This study examined the effects of different, statistically designed, enzymatic maceration treatments on juice yield, turbidity, and phenol yield (total phenols and total anthocyanins) in experimentally produced elderberry juice. Increased pectinolytic enzyme dose, longer maceration time, and elevated reaction temperature all had significantly positive effects on the juice yield. Increased enzyme dose and maceration temperature also increased the yields of anthocyanins in the elderberry juice, while none of the reaction parameters affected the juice turbidity. The juice yield was optimized further in a new experimental template, made by using the statistical steepest ascent optimization method. In the new response surface design template an optimal maceration treatment giving maximal juice yield and anthocyanin yields and low turbidity was identified. With the optimal treatment with a pectinolytic enzyme preparation, Pectinex BE 3L, produced by a cloned Aspergillus strain, a maximal juice yield of 77% w/w of the berry mash, an anthocyanin yield of 2380 mg/kg fresh berry mash, and a turbidity level of 128 formazin nephelometric units (FNU) were obtained. Enzymatic prepress treatment generally decreased turbidity levels by 30% as compared to pressing without prior enzymatic treatment. A comparison of the responses obtained after the optimal enzymatic treatment with five different pectinolytic enzyme preparations showed that the Aspergillus niger preparation Pectinex BE Color gave slightly better juice and phenol yields, and lower turbidity levels than the other enzyme preparations tested. In conclusion, the results demonstrated that juice yields and phenolic yields in elderberry juice could be improved with enzyme treatment and that the optimal reaction conditions for obtaining the best juice yield, highest phenolics, and lowest turbidity levels could be rationally identified via statistical factor level optimization.Industrial relevanceThis paper describes an optimization study on enzymatic pre-press treatment of elderberries for elderberry juice production. The aim is to increase juice yield, lower juice turbidity, and enhance the release of anthocyanins and other co-existing phenolics into the juice via enzyme catalyzed degradation of the cell wall polysaccharides in the elderberries. The work defines an optimal enzymatic treatment for increased elderberry juice yield, anthocyanin yield, and lowered juice turbidity, and also demonstrates how to shift a statistical design template to obtain improved results on selected parameters. Although the work is focused on elderberry juice the enzymatic as well as the statistical design strategy can easily be adapted for other fruit juice and food processes.  相似文献

A series of experiments was conducted to demonstrate the applicability of a new Filtomat® thread filtration principle for microfiltration of semiprocessed blackcurrant juice and cherry juice. The effect of juice temperature (3–20C), flow (20–80 L/h), and filter pore size (3–10 μm) on the transmembrane pressure, juice turbidity, protein, sugar, and total phenols levels was evaluated in a lab scale microfiltration unit employing statistically designed factorial experiments. Thread microfiltration reduced significantly the turbidity of both juices. For blackcurrant juice, in all experiments, the turbidity was immediately reduced to the level required for finished juice without compromising either the protein, the sugar or the phenols content. High flow rates increased the turbidity in blackcurrant juice, but did not affect cherry juice quality. Filtomat® thread microfiltration therefore appears suitable as a novel technology for berry juice processing, especially for blackcurrant juice filtration.  相似文献

Pre-press maceration treatments with 10 different pectinolytic enzyme preparations were investigated in experimental black currant juice production using response surface design templates. Enzyme dosage, maceration time, reaction temperature, and degree of berry crushing were varied, and the juice yields, anthocyanins, total phenols, and turbidity in the resulting juices were compared for a total of 250 different enzymatic treatments. The yields of anthocyanins and total phenols in the juices ranged from 900 to 2200 and 3050 to 5100 mg/kg wet weight black currant mash, respectively. Juice yields ranged from 66.4% to 78.9% by wet weight of mash. Turbidity levels ranged from 25 to 916 formazan nephelometric units (FNU). The reaction parameters induced larger variations in the responses than the different enzyme preparations, but the cloned Aspergillus niger/Aspergillus aculeatus preparation Pectinex BE® consistently tended to be among those giving the best responses regarding anthocyanin yields, phenols, and low juice turbidity. The optimal maceration was achieved using an enzyme dosage of 0.18% by wet weight of berries with a reaction at 60 °C for 30 min on the most finely crushed berry mash. This treatment gave similar profiles of anthocyanins in the juices with all the 10 enzyme preparations. The same 10 juices all exhibited antioxidant activity against human low-density lipoprotein oxidation in vitro, but the antioxidant potency varied depending on the enzyme preparation used in the pre-press maceration.  相似文献

Incorporation of bioactive-containing fruits and/or vegetables into the carbohydrate-rich food matrix are effective strategies to develop food products with functional health benefits. In this study, blackcurrant powder was added into pea starch and mung bean starch to form pastes to investigate the effects of blackcurrant powder on physicochemical and nutritional properties of the starch pastes. The predictive in vitro glycaemic response of the pastes was highlighted by in vitro glycaemic glucose equivalent assay and alpha-amylase inhibitory activity assay. Both assays showed that blackcurrant powder attenuated (P < 0.05) reducing sugar released, through the inhibition of α-amylase. The colour profiles and textural properties of the pastes were modified by the additions of blackcurrant berry powder at different levels. Nutritional characteristics of the pastes, including total phenolic content and antioxidant activity, were significantly changed (P < 0.05) with the addition of blackcurrant powder.  相似文献

The effects of different enzymatic treatments on the sensory profile and chemical composition, especially various phenolic compounds, sugars and fruit acids, of blackcurrant juices were investigated. In addition to pectinolytic activities, the enzymes had different cellulolytic, hemicellulolytic and glycolytic activities. The enzymatic treatments improved significantly the yield of juice, but with more fermented and astringent characteristics than juices without enzymatic hydrolysis. Enzymes with higher glycosidase activity increased astringency and the content of phenolic compounds in the juice. When the incubation temperature was elevated the contents of fruit acids increased whereas sugars and the perceived sweetness were the highest in the juices without enzymatic treatments. Despite of the significant changes in chemical composition, the sensory properties did not change significantly during 6 weeks of storage. Optimisation of the enzymatic process is crucial for juice making as maximising the juice yield or the content of phenolic compounds may produce unwanted sensory properties to the juice.  相似文献

A new biocatalyst was prepared by the immobilization of a Trametes pubescens laccase, into a wide-pore poly(vinyl alcohol) cryogel. The known enzyme was produced and purified by the previously described procedure. The resulted laccase (yield 40%) has an activity of 46.4 U mg−1 and 12.51 mg mL−1 protein content. The enzyme was subsequently immobilized in a functionalized macroporous cryogel beads by a covalent immobilization technique. The time dependence of the immobilization process and the enzyme loading of the carrier material (5.2 mg g−1 cryogel) were determined by measuring the decrease of protein amount in the enzyme solution. In conversion experiments, a higher stability of the immobilized biocatalyst compared to the free enzyme was evidenced. Steady-state kinetic characterization of four phenols (catechol, caffeic and chlorogenic acids, and catechin) has been performed with free and immobilized laccase, the catalytic parameters being determined and compared. The effect of both laccases (free and immobilized) on the phenol content of retailed apple juice samples, having the same initial composition, was also investigated by working in batch conversion. The variation in phenolic compound content has been compared with that of an untreated apple juice sample having initially the same content of phenolic compounds. A number of advantages resulted in using the immobilized laccase for the apple juice treatment (conservation to some extent of enzyme activity, higher content of phenols preserved, easy separation of the enzyme from the apple juice, therefore avoiding the possible unhealthy effects due to the remaining protein, etc.).  相似文献

果实粒径普遍被认为是影响果实品质的一项重要指标。本实验以宁夏玉泉营地区的‘美乐’和‘蛇龙珠’葡萄为实验材料,将‘美乐’果粒分为大于14 mm(大果粒)、12~14 mm(中等果粒)、小于12 mm(小果粒)3 个粒径等级;将‘蛇龙珠’果粒分为大于15 mm(大果粒)、14~15 mm(中等果粒)、小于14 mm(小果粒)3 个粒径等级,连续两年测定不同粒径葡萄的可溶性固形物质量分数、可滴定酸质量浓度、花色苷单体含量以及单体酚含量。结果表明:连续两年两个葡萄品种均表现为中等果粒所占比例较高;2014年‘美乐’和‘蛇龙珠’葡萄果实的可溶性固形物质量分数随粒径的增大而降低,而2015年均表现为中等果粒较高;连续两年葡萄的种/果质量比和单果粒种子数均随粒径的减小而降低;2014、2015年酚类物质含量在‘美乐’中表现为小果粒和中等果粒较高,‘蛇龙珠’则为大果粒和中等果粒较高;‘美乐’中等果粒葡萄的花色苷单体总量最高,‘蛇龙珠’果实的花色苷单体含量受年份影响较大;单体酚总量以及黄酮醇类物质总量在‘美乐’葡萄中表现为在中等果粒中较高,‘蛇龙珠’葡萄则表现为在大果粒中较高。结论:中等果粒的‘美乐’葡萄和大果粒的‘蛇龙珠’葡萄酚类物质含量较高。  相似文献

Bilberries (Vaccinium myrtillus) and blackcurrants (Ribes nigrum) were treated with extensive dosages of commercial cell wall degrading enzyme preparations, i.e. Econase CE, Pectinex Ultra SP‐L, Pectinex Smash, Pectinex BE 3‐L and Biopectinase CCM. The enzymes were dosed based on the polygalacturonase activity. The juice yield was improved in both berries as a result of the enzymatic treatment. The improvement was more pronounced with blackcurrants owing to their thicker cell walls. The impact of the enzymatic treatment on anthocyanins present in the juices was investigated using HPLC‐DAD. The enzyme preparations affected the contents and composition of anthocyanins in the juices. Pectinex Ultra SP‐L, Pectinex Smash, Pectinex BE 3‐L and Biopectinase CCM increased the total content of anthocyanins by 13–41% in the bilberry juices and by 18–29% in the blackcurrant juices. Econase CE, however, produced a dramatic decrease in the total anthocyanin content in the bilberry juice due to its enzyme profile, whereas no such effect was observed with the blackcurrant juice. All the enzyme mixtures tested produced a total or extensive loss of anthocyanidin galactosides in bilberry juice. Commercial enzyme preparations used in the production of berry juices can improve extraction of anthocyanins into the juice. Phenolic profiles of nine red fruit commercial juice concentrates were characterised by means of high -performance liquid chromatography–diode array detection–mass spectrometry–mass spectrometry. Flavonoids such as anthocyanins, flavonols, hydroxycinnamic acid derivatives, stilbenoids, flavan 3-ols, ellagic acid derivatives, and other phenolic acids, were both identified and quantified in chokeberry, elderberry, blackcurrant and redcurrant, strawberry, red grape, cherry, plum, and raspberry commercial juice concentrates. Once the characterisation was carried out, the antioxidant capacity of each concentrate was assessed in vitro by means of two different methods: the 2,2-azinobis(3-ethylbenzthiazoline-6-sulfonic acid) (Trolox equivalent antioxidant capacity) method and the 2,2-diphenyl-1-picrylhydrazyl radical method. Elderberry, chokeberry and blackcurrant concentrates were the richest in total phenolics and they had the strongest antioxidant capacity; therefore, these three juices may have huge interest as ingredients in the design of functional juices.  相似文献

Aronia berry has a deep red colour, and as it is cheaper than blackcurrant, it may be used to adulterate blackcurrant juice and drinks made therefrom. The aim of the study was to produce a model using multivariate tools to quantify the level of adulteration of blackcurrant concentrates with aronia berry concentrates. Samples of various blends between the two fruits were analysed after dilution on an ultra high pressure liquid chromatography-time of flight mass spectrometry (UPLC-TOF-MS) to create partial least squares (PLS) models. Moreover, variable selection was investigated with the recursive weighted PLS (rPLS) method to increase PLS model quality, and different levels of fusion between data from the two ionisation modes were also compared. The best model was obtained from fusion of ionisation modes with variable selection using the rPLS method. The limit of detection of 5 % aronia berry concentrate in blackcurrant concentrate was achievable with ±1 % error on the adulteration level.  相似文献

Black chokeberry juice (Aronia melanocarpa, Elliot), blackcurrant juice (Ribes nigrum, Ben Lomond) and α-tocopherol were found to protect phosphatidyl choline against oxidation in a peroxidating liposome system as evidenced by lag phases for formation of conjugated dienes. When present together, black chokeberry juice and α-tocopherol showed a clear synergistic effect on the length of the lag phase, while effects of blackcurrant juice and α-tocopherol were additive. The concentration of total phenolics in black chokeberry juice was six times higher than in blackcurrant juice (gallic acid equivalents). Ascorbic acid corresponded to 1% of total phenolics in black chokeberry juice and 10% in blackcurrant juice. Based on the length of the lag phase, the phenolics present in black chokeberry were on an average, twice efficient as scavengers of lipid peroxyl radicals as phenolics in blackcurrant. Black chokeberry was by HPLC analysis of peroxidating liposomes, in contrast to blackcurrant, found to protect α-tocopherol efficiently against oxidation to the end of the lag phase. The phenolics present in black chokeberry juice were concluded to be more efficient in regenerating or protecting α-tocopherol than ascorbic acid or the phenolics in blackcurrant. As for the phenolics, this was further evidenced by ranking of their radical scavenging activity as studied by ESR-spectroscopy.  相似文献

The antioxidant capacities of cranberry juice and three extracts isolated from frozen cranberries containing anthocyanins, water-soluble and apolar phenolic compounds, were evaluated at pH 2.5 and 7, respectively. The free radical-scavenging (FRS) and the lipid peroxidation inhibition (LPI) activities of each samples, and extracts, were studied using the N,N-diethyl-p-phenylenediamine (DPD) decoloration test and the thiobarbituric acid reactive substances (TBARS) assay, respectively. The cranberry phenols displayed good free radical-scavenging properties, but were less efficient at inhibiting the peroxidation of lipids. Of all the samples tested, the water-soluble phenolic compounds showed the greatest free radical-scavenging (68.2 mmol TE/mg phenol) and antioxidant (13.4 mmol TE phenol) activities. The polarity of the phenols, the pH of the medium and the juice process had a great influence on the antioxidant activities. The phenols isolated from cranberries with an aqueous solvent have greater antioxidant properties than those extracted with an organic solvent mixture. The antioxidant activity of the cranberry samples adjusted at pH 2.5 was greater than those adjusted at pH 7. Compared to the cranberry extracts, the juice exhibited a much lower antioxidant activity, especially when compared with the extract containing water-soluble compounds which the extraction conditions were similar to those used to obtain the juice.  相似文献

该试验以红枣为原料,研究乳酸菌、酵母菌单一及偶联发酵对枣汁总糖、总酸、总酚、黄酮和酒精含量的影响,使用液相色谱-质谱联用(Liquid Chromatograph-Mass Spectrometer,LC-MS)技术进行游离型酚酸差异分析,气相色谱-离子迁移谱联用(Gas Chromatography-Ion Migration Spectrum,GC-IMS)技术进行挥发性有机物差异分析。结果表明,原枣汁无论是单一菌种发酵还是混合菌种发酵,总糖、黄酮含量降低,总酸、总酚含量、酒精度均增加,反式阿魏酸、4-羟基苯甲酸、氢化肉桂酸、香草酸、苯甲酸、丁香酸增加;酵母菌发酵枣汁总酚、黄酮、酒精度、苯丙氨酸、儿茶素、表儿茶素、丁香酸 、芥子酸含量均高于乳酸菌发酵枣汁,总酸含量低于乳酸菌发酵枣汁;经混菌偶联发酵,糖类物质降解效率、产酸效率、总酚生成率、黄酮留存率以及产酒精效率更高;游离型酚酸反式阿魏酸、反式肉桂酸、香草酸、没食子酸、对香豆酸、苯甲酸、原儿茶酸、咖啡酸、丁香酸较单菌发酵样品高;挥发性成分53种,包括11种醇类化合物、9种酯类化合物、8种酮类化合物、8种醛类化合物、2种酸类化合物、2种吡嗪吡嗪类化合物,混菌发酵样品中风味物质酯类、醇类、醛类分别占17.2%、58.2%、和1.3%,酯类和醛类显著高于乳酸菌发酵样品、酵母菌发酵样品及原枣汁,香味浓烈,因此采用酵母菌、乳酸菌混合发酵,可以有效地增强发酵枣汁的品质,丰富口感,该研究为推动混菌发酵在红枣酵素发酵生产行业的应用,提供相应的理论基础和科学实践依据。  相似文献

The effects of raspberry, strawberry and bilberry juices and green and black tea on early atherosclerosis in hamsters were investigated. They received an atherogenic diet and at the same time either a juice or a tea at a daily dose corresponding to the consumption of 275 ml by a 70 kg human. After 12 weeks berry juices and teas inhibited aortic lipid deposition by 79–96% and triggered reduced activity of hepatic antioxidant enzymes, not accompanied by lowered plasma cholesterol. These findings suggest that moderate consumption of berry juices and teas can help prevent the development of early atherosclerosis. There were substantial differences between the five beverages in terms of composition and concentration of individual phenolic compounds that were present. This indicates that anti-atherosclerotic effects can be induced by a diversity of phenolic compounds rather than a few specific components. The possible mechanisms by which this is brought about are discussed.  相似文献

Extrusion process was developed to exploit blackcurrant juice press residues from industrial side-streams. Press residues obtained from conventional enzymatic pressing, with high content of fiber and seed oil, and novel non-enzymatic juice processing, with high content of sugars, fruit acids and anthocyanins, were extruded with barley flour, oat flour or oat bran. The recipes consisted of blackcurrant press residues (30%), cereal materials (40%) and potato starch (30%) and small amount of sugar and salt. When compared to enzymatic press residue and oat bran, the novel non-enzymatic press residue extruded with barley or oat flour had higher expansion, lower hardness and density, higher redness (a*), lower pH, and higher contents of fructose, glucose and fruit acids, all contributing positively to liking of texture, appearance, and flavor as well as berry-like experience. These characteristics were obtained with more gentle processing parameters, consisting of a lower total mass flow, screw speed and barrel temperature. Female consumers gave lower ratings in flavor, appearance and overall pleasantness for blackcurrant snacks than males. The study presented a sustainable way of utilizing industrial press residues from different processes of berry juice pressing for production of healthy snacks and breakfast cereals.  相似文献

The effects of enzyme concentration (50–650 mg/kg grated carrot), pectolytic and cellulolytic enzyme ratio (3:7–7:3), incubation time (30–150 min) and temperature (25–65 °C) on juice recovery and viscosity from grated carrot were studied. A central composite rotatable design (CCRD) was used in designing the treatment combinations of four variables at five levels. The process involved in treating the blanched grated carrot with mixture of crude pectolytic enzyme from Aspergillus foetidus and crude cellulolytic enzyme from Trichoderma ressi, keeping the samples at the desired time, followed by extraction of juice. Enzyme-treated grated carrot sample showed increased juice recovery as compared to control. A second-order response surface model adequately fitted the data. All the variables affected juice recovery and viscosity significantly. Enzyme concentration, pectolytic and cellulolytic enzyme ratio, incubation time and temperature had total and combined effect at linear, square and interactive level on both responses. The optimum condition was enzyme concentration, 210.7 mg/kg of grated carrot; pectolytic and cellulolytic enzyme ratio, 3.84:6.16; incubation time, 130 min and incubation temperature 47 °C. Under the optimal conditions, juice extracted from enzyme-treated grated carrot was 74.3% having juice viscosity 1.07 cP, corresponding to the increase in yield by 13.95% and decrease in viscosity by 0.45 cP.  相似文献

Cancer chemopreventive properties were evaluated in HPLC fractions of different polarity obtained from two cranberry juices and three extracts isolated from frozen cranberries and pomace containing anthocyanins, water-soluble and apolar phenolic compounds, respectively. Compounds with close polarities were collected in order to obtain between three and four fractions from each juice or extract. Cranberry fractions were screened for their ability to induce the phase II xenobiotic detoxification enzyme quinone reductase (QR). The results showed that there was no cytotoxicity against the cells used in the test. All samples stimulated the quinone reductase activity except the highest concentrations of the less polar fraction of anthocyanin-rich extract from pomace, which inhibited the QR activity. The QR induction for all samples varied with the concentration and there was an optimal concentration for which the QR induction was maximal. The technological process to manufacture cranberry juice had little influence on the overall QR inducer potencies of cranberry fractions, whereas the ability of phenols in fractions to stimulate the QR activity has been reduced significantly (P  0.05) during the technological process. Among all samples, phenolic compounds of eight fractions presented a maximum QR induction greater than 100 II(QR)/mg phenol. The phenolic compounds of the most polar fraction (rich in phenolic acids) and those of the less polar fraction (rich in proanthocyanidins) showed stronger induction than those observed with phenols from intermediate fractions.  相似文献

Abstract: It remains important to establish the stability of anthocyanins throughout commercial processing in order to maintain the bioactivity of the processed food/s. The present study aimed to assess the recovery and formation of anthocyanins and their free phenolic acid degradation products during the commercial processing of blackcurrant juice concentrate. A bench‐scale processing model was also established to allow for alteration of predefined parameters to identify where commercial processes could be modified to influence anthocyanin yield. No significant loss in anthocyanins was observed throughout the commercial processing of blackcurrants, from whole berry through milling, to pectin hydrolysis and sodium bisulphite addition (P ≥ 0.7). No significant loss in anthocyanins was observed following the subsequent processing of pressed juice, through pasteurization, decantation, filtration, and concentration (P ≥ 0.9). Similarly, the bench‐scale model showed no significant losses in anthocyanin content except during pasteurization (22%± 0.7%, P < 0.001). In the full‐factorial Design of Experiment model analysis, only sodium bisulphite concentration had an impact on anthocyanin recovery, which resulted in an increase (23% to 27%; P < 0.001) in final anthocyanin concentration. No phenolic degradation products (free protocatechuic acid or gallic acid derived from cyanidin and delphinin species, respectively) were identified in any processed sample when compared to authentic analytical standards, analyzed by ultra‐performance liquid chromatography DAD. Practical Application: This article provides crucial data directly applicable to commercial juice processing, such as improving anthocyanin yield and practical considerations for anthocyanin stability and degradation. This aspect is particularly pertinent considering the current commercial interest in anthocyanin‐derived phenolic acids and their health‐related benefits. Further research and development targets in the area of commercial juice product development are identified.  相似文献

以红地球葡萄为研究对象,对葡萄进行膨大处理后,定期采样,测定处理组和对照组的总酚、总黄酮、总黄烷醇以及铜离子还原力、铁氰化钾还原力、DPPH自由基清除率、金属螯合力、羟自由基(·OH)清除率,用DPS软件对处理组和对照组之间的差异及酚类物质与抗氧化活性之间的相关性进行分析。结果表明:膨大处理可增加红地球葡萄酚类物质含量并增强其抗氧化活性;膨大处理对红地球葡萄总酚、总黄酮、总黄烷醇、铜离子还原力、铁氰化钾还原力的影响较大,对DPPH自由基清除率、金属螯合力、·OH清除率的影响较小。铜离子还原力、铁氰化钾还原力与酚类物质(总酚、总黄酮、总黄烷醇)含量均呈极显著正相关;金属螯合力、DPPH自由基清除率与酚类物质含量均无显著相关性,·OH清除率与酚类物质含量呈极显著负相关。铜离子还原力与铁氰化钾还原力呈极显著正相关;与DPPH自由基清除率呈显著正相关。铜离子还原力与·OH清除率、铁氰化钾还原力与·OH清除率呈极显著负相关。  相似文献

Summary. The anthocyanins, flavonols and other phenolic compounds present in blackcurrant juice have been characterized and estimated using column, paper and thin-layer chromatography, and absorption spectrophotometry. New minor phenolic compounds isolated and identified include 5-methyl quercetin, quercitrin, dactylifric acid and p -coumaric acid. the presence of previously reported phenolic compounds in blackcurrant juice has been confirmed. A quantitative assessment of anthocyanin and other phenolic substances has been made.  相似文献
