阴离子交换树脂及气相色谱法测定腊肉中的游离脂肪酸   总被引:6,自引:0,他引:6  
建立一种测定腊肉中游离脂肪酸(FFA)组成及含量的方法,以Amberlyst-26阴离子交换树脂分离纯化腊肉中的FFA,用十五烷酸(C15:0)作内标,以BF3-CH3OH、苯、甲醇为甲酯化试剂,将得到的甲酯化产物用正己烷萃取后进行气相色谱分析,所采用的色谱柱为毛细管石英柱(FFAP型)。各脂肪酸浓度在4~32mg/mL范围内具有良好线性关系,对各次加入不同量C12:0、C14:0、C16:0、C18:0、C18:1的回收率为93.91%~95.71%,重复性实验中各脂肪酸的变异系数CV%为1.77%~3.82%,方法可靠。  相似文献

Two concurrent trials were conducted to investigate the influence of low‐protein amino acid‐supplemented diets on the performance, carcass characteristics, whole‐body composition and efficiencies of nutrient utilisation by the male broiler chicken from age 3 to 6 weeks. The first trial comprised five isoenergetic (13.0 MJ kg−1) diets containing 225 (control), 210, 190, 172 or 153 g kg−1 crude protein (CP) supplemented with essential amino acids (EAAs) to meet the minimum National Research Council recommendations. In the second trial a composite mixture of non‐essential amino acids (NEAAs) was added to the lower‐CP diets (ie 210–153 g kg−1) such that they became isoproteinous (N × 6.25) with the 225 g kg−1 control. Neither the lowering of dietary CP nor NEAA supplementation had any significant influence on weight gain or the relative weights of the various carcass cuts. However, chicks fed the lowest‐CP diets consumed more feed (P ≤ 0.05) and had poorer (P ≤ 0.05) feed conversion efficiency (FCE). NEAA supplementation enhanced FCE to the control levels. Whole‐body compositional analysis showed that lowering dietary CP increased (P ≤ 0.01) total body fat in a linear fashion (P ≤ 0.001; r = −0.72). Equalising dietary CP with the control (ie maintaining identical energy/protein ratio) by NEAA supplementation did not correct for the fat deposition. Total body protein (g kg−1) was identical with the control with or without NEAA supplementation. Dietary energy, protein retention efficiency (PRE) and protein efficiency ratio (PER) were more efficient (P ≤ 0.01) in the lower‐protein diets, while NEAA supplementation significantly (P ≤ 0.01) decreased the efficiency of N utilisation. Reducing dietary CP from 225 to 153 g kg−1 decreased N excretion in a highly significant linear fashion (P ≤ 0.001; r = 0.73). The nutritional and environmental implications of the increased body fat deposition on the one hand and the decreased N excretion on the other in the low‐protein‐fed chickens are discussed and the need to harmonise these apparently conflicting interests is emphasised. © 2000 Society of Chemical Industry  相似文献

Probiotic characteristics (deconjugation of bile salts, hydrophobicity and β-galactosidase activity) and the resistance to biological barriers (gastric juice and bile salts) of 24 strains of lactic acid starter bacteria (Streptococcus thermophilus, Lactobacillus delbrueckii subsp. bulgaricus and Lactococcus lactis) and 24 strains of probiotic bacteria (Lactobacillus acidophilus, Lactobacillus casei, Lactobacillus rhamnosus and bifidobacteria) were compared. Among the probiotic bacteria tested, Lactobacillus acidophilus was the most interesting species since it showed high values of resistance to gastric juice and bile, hydrophobicity and β-galactosidase and bile salts deconjugation activities. Bifidobacterium bifidum strains showed the same behavior, although the values of the parameters investigated were slightly lower than those obtained for Lactobacillus acidophilus. On the other hand, it was demonstrated that Lactobacillus delbrueckii subsp. bulgaricus was the lactic acid starter species with the best probiotic characteristics among the starter species assessed. It was resistant to gastric juice and bile, and showed high values for β-galactosidase activity. On the other hand, lactic acid starter bacteria showed hydrophobicity values similar to or higher than those obtained for the strains of the Lactobacillus casei . According to the results found, the total probiotic value of a fermented dairy product should take into account not only the intestinal probiotic cultures used in the formulation but also the probiotic contribution of the lactic acid starter microflora.  相似文献

The aim of this work was to investigate the effects of 1,18‐octadecen‐9‐dioic acid (dioic acid) and a Rumex occidentalis extract complex for their skin‐lightening action in an Indian population. Prior to the clinical study, the efficacy of dioic as an inhibitor of melanogenesis was confirmed on dark‐pigmented human melanocytes. As part of a 12‐week vehicle‐controlled clinical study, the skin‐lightening effect of a test product containing 1% dioic acid, 2% of a Rumex occidentalis extract and sunscreens (SPF 15) was assessed on the facial skin of 71 Indian female volunteers. Change in skin colour was monitored by (A) Chroma Meter® measurement (L*, a*, b*) and Individual Typology Angle (ITA?) calculation and (B) Visual grading of standardized photographs by a dermatologist. Colorimetric measurements on volunteers' cheeks showed a significant increase of L* and ITA? compared to baseline after 4, 8 and 12 weeks of test product application. For both L* and ITA? measurements, changes were significantly different than the SPF 15‐containing vehicle at weeks 4 and 12. These results were confirmed by the dermatological visual grading. The overall skin‐lightening action of the test product was beyond the one observed with the SPF 15 vehicle. These findings show that a dioic acid and Rumex occidentalis complex deliver a significant skin‐lightening effect on facial skin in an Indian population.  相似文献

The objective of this work was to evaluate the effect of the supplementation of conjugated linoleic acid (CLA; 4.5 g of cis-9,trans-11 C18:2 and 4.5 g of trans-10,cis-12 C18:2) on milk performance, milk fatty acid (FA) composition, and adipose tissue reactivity in dairy goats fed a high-concentrate diet based on corn silage. Twenty-four multiparous dairy goats in early to mid lactation were used in a 10-wk trial, with a 3-wk adaptation to the experimental total mixed ration that contained corn silage (35%, dry matter basis), beet pulp (20%), barley (15%), and a commercial concentrate (30%). Goats were randomly allocated to 2 experimental groups and they were fed 45 g/d of a lipid supplement (either CLA or Ca salts of palm oil added on top of the total mixed ration). Individual milk production and composition were recorded weekly, and milk FA composition was analyzed in wk 2, 5, and 6. In the last week of the trial, an isoproterenol challenge was performed for 12 goats before morning feeding. The CLA supplementation had no effect on dry matter intake (DMI), body weight (BW), milk yield, milk protein content, and lactose yield and content, but it significantly decreased milk fat yield and content by 18 and 15%, respectively. The decrease in milk fat yield was related to a lower secretion of FA synthesized de novo, of the medium-chain FA, and to a lesser extent of the long-chain FA that are taken up from the peripheral circulation. The CLA supplementation decreased the proportion of the sum of C16:0 and C16:1 and the sum of total cis C18:1, and it increased the proportions of the sum of long-chain (C >16) and the sum of iso FA without modification of the total trans C18:1 and the sum of FA synthesized de novo (C <16). During the first 25 min relative to isoproterenol injection, the maximal concentrations, the increases above basal concentration, the changes in area under the curve, and the total area under the curve for glucose and nonesterified FA were not affected by CLA treatment. In conclusion, CLA supplementation associated with a high-concentrate diet based on corn silage resulted in decreased milk fat yield, increased net energy balance, and it did not affect the sensitivity of the adipose tissue to lipolytic challenge in lactating goats.  相似文献

The aim of the present study was to produce pork with enhanced nutritive value for humans, both in terms of fatty acid profile (mainly long chain n − 3 fatty acids by feeding fish oil) and selenium. Forty-eight female pigs were allocated to one of six treatment groups: animal by-products and plant feedstuffs with tallow, plant feedstuffs with a blend of soybean oil and linseed oil with or without a supplement (CLA, selenium, vitamin E and vitamin C), plant feedstuffs with tallow and supplement, plant feedstuffs with fish oil and supplement. The diets containing the fish oil were fed up to either 49 days or 28 days before slaughter. The dietary treatments had no significant effects on growth performance, carcass characteristics and meat quality. When fish oil was included in the diet, higher levels of EPA, DPA and DHA were measured in the subcutaneous fat (up to 3.74%).  相似文献

BACKGROUND: Chitosan (CTS), as an ideal coating material, has been used for post‐harvest treatment of fruits and vegetables. The effects of coating with 1.5% CTS, alone or combined with 10 mmol L?1 ascorbic acid (AsA), on post‐harvest quality and core browning (CB) incidence in ‘Yali’ pears during storage at room temperature (25 ± 1 °C) were investigated in this study. RESULTS: It was found that both CTS coating and CTS + AsA coating delayed the increase of weight loss, retained greater firmness, total soluble solids and titratable acidity content than controls; coatings decreased respiration rate and membrane permeability, and also effectively inhibited CB after 60 days of storage. Compared with CTS coating alone, CTS + AsA coating increased these beneficial effects and also helped to retain a much higher AsA content and the activities of antioxidative enzymes (superoxide dismutase, catalase and ascorbate peroxidase). CONCLUSION: The CTS coating has the potential to maintain post‐harvest quality and control CB which is the main problem in ‘Yali’ pears during storage. The CTS + AsA coating has more effective application in this study, which could not only be due to reducing the respiration rate and inhibiting the senescence process, but also to increasing the antioxidant capability of the fruit. Two batches of Tenerife cheese were produced from pasteurised goats’ milk, one with a commercial starter and another with an autochthonous starter consisting of three selected strains of lactic acid bacteria isolated from artisanal Tenerife cheese. Influence of starter on the characteristics of the cheeses was evaluated. Owing to the composition of the starters, lactobacilli and leuconostocs varied significantly between both cheeses. However, the type of starter did not significantly affect the physicochemical and proteolytic properties of the cheeses. In general, cheeses made with autochthonous starter received higher scores for sensory attributes, especially for aroma, than those made with commercial starter.  相似文献

Forty-eight young Holstein bulls (slaughtered at 458.6 ± 9.79 kg body weight) were used to evaluate the effect of whole linseed and conjugated linoleic acid (CLA) supplementation on animal performance, adipose tissue development, and carcass characteristics. The animals were fed with one of four isoenergetic and isoproteic diets: control (0% linseed, 0% CLA), linseed (10% linseed, 0% CLA), CLA (0% linseed, 2% CLA), and linseed plus CLA (10% linseed, 2% CLA). Animal performance and carcass characteristics were unaffected by diet composition. Adding linseed or CLA to the concentrate diet did not result in significant differences in adipocyte size and number or lipogenic enzyme activity. However, while the frequency distribution of subcutaneous adipocyte diameters followed a normal distribution, the frequency distribution of intramuscular adipocyte diameters was not normal in any dietary group (skewness coefficients: 0.8, 1.2, 0.9, 0.8 for control, linseed, CLA, and linseed plus CLA, respectively; P < 0.05), indicative of adipocyte proliferation in the intramuscular adipose tissue.  相似文献

The objective of these studies was to determine the effects of feeding a novel rumen-protected Lys (RP-Lys) product on plasma AA, lactational performance, and Lys bioavailability. To evaluate RP-Lys on lactation performance a corn-based diet (42.5% of corn silage and 21.9% of corn and corn by-products, on DM basis) was formulated to be Lys deficient but adequate in Met, energy, and metabolizable protein. Thirty-six lactating Holstein cows were fed either a Lys-deficient control diet (CON) with no added RP-Lys, or diets containing 0.3% of RP-Lys (0.3RP-Lys) or 0.6% of RP-Lys (0.6RP-Lys) for 8 wk. There were no effects on dry matter intake (mean ± SD; 26.1 ± 0.58 kg/d), milk yield (37.9 ± 0.72 kg/d), or milk composition to the RP-Lys supplementation. No effect was observed on plasma AA concentrations except for His. Plasma His was linearly reduced by Lys feeding (42.6, 41.2, 30.0 ± 4.09 μM, for CON, 0.3RP-Lys, and 0.6RP-Lys, respectively). Calculated efficiency of Lys utilization decreased linearly with RP-Lys supplementation. In the companion study, 3 rumen-cannulated lactating dairy cows were used in a 3 × 3 Latin square design to assess the bioavailability of the RP-Lys. Free Lys (HCl-Lys), RP-Lys, and water were administered separately by postruminal bolus dosing. The Lys bioavailability was assessed by the ratio of area under the curve of Lys plasma concentration for RP-Lys compared with HCl-Lys and discounted for the area under the curve for water bolus dose. The estimated bioavailability of the RP-Lys was 24.4% ± 4.61. In summary, increased supplemental doses of Lys had no effect on Lys plasma concentration and lactational performance when fed to dairy cows on a corn-based diet, although altered Lys as % of essential AA was observed. However, the lack of effects should be considered in light of the lower-than-expected bioavailability of the RP-Lys.  相似文献
