Inspired by the asymmetric structure and responsive ion transport in biological ion channels, organic/inorganic hybrid artificial nanochannels exhibiting pH‐modulated ion rectification and light‐regulated ion flux have been constructed by introducing conductive polymer into porous nanochannels. The hybrid nanochannels are achieved by partially modifying alumina (Al2O3) nanopore arrays with polypyrrole (PPy) layer using electrochemical polymerization, which results in an asymmetric component distribution. The protonation and deprotonation of Al2O3 and PPy upon pH variation break the surface charge continuity, which contributes to the pH‐tunable ion rectification. The ionic current rectification ratio is affected substantially by the pH value of electrolyte and the pore size of nanochannels. Furthermore, the holes (positive charges) in PPy layer induced by the cooperative effect of light and protons are used to regulate the ionic flux through the nanochannels, which results in a light‐responsive ion current. The magnitude of responsive ionic current could be amplified by optimizing this cooperation. This new type of stimuli‐responsive PPy/Al2O3 hybrid nanochannels features advantages of unique optical and electric properties from conducting PPy and high mechanical performance from porous Al2O3 membrane, which provide a platform for creating smart nanochannels system.  相似文献

Local hypoxia in tumors results in undesirable impediments for the efficiencies of oxygen‐dependent chemical and photodynamic therapy (PDT). Herein, a versatile oxygen‐generating and pH‐responsive nanoplatform is developed by loading MnO2 nanodots onto the nanosystem that encapsulates g‐C3N4 and doxorubicin hydrochloride to overcome the hypoxia‐caused resistance in cancer therapy. The loaded MnO2 nanodots can react with endogenous acidic H2O2 to elevate the dissolved oxygen concentration, leading to considerably enhanced cancer therapy efficacy. As such, the as‐prepared nanoplatform with excellent dispersibility and satisfactory biocompatibility can sustainably increase the oxygen concentration and rapidly release the encapsulated drugs in acid H2O2 environment. In vitro cytotoxicity experiments show a higher therapy effect by the designed nanoplatform, when compared to therapy without MnO2 nanodots under hypoxia condition, or chemical and photodynamic therapy alone with the presence of MnO2 nanodots. In vivo experiments also demonstrate that 4T1 tumors can be very efficiently eliminated by the designed nanoplatform under light irradiation. These results highlight that the MnO2 nanodots‐based nanoplatform is promising for elevating the oxygen level in tumor microenvironments to overcome hypoxia limitations for high‐performance cancer therapy.  相似文献

A simple preparation of thermoreversible gelatin‐based ferrogels in water provides a constant structure defined by the crosslinking degree for gelatin contents between 6 and 18 wt%. The possibility of varying magnetite nanoparticle concentration between 20 and 70 wt% is also reported. Simulation studies hint at the suitability of collagen to bind iron and hydroxide ions, suggesting that collagen acts as a nucleation seed to iron hydroxide aggregation, and thus the intergrowth of collagen and magnetite nanoparticles already at the precursor stage. The detailed structure of the individual ferrogel components is characterized by small‐angle neutron scattering (SANS) using contrast matching. The magnetite structure characterization is supplemented by small‐angle X‐ray scattering and microscopy only visualizing magnetite. SANS shows an unchanged gelatin structure of average mesh size larger than the nanoparticles with respect to gel concentration while the magnetite nanoparticles size of around 10 nm seems to be limited by the gel mesh size. Swelling measurements underline that magnetite acts as additional crosslinker and therefore varying the magnetic and mechanical properties of the ferrogels. Overall, the simple and variable synthesis protocol, the cheap and easy accessibility of the components as well as the biocompatibility of the gelatin‐based materials suggest them for a number of applications including actuators.  相似文献

A pH responsive, chitosan‐based hydrogel film is used to cap the pores of a porous SiO2 layer. The porous SiO2 layer is prepared by thermal oxidation of an electrochemically etched Si wafer, and the hydrogel film is prepared by reaction of chitosan with glycidoxypropyltrimethoxysilane (GPTMS). Optical reflectivity spectroscopy and scanning electron microscopy (SEM) confirm that the bio‐polymer only partially infiltrates the porous SiO2 film, generating a double layer structure. The optical reflectivity spectrum displays Fabry–Pérot interference fringes characteristic of a double layer, which is characterized using reflective interferometric Fourier transform spectroscopy (RIFTS). Monitoring the position of the RIFTS peak corresponding to the hydrogel layer allows direct, real‐time observation of the reversible volume phase transition of the hydrogel upon cycling of pH in the range 6.0–7.4. The swelling ratio and response time are controlled by the relative amount of GPTMS in the hydrogel. The pH‐dependent volume phase transition can be used to release insulin trapped in the porous SiO2 layer underneath the hydrogel film. At pH 7.4, the gel in the top layer effectively blocks insulin release, while at pH 6.0 insulin penetrates the swollen hydrogel layer, resulting in a steady release into solution.  相似文献

In this study, we report on a novel composite membrane system for pH‐responsive controlled release, which is composed of a porous membrane with linear grafted, positively pH‐responsive polymeric gates acting as functional valves, and a crosslinked, negatively pH‐responsive hydrogel inside the reservoir working as a functional pumping element. The proposed system features a large responsive release rate that goes effectively beyond the limit of concentration‐driven diffusion due to the pumping effects of the negatively pH‐responsive hydrogel inside the reservoir. The pH‐responsive gating membranes were prepared by grafting poly(methacrylic acid) (PMAA) linear chains onto porous polyvinylidene fluoride (PVDF) membrane substrates using a plasma‐graft pore‐filling polymerization, and the crosslinked poly(N,N‐dimethylaminoethyl methacrylate) (PDM) hydrogels were synthesized by free radical polymerization. The volume phase‐transition characteristics of PMAA and PDM were opposite. The proposed system opens new doors for pH‐responsive “smart” or “intelligent” controlled‐release systems, which are highly attractive for drug‐delivery systems, chemical carriers, sensors, and so on.  相似文献

Inorganic nanoparticles (NPs) are promising drug delivery carriers owing to their high drug loading efficiency, scalable preparation, facile functionalization, and chemical/thermal stability. However, the clinical translation of inorganic nanocarriers is often hindered by their poor biodegradability and lack of controlled pH response. Herein, a fully degradable and pH‐responsive DOX@ACC/PAA NP (pH 7.4–5.6) is developed by encapsulating doxorubicin (DOX) in poly(acrylic acid) (PAA) stabilized amorphous calcium carbonate (ACC) NPs. The DOX‐loaded NPs have small sizes (62 ± 10 nm), good serum stability, high drug encapsulation efficiency (>80%), and loading capacity (>9%). By doping proper amounts of Sr2+ or Mg2+, the drug release of NPs can be further modulated to higher pH responsive ranges (pH 7.7–6.0), which enables drug delivery to the specific cell domains of tissues with a less acidic microenvironment. Tumor inhibition and lower drug acute toxicity are further confirmed via intracellular uptake tests and zebrafish models, and the particles also improve pharmacokinetics and drug accumulation in mouse xenograft tumors, leading to enhanced suppression of tumor growth. Owing to the excellent biocompatibility, biodegradability, and tunable drug release behavior, the present hybrid nanocarrier may find broad applications in tumor therapy.  相似文献

To face the increasing demand of self‐healing hydrogels with biocompatibility and high performances, a new class of cellulose‐based self‐healing hydrogels are constructed through dynamic covalent acylhydrazone linkages. The carboxyethyl cellulose‐graft‐dithiodipropionate dihydrazide and dibenzaldehyde‐terminated poly(ethylene glycol) are synthesized, and then the hydrogels are formed from their mixed solutions under 4‐amino‐DL‐phenylalanine (4a‐Phe) catalysis. The chemical structure, as well as microscopic morphologies, gelation times, mechanical and self‐healing performances of the hydrogels are investigated with 1H NMR, Fourier transform infrared spectroscopy, atomic force microscopy, rheological and compression measurements. Their gelation times can be controlled by varying the total polymer concentration or 4a‐Phe content. The resulted hydrogels exhibit excellent self‐healing ability with a high healing efficiency (≈96%) and good mechanical properties. Moreover, the hydrogels display pH/redox dual responsive sol‐gel transition behaviors, and are applied successfully to the controlled release of doxorubicin. Importantly, benefitting from the excellent biocompatibility and the reversibly cross‐linked networks, the hydrogels can function as suitable 3D culture scaffolds for L929 cells, leading to the encapsulated cells maintaining a high viability and proliferative capacity. Therefore, the cellulose‐based self‐healing hydrogels show potential applications in drug delivery and 3D cell culture for tissue engineering.  相似文献

To apply siRNA as a therapeutic agent, appropriate attention should be paid to the optimization of the siRNA gene silencing effect, both in terms of magnitude and duration. Intracellular time‐controlled siRNA delivery could aid in tailoring the kinetics of siRNA gene knockdown. However, materials with easily tunable siRNA release properties have not been subjected to thorough investigation thus far. This report describes cationic biodegradable dextran microgels which can be loaded with siRNA posterior to gel formation. Even though the siRNAs are incorporated in the hydrogel network based on electrostatic interaction, still a time‐controlled release can be achieved by varying the initial network density of the microgels. To demonstrate the biological functionality of the siRNA loaded gels, we studied their cellular internalization and enhanced green fluorescent protein (EGFP) gene silencing potential in HUH7 human hepatoma cells.  相似文献

The ideal theranostic nanoplatform for tumors is a single nanoparticle that has a single semiconductor or metal component and contains all multimodel imaging and therapy abilities. The design and preparation of such a nanoparticle remains a serious challenge. Here, with FeS2 as a model of a semiconductor, the tuning of vacancy concentrations for obtaining “all‐in‐one” type FeS2 nanoparticles is reported. FeS2 nanoparticles with size of ≈30 nm have decreased photoabsorption intensity from the visible to near‐infrared (NIR) region, due to a low S vacancy concentration. By tuning their shape/size and then enhancing the S vacancy concentration, the photoabsorption intensity of FeS2 nanoparticles with size of ≈350 nm (FeS2‐350) goes up with the increase of the wavelength from 550 to 950 nm, conferring the high NIR photothermal effect for thermal imaging. Furthermore, this nanoparticle has excellent magnetic properties for T2‐weighted magnetic resonance imaging (MRI). Subsequently, FeS2‐350 phosphate buffer saline (PBS) dispersion is injected into the tumor‐bearing mice. Under the irradiation of 915‐nm laser, the tumor can be ablated and the metastasis lesions in liver suffer significant inhibition. Therefore, FeS2‐350 has great potential to be used as novel “all‐in‐one” multifunctional theranostic nanoagents for MRI and NIR dual‐modal imaging guided NIR‐photothermal ablation therapy (PAT) of tumors.  相似文献

In this paper, a novel bioinspired stem cell‐laden microgel and related in vivo cartilage repair strategy are proposed. In particular, herein the preparation of new stem cell‐laden microgels, which can be injected into the chondral defect site in a minimally invasive way, and more importantly, capable of in situ self‐assembly into 3D macroporous scaffold without external stimuli, is presented. Specifically, thiolated gelatin (Gel‐SH) and vinyl sulfonated hyaluronic acid (HA‐VS) are first synthesized, and then stem cell‐laden gelatin/hyaluronic acid hybrid microgels (Gel‐HA) are generated by mixing Gel‐SH, HA‐VS, and bone mesenchymal stem cells (BMSCs) together via droplet‐based microfluidic approach, followed by gelation through fast and efficient thiol‐Michael addition reaction. The encapsulated BMSCs show high viability, proliferation, and chondrogenic differentiation potential in the microgels. Moreover, the in vitro test proves that BMSC‐laden Gel‐HA microgels are injectable without sacrificing BMSC viability, and more importantly, can self‐assemble into cartilage‐like scaffolds via cell–cell interconnectivity. In vivo experiments further confirm that the self‐assembled microgels can inhibit vascularization and hypertrophy. The Gel‐HA microgels and relevant cartilage repair strategy, i.e., injecting BMSC‐laden microgels separately and reconstructing chondral defect structure by microgel self‐assembly, provides a simple and effective method for cartilage tissue engineering and regenerative medicine.  相似文献

A bioinorganic nanohybrid glucose‐responsive membrane is developed for self‐regulated insulin delivery analogous to a healthy human pancreas. The application of MnO2 nanoparticles as a multifunctional component in a glucose‐responsive, protein‐based membrane with embedded pH‐responsive hydrogel nanoparticles is proposed. The bio‐nanohybrid membrane is prepared by crosslinking bovine serum albumin (BSA)–MnO2 nanoparticle conjugates with glucose oxidase and catalase in the presence of poly(N‐isopropyl acrylamide‐co‐methacrylic acid) nanoparticles. The preparation and performance of this new nanocomposite material for a glucose‐responsive insulin release system is presented. The activity and stability of immobilized glucose oxidase and the morphology and mechanical properties of the membrane are investigated. The enzymatic activity is well preserved in the membranes. The use of MnO2 nanoparticles not only reinforces the mechanical strength and the porous structure of the BSA‐based membrane, but enhances the long‐term stability of the enzymes. The in vitro release of insulin across the membrane is modulated by changes in glucose concentration mimicking possible fluctuations of blood‐glucose level in diabetic patients. A four‐fold increase in insulin permeation is observed when the glucose concentration is increased from normal to hyperglycemic levels, which returns to the baseline level when the glucose concentration is reduced to a normal level.  相似文献

A versatile targeted etching strategy is developed for the large‐scale synthesis of urchin‐like mesoporous TiO2 hollow spheres (UMTHS) with tunable particle size. Its key feature is the use of a low‐temperature hydrothermal reaction of surface‐fluorinated, amorphous, hydrous TiO2 solid spheres (AHTSS) under the protection of a polyvinylpyrrolidone (PVP) coating. With the confinement of PVP and water penetration, the highly porous AHTSS are selectively etched and hollowed by fluoride without destroying their spherical morphology. Meanwhile TiO2 hydrates are gradually crystallized and their growth is preferentially along anatase (101) planes, reconstructing an urchin‐like shell consisting of numerous radially arranged single‐crystal anatase nanothorns. Complex hollow structures, such as core–shell and yolk–shell structures, can also be easily synthesized via additional protection of the interior by pre‐filling AHTSS with polyethylene glycol (PEG). The hollowing transformation is elucidated by the synergetic effect of etching, PVP coating, low hydrothermal reaction temperature, and the unique microstructure of AHTSS. The synthesized UMTHS with a large surface area of up to 128.6 m2 g‐1 show excellent light‐harvesting properties and present superior performances in photocatalytic removal of gaseous nitric oxide (NO) and photoelectrochemical solar energy conversion as photoanodes for dye‐sensitized mesoscopic solar cells.  相似文献

Efficient nuclear delivery of anticancer drugs evading drug efflux transporters (DETs) on the plasma and nuclear membranes of multidrug‐resistant cancer cells is highly challenging. Here, smart nanogels are designed via a one‐step self‐assembly of three functional components including a biocompatible copolymer, a fluorescent organosilica nanodot, and a photodegradable near‐infrared (NIR) dye indocyanine green (ICG). The rationally designed nanogels have high drug encapsulation efficiency (≈99%) for anticancer drug doxorubicin (Dox), self‐traceability for bioimaging, proper size for passive tumor targeting, prolonged blood circulation time for enhanced drug accumulation in tumor, and photocontrolled disassemblability. Moreover, the Dox‐loaded nanogels can effectively kill multidrug‐resistant cells via two steps: 1) They behave like a “Trojan horse” to escape from the DETs on the plasma membrane for efficiently transporting the anticancer “soldier” (Dox) into the cytoplasm and preventing the drugs from being excreted from the cells; 2) Upon NIR light irradiation, the photodegradation of ICG leads to the disassembly of the nanogels to release massive Dox molecules, which can evade the DETs on the nuclear membrane to exert their intranuclear efficacy in multidrug‐resistant cells. Combined with their excellent biocompatibility, the nanogels may provide an alternative solution for overcoming cancer multidrug resistance.  相似文献

Polymeric semiconductors are emerging as a kind of competitive photocatalysts for hydrogen evolution due to their well‐tunable structures, versatile functionalization, and low‐cost processibility. In this work, a series of conjugated porous polymers with substantial cyano‐substituted fully sp2‐carbon frameworks are efficiently synthesized by using electron‐deficient tricyanomesitylene as a key building block to promote an organic base‐catalyzed Knoevenagel condensation with various aldehyde‐substituted arenes. The resulting porous polymers feature donor‐acceptor structures with π‐extended conjugation, rendering them with distinct semiconducting properties. They possess hierarchically porous structures, nanoscale morphologies, and intriguing wettability. These promising physical characters, finely tailorable by varying the arene units, are essentially relevant to the abundant cynao substituents over the whole frameworks. The as‐prepared porous polymers exhibit excellent visible‐light‐driven photocatalytic activity for water‐splitting hydrogen evolution with apparent quantum yield up to 2.0% at 420 nm or 1.9% at 470 nm, among the highest values yet reported for porous polymer‐based photocatalysts, also representing the first example of such kinds of catalysts formed through a metal‐free‐catalyzed carbon–carbon coupling reaction.  相似文献

In this paper, a preparation of stimuli‐responsive capsules based on aqueous microgels is described. Microgel particles act as stabilizers for oil‐in‐water emulsion and organize themselves on the surface of chloroform droplets containing the biodegradable polymer poly(4‐hydroxybutyrate‐co‐4‐hydroxyvalerate) (PHBV). After chloroform evaporation, composite capsules consisting of a thin PHBV wall with integrated microgels are obtained. Due to the presence of microgels acting as sensitive building blocks, the capsules respond to different stimuli (temperature, solvent concentration). Preliminary results indicate that the capsule dimensions and morphology can be tuned by microgel and PHBV concentration in water and chloroform, respectively.  相似文献

A facile method to control the volume‐phase transition kinetics of thermo‐sensitive poly(N‐isopropylacrylamide) (PNIPAM) microgels is presented. Monodisperse PNIPAM microgels with spherical voids are prepared using a microfluidic device. The swelling and shrinking responses of these microgels with spherical voids to changes in temperature are compared with those of voidless microgels of the same size and chemical composition prepared using the same microfluidic device. It is shown that the PNIPAM microgels with voids respond faster to changes in temperature as compared with their voidless counterparts. Also, the induced void structure does not have a detrimental effect on the equilibrium volume change of the microgels. Thus, the volume phase transition kinetics of the microgels can be finely tuned by controlling the number and size of the voids. The flexibility, control, and simplicity in fabrication rendered by this approach make these microgels appealing for applications that range from drug delivery systems and chemical separations to chemical/biosensing and actuators.  相似文献

Freestanding ultrathin 2D noble metal nanosheets have drawn enormous attention due to their potential applications in various fields. However, the synthesis of 2D noble metal nanosheets still remains a great challenge due to the lack of an intrinsic driving force for anisotropic growth of 2D structures. Here, a facile one‐pot synthesis of ultrathin freestanding porous Pd nanosheets (≈2.5 µm in lateral size and 10 nm in thickness) flexibly knitted by interweaved ultrathin nanowires with the assistance of poly(diallyldimethylammonium chloride) is presented. Nanoparticles attachment and subsequent self‐assembly in the synthetic process are responsible for the formation of such intriguing nanostructures. Moreover, finely controlling the pH value of the precursor solution leads to yield different Pd nanostructures with tunable dimensionalities, including 3D nanoflowers, 2D nanosheets, and 1D nanochains. Owing to the unique structural features, the obtained freestanding porous Pd nanosheets exhibit excellent electrocatalytic activity and stability towards formic acid oxidation compared to those of other dimensional counterparts and commercial Pd black.  相似文献

Self‐assembled nanocapsules containing a hydrophilic core and a crosslinked yet thermosensitive shell are successfully prepared using poly(ethylene‐oxide)‐poly(propylene‐oxide)‐poly(ethylene‐oxide) block copolymers, 4‐nitrophenyl chloroformate, gelatin, and 1‐ethyl‐3‐(3‐dimethylaminopropyl) carbodiimide. The core is further rendered magnetic by incorporating iron oxide nanoparticles via internal precipitation to enable externally controlled actuation under magnetic induction. The spherical nanocapsules exhibit a hydrophilic‐to‐hydrophobic transition at a characteristic but tunable temperature reaching 40 °C, triggering a size contraction and shrinkage of the core. The core content experiences very little leakage at 25 °C, has a half life about 5 h at 45 °C, but bursts out within a few minutes under magnetic heating due to iron oxide coarsening and core/shell disruption. Such burst‐like response may be utilized for controlled drug release as illustrated here using a model drug Vitamin B12.  相似文献

Up‐conversion (UC) luminescent and porous NaYF4:Yb3+, Er3+@SiO2 nanocomposite fibers are prepared by electrospinning process. The biocompatibility test on L929 fibrolast cells reveals low cytotoxicity of the fibers. The obtained fibers can be used as anti‐cancer drug delivery host carriers for investigation of the drug storage/release properties. Doxorubicin hydrochloride (DOX), a typical anticancer drug, is introduced into NaYF4:Yb3+, Er3+@SiO2 nanocomposite fibers (denoted as DOX‐NaYF4:Yb3+, Er3+@SiO2). The release properties of the drug carrier system are examined and the in vitro cytotoxicity and cell uptake behavior of these NaYF4:Yb3+, Er3+@SiO2 for HeLa cells are evaluated. The release of DOX from NaYF4:Yb3+, Er3+@SiO2 exhibits sustained, pH‐sensitive release patterns and the DOX‐NaYF4:Yb3+, Er3+@SiO2 show similar cytotoxicity as the free DOX on HeLa cells. Confocal microscopy observations show that the composites can be effectively taken up by HeLa cells. Furthermore, the fibers show near‐infrared UC luminescence and are successfully applied in bioimaging of HeLa cells. The results indicate the promise of using NaYF4:Yb3+, Er3+@SiO2 nanocomposite fibers as multi‐functional drug carriers for drug delivery and cell imaging.  相似文献

Nanometerization of liquid metal in organic systems can facilitate deposition of liquid metals onto substrates and then recover its conductivity through sintering. Although having broader potential applications, producing stable aqueous inks of liquid metals keeps challenging because of rapid oxidation of liquid metal when exposing to water and oxygen. Here, a biocompatible aqueous ink is produced by encapsulating alloy nanodroplets of gallium and indium (EGaIn) into microgels of marine polysaccharides. During sonicating bulk EGaIn in aqueous alginate solution, alginate not only facilitates the downsizing process via coordination of their carboxyl groups with Ga ions but also forms microgel shells around EGaIn droplets. Due to the deceasing oxygen‐permeability of microgel shells, aqueous ink of EGaIn nanodroplets can maintain colloidal and chemical stability for a period of >7 d. Crosslinked alginate‐gel with tunable thickness can retard the generation and release of toxic cations, thereby affording high biocompatibility. The soft alginate shells also enable to recover electric conductivity of EGaIn layers by “mechanical sintering” for applications in microcircuits, electric‐thermal actuators, and wearable sensors, offering huge potential for electronic tattoos, artificial limbs, electric skins, etc.  相似文献
