The development of resistance to Boophilus microplus by cattle was studied using sets of cattle twins, in stalls. One twin from each set received 3 infestations of 40,000 larvae and the other a continuous infestation of 1,000 larvae a day over the same period. Sets of twins were then challenged with 1,000 larvae a day for 40 days and 2 field infestations of 20,000 larvae. Correlations of ranking for resistance made at 40-day periods during daily infestations of 1,000 larval ticks or for corresponding intermittent infestations of 40,000 larvae were low until animals had received 120,000 larvae. Thereafter irrespective of method of infestation correlations of ranking were relatively high (r = 0.61-0.96) between periods of infestations or method of infestation including 2 field infestations. Fewer adult female ticks matured on the daily infested animals than on the intermittently infested animals during the treatment period, but animals developed a similar resistance level whether infested with either technique.  相似文献

This study examined the relationships among louse density, pruritus and dermal response to insect antigens in sheep infested with Bovicola ovis. Polypay and Columbia ewes were allocated to two groups, infested and naive, and louse densities and pruritus were monitored for 15 months. Ten months after the initial infestation, all sheep were tested for hypersensitivity on the midside and ears by intradermal injection of soluble extracts of B. ovis, Stomoxys calcitrans and Musca autumnalis. The areas of skin reactions were measured at 20 min, 1, 3 and 24 h after injection and skin thickness was measured at 24 h. Louse densities on Polypays were approximately 10 times greater than on Columbias, and pruritus was correlated with louse numbers at most inspections. Most pruritic behaviour was directed to the sides of infested sheep. Wheal and flare reactions developed rapidly to all extracts in both infested and naive ewes. Reactions to louse extract were larger in infested than naive sheep at all four times after injection. In the infested Polypays, reactions to louse extract were greater than to the fly extracts, but in naive sheep there was little difference among extracts. Reactions in naive Columbias were larger than in naive Polypays at 20 min, 1 and 3 h, but had almost completely abated in both groups at 24 h. Reactions in infested Columbias were greater than in infested Polypays at 20 min, but at 24 h reactions in the Polypays were larger. Louse numbers and pruritus were correlated with wheal areas and skin thickness at 24 h, but there was little relationship with the size of reactions at earlier times. These findings are consistent with the development of a hypersensitive response to B. ovis and suggest that dermal reactions to lice may influence sheep susceptibility.  相似文献

In all, 2 groups of lambs were infected either with Oestrus ovis first-instar larvae or with 10,000 third-stage larvae (L3) of Haemonchus contortus. Another group of lambs was infected with both parasites. Fecal nematode egg counts, plasma pepsinogen concentrations, specific O. ovis enzyme-linked immunosorbent assay (ELISA) antibodies, and blood eosinophil counts were monitored and compared to with the values recorded for a control group of uninfected lambs. There was no significant difference between the burden of H. contortus found in mixed and single infections. However, the nematode egg production was significantly depressed in mixed infections. O. ovis affects the population of H. contortus at least by decreasing the parasite egg output. This effect may be mediated through the increase in eosinophil production stimulated by the presence of O. ovis.  相似文献

The efficacy of ivermectin administered orally at a dosage rate of 0.2 mg/kg liveweight against naturally acquired larval infestations of Oestrus ovis in sheep was 100% in a field trial. Ten sheep were free from infestation by first, second and third instar larvae of O. ovis 12 days post treatment, whereas 10 control sheep harboured 37.7 larvae on average, most of them first stage.  相似文献

The effect of a program to control Teania ovis cysticercosis was investigated on 32 adjacent farms at Mt Manypeaks in Western Australia. Farmers were advised not to feed dogs any raw sheep meat or offal and they were supplied with sufficient cestocide to treat all their dogs every 2 months. Other aspects such as the need for control of the movement of dogs and the correct disposal of offal from home killing of sheep were discussed and recommendations made. The success of the program was determined by recording the incidence of Cysticercus ovis in lambs born on the farms and killed at a local abattoir, and the incidence of T. ovis in dogs on the farms. Before the trial 6.9% of lambs were infected with C. ovis and 11 of the 32 farms had a T. ovis infected dog. The C. ovis incidence figures fell to 2.8%, 0.5%, 1.8% and 0.3% for the four years of the trial, and only 1 dog, a recently introduced puppy, was found to be infected with T. ovis. The results are discussed and reasons for the success of the program as a whole and apparent individual farm failures are discussed.  相似文献

A trial was designed to test the efficacy of vaccination of lambs against field challenge with T. ovis eggs using 3 different levels of pasture contamination. Three concentric paddocks (1.62 hectares each) were used and the inner paddock was heavily contaminated with T. ovis eggs by tethering 2 dogs, experimentally infected with T. ovis, at 5 different points in the paddock over a period of 1 month. Four groups of lambs were used. Sixty lambs, from a property known to be free of T. ovis for 2 years previously, were divided into 2 groups of 30 each; one group was vaccinated with T. ovis culture antigen (vaccinated group=V) while the other was vaccinated with culture medium without antigen (sham-vaccinated group=SV). Thirty mature sheep were obtained from a property with a recent record of T. ovis infection (previous natural exposure to infection group=PNE) and 15 lambs were artificially reared from birth under cestode-egg-free conditions (artificially reared groups= AR). Ten animals from groups V, SV, and PNE, and 5 AR lambs were grazed on each of the 3 concentric paddocks for 6 weeks, and then all animals were slaughtered and their total muscle mass (including heart, diaphragm, tongue and masseters) examined, by slicing, for T. ovis cysticerci. The mean total numbers of T. ovis cysticerci recovered were as follows: Inner paddock, PNE 45.5, AR 47.6, SV 107.8, V 6.5; Middle paddock, PNE 5.2, AR 9.0, SV 8.2, V 0.3; Outer paddock, PNE 2.9, AR 2.8, SV 2.3, V 0.2. There were 4 major conclusions from this study. Firstly, vaccination was more effective than previous natural exposure to infection in preventing establishment of new cysticerci. Secondly, although vaccination effectively reduced the numbers of cysticerci, those that became established were mostly viable at the time of slaughter. In contrast, PNE sheep had greater numbers of cysticerci but the majority were degenerating. Vaccination, therefore, did not seem to stimulate a complete immunological response. Thirdly, the presence of T. hydatigena in lambs did not prevent subsequent infection with T. ovis. Fourthly, T. ovis eggs were dispersed from the inner paddock to the outer paddock.  相似文献

Activated oncospheres of Taenia ovis were incubated for 24 h or 48 h at 37-5 degress C in Medium 858 alone or in Medium 858 enriched with 20% lamb serum or foetal calf serum. Antigens contained in the supernatant medium from these incubations were concentrated, emulsified in Freund's incomplete adjuvant and injected intramuscularly into lambs. These lambs developed a very high level of immunity to subsequent challenge infection with T. ovis eggs.  相似文献

Attempts to immunise sheep against natural infestations by Lucilia cuprina larvae have not been effective. Yet it is known that the larvae excrete the immunosuppressant ammonium bicarbonate. The effect of larval ammonium and nonionic ammonia on immunopathobiology was evaluated in 12 infested sheep. The concentration of ammonium in veins draining infested sites was measured in another group of four sheep. Mean jugular unionized ammonia concentration increased 3.5 to 5.6 times above pre-infested control levels. Mean venous ammonium concentrations draining infested sites were 13 times higher than pre-infested jugular or carotid levels. Increases in jugular ammonia concentrations correlated with increased number of larvae, area of infestation, earlier death, neutropenia, eosinopenia, lymphocytopenia, large declines in serum globulins and zinc, and large rises in toxic neutrophils. The high concentrations of toxic unionized ammonia in blood directly permanently damaged neutrophils and lymphocytes and depressed serum globulin production. The results show that the ammonium from the excreta of larvae of L. cuprina may be highly immunosuppressive.  相似文献

The contribution of migratory and resident birds to the introduction of Lyme disease will vary with the degree to which various species expose themselves to, and are infested by, juvenile vector ticks, and their ability to support and transmit the infectious agent. To examine the relative contribution of various passerine species during the emergence of this disease, we compared the abundance and infection rates of the blacklegged tick, Ixodes scapularis Say, removed from mist-netted birds with those from live-trapped mice at a coastal study site in southern Maine, collected during an 8-yr period in which the range of this tick and the incidence of Lyme disease increased in the state. Weekly bird-banding sessions using six 12-m Japanese mist nets were carried out from May through August 1989-1996. In 1989, 1991 and 1993, mice were live-trapped in a Sherman trap grid (7 by 7 m) during five 3-night sessions, June through August; in 1994-1996, 2 such grids were similarly trapped. Annual adult tick abundance was estimated by flagging vegetation. We removed 2,633 juvenile deer ticks from 1,713 of 1,972 birds examined. Twenty-five of 64 bird species were infested. The percentages of birds infested and the rate of infection among removed larvae and nymphs increased over the years, but species varied markedly in their ability to infect ticks. No infected larvae were removed from catbirds or towhees. The larval to nymphal ratio was higher in mice than in birds. Infection rates among bird-derived larvae were less than among mice-derived larvae, but increased with time. Because of the different ways in which individual species of passerine birds contribute to the availability of vector ticks and respond to the agent of Lyme disease in emerging areas, further research into host competency and borreliacidal mechanisms is needed.  相似文献

Investigations into the incidence of Salmonella abortus ovis (S. abortus ovis) infections should not be neglected in diagnosis of ovine abortion cases. For detection of this pathogen agent, direct cultivation, as well as pre-enrichment combined with enrichment procedures in tetrathionate or Rappaport-Vassiliadis medium, should be performed with respect to the features of S. abortus ovis. The detection limit of S. abortus ovis using pre-enrichment and enrichment media could be determined using 6.5 x 10(3)-6.5 x 10(4) bacteria. For subsequent cultivation of S. abortus ovis Gassner, XLD or Rambach agar are suitable. In infected sheep showing no excretion of S. abortus ovis, the pathogen can be detected by serological studies using microagglutination or the ELISA test. The ELISA test proved to be more sensitive than the microagglutination test, detecting antibodies against S. abortus ovis in 17% of the 814 sheep tested. The microagglutination test revealed positive results in only 2% of the sheep tested.  相似文献

The immunosuppressive effect of experimental Boophilus microplus infestation on bovine peripheral blood lymphocytes (PBL) and on host antibody production to a protein antigen (ovalbumin) was examined. Boophilus microplus infestation caused a marginal decrease in the percentage of T lymphocytes in PBL, which was observed in both lightly (5000 larvae) and heavily (40,000 larvae) infested cattle, and began at the second infestation and continued until the end of the fourth infestation. The percentage of B lymphocytes in heavily tick-infested cattle was less than that in non-infested control cattle after the fourth infestation. The response of PBL from tick-infested cattle to phytohemagglutinin (PHA) was always less than that of tick-free cattle after the second infestation. No noteworthy differences were detected between the three stages of tick infestation, that is, 1 week before the peak of adult engorgement, the middle of the peak and 1 week after all ticks had dropped. Boophilus microplus saliva (100 microliters ml-1) suppressed 47% of the response of bovine PBL to PHA in vitro. This suppressive effect of saliva may contribute to the lower responsiveness of PBL from tick-infested cattle. Antibody production by tick-infested cattle was examined during the third and fourth heavy tick infestation. Tick-infested cattle showed a diminished response against ovalbumin after the second immunization. The immunosuppressive effects of tick infestation may play an important role in tick survival or in the transmission of tick-borne diseases in the field.  相似文献

Brucellosis in sheep, caused by Brucella ovis, is primarily a chronic infectious disease of rams with epididymitis as its most characteristic lesion. Six hundred rams from an infected farm were clinically and serologically examined once a year, over a 3-year period. An increase from 2.1% to 6.3% in the prevalence of animals serologically positive to B. ovis occurred over the 3 years. However, the prevalence of rams with lesions in the reproductive tract declined from 14.2% to 6.5% in the third year following one year of strict culling of clinically affected and rams that were serologically positive for B. ovis. Clinical lesions found in the 179 affected rams fell into two main categories: rams with epididymitis and rams with affected lymph nodes. These results suggest that the prevalence of the disease relates mainly to the sexual activity of the animal and not to age in itself. A single cull based on the results of clinical examination and serological test results was unable to decrease the prevalence of B. ovis in an extensive Corriedale sheep flock.  相似文献

Rickettsia rickettsii, the causative agent of Rocky Mountain spotted fever, was lethal for the majority of experimentally and transovarially infected Rocky Mountain wood ticks (Dermacentor andersoni). Overall, 94.1% of nymphs infected as larvae by feeding on rickettsemic guinea pigs died during the molt into adults and 88. 3% of adult female ticks infected as nymphs died prior to feeding. In contrast, only 2.8% of uninfected larvae failed to develop into adults over two generations. Infected female ticks incubated at 4 degreesC had a lower mortality (80.9%) than did those held at 21 degreesC (96.8%). Rickettsiae were vertically transmitted to 39.0% of offspring, and significantly fewer larvae developed from infected ticks. The lethal effect of R. rickettsii may explain the low prevalence of infected ticks in nature and affect its enzootic maintenance.  相似文献

The helminth fauna of the gastrointestinal tract of 25 and the respiratory organs and the livers of 6 German goats was qualitatively and quantitatively examined. One trematode species (Dicrocoelium dendriticum), 2 species of cestodes (Moniezia expansa and metacestodes of Taenia hydatigena) and 28 species of nematodes (24 in the gastrointestinal tract and 4 in the lungs) were recorded. Two goats were infested with Oestrus ovis larvae. The most prevalent species were Ostertagia circumcincta and Chabertia ovina (84% each), Ostertagia trifurcata and Oesophagostomum venulosum (76% each), Ostertagia pinnata and Trichostrongylus colubriformis (64% each), Trichuris ovis (60%) and Skrjabinema ovis and Trichuris globulosa (56% each). The highest mean worm counts were seen in goats infected with Skjabinema ovis (4003), Ostertagia circumcincta (2501), Trichostrongylus axei (1825), Trichostrongylus colubriformis (1578) and Nematodirus battus (1050). Totally, the goats did harbour more nematodes in the abomasum (3734) than in the small intestine (1707) or the large intestine (2343). The lungs were parasitized by Cystocaulus ocreatus, Muellerius capillaris, Neostrongylus linearis and Protostrongylus rufescens.  相似文献

Six adult sheep and four newborn lambs (5 days old) were implanted with stimulator leads into the latissimus dorsi muscle and connected to a Myostim 7220 pacing system (Telectronics Pacing Systems, Inc., Englewood, CO). Electrical stimulation was started immediately after the operation. After 8 weeks of electrical stimulation, contractile force (CF) in adult sheep decreased to 76-81%, and to 78-82% in lambs. After 2 weeks' delay, CF in adults was 96-98%, and only 89-93% in lambs. After a 30 min intensive stress test, unconditioned control muscle lost 39% in lambs and 43% in adults. Muscle conditioned for 8 weeks lost 7-8% CF. However, after 2 weeks' delay, CF in adult muscle lost 33%, but only 12% in lambs. After cessation of electrical stimulation, the LDH-5 and LDH-1 + 2 fractions reverted to initial levels in adults, whereas in lambs, these levels continued to follow trends established during electrical stimulation. In both adults and lambs, the percent area occupied by the mitochondria increased during electrical stimulation by 6.9% in adults and 6.5% in lambs. After electrical stimulation cessation, the percent area in adults returned to baseline levels, whereas it continued to be elevated in lambs (3.3% vs 5.1%, respectively). The transformed muscle of the lamb did not revert to baseline levels after a delay period.  相似文献

An eight-year study was conducted to define the epidemiology of gastrointestinal nematode infection in Suffolk and Gulf Coast Native (Native) breeds of sheep, and to determine if the Native sheep is more resistant to infection. For the initial three years, each breed grazed separate pastures where anthelmintic treatments were administered to individual animals on a salvage basis. For the last five years, both breeds grazed concurrently; anthelmintic treatments were administered to individual animals on a salvage basis for the first three years, and to all animals, when treatment criteria were met, for the last two years. The fecal egg count (FEC) and blood packed cell volume (PCV) were monitored, and tracer lamb nematode burdens were determined. Overall, FEC for both breeds increased in the spring (periparturient rise) for most years and in the summer for all years. Under separate grazing conditions, Native ewes and lambs had consistently lower infection levels than Suffolk ewes and lambs. During the haemonchosis season (June-September) each year, Suffolk ewe and lamb PCV decreased, and Native ewe and lamb PCV remained relatively stable. The salvage treatment protocol resulted in 27 treatments for Suffolk and one for Native ewes; similarly for lambs, 13 for Suffolk and zero for Native. Tracer lambs grazed with their respective breed, and the FEC and mean total nematode burden corresponded with the pattern of infection for their respective breed. The predominant nematodes found in Suffolk and Native tracer lambs were Haemonchus contortus and Trichostrongylus spp., respectively. Under concurrent grazing conditions, the same seasonal repeatable pattern of infection was present and was exhibited by both breeds, with the Native ewes and lambs being consistently and significantly (p < or = 0.05) lower for FEC and higher for PCV. The salvage treatment protocol resulted in 57 and zero treatments for Suffolk and Native ewes, respectively; for lambs, 46 and 11. Tracer lamb nematode burdens again corresponded to their respective breed pattern of infection, with H. contortus and Trichostrongylus spp. being predominant in Suffolk and Native lambs, respectively. Data from all tracer lambs showed a relatively low level of hypobiosis (H. contortus only), and, although there was no consistent hypobiosis season, the tendency was for a higher level to occur in the fall. These results showed that the classic repeatable seasonal pattern of gastrointestinal nematode infection occurred in both breeds of sheep, and that Native sheep were more resistant to infection (specifically H. contortus) than Suffolk sheep.  相似文献

Certain aspects of the development of Ehrlichia canis, causative agent of canine ehrlichiosis (tropical canine pancytopenia) in Rhipicephalus sanguineus ticks were studied. It was found that partial feeding of nymphs infected as larvae with E canis was a desirable, if not necessary, preliminary treatment for successful infection of dogs with ground-up ticks. It remains unclear whether feeding increased the number or altered the virulence of ehrlichiae within tick tissues. Ehrlichia canis organisms were detected by immunofluorescent microscopy in the midgut and hemocytes and by electron microscopy in the midgut and salivary glands of partially engorged adult ticks which had been infected as larvae and nymphs. Organisms were not observed in the ovary. Intracytoplasmic inclusions contained 1 to 80 elementary bodies, each provided with 2 distinct membranes. Infection of the midgut and salivary gland was confirmed by injecting homogenates of these tissues into susceptible dogs. Staining of gut smears of partially engorged adult ticks by fluorescein-conjugated anti-E canis antibody was found to be a reliable indicator of the infection.  相似文献

The frequency of fascioliasis in sheep of south-western Jutland has been determined on the basis of slaughterhouse registration of infected livers over a 5-year period (1969-1973). The average yearly percentage of adult sheep found to be infected ranged from 83.9 to 91.2%, whereas less than 1 % of the slaughtered lambs proved infected. The high infection percentage among adult animals and the strikingly low frequency among slaughter lambs could be explained by the characteristic management system of the marshland: In summer the sheep graze the dike and the foreland on its seaward side, and in winter the animals graze in the marshland. Lymnaea truncatula is not found on or at the seaward side of the dike, whereas it is abundant all over the marshland. Thus the lambs graze exclusively the snail-free area from which they are taken directly to the slaughterhouse. The summer grazing also protects the adult animals as they avoid the massive uptake of metacercariae which normally takes place in fluke infected areas in late summer.  相似文献

Variation between hosts of different ages and genotypes in the developmental success of trichostrongylid parasite eggs from sheep was investigated in two trials. The percentage development to infective third-stage larvae of eggs collected from lambs and adult ewes infected with Ostertagia circumcincta was compared in an indoor trial. In addition, sheep previously bred for either high or low faecal egg count and grazed outdoors on parasite contaminated pasture were sampled; egg development, faecal egg count, generic profile and faecal dry matter were measured. In both trials, development to L3 was significantly lower in eggs derived from adult ewes than from lambs and, in the field trial, from animals selected for low faecal egg count. The observed differences could not be accounted for by variations in faecal egg count, faecal dry matter content nor by differences in the generic composition of worm egg output. Although not shown conclusively, the results are consistent with an immune mechanism influencing the development of free-living stages of gastrointestinal nematodes outside the host. These results have significant implications for our understanding of parasite epidemiology, particularly as it relates to sources of pasture contamination.  相似文献

Four groups of 8 parasite-naive Dorset-crossbred lambs, 3-4 months old, were turned out on infected pasture on 2 May and allocated to 4 separate paddocks. From May to September, 2 groups received Duddingtonia flagrans (10(6) chlamydospores per kg body weight per lamb per day) mixed in 100 g of barley, while the other 2 groups received barley only. All groups remained set-stocked until slaughter for worm counts on 10 October. In late June, all lambs were treated with fenbendazole due to severe parasitic gastroenteritis in all groups. The faecal egg counts were comparable for the 2 treatments throughout the grazing period. Larval development of Ostertagia/Trichostrongylus spp. in faecal cultures was 1-28% in the fungi-fed groups compared with 60-80% in the untreated groups (P < 0.05). In September, pasture larval counts of Ostertagia/Trichostrongylus were 930 and 4400 L3 kg-1 on paddocks of fungi-fed and untreated groups, respectively. Corresponding figures for Nematodirus spp. were 7200 and 11600 L3 kg-1, respectively. At slaughter, the number of immature Ostertagia spp. was 62% lower in the fungi-fed groups compared with the untreated groups (P < 0.05). Four parasite-free lambs were introduced to each paddock during the period 3-23 October and slaughtered for worm counts after 3 weeks of housing. The total worm burden of tracers on paddocks previously grazed by fungi-fed lambs was reduced 86% (P < 0.05; geometric means) compared with control groups, while significant reductions were also seen in abomasal worm counts (68%; P < 0.05), N. spathiger (98%; P < 0.05) and for N. battus (97%; P < 0.01). It is concluded that dosing sheep with D. flagrans while grazing may limit the build up of pasture contamination in the late grazing season and subsequently limit the intake of larvae in sheep.  相似文献
