Production of the phytase (EC 3.1.3.8) from Aspergillus ficuum in a submerged batch process was inhibited by high concentrations of glucose. The inhibition was overcome by applying a fed batch technique in the production of the enzyme. Tests carried out at different oxygen concentrations revealed that aeration had a beneficial effect on the production of the enzyme. The enzyme showed an optimum pH and temperature of 5·0 and 60°C, respectively. Preincubation of the enzyme preparation at 60°C resulted in relatively fast denaturation of the enzyme. Upon storage at 4°C it lost only 15% of its activity in 5 weeks. Aspergillus ficuum also produced phytase when grown on canola meal by a solid state technique. The enzyme catalysed degradation of the phytic acid present in the meal and completely eliminated it, rendering the commodity more suitable for animal feed. An apparent 10% increase in protein content of the canola meal was noted as a result of the growth of the microorganism.  相似文献

Canola meal (CM) or by-products of ethanol production (dried distillers grain, DDG) may offer an economical alternative to soybean meal (SBM) in North American dairy rations. These protein supplements can effectively replace SBM and, in 2 recent meta-analyses, CM had a positive effect on milk and milk protein yields compared with SBM. The objective of this study was to determine if the positive responses observed with inclusion of CM in dairy rations could be explained by an increased availability of His, Lys, Met, or glucose. Eight Holstein dairy cows were used in a replicated 4 × 4 Latin square with 14-d periods. Cows were fed isonitrogenous (17.2% crude protein) and isoenergetic (1.56 Mcal/kg of net energy of lactation) diets formulated to slightly exceed nutrient requirements. Diets contained 38% grass hay and 62% corn-based concentrate including SBM, CM, corn high-protein DDG (HPDDG), or wheat DDG plus solubles (WDDGS) as the single protein supplement. The effect of protein supplements on availability of His, Lys, Met, and glucose was estimated using variations in the whole-body (WB) flux of these nutrients, determined by isotopic dilution. As planned, dry matter intake and milk and milk protein yields were not affected by treatments and averaged 23.7, 31.4, and 1.14 kg/d, respectively. Lactose yield did not differ among diets although milk lactose content tended to be lower with CM and WDDGS diets than with SBM and HPDDG diets. Lysine availability was affected by treatments: the highest WB irreversible loss rate (ILR) was observed for the CM diet (371 g/d) and the lowest for HPDDG diet (290 g/d); values for SBM and WDDGS were intermediate (330 and 316 g/d, respectively). Availability of His and Met did not vary among diets and WB ILR averaged, respectively, 129 and 124 g/d; the CM diet, however, had numerically the highest His and Met ILR. Plasma concentrations of most of the essential AA were higher with the CM diet and lower with the HPDDG diet, the exception being Leu for which the concentration was highest for the HPDDG diet. Glucose WB rate of appearance was altered by diet, with the highest mean observed for SBM (3,036 g/d) and the lowest for CM (2,795 g/d); the 2 diets with the lowest WB glucose rate of appearance (CM and WDDGS) also had the lowest dietary starch concentration. Overall, this study suggested that positive responses in milk and milk protein yields observed with inclusion of CM in dairy rations could be linked to a greater supply of metabolizable protein, including some essential AA, especially His, Lys, and Met, as glucose availability was certainly not increased in cows fed the CM diet.  相似文献

The objective of this experiment was to investigate the effects of replacing conventional, solvent-extracted canola meal (control; CTRL) with high oil content; conventional, mechanically extracted canola meal (CMEC); high-oleic, low polyunsaturated fatty acid (FA) canola meal (HOLL); and high-erucic acid, low-glucosinolate rapeseed meal (RPS) on rumen function, digestibility, milk production, and milk FA composition in lactating dairy cows. The experimental design was a replicated 4 × 4 Latin square with 8 lactating dairy cows. Four of the cows were ruminally cannulated. All oilseed meals were included at approximately 12 to 13% of dietary dry matter (DM). Crude protein and fat concentrations (% of DM) of the meals were 43 and 3.1%, 32.8 and 16.1%, 45.2 and 13.7%, and 34.3 and 17.9% for CTRL, CMEC, HOLL, and RPS, respectively. All diets were formulated to supply net energy of lactation in excess of requirements. The CMEC and RPS diets were predicted to be about 1% deficient in metabolizable protein. Relative to the CTRL, inclusion of high-oil seed meals in the diet lowered ruminal acetate concentration and the molar acetate:propionate ratio and decreased DM intake. Milk yield generally followed DM intake and was lower for CMEC and RPS than the CTRL. Treatments had no effect on milk composition, other than an increase in milk urea nitrogen concentration for HOLL. Fat-corrected milk (3.5%) feed efficiency was increased by HOLL and RPS compared with CTRL. Urinary urea nitrogen losses were increased by HOLL, which, as a consequence, increased the ammonia-emitting potential of manure. The ratio of milk N-to-N intake was greater for CMEC and RPS. Replacing solvent-extracted canola meal with the high-oil meal decreased milk fat 12:0, 14:0, 16:0, and total saturated FA content and enhanced cis-9 18:1 and total monounsaturated FA concentrations. Relative to the CTRL, canola increased total trans FA in milk, whereas inclusion of HOLL in the diet increased trans-11 18:1 and cis-9, trans-11 CLA content. The RPS increased milk fat cis-13 22:1 content from 0.07 to 2.33 g/100 g of FA. In conclusion, HOLL or RPS, which are likely to come from small-scale biodiesel plants where oil is cold pressed without hexane extraction, fed at levels at or above 12 to 13% of dietary DM may decrease feed intake and milk production, but can be used to alter milk FA composition in lactating dairy cows.  相似文献

Decolourization and degradation of the diazo dye Reactive Black 5 was carried out by the yeast Trichosporon akiyoshidainum. A nine-factor Plackett-Burman design was employed for the study and optimization of the decolourization process and production of manganese peroxidase (MnP) and tyrosinase activities. In the present study, 26 individual experiments were conducted and three responses were evaluated. Raising yeast extract concentration significantly enhanced decolourization and MnP production. Carbon and nitrogen sources, glucose and (NH4)2 SO4, showed no significant effect on any response over the concentration range tested. Other culture medium components, such as CaCl2 or MgSO4, could be excluded from the medium formula, as they had no effect on the evaluated responses. Metal ions (Fe, Cu and Mn) showed different effects on decolourization and enzymatic activities. Addition of copper significantly enhanced MnP activity and decreased dye decolourization. On the contrary, iron had a positive effect on decolourization and no effect on enzyme production. Oddly, increasing manganese concentration had a positive effect on tyrosinase production without affecting decolourization or MnP activity. These results strongly suggest that dye decolourization should be regarded as a complex multi-enzymatic process, where optimal medium composition should arise as a compromise between those optimal for each implied enzyme production.  相似文献

The influence of acetic acid preservation of Chilean mackerel (Trachurus murphy) on its suitability of processing a fish meal quality was studied on a pilot-plant scale and on the industrial level. In particular, the influence of acetic acid preservation of raw material on water-oil separation during cooking-pressing and resulting quality changes in fish meal produced was investigated. The results have shown that preservation of raw fish by acetic acid may improve water-oil separation after cooking, whereas the quality of oil and fish meal measured as acid value and available lysine, respectively, remain constant. A concentration of 1000–3000 μg kg?1 of acetic acid in the raw material is recommended.  相似文献

The effect of short-term storage on the protein, phosphorus and phenolic content as well as peroxidase and o-diphenolase activities of cut, harvested Jamaican yam (Dioscorea sp) tubers (D rotundata. D alata and D cayenensis) was studied. There was an initial increase in the total phenolic content up to the third week of storage followed by a gradual decrease to the sixth week. Phenolic content was found to be highest in D cayenensis followed by D rotundata and D alata. The activities of peroxidase (EC 1. 11. 1. 7) and o-diphenolase (EC 1. 10.3.1) increased steadily up to the third week of storage and thereafter decreased to the fifth week. The intensity and rapidity of browning in tubers when cut, correlated very closely with the tuber o-diphenolase and phenolic content levels while the onset of rotting correlated with the peroxidase activity levels in the species studied.  相似文献

Polyphenoloxidase and peroxidase enzyme activities were evaluated following combined pressure, temperature and holding time treatment in banana (Musa acuminata). Using pressures of up to 110 MPa, temperatures of up to 70 °C and holding times of up to 25 min, based on a 23 central composite design, the interactive effects were found to significantly influence the activity of both enzymes in prepared banana pulp. Temperature and pressure were found to influence the inactivation of polyphenoloxidase separately, while temperature, pressure and holding time were found to influence the loss of peroxidase in the banana, although no significant interactive effects were found. The reduction in polyphenoloxidase activity was found to be less influenced by the combined treatment than peroxidase activity, thought to be due to solubilisation of the enzyme and effects of the soluble solids content. © 2000 Society of Chemical Industry  相似文献

在pH值为9.5,阿魏酸存在和37℃的条件下,利用辣根过氧化物酶(EC1.11.1.7)和过氧化氢对酪蛋白进行氧化交联,并用十二烷基磺酸钠-聚丙烯酰胺凝胶电泳和紫外吸收分析确认交联作用。在蛋白质质量分数为0.04%时,交联酪蛋白的乳化活性指数、乳化稳定性分别比酪蛋白提高25.2%和7.0%。利用葡萄糖酸–δ–内酯制备酪蛋白和交联酪蛋白的酸化凝胶,扫描电子显微镜观察可以看出交联酪蛋白凝胶的微观结构中空穴减少,形成了更为致密的结构。  相似文献

Antarctic krill meal as well as mixed krill meal and starches were extruded through a twin‐screw extruder. The extrudates were hard and porous agglomerates. The pellets still showed intact structure without obvious disintegration in response to hexane extraction. The oil recovery rates for raw krill meal and the extrudate of single krill meal were 55.08% and 59.08%, respectively. For extrudates, oil recovery rate increased along with the increasing additive amount of starches, with a maximum of 83.13%. Oil extracted from raw krill meal contained 49.30% of triglycerides (TG), 44.16% of phospholipids (PL), 1.45% of free fatty acids (FFA) and 4.69% of cholesterols (CHO). Meanwhile, polyunsaturated fatty acids (PUFA), monounsaturated fatty acids (MUFA) and saturated fatty acids (SFA) account for 20.89%, 37.84% and 41.27% of the total fatty acids, respectively. By contrast, oils extracted from extrudates contained more TG but less PL and more SFA but less PUFA.  相似文献

BACKGROUND: Physicochemical properties and digestibility of proteins can be modified by covalent interactions with oxidized phenolic compounds, i.e., quinones. In order to control these interactions in food products, the covalent interactions between quinones from caffeoylquinic acid (CQA) and amino acid side chains were studied with mass spectrometry using N‐terminally protected amino acids. RESULTS: The addition of two molecules of CQA, presumably in the form of a pre‐formed dimer, was observed for lysine, tyrosine, histidine and tryptophan. A monomer of CQA seemed to be able to react with histidine and tryptophan, whereas no interaction with a CQA monomer was observed for lysine and tyrosine. Serine and threonine showed no covalent interactions with CQA. Cross‐linking between CQA and the side chains of two molecules of lysine is likely to occur also in proteins. The results show that protein cross‐linking may also be expected to occur via two tyrosine residues in the absence of other phenolic substrates. The side chains of lysine and tyrosine are more reactive than that of histidine and tryptophan. CONCLUSIONS: These results show that covalent protein modification by oxidized phenolics occurs preferentially via an initial dimerization and encompasses not only lysine and cysteine residues. Selected high‐phenolic lines of spearmint were subjected to a constant 30 °C heat regimen for a period of 4 weeks to determine the effects of heat stress on soluble phenolics, phenols and rosmarinic acid biosynthesis and antioxidant capacity. Heat stress significantly reduced levels of total phenolic acids (71–87%) and soluble phenols (75–87%). This loss was concomitant with a loss of total antioxidant capacity of 21–60% after week 1 and up to 95% by week 4. High‐performance liquid chromatography profiling of heat‐stressed plants at 270 and 320 nm detected nearly a complete loss of rosmarinic acid in all seven chemotypes. High‐temperature drying of non‐heat‐stressed plants at 80 °C resulted in a similar loss of total antioxidant capacity and rosmarinic acid content an effect not observed in material that was subjected to low‐temperature drying first, followed by exposure to high temperature. This suggests that heat stress negatively regulates rosmarinic acid biosynthesis and causes a potential rapid biological breakdown of rosmarinic acid in tissues. 2,2‐Diphenyl‐1‐picrylhydrazyl radical assays of heat‐stressed and non‐stressed plants clearly show that rosmarinic acid is the major contributor to the antioxidant capacity in spearmint. Twelve multiparous Holstein cows averaging 65 (33 to 122) DIM were used in a 4 x 4 Latin square for 4-wk periods to determine whether feeding fish oil as fish meal would stimulate increased amounts of milk conjugated linoleic acid (cis-9, trans-11 C18:2; CLA) and transvaccenic acid (trans-11 C18:1; TVA) when the cows were fed extruded soybeans to supply additional linoleic acid. Treatment diets were 1) control; 2) 0.5% fish oil from fish meal; 3) 2.5% soybean oil from extruded soybeans; and 4) 0.5% fish oil from fish meal and 2% soybean oil from extruded soybeans. Diets were formulated to contain 18% crude protein and were composed (dry basis) of 50% concentrate mix, 25% corn silage, and 25% alfalfa hay. Intake of DM was not affected by diet. Milk production was increased by diets 2, 3, and 4 compared with diet 1 (control). Milk fat and milk protein percentages decreased with diets 3 and 4. Milk fat yield was not affected by treatments, but yield of milk protein was increased with supplemental fish meal and extruded soybeans or their blend. When diets 2, 3, or 4 were fed, concentrations of cis-9, trans-11 CLA in milk fat increased by 0.4-, 1.4-, and 3.2-fold, and TVA concentrations in milk fat increased by 0.4-, 1.8-, and 3.5-fold compared with the control milk fat. Increases in TVA and cis-9, trans-11 CLA were 91 to 109% greater when a blend of fish meal and extruded soybeans was fed than the additive effect of fish meal and extruded soybeans. This suggested that fish oil increased the production of CLA and TVA from other dietary sources of linoleic acid such as extruded soybeans.  相似文献

Strawberries are a good source of micronutrients, especially antioxidant phenolics. More information is needed to better exploit the health-promoting effect of such fruits. Several studies focused on the effects of genotype, cultural practices, and seasonal variation on the antioxidant potential of strawberries, but often yielding contradictory results and/or focusing on only 1 source of variability. In the present study, we showed that total phenolic compounds, ascorbic acid, and antioxidant capacity strongly differed between genotypes of strawberry. The precise results revealed the importance of genetic background for the antioxidant capacity and for the content of total phenolics (with up to 3.3-fold variations). Other parameters may also influence the antioxidant capacity in strawberry such as harvest time, culture conditions, and environmental factors. Moreover, in this study, the harvesting time (at the same ripening stage) appeared to be very important, more important than genotype. Variations of the antioxidant capacity of up to 4.1-folds were observed following the harvesting time of "Elsanta" cultivar. PRACTICAL APPLICATION: This article compares the antioxidant capacity and the content in ascorbic acid and phenolic compounds of strawberries of different varieties and of fruits harvested from April to December at the same ripening stage. The importance of strawberry antioxidant capacity resides in its benefits for human health.  相似文献

The reductive dissolution of MnO2 by Fe(II) under conditions simulating acid mine drainage (pH 3, 100 mM SO4(2-)) was investigated by utilizing a flow-through reaction cell and synchrotron X-ray absorption spectroscopy. This configuration allows collection of in situ, real-time X-ray absorption near-edge structure (XANES) spectra and bulk solution samples. Analysis of the solution chemistry suggests that the reaction mechanism changed (decreased reaction rate) as MnO2 was reduced and Fe(III) precipitated, primarily as ferrihydrite. Simultaneously, we observed an additional phase, with the local structure of jacobsite (MnFe2O4), in the Mn XANES spectra of reactants and products. The X-ray absorbance of this intermediate phase increased during the experiment, implying an increase in concentration. The presence of this phase, which probably formed as a surface coating, helps to explain the reduced rate of dissolution of manganese(IV) oxide. In natural environments affected by acid mine drainage, the formation of complex intermediate solid phases on mineral surfaces undergoing reductive dissolution may likewise influence the rate of release of metals to solution.  相似文献

Total volatile basic nitrogen (TVB-N) as a measure of protein degradation and total microbial counts or stored raw mackerel (Trachurus murphy), which had been treated with chemical preservatives, were compared with the TVB-N and microbial counts of untreated fish. In unpreserved fish, decomposition of fish protein was rapid. Treatment of mackerel with acetic (1000-5000 μg kg?1), which was the most effective of the parameters studied, approximately halved the rate of increase in TVB-N compared with the untreated samples. Changes in the total microbial counts presented a similar trend.  相似文献

BACKGROUND: The interactions between phenolic compounds and proteins can modify protein properties important in the food industry. To understand the effects of these interactions, the covalent interactions between caffeoylquinic acid (chlorogenic acid, CQA) oxidised by polyphenol oxidase (PPO) at acidic pH 6 (pH 6) and α‐lactalbumin, lysozyme and bovine serum albumin (BSA) were compared with non‐enzymatically induced covalent interactions at alkaline pH (pH 9). The effects of these modifications on protein properties were examined. RESULTS: Both ways of modification seemed to result in protein modification mainly via dimeric rather than monomeric CQA quinones. These modifications led to a decrease in the number of free primary amino groups of the proteins. Modification with CQA alone induced a low degree of protein dimerisation, which also occurred through the action of PPO alone. Modification drastically reduced the solubility of lysozyme over a broad pH range, whereas that of α‐lactalbumin was strongly reduced only at pH values close to its pI. The solubility of BSA was much less affected than that of the other proteins and only at acidic pH. CONCLUSION: These results indicate some similarities between modifications at pH 6 and 9 and that both modifications clearly change the functional properties of globular proteins.
