本文以姬松茸菌丝体的深层发酵液为原料,研究了姬松茸保健酒的制备工艺。先将姬松茸菌丝体进行深层发酵、分离,并对分离后菌丝进行超声破碎30min,然后重新将菌丝体加入到分离后的发酵液中并添加193g/L庶糖进行发酵,其发酵参数为:酒精发酵酵母接种量为1.5%,发酵温度为26℃,后酵时间为6d。此工艺下制得的保健酒的酒精度为8-9°,粗多糖含量大于3.00%,总酸度为0.5g,L,酒质柔顺爽口,具有姬松茸独特的香气。  相似文献

研究了不同种龄对姬松茸液体发酵过程中菌丝体干质量、胞外多糖含量、蔗糖利用率的影响,并运用动力学分析的方法对姬松茸菌体发酵菌丝体干质量、胞外多糖含量、蔗糖利用率、溶氧进行相关分析。结果表明,4 d种龄的姬松茸发酵液的菌丝体干质量、胞外多糖含量和蔗糖利用率均优于其他种龄的姬松茸发酵液,且通过5 L发酵罐放大试验菌丝体干质量、胞外多糖含量、蔗糖利用率分别为1.417 g/100 mL、6.967 g/L、82.5%,较摇瓶发酵培养分别提高了45.37%、20.41%、5.87%。通过各发酵参数的相关性分析表明,姬松茸液体深层发酵菌丝体干质量、胞外多糖含量同蔗糖利用率、溶氧之间存在一定的相关性。  相似文献

以姬松茸胞外多糖含量为评价指标,采用正交试验法优化液体发酵培养基配方,通过测定小鼠血清中谷丙转氨酶(ALT)和谷草转氨酶(AST)水平,研究姬松茸发酵液对酒精性肝损伤小鼠的保护作用。结果表明,姬松茸液体发酵培养基的最佳配方为:麸皮10 g/L,马铃薯250 g/L,葡萄糖20 g/L。在此条件下,获得姬松茸胞外多糖含量为(3.58±0.13) g/L。与模型组相比,姬松茸发酵综合液组的ALT、AST水平显著降低(P<0.05),姬松茸发酵液组ALT水平显著降低(P<0.05)。表明姬松茸发酵液对小鼠酒精性肝损伤具有一定的保护作用。  相似文献

通过响应面分析(RSA)法,优化了姬松茸深层发酵菌丝体胞内多糖的热水浸提条件,即提取温度为95℃,时间为3·5h,料(湿菌体)水比为1∶3·5(w/w)。结合得率及对体外静止脾淋巴细胞增殖的促进实验结果,确定了菌丝体粗多糖的最适醇沉浓度为85%。在上述工艺条件下实际测得姬松茸深层发酵菌丝体粗多糖的提取得率为77·26g/kg,所得粗多糖在20mg/(kg·d)的剂量下对小鼠的S180移植瘤的抑瘤率为59·84%。  相似文献

研究了锌对食药用菌姬松茸生长代谢的影响,考察了姬松茸耐锌能力和富集能力以及锌对姬松茸多糖和氨基酸代谢的影响.结果表明:液体培养姬松茸对锌的最适富集质量浓度为300mg/L,在锌质量浓度为300 mg/L以内,锌对姬松茸菌丝量、多糖、氨基酸均有促进作用.锌的有机化分析表明82.2%的ZnSO4在姬松茸菌丝内以有机锌的形式存在,氨基酸分析结果显示,富锌后菌丝体和发酵液必需氨基酸含量与对照相比分别提高了12.3%和3.6%.  相似文献

目的:研究姬松茸发酵保健饮液的制备方法并对其免疫调节活性进行评价.方法:将姬松茸液态深层发酵液浓缩后,与菌丝体提取液进行混合,再经精滤、调配、装瓶、杀菌、冷却等工序制得姬松茸保健饮液.采用昆明种小鼠经口灌胃方式给与5、10和30mg/kg·d三种多糖剂量的保健饮液,通过测定免疫器官重量、腹腔巨噬细胞吞噬鸡红细胞指数、鸡红细胞作免疫源的溶血素含量及小鼠外周血中淋巴细胞酶标记染色测定、2,4-二硝基氟苯诱发的迟发型变态反应来评价该保健饮液对小鼠免疫功能的影响.结果:姬松茸发酵保健饮液能增强小鼠的胸腺指数和脾指数,提高腹腔巨噬细胞的吞噬功能和溶血素含量,增强2,4-二硝基氟苯所致的迟发型变态反应,其最适作用剂量为10mg/kg·d.结论:姬松茸发酵保健饮液可以提高小鼠非特异性免疫、特异性免疫和细胞免疫功能.  相似文献

目的:研究金针菇深层发酵的条件、培养菌丝体及菌丝体多糖的提取工艺。方法:通过发酵罐发酵研究金针菇深层发酵的条件及培养菌丝体;通过单因素试验及正交试验研究菌丝体多糖的水浴提取工艺。结果:试验确定了金针菇发酵的最佳种龄为48h、最适接种量是15%,菌丝生长曲线测定确定了发酵周期为培养时间96h,溶氧30%-50%,25℃培养;水浴提取金针菇菌丝体多糖的最佳工艺是:料液比1:60,水浴时间90min,水浴温度60℃。结论:明确了金针菇深层发酵的培养条件,通过发酵罐发酵每升发酵液可得到40g金针菇菌丝体(干重、得到了金针菇菌丝体多糖的水浴提取工艺。  相似文献

利用响应面分析法(Response Surface Methodology)对姬松茸液体发酵培养条件进行优化。方法:以土豆用量、葡萄糖浓度以及麸皮用量作为影响因子,姬松茸发酵液中多肽的含量作为响应值,采用3因素3水平的Box-Behnken中心组合设计进行响应面法分析。结果:姬松茸液体发酵液中多肽含量达最大值时的最佳培养条件为:土豆39.8g/100mL,葡萄糖3.1g/100mL,麸皮5.7g/100mL,多肽含量预测最大值为103.82mg/100mL。结论:用响应面方法优化的最佳培养条件下,姬松茸液体发酵液中多肽的含量可达最大值。  相似文献

Since grapevine ( Vitis spp .) rootstock material is being traded increasingly as disbudded woody material a lack of distinctive morphological features on such material necessitates an alternative and reliable means of identification. Methods described here were developed for rapid and efficient extraction of DNA from woody samples rich in phenolic compounds and polysaccharides, and for subsequent identification of varieties by RAPD PCR. Using these methods, and with the application of only one selected RAPD primer, we were able to differentiate sixteen rootstock varieties, including the seven varieties most commonly used in Germany. Problems commonly encountered with reproducibility of RAPD patterns were avoided by choosing primers with a dinucleotide sequence and a high G/C content that allowed a rather high annealing temperature of 45°C. Methods described here should also be useful for other horticultural crops, especially those with woody tissues rich in phenolic compounds and polysaccharides.  相似文献

An internet website (http://cpf.jrc.it/smt/) has been produced as a means of dissemination of methods of analysis and supporting spectroscopic information on monomers and additives used for food contact materials (principally packaging). The site which is aimed primarily at assisting food control laboratories in the European Union contains analytical information on monomers, starting substances and additives used in the manufacture of plastics materials. A searchable index is provided giving PM and CAS numbers for each of 255 substances. For each substance a data sheet gives regulatory information, chemical structures, physico-chemical information and background information on the use of the substance in particular plastics, and the food packaging applications. For monomers and starting substances (155 compounds) the infra-red and mass spectra are provided, and for additives (100 compounds); additionally proton NMR are available for about 50% of the entries. Where analytical methods have been developed for determining these substances as residual amounts in plastics or as trace amounts in food simulants these methods are also on the website. All information is provided in portable document file (PDF) format which means that high quality copies can be readily printed, using freely available Adobe Acrobat Reader software. The website will in future be maintained and up-dated by the European Commission's Joint Research Centre (JRC) as new substances are authorized for use by the European Commission (DG-ENTR formerly DGIII). Where analytical laboratories (food control or other) require reference substances these can be obtained free-ofcharge from a reference collection housed at the JRC and maintained in conjunction with this website compendium.  相似文献

The characterization of the aromatic profile of several apricot cultivars with molecular tracers in order to obtain objective data concerning the aromatic quality of this fruit was undertaken using headspace–solid phase microextraction (HS–SPME). Six apricot cultivars were selected according to their organoleptic characteristics: Iranien, Orangered, Goldrich, Hargrand, Rouge du Roussillon and A4025. The aromatic intensity of these varieties measured by HS–SPME–Olfactometry were defined and classified according to the presence and the intensity of grassy, fruity and apricot like notes. In the six varieties, 23 common volatile compounds were identified by HS–SPME–GC–MS. Finally, 10 compounds, ethyl acetate, hexyl acetate, limonene, β-cyclocitral, γ-decalactone, 6-methyl-5-hepten-2-one, linalool, β-ionone, menthone and (E)-hexen-2-al were recognized by HS–SPME–GC–O as responsible of the aromatic notes involved in apricot aroma and considered as molecular tracers of apricot aromatic quality which could be utilized to discriminate apricot varieties.  相似文献

The advent of the functional barrier concept in food packaging has brought with it a requirement for fast tests of permeation through potential barrier materials. In such tests it would be convenient for both foodstuffs and materials below the functional barrier (sub-barrier materials) to be represented by standard simulants. By means of inverse gas chromatography, liquid paraffin spiked with appropriate permeants was considered as a potential simulant of sub-barrier materials based on polypropylene (PP) or similar polyolefins. Experiments were performed to characterize the kinetics of the permeation of low molecular weight model permeants (octene, toluene and isopropanol) from liquid paraffin, through a surrogate potential functional barrier (25 μm-thick oriented PP) into the food simulants olive oil and 3% (w/v) acetic acid. These permeation results were interpreted in terms of three permeation kinetic models regarding the solubility of a particular model permeant in the post-barrier medium (i.e. the food simulant). The results obtained justify the development and evaluation of liquid sub-barrier simulants that would allow flexible yet rigorous testing of new laminated multilayer packaging materials.  相似文献

This paper describes the second part of a project undertaken to develop certified mussel reference materials for paralytic shellfish poisoning toxins. In the first part two interlaboratory studies were undertaken to investigate the performance of the analytical methodology for several PSP toxins, in particular saxitoxin and decarbamoyl-saxitoxin in lyophilized mussels, and to set criteria for the acceptance of results to be applied during the certification exercise. Fifteen laboratories participated in this certification study and were asked to measure saxitoxin and decarbamoyl-saxitoxin in rehydrated lyophilized mussel material and in a saxitoxin-enriched mussel material. The participants were allowed to use a method of their choice but with an extraction procedure to be strictly followed. The study included extra experiments to verify the detection limits for both saxitoxin and decarbamoyl-saxitoxin. Most participants (13 of 15) were able to meet all the criteria set for the certification study. Results for saxitoxin.2HCl yielded a certified mass fraction of <0.07 mg/kg in the rehydrated lyophilized mussels. Results obtained for decarbamoyl-saxitoxin.2HCl yielded a certified mass fraction of 1.59+/-0.20 mg/kg. The results for saxitoxin.2HCl in enriched blank mussel yielded a certified mass fraction of 0.48 +/- 0.06 mg/kg. These certified reference materials for paralytic shellfish poisoning toxins in lyophilized mussel material are the first available for laboratories to test their method for accuracy and performance.  相似文献
