Twenty Holstein cows were assigned to one of four groups according to milk production and beta-carotene supplementation to evaluate effects of supplementation and milk production on reproductive performance and response of corpus luteum to human chorionic gonadotropin indicated by release of progesterone and secretion of luteinizing hormone in blood plasma. Cows were divided into groups of high and low production and divided further into beta-carotene supplemented and unsupplemented groups. Blood samples were taken 10 days postestrus at 10-min intervals from 1 h prior to injection of 5000 IU human chorionic gonadotropin until 5 h postinjection. Mean days to first detected estrus were 33, 37, 34, and 36 in supplemented, unsupplemented, high-production, and low-production cows. Treatment with beta-carotene and milk production did not affect days to first breeding (63, 67, 62, and 67), or services per conception (3.2, 5.1, 4.6, and 3.7). However, days open were reduced by supplementation (116 versus 186), but milk production had no effect (173 versus 129). Regressions of progesterone on sampling time within treatment and within milk production were significant. Regression of luteinizing hormone on time within production group was also significant. Neither beta-carotene nor milk production affected most reproductive measures, but differences were subtle in corpus luteum response to human chorionic gonadotropin, which may alter endocrine function.  相似文献

Gonadotropin-releasing hormone administered at breeding enhances fertility of dairy cows, so a study was designed to evaluate the mechanism for enhanced fertility following administration of gonadotropin-releasing hormone at first postpartum breeding. Twenty-four cows were assigned randomly to one of two treatments, 100 micrograms of gonadotropin-releasing hormone intramuscular or saline vehicle intramuscular at insemination. Blood samples for luteinizing hormone assay were taken at 2-h intervals prior to breeding and .5-h intervals for 3 h after insemination. Composite morning milk samples for progesterone assay were collected for 30 days after insemination or until next estrus. Cows given gonadotropin-releasing hormone had higher luteinizing hormone concentrations in blood serum following treatment than cows given saline, 13.2 versus 3.0 ng/ml. There was no relationship between luteinizing hormone and subsequent conception. Progesterone for cows that became pregnant was higher throughout sampling days. Mean progesterone concentrations were 4.6 versus 2.2 ng/ml in pregnant and nonpregnant cows during the first 4 days after insemination. Cows treated with gonadotropin-releasing hormone that conceived had higher progesterone than other cows, and that was evident at the first 4 days postbreeding.  相似文献

Concentrations of glucose, growth hormone, insulin, and glucagon in blood plasma relative to days in milk, milk production, type of housing, and season were measured. Blood samples were obtained from 133 to 150 lactating Holstein cows on 3 consecutive days in July, October, January, and April. Glucose, insulin, and ratio of insulin to glucagon increased with increasing days in milk. Growth hormone and ratio of growth hormone to insulin decreased with increasing days in milk. Glucagon concentrations were similar throughout lactation. Above average milk production throughout lactation was associated with above average glucagon, lower insulin, and lower glucose. The relationship was not significant between growth hormone concentration and milk production, however. Cows on summer pasture with limited grain supplement had higher growth hormone and lower glucose, insulin, and glucagon than cows eating ad libitum in barns or feedlot. In general, however, all cows had higher glucose and lower growth hormone, insulin, and ratio of insulin to glucagon in July than in cooler months.  相似文献

The relationship between concentrations of plasma vitamin A and c-carotene and corpora lutea was studied using 52 Holstein cows. Bovine luteinizing hormone was added to incubation tubes in doses of 0, 10, or 100 ng/ml. Regression of progesterone secretion by luteal cells in vitro on plasma beta-carotene was positive and significant for corpora lutea collected during the winter months when plasma beta-carotene was low. The two were unrelated during the summer months when beta-carotene was higher. Similar regressions for in vitro progesterone production and vitamin A were not significant in either season. These results suggest that in vivo beta-carotene status is related to bovine luteal function in vitro.  相似文献

Thirteen lactating Holstein cows were assigned randomly to either a control or beta-carotene (600 mg/d) treatment group to determine the effect of long-term beta-carotene supplementation on releasable luteinizing hormone in response to gonadotropin-releasing hormone challenge. The experimental period was 28 wk. Milking was terminated at wk 16, ovariectomy performed at wk 20, and response to gonadotropin-releasing hormone determined at wk 27. Serum beta-carotene concentrations reflected dietary intake and were higher in the cows fed beta-carotene after 2 wk of supplementation and remained higher for the duration of the trial. Feeding supplemental beta-carotene had no effect on circulating progesterone concentration, corpora lutea size or corpora lutea progesterone concentration, or basal concentrations of luteinizing hormone, frequency, and amplitude of luteinizing hormone pulses, or the release of luteinizing hormone in response to gonadotropin-releasing hormone. Thus, pituitary responsiveness to exogenous gonadotropin-releasing hormone was not affected by feeding supplemental beta-carotene.  相似文献

The objective of this study was to determine whether organically complexed Co, Cu, Mn, and Zn would improve the reproductive performance and milk and milk component production in lactating dairy cows that began receiving bovine somatotropin in the ninth week of lactation. Holstein (n = 50) and Jersey (n = 10) cows were blocked by breed, lactation number, and incidence of retained fetal membranes. Two diets assigned within blocks and fed from parturition until 154 d of lactation were control or control supplemented daily with 26 mg of Co as Co glucoheptonate, 125 mg of Cu as Cu-Lys, 199 mg of Mn as Mn-Met, and 359 mg of Zn as Zn-Met. Cows were fitted with electronic pressure-sensing devices in the second week of lactation for detection of estrus. Ovarian structures were determined via transrectal ultrasonography at 7-d intervals from parturition until observation of the first corpus luteum. Blood samples were taken at 7-d intervals and analyzed for plasma concentrations of progesterone, insulin, and urea nitrogen. Onset of luteal activity was identified by progesterone concentrations > or = 1 ng/ml. Retained fetal membranes increased days to first estrus (detected via electronic estrous detection), first luteal activity, and first corpus luteum in control cows but not in supplemented cows. Days to first observed estrus were greater for control cows than for supplemented cows. Days to first service, days open, days from first service to conception, services per conception, milk yield, milk components, and somatic cell counts were similar for control and supplemented cows. Supplementation with complexed trace minerals effectively reduced days to first estrus.  相似文献

Eighty-five postpartum Holstein cows were randomly assigned to receive 0, 50, 100, or 250 micrograms of the gonadotropin-releasing hormone product, Procystin when follicular cysts were diagnosed by palpation per rectum. Accurate reproductive records were maintained, and milk samples were collected at the time of diagnoses for assay for progesterone. An additional 101 cows were injected with only the 100 micrograms dose of Procystin when cysts were identified. Data showed that days from treatment to first observed estrus decreased with increasing doses of Procystin with no advantage of 250 micrograms over 100 micrograms. Days open and conception rates were similar among the treatment groups. Cows with less than 1 ng/ml progesterone in their milk at the time of treatment returned to estrus sooner than cows with progesterone concentrations greater than 1 ng/ml. In addition, gonadotropin-releasing hormone administered to those cows with low progesterone at the time of treatment led to significantly increased progesterone concentrations by 7 and 14 d posttreatment. We conclude that although Procystin administration hastened estrus of cows with ovarian cysts, breeding practices on the farms did not lead to an improvement in reproductive efficiency of the cows that possessed cysts.  相似文献

Effect of feeding high fat diets on peripheral plasma concentrations of immunoreactive insulin, glucagon, and growth hormone following intravenous glucose challenge (100 mg D-glucose/kg body weight) at 50 and 100 d of lactation in 16 multiparous Holstein cows was evaluated. The high fat diet contained 18.5% whole cottonseed on a dry matter basis as the source of extra dietary fat. Feeding the high fat diet had no apparent effect on energy balance. Basal plasma insulin and insulin:glucagon ratio were increased in cows fed the high fat diet relative to those of controls. Glucagon, insulin, and insulin:glucagon ratio response to glucose challenge were not affected by diet. Response of growth hormone to glucose challenge at 50 d of lactation was depressed in cows fed the high fat diet. Plasma glucose peaked at greater concentrations in cows fed the high fat diet. Feeding a high fat diet to dairy cows appears to increase basal insulin concentration and insulin:glucagon ratio, which has actions opposed to glucose synthesis. Thus, endocrine effects of a high fat diet appear to favor decreased milk production.  相似文献

Concentrations of reproductive hormones and ovarian changes were monitored every 4 days during the postpartum period in 35 dairy cows. The cows were classified as either control (22 cows) or cystic (13 cows) based on per rectum examination of the ovaries. Control cows were detected with a corpus luteum after the first postpartum ovulation while in cystic cows, ovarian follicles of at least 2.5 cm in diameter persisted in the absence of a corpus luteum for at least 14 days. The mean postpartum interval to the first follicle 1.5 to 2.0 cm in diameter was about 16 days for both groups. Ovulation was detected earlier postpartum for cows in the control group (18.4 +/- 1.7 days) than for cows in the cystic group (36.3 +/- 2.7 days). Luteinizing hormone in plasma increased during the early postpartum period (days 1 to 10) for both groups, but mean concentrations of luteinizing hormone, progesterone, and estradiol-17 beta in plasma were not different between groups then or on days -8, -4, or 0 (day of the first postpartum follicle 1.5 to 2.0 cm in diameter). During this period (days -8 to 0), estradiol-17 beta and luteinizing hormone in plasma were correlated positively, but regression coefficients differed between groups. These results suggest a partial hypothalamic and/or pituitary failure in releasing pituitary luteinizing hormone as a cause for postpartum ovarian cysts.  相似文献

Thirty-six Holstein cows fed a corn silage-based ration and 34 fed an alfalfa-grass silage-based ration were assigned according to calving date to receive either 300 mg/head per day of synthetic beta-carotene in a gelatin capsule or an empty gelatin capsule daily for the first 100 days postpartum. Supplemental vitamin A was provided at 3919 IU/kg of ration dry matter. Cervix diameters for cows supplemented with beta-carotene were smaller at 21 days and 28 days postpartum. Days from parturition to first observed estrus were less when beta-carotene was added and less when corn silage was fed. Means of other reproductive traits were more favorable for cows treated with beta-carotene (fewer services per conception and shorter intervals between parturition and conception). Milk progesterone at 21 and 28 days postpartum was not affected by either roughage or beta-carotene treatment. Milk from cows receiving alfalfa-grass silage contained lower somatic cell concentrations than that produced by cows fed corn silage. Milk somatic cell concentrations were lower in both roughage treatment groups for cows supplemented with beta-carotene. No effects of roughage or beta-carotene on milk production or milk fat percentages were significant.  相似文献

The influence of plasma concentrations of beta-carotene and vitamin A on in vivo progesterone production by bovine corpora lutea after gonadotropin-releasing hormone-induced LH release was assessed in 39 postpartum dairy cows. Thirty Holsteins and nine Jerseys were given 100 micrograms gonadotropin-releasing hormone on d 12 of an estrous cycle, which began from 30 to 49 d postpartum. Concentrations of beta-carotene and vitamin A in plasma and progesterone and LH in serum were determined prior to gonadotropin-releasing hormone injection (0 h); serum progesterone and LH concentrations were also determined 1, 2, and 3 h after injection of gonadotropin-releasing hormone. Serum concentrations of progesterone and LH were increased by gonadotropin-releasing hormone. Incremental progesterone production in an analysis of covariance was influenced by breed as well as the interactions of breed with vitamin A, of season with beta-carotene, and of season with vitamin A. The regression coefficients were positive for beta-carotene and negative for vitamin A in all cases. In conclusion, luteal function in the postpartum cow appears to be related to plasma concentrations of beta-carotene and vitamin A.  相似文献

This study examined the effects of multiple subcutaneous glucagon injections with or without co-administration of oral glycerol on energy status-related blood metabolites and hormones of Holstein dairy cows in the first 2 wk postpartum. Twenty multiparous cows were fed a dry cow ration supplemented with 6 kg of cracked corn during the dry period to increase the likelihood of developing postpartal fatty liver syndrome. Cows with a body condition score of ≥3.5 points (1- to 5-point scale) were assigned randomly to 1 of 4 treatment groups: saline, glucagon, glycerol, or glucagon plus glycerol. Following treatment, serial blood samples were collected over an 8-h period to determine the effects of glucagon and glycerol on blood metabolites and hormones. Treatment effects were determined by comparing the concentrations of metabolites and hormones during the first 4-h period and the entire 8-h period after treatment administration (time 0) with the concentration of the same compounds at time 0 on d 1, 7, and 13 postpartum. Administration of glucagon alone increased concentrations of plasma glucagon and insulin on d 1, 7, and 13 and increased plasma glucose and decreased plasma nonesterified fatty acids (NEFA) on d 7 and 13 postpartum relative to the saline group. Administration of glycerol alone increased plasma glucose on d 7 and plasma triacylglycerols on d 1 postpartum. Glycerol administration also decreased plasma glucagon and NEFA on d 1, 7, and 13 and plasma β-hydroxybutyrate (BHBA) on d 1 postpartum relative to the saline group. Administration of glucagon plus glycerol increased and sustained concentrations of plasma glucagon, glucose, and insulin on d 1, 7, and 13 and decreased plasma NEFA on d 1, 7, and 13 and BHBA on d 1 and 7. Early postpartal treatment of dairy cows with glucagon plus glycerol increased plasma glucose and insulin, decreased plasma NEFA and BHBA, and increased secretion of liver NEFA as plasma triacylglycerols. This suggests that glucagon and glycerol, when co-administered, act to decrease the likelihood of metabolism-related syndrome development in dairy cows.  相似文献

We examined the long-term effects of daily subcutaneous injections of 15 mg of glucagon during the first 14 d postpartum with or without coadministration of 400 mL of pure glycerol orally on blood metabolites and hormones and liver composition of Holstein dairy cows during early lactation. Fourteen multiparous cows with body condition score of ≥3.5 points (1-5 point scale) were assigned randomly to one of 4 treatment groups—saline, glucagon, glycerol, or glucagon plus glycerol. Fatty liver syndrome was induced by feeding cows a dry-cow ration supplemented with 6 kg of cracked corn daily during the last 6 wk of the dry period. Compared with saline treatment (n = 3), coadministration of glucagon and glycerol (n = 4) increased plasma glucose and insulin and decreased plasma nonesterified fatty acid concentrations in both treatment weeks, whereas glucagon alone (n = 3) produced similar changes plus a decrease in plasma β-hydroxybutyrate in the second week only. No significant changes were observed for the glycerol alone treatment (n = 4). We conclude that a single daily dose of glycerol for the first 14 d postpartum may potentiate the action of glucagon in the first treatment days to alleviate some symptoms of fatty liver syndrome, such as the increase in plasma nonesterified fatty acids and the decrease in plasma glucose and insulin, in Holstein dairy cows after parturition.  相似文献

Fifty-six Holstein cows were used in a replicated study to determine whether supplemental beta-carotene improved reproductive performance. Each of two replicates was of completely randomized design with 2 X 2 factorial arrangement of two diets with or without beta-carotene supplementation. On a dry matter basis, diet 1 was 5% hay, 20% haylage, 25% corn silage, and 50% concentrate. Diet 2 was 7.5% hay, 42.5% corn silage, and 50% concentrate. The diets contained adequate amount of vitamins A, D, and E. From 10 d postpartum until pregnancy was confirmed by rectal palpation, half the cows on each diet received a supplement of 400 mg beta-carotene per head daily. The remaining cows on each diet received a supplement of 160,000 IU vitamin A per head daily. Supplemental beta-carotene increased plasma beta-carotene throughout the trials. Median days to first ovulation, first service, days open, and mean services per conception were: 22, 77, 97, and 1.6 for cows receiving beta-carotene supplement compared with 19.5, 73, 82, and 1.9 in controls. Supplementation did not affect first service conception rate, uterine involution, or milk yield. Incidence of follicular cysts, luteal cysts, pyometra, and endometritis in cows fed beta-carotene were 11, 7, 0, and 7% compared with 8, 21, 4, and 13% in control cows. Supplemental beta-carotene did not improve the fertility of Holstein cows.  相似文献

Sixty-two multiparous and 35 primiparous Holstein cows were assigned randomly at 10 days postpartum to receive a ration with or without 300 mg beta-carotene/cow per day. Multiparous and primiparous cows were grouped separately and group-fed complete rations once daily. Incidence of ovarian cysts (26% by rectal palpation) was not affected by beta-carotene fed. Multiparous cows had greater incidence (39%) of ovarian cysts than primiparous cows (11%). Fifty-seven percent of cysts were classified follicular by rectal palpation. Progesterone concentration of milk also was used for diagnosis of type of cyst. Cows with ovarian cysts and with progesterone concentrations in milk less than 1 ng/ml were classified follicular, and those having concentration greater than 1 ng/ml were classified luteal. As determined by milk progesterone, rectal palpation was more accurate for diagnosis of luteal cysts than for diagnosis of follicular cysts. Progesterone concentrations of milk for animals with luteal and follicular cysts were 10.66 +/- 1.29 and .37 +/- .07 ng/ml. beta-Carotene did not affect response or days to respond to treatment with human chorionic gonadotropin or gonadotropin-releasing hormone. Supplemental beta-carotene was not beneficial for reducing incidence of ovarian cysts in cows receiving an adequate supply of beta-carotene in their diet.  相似文献

Four lactating Holstein cows averaging 155 d postpartum were used in a crossover design to examine the influence of somatotropin administration (33 mg/d for 28 d) and dietary CP (11 vs. 16% CP) concentration on physiological responses to single intravenous injections of glucagon, insulin, glucose, and somatotropin. Dietary CP was without influence on plasma hormone or metabolite responses to the challenges but increased basal concentrations of FFA. Basal concentrations of glucagon, glucose, and insulin were not significantly increased by somatotropin treatment; however, there was a tendency for insulin concentrations to be higher in somatotropin-treated cows. Somatotropin treatment increased basal concentrations of somatotropin and FFA. Increases in glucose concentrations after glucagon challenge occurred more quickly, reached a greater peak, and lasted longer for cows treated with somatotropin than for control cows. Insulin concentrations were decreased after somatotropin challenge in control cows, but not in somatotropin-treated cows. Responses of metabolites and hormones to glucose or somatotropin challenges were not influenced by somatotropin treatment. Changes in the metabolism of glucose and insulin suggest that cows treated with somatotropin have a greater ability to synthesize and conserve glucose than control cows. Data obtained support the concept of the homeorhetic action of somatotropin.  相似文献

This study investigated the effects of short-term food restriction or supplementation on folliculogenesis and plasma and intrafollicular metabolite and hormone concentrations. Ewes were randomly assigned to three groups: the control group received a maintenance diet (M) while the supplemented group and restricted group received 1.5×M and 0.5×M respectively on days 6-12 of their estrous cycle. Estrus was synchronized by intravaginal progestogen sponges for 12 days. On days 7-12, blood samples were taken. After slaughter, the ovarian follicles were classified and the follicular fluid was collected. Compared with restriction, supplementation shortened the estrous cycle length, decreased the number of follicles 2.5-3.5?mm and follicular fluid estradiol (E2) concentration, increased the number of follicles>3.5?mm and plasma glucose, insulin and glucagon concentrations, and augmented the volume of follicles>2.5?mm. Restricted ewes had higher intrafollicular insulin concentration, but it was similar to that of supplemented ewes. Compared with follicles≤2.5?mm, the intrafollicular glucose and E2 concentrations were increased and the testosterone, insulin, and glucagon concentrations and lactate dehydrogenase (LDH) activity were decreased in follicles>2.5?mm. Only in restricted ewes were intrafollicular LDH and testosterone concentrations in follicles≤2.5?mm not different from those in follicles≤2.5?mm. In conclusion, the mechanism by which short-term dietary restriction inhibits folliculogenesis may involve responses to intrafollicular increased E2, testosterone, and LDH levels in late-stage follicles. This may not be due to the variation of intrafollicular insulin level but rather due to decreased circulating levels of glucose, insulin, and glucagon.  相似文献

A total of 155 high yielding dairy cows were used to study the effect of dietary beta-carotene (carotene) on fertility. The diet of group 1 was supplemented with 69 mg retinyl acetate per cow daily before calving and 96 mg retinyl acetate after calving: diet of group 2 was supplemented with 500 mg carotene per cow before calving and 96 mg retinyl acetate after calving; and that of group 3 with 500 mg carotene before and 700 mg after calving. Conception rates were 53.9, 42.2, and 27.9% for cows of groups 1, 2, and 3. Days open were 110 and 112 for young (second and third lactation) and old (fourth and later lactations) cows of the group 1; 130 and 129 for young and old cows of group 2; and 133 and 166 for young and old cows of group 3. Number of inseminations per conception was 1.8 and 1.8, 2.2 and 2.2, and 2.5 and 3.4 for the same groups, respectively. Within group 3 the percent of cows pregnant 120 d after calving was 57 and 20 for the young and old cows. Correlation coefficients between plasma carotene of 53 old cows of all groups 4 and 8 wk after calving, and the number of open days and the number of inseminations per conception were significant. The intake of high carotene may have adverse effects on the fertility of dairy cows.  相似文献

Twenty-five dairy cows were assigned randomly by breed to flumethasone treatment (10 mug/day, 13 cows) or placebo control (12 cows) from 4 to 44 wk of lactation. Lactation means and trends with stage of lactation were treatment responses. Mean milk yields of cows supplemented with flumethasone were not significantly different from controls. Mean metabolite (glucose, nonesterified fatty acids, and total esterified fatty acids) and hormone concentrations (corticoids, insulin, and prolactin) of blood plasma and their trends throughout lactation were unaffected by supplementation. Data were pooled to determine effects of stage of lactation, temperature, pregnancy, and month on these variables. Changes during lactation were systematic for feed intake, body weight, milk yield, milk components except somatic cells, blood metabolites, and prolactin. At environmental temperatures above 18 C, effects were consistently negative for milk yield and composition, but only above 26 C for feed intake. Prolactin of plasma increased with increasing temperature to 18.2 C and then decreased. No other plasma metabolite or hormone was affected by days pregnant, age, or temperature. Months affected feed intake, milk yield, milk fat percentages, all blood metabolites, and prolactin. Prolactin concentrations increased as daylight hours increased. Our inability to augment established lactation by feeding a supplement of synthetic glucocorticoid to lactating cows is consistent with the view that a lack of avialable corticoids does not limit persistency of lactation in the cow.  相似文献

The effect of feeding Ca soaps of fatty acids for 120 d from parturition on productive and reproductive performance of dairy cows was examined, together with changes in blood lipids and progesterone. Cows fed Ca soaps received 1.5 Mcal NE1 d more than controls fed isonitrogenous and equi-forage rations. Cows fed Ca soaps produced more milk and fat for 60 d, and more FCM for 90 d after calving. In these cows, BW decreased more, and reached a minimum later than controls. Plasma free fatty acids were higher until 50 d postpartum, when phospholipid, and later triglyceride, concentrations were elevated. Cows fed soaps commenced ovarian cyclicity later than controls as shown by frequency distribution of the time required to reach cyclic plasma progesterone concentrations, 2 ng/ml progesterone in plasma, or behavioral estrus. However, once cyclicity commenced, more fat-fed cows had normal cycle length (18 to 26 d) than controls. Progesterone concentrations were higher in Ca soap-fed cows in the luteal phase before AI and were higher 9 and 24 d after AI in percent that conceived. Conception rate was higher in cows fed Ca soaps for 2nd to 4th AI, a higher proportion of cows were pregnant at 150 d after calving and number of open days was reduced. Feeding Ca soaps of fatty acids resulted in higher production at the expense of body reserves close to calving, and subsequently enhanced lipid concentrations in plasma. Higher progesterone production was accompanied by increased pregnancy rate and reduced open days.  相似文献
