This research aimed at evaluating the protective effects of almond oil against acute hepatic injury induced by carbon tetrachloride in rats. The study results showed that animals received almond oil prior to the administration of CCl4 significantly decreased serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP) and lactate dehydrogenase (LDH) activities, and total cholesterol (TC), triglyceride (TG) and low density lipoprotein (LDL) content, and increased serum high density lipoprotein (HDL) content. Whereas, pre-treatment with almond oil markedly increased rat hepatic superoxide dismutase (SOD), catalase and glutathione peroxidase (GPx) levels, and decreased the malondialdehyde (MDA) level. These results combined with liver histopathology demonstrated that almond oil has potent hepatoprotective effects, and could be developed as a functional food for the therapy and prevention of liver damage.  相似文献

Pleurotus ostreatus (PO), which is also called the oyster mushroom, is the secondly most popular cultivated mushroom in the world and has been shown to display antioxidant activity. In this study, the hepatic protective effects of extracts of PO cultured in wheat (POW) on carbon tetrachloride (CCl4)-induced liver injury in rats were evaluated by analyzing blood markers of liver injury. Sprague-Dawley rats were pretreated with POW extracts (0, 0.5, and 1.0 g/kg) orally for 14 days prior to the administration of CCl4 for 3 days. Marked evaluation of alanine and aspartate aminotransferases, lactate dehydrogenase, alkaline phosphatase, and γ-glutamyltransferase were observed in the plasma from control rats after CCl4 treatment. This increased liver injury markers were significantly reduced in a dose-dependent manner by pretreatment with the POW extract, suggesting that POW prevented acute liver damage in CCl4-intoxicated rats by suppressing cellular leakage and loss of the functional integrity of cell membranes in the liver. POW extracts could also improve lipid profiles damaged caused by CCl4 in liver through a reducing plasma triglyceride and total cholesterol and a recovering plasma HDL-cholesterol. These results suggest that the extracts of POW ameliorate hepatotoxicity induced by CCl4 in the rat model.  相似文献

The vast biodiversity of nature provides bioactive compounds that may be useful in the fight against chronic diseases. This study was designed to investigate the protective effects of the ethanol extract of Spirulina laxissima West (Pseudanabaenaceae) (EESL) against carbon tetrachloride (CCl(4)) induced hepatotoxicities in rats. Male albino rats of Sprague-Dawley strain were treated orally with the ethanol extract of S. laxissima (50, 100 mg kg(-1) body wt.) 1 h before each CCl(4) administration. The ethanol extract of S. laxissima showed the maximum antioxidant property in vitro. There were statistically significant losses in the activities of antioxidant enzymes and an increase in TBARS and liver function marker enzymes in the serum of the CCl(4)-treated group compared with the control group. However, all the tested groups were able to counteract these effects. The antioxidant activity of the extracts might be attributable to its proton-donating ability, as evidenced by DPPH. In the present study, the decline in the level of antioxidant observed in CCl(4)-treated rats is a clear manifestation of excessive formation of radicals and activation of the lipid peroxidation system resulting in tissue damage. The significant increases in the concentration of antioxidant enzymes in tissues of animals treated with CCl(4) + EESL indicate the antioxidant effect of EESL. This study suggests that EESL can protect the liver against CCl(4)-induced oxidative damage in rats, and the hepatoprotective effect might be correlated with its antioxidant and radical-scavenging effects.  相似文献

在体外条件下模拟二甲基亚硝胺(NDMA)的生成反应,对八角、丁香、洋葱、香菜、小茴香、大蒜这6种香辛料提取液的抑制NDMA生成的作用和加入剂量的关系进行研究.结果表明:在加入量逐渐加大的情况下,八角和丁香的抑制效果随之增强,最高可达到90%以上,大蒜和洋葱的抑制效果不明显,芫荽和小茴香的作用则是先强后弱,最终稳定在一定水平.  相似文献

The dechlorination of carbon tetrachloride (CT) by Fe(II) associated with goethite in the presence of transition metal ions was investigated. X-ray photoelectron spectroscopy (XPS) and X-ray powder diffraction (XRPD) were used to characterize the chemical states and crystal phases of transition metals on solid phases, respectively. CT was dechlorinated to chloroform (CF) by 3 mM Fe(II) in 10 mM goethite (25.6 m2 L(-1)) suspensions. The dechlorination followed pseudo-first-order kinetics, and a rate constant (k(obs)) of 0.036 h(-1) was observed. Transition metal ions have different effects on CT dechlorination. The addition of Ni(II), Co(II), and Zn(II) lowered the k(obs) for CT dechlorination, whereas the amendment of 0.5 mM Cu(II) into the Fe(II)-Fe(III) system significantly enhanced the efficiency and the rate of CT dechlorination. The k(obs) for CT dechlorination with 0.5 mM Cu(II) was 1.175 h(-1), which was 33 times greater than that without Cu(II). Also, the dechlorination of CT by surface-bound iron species is pH-dependent, and the rate constants increased from 0.008 h(-1) at pH 4.0 to 1.175 h(-1) at pH 7.0. When the solution contained Cu(II) and Fe(II) without goethite, a reddish-yellow precipitate was formed, and the concentration of Fe(ll) decreased with the increase in Cu(II) concentration. XPS and XRPD analyses suggested the possible presence of Cu2O and ferrihydrite in the precipitate. Small amounts of aqueous Cu(I) were also detected, reflecting the fact that Cu(II) was reduced to Cu(I) by Fe(II). A linear relationship between k(obs) for CT dechlorination and the concentration of Cu(II) was observed when the amended Cu(II) concentration was lower than 0.5 mM. Moreover, the k(obs) for CT dechlorination was dependent on the Fe(II) concentration in the 0.5 mM Cu(II)-amended goethite system and followed a Langmuir-Hinshelwood relationship. These results clearly indicate that Fe(II) serves as the bulk reductant to reduce both CT and Cu(II). The resulting Cull) can further act as a catalyst to enhance the dechlorination rate of chlorinated hydrocarbons in iron-reducing environments.  相似文献

Natural attenuation processes of chlorinated solvents in soils and groundwaters are increasingly considered as options to manage contaminated sites. Under anoxic conditions, reactions with ferrous iron sorbed at iron(hyro)xides may dominate the overall transformation of carbon tetrachloride (CCl4) and other chlorinated aliphatic hydrocarbons. We investigated mechanisms and product formation of CCl4 reduction by Fe(II) sorbed to goethite, which may lead to completely dehalogenated products or to chloroform (CHCl3), a toxic product which is fairly persistent under anoxic conditions. A simultaneous transfer of two electrons and cleavage of two C-Cl bonds of CCl4 would completely circumvent chloroform production. To distinguish between initial one- or two-bond cleavage, 13C-isotope fractionation of CCl4 was studied for reactions with Fe(II)/ goethite (isotopic enrichment factor epsilon = -26.5% percent per thousand) and with model systems for one C-Cl bond cleavage and either single-electron transfer (Fe(II) porphyrin, epsilon = -26.1 percent per thousand) or partial two-electron transfer (polysulfide, epsilon = -22.2 percent per thousand). These epsilon values differ significantlyfrom calculations for simultaneous cleavage of two C-Cl bonds (epsilon approximately equal to -50 percent per thousand), indicating that only one C-Cl bond is broken in the critical first step of the reaction. At pH 7, reduction of CCl4 by Fe(II)/ goethite produced approximately 33% CHCl3, 20% carbon monoxide (CO), and up to 40% formate (HCOO-). Addition of 2-propanol-d8 resulted in 33% CDCl3 and only 4% CO, indicating that both products were generated from trichloromethyl radicals (*CCl3), chloroform by reaction with hydrogen radical donors and CO by an alternative pathway likely to involve surface-bound intermediates. Hydrolysis of CO to HCOO-was surface-catalyzed by goethite butwastoo slow to account for the measured formate concentrations. Chloroform yields slightly increased with pH at constant Fe(II) sorption density, suggesting that pH-dependent surface processes direct product branching ratios. Surface-stabilized intermediates may thus facilitate abiotic mineralization of CCl4, whereas the presence of H radical donors, such as natural organic matter, enhances formation of toxic CHCl3.  相似文献

This study was performed to examine the anti-fibrotic activity of HV-P411, an herbal mixture of seeds of Vitis vinifera, Schisandra chinensis and Taraxacum officinale extract, against carbon tetrachloride (CCl4)-induced liver fibrosis. Hepatic fibrosis was induced by intraperitoneal injection of CCl4 (0.5 ml/kg, twice weekly) for 8 weeks. Rats were treated orally with HV-P411 at 50, 100, 200, and 400 mg/kg once a day. After chronic exposure to CCl4, the levels of hydroxyproline were markedly increased; these were significantly reduced by HV-P411 at all dose levels. The level of serum aminotransferases and lipid peroxidation were increased after the CCl4 treatment, while reduced glutathione was decreased. These changes were attenuated by HV-P411. In addition, HV-P411 attenuated CCl4-induced raised serum concentration of transforming growth factor-β1, and the levels of matrix metalloprotease-2 and tissue inhibitor of metalloprotease-1 mRNAs. Our results suggest that HV-P411 may prevent liver fibrosis by modulating fibrogenesis and fibrolysis.  相似文献

Rice bran was extracted with water and its effects on potato and banana polyphenol oxidase (PPO) were investigated. Rice bran extract (RBE), conc. 0.3 g mL?1, exhibited PPO inhibition in potato and banana PPO with % inhibition of 69.31% and 47.63%, respectively (P 0.05). RBE showed a concentration‐dependent inhibition on potato and banana PPO. RBE (conc. 0.3 g mL?1) inhibited potato PPO higher than ascorbic acid, citric acid, NaCl and EDTA (final conc. 20 mg L?1); and it also inhibited banana PPO higher than citric acid, NaCl and EDTA (final conc. 20 mg L?1), respectively. The combination of RBE with citric acid or ascorbic acid appeared to be additive inhibitory effect on banana and potato PPO. Kinetic study of the inhibition on potato and banana PPO by RBE showed that RBE was a mixed‐type inhibitor; however, RBE appeared to be able to act directly on enzyme structure rather than substrate structure.  相似文献

Effect of chicory root extract (CRE) on the triglyceride metabolism in orotic acid (OA)-fed rats was investigated. Liver weights and hepatic triglyceride concentrations were markedly increased by OA-feeding rats. These results were attributed to the significant increase in the activity of hepatic phosphatidate phosphohydrolase (PAP), diacylglycerol acyltransferase (DGAT), and ratelimiting enzymes for triglyceride synthesis. Supplementation of CRE to OA did significantly reduced the hepatic triglyceride concentrations and DGAT activity without affecting PAP activity. Furthermore, OA treatment was significantly decreased plasma triglyceride (TG) and increased hepatic TG concentrations and reduced microsomal triglyceride transfer protein (MTP) activity without diminishing MTP mRNA expression in rats. However, hepatic TG concentration was significantly decreased and MTP activity was also reduced without diminishing MTP mRNA expression in rats fed simultaneous with OA and CRE diet. The hepatocytes in the OA-feeding rats contained numerous largely fat droplets, but CRE feeding prevented the OA-induced fat accumulation. Present study demonstrates that CRE reduces the liver TG accumulation by reduced DGAT and MTP activities without diminishing MTP mRNA expression by OA administration.  相似文献

Aflatoxin B1 has been considered as the most potent liver carcinogen for humans. One factor that stimulates the production of aflatoxins in oilseed products, such as sesame seeds and their products is the presence of high levels of fatty acids. This work presents further evidence that sesame is a favorable substrate for aflatoxin B1 production by Aspergillus parasiticus. Moreover, the use of wild artichoke (Cynara cardunculus L.) extract was examined showing inhibition of aflatoxin B1 production in both sesame and yeast extract sucrose medium, inoculated with A. parasiticus. More specifically, the inhibition of aflatoxin B1 production by A. parasiticus in sesame seeds paste was 99.6% and in yeast extract sucrose medium it was 99.4%.  相似文献

The inhibitory effect of an aqueous extract from spinach on degranulation of RBL-2H3 cells is herein reported. The extract significantly suppressed antigen-induced degranulation in a dose-dependent manner without affecting cell viability. Active substances in the extract were heat-stable and trypsin-resistant with molecular weights ranging from 500 Da to 14 kDa. The extract inhibited elevation of the intracellular Ca2+ concentration caused by stimulation by antigen, while not suppressing degranulation induced by a calcium ionophore A23187. Immunoblot analysis revealed that the inhibitory effect results from downregulation of phosphorylation of both Syk kinase and phosphatidylinositol 3-kinase in the signalling pathways involved in degranulation caused by the antigen–antibody interaction. Taken together, these findings suggest that aqueous spinach extract has an anti-allergic activity that controls degranulation.  相似文献

Green rusts (GRs), mixed iron(II)/iron(III) hydroxide minerals found in many suboxic environments, have been shown to reduce a range of organic and inorganic contaminants, including several chlorinated hydrocarbons. Many studies have demonstrated the catalytic activity of transition metal species in the reduction of chlorinated hydrocarbons, suggesting the potential for enhanced reduction by GR in the presence of an appropriate transition metal catalyst. Reductive dechlorination of carbon tetrachloride (CT) was examined in aqueous suspensions of GR amended with Ag(I), Au(III), or Cu(II). The CT reduction rates were greatly increased for systems amended with Cu(II), Au(III), and Ag(I) (listed in order of increasing rates) relative to GR alone. Observed intermediates and products included chloroform, dichloromethane, chloromethane, methane, acetylene, ethene, ethane, carbon monoxide, tetrachloroethene, and various nonchlorinated C3 and C4 compounds. Product distributions for the reductive dechlorination of CT were highly dependent on the transition metal used. A reaction pathway scheme is proposed in which CT is reduced primarily to methane and other nonchlorinated end products, largely through a series of one-electron reductions forming radicals and carbenes/carbenoids. Recently, X-ray absorption fine structure analysis of aqueous GR suspensions amended with Ag(I), Au(III), or Cu(II) showed that the metals were reduced to their zerovalent forms. A possible mechanism for CT reduction is the formation of a galvanic couple involving the zerovalent metal and GR, with reduction of CT occurring on the surface of the metal and GR serving as the bulk electron source. The enhanced reduction of CT by GR suspensions amended with Ag(I), Au(III), or Cu(II) may prove useful in the development of improved materials for remediation of chlorinated organic contaminants.  相似文献

The sensory quality of canned liver sausages is limited by a negative burnt high-temperature-heated flavour, caused by Maillard reaction products between amino acids and sugars. In experiments with liver sausage, reduced glutathione in concentrations of 0.01-0.30% had no positive effect on taste and color. N-acetyl-L-cysteine, an acylated derivative of the amino acid L-cysteine, was tested in concentrations between 0.03-0.50%; a concentration of 0.15% was optimal for the inhibition of the burnt flavour. An acid taste caused by lower pH was corrected by the addition of 0.25% diphosphate. When 0.15% N-acetyl-L-cysteine and 0.25% diphosphate were added to the sausage preparations, the heat-induced changes in amino acids and glucose were the same as in the control. Moreover, under different sterilization conditions, the additive mixture was shown to be an excellent inhibitor of the burnt flavour.  相似文献

The inhibitory effect of onion extract on polyphenol oxidase and enzymatic browning of taro was investigated. The polyphenol oxidase from taro was strongly inhibited by various reducing agents, such as l-ascorbic acid, l-cysteine, dithiothreitol, glutathione and sodium pyrosulfite. The enzyme was also inhibited by addition of onion extract. Regardless of substrates used, the addition of heated onion extract at 100 °C for 10 min, gave a stronger inhibitory effect on taro polyphenol oxidase activity than did fresh unheated extrtact. The inhibitory effect of onion extract was dependent on heating temperature and time. The addition of glucose, glycine, or both to the onion extract, during heating, stimulated the inhibitory effect of the onion extract, suggesting that non-enzymatic browning products, produced during heating, might be responsible for the stronger inhibitory action of the heated onion extract.  相似文献

This study was conducted to investigate the effect of grapevine leaf extract (GLE) on the oxidative stress of rats fed a high-fat diet. Rats were divided into six groups: a normal diet with 0% (NC), 1.0% (NG1.0), and 1.5% GLE (NG1.5); and a high-fat diet with 0% (HFC), 1.0% (HFG1.0), and 1.5% GLE (HFG1.5). Treatment with GLE to the high-fat diet reduced lipid peroxide concentrations in plasma and liver compared to those of HFC rats. Total glutathione and GSH/GSSG of the groups with GLE were higher than those of untreated groups. Hepatic glutathione peroxidase, glutathione-S-transferase, and glutathione reductase activities along with catalase and glutathione peroxidase activities in erythrocytes increased after GLE treatment to high-fat diet compared with HFC rats. Hepatic retinol and tocopherol increased in HFG1.5 group compared to those of HFC group. These results indicate that GLE can protect against oxidative stress induced by a high-fat diet in rats.  相似文献

Ethanol metabolism-associated oxidative stress contributes to the pathogenesis of alcoholic liver disease. We examined the effect of a Mate tea extract on ethanol-induced liver injury in vitro and in vivo models. Isolated hepatocytes were incubated with ethanol. An extract of Yerba-Mate tea (EMT) was added to the cultures simultaneously with ethanol. EMT treatment suppressed the ethanol-induced increase in cell death by inhibiting cytochrome p450 2E1 (CYP2E1) activity, which is related to the production of reactive oxygen species. Furthermore, we examined the effects of EMT on serum transaminase activity, and the progression of liver fibrosis in rats treated with ethanol and CCl4. Rats were fed a diet that included 0.005% or 0.02% EMT or no EMT. For a period of 3 weeks, the animals were provided drinking water containing 5% ethanol and were also treated with carbon tetrachloride (CCl4) (0.1 ml/kg of body weight). EMT treatment suppressed plasma ALT and AST activities in the ethanol- and CCl4-treated rats. EMT treatment also decreased CYP2E1 expression and increased ADH expression in the ethanol- and CCl4-treated rats. EMT treatment fully protected the rats against ethanol- and CCl4-induced liver injury. These results suggest that EMT may serve as a candidate for preventing ethanol-induced liver injury.  相似文献

The inhibitory effect of onion extract on banana polyphenol oxidase activity during ripening of banana when stored at room temperature for 10 days was investigated. The addition of the onion extract that had been heated at 100 °C for 10 min exhibited a higher inhibitory effect on the banana polyphenol oxidase activity during ripening of banana than that of the fresh onion extract. When the onion extract that had been treated at a high temperature was added, the banana polyphenol oxidase activity was markedly inhibited. It was found that heat treated onion extract inhibited the banana polyphenol oxidase non-competitively. The MRP synthesized from arginine, cysteine, histidine and lysine significantly inhibited banana polyphenol oxidase. The enzyme activity was inhibited by addition of various anti-browning agents.  相似文献
