants learned to move a mobile by kicking and then were exposed to a different mobile (Exp 1) or context (Exp 2) immediately or 3 days afterward (Exp 3). They were tested after increasing delays with the original stimulus, the exposed stimulus, or a completely novel stimulus. Retroactive interference was temporary and unrelated to the exposure delay. The data are consistent with a retrieval-based account of interference. Memory updating (i.e., responding to the interfering stimulus) was coincident with retroactive interference, suggesting that retroactive interference is an adaptive mechanism that facilitates memory updating within a narrow time window. Two experiments investigated the effects on instrumental responding of a sequence of training stages similar to that followed in demonstrations of sensory preconditioning. In Exp I, 62 male hooded rats that experienced response-correlated presentations of a neutral stimulus, followed by pairings of the neutral stimulus with food, showed an enhanced tendency to emit the response during a test in which responding had no programmed consequences. In Exp II, in which the 2nd stage of training involved pairings of the neutral stimulus with shock, responding by the 24 Ss during the test was suppressed. Two interpretations of the results are discussed: (a) Enhancement and suppression of the operant response were mediated by Pavlovian processes (viz., sensory preconditioning); (b) the critical association formed was between the response and the neutral stimulus, and the observed variations in instrumental responding reflected response preconditioning. The comparator hypothesis posits that conditioned responding is determined by a comparison at the time of testing between the associative strengths of the conditioned stimulus/stimuli (CS) and stimuli proximal to the CS at the time of conditioning. The hypothesis treats all associations as being excitatory and treats conditioned inhibition as the behavioral consequence of a CS that is less excitatory than its comparator stimuli. Conditioned lick suppression by rats was used to differentiate 4 possible sources of retarded responding to an inhibitory CS. These include habituation to the unconditioned stimulus/stimuli (UCS), latent inhibition to the CS, blocking of the CS-UCS association by the conditioning context, and enhanced excitatory associations to the comparator stimuli. Prior research has demonstrated the 1st 3 phenomena. Therefore, we employed parameters expected to highlight the 4th one—the comparator process. In Exp I, our negative contingency training produced a conditioned inhibitor that passed inhibitory summation and retardation tests. In Exp II we found transfer of retardation from an inhibitory CS to a novel stimulus when the location where retardation-test training occurred was excitatory. In Exp III, extinction of the conditioning context attenuated retardation regardless of whether extinction occurred before or after the CS-UCS pairings of the retardation test. Two experiments examined habituation and dishabituation of behavioral responding to repeated presentations of a tactile stimulus (brush stroke) in 48 newborns during the 1st epoch of active-quiet sleep following a feeding. Exp I demonstrated habituation to a repeatedly presented brush stroke to the ear but failed to demonstrate dishabituation (i.e., response recovery) to the original brush stroke following an intense auditory stimulus (86 dB rattle sound). A post hoc control group showed that the intense auditory stimulus had suppressed subsequent responding to the tactile stimulus. Exp II replicated the habituation phase of Exp I and demonstrated response recovery to stimulation at a novel tactile site and to an auditory probe. Results indicate that the habituation paradigm used in infancy research could be successfully extended to the tactile modality. It is also suggested that prior auditory stimulation, but not prior tactile stimulation, might direct attention away from a subsequently presented tactile stimulus. Attempted to replicate previous demonstrations of classical conditioning of morphine analgesic tolerance and to determine whether stimulus overshadowing effects might explain previous conflicting findings. In Exp I, 8 groups of male Sprague-Dawley rats received a series of 10 morphine (5 mg/kg) and/or saline injections, differing only with respect to the contingency between a compound visual-auditory CS and the substance injected. When tested for analgesic responding to morphine in the presence of the compound CS, only those groups for which the CS and morphine injections were paired during the acquisition sequence evidenced tolerance. In Exp II, tolerant Ss were tested in the presence of 1 component of the compound CS. When a loud tone (85 db) was used in the compound, less analgesic tolerance was elicited later by the weaker visual stimulus alone. This differential stimulus control of the analgesic response suggests that overshadowing may contribute to failures to replicate conditioned morphine tolerance. Rats responded on 2 levers delivering brain stimulation reward on concurrent variable interval schedules. Following many successive sessions with unchanging relative rates of reward, subjects adjusted to an eventual change slowly and showed spontaneous reversions at the beginning of subsequent sessions. When changes in rates of reward occurred between and within every session, subjects adjusted to them about as rapidly as they could in principle do so, as shown by comparison to a Bayesian model of an ideal detector. This and other features of the adjustments to frequent changes imply that the behavioral effect of reinforcement depends on the subject's perception of incomes and changes in incomes rather than on the strengthening and weakening of behaviors in accord with their past effects or expected results. Models for the process by which perceived incomes determine stay durations and for the process that detects changes in rates are developed. Conducted 2 experiments with a total of 8 male Charles River albino rats. In Exp I, using a runway paradigm, pretrial electric stimulation of the brain (ESB), food deprivation, and to a lesser extent water deprivation increased the running speed to an ESB reward. The trial-by-trial course of the change in running speed after the manipulation of food deprivation resembled the steplike performance effect seen when pretrial priming ESB was changed rather than the incremental learning effect seen when the response contingent ESB was changed. In Exp II, pretrial ESB inhibited running for food. Results suggest that hunger and pretrial ESB enhance rats' performance for an ESB reward, but act in opposing directions on their performance for a food reward. 31 male Wistar rats self-administering cocaine were treated with the dopamine receptor blocker pimozide or the noradrenergic blockers phentolamine or phenoxybenzamine. Pimozide caused a dose-related (.0625–.5 mg/kg) acceleration of responding; at the higher doses responding subsequently ceased. These effects of pimozide parallel the known effects of reward (unit dose) reduction and reward termination and thus suggest an important role for dopaminergic brain mechanisms in the mediation of cocaine reinforcement. Neither phenoxybenzamine given systemically nor phentolamine given intraventricularly had similar effects; thus no similar role for noradrenergic brain mechanisms is suggested by these experiments.
