Ips typographus beetles were collected in the field, separated into eight attack phases (from beetles walking on the trunk of a tree under attack to those excavating gallery systems with a mother gallery longer than 4 cm), and immediately frozen in liquid nitrogen. 2-Methyl-3-buten-2-ol,cis- andtrans-verbenol, verbenone, myrtenol, trans-myrtanol, ipsenol, ipsdienol, and 2-phenylethanol were quantified from excised hindguts against an internal standard, heptyl acetate, in the extraction solvent. Methylbutenol, the pinene alcohols, and 2-phenylethanol showed the same pattern of variation between attack phases in males, with the largest amounts present before accepting females and then a fast decline. Ipsenol and ipsdienol were not detected in males before the females were accepted, and the amounts increased when the females start their egg laying. Verbenone occurred only in trace amounts. The beetles were sampled from five Norway spruce trees (Picea abies) of differing resin flow. The correlations between the nine pheromone components and five major host monoterpenes in the gut showed that the variation in the amount of methyl-butenol, ipsenol, and ipsdienol could not be explained by the variation in the amounts of host monoterpenes. In contrast over 80% of the quantitative variation ofcis-verbenol,trans-verbenol, and myrtenol was explained by the amount of -pinene. The nine pheromone components from 36 individual males were also quantified. Both methylbutenol andcis-verbenol showed a large variation in both amounts and proportions. Females containedtrans-verbenol and traces of most other components found in males. When accepted by the male, they also contained a female-specific compound, -isophorone. The total amounts of, and proportions among, components of the aggregation pheromone produced byIps typographus were found to vary considerably among individuals excised from attacks on standing spruce trees. Chemical analyses of 392 individual male beetles were made by GC-MS. Both unmated and mated males had log-normal frequency distributions in their content of the pheromone components 2-methyl-3-buten-2-ol (MB) andcis-verbenol (cV), since a large fraction of males had a low content. The amount of MB in male hindguts varied independently of cV and the other oxygenated monoterpenes, while the amount of cV covaried with the other pinene alcohols and showed a variation between beetles from different spruce trees. Mated males had, on average, lower amounts of MB than unmated, while the average content of cV in mated males varied with the resin content of their host trees. Ipsdienol and ipsenol were only found in mated males, but in less than 40% and 10%, respectively, of these mated males. Even-aged males exposed to -pinene in the laboratory showed slightly less variation in the amounts of verbenols, and the variations in ratio between cV and tV were similar to those among males attacking the same spruce tree.Coleoptera, Scolytidae.This study was made within the Swedish project Odour Signals for Control of Pest Insects.  相似文献

The density of bark-beetle colonization of a tree could be regulated by a quantitative effect of the pheromone signal from beetles in the tree (cessation of release of attractive pheromone) or by a qualitative effect (production of pheromone components inhibiting attraction). The quantitative hypothesis was tested onIps typographus by varying the release rate of the two known attractive compounds, 2-methyl-3-buten-2-ol (MB) and (4S)-cis-verbenol (cV). The highest number of beetles were captured at traps with the highest release rates. The catch was nearly proportional to the release of MB and cV at a distance between traps of 12 m or more. At 6-, 3-, and 1.5-m distances between traps deployed in a triangular arrangement there was still a good discrimination between release rates, but relatively more beetles, especially males, were caught on the blank. The lower release rates caught an equal sex ratio while the highest release rate caught only about 30% males. The qualitative hypothesis was tested by releasing the suspected inhibitors ipsdienol (Id) and ipsenol (Ie), from traps in the same amounts as cV. Only small effects were noted forI. typographus. However, the competitorI. duplicatus was attracted to Id and inhibited by Ie, while the predatorThanasimus formicarius was attracted to both compounds. On the other hand, when the ratio of Id or Ie to cV was 101 or 0.11 rather than 11, they affected the numbers ofI. typographus attracted. A small amount of Id combined with the attractants increased trap catch, while large amounts of Id or Ie decreased attraction, especially when combined. Six yeast strains have been isolated and identified from the spruce bark beetle,Ips typographus. We have studied the ability of the yeasts to interconvertcis-verbenol,trans-verbenol, and verbenone. (1S)-cis-Verbenol is an active component in the aggregation pheromone ofIps typographus. The isolatedCandida molischiana/ Hansenula capsulata strain can convert both (1R)- and (1S)-cis-verbenol to verbenone. TheCandida nitratophila strain converts (1R)-cis-verbenol totrans-verbenol and (1S)-cis-verbenol to verbenone. Some of the yeast strains produce 3-methylbutanol, 2-methylpropanol, and 2-phenylethanol after growth in Sabouraud medium.  相似文献

Thanasimus formicarius (L.) responds to racemic ipsdienol and ipsenol and less to (S)-cis-verbenol. All three are pheromone components in several bark beetles of the genusIps. Synergistic effects appeared when the components were combined. Methylbutenol alone, the specific pheromone component ofIps typographus, elicited no response, but synergestic effects appeared when methylbutenol was combined withcis-verbenol and ipsdienol. Volatiles released from individual entrance holes of eight spruce bark beetles (Ips typographus) were collected during the first week of attack on a resistant host tree. In order to quantify the release of the highly volatile 2-methyl-3-buten-2-ol (MB) from attacking males, a new method was developed with deuterated quantification standard released at the time of collection. The amounts of collected volatiles, as analyzed by GC and GC-MS, showed a large variation between individual holes and also between subsequent entrainments from the same hole. Most of the quantified compounds on the average have two maxima, with a pronounced intervening depression. The amounts of releasedcis-verbenol (cV) increased five times during the first two days, while the amounts of MB were consistently high. The attacked spruce tree was not taken by the beetles, and the average amounts of the two aggregation pheromone components, MB and cV, increased again after the first maxima. The first peak of oxygenated monoterpene, released in the beginning of the attack containing -terpineol, terpinen-4-ol, bornyl acetate,trans-pinocarveol, and verbenone, was possibly due to spontaneous oxidation of monoterpene hydrocarbons from the tree. Microorganisms established in the gallery wall phloem probably participated in the production of oxygenated monoterpenes during the second increase.Ips typographus (Coleoptera: Scolytidae).  相似文献

Porapak Q-captured volatiles of malePolygraphus rufipennis (Kirby) boring in black spruce and white spruce from Newfoundland and British Columbia, respectively, contained 3-methyl-3-buten-1-ol. Volatiles from logs in which the males had been joined by females contained the compound in reduced quantities. Hindgut extracts from male and femaleP. rufipennis disclosed no male-specific volatiles, but 3-methyl-3-buten-1-ol was detected in extracts of male-produced frass. The compound was not present in extracts from fresh phloem tissue.P. rufipennis of both sexes responded strongly in the field to traps baited with 3-methyl-3-buten-1-ol released at 4390 g/day. There was little response to unbaited traps, fresh uninfested black spruce logs, or to 3-methyl-3-buten-1-ol released at lower rates. Combination of 3-methyl-3-buten-1-ol with either one of five terpenes prevalent in black spruce did not enhance beetle catch. Approximately half of 20 black spruce trees baited with 3-methyl-3-buten-1-ol were attacked, compared to 20.0% of 10 unbaited control trees. Attraction of maleIps paraconfusus to male-infested ponderosa pine logs was inhibited by volatiles from logs infested with mated males and females over an 8-day period in the field. The response of females during this time was not significantly inhibited by these volatiles. Synthesis of the male-specific pheromones, ipsenol and ipsdienol, appeared negligible after 8 days in males allowed to mate with 3 females in these logs while males alone contained levels of these pheromones at about half their maximum rate. The decline in pheromone production in mated males appears to result, at least in part, from a reduction in the activity of the biosynthetic system that converts the host monoterpene, myrcene, to ipsenol and ipsdienol. Mating and feeding have apparently no effect on the biosynthetic system that converts the host monoterpene, (–)--pinene, to the pheromone,cis-verbenol in either males or females. The reduced production and release of pheromones by males after mating appears to play a major function in the process of terminating the aggregation phase of host colonization.  相似文献

The male-produced aggregation pheromone inIps paraconfusus is composed of three compounds. Female bark beetles were exposed to combinations of these compounds, presented as point sources in an enclosed, circular arena. By itself,cis-verbenol (cV) had no effect on the number of beetles that reached the source. Either ipsenol (Ip) alone or ipsdienol (Id) alone strongly increased the number that reached the source, with Id producing a dose-response curve with a much steeper slope. cV moved the onset of the response to Id to higher doses of Id, but the response rose more rapidly after onset than when cV was absent. Overall, cV inhibited the effect of Id except at the highest dose. cV affected the onset of the response to Ip little or none, but strongly increased the slope of the response, synergizing the effect of Ip. The responses to combinations of Id and Ip were related to the log of a linear combination of their doses. The results are consistent with a model where Id and Ip act at a single site of action, but with different potencies, while cV appears to modify the effects of Id and Ip, rather than affecting the site of action directly.  相似文献

The enantiomeric composition of the pheromone components (+)-ipsdienoI, e.e. 87.6%, and (–)-ipsenol, e.e. 93.8%, produced by the male bark beetleIps paraconfusus (Scolytidae) under natural conditions was determined by HPLC separation of their diastereomeric ester derivatives. Males confined in an atmosphere of ipsdienone produced (–)-ipsdienol, e.e. 28%, and (–)-ipsenol, e.e. 86%, indicating an enantiomeric selectivity in the conversion of the ketone to the alcohols. These findings demonstrate an enantioselective conversion mechanism in the biosynthetic pathway to the pheromones from myrcene, a host-plant terpene.  相似文献

Responses of the European spruce bark beetle, I. typographus (Coleoptera: Scolytidae), to low release-rate pheromones were investigated in two experiments in a spruce forest at Wellin, southern Belgium. Dose–response of the beetle was first examined in a trapping experiment in August 1999. The major pheromone components of I. typographus, (S)-cis-verbenol (cV) and 2-methyl-3-buten-2-ol (MB) were released from window traps. Five treatments were replicated five times: (1) blank trap, (2) 0.03 and 1.2, (3) 0.11 and 4.5, (4) 0.34 and 15.2 mg/day of cV and MB, respectively, and (5) Pheroprax. Trap catches increased linearly as a function of increasing release rates of cV and MB. A second study aimed at making preliminary observations on the attraction range of the pheromones as compared to Pheroprax. A release–recapture experiment was carried out in May 2000: four trap-trees located 50 m away from a central release platform were equipped with window traps baited with increasing release rates, tested in separate releases: 0.03 and 1.2; 0.11 and 4.5; 0.34 and 15.2 mg/day of cV and MB, respectively, and Pheroprax. Recaptures increased with the release rate. Trap catches were different at lower release rates, but not between the release rate of 15.2 mg/day of MB and Pheroprax. Variations in captures in relation to wind parameters showed persistently random flight at the lower release rates. Significant upwind flight was observed for Pheroprax only. These results imply that trap interference existed for Pheroprax and suggest that its attraction range may be greater than 50 m.  相似文献

Ips cembrae adults were collected from larch log piles in northeast China, separated into six attack phases, and immediately frozen in liquid nitrogen. Three previously described aggregation pheromone components [ipsenol, ipsdienol, and 3-methyl-3-buten-1-ol (331-MB)] and six other volatiles of beetle origin (2-phenylethanol, geraniol, ipsenone, ipsdienone, myrtenol and verbenone) were identified and quantified by GC-MS from excised male hindguts. No amitinol, a recently reported aggregation pheromone component, was detected in our samples. The amounts of these volatiles (except the last two) showed a similar pattern of variation between attack phases in males. The largest amounts of most male volatiles were present in phases 1–2, when the nuptial chamber was being constructed or only one female was accepted. The amounts of the volatiles declined sharply in the following phases. The hindgut volatiles, mainly the pheromone components, from 46 individual males in phase 1 were also quantified. Ipsenol, ipsdienol, and 331-MB showed a large variation in both amounts and proportions. The chirality of these two dominant aggregation pheromone components was determined as 100% (–)-enantiomer of ipsenol and 96% (+) enantiomer of ipsdienol. No male aggregation pheromone components were detected from mated females, except three extracts that were probably contaminated by male tissues.  相似文献

In bioassays conducted with walking beetles in the laboratory (S)–(+)–, (R)-(–)-, and (±)-ipsdienol were attractive alone, but reduced the attraction of both sexes of the mountain pine beetle,Dendroctonus ponderosae Hopkins, to extracts of female frass. Field trapping studies indicated that attraction ofD. ponderosae to a stimulus composed of myrcene +trans-verbenol +exo-brevicomin was significantly reduced with the addition of (±)- and sometimes (S)-(±)-ipsdienol. Thus, (S)-(+)-ipsdienol produced by males of this species may act as an antiaggregation pheromone. (S)-(+)-Ips-dienol is thought to function as a repellent allomone against the pine engraver,Ips pini (Say), in regions whereI. pini utilizes (R)-(–)-ipsdienol as an aggregation pheromone. However, in southwestern British ColumbiaI. pini was attracted to the (±)-ipsdienol used in field bioassays ofD. ponderosae, a finding consistent with the production of both enantiomers byI. pini in this region. When presented with the ternary semiochemical bait forD. ponderosae, (±)-ipsdienol was not attractive toI. pini. MaleIps paraconfusus Lanier bored and fed in cellulose powder substrates treated with solvent extracts of ponderosa pine phloem in preference to cellulose powder alone. Stimuli that elicit boring and feeding behavior occurred in the water extracts and the combined solvent extracts. No significant boring or feeding occurred in the methanol extract. There was a preference for, but no significant feeding in, the water partition of the ether extract. A synthetic mixture of nine green leaf volatiles (GLVs) including linalool was tested on antennae of Ips typographus (L.) with coupled gas chromatographic–electroantennographic detection (GC-EAD). Strong responses were found to 1-hexanol, (Z)-3-hexen-1-ol, and (E)-2-hexen-1-ol. Weak responses were recorded to (E)-3-hexen-1-ol, (Z)-2-hexen-1-ol and linalool, while hexanal, (E)-2-hexenal and (E)-3-hexenyl acetate elicited no EAD responses. In a laboratory walking bioassay, the attraction of I. typographus females to a synthetic pheromone source was significantly reduced when a mixture of the three most EAD-active GLV alcohols was added to the source. Further reduction in response was obtained when these three alcohols were combined with verbenone (Vn). In field trapping experiments, a blend of 1-hexanol, (Z)-3-hexen-1-ol, and (E)-2-hexen-1-ol reduced I. typographus trap catches by 85%, while ca. 70% reduction of trap catch was achieved by Vn or a blend of (E)-3-hexen-1-ol, (Z)-2-hexen-1-ol, and linalool. The strongest disruptive effect was found when Vn plus a blend of the three most EAD active GLV alcohols was added to the pheromone trap (95% catch reduction). Adding the blend of the three most EAD active alcohols to pheromone-baited traps significantly reduced the proportion of males captured. These three GLV alcohols were also disruptive in the laboratory and in the field when tested individually. Hexanal, (E)-2-hexenal, and (Z)-3-hexenyl acetate were inactive both in the lab and in the field. Our results suggest that these nonhost green leaf alcohols may explain part of the host selection behavior of conifer-attacking bark beetles and may offer a source of inhibitory signals for alternative management strategy for forest protection.  相似文献

The most prominent beetle-produced volatiles identified in the abdominal extracts of maleDryocoetes confusus Swaine after they had bored for 24 hr in logs of subalpine fir,Abies lasiocarpa (Hook.) Nutt. were:exo andendo-brevicomin,trans-verbenol, verbenone, myrtenol,trans-pinocarveol,cis- andtrans-p-menthen-7-ol, 3-caren-10-ol, and several monoterpenes and sesquiterpenes. Myrtenol was the only conspicuous compound in extracts from males that had been exposed toA.lasiocarpa resin volatiles for 24 hr. Laboratory bioassays indicated that both (–)- and (+)-exo-brevicomin were attractive to femaleD. confusus, and that the (–) enantiomer did not inhibit response to its antipode. Volatiles from hindgut extracts of males of the Qinghai spruce bark beetle, Ips nitidus, from different attack phases (phase 1: unpaired males and phases 2–4: males joined with one to three females) and hindgut extracts of mated females were analyzed by gas chromatography–mass spectrometry (GC–MS)/flame ionization detection (FID) with both polar and enantioselective columns. The GC–MS/FID analyses demonstrated that unpaired males from attack phase 1 (nuptial chamber constructed) produced 2-methyl-3-buten-2-ol, approx. 74%-(−)-ipsdienol, and (−)-cis-verbenol as major hindgut components, and (−)-trans-verbenol, (−)-ipsenol, (−)-verbenone, myrtenol, and 2-phenylethanol as minor or trace components. The quantities of 2-methyl-3-buten-2-ol and especially ipsdienol decreased after mating during phases 2–4, whereas the quantities of (−)-cis- and (−)-trans-verbenol did not change. In contrast, the quantity of (−)-ipsenol seemed to increase as mating activity progressed. After mating with three females (harem size = 3; phase 4), only trace to small amounts of male-specific compounds were detected from I. nitidus male hindguts. Chemical analysis of the hindgut extracts of mated females showed only trace amounts of semiochemicals. A field-trapping bioassay in Qinghai, China showed that the four-component “full blend” containing the three major components, 2-methyl-3-buten-2-ol, (±)-ipsdienol, and (−)-cis-verbenol, plus a minor component, (−)-trans-verbenol, caught significantly more I. nitidus (♂/♀ = 1:2.2) than did the unbaited control and two binary blends. The replacement of (±)-ipsdienol with nearly enantiomerically pure (−)-ipsdienol in the “full blend” significantly reduced trap catches, which suggests that both enantiomers are needed for attraction. On the other hand, removal of (−)-trans-verbenol from the active “full blend” had no significant effect on trap catches. Our results suggest that the three major components, 2-methyl-3-buten-2-ol, 74%-(−)-ipsdienol, and (−)-cis-verbenol (at 7:2:1), produced by unpaired fed males, are likely the aggregation pheromone components of I. nitidus, thus representing the first characterization of an aggregation pheromone system of a bark beetle native solely to China.  相似文献

Females of a pinyon pine bark beetle,Ips hoppingi Lanier, were less attracted by the aggregation pheromone produced by conspecific males than by the pheromone produced by the neighboring sibling species,I. confusus (LeConte). Cross-attraction was elicited by males infesting the regional pinyon pine hosts (P. discolor andP. edulis) of eitherIps species in south-eastern Arizona. The movement of bark beetles near an attractive pheromone source is described in terms of mathematical models of the diffusion type. To test the models, two release experiments involving 47,000 marked spruce bark beetles [Ips typographus (L.)] were performed. The attractive source was a pheromone trap, surrounded by eight concentric rings with eight passive trap stations on each ring. Captures were recorded every 2–10 minutes for the pheromone trap and once for the passive traps. The models were fitted to the distribution in time of the central pheromone trap catch and to the spatial distribution of catch among the passive traps. The first model that gives a reasonable fit consists of two phases: Phase one—After release the beetles move according to a diffusion process with drift towards the pheromone trap. The strength of the drift is inversely proportional to the distance from the traps. Phase two—those beetles attracted to, but not caught by, the pheromone trap are no longer influenced by the pheromone, and their movement is described by a diffusion process without drift. In phase two we work with a loss of beetles, whereas the experiment seems to indicate that the loss of beetles in phase one is negligible. As a second model, the following modification of phase one is considered: After release the beetles move according to a diffusion process without drift, until they start responding to the pheromone (with constant probability per unit time), whereafter they start moving according to a diffusion process with drift. This study, like other release experiments, shows that the efficiency of the pheromone trap is rather low. What is specific for the present investigation is that we try to explain this low efficiency in terms of dynamic models for insect movement. Two factors seem to contribute: Some beetles do not respond to pheromone at all, and some beetles disappear again after having been close to the pheromone trap. It also seems that the motility of the beetles decreased after they ceased responding to the pheromone. Furthermore, the data lend some support to the hypothesis that flight exercise increases the response of the beetles to pheromone.  相似文献
