桑黄是一种珍稀药用真菌,其黄酮可入药。为优化桑黄黄酮发酵的液体培养基,以菌丝体黄酮产量为指标,通过正交试验,对桑黄液体培养基进行优化。研究结果显示黄酮高产培养基最优条件:玉米粉3%+麸皮7%,蛋白胨2.0%,KH2PO40.10%,MgSO40.15%,菌丝体黄酮产量为212.35mg/L。 相似文献

以玉米渣和葡萄糖为复合碳源,以麸皮和蛋白胨及酵母粉为复合氮源,采用L 9(34)正交设计法优化桑黄菌丝体液体培养基,并为此初步确定了液体培养条件.实验结果表明:最佳培养基为玉米渣3%、麸皮3%、葡萄糖3%,KH2PO4 0.3%、MgSO4·7H2O 0.15%、VB1 20μg/100ml、VB2 30μg/100ml.在32.5℃、pH自然、摇床转速130r/min、摇床培养192h的条件下,其菌丝体生物量平均得率为4.4g/100ml. 相似文献

目的:探讨桑黄菌液体发酵的适宜培养基,为进一步研究其液体发酵工艺及提取有效成分做准备。方法:首先用单因素单指标法筛选,再用正交试验双指标法筛选。结果:桑黄菌生长所需最适C/N比为24;最适的碳源为黄豆粉和玉米粉,适宜的含量分别为4%和3%;最适的氮源为小麦麸粉,适宜的含量为0.5%;对菌丝生长发育影响显著的物质有FeSO4、ZnSO4、K2SO4、CaSO4、Na2HPO4、VB1等。结论:桑黄菌液体发酵适宜的培养基配方(g/L):黄豆粉40、玉米粉30、小麦麸粉5、酵母浸出汁4、CaSO410、K2SO41、Na2HPO41.5、ZnSO4.7H2O0.1、Fe-SO4.7H2O0.5、VB10.1。 相似文献

ABSTRACT: Dried mycelia and mycelia-free broths produced by submerged cultures of Phellinus igniarius under optimal culture conditions were extracted using methanol and hot water and investigated for antioxidant properties. Methanolic extracts from dried mycelia (MEM) and mycelia-free broth (MEB) showed significant antioxidant properties for all EC50 values less than 10 mg/mL except for MEB in scavenging effects on DPPH radicals. Hot water extracts from dried mycelia (HWEM) were evidenced by their low EC50 values (<10 mg/mL) to be effective in reducing power, chelating effect on ferrous ions, and scavenging effect on superoxide anions. α-tocopherol was mainly found in MEM and γ-tocopherol in MEB. Ascorbic acid and total flavonoids were abundant in methanolic extracts (MEM + MEB), whereas total phenols were rich in HWEM. An excellent correlation between contents of total phenols and EC50 values was accomplished for antioxidant activity ( R 2 = 0.996) and chelating ferrous ions ability ( R 2 = 0.922).
Practical Application: In our paper, the products by submerged culture of Phellinus igniarius exhibited powerful antioxidant properties. Results told that extracts from fermenting products by P. igniarius might be good sources for antioxidant-related functional foods and pharmaceutical industries. 相似文献

为更好地再利用酱渣废弃物,以桑黄菌为试验菌种,研究一种适合深层发酵生产富含黄酮、可溶性蛋白和菌丝体的培养基配方。以黄酮和可溶性蛋白的含量及菌丝体干重为考察指标,采用正交试验设计和多指标综合平衡法筛选出黄酮和可溶性蛋白的含量及菌丝体干重均较高的培养基配方。结果表明:最优培养基配方为酱渣12%、酵母浸膏0.4%、KH_2PO_4 0.25%、MgSO_4 0.05%。最优培养基配方经过3次验证试验,得到黄酮含量为2.033 mg/mL,可溶性蛋白的含量为1.667 mg/mL。菌丝体平均干重为1.861 g。 相似文献

以菌丝生物量、胞内多糖含量及胞内多糖产量为评价指标,对鲍氏针层孔菌液体深层发酵培养的培养基配方及发酵条件进行了优化。通过单因素实验和正交实验筛选液体发酵培养基,结果表明,最适培养基为:玉米粉4%,麸皮2%,KH2PO40.4%,MgSO4·7H2O0.05%。进一步对培养条件进行优化,得到最适生长的液体发酵条件为:培养温度28℃、pH6.5、接种量12%、装液量150mL/500mL、转速140r/min、发酵时间168h。在此优化工艺条件下,菌丝生物量、胞内多糖含量及产量由优化前的12.837g/L,15.930mg/g,204.493mg/L分别提高到19.410g/L,20.935mg/g,406.348mg/L。 相似文献

主要以液态深层发酵培养云芝 ,在未控制和控制 pH的条件下观察对云芝多糖形成及菌体生长的影响 ,结果发现在未控制pH的条件下 ,云芝的 pH变化具有双阶段变化即 pH骤降阶段和 pH自我调整阶段 ,在前一阶段菌体大量生长 ,多糖合成很少 ;在后一阶段 ,多糖大量合成。其菌体干重可达 13kg/m3左右 ,胞外多糖最大可达 5 8kg/m3左右。在控制 pH分别为 4 0、4 5、5 0的条件下 ,发现随着pH的增加 ,菌体量逐渐减少 ,大约减少了 2 9%左右 ,但胞外多糖合成量却随着 pH的增加而增加 ,大约增加了 5 0 % ,达 8 8kg/m3。因此 ,pH对云芝菌体生长和胞外多糖合成有重要影响 相似文献
