The influence of hydroxamic acids (Hx), plant secondary metabolites associated with aphid resistance in wheat, on the host acceptance and suitability of the aphid Sitobion avenae to the cereal aphid parasitoid Aphidius rhopalosiphi was evaluated. Aphids showed a reduction in mean relative growth rate and in body size in the wheat cultivar with higher Hx level. Reduction in aphid size was related to a decreased success in avoiding parasitoid oviposition. A minor increase in A. rhopalosiphi developmental time was observed in aphids feeding on the higher Hx cultivar. Experiments with different concentrations of DIMBOA glucoside, the main Hx in wheat, in artificial diets showed an increase in parasitoid developmental time at the highest concentration, with no change in other performance variables. The evidence is discussed in relation to the compatible utilization of host-plant resistance and biological control in integrated pest management.  相似文献

We hypothesized that aphids after previous exposure to hydroxamic acids (Hx), a family of secondary plant compounds deleterious to aphids, are able to reduce their subsequent exposure to them. This hypothesis was tested by evaluating the time to produce salivation into a sieve element (SSE) by the aphid Sitobion fragariae on seedlings of two wheat cultivars of Triticum aestivum differing in their concentration of Hx. The total time to produce a first SSE was significantly longer in the high-Hx cultivar; however, the subsequent, second SSE (first SSE after interruption of probing) in this cultivar was significantly reduced, reaching the level observed in the low-Hx plants. Therefore, a strategy to reduce the exposure to secondary compounds was observed only in the second SSE in high-Hx plants. When the experimental plant was replaced by a new unattacked plant after the first SSE, aphids did not change the behavior described, thus excluding an aphid-induced plant susceptibility. The number of cell punctures and accumulated duration was not affected by previous exposure to Hx, either in low or high Hx cultivars. Total time and pathway time but not cell punctures, seem to be the variables affected by previous exposure to Hx.  相似文献

2,4-Dihydroxy-7-methoxy-1,4-benzoxazin-3-one (DIMBOA), a hydroxamic acid involved in the resistance of cereals to aphids, was administered to adult individuals of the aphid Sitobion avenae in artificial diets. Effects on the cellular metabolism were inferred from the evaluation of several organelle marker enzymes. Catalase from peroxisomes and cytochrome c oxidase from mitochondria increased their activities about twofold when aphids were fed with 2 mM DIMBOA. The role of these enzymes in the metabolizing of xenobiotics by aphids is discussed. Biochemical and cytochemical evidences for the presence of peroxisomes in aphids are reported here for the first time.  相似文献

DIMBOA (2,4-dihydroxy-7-methoxy-1,4-benzoxazin-3-one), a secondary metabolite found in cereal extracts, confers resistance in wheat to aphids. Its effect on beneficial organisms was tested on larvae of the aphid predatorEriopis connexa Germar. Larvae were fed until pupation on artificial diets to which different concentrations of DIMBOA (2–200g/g diet) were added, as well as on aphids that had been feeding on wheat seedlings with different DIMBOA levels (140–440 g/g fresh tissue). In diets, the effect of DIMBOA was greatest on survival of third-instar larvae and on the duration of the second and fourth instars. When aphids were provided as food, those that had fed on a wheat cultivar with an intermediate DIMBOA level led to a significantly longer larval duration in the predator than did those that fed on either low or high DIMBOA cultivars. Shortest predator development times were obtained with aphid prey that had fed on high DIMBOA seedlings. Differential Sensitivity of Mosquito Taxa to Vegetable Tannins   总被引:1,自引:0,他引:1  
The sensitivity of larval Culicidae to vegetable tannins was investigated in different taxa representative of the fauna from alpine hydrosystems (Aedes rusticus, Culex pipiens) and foreign noxious fauna (Aedes aegypti, A. albopictus). Bioassays reveal that tannic acid at concentrations of 0.1–6 mM is significantly more toxic for C. pipiens than for Aedes taxa, and A. aegypti is more sensitive than A. albopictus and A. rusticus. Comparison of the rank order of sensitivity among taxa with the associated levels of cytochrome P-450, esterase, and glutathione-S-transferase activities suggests that cytochrome P-450 and esterases may be involved in the detoxification of tannins. A possible involvement of these detoxifying enzymes is also revealed in vivo by the synergistic effects of S,S,S-tributyl phosphorotrithioate (esterase inhibitor) and piperonyl butoxide (P-450 inhibitor). The differential sensitivity to tannins among taxa is discussed in terms of ecological implications within mosquito communities from alpine hydrosystems, where the acquisition of tannins–detoxifying enzymatic systems may be considered as a key innovation.  相似文献

Hydroxamic acids (Hx) are natural products of Gramineae that are associated with cereal resistance to pests. We aimed at characterizing the induction of Hx accumulation in seedlings of wheat,Triticum aestivum, by short-term infestation of the cereal aphid,Rhopalosiphum padi. A load of 25 aphids increased significantly the Hx levels in the infested primary leaf in comparison with control levels. Lower loads did not increase Hx concentration. Aphid infestation lasting 16 hr did not elicit induction of Hx, even after a time-lag of 32 hr to allow the expression of any induced response. Forty-eight hours was the minimum duration of aphid infestation required to trigger Hx induction. The age of the infested tissue (the primary leaf) did not affect induction. Similar increases of Hx were found in unfolding, expanding, and totally expanded primary leaves. It was determined that the regime of nutrient supply (N-intensive nutritive solutions at low and high concentration) to wheat seedlings had no effect on the magnitude of the aphid-induced Hx (N-based secondary metabolites). Results obtained are discussed in the framework of general theories of plant defense allocation.  相似文献

DIMBOA glucoside (2-O-/gb-D-glucopyranosyl-4-hydroxy-7-meth-oxy-1,4-benzoxazin-3-one), the main hydroxamic acid (Hx) in intact wheat plants, was detected in the honey dew ofRhopalosiphum padi feeding on seedlings of six wheat cultivars that differed in their concentration of Hx, suggesting that the chemical circulates in the phloem. Neither the aglucone (DIMBOA) nor its main breakdown product were found in any of the honeydew samples. Honey dew production by aphids caged on seedlings of the wheat cultivars and DIMBOA glucoside concentrations in the honeydew followed biphasic curves when plotted against Hx concentration, suggesting passive ingestion of the chemical from the phloem at low Hx concentrations and limited ingestion due to feeding deterrency by Hx in mesophyll cells at high Hx concentrations. The presence of plant toxins such as Hx glucosides in the phloem sap, the main ingesta of aphids, and in the mesophyll cells, has major implications for plant defense, through a feeding deterrent effect during stylet penetration, and deterrency (antixenosis) along with antibiosis during feeding.  相似文献

Brassica nigra plants, characterized by high levels of sinigrin, and artificial aphid diets to which sinigrin was selectively added were used to rear the crucifer specialist, Brevicoryne brassicae. Aphids were provided as a food source to two species of polyphagous ladybird, Adalia bipunctata and Coccinella septempunctata. First instar A. bipunctata were unable to survive when fed with B. brassicae reared on B. nigra or diets containing 0.2% sinigrin, but when fed with aphids reared on diets containing 0% sinigrin, survival rates were high. By contrast, first instar C. septempunctata were able to survive when fed with aphids reared on B. nigra or artificial diets containing up to 1% sinigrin. However, the presence of sinigrin in the aphid diet decreased larval growth and increased the time necessary for larvae to reach second instar for this species of ladybird. These results indicate that the presence of sinigrin in the diet of B. brassicae makes this aphid unsuitable as a food source for A. bipunctata but not for C. septempunctata, although for this ladybird species, there appear to be costs associated with feeding on aphids that contain this secondary metabolite.  相似文献

Diaeretiella rapae (M'Intosh) (Hymenoptera: Aphidiidae) is a parasitoid of several aphid species, including the Russian wheat aphid (RWA),Diuraphis noxia (Mordvilko), and the cabbage aphid (CA).Brevicoryne brassicae (L.). The response of matedD. rapae females to odors from wheat, cabbage, and plant-host complexes was investigated using a four-choice olfactometer. Experienced parasitoids, but not inexperienced females, responded positively to odors of the wheat-RWA complex in a no-choice test. In choice tests, experienced parasitoids did not respond to odors of uninfested cabbage and wheat leaves, but did respond positively to aphid-infested plants and to aphids alone. The response ofD. rapae to the cabbage-CA complex and to CA alone was significantly greater than to the wheat-RWA complex and RWA alone, suggesting an innate odor preference for crucifer-feeding aphids.  相似文献

Chemical analysis of the volatiles released by sexual females (oviparae) of the peach aphid, Tuberocephalus momonis, identified two ubiquitous aphid sex pheromone components, (4aS,7S,7aR)-nepetalactone and (4aS,7S,7aR)-nepetalactol, in a ratio of 4 : 1. In field trials in Korea, employing traps releasing the two compounds in differing ratios, the most effective sex pheromone blend for trapping male T. momonis was found to be 85 : 15 nepetalactone–nepetalactol. Surprisingly, large numbers of presexual females (gynoparae) of this species were also collected when the catching rates were highest. In addition to T. momonis, over 20 other species of aphids were caught, particularly Myzus lythri, M. dycei, Lachnus tropicalis and M. persicae, in descending order of abundance.  相似文献

Phloem sap of wheat seedlings differing in whole leaf hydroxamic acid (Hx) concentrations was collected by cutting stylets of feeding aphids. DIMBOA-glucoside was the only Hx-related product found. Two toxic and bitter-tasting cardenolides (cardiac-active steroids) were sequestered by the brightly colored oleander aphid,Aphis nerii B. de F., from the neotropical milkweed host plantAsclepias curassavica L. After feeding on milkweed-reared aphids, the orb-web spiderZygiella x-notata (Clerck) built severely disrupted webs and attacked fewer nontoxic, control aphids, whereas the webs of spiders fed only nontoxic aphids remained intact. The regularity and size of the prey-trapping area of webs were reduced significantly in proportion to the amount of toxic aphids eaten. The effects of toxic aphids on spider web structure were mimicked by feeding spiders the bitter-tasting cardenolide digitoxin, a cardenolide with similar steroidal structure and pharmacological activity to the two aphid cardenolides. These results show that the well-known effects of psychoactive drugs on spider web structure are more than interesting behavioral assays of drag activity. Similar effects, produced by plant-derived chemicals in the spider's aphid prey, are relevant to the ecology and evolution of interactions between prey defense and predator foraging.  相似文献

UPD-glucose transferases are found in the cytosolic and microsomal fractions of the grain aphidSitobion avenae F. Gel filtration and SDSPAGE revealed that the microsomal fraction contained several forms of the enzyme. The molecular weights of the three most active fractions might be 68,000, 66,000, and 36,500. There was a negative correlation between the enzymes' activity in extracts of aphids and the concentration of DIMBOAaglucone in the winter wheat variety fed on by the aphid. A strong inhibition of the activity of the UPD-glucose transferases was observedin vitro at a concentration of DIMBOA as low as 0.01 mM. There was a greater activity of the enzymes in aphids fed on seedlings of susceptible than on moderately resistant wheat cultivars. Prolonged feeding on resistant cultivars resulted in a further reduction in the activity of the aphid's enzymes. The significance for cereal aphids of the role of their UDP-glucose tranferases in the detoxification of plant allelochemicals and adaptation to resistant varieties of cereals is discussed.  相似文献

Secondary metabolites play an important role in host plant resistance to insects, and insects, in turn, may develop mechanisms to counter plant resistance mechanisms. In this study, we investigated the toxicity of gramine to the cereal aphid Sitobion avenae and some enzymatic responses of S. avenae to this alkaloid. When S. avenae fed on an artificial diet containing gramine, mortality occurred in a dose-dependent manner. The LC50 of gramine was determined to be 1.248 mM. In response to gramine, S. avenae developed increased activities of carboxylesterase and glutathione S-transferase, two important detoxification enzymes. The activities of both enzymes were positively correlated with the concentration of dietary gramine. In addition, the activities of peroxidase and polypheolic oxidase, two important oxidoreductase enzymes in S. avenae, increased in response to gramine; however, catalase activity decreased when insects were exposed to higher levels of dietary gramine. The potential role of gramine in host plant resistance and S. avenae counter-resistance is discussed.  相似文献

The biochemical basis for the tolerance of the European corn borer,Ostrinia nubilalis, to the phototoxin-terthienyl was investigated by measuring the midgut polysubstrate monooxygenases and glutathioneS-transferase activities.-Terthienyl administered in the diet to the corn borers increased the level of cytochromeb 5, NADH-cytochromec reductase,O-demethylase, and glutathioneS-transferase activities. The induced detoxification enzyme activities should enable the corn borer to metabolize-terthienyl more efficiently and therefore render the corn borer highly tolerant to-terthienyl.  相似文献

Hydroxamic acids (Hx) produced by some cereal crops have been associated with allelopathy. However, the release of Hx to the soil by the producing plant-an essential condition for a compound to be involved in allelopathy-has not been shown. GC and HPLC analysis of roots and root exudates of wheat (Triticum aestivum L.) and rye (Secale cereale L.) cultivars, with high Hx levels in their leaves, demonstrated the presence of these compounds in the roots of all cultivars analyzed and in root exudates of rye. Moreover, bioassays employing root exudates collected from wheat and rye seedlings demonstrated that only rye exudates inhibited root growth of wild oats,Avena fatua L., a weed whose root growth is inhibited by Hx. These results suggest that rye could potentially interfere with the growth ofAvena fatua in nature and that this interference could be due to the release of Hx to the soil by way of roots.  相似文献

The bird cherry-oat aphid, Rhopalosiphum padi (L.), and the damson-hop aphid, Phorodon humuli (Schrank), migrate at the same time of year and colonize closely related Prunus spp. as primary hosts, but utilize (1R,4aS,7S,7aR)-nepetalactol and (1RS,4aR,7S,7aS)-nepetalactol, respectively, as sex pheromones. Interactions between these sex pheromones and benzaldehyde and methyl salicylate, plant volatiles common to primary hosts of both species, were investigated to assess whether they confer reproductive isolation between these species. Female autumn migrants (gynoparae) and males of these two species were caught in the field with water traps baited with their respective sex pheromones. Rhopalosiphum padi gynoparae and males also responded positively to benzaldehyde. Release of either benzaldehyde or methyl salicylate with the conspecific sex pheromone increased catches of both species of aphid. However, releasing both plant volatiles with the sex pheromone of R. padi increased catches of gynoparae and males, but reduced those with the sex pheromone of P. humuli. These results support the hypothesis that specific plant volatiles synergize responses of autumn migrating aphids to their sex pheromone. Because these interactions are species-specific, they may be important in allowing males to discriminate between conspecific sexual females (oviparae) and those of other aphid species.  相似文献

Diaeretiella rapae, a parasitoid that predominately specializes in the parasitism of Brassica-feeding aphids, attacks Lipaphis erysimi, a specialist feeding aphid of the Brassicaceae and other families in the Capparales, at a greater rate than the generalist-feeding aphid, Myzus persicae. In this study, we investigated the orientation behavior of D. rapae to the volatile chemicals produced when these two aphid species feed on turnip (Brassica rapa var rapifera). We showed no significant preference orientation behavior to either aphid/turnip complex over the other. Isothiocyanates are among the compounds emitted by plants of the Brassicaceae in response to insect feeding damage, including by aphids. We assessed parasitoid orientation behavior in response to laboratory-formulated isothiocyanates. We tested two formulations and discovered significant orientation toward 3-butenyl isothiocyanate. We also assessed plant and aphid glucosinolate content, and showed large levels of glucosinolate concentration in L. erysimi, whereas there was little change in plant content in response to aphid feeding. Our results suggest that during the process of host location, similar cues may be utilized for locating L. erysimi and M. persicae, whereas the acceptance of hosts and their suitability may involve aspects of nonvolatile aphid chemistry.  相似文献

The response of the bird cherry-oat aphid, Rhopalosiphum padi, to barley plants was investigated following exposure of the plants to root allelochemicals from the aggressive weed couch-grass, Elytrigia (Agropyron) repens. Plants were treated either with root exudates from living couch-grass plants or with previously identified couch-grass root compounds [5-hydroxyindole-3-acetic acid, DL-5-hydroxytryptophan, L-5-hydroxytryptophan hydrate, and 6-hydroxy-1,2,3,4-tetrahydro--carboline-3-carboxylic acid (carboline)] either separately or in mixtures. In choice and no-choice settling tests, aphid acceptance of barley plants was significantly reduced following treatment with root exudates, and the carboline when tested alone or in combination with the other compounds. In contrast, the other compounds without the carboline were less active in reducing aphid acceptance. In a probing bioassay, individual substances were either neutral or stimulatory to aphids, indicating that the reduced settling was probably not due to direct effects on aphids, but rather due to effects on the plant. This was confirmed in olfactometer assays, in which aphids were repelled by odors from barley plants following treatment with a mixture containing all four chemicals.  相似文献

ACQUIRED AND R-GENE-MEDIATED RESISTANCE AGAINST THE POTATO APHID IN TOMATO   总被引:4,自引:0,他引:4  
We examined the effects of three forms of host plant resistance in tomato, Lycopersicon esculentum, on the potato aphid, Macrosiphum euphorbiae. Mi-1.2, a resistance gene (R-gene) in tomato that deters aphid feeding, reduced the population growth of both potato aphid isolates tested, although it appeared to have a greater impact on isolate WU11 than on isolate WU12. The results suggest that there may be quantitative differences in virulence between these two aphid isolates. We also examined two distinct forms of acquired resistance in tomato, jasmonic acid (JA)-dependent and salicylic acid (SA)-dependent induced defenses. Exogenous foliar application of JA triggered expression of a JA-inducible proteinase inhibitor in tomato cultivars with and without Mi-1.2, although the effects of treatment on aphid performance differed between these cultivars. JA-treatment reduced aphid population growth on a susceptible tomato cultivar that lacks Mi-1.2, but did not significantly enhance or inhibit aphid control on a near-isogenic resistant tomato cultivar that carries this gene. Foliar application of an SA analog, benzothiadiazole (BTH), was used to induce SA-dependent defenses. BTH treatment reduced the population growth of both aphid isolates on a susceptible tomato cultivar, and also enhanced aphid control on a resistant cultivar. The results indicate that both SA- and JA-dependent acquired resistance in tomato have a direct negative effect on a phloem-feeding insect. Furthermore, this study demonstrates that acquired resistance and R-gene-mediated resistance can interact for enhanced suppression of insect herbivores.  相似文献
