The aim of the investigation was evaluation of phenolic compounds content, measured by HPLC method at selected stages of clear apple juice production with pectolytic mash enzymation from ‘Shampion’ and ‘Idared’ apples. Sum of phenolic compounds in ‘Shampion’ mash was significantly higher than in ‘Idared’ mash, 520 and 352 mg kg?1, respectively. Enzymation of ‘Idared’ mash resulted in a higher phenolic compounds decrease compared to ‘Shampion’, 23% and 14% respectively. Sum of phenolics in juices from ‘Shampion’ for Panzym at 50 °C (366 mg kg?1) was significantly higher than in juices from ‘Shampion’ for Rohapect at 20 °C and ‘Idared’ for Panzym at 50 °C (256 and 234 mg kg?1, respectively). The lowest phenolics (95 mg kg?1) was in juices from ‘Idared’ for Rohapect at 20 °C. There was a positive correlation (R2 = 0.933) between phenolics content and antioxidant activity of juices (ABTS?+). In general, enzymation at 50 °C with Panzym lead to a higher content of the phenolic compounds in juices compared to Rohapect at 20 °C.  相似文献

Folin‐Ciocalteu colorimetric assay (FC) is the most widely used assay to estimate the total phenolic content in foods, beverages, herbs and other plant extracts, but many chemical compounds may act as interfering agents, producing inaccurate estimations of the real concentration of phenolic compounds in the matrix. Based on this limitation, the objective of this study was to compare, quantitatively, the Folin‐Ciocalteu and Prussian Blue (PB) assays in estimating the total phenolic content in purple grape juices (n = 20; Vitis labrusca L.) and teas (n = 25) from different botanical origins using 96‐well microplates. PB assay presented a low limit of detection (PB = 0.27 mg/L; FC = 0.25 mg/L) and quantification (PB = 0.92 mg/L; FC = 0.82 mg/L), showing its suitability in screening the total phenolic content in grape juices and teas. FC and PB assays presented a high association (P < 0.0001) for teas (r = 0.887) and grape juices (r = 0.923). The advantages of PB over FC assay are its simplicity, low time consumption (15 min reaction as compared to 60 min reaction for the FC assay), lower usage of reagents (solutions are prepared in a mM base), and higher selectivity. Additionally, PB assay was proven to be reproducible and repeatable and, therefore, may be used as an alternative to FC assay.  相似文献

Effects of enzymatic and non-enzymatic juice pressing on key orosensory and chemical quality factors of blackcurrant juices were studied in laboratory scale using berries of five different cultivars (Mortti, Mikael, Marski, Ola and Breed15). Enzymatic processing increased the juice yield by 10–22% and the content of various phenolic compounds in juice by 4–10-fold as compared to the non-enzymatic process. Higher intensity of the mouth-drying astringency of the enzyme-aided juice was the most significant orosensory difference between the processes. Juices of different blackcurrant cultivars varied in sweetness, sourness and bitterness. The most intensive sensory attributes of the juices were sourness and puckering astringency regardless of processing method. They correlated positively with each other and were contributed by acid content and pH. In enzyme-aided juices, the contents of flavonol glycosides and hydroxycinnamic acids were associated with mouth-drying astringency, and sugar/acid ratio correlated with sweetness. These correlations were less clear in non-enzyme juices possibly due to lower content of phenolic compounds and the high content of pectin.  相似文献

Using a modified method for the determination of the18O/16O ratio in water the δ18O values for authentic apple juice concentrates (70 °Brix) of known origin, for industrially produced invert sugar syrups, for commercial apple juice concentrates, and for spiked apple juice concentrates have been measured. The δ18O values for the authentic apple juice concentrates were correlated to the δ18O values of the related pressed (single strenght) apple juices and to the sugar content of the concentrates in °Brix. The results for the samples were compared to and evaluated together with the δ13C values for the sugars. Concluding from the results, a minimum value for authentic apple juice concentrate of 0 to 1%., depending on the regional origin of the material, is suggested, while the determination of the δ18O value of apple juice concentrate is preferably used in addition to the already well known methods on the basis of stable isotope measurement.  相似文献

Reverse osmosis (RO) concentration of apple juices from 10-20 Brix using a commercial plate and frame RO system at various temperatures and pressures demonstrated that high resistance (HR) membranes are more efficient in sugar retention than cellulose acetate (CA) membranes. Both temperature and pressure had a significant effect on solute recovery. Optimum processing conditions for HR membranes were 40°C at 40 bar, 98% sugar recovery and 16.7L/m2/hr average processing capacity (flux). An increase of 3-4% average processing capacity per 1°C increase in temperature was observed. Operating conditions and membranes did not modify fructose content or glucose/fructose ratio of the RO concentrated juices.  相似文献

The purpose of this study was to investigate the changes that occur during conventional clarification using gelatin, bentonite, silica sol, and water-soluble chitosan on the phenolic compounds, antioxidant activity, and color of apple juice. The apple material used in this study was of two varieties: Sampion and Idared. The changes in the polyphenols composition (procyanidins, hydroxycinnamic derivatives, and dihydrochalcones) were monitored through the clarification process. Sampion apple control juices contained more total polyphenols than do Idared apple juices. In Sampion variety apple juice, the dominant polyphenols are the flavan-3-ols (86% of total polyphenols), followed by hydroxycinnamic acids (9.7%), dihydrochalcones (3.0%), and flavonols (1.3%). In Idared apple juice the hydroxycinnamic acids (especially chlorogenic acid) are dominant (about 48% of total polyphenols), followed by flavan-3-ols (40%). However, the concentration of polymeric procyanidins in Sampion apple juices was 62.8 and 46.3% less when the Profloc (chitosan) and gelatin treatments were used, respectively. Aktivbentonit and Puranit (bentonite) supplementary added in juices clarification have some protective effect on polymeric procyanidins only with Profloc treatment. That kind of effect was not observed in Idared apple juices with almost eight times smaller polymeric procyanidins concentration than in Sampion apple juices. The antioxidant activity, measured by the DPPH (1,1-diphenyl-2-picrylhydrazyl radical) method, ranged from 0.20 mg TEAC/mL in Idared apple juice to 0.30 mg TEAC/mL in Sampion apple juice, measured by the ABTS method, from 0.17 to 0.48 mg TEAC/mL, respectively. Clarification of apple juices with chosen clarifying agents has statistically no significant (p>0.05) influence on antioxidant capacity. This study suggests that chitosan can be used as a conventional clarifying aid of apple juices and that treatment has no impact on their biochemical parameters.  相似文献

The present paper reports the application of an optimized pre-column derivatization procedure with aminoquinolyl-N-hydroxysuccinimidyl carbamate (AQC) to the determination of 21 free amino acids in fruit juices. The method permitted the optimal separation of 21 amino acids and ammonium in 30 min. Excellent response linearity was obtained from 2 to 1,000 μM for all amino acids, except tryptophan, for which the linearity was 5–1,000 μM. The fluorescence detection limits ranged from 0.08 pmol (phenylalanine) to 0.69 pmol (cysteine), except for tryptophan (7.89 pmol), for a signal-to-noise ratio of 3. The relative standard deviations of peak areas ranged from 0.1 to 0.4 % for intraday analysis and from 2.1 to 5.0 % for interday analysis. The method was applied to analyze the free amino acids in six fruit juices, and the recoveries of the proposed method were 90.4 to 103.3 %. The total free amino acid content ranged from 56.97 mg L?1 (apple) to 469.45 mg L?1 (longan) in six fruit juices. Aspartic acid (Asp), glutamic acid (Glu), asparagine (Asn), serine (Ser), and glutamine (Gln) were present in higher levels in most samples, whereas cysteine (Cys) was below the detection limit in the selected samples. Notable advantages of this method include its speed, ease, low cost, good repeatability, high sensitivity, excellent precision and accuracy, and anti-interference in high-sugar matrices.  相似文献

The in vitro digestive stability of phenolic compounds and the antioxidant capacity of five kinds of commonly consumed fruit juices in the daily diet, including apple juice (AJ), orange juice (OJ), grape juice (GJ), pomelo juice (PJ) and kiwifruit juice (KJ), were studied. Following in vitro digestion, the total phenolic (TP) content of fruit juices decreased to different extents by 35%, 25.3%, 23.5%, 22.2% and 7.8% for KJ, OJ, PJ, GJ and AJ, respectively. The individual phenolic content showed similar changes to the TP content, showing reductions of naringenin‐trisaccharide in OJ and PJ, epicatechin in GJ, and chlorogenic acid in AJ by 43.74%, 27.59%, 47.11% and 33.28%, respectively. Conversely, the antioxidant capacity of fruit juices during digestion measured by ABTS assay increased from 4.79% to 35.53%, except in KJ, which decreased by 19.34%. These results show the health benefits of fruit juices after processing and contribute towards establishing suitable dietary recommendations.  相似文献

以来自7?个不同主产区的21?个主栽品种,共41?份苹果鲜果为研究对象,对每个品种鲜榨浊汁的出汁率、浊度、果汁颜色、褐变度、透光率、可溶性固形物、可滴定酸、总酚、糖、有机酸、VC、矿物质、抗氧化性等品质指标进行测定。运用描述性分析、逐步线性判别分析进行数据处理。结果表明:不同品种或不同产地的苹果浊汁,品质差异较大。总体来看,a*值、原始浊度、褐变度、可滴定酸、葡萄糖、蔗糖、苹果酸等指标的变幅均较大,分别为94.07%、70.41%、42.86%、35.26%、42.02%、42.40%、37.69%。对于不同品种的苹果浊汁,由于褐变度、总酚、抗氧化性等色泽品质指标以及糖、酸等营养品质指标间具有较大差异,其分布呈现分散状态。其中新红星品种的褐变度、总酚含量、DPPH自由基清除率等品质指标普遍居高,色泽品质差异最显著。富士品种大部分样品的果糖、葡萄糖、蔗糖、苹果酸等指标含量较高,具有较好的营养品质。然而对于不同产地间的样品,其浊度、可滴定酸、可溶性固形物等加工品质指标具有较大差异,样品分布较为分散。河北省样品的出汁率、原始浊度、可滴定酸等指标普遍较低,加工品质较差。来自山东省不同品种苹果浊汁的样品以及来自不同产地富士苹果浊汁的样品间均具有较大差异,并且分别获得了77.8%和72.7%的判别准确率。以上结果表明,不同产地、不同品种苹果原料制得的苹果浊汁存在较大差异,通过对其品质指标进行差异性分析,可为上游育种与种植产业结构调整提供数据基础;通过判别模型的建立实现品种与产地的识别与预测,可以指导企业对苹果原料进行制汁专用化筛选。  相似文献

以绿豆芽和苹果为原料,研制复合型澄清果汁饮料。用两种汁液比、汁液量、蔗糖量、柠檬酸量为因素,采用4因素3水平L9(3^4)正交实验设计,筛选最佳实验配方。结果表明,最佳配方为绿豆芽:苹果汁为1:1、汁液量22%、蔗糖用量7%、柠檬酸用量0.02%。  相似文献

Focusing on 17 constituents, the polyphenol profiles of juices freshly made from various dessert (n = 4) and cider apple cultivars (n = 7) as well as commercially available apple juices (n = 24) were investigated using high-performance liquid chromatography-photodiode array detection (HPLC-DAD) and (HPLC)-electrospray ionization-tandem mass spectrometry (ESI(neg)-MS/MS) analyses. Significant differences in the total polyphenol content as well as the profiles of the apple cultivars under study were observed. For dessert apples the total polyphenol content ranged from 154 to 178 mg/L, whereas for 'old' German cider apple cultivars 261-970 mg/L were determined. Boskoop showed the highest (970 mg/L) and Granny Smith the lowest (154 mg/L) polyphenol content of the freshly prepared samples under study. Hydroxycinnamic acids, with chlorogenic acid as dominating constituent, ranged from 57 to 68 mg/L as well as from 134-593 mg/L in juices made from dessert apples and that from cider apples, respectively. Dessert apple juices showed lower contents of dihydrochalcones (10-35 mg/L) and flavan-3-ols (50-95 mg/L) compared to that of cider apples (34-171 mg/L and 70-393 mg/L, respectively). Quercetin and its derivatives were found from 0.4-4 mg/L and 0.4-27 mg/L in juices made from dessert apples and that of cider apples, respectively. Compared with freshly made juices, lower contents of polyphenols were determined in the commercial samples under study. Amounts ranging from 110-459 mg/L, dominated by chlorogenic acid with concentrations from 53-217 mg/L, were determined. Information about cultivar-typical apple polyphenol content and profile is important for bioactivity studies and, consequently, essential for the development of consumer-relevant products with particular nutritional functionalities.  相似文献

Abstract The effects of conventional clarification and ultrafiltration techniques on the phenolic composition of apple juice was studied. Since the health benefit of fruits and fruit juices have been ascribed, in part, to the phenolic antioxidants, it is very important to know what the effects of processing steps are on the level of phenolic compounds in fruit juices. The total losses of phenolic compounds were determined to be 28% and 7% during conventional clarification using gelatin and bentonite, and ultrafiltration, respectively. A total of 14 phenolic compounds were identified in apple juice prepared from Golden Delicious apples. The phenolic compounds were found to be based mainly on hydroxycinnamic acids (∼ 50% of total phenolics) and chlorogenic acid was found to be the major phenolic compound with its concentration as high as 465.45 mg 1−1 in raw apple juice.  相似文献

精选新疆地区、黄河流域16种红富士苹果,进行果实性状、营养成分分析和感官评价:新疆阿克苏、伊犁特克斯和陕西洛川的红富士苹果表现最佳。4地红富士苹果平均单果重甘肃陕西新疆山东,种群内变异性大(CV15%),具有显著性差异(p=00.05)。16种红富士苹果种群内部营养成分变异性较大(CV10%),尤其Vc、蛋白、膳食纤维(CV25%);不同产地间红富士还原糖(p=00.05)、可溶性固形物(p=0.05)、膳食纤维(p=0.0230.05)具有显著性差异。风味评分与固酸比之间具有显著正相关关系(p=00.05,r=0.886),新疆红富士具有更高的固酸比,且与其它地区具有显著性差异(p=0.0090.05),使得新疆红富士苹果更能满足人们对于风味的较高要求且与其它地区具有显著性差异(p=0.0030.05)。此外,还原糖与可溶性固形物具有显著正相关关系(p=0.0010.05,r=0.75),总酸和可溶性固形物之间存在显著正相关关系(p=0.0140.05,r=0.602)。  相似文献

Fifty-one samples from 12 geographic growing regions of Turkey and 3 varieties common for production of apple juice concentrate were processed to apple juice and analysed for their sugar composition. Enzymatic analyses was used to determine soluble sugars of apple juice. Glucose, fructose, sucrose, total sugar concentrations (g/l) and total soluble solids (%) of apple juices were ranged, respectively, as follows: 9.30-32.2, 66.10-96.00, 8.5-55.10, 110.90-164.40, 11.80-18.60. Cultivar significantly influenced the content of all the sugar analysed. Significant differences in fructose and sucrose concentrations have also been found between some growing areas.  相似文献

The antioxidative capacities of a number of Rubus species of varied pigmentation have been investigated. In addition, total phenol, anthocyanin and ascorbic acid contents have been determined. Two methods to assess the antioxidant potential of fruit juices have been used. The antioxidant capacities of the fruit ranged from 0 to 25.3 µmol Trolox equivalents g−1 (TEAC) or from 190 to 66 000 µmol l−1 ferric reducing antioxidant power (FRAP). Ascorbic acid contributes only minimally to the antioxidant potential of Rubus juices (<10%, TEAC). There are apparent linear relationships between antioxidant capacity (assessed as both TEAC and FRAP) and total phenols (rxy = 0.6713 and 0.9646 respectively). Also, anthocyanin content has a minor influence on antioxidant capacity (rxy = 0.3774, TEAC; rxy = 0.5883, FRAP). The sample with the highest antioxidant capacity (Rubus caucasicus) had the highest phenol content, but only a low percentage was represented by anthocyanins. The present study demonstrates the potential of certain wild Rubus species, notably R caucasicus, for improvement of nutritional value through germplasm enhancement programmes. © 2000 Society of Chemical Industry  相似文献

Effects of pomace maceration on yield, turbidity, cloud stability, composition of phenolics, antioxidant activity and colour properties were studied, to evaluate the potential applicability of enzyme preparations in puree-enriched cloudy apple juice production. The yield of mixed juice and puree from pomace obtained in the enzymatic processing of apple ranged from 92.3% to 95.3%, significantly higher than the yield from the control without enzymatic pomace treatment (81.8%). Higher turbidity was obtained upon pomace treatment with Pectinex XXL and Pectinex Ultra SPL enzymes. The total content of phenolic compounds in apple pomace was higher than in raw juices (1520 mg/kg and 441 mg/L, respectively). The total polyphenol yields were higher in juices treated with Pectinex AFP L-4, Pectinex Yield Mash and Pectinex XXL, as compared to the control treatment. During 6 months of storage, a significant change was observed in the content of polyphenols, especially in procyanidin fractions.  相似文献

BACKGROUND: The effect of high‐intensity pulsed electric field (HIPEF) processing (35 kV cm?1 for 1500 µs using 6‐µs bipolar pulses at 200 Hz) on the antioxidant features (vitamin C, β‐carotene, total phenolic compounds and antioxidant capacity) of carrot juice as well as on peroxidase activity was investigated and compared to the observed in heat pasteurised juices (90 °C for 60 s or 30 s) having the fresh juice as a reference. RESULTS: HIPEF and heat‐treated carrot juices had higher β‐carotene and lower vitamin C contents than the untreated juices immediately after processing. The antioxidant capacity of the juices was significantly modified neither by HIPEF nor by thermal treatments. POD activity decreased drastically (≥93.3%) after processing irrespective of the treatment applied. Vitamin C and β‐carotene content decreased throughout the storage following an exponential trend (R2 = 0.801–0.984) with degradation rates between 1.7 × 10?2 and 3.5 × 10?2 day?1. Vitamin C and β‐carotene contents were better maintained in HIPEF‐treated than in heat‐pasteurised juices throughout the storage. Total phenolic content and the antioxidant capacity of the HIPEF‐treated juice did not substantially differ from that of the thermally treated juice for 56 days. The quality of 7 commercial apple juices, after 11 ‐mo storage, was assessed on the basis of polyphenol and vitamin C contents, and also on the Trolox® equivalent antioxidant capacity (TEAC) value. Phenolic acids, flavonoids, and vitamin C were quantified using high‐performance liquid chromatography. It was shown that the content of phenolic acids in juices varied up to 2‐fold and flavonoids up to 4.5‐fold. Moreover, storage of juices resulted in a decrease of phenolic acids (5% to 21%), flavonoids (8% to 19%), and also of TEAC values (6% to 14%). Significant correlations between polyphenol content and TEAC values for fresh, stored, and all juices (fresh plus stored) and no correlation for vitamin C indicate that the TEAC value of apple juices depends mainly on their polyphenol content. Based on these results, it is concluded that the TEAC value, directly reflecting polyphenol content, may serve as a useful parameter to assess the quality of apple juice at any stage of its shelf life.  相似文献
