无菌钝顶螺旋藻单细胞的制备和再生   总被引:7,自引:1,他引:6  
用含有0.75mol/LNaCl的Zarrouk培养基洗去藻丝体表面的角质鞘,可获得大量有活性并能再生的单细胞,再生率为28.6%。用0.4%的次氯酸钠溶液消毒藻丝体5分钟,恢复培养后再置于含有终浓度均为150μg/μl的卡那霉素、庆大霉素和新霉素Zarrouk培养基中35℃48h杀菌,最后用含1.5mol/LNaCl的Zarrouk培养基对藻丝体洗壁处理1min,可得无菌螺旋藻品系。  相似文献

螺旋藻对六种抗生素的敏感性研究   总被引:5,自引:0,他引:5  
通过6种基因工程中常用抗生素对螺旋藻生长抑制的研究发现,钝顶螺旋藻S6和极大螺旋藻SM对卡那霉素和新霉素不敏感,10-700μg/ml的卡那霉素和10-300μg/ml新霉素仍不能抑制其生长;S6和SM对庆大霉素的敏感性存在较大差异,液体培养中的抑制浓度分别是300μg/ml和50μg/ml;S6和SM品系对氨苄青霉素和链霉素较敏感,固体培养中5.0-50.0μg/ml和50μg/ml;S6和SM品系对氨苄青霉素和链霉素较敏感,固体培养中5.0-50.0μg/ml的氨苄青霉素对它们有致死作用;液体培养中链霉素的的抑制浓度是5.0μg/ml,固体培养中链霉素的致死浓度是50μg/ml;S6和SM对氯霉素最敏感,液体培养时0.1μg/ml的氯霉素可抑制螺旋藻的生长,固体平板致死浓度是1.0μg/ml。应用电转化法将带有CAT基因片段的同源重组质粒导入螺旋藻细胞,用含有20μg/ml氯霉素Zarrouk固体平板筛选出了抗性转化子,研究结果表明,氯霉素是螺放藻转化系统中理想的抗性选择试剂。  相似文献

建立栀子金花丸中黄芩苷的含量测定方法。方法采用未涂层弹性融硅石英毛细管柱(60cm×75μm,有效长度52cm),以40mmol·L^-1硼砂溶液(pH9.0)为运行缓冲液;分离电压为12kV;重力进样10s(高度15cm)l检测波长为280nm;以对硝基苯甲酸为内标。结果黄芩苷质量浓度在7.5~52.5μg.mL—1范围内具有良好的线性关系(r=O.9996),黄芩苷平均回收率为96.7%,RSD唐为1.62%(n=6)。结论该方法专属性强,结果准确可靠、重现性好,可用于栀子金花丸的质量控制。  相似文献

目的建立快速测定齐墩果酸片含量的方法。方法采用毛细管电泳法,未涂层弹性石英毛细管(75μm×50cm,有效长度40cm);压力进样,压力:3.0kPa;进样时间:5s;分离电压:25kV;柱温:25℃t检测波长:200nm;运行缓冲液:含5%(体积分数)甲醇的20mmol·L-1硼砂溶液(pH9.4)I运行时间:10min。结果齐墩果酸的线性范围为2.58~660μg·mL-1(r=0.9997),平均回收率为99.47%,RSD为1.28%(n=6)。结论方法准确、快速、简便,适于齐墩果酸片的含量测定。  相似文献

目的;建立凝胶色谱法测定头孢唑林钠的聚合物方法。方法;采用葡聚糖凝胶Sephadex G-10柱(16ram×33cm),流动相:A:0.01mol/L磷酸盐缓冲液(pH7.0),B:0.01%十二烷基硫酸钠溶液。流速为1mol/min;检测波长为254nm;进样量1200μL。结果:头孢唑林钠在6.696—29.54mg/mL范围内,浓度与聚合物的峰面积呈良好线性关系(r=0.9986),可精确对高分子杂质峰进行定量。结论:该方法简便,准确。重现性好。  相似文献

采用恒初始应变速率法对工业用铬青铜QCr0.5进行了电致压缩超塑性试验研究,考察了电场强度对流变应力及超塑压缩变形后QCr0.5显微组织的影响。结果表明,QCr0.5在750℃,初始应变速率为1.5×10^-4S^-1,电场强度为4KV/cm情况下具有较好的超塑性,且外加电场能有效细化QCr0.5显微组织。  相似文献

目的:建立高效液相色谱法测定拉坦前列素滴眼液中苯扎铵的含量。方法:色谱柱为Chiralcel OD—R(250*4.6mm,5μm)0.025mol/L磷酸盐缓冲液(0.025mol/L磷酸二氢钠溶液用0.025mol/L磷酸调节pH值至2.5)-乙腈(55:45)为流动相:检测波长:200nm,柱温35℃,流速:0.5mL/min。结果:苯扎氯铵(C21H38C1N)在81.6~122.4ng范围内呈良好的线性关系(r=0.9999,n=5),回归方程为A:1414W-688,平均回收率为99.74%,RSD=0.57%(n=9)。结论:本法简便、灵敏、准确、重复性好,可用于拉坦前列素中的苯扎氯铵的质量控制。  相似文献

合成了壳聚糖接枝聚苯胺,并采用静电自组装法制备了壳聚糖接枝聚苯胺/葡萄糖氧化酶生物传感器。讨论了温度、pH值、离子强度以及自组装膜层数对传感器电流响应的影响。制备的葡萄糖生物传感器检测范围为0.5mmol/L~16mmol/L,电流响应时间仅为5.2s。  相似文献

利用电激法将GUS基因转入杜氏盐藻细胞内进行瞬间表达,研究了盐藻的生长状态和电激转化条件对转化率的影响,并比较了CaMV35S,Ubil,Ubil-Ω5种启动子的转化效率。结果表明,培养5d的盐藻在6kV的脉冲电压,0.05s的脉冲持续时间和210的脉冲次数下电激可获得较高的转化率,为转化最佳条件。5种启动子中,Ubil-Ω启动子的转化效率最高,适合作为外源基因在盐藻细胞中高效表达的启动子。  相似文献

In this paper, we present a new method for inserting several triangulated surfaces into an existing tetrahedral mesh generated by the meccano method. The result is a conformal mesh where each inserted surface is approximated by a set of faces of the final tetrahedral mesh. First, the tetrahedral mesh is refined around the inserted surfaces to capture their geometric features. Second, each immersed surface is approximated by a set of faces from the tetrahedral mesh. Third, following a novel approach, the nodes of the approximated surfaces are mapped to the corresponding immersed surface. Fourth, we untangle and smooth the mesh by optimizing a regularized shape distortion measure for tetrahedral elements in which we move all the nodes of the mesh, restricting the movement of the edge and surface nodes along the corresponding entity they belong to. The refining process allows approximating the immersed surface for any initial meccano tetrahedral mesh. Moreover, the proposed projection method avoids computational expensive geometric projections. Finally, the applied simultaneous untangling and smoothing process delivers a high‐quality mesh and ensures that the immersed surfaces are interpolated. Several examples are presented to assess the properties of the proposed method.  相似文献

A four-ball tester was used to evaluate the anti-wear performance of three kinds of organomolybdemun compounds in the engine oils, i. e., molybdenum dialkyldithiophosphate (MoDDP), molybdenum dialkyldithiocarbamate ( MoDTC), and sulphur and phosphorus freeorganomolybdeum (Molybdate). The results indicate that a low concentration of MoDDP doesn' t improve the anti-wear properties of the commercial engine oils, but a high concentration of MoDDP can obviously improve the anti-wear properties and the load-carrying capacity of the engine oils. MoDTC doesn' t improve the antiwear properties of the engine oils, but worsens the anti-wear properties of the oils. A flow calorimeter for enthalpy increment measurements on condensed gases is presented. A better knowledge of the properties of the liquefied natural gas is needed, and therefore a liquid loop has been designed for our flow calorimeter. The fluid loop in the calorimeter is designed in order to avoid the two-phase region, since two phases would give compositional disturbances in the measurements. The avoidance of the two-phase region is made possible by increasing the pressure of the test fluid after the measurement section, then heating the fluid at super-critical pressure past the critical point. Finally, the fluid is throttled to the low-pressure gas state at the inlet condition of the compressor that circulates the fluid. To perform the pressure increase, a new cryogenic pump has been designed. To evaluate the new equipment, measurements were taken on liquid ethane over the temperature range 146–256 K at pressure between 0.9 and 5.1 MPa.  相似文献

In the present study a high‐boron high speed steel (HSS) roll material was designed. Many expensive alloy elements have been substituted by cheap boron alloy, and high‐boron high speed steel roll has been manufactured by centrifugal casting method. The microstructures, mechanical properties and wear resistance of centrifugal casting high‐boron high speed steel roll have been investigated by optical microscopy (OM), scanning electron microscopy (SEM), and X‐ray diffraction (XRD) analysis, hardness test, impact test and wear test. The results indicated that the solidification microstructures of high‐boron high speed steel roll consisted of M2(B,C), (W,Mo)2(B,C), M3(B,C), M23(B,C)6 type borocarbides and martensite, a small amount of retained austenite. Borocarbides were continuously distributed over the grain boundary. After quenching from 1050 °C, local broken network appeared in partial borocarbides, and fine secondary borocarbide precipitated from the matrix. After tempering from 525 °C, the amount of precipitated borocarbide increased significantly. After heat treatment, the hardness of high‐boron high speed steel roll excelled 60 HRC, and its impact toughness excelled 8.0 J/cm2. The single groove steel rolling amount of high‐boron high speed steel rolls increases by 500% than that of bainite cast iron roll, when the rolls are used in K1 mill housing of bar mill.  相似文献

Zusammenfassung Bei der Gestaltung von Ernteprozessen — beispielsweise des Prozesses Mähdrusch, Körnertransport und -abnahme — als transportverbundene Fließarbeitsverfahren mit mindestens zwei verschiedenen Arbeitsmitteln gibt es einen großen Optimierungsspielraum. Solche Prozesse sind mehrstufige Bedienungsprozesse mit gemischter Anordnung der Bedienungseinrichtungen. Außerdem besitzen sie stark ausgeprägte räumliche Aspekte. Es wird vor allem gezeigt, welche Merkmale solcher Prozesse in Simulationsmodellen abgebildet sein müssen, welche Zielgrößen in Abhängigkeit von welchen Einflußgrößen unter Berücksichtigung welcher Prozeßbedingungen ermittelt werden sollten und wie der Bewertungsprozeß zur Ermittlung der gewünschten Informationen gestaltet werden muß.  相似文献

Surface characterization and microstructure studies are performed on chemical vapor deposited (CVD) tungsten coating. There is about 2 μm thickness diffusion layer of tungsten in the molybdenum substrate. The thermal shock test shows tungsten coating has good adhesion with molybdenum substrate, but the elements of oxygen and carbon in the tungsten coating have the bad affection to the adhesion. The result of high-temperature diffusion experiment is the diffusion rate from molybdenum substrate to tungsten coating is faster.  相似文献
