Noradrenergic (NA) involvement in acquisition (AQ) and retention (RET) of a shock-motivated 14-unit T-maze after intraperitoneal (ip) administration of DSP4, a neurotoxin that depletes forebrain NA, was evaluated in 2 experiments using 3-month-old male F-344 rats. In Experiment 1, DSP4 rats did not differ from saline-treated (SAL) rats on any measure of maze performance during AQ (DSP4 2 weeks prior to testing) nor during a RET test 2–3 weeks later (DSP4 immediately after the last AQ trial or 1 week after AQ). In Experiment 2, DSP4 and scopolamine (SCOP), a cholinergic (ACh) antagonist, were administered in combination to test for an NA-ACh interaction. DSP4 again had no effect on AQ performance nor did DSP4 interact with either dose of SCOP to further impair performance. SCOP impaired all measures of maze performance except shock duration. These experiments suggest that central NA systems may not be involved directly or interactively with ACh systems required for efficient performance in this maze. Male rats that received bilateral injections of colchicine into 2 rostrocaudal sites showed relatively long-lasting alterations in a previously acquired radial arm maze task and specific destruction of dentate granule cells. Subsequent experiments with cholinergic drugs indicate that physostigmine or nicotine had no effect on number of errors made in the maze, although other signs of cholinergic or pharmacological activity were present. RS-86, an analog of arecoline, decreased errors in colchicine-treated Ss, but effects were associated with signs of parasympathetic overstimulation and behavioral sedation. Pretreatment with scopolamine increased errors in controls but had no effect in colchicine-treated Ss. Colchicine-treated Ss were less sensitive to the motor stimulant effect of scopolamine. Effects appeared to be associated with increased levels of choline acetyltransferase in the hippocampus and a down regulation of muscarinic postsynaptic receptors. Intradentate colchicine may destroy granule cells, leading to compensatory reinnervation of cholinergic nerve terminals having cell bodies in the septum. A recent animal model that has been particularly useful in the neurobiology of aging has been the age-related decline of spatial information processing capacity in Sprague-Dawley rats measured in the place-learning water task developed by Morris (1981). In the first experiment of the present study, place behavior was examined in young (6 months), old (23–24 months), and very old (28 months) rats of another strain, Long-Evans. As an analogue of aging-related cholinergic dysfunction the effects of atropine sulfate (5–50 mg/kg), an anticholinergic drug that is known to disrupt behavior in this task, also was determined. Place navigation was not impaired in undrugged rats, even those in the oldest age group. Rats treated with atropine showed dose-dependent deficits. In a second experiment, young (4–5 months), old (18–20 months), and very old (28 months) Fischer-344 rats were examined. Place navigation was impaired in the old rats. The very old (28 months) rats could not swim well enough to be tested adequately. Although nonspatial deficits associated with aging may be found across most strains tested, there appear to be very large strain-related differences in spatial processing ability as a function of age. A group of novel neuroleptics (e.g. olanzapine, seroquel, sertindole and ziprasidone) and already marketed compounds (e.g. clozapine, haloperidol and risperidone) were tested for acute effect on spatial learning and memory in Morris' water maze task. Young rats were trained for 4 consecutive days (three trials/day) to find a platform situated beneath the water surface. Two compounds, sertindole and seroquel, were without effect on spatial performance, whereas clozapine impaired performance on the first 2 test days but showed no effect compared to the controls on the last 2 test days. Ziprasidone and olanzapine markedly impaired spatial memory without affecting motor function (measured by the swimming speed). Risperidone and haloperidol also impaired performance but in addition both compounds significantly lowered the swimming speed. The present study indicates that several of the compounds impair spatial learning in Morris water maze. This might be of clinical importance in the treatment of schizophrenics, as many of these patients already show severe cognitive deficits. Therefore, certain antipsychotics could worsen the preexisting memory deficits in schizophrenic patients and this aspect should be considered before antipsychotic treatment.  相似文献

A within-subject design was used to examine delayed-non-match-to-sample radial arm maze performance in aging (6–18 months) male Sprague-Dawley rats. A decrease in correct choices and an increase in retroactive errors were observed at all retention intervals at 18 months of age compared with performance at 6 or 12 months. No age by retention interval interaction was observed. Neither age nor increasing retention interval influenced proactive errors during the retention test. The observation of an age- and delay-dependent increase in retroactive errors, but not proactive errors, suggests that the deficit relates to a memory dysfunction as opposed to a generalized performance deficit. Adult rats show a preference for directional navigation over place navigation in the Morris water task. Here, the authors investigated whether preweanling rats with a newly developed ability to perform the water task also solve the task via directional navigation. After 24-day-old rats were trained to find a hidden platform in a fixed spatial location, a no-platform probe trial was conducted with the pool either in the same position as that used during training (no shift group) or shifted to a new position in the room (shift group). The authors found that rats in the shift group did not search for the platform at its absolute spatial location but rather navigated in the same direction that the platform was located during training and searched at the correct distance from the pool wall, resulting in a search at a location that was never trained. This pattern of results suggests that young rats learn to solve the water task by navigating in a particular direction rather than navigating to a precise place--a finding that may have implications for understanding hippocampal development. The assumption that blockade of long-term potentiation by N-methyl-D-aspartate antagonists interferes with spatial memory was supported by experiments showing that 15 mg/kg ketamine impairs acquisition of navigation to a hidden platform but not to a visible platform. Higher doses were required to impair retrieval of overtrained place navigation. In a working memory version of the task, retrieval latencies were shorter than acquisition latencies with 4- to 15-min but not with 30- and 60-min delays. Latent learning was only effective with the 4-min delay. Ketamine prolonged the initial search of the hidden platform at 3 mg/kg and impaired latent learning but not active acquisition at 1.5–20 mg/kg. Comparison of behavioral and synaptic effects of ketamine suggests that long-term potentiation is a necessary condition, but not a sufficient condition, for acquisition of place navigation, because search strategy and latent place learning are impaired by ketamine doses not interfering with this synaptic phenomenon. Male Long-Evans rats were injected with 32 ng/μl of the N-methyl-D-aspartate (NMDA) receptor antagonist 3-(2-carboxypiperazin-4-yl) propyl-1-phosphonic acid (CPP) or vehicle and trained to locate a hidden platform in a different location (reversal training) than used on the initial 4 days of training. Rats treated with vehicle or CPP into the dorsal hippocampus, basolateral amygdala, or mediodorsal striatum had similar latencies to locate the platform on the reversal day. Rats infused with CPP into the dorsal hippocampus or mediodorsal striatum failed to search preferentially in the novel location during a 24-hr, drug-free retention test, whereas all other groups searched preferentially in this location. Therefore, blocking dorsal hippocampal or mediodorsal striatal NMDA receptors selectively blocked long-term spatial retention without producing short-term performance deficits. Using a radial maze analog task, T. R. Zentall, J. N. Steirn, and P. Jackson-Smith (1990) found evidence that when a delay was interpolated early in a trial, pigeons coded locations retrospectively, but when the delay was interpolated late in the trial, they coded locations prospectively (support for a dual coding hypothesis). In Experiment 1 of the present study, the authors replicated the original finding of dual coding. In Experiments 2 and 3, they used a 2-alternative test procedure that does not require the assumption that pigeons' choice criterion, which changes over the course of the trial, is the same on delay and control trials. Under these conditions, the pigeons no longer showed evidence for dual coding. Instead, there was some evidence that they showed prospective coding, but a more parsimonious account of the results may be that the delay produced a relatively constant decrement in performance at all points of delay interpolation. The original finding of dual coding by Zentall et al. might have been biased by more impulsive choices early in control trials but not in delay trials and by a more stringent choice criterion late in delay trials. The present study provides the 1st report on the effects of selective lesions of the dysgranular portion of the retrosplenial cortex in rats. Excitotoxic lesions of the dysgranular area were sufficient to impair behavior in the radial-arm maze by biasing the strategy used to solve the task. In particular, rats with dysgranular retrosplenial lesions were less reliant on distal visual cues to control performance of a working memory task in the radial-arm maze. Instead, they were more reliant on using a motor turning strategy to solve the task. This change in strategy is consistent with anatomical data showing that the dysgranular region is the primary recipient of visual inputs to the rat retrosplenial cortex. Hippocampal damage impairs navigation with respect to information provided by the shape of an arena. Recent evidence has suggested that normal rats use local geometric information, as opposed to a global geometric representation, to navigate to a correct corner. One implication of this pattern of results is that hippocampal lesions may impair processing of 1 or more of the local geometric features of an environment. The authors therefore investigated the effects of hippocampal cell loss in rats on navigation to a hidden goal with respect to a variety of local cues in an environment with a distinctive shape. Rats with lesions of the hippocampus were impaired in discriminating a right-angled corner from its mirror image. However, they were able to use cues provided by an acute-angled corner (Experiment 1) or a local polarizing cue (Experiment 2). In contrast, lesioned rats were impaired in discriminating long versus short walls (Experiment 3). Results indicate that the hippocampus plays a role in disambiguating locations by processing (metric) information related to the distance between corners. This study examined the effects of disorientation on the acquisition of different spatial reference memory tasks. In an appetitively motivated radial arm maze task in which 1 arm was consistently baited, rats that were disoriented before each trial were impaired in their ability to acquire the task relative to rats placed in a clear container and not disoriented. However, disoriented rats were able to learn a Morris water maze and a water version of the radial arm maze under similar training conditions, suggesting that the effects of disorientation may interact with the quality or quantity of motivation involved in a given task. These results suggest that appetitive and aversive spatial tasks are dissociable, and that any impairment that is due to disorientation is specific to the appetitive radial arm maze task. Nonspatial pretraining (NSP) enables rats to learn the general strategies of the water maze task (WMIs; e.g., learning to swim away from the wall and to climb onto the hidden platform), reduces sensorimotor disturbances, and eliminates acquisition impairments caused by scopolamine hydrobromide, a muscarinic antagonist. To evaluate the contributions of the components of NSP to these effects, NSP was fractionated so that different groups of male rats swam, were placed onto the hidden platform, climbed onto the hidden platform, or were placed into an empty maze before spatial training under scopolamine. No single component of the NSP procedure was sufficient to produce its full effects on sensorimotor disturbances and WMT acquisition. Experience with most or perhaps all of the specific behaviors required in the WMT appears to be important for NSP to produce its full effects. The role of the mediodorsal thalamic nucleus (MD) in spatial memory processes was assessed. Animals were preoperatively trained on an 8-arm maze placed in a visually deprived environment. Following 50 acquisition trials, one group received bilateral electrolytic lesion of the MD thalamus, whereas the other group received sham lesions. On postoperative tests of radial maze performance, MD lesioned animals made significantly more errors, made more errors sooner, and emitted fewer correct responses before making an error than did sham controls. The lesioned subjects also exhibited considerable perseveration immediately postoperation and developed response patterning on postlesion trials. Lesions of the mediodorsal thalamus may fundamentally compromise memory systems and alter ability to respond appropriately in a minimally cued environment. Previous studies have shown double dissociations between win–stay and win–shift radial maze learning in terms of their underlying neural substrates. To examine the content of the associations formed in the two tasks, the authors devalued the food unconditioned stimulus (UCS) by taste aversion to differentiate stimulus–stimulus(CS–UCS) and stimulus–response (CS–CR) learning. UCS devaluation was performed in rats that were over- or undertrained on the win–stay task. Devaluation substantially reduced food consumption on the maze but failed to disrupt choice accuracy, regardless of the amount of training. Devaluation did not affect latency in overtrained rats but did increase latency in undertrained rats. In the win–shift task, devaluation caused rats to reject the reinforcer, yet they continued to accurately win–shift, but with significantly longer latencies (Experiment 3). The results suggest that an S–R association may mediate performance after extended win–stay training. Spatial navigation in the real-world is a complex task that involves many functions, such as landmark identification, orientation, and the calculation of heading vectors. This study uses a 2 × 2 experimental design with fMRI to isolate mnemonic and navigational processes that accompany the calculation of heading vectors. The conditions are based on a working memory version of the Morris water maze task and navigation takes place in a visually austere virtual environment. In an allocentric condition, subjects navigate around a circular arena where there is one small red square on the wall. Each trial begins with an encoding phase in which subjects locate and navigate to a visible coin. Then, in a test phase, after being randomly repositioned, they retrieve the coin when it is invisible. In a control task, there are eight distinct cues around the arena that provide direct cue-place information. Results show significant interaction effects in bilateral posterior parietal cortex, which is compatible with evidence that parietal cortex helps translating between allocentric coordinates and egocentric directions. There was also greater activation for the allocentric task in right posterior hippocampus and left retrosplenial cortex, which could be related to self-localization and orientation. The findings are also compatible with the recent proposal by Kubie and Fenton (2009) that navigation primarily depends on heading vectors between salient places. 102 male Sprague-Dawley rats with selective lesions involving different hippocampal cell fields and/or projections either learned a complex spatial maze postoperatively or were trained preoperatively and tested after the operations. Following damage to anterdorsal CA1 cells and the alveus, acquisition was impaired, but performance was normal when the task was learned preoperatively; postoperative acquisition and performance of the preoperatively learned task were impaired in animals with fimbrial lesions. Data suggest that the CA1 cell field and the projections to the subiculum play an important role in the acquisition of new spatial information but that these connections are not necessary for the utilization of spatial information learned preoperatively. Birds were tested in an open-room radial maze with learned spatial locations that varied from trial to trial (working memory) and locations that remained spatially stable (reference memory). Three of the species, the Clark's nutcracker (Nucifraga columbiana), pinyon jay (Gymnorhinus cyanocephalus), and Western scrub jay (Aphelocoma coerulescens) store food to varying degrees. The other species, the Eurasian jackdaw (Corvus monedula) does not. Pinyon jays and scrub jays performed better than the nutcrackers and jackdaws in both working and reference memory components of the maze. The pinyon jay and jackdaw performed as would be expected on the basis of their natural history and previous research, but the scrub jay and nutcracker did not. Results are consistent with phylogenetic relationships among the 4 species, but could also be explained by differences in response strategies or interference in processing both types of memory components of the maze. For many disorders of the retinal pigment epithelium (RPE) for which there are no effective treatments, transplantation of RPE cells may provide a viable means of restoring function. Using a solvent casting technique, we have manufactured thin films of poly(L-lactic acid) and poly(DL-lactic-co-glycolic acid) 75:25 and 50:50. Non-porous, flexible films with controlled thickness as thin as 12 +/- 3 microns and reproducible surface morphologies and flexural properties were produced. Fetal human RPE cells were found to attach to these substrates when cultured in vitro. The films made using this technique may provide a means of transplanting allogeneic RPE cells as a therapy for a number of ocular diseases related to RPE dysfunction.  相似文献

The spatial memory of adult rats prenatally exposed to cocaine and that of control offspring was assessed using the Morris water maze. Offspring were derived from Sprague Dawley dams that received subcutaneous injection of 40 mg/kg/3 cc cocaine hydrochloride (C40) daily on gestational Days 8-20, pair-fed dams injected with saline, or nontreated control dams. After acquisition, the platform was moved to a new location (reversal phase). Probe trials were conducted at the end of acquisition and reversal training. On the 1st acquisition day, adult male and female offspring prenatally exposed to cocaine required significantly more time and traversed a greater distance to find the hidden platform than did control offspring. Despite these initial differences observed in C40 offspring performance, all of the rats were performing at equivalent levels at the time probe trials were conducted.
