In the last years, there has been a growing interest in the recovery of bioactive compounds from natural sources for the development of novel functional foods.In this study, natural extracts, previously obtained by fractioned high pressure extraction from “Saco” cherry culls, were characterized in terms of anticancer activity. The product derived from CO2:EtOH (90:10, v/v) extraction after a pre-treatment of raw material with supercritical CO2 during 1 h, exhibited the highest antiproliferative activity in human HT29 colon cancer cells. In addition, when compared with doxorubicin, cherry extracts induced cell cycle arrest in a different cell cycle checkpoint.In order to obtain extracts with enhanced antiproliferative activity, the extraction process was further explored. Using highly pure CO2 and EtOH 96% the inhibition of cancer cell growth was significantly enhanced by 16-fold. In addition, the incorporation of a conventional extraction step with MetOH or EtOH:H2O (50:50, v/v) prior to fractioned process allowed to obtain cherry extracts more concentrated in antiproliferative compounds. Perillyl alcohol present in cherry extracts was pointed to be one of the major responsible for anticancer properties of cherry extracts.  相似文献

The management of agro-industrial residues is an important issue for environmental reasons and the reuse of byproducts represents a good alternative, especially if it is conjugated with green technologies and the production of valuable products. Portuguese elderberry pomace is rich in anthocyanins with therapeutic properties that confer to this byproduct potential to be applied in the food and pharmaceutical industries. Fractionated high pressure extractions from elderberry pomace were performed using supercritical CO2 extraction, followed by enhanced solvent extraction (ESE) with diverse CO2/ethanol/H2O solvent mixtures (0-90%, 0.5-100%, 0-95%, v/v/v), at 313 K and 21 MPa, in order to obtain anthocyanin-rich fractions. The ESE solvent mixtures had a substantial effect on extracts yield and composition. The maximum extraction yield (24.2%), total phenolic compounds (15.8% gallic acid equivalents), total flavonoids (8.9% epicatechin equivalents), total anthocyanins (15.0% cyanidin-3-glucoside equivalents) and antioxidant activity (IC50 of 21 μg) achieved highlight the great potential of elderberry pomace for valuable applications.  相似文献

The possibility of using the leaves of Cordia verbenacea as a new source of natural antioxidant compounds was investigated. In the present work, extracts from C. verbenacea were obtained using different extraction methods: supercritical fluid extraction (SFE), Soxhlet (SE), hydrodistillation and maceration, with the objective to evaluate the methods in terms of yield and antioxidant potential. The high-pressure technique was applied using pure CO2 and CO2 with co-solvent at different temperatures and pressures (30, 40 and 50 °C and 100, 200, and 300 bar). Organic solvents with different polarities were used to obtain extracts by low-pressure extraction processes. The extracts were evaluated according to their antioxidant activity using total phenolic content, scavenging abilities on DPPH radical, total antioxidant activities (ABTS•+), superoxide anion radical-scavenging (O2) and protection against lipid peroxidation in vitro (LPO). Ethyl acetate fraction obtained by maceration and extract isolated by SE using 25% aqueous mixture of ethanol possessed the highest scavenger activity against DPPH radical (IC50 = 9.2 ± 0.4 μg/ml, IC50 = 27.4 ± 0.1 μg/ml, respectively). The SFE with 8% ethanol as a co-solvent produced extracts with distinguished increase in the antioxidant activity. The Soxhlet extract with ethyl acetate exhibited a strong reduction of lipid peroxidation (IC50 = 209 ± 3 μg/ml) value comparable to the standard rutin (IC50 = 203 ± 2 μg/ml). The results indicate that extracts of C. verbenacea have important potential as a source of bioactive compounds with antioxidant activity.  相似文献

In this work the supercritical fluid extraction (SFE) with carbon dioxide (CO2) and with ethyl acetate (EtAc) and ethanol (EtOH) as co-solvents was applied to obtain the phenolic fraction from guava seeds (Psidium guajava L.). The extraction was explored at various operating conditions, using 10, 20 and 30 MPa and 40, 50 and 60 °C. The use of EtAc and EtOH as co-solvents in SFE was also studied. The supercritical process was compared with traditional techniques such as Soxhlet extraction using EtAc and EtOH as solvents. The quality of the different extracts, obtained using SFE and Soxhlet methods and different solvents, was evaluated through the antioxidant activity, obtained by the collection methods of scavenging DPPH and bleaching of β-carotene, and also through the total phenolic content (TPC) of the samples, by the Folin-Ciocalteu method. The antioxidant potential indicates the use of ethanol as co-solvent as the best modifier in SFE, used in concentration of 10% (w/w) at 50 °C and 30 MPa. The quality of the extracts obtained by SFE with EtOH varied with the operating conditions of temperature and pressure, with higher values obtained at 10 and 20 MPa for TPC results and also antioxidant methods. The process yield of the phenolic fraction was also evaluated for all the extraction procedures studied (SFE and Soxhlet), with results varying from 0.380 to 1.738% (w/w).  相似文献

Pandan (Pandanus amaryllifolius Roxb.) leaf is a source of natural flavoring widely used in South-east Asia. The major compound contributing to the characteristic flavor of Pandan is 2-acetyl-1-pyrroline (2AP). This highly volatile compound also contributes significantly to the flavor of aromatic rice such as basmati and jasmine rice. As the consumer requirement for use of natural flavors, extraction of components from natural sources has been sought. In this study, supercritical carbon dioxide (SC-CO2) and solvent extraction of components from Pandan leaves were performed. Experimental parameters included particle size and drying method (oven and freeze drying). Results indicated that the initial value of moisture content and particle size of Pandan leaves had the greatest effect on the total yield and 2AP concentration of the extracts. Almost 80% of water in Pandan leaves can be removed by drying. Yields of supercritical extracts were 10 times lower when compared to the hexane extracts. The total yield of extracts was increased up to 50% with decreasing particle size of Pandan leaves. Extraction of coarsely ground freeze-dried Pandan leaves by SC-CO2 obtained the highest yield (0.88 ± 0.06%) followed by oven dried (0.38 ± 0.09%) and fresh leaves (0.34 ± 0.01%). The 2AP was identified by GC-MS and analyzed by GC-FID. Supercritical and hexane extracts of pre-treated Pandan leaves were found to have a small quantity of 2AP ranging between 0.04 ± 0.01 and 0.45 ± 0.01 ppm. Grinding pre-treatment was the best method for both SC-CO2 and hexane extractions while the freeze drying method was the best for SC-CO2.  相似文献

Extraction of the fresh flowers of Michelia champaca L. with liquid CO2 provided a floral extract in 1.0 ± 0.04 wt% yields. The extract so obtained contains far less waxes and is organoleptically very superior. Similarly extraction with pentane gave the so-called ‘Concrete’ in 1.58 ± 0.06 wt%. While the concrete contains co-extracted floral waxes that make it unsuitable for blending with other perfumes, direct extraction with CO2 is an expensive process mainly due to low bulk density of flowers and their availability during short flowering season. On the other hand, fractionation of the concrete with liquid CO2 to separate the waxy components has provided solvent and almost wax free fractions. The duration of extractive fractionation has been optimized for selective extraction with liquid CO2 at 62 bar. These liquid CO2 fractions of concrete and liquid CO2 extract of flowers were analyzed by GC and GC/MS and their composition compared with that of concrete and partially de-waxed absolute obtained in the conventional way. The major fragrance compounds enriched in the direct liquid CO2 extract were methyl benzoate (11.5 ± 0.8%), phenyl ethyl alcohol (5.0 ± 0.6%), phenyl acetonitrile (10.4 ± 1.1%), indole (1.2 ± 0.3%), methyl anthranilate (1.3 ± 0.5%), E-β-ionone (1.5 ± 0.4%), and Z-methyl jasmonoate (1.0 ± 0.3%). The liquid CO2 fractionation of concrete is a practical process and the first fraction is comparable with direct liquid CO2 flower extract in terms of composition of the major compounds.  相似文献

Instant active dry baker's yeast is a well-known product widely used for leavening of bread, produced by fermentation, and usually dried by hot air to 94-96% dry matter content. Multi-stage fluidized bed drying process is a commercial effective method for yeast drying. In this work, optimum operating parameters of an industrial continuous fluidized bed dryer for the production of instant active dry yeast were investigated. The dryer contained four zones separated with moving weirs. The operating conditions such as temperature, loading rate of compressed yeast granules, and hot air humidity had direct effects on both yeast activity and viability. The most important factors that affected the quality of the product were loading rate and the operational temperature in each zone on the bed. Optimization was performed for three loading rates of the feed to the dryer, using response surface methodology for the experimental design. The most significant factor was shown to be the loading rate with mean fermentation activity values of 620, 652, and 646 cm3 CO2/h for 300, 350, and 400 kg/h loading rates, respectively. The data analysis resulted in an optimal operating point at a loading rate of 350 kg/h and temperatures of zones 1, 2, 3, and 4 controlled at 33, 31, 31, and 29 °C, respectively. The best activity value was predicted as 668 ± 18 cm3 CO2/h, and confirmation experiments resulted in 660 ± 10 cm3 CO2/h. At the same operating point, the average viability of the cells was predicted as 74.8 ± 3.7% and confirmed as 76.4 ± 0.6%. Compared with the normal operating conditions at the plant, the optimization resulted in more than 12% and 27% improvement in the yeast activity and viability, respectively.  相似文献

The objective of this study was to obtain orange (Citrus sinensis L. Osbeck) pomace extract using supercritical fluid extraction (SFE) with CO2 and with CO2 and co-solvent. In order to evaluate the high pressure method in terms of process yield, extract composition and biological activity, low pressure methods were also applied to obtain orange extracts, such as ultrasound (UE) and soxhlet (SOX), with different organic solvents, and hydrodistillation (HD). The SFE conditions were temperatures of 313.15 K and 323.15 K and pressures from 100 to 300 bar. The SFE kinetics and mathematical modeling of the overall extraction curves (OEC) were also investigated. The antioxidant potential of the extracts was evaluated by the DPPH method, by the Folin-Ciocalteau method and by the β-carotene/linoleic acid bleaching method. The antimicrobial activity of the extracts was also studied. The main compounds identified were l-limonene, palmitic and oleic acids, n-butyl benzenesulfonamide and β-sitosterol.  相似文献

Leaves of Arrabidaea chica (Humb. Bonpl.) Verlot are rich in anthocyanins and have been used as a medicinal plant in the Amazon region. In order to obtain different extracts from this plant, a sequential extraction in fixed bed was carried out at 40 °C and 300 bar, using supercritical carbon dioxide (scCO2) in a first step, and a mixture containing CO2/ethanol/water at mass ratios of approximately 80/20/0, 80/14/6 and 80/10/10 in a second extraction step. The residue from the second step was extracted with water at 40 °C and atmospheric pressure. Ethanolic, aqueous and hydroalcoholic (70:30, v:v) extracts were also obtained by conventional extraction methods at atmospheric pressure. All extracts were analyzed for global extraction yield, total phenolic content, total flavonoids, and carajurin content. High performance liquid chromatography (HPLC) was used both to quantify carajurin, which is the main anthocyanin component of A. chica, and to monitor qualitatively two other anthocyanin pigments found in that plant. The extraction yield in the first step was only 0.65% using pure scCO2, but this extraction was highly selective to extract carajurin from the three main anthocyanins. The accumulated global yield of the two steps ranged from 3% when the solvent ratio (80/20/0) was used in the second step to about 50% when 6 or 10% water was used, showing the highest yield when the extraction was done with water. The highest contents of total phenolic compounds (178 mg GAE/g extract) and total flavonoids (373 mg EC/g extract) were found in the process performed with the extraction mixture (80/20/0), and the highest carajurin content was obtained in the ethanolic extracts.  相似文献

The hydrothermal treatment of two different wood biomass samples such as cherry (hard wood) and cypress (soft wood), whose composition is different i.e. lignin, cellulose and hemicellulose were performed at 280 °C for 15 min with aq. K2CO3 with different concentrations (0–1 M). The soft wood biomass contains higher lignin content than hard wood biomass. The cellulose rich cherry wood biomass produced higher proportion of acetic acid than cypress. The lignin rich cypress produced the hydrocarbons with major portion of phenolic hydrocarbons and derivatives than cherry. The total oil yields from both cherry and cypress wood biomass produced 50 wt% of liquid hydrocarbons at 280 °C for 15 min with 0.5 M K2CO3 solution. The volatility distribution of liquid hydrocarbons showed the characteristic features of soft and hard wood biomasses.  相似文献

Brazilian Ginseng extracts of two species, Pfaffia paniculata and Pfaffia glomerata, were obtained by supercritical fluid extraction (SFE) with CO2 and by low-pressure solvent extraction (LPSE) with methanol, hexane and ethanol. The SFE assays were conducted at pressures of 100, 200 and 300 bar, and temperatures of 30 and 50 °C. The qualitative chemical compositions of the extracts were determined by thin layer chromatography (TLC). One of the active principles of interest from P. glomerata extract, β-ecdysone, was identified and quantified by HPLC. The antioxidant activities of Brazilian Ginseng extracts were determined by the coupled reaction of linolenic acid and β-carotene. For P. paniculata, the highest SFE yield was obtained at 200 bar/50 °C (0.22%, dry basis—d.b.), while the best extraction condition for P. glomerata was obtained at 200 bar/30 °C (0.18%, d.b.). The higher extract yields obtained by LPSE were 2.0% and 5.8% (w/w, d.b.) for P. paniculata and P. glomerata, respectively, both obtained with methanol as extraction solvent. From the overall extraction curve of P. glomerata, it was possible to obtain the kinetic parameters of extraction; the duration of the CER (constant extraction rate) period was determined as 134 min. The TLC plates showed the possible presence of flavonoids in the ethanolic extract for both Pfaffia species. The antioxidant activity analysis detected that LPSE extracts had higher activity than SFE extracts.  相似文献

A procedure for the recovery of aromatic extracts from raw grape-spirit (alcohol strength 73 vol.%) by means of a countercurrent supercritical fluid extraction (CC-SFE) on a pilot plant scale is presented. The effect of the solvent-to-feed ratio at 7, 8, 10, and 20 kgCO2/kgFeed on the extraction of flavor compounds, operating at constant pressure of 100 bar and extraction temperature of 50 °C have been studied. Increasing the solvent-to-feed ratio from 7 to 20 kgCO2/kgFeed, the total volatile compounds yield decreases from 64 to 5% and that of ethanol from 85 to 49%. The highest concentration of volatile compounds has been obtained at 7 and 8 kgCO2/kgFeed solvent-to-feed ratio. Mixtures containing grape-spirit (alcohol strength 40 vol.%) added with 30% (v/v) of different extracts have been tested and judged by a panel. Sensory evaluation indicated as preferred the mixtures added with the extracts obtained at lowest solvent-to-feed ratio.  相似文献

Ni modified K2CO3/MoS2 catalyst was prepared and the performance of higher alcohol synthesis catalyst was investigated under the conditions: T = 280–340 °C, H2/CO (molar radio) = 2.0, GHSV = 3000 h 1, and P = 10.0 MPa. Compared with conventional K2CO3/MoS2 catalyst, Ni/K2CO3/MoS2 catalyst showed higher activity and higher selectivity to C2+OH. The optimum temperature range was 320–340 °C and the maximum space-time yield (STY) of alcohol 0.30 g/ml h was obtained at 320 °C. The selectivity to hydrocarbons over Ni/K2CO3/MoS2 was higher, however, it was close to that of K2CO3/MoS2 catalyst as the temperature increased. The results indicated that nickel was an efficient promoter to improve the activity and selectivity of K2CO3/MoS2 catalyst.  相似文献

The olive leaf phenolic composition of the Greek cultivars koroneiki, megaritiki and kalamon was determined using LC/MS. Furthermore, the antioxidant activity of olive leaf extracts from the above three cultivars, using solvents of increasing polarity (petroleum ether, dichloromethane, methanol and methanol/water: 60/40) was evaluated using the stable free radical diphenylpicrylhydrazyl (DPPH) test. Furthermore the oxidative stability index (OSI) was compared to that of the synthetic antioxidant TBHQ and commercial oleoresin (rosemary extract). The ability of phenolic compounds to inhibit the lipoxygenase (LOX) activity was also investigated. The ten main components determined in the olive tree leaf extracts for the cultivars koroneiki and kalamon were: secologanoside, dimethyloleuropein, oleuropein diglucoside, luteolin-7-O-glucoside, rutin, oleuropein, oleuroside, quercetin, ligstroside and verbascoside. Respective compounds for the cultivar megaritiki were: secologanoside, dimethyloleuropein, oleuropein diglucoside, luteolin7-O-glucoside, oleuropein, oleuroside, quercetin and ligstroside. In all three cultivars, oleuropein represented the main phenolic component. The solvent polarity influenced the total amount of the phenolic compounds determined. When methanol/water (60/40) was used, as solvent, more phenolic compounds were determined. The total amounts of phenols determined in the extracts, obtained by successive extractions using the above solvents, were 6,094, 5,579 and 6,196 mg/kg (mg gallic acid/kg dried olive leaves) for the cultivars megaritiki, kalamon and koroneiki, respectively. Among all extracts, methanol/water extracts exhibited the highest antioxidant activity as shown through the application of the DPPH and OSI methods. The OSI antioxidant activity followed the sequence: synthetic antioxidant TBHQ > commercial oleoresin > olive tree leaf extracts > control. Likewise, methanol/water olive leaf extracts significantly inhibited soybean lipoxygenase, although some small differences in the activity among the olive leaf extracts of the different cultivars were observed. The solvent polarity as well as the amount of the extract influenced the inhibitory activity. A positive correlation was shown between the antioxidant activity of leaf extracts and the total phenol content.  相似文献

In this work we designed and built a homemade supercritical fluid extraction (HM-SFE) system, in which pure CO2 and CO2 with co-solvents were used. The HM-SFE was made by means of thermal dilatation-contraction (TDC). This HM-SFE system was used for obtaining guava (Psidium guajava L.) seed oil, using supercritical CO2 adding ethanol as co-solvent (CO2 SC/EtOH), extractions were performed at 313 K and different pressures (10, 20 and 30 MPa), each one in four stages of 30 min, the extract with higher yield was subjected to transesterification and high-resolution gas chromatography (HRGC) analysis. The highest extraction yield was obtained at 30 MPa (17.30% w/w), this yield was higher than one observed in a previous work using SC-CO2, and near to the one obtained by Soxhlet extraction (20.2% w/w). HRGC enabled the identification of components of the derivatized extract as methyl esters of palmitic, oleic, linoleic, and stearic fatty acids. The results obtained with HM-SFE system was compared with a commercial SFE system, obtained very similar results. In this work was possible to construct a low cost and simple manner HM-SFE system which was employed for obtaining guava seed oil, using CO2 SC/EtOH.  相似文献

The extraction of biologically active compounds from eggplant pulp by modified supercritical CO2 extraction was investigated. Response surface methodology was used to optimize the extraction of phenolic compounds and antioxidant capacity by assessing the pressure, temperature, and cosolvent percentage. The results demonstrated that the maximum phenolic content (3,530.79 mg CAE/100 g extract) and antioxidant capacity (4,593.22 μmol TE/g extract) were observed at 56.8°C, 280 bar, and 1.22% of ethanol and were higher than those obtained by conventional solvent extraction. Four phenolic acids were identified in the supercritical extracts and not in the conventional extracts using HPLC-DAD analysis, suggesting that modified supercritical CO2 extraction is more efficient and selective.  相似文献

The extraction of polyphenol compounds from jatoba (Hymenaea courbaril L. var stilbocarpa) bark using supercritical fluid extraction (SFE) with CO2 and cosolvents has been investigated. Among the solvent systems studied, SFE using CO2 and water (9:1, v/v), at 323 K and 35 MPa, presented the best results, with extract yield of 24%, and with high antioxidant activity (IC50 of 0.2 mg/cm3). This solvent system was used to determine global yield isotherms, which were built at 323 and 333 K, and 15, 25, and 35 MPa, using a second lot of jatoba. The highest yield was 11.5% at 15 MPa and 323 K, with maximum total phenolic compounds (TPC) of 335.00 mg TAE/g extract (d.b.) and total tannins content of 1.8 g/100 g raw material. A kinetic experiment was performed using optimized conditions, yielding 18% extract, and the kinetic parameters were used to scale-up the process from laboratory to pilot scale. Chemical analyses showed high content of phenolic compounds in the extracts of jatoba bark mostly due to the presence of procyanidins.  相似文献

In this study extraction of polyphenols and flavonoids from cultivated hybrid Sideritis scardica × Sideritis syriaca, known for its rich content of phenolics and flavonoids with antioxidant activity, was investigated. Extractions have been done by ethanol and water-ethanol, respectively. High equilibrium values of the extracted species were obtained—17.55 mg/(g solid) total phenolics and 5.7 mg/(g solid) total flavonoids with ethanol as solvent. The influence of the solvent on the total yield and the content of biologically active compounds were studied. Maximum polyphenolics and flavonoids extraction was observed for water-ethanol solvent ratio 20/80. Increase of the content of ethanol in the solvents led to lower total yield of extracts but higher percentage of polyphenolics. The extraction kinetics showed that 90% of the phenolic compounds were extracted during the first 2.5 h. The experimental kinetics was described by a constant effective diffusion coefficient De = 1.5 × 10−12 m2/s in the solid, accounting for the actual particle size distribution.  相似文献

Microalgae represent diverse branch of microorganism that can produce a wide range of unique functional ingredients that can be used in food, cosmetics, pharmaceuticals, and energy. Among them, Haematococcus pluvialis is known for accumulating the highest levels of a potent natural antioxidant, astaxanthin, which has demonstrated positive health effects. Therefore, the aim of numerous studies has been to develop novel and efficient extraction techniques to produce high-quality (purity and antioxidant activity) extracts, while complying with the Green Chemistry Principles. Supercritical CO2 (scCO2) emerges as an alternative to organic solvents because of its high selectivity and bioactivity-preserving qualities. Nevertheless, astaxanthin is a large molecule with low solubility in scCO2 that usually requires long extractions at high pressures. Ethanol has been used as co-solvent to increase astaxanthin solubility in scCO2. In this work, a Box–Behnken experimental design was used to study the effects of operating pressure (20–35 MPa), temperature (40–70 °C), and ethanol content in scCO2 (0–13%, w/w) on the yield, astaxanthin content, and antioxidant activity of H. pluvialis extract. Results showed that ethanol content in CO2 has a more significant effect on all responses than pressure and temperature. These results lead us to investigate the effect of a further increase in ethanol content, up to the region of gas-expanded liquids. We studied the effects of temperature (30–60 °C) and ethanol content (50–70%, w/w) at a fixed pressure (7 MPa) on the same response variables using CO2-expanded ethanol (CXE). Results showed that temperature and ethanol content had a significant influence on astaxanthin yield and antioxidant activity. Also, the overall responses of CXE surpassed scCO2 extractions to match conventional extraction with acetone, maintaining high quality extracts, thus validating the use of this new type of green technology for extraction of high-value compounds.  相似文献

The aims of this work were to study the chemical composition of the essential oil from the leaves of Pereskia aculeata and to evaluate some biological activities of three leaf extracts. The phenolic content, antioxidant activity, and in vitro antimicrobial and antifungal activities were determined. The methanol extract showed antioxidant activity (EC50 7.09 mg/mL) and high polyphenols content (15.04 ± 0.31 mg gallic acid equivalents (GAE)/g). The petroleum ether extract exhibited potent antibacterial activity against Escherichia coli, whereas the chloroform extract showed inhibitory activity against Bacillus cereus and Staphylococcus aureus. The petroleum ether and methanol extracts were more effective in inhibiting the growth of Aspergillus versicolor. The possible cytotoxicity of extracts on neuroblastoma SH-SY5Y cancer cell line and the influence on adenylate cyclase (ADCY) expression was also studied. P. aculeata chloroform extract showed antiproliferative activity with an IC50 value of 262.83 µg/mL. Treatments of SH-SY5Y neuroblastoma cells with 100 µg/mL of methanol extract significantly reduced ADCY1 expression.  相似文献
