Mammary arteriovenous differences of prolactin concentration and net mammary uptake of prolactin from blood were quantified near parturition in 9 dairy cows. Six cows were milked once daily for at least 6 d before parturition, and prepartum lactogenesis occurred in 3 of 6 cows. Prepartum milking 2 or more d before parturition abruptly increased secretion of prolactin into blood, but milkings within 1 d before or after parturition did not increase prolactin secretion. Concentrations of prolactin in whole milk sampled over 8 d before parturition (64.5 ng/ml) were substantially greater than those occurring several days after parturition (19 ng/ml). Milk prolactin concentrations were unaffected by the successful induction of prepartum lactogenesis, which greatly increased prepartum yields of milk (2 to 8 kg/milking). Therefore, the alveolar lumenal content of prolactin was greatest in pregnant cows with prepartum lactogenesis. This enhanced content of intraalveolar prolactin before parturition was associated with an absence of mammary uptake of prolactin immediately prior to ejection of the prolactin-containing milk from the alveoli. However, prolactin uptake was quickly restored to about 2 micrograms/min per half udder shortly after milk ejection. During the prepartum period, an enhanced intraalveolar reservoir of 200 to 400 micrograms prolactin, due to induction of prepartum lactogenesis, appears to saturate temporarily all putative sites for uptake of prolactin from blood.  相似文献

Mammary uptake of insulin from blood was quantified near parturition in nine dairy cows, some of which were milked prepartum to induce prepartum lactogenesis. Milk samples were immunoassayed for concentrations of insulin, insulin-like growth factor I, and insulin-like growth factor II. Cows milked prepartum, but in which prepartum lactogenesis did not occur (denoted as unsuccessful group), had higher arterial concentrations and greater mammary uptake of insulin during several days immediately preceding parturition than did those with prepartum lactogenesis. Although mammary uptake of insulin was measurable before parturition, estimated uptake on postpartum d 4 through 6 was negligible. Colostrum and milk contained very substantial quantities of insulin, insulin-like growth factor I, and insulin-like growth factor II. Cows in which prepartum lactogenesis was induced (denoted as successful group) had lower concentrations of both forms of insulin-like growth factor in prepartum samples of milk. Cows of the successful group also had a decreasing prepartum profile of milk insulin, but the unsuccessful group had an increasing trend until parturition. Milk concentrations of insulin and both forms of insulin-like growth factor declined rapidly after parturition and completion of lactogenesis.  相似文献

A field study using seven Holstein herds was conducted to determine effects of prepartum milking on milk production, health disorders, and reproductive performance. In each herd, 80 cows (30% first lactation cows) were assigned 1 mo prior to expected calving date to one of two treatments: postpartum or prepartum milking. The group milked prepartum was machine-milked twice daily at regular milking intervals beginning 14 d prior to date of expected calving. The group milked postpartum was milked for the first time after calving. The day prior to calving, 36, 33, and 31% of the cows milked prepartum produced less than 4.5 kg, 4.5 to 9 kg, and greater than 9 kg of milk, respectively. No relationship existed between days milked prepartum and prepartum milk yield. Lactation milk yield and persistency were not affected by prepartum milking. Prepartum milking reduced incidence of milk fever and mastitis during the 1st mo after parturition. Treatment was not a significant source of variation for reproductive performance or body condition; however, culling was higher for cows milked postpartum. Results indicate no adverse effects on cow performance due to prepartum milking nor increase in lactation milk yield.  相似文献

Colostrum formation and lactogenesis in the mammary gland and the timing of parturition are regulated by endocrine signals. Changes in progesterone (P4) and prolactin (PRL) are considered key events that inhibit colostrum formation, trigger parturition, and signal the onset of lactation. The goal of our study was to determine if colostrum yield and composition and immunoglobulin transfer are affected by prepartum milking relative to the decrease in P4, peak of PRL, or occurrence of parturition. Twenty-three multiparous cows were randomly assigned to 1 of 2 groups: (1) control with first milking at 4 h postcalving (CON, n = 11), and (2) treatment group with first milking approximately 1 d before calving and second milking at 4 h after parturition (APM, n = 12). Colostrum yields were recorded and proportional samples were analyzed for immunoglobulin G (IgG) concentration. Blood plasma samples for the analyses of P4 and PRL were collected 3 times daily at 8-h intervals for 4 d prepartum and again taken at 4 h after parturition. Total colostrum mass of APM cows was higher than that of CON cows. Immunoglobulin G concentration and protein content did not differ between antepartum milking in APM cows and postpartum milking in CON cows. Colostrum IgG concentration and protein content in APM cows at the postpartum milking were lower compared with the IgG concentration established at the prepartum (APM) and postpartum milkings of CON cows. Immunoglobulin G mass did not differ in first and second colostrum collection in APM cows but was lower compared with that of CON cows. The sum of IgG mass in APM cows (prepartum + postpartum collections) did not differ from that of CON cows. Lactose and fat in milk (concentration and mass) increased from first to second milking in APM cows. Total mass of lactose and fat in APM cows (prepartum + postpartum collections) was greater compared with that of CON cows. The finding that the time of milking relative to parturition, P4 decrease, and PRL peak slightly affected yield and quality of colostrum emphasizes the complex interactions of numerous endocrine and morphological changes occurring during colostrogenesis and lactogenesis in dairy cows. The considerably rapid transfer of immunoglobulins into colostrum of prepartum-milked cows within a few hours leads to the hypothesis that the transfer of IgG can be very fast and—contrary to earlier findings—persist at least until parturition.  相似文献

Four multiparous cows were used to determine whether continuous twice daily milking of half the udder throughout the dry period would alter the prepartum accumulation of prolactin in secreted milk. The control half of each udder was allowed a dry period of 43 d, and twice daily milking was reinitiated in this half udder 11 d before parturition, whereas the treated half was milked continuously throughout. Milk from the treated half decreased to very low yields despite continuous milking, whereas concentrations of prolactin in milk remained low and without much day-to-day variation within each cow. Concentrations of prolactin in milk and yields of milk increased during the week before parturition in both halves of the udder, but there were no significant differences due to previous continuous milking. Therefore, prepartum accumulation of high concentrations of prolactin in secreted milk did not appear to depend on prior occurrence of a dry period. Elevated concentrations of prolactin in milk declined rapidly and equally in each half udder after parturition.  相似文献

This study investigated the immediate and long-term effects of temporary alterations to postpartum milking frequency (MF) on milk production, body condition score (BCS), and indicators of energy status in pasture-grazed cows supplemented with concentrates. Multiparous Holstein-Friesian cows (n = 150) were randomly assigned to 1 of 5 groups at calving: milked twice daily (2×) throughout lactation (control), or milked either once daily (1×) or 3 times daily (3×) for 3 or 6 wk immediately postpartum, and then 2× for the remainder of lactation. During wk 1 to 3 postpartum, cows milked 1× produced 15% less milk and 17% less energy-corrected milk (ECM) than cows milked 2×. This immediate production loss increased to 20% less milk and 22% less ECM during wk 4 to 6 postpartum for cows that remained on 1× milking; these animals also produced less than 1× cows switched to 2× milking after 3 wk. During wk 8 to 32, when all cows were milked 2×, those previously milked 1× had sustained reductions in milk (−6%) and ECM (−8%) yields, which were not affected by the duration of reduced postpartum MF. In contrast, cows milked 3× postpartum had 7% greater milk yields during wk 1 to 6 compared with 2× controls, irrespective of the duration of increased MF. Milk yields also remained numerically greater (+5%) during wk 8 to 32 in cows previously milked 3×. Nevertheless, yields of ECM were not increased by 3× milking, because of lower milk fat and protein contents that persisted for the rest of lactation. In addition, indicators of cow energy status reflected an increasing state of negative energy balance with increasing MF. Cows milked 1× postpartum had greater plasma glucose and lower plasma nonesterified fatty acid concentrations during the reduced MF, and plasma glucose remained lower for 2 wk after cows had switched to 2× milking. Moreover, BCS was improved relative to 2× controls from wk 5 to 6. In contrast, cows milked 3× had lower plasma glucose concentrations, greater plasma nonesterified fatty acid concentrations, and greater BCS loss during wk 1 to 3; however, greater body fat mobilization was not sustained, indicating that additional energy supplements may be required to achieve better milk production responses. In conclusion, temporary 1× milking had lactation-long negative effects on milk and milk component yields but improved cow energy status and BCS, whereas temporary 3× milking immediately increased milk yield but did not improve milk fat and protein yields in pasture-grazed cows.  相似文献

Reports over the past decade have indicated that normal lactational performance can be achieved in genetically superior and high-producing dairy cows, even when the dry period between 2 lactations is omitted. The hypothesis tested in this experiment was that normal lactogenesis I and metabolic function may be achievable in continuously milked high-yielding dairy cows as a result of the genetic selection for lactation performance and hence longevity of mammary epithelial cells. The milk production and mammary nutrient uptake in response to omission of the dry period for cows with an expected peak milk yield higher than 45 kg/d were studied in 28 Holstein dairy cows managed without bovine somatotropin. Performance and metabolic parameters were followed in late gestation and in the following early lactation. Fourteen cows were milked continuously throughout late gestation, and another 14 dairy cows experienced a 7-wk prepartum dry period. Continuous milking during the prepartum period reduced milk production in the following early lactation period by >20%. The reduced milk production could not be readily ascribed to inefficiency of the mechanisms responsible for nutrient uptake by the lactating mammary epithelial cells, nor to systemic endocrine changes. This suggests that lowered mammary nutrient uptake must have been associated with reduced mammary blood flow, metabolic activity, or both, most likely as a result of disturbed lactogenesis I prepartum or lactogenesis II postpartum triggered by as yet unknown local mechanisms. Milk protein content was elevated by 0.4 percentage units in the continuously milked cows. The underlying reason is unknown, but given the current pricing system for milk, it deserves to be further investigated.  相似文献

Postpartum production performance of dairy heifers may be enhanced by prepartum milking by alleviating the stress of the periparturient period. The objective of this study was to determine the impact of prepartum milking of dairy heifers on postpartum reproduction, udder health, milk production and other associated production characteristics. Pregnant heifers (Holstein, n=21; Jersey n=10) were assigned to either a prepartum milked (premilked, n=15) or control (n=16) group. Premilked heifers were milked twice daily starting 3 weeks prior to anticipated calving dates, and milk yield recorded at each milking. All heifers were evaluated on days 21, 14 and 7 before calving, and udder oedema scores and milk conductivity readings were recorded. Following calving, measurements were taken twice weekly to assess udder oedema, milk conductivity (indicative of udder infection), and reproductive health, which included palpation for uterine tone and uterine position, vaginal electrical impedance (VEI) and the quantification of cross-sectional area of the uterine horns (uterine difference) by transrectal ultrasonography. Uterine tone, uterine position, uterine difference, and VEI did not differ (P>0.10) with treatment. Overall, up to week six inclusive, postpartum premilked heifers had lower (P<0.01) udder oedema scores than control heifers and up to week five inclusive, had lower (P<0.01) milk conductivity readings (indicative of fewer incidences of udder infections) than control heifers. The premilked heifers of both breeds produced more milk (P<0.01) at calving and more milk overall from calving to day 60 postpartum than the control heifers. In summary, udder health and milk production were improved post calving in premilked heifers compared with controls. However, no overt differences in reproductive characteristics were observed between the premilked and control heifers.  相似文献

We hypothesized that early-lactation increased milking frequency, in combination with bovine somatotropin (bST), would improve milk yield in continuously milked (CM) primiparous glands through greater mammary epithelial cell (MEC) function, proliferation, and reduced apoptosis (cell turnover). Primiparous cows were randomly assigned to a 2 × 2 × 2 factorial with a split-plot design to either a continuous bST (+bST, n = 4) or no bST (−bST, n = 4) treatment throughout the study. Within each animal, udder halves were randomly assigned to either a CM or a 60-d dry period (control). During late gestation, CM glands were milked twice daily until calving or until spontaneous dry-off. At calving, cows were milked either twice or 4 times daily and udder-half milk yield was recorded until 30 d postpartum. Mammary biopsies were conducted on −19 ± 13, −8 ± 6, +2, +7, and +20 d relative to calving. Postpartum milk yield was reduced in CM udder halves. Reduced milk yield in CM half udders from cows administered bST and milked 4 times daily was 35% compared with 65% in CM half udders in cows not provided bST and milked twice daily. Proliferation of MEC tended to be greater in control vs. CM tissue at 8 ± 6 d prepartum. Mammary epithelial cell proliferation was greater during the prepartum period (d −19, −8) compared with postpartum time points (d 2, 7, 20). Apoptosis of MEC was not affected by dry period length, but was elevated during the first 7 d postpartum compared with levels measured at −19, −8, and 20 d. Bovine somatotropin did not alter MEC turnover in primiparous CM or control glands. The use of increased milking frequency and bST alleviated, but did not prevent, reductions in milk yield of CM primiparous cows.  相似文献

We used 48 multiparous Holstein cows to compare the response of dairy cows to a direct-fed mixture of cellulase and xylanase enzymes (1.25 L of enzyme concentrate/tonne of forage dry matter) applied to the forage portion (60% corn silage and 40% alfalfa hay) of a total mixed diet starting either in the close-up dry period, at calving, or at peak milk production. Cows were blocked by calving date and, within blocks, randomly assigned to one of four treatment diets. Treatments were: 1) an untreated control diet, 2) enzyme addition to the forage from wk 6 to 18 postpartum, 3) enzyme addition to the forage from calving to wk 18 postpartum, and 4) enzyme addition to the forage from wk 4 prepartum to wk 18 postpartum. Total mixed diets were 65% forage and 35% concentrate prepartum, and 50:50 forage:concentrate postpartum. The production of milk, solids-corrected milk, fat-corrected milk, and energy-corrected milk was higher for cows fed enzyme-treated diets than for cows fed control diet. Production was similar for cows in all enzyme-treated groups, although numerically highest for cows that started receiving enzyme-treated forages right after parturition and numerically lowest when started prepartum. Concentrations of fat, protein, and lactose in milk were similar for all treatments; yields of protein and fat were higher for cows fed enzyme-treated forages. Dry matter intake and body condition scores, both prepartum and postpartum, were similar for all diets. Eating rates, as determined in two 24-h studies, were similar for control and enzyme-treated diets. The feeding of enzyme-treated forages increased milk production. While the effect of when the feeding of enzyme-treated forages started was not statistically significant, we recommend starting soon after parturition because of the greatest total milk production when starting at that time.  相似文献

Our objective was to investigate the effects of prepartum metabolizable protein (MP) supply and management strategy on milk production and blood biomarkers in early lactation dairy cows. Ninety-six multigravida Holstein cows were used in a randomized complete block design study, blocked by calving date, and then assigned randomly to 1 of 3 treatments within block. Cows on the first treatment were fed a far-off lower MP diet [MP = 83 g/kg of dry matter (DM)] between ?55 and ?22 d before expected calving and then a close-up lower MP diet (MP = 83 g/kg of DM) until parturition (LPLP). Cows on the second treatment were fed the far-off lower MP diet between ?55 to ?22 d before expected parturition and then a prepartum higher MP diet (MP = 107 g/kg of DM) until calving (LPHP). Cows on the third treatment had a shortened 43-d dry period and were fed the prepartum higher MP diet from dry-off to parturition (SDHP). After calving, cows received the same fresh diet from d 0 to 14 and the same high diet from d 15 to 84. Data were analyzed separately for wk ?6 to ?1 and wk 1 to 12, relative to parturition. Dry matter intake from wk ?6 to ?1 was not different between LPHP and LPLP and increased for SDHP compared with LPLP. In contrast, dry matter intake for wk 1 to 12 postpartum did not change for LPHP versus LPLP or for SDHP versus LPLP. Compared with LPLP cows, LPHP cows had lower energy-corrected milk yield and tended to have decreased milk fat yield during wk 1 to 12 of lactation. Conversely, yields of energy-corrected milk and milk fat and protein were similar for SDHP compared with LPLP. Plasma urea N during wk ?3 to ?1 increased for LPHP versus LPLP and for SDHP versus LPLP; however, no differences in plasma urea N were observed postpartum. Elevated prepartum MP supply did not modify circulating total fatty acids, β-hydroxybutyrate, total protein, albumin, or aspartate aminotransferase during the prepartum and postpartum periods. Increased MP supply prepartum combined with a shorter dry period (SDHP vs. LPLP) tended to increase whole-blood β-hydroxybutyrate postpartum; however, other blood metabolites were not affected. Taken together, under the conditions of this study, elevated MP supply in close-up diets reduced milk production without affecting blood metabolites in multiparous dairy cows during early lactation. A combination of a shorter dry period and increased prepartum MP supply (i.e., SDHP vs. LPLP) improved prepartum dry matter intake without modifying energy-corrected milk yield and blood biomarkers in early lactation cows.  相似文献

The effects of hypocalcemia at parturition on concentrations of serum immunoglobulin and conglutinin, number of bacteria shed into milk, and leukograms of dairy cows were investigated from -4 wk prepartum to 4 wk postpartum. Ten healthy multiparous Holstein cows were fed a high calcium diet to induce hypocalcemia at parturition. Five cows received intramuscular parathyroid hormone to prevent hypocalcemia at parturition. All cows experienced a leukopenia (attributable to an absolute and relative neutropenia) during the 1st wk after calving, decreased serum conglutinin activity during the first 3 wk postpartum, and decreased concentration of serum IgG1 during the 3 wk before calving. At parturition, a large increase in organisms was found in foremilk (1000 to 10,000 times more than prepartum values). Neither the hematological changes nor the decreased immunoglobulin concentration was influenced by hypocalcemia or the development of milk fever. This implies that the degree of hypocalcemia observed did not have a large or irreversible influence on bacterial infection, hematological, or humoral immunity changes in periparturient cows.  相似文献

Adrenal responsiveness was evaluated by injecting 10 multiparous dairy cows with 200 IU adrenocorticotropin between -13 and -2 days prepartum (I) and postpartum between 24 and 40 h (II) and 21 and 24 days (III). Concentrations of glucocorticoids following injection were influenced by day of injection, temperature, and minimum percent relative humidity but not by breed, breed X injection day interaction, or age of cow. Likewise differences in regressions for adrenal response and mean response (ng/ml) for the three injections were nil. Mean concentrations at peak (45, 60, and 120 min postinjection samples) adjusted for preinjection concentrations also did not differ for the three periods of injection. Mean concentrations of glucocorticoids in plasma for daily samples between -13 and -2 days prepartum were 5.3 +/- .4 (n = 61), reached a peak of 14.8 +/- .3 ng/ml the day of calving, and remained high for 2 days postpartum. Estradiol increased through prepartum sampling from 23.3 to 339.6 +/- 94.1 pg/ml the day of calving, then declined abruptly. Progestins began to decline about -5 days prepartum from mean concentration of 4.09 +/- .62 (n = 25) and attained low concentrations (.30 +/- .06 ng/ml) 2 days postpartum. Although there was a surge of glucocorticoids at parturition, this was not associated with a modification in adrenal responsiveness or with prepartum concentrations of other steroid hormones of plasma. Adrenal potential in prepartum and postpartum dairy cows appears well maintained.  相似文献

Thirty-four multiparous Holstein dairy cows (780 +/- 17.2 kg body weight; 3.4 +/- 0.08 body condition score) were used in a completely randomized design to evaluate the effects of fermentable carbohydrate source on dry matter (DM) intake, milk production, and blood metabolites of transition cows. Treatments were initiated 30 d before expected calving date. After calving, all cows went onto a similar lactating cow diet. Dry matter intake was measured daily, and milk production and composition were measured weekly for 56 d after calving The control diet consisted of 11.5% ground corn, whereas the treatment diet consisted of sucrose replacing 2.7% of ground corn on a DM basis. Prepartum plasma glucose concentrations were higher (66.3 vs. 69.3 +/- 1.13 mg/dl) for cows fed the diet containing sucrose. Glucose concentrations were not different postpartum. Prepartum and postpartum nonesterified fatty acids, blood urea N, and insulin did not differ between treatments. Substitution of sucrose as a readily available carbohydrate source for ground corn did not affect prepartum or postpartum DM intake. Milk yield, 3.5% fat-corrected milk yield, and milk components did not differ between treatments. Results from this research demonstrated that partially replacing ground corn with sucrose did not enhance prepartum or postpartum intake or performance.  相似文献

Multiparous Holstein cows (n = 300) were assigned to 1 of 2 milking frequency treatments at parturition. Cows were either milked 6 times (6×) or 3 times (3×) daily to determine effects on early lactation milk yields and subsequent lactation persistency with or without use of recombinant bST (rbST). Treatments included a control group milked 3× and 3 groups milked 6× for either the first 7, 14, or 21 days in milk (DIM). Those 4 groups of cows all received rbST starting at 63 DIM. The fifth treatment group was also milked 6× for the first 21 DIM but those cows received no rbST during the entire lactation. All cows returned to 3× milking after their respective treatment periods ended. Cows milked 3× tended to produce more milk (43.2 vs. 41.5 and 41.0 ± 1.1 kg/d) during the first 9 wk of lactation compared with cows milked 6× for 7 or 21 DIM, respectively. Group milk yields after wk 9 averaged 38.3 ± 0.7 kg/d and did not differ among various groups assigned to an increased milking frequency in early lactation. Percentages of milk fat (3.8 ± 0.12%) and protein (2.9 ± 0.06%) did not differ among treatments during the first 9 wk after calving. Early lactation milk yield (41.9 ± 1.2 kg/d) did not differ between the 2 groups of cows milked 6× for 21 DIM. However, cows subsequently administered rbST (at 63 DIM) produced more milk (38.8 vs. 34.2 ± 0.9 kg/d) from wk 10 to 44. The number of cows sent to the hospital during the 305-d trial for mastitis (97), digestive disorders (14), respiratory issues (9), lameness (22), or retained placenta (16), were not affected by treatments (χ2 = 0.49). Under the conditions of this commercial dairy herd in Arizona, increasing milking frequency to 6 times daily for 7 to 21 d at the start of lactation conditions did not increase milk yield nor improve lactation persistency.  相似文献

Our objectives were to evaluate the effects of prepartum monensin supplementation and dry-period nutritional strategy on the postpartum productive performance of cows fed monensin during lactation. A total of 102 Holstein cows were enrolled in the experiment (32 primiparous and 70 multiparous). The study was a completely randomized design, with randomization restricted to balance for parity, body condition score, and expected calving date. A 2 × 2 factorial arrangement of prepartum treatments was used; the variables of interest were prepartum feeding strategy [controlled-energy diet throughout the dry period (CE) vs. controlled-energy diet from dry-off to 22 d before expected parturition, followed by a moderate-energy close-up diet from d 21 before expected parturition through parturition (CU)] and prepartum monensin supplementation [0 g/t (control, CON) or 24.2 g/t (MON); Rumensin; Elanco Animal Health, Greenfield, IN]. Lactation diets before and after the dry period contained monensin at 15.4 g/t. During the close-up period, cows fed CU had greater DM and NEL intakes than cows fed CE. Calf BW at birth tended to be greater for cows fed CU than for those fed CE but was not affected by MON supplementation. Diet did not affect calving difficulty score, but cows supplemented with MON had an increased calving difficulty score. We found a tendency for a MON × parity interaction for colostral IgG concentration, such that multiparous MON cows tended to have lower IgG concentration than CON cows, but colostral IgG concentration for primiparous MON and CON cows did not differ. Postpartum milk yield did not differ between diets but tended to be greater for cows supplemented with MON. Milk fat and lactose content were greater for cows fed CU than for those fed CE, and lactose content and yield were increased for cows supplemented with MON. Solids-corrected and fat-corrected milk yields were increased by MON supplementation, but were not affected by diet. Overall means for postpartum DMI did not differ by diet or MON supplementation. The CU diet decreased the concentration of nonesterified fatty acids during the close-up period but increased it postpartum. Neither diet nor monensin affected β-hydroxybutyrate or liver composition. Overall, postpartum productive performance differed little between prepartum dietary strategies, but cows fed MON had greater energy-corrected milk production. In herds fed monensin during lactation, monensin should also be fed during the dry period.  相似文献

The effect of induced parturition and estradiol on feed intake, liver triglyceride, plasma metabolites, and milk yield was evaluated in fifty-six Holstein cows and heifers. Cows were assigned to treatments on d 260 of gestation and were on trial until d 10 postpartum for measurement of dry matter intake (DMI), plasma metabolites, and liver triglyceride and until d 31 postpartum to measure milk yield. Fourteen animals per group (9 cows and 5 heifers) received either a placebo, 1 mg of fenprostalene, 50 mg of estradiol-17 beta benzoate, or both on d 276 of gestation. Cows that received fenprostalene consumed more dry matter (DM) for the last 8 d prepartum than did cows that did not receive fenprostalene (9.6 kg/d vs. 8.5 kg/d, respectively) but consumed less DM for the first 10 d postpartum (10.9 kg/d vs. 13.1 kg/d, respectively). Cows injected with estradiol-17 beta benzoate tended to consume less DM postpartum than did cows not injected with estradiol-17 beta benzoate (11.3 kg/d vs. 12.7 kg/d, respectively). There was no effect of treatment on milk yield; however, a fenprostalene by day interaction resulted from lower milk yield on d 3, 4, 5, 7, and 10 relative to calving in cows that received fenprostalene. Administration of fenprostalene resulted in a delay in the peak plasma nonesterified fatty acid (NEFA) concentration until 2 d after calving. Plasma glucose concentrations were greatest 1 d prior to calving for cows that received fenprostalene, whereas plasma glucose concentrations peaked on the day of calving for cows that did not receive fenprostalene. Liver triglyceride increased over time; however, there was no effect of treatment on liver triglyceride. Calving induction improved DMI for the last 8 d prepartum, but a concomitant decrease in liver triglyceride after calving did not result. Estradiol-17 beta benzoate had no effect on plasma metabolites or liver triglyceride, indicating that the physiological rise in estradiol prior to calving does not have a primary role in lipolysis or hepatic fatty acid metabolism in the dairy cow.  相似文献

Objectives were to evaluate the effects of a low dose of bovine somatotropin (bST) injected prepartum and postpartum on body condition score (BCS), body weight (BW), and milk yield (MY) in cows as well as somatotropin insulin, insulin-like growth factor-I (IGF-I), glucose, and nonesterified fatty acids (NEFA) in plasma. Holstein cows nearing second or later parities were assigned randomly to control (CON = 98) or bST-treated (TRT = 95) groups. Biweekly injections of bST began 21 +/- 3 d before expected calving and continued through 42 d postpartum (CON vs. TRT; 0 vs. 10.2 mg of bST/ d). From 42 to 100 d postpartum, no cows received bST. During yr 1, somatotropin, IGF-I, insulin, NEFA, and glucose were measured in plasma samples from 82 cows. During yr 2, effects of bST on BCS and BW of 111 cows were evaluated, but no blood samples were collected. Milk yields through 100 d of all 193 cows were analyzed. Prepartum treatment with bST resulted in greater prepartum plasma concentrations of somatotropin, insulin, and numerically greater NEFA but did not affect glucose or IGF-I. Postpartum bST increased mean plasma concentrations of somatotropin and NEFA, but not INS, IGF-I, or glucose. Mean BCS of cows did not differ prepartum, around parturition, or postpartum. Although mean BW did not differ prepartum or around calving, cows receiving bST maintained greater BW postpartum. Cows receiving bST tended to have higher MY (6.6%) in the first 60 d of lactation, but differences did not persist through 100 d, including approximately 40 d when no cows received bST. Number of cows that were culled due to health (CON = 3 vs. TRT = 2) or died (CON = 3 vs. TRT = 1) were not affected by treatment. Low doses of bST in the transition period resulted in higher postpartum BW, quicker recovery of body condition during lactation, and significantly more milk during treatment.  相似文献

Transition heifers face multiple stressors during the periparturient period, including first exposure to milking, that may adversely impact dry matter intake (DMI), reduce milk production, compromise immune function, and increase susceptibility to disease. It was hypothesized that reducing the combined stressors experienced at calving would improve the periparturient performance, health, and well-being of heifers. The objective of this study was to determine the effect of initiating the milking procedure 3 wk before expected calving on production, DMI, body weight, energy balance, udder health, calving traits, and health status, as indicated by plasma acute phase protein concentrations. Twenty-two primigravid heifers, blocked by expected calving date, were assigned randomly either to a prepartum milking (PM) group or control group. The PM heifers were milked twice daily beginning at 21 d before expected calving, and control heifers were not milked until after calving. All heifers had access to the same precalving and post-calving diets. Results indicated that PM heifers produced more milk during the first 2 wk after calving and had greater DMI as a percentage of body weight during the first month after calving than did control heifers, although energy balance was more negative for PM heifers. The PM heifers had reduced somatic cell counts through the first month after calving and lower average somatic cell scores during lactation despite having more quarters with mastitis infection at calving. The PM heifers had less udder edema at the third milking postcalving, and had reduced concentrations of haptoglobin in blood sooner than did control heifers. These results indicate that prepartum milking is an alternative management practice that has beneficial effects on the production, health, and well-being of first-lactation cows.  相似文献

Roles of the lactogenic hormones prolactin and placental lactogen in mammary development in ruminants were reviewed. In contrast with other ruminants, failure to detect lactogenic activity in the serum of pregnant cows (in excess of that attributed to prolactin) suggests that placental lactogen may have little direct effect on mammary growth or lactogenesis. However, replacement and ablation experiments using ergocryptine provide definitive evidence that increased periparturient secretion of prolactin is necessary for maximal milk production in cattle. Quantitative microscopy indicates a relative failure of mammary cells in cows with inhibited secretion of prolactin to differentiate structurally. Prolactin induces synthesis and secretion of alpha-lactalbumin in prepartum bovine mammary tissue. Temporary disruption of mammary microtubules immediately prepartum in pregnant heifers reduced subsequent milk production, biosynthetic capacity, and cellular differentiation. For maximal milk production, mammary secretory cells apparently must respond to lactogenic hormone stimulation during the immediate periparturient period. Colchicine may desensitize the mammary epithelium to prolactin action. Membrane binding of radiolabeled human growth hormone to ruminant mammary gland provides a measure of lactogenic hormone binding sites. Specific binding to 600 micrograms of mammary membrane protein was 296% greater in lactating, compared with nonlactating, pregnant (65 days of gestation) ewes. Binding capacity (fmol/mg membrane protein) averaged 275 +/- 57 in mammary membranes from nonlactating, pregnant ewes (100 days gestation, n = 2) and 2,325 +/- 521 in mammary membranes from lactating ewes (n = 6, 14 to 21 days postpartum). Greater understanding of hormonal regulation of the ruminant mammary gland likely will result in development of techniques to produce milk more efficiently and perhaps capability to evaluate production potential of young animals.  相似文献
