Mountain pine beetles,Dendroctonus ponderosae Hopkins, and California five-spined ips,Ips paraconfusus Lanier, were reared axenically from surface-sterilized eggs on aseptic pine phloem. After 24 hr in host logs, axenip femaleD. ponderosae and maleI. paraconfusus produced the aggregation pheromones,trans-verbenol (D. ponderosae), and ipsenol and ipsdienol (I. paraconfusus). Emergent, axenically reared maleD. ponderosae contained normal amounts of the pheromoneexo-brevicomin. Axenic femaleD. ponderosae treated with juvenile hormone or exposed to vapors of -pinene, produced the pheromonetrans-verbenol. By 25–35 days after eclosion, axenic females exposed to -pinene vapors produced over six times as muchtrans-verbenol as wild females, suggesting that while microorganisms in wild females may producetrans-verbenol, they may also inhibit production of the pheromone or use it as a substrate.Research supported in part by Natural Sciences and Engineering Research Council, Canada, Operating Grants A3881 and A3706, and H.R. Attraction of maleIps paraconfusus to male-infested ponderosa pine logs was inhibited by volatiles from logs infested with mated males and females over an 8-day period in the field. The response of females during this time was not significantly inhibited by these volatiles. Synthesis of the male-specific pheromones, ipsenol and ipsdienol, appeared negligible after 8 days in males allowed to mate with 3 females in these logs while males alone contained levels of these pheromones at about half their maximum rate. The decline in pheromone production in mated males appears to result, at least in part, from a reduction in the activity of the biosynthetic system that converts the host monoterpene, myrcene, to ipsenol and ipsdienol. Mating and feeding have apparently no effect on the biosynthetic system that converts the host monoterpene, (–)--pinene, to the pheromone,cis-verbenol in either males or females. The reduced production and release of pheromones by males after mating appears to play a major function in the process of terminating the aggregation phase of host colonization.  相似文献

MaleIps paraconfusus Lanier bored and fed in cellulose powder substrates treated with solvent extracts of ponderosa pine phloem in preference to cellulose powder alone. Stimuli that elicit boring and feeding behavior occurred in the water extracts and the combined solvent extracts. No significant boring or feeding occurred in the methanol extract. There was a preference for, but no significant feeding in, the water partition of the ether extract. The parasitic dwarf mistletoeArceuthobium vaginatum attacksPinus ponderosa as its primary host andP. contorta as an occasional host. Within ponderosa pine stands there is also differential parasitism among individual trees. We compared biochemical features of phloem and xylem oleoresin between infected individuals of the two pine species (N=15 for each species) and also between infected (N=30) and nearby uninfected (N=30) ponderosa pine conspecifics. There were significant differences in chemical features, both at the interspecific (P. ponderosa vs.P. contorta) and intraspecific (P. ponderosa) levels. Discriminant function analysis based on chemical features of phloem correctly classified all trees used in the analysis as eitherP. ponderosa orP. contorta, and 95% of all ponderosa pine trees as either parasitized or nonparasitized. Monoterpene composition of oleoresin was distinct between species, and differences between parasitized and nonparasitizedP. ponderosa were also significant. Many of the observed chemical differences are probably constitutive, although levels of nonstructural carbohydrates and -pinene may change in response to dwarf mistletoe infection. Biochemical differences at the intraspecific level were distinct from interspecific differences. Patterns of differential attack can have genetic consequences upon both the parasite and its hosts, and, in the process, may contribute to the evolution of host races of the parasite and to the evolution of host resistance within ponderosa pine.  相似文献

Linalool, a compound from maleI. pini, previously suggested as an inhibitor forI. paraconfusus, has no obvious effect on the response ofI. paraconfusus to ponderosa pine bolts containing maleI. paraconfusus. I. pini from California and New York equally inhibit the response ofI. paraconfusus to maleI. paraconfusus. Ipsenol, one component of the attractant pheromone ofIps paraconfusus, inhibits attacks byIps pini on ponderosa pine logs baited with maleI. pini. MaleI. paraconfusus confined to artificial entrance tunnels in white fir logs produced the pheromone compounds ipsenol and ipsdienol in their hindguts. The hindguts were attractive to females in a laboratory olfactometer and the male infested logs were attractive in field bioassay. The amount of pheromones produced and the amount of feeding and boring activity is much less in white fir than in ponderosa pine. This study characterized the chemical response of healthy red pine to artificial inoculation with the bark beetle-vectored fungusLeptographium terebrantis. In addition, we sought to determine whether stress altered this induced response and to understand the implications of these interactions to the study of decline diseases. Twenty-five-year-old trees responded to mechanical wounding or inoculation withL. terebrantis by producing resinous reaction lesions in the phloem. Aseptically wounded and wound-inoculated phloem contained higher concentrations of phenolics than did constitutive tissue. Trees inoculated withL. terebrantis also contained higher concentrations of six monoterpenes,-pinene,-pinene, 3-carene, limonene, camphene, and myrcene, and higher total monoterpenes than did trees that were mechanically wounded or left unwounded. Concentrations of these monoterpenes increased with time after inoculation. Total phenolic concentrations in unwounded stem tissue did not differ between healthy and root-diseased trees. Likewise, constitutive monoterpene concentrations in stem phloem were similar between healthy and root-diseased trees. However, when stem phloem tissue was challenged with fungal inoculations, reaction tissue from root-diseased trees contained lower concentrations of-pinene, the predominant monoterpene in red pine, than did reaction tissue from healthy trees. Seedlings stressed by exposure to low light levels exhibited less extensive induced chemical changes when challenge inoculated withL. terebrantis than did seedlings growing under higher light. Stem phloem tissue in these seedlings contained lower concentrations of-pinene than did nonstressed seedlings also challenge inoculated withL. terebrantis. It is hypothesized that monoterpenes and phenolics play a role in the defensive response of red pine against insect-fungal attack, that stress may predispose red pine to attack by insect-fungal complexes, and that such interactions are involved in red pine decline disease. The antennal response ofDendroctonus valens to host monoterpenes from the resin of ponderosa pine was studied using the electroantennogram (EAG) technique. Male and female beetles were given a single dose of each of 11 different monoterpenes. Response amplitude to the different compounds did not vary between sexes and was generally well correlated with results from field attraction studies. Response to (S)-(–)--pinene was greatest. The relative amplitude of the responses to the (R)-(+) and (S)-(–) enantiomers of a-pinene, however, were reversed from their relative attractiveness in the field. A dose-response study was conducted for the (R)-(+) and (S) -(–) enantiomers of a-pinene, plus a reciprocal differential saturation test with successive doses of first one enantiomer of-pinene and then the other. Comparison of EAG traces suggests different receptors for the two stereoisomers of-pinene. Differential saturation curves suggest that while one set of receptors may respond to both enantiomers, some receptors respond only to the (S)-(–) enantiomer.  相似文献

When yeasts associated withIps typographus beetles were grown in an aqueous phloem medium for two days, the main oxygenated monoterpenes produced were -terpineol and borneol. Terpinene-4-ol, myrtenol, andtrans-pinocarveol were also found but in lesser amounts. Of the six strains used in this study,Hansenula capsulata andCandida nitratophila produced the largest amounts of oxygenated monoterpenes. Addition of -pinene to the phloem medium generally reduced the amounts of oxygenated monoterpenes, probably because this substance is toxic to all tested yeast species. OurCandida diddensii strain seemed to be particularly sensitive to -pinene. None of the yeast strains producedcis-verbenol,trans-verbenol, or verbenone from the medium or from added -pinene.This study was made within the project Odour Signals for Control of Pest Insects.  相似文献

Concentrations of soluble phenolics and lignin in the phloem of ponderosa pines inoculated with the pathogen Heterobasidion annosum were assessed over a period of 2 years in a 35-year-old plantation in northern California, USA. The major effect of the pathogen on phloem-soluble phenolics consisted of a significant accumulation of ferulicacid glucoside: 503 ± 27 g/g fresh weight (FW), compared with 366 ± 26 g/g FW for mock-treated and 386 ± 27 g/g FW for control trees. Lignin content was negatively correlated with ferulic acid glucoside concentration, and there was an indication of lignin reduction in the cell walls of inoculated trees. Lignin had a negative effect on the in vitro growth of two common bark beetle fungal associates, Ceratocystiopsis brevicomi and Ophiostoma minus. For this reason it, is hypothesized that lower lignification may facilitate the growth of beetle-associated fungi, resulting in greater susceptibility of the presymptomatic host to bark beetle colonization.  相似文献

Ponderosa pine logs infested withIps paraconfusus males inhibited the attraction ofDendroctonus brevicomis in the field to either attractive logs cut from a ponderosa pine tree under attack byD. brevicomis or to their synthetic pheromones,exo-brevicomin, frontalin, and myrcene. Logs cut from trees under attack byD. brevicomis inhibited the response ofI. paraconfusus to logs infested with maleI. paraconfusus.Exo-brevicomin, frontalin, and myrcene did not inhibit their response but verbenone did. Verbenone was found in maleD. brevicomis dissected from attractive logs under attack during the same time the response ofI. paraconfusus was inhibited by these logs.Trans-verbenol andexo-brevicomin were found in femaleD. brevicomis while verbenone,trans-verbenol, and frontalin were found in maleD. brevicomis in relatively large amounts near the beginning of the aggregation phase of host colonization. All of these compounds had decreased at a similar rate 5 days later. This gradual decrease inexo-brevicomin and frontalin probably caused the observed reduction in attraction. The ecological significance of these compounds in relation to termination of the aggregation phase ofD. brevicomis and reduction of interspecific competition is discussed.Coleoptera: Scolytidae. The exotic longhorn beetle Arhopalus tristisis a pest of pines, particularly those damaged by fire, and a major export quarantine issue in New Zealand. Actinograph recordings of caged individuals showed that males and females were most active from dusk to midnight. Olfactometer experiments indicated that females moved upwind toward odors from burnt pine (80%, N= 75), compared to unburnt pine (20%). Oviposition choice tests showed that eggs were predominantly laid on burnt logs (79%, N= 20), compared to unburnt logs. Beetles were trapped by funnel traps baited with burnt (mean catch per trap 7.8) and unburnt (mean catch 4.1 per trap) pine bark from inside a screen cage (4 × 3 m), while unbaited traps had a mean catch 0.1 beetles (N= 8 replicates). The treatment of burnt pine bark with a 1:1:2 mixture of green leaf volatiles (E)-2-hexen-1-ol and (E)-2-hexenal) in mineral oil as a repellent reduced trap catch by fivefold in a similar experiment (mean catches of 1.2 beetles per trap to burnt pine bark plus repellent treatment and 6.2 beetles per trap to burnt pine bark alone). The treatment of burnt pine bark with this solution also reduced oviposition by 98.5% (mean eggs per log of 11.1 on burnt pine and 0.3 on burnt pine plus repellent), indicating that oviposition cues have the potential to be significantly disrupted. The electrophysiological responses of adult beetles were recorded to a range of odorants. Normalized responses to monoterpenes known to occur in Pinus radiataranged from about 20 to about 150, with -terpineol giving the greatest responses in both sexes. Green leaf volatiles also gave high responses. The potential exists to improve the management of this insect using chemical cues in various ways.  相似文献

The effects of host tree monoterpenes on primary and secondary attraction of the bark beetles, Ips pini and Ips grandicollis, and their associated phloeophagous insects were investigated. Monoterpenes alone were not attractive to I. pini. However, monoterpenes mediated the attraction of I. pini to its aggregation pheromones. With the exception of 3–carene, the effect of monoterpenes on I. pini's response to its pheromone, ipsdienol plus lanierone, was inhibitory. In contrast, (–)--pinene both attracted I. grandicollis and enhanced the attraction of I. grandicollis to its pheromone, ipsenol. No monoterpene inhibited the response of I. grandicollis to its pheromone. The inhibitory effect of host monoterpenes on I. pini response to its aggregation pheromone differs from previous work, in which monoterpenes either synergized responses or had no effect. In addition to possible geographic differences, the concentrations used in our study simulated trees that had begun to respond to attack, whereas previous studies deployed lower concentrations, which simulated constitutive phloem from unattacked trees. These results support the view that trees that undergo induced responses to bark beetles sometimes inhibit attraction of additional beetles, despite the beetles' production of aggregation pheromones. Neither species displayed cross-attraction to the pheromone of the other. The red turpentine beetle, Dendroctonus valens, showed weak and consistent attraction to (+)--pinene and in some cases to (–)--pinene. Attraction to (–)--pinene was usually enhanced by Ips spp. pheromones. The absence of strong attraction to (+)--pinene and partial attraction to (–)--pinene suggest that the effects of different stereoisomers of -pinene on D. valens vary throughout its geographical range. Hylastes porculus was also attracted to some monoterpenes, particularly (–)--pinene. An additional 10 species of phloeophagous insects were caught in response to monoterpenes and/or pheromones, including the pine root weevils, Hylobius pales and Pachylobius picivorus, and the longhorned beetle, Monochamus carolinensis.  相似文献

This study investigated the influence of the plant antifeedants, pinosylvin and pinosylvin methyl ether (PME), on suppression of feeding by snowshoe hares (Lepus americanus). Screening bioassays showed clearly that pinosylvin suppressed feeding by hares when sprayed directly on coniferous tree seedlings. Pinosylvin odor, when associated with food (but not mixed with it), also significantly reduced consumption of rabbit chow by hares. Large pen bioassays indicated that both pinosylvin and PME significantly reduced feeding on tree seedlings by hares when the antifeedants were sprayed directly on trees. In field bioassays near Prince George, British Columbia, Canada, pinosylvin sprayed on trees and encapsulated in controlled-release devices of PVC plastic, with an internal wire as a twist-tie for attachment to tree, significantly reduced feeding by hares. Thus, pinosylvin will generate an avoidance response in terms of feeding by snowshoe hares. This response is likely triggered by an olfactory pathway based on positive results with controlledrelease devices attached to seedlings. Our study reports the first practical utilization of plant antifeedants for forest crop protection and wildlife management.  相似文献

We surveyed variation in both the concentration and composition of monoterpenes in six tissues within individuals of ponderosa pine to determine (1) whether variation exists between different tissues; (2) whether variation occurs between samples from the north and south sides of the tree within a tissue, and (3) whether composition of one tissue is correlated with that of others. Wood, roots, and resin exuded from the trunk have similar monoterpene compositions within trees. Phloem and cones contain a higher proportion of -pinene with less carene than resin, while needles are distinguished by high proportions of -pinene. Samples of the same tissue taken from the north and south sides of the tree differed little and were strongly correlated. Correlations were moderate across tissues within the groups sharing similar monoterpene compositions (e.g., phloem vs. cones), but correlations between tissues in different groups were weak (e.g., phloem vs. needles).  相似文献

Short-range attraction/feeding stimulation of male Mediterranean fruit flies [Ceratitis capitata (Wiedemann), (Diptera: Tephritidae)] to a stem extract of a host plant,Litchi chinensis Sonn. (Sapindaceae), and to milky exudates from stems of nonhost plants,Ficus retusa L. andF. benjamina L. (Moraceae), were attributed to the presence of the sesquiterpene -copaene. The presence of -copaene in the milky exudate from stems ofF. benghalensis L. is also suggested as eliciting similar behavioral responses in male medflies. The presence of minor quantities of -ylangene in the plants and its contributory effects to the behavioral response of male medflies is discussed. Short-range attraction/feeding stimulation of male medflies to equal amounts of -ylangene-free -copaene samples (94.5%+), prepared from -copaene-enriched angelica seed oil and copaiba oil, respectively, showed no difference in intensity of response. -Ylangene elicited a slightly less intense response for male medflies than -copaene.Names of products in this paper are included for the benefit of the reader and do not imply endorsement or preferential treatment by USDA.  相似文献

The influence of mustelid (weasel) anal-gland compounds (semiochemicals) and a plant antifeedant (pinosylvin) on snowshoe hare (Lepus americanus) and meadow vole (Microtus pennsylvanicus) feeding damage was investigated. This study has shown conclusively that when applied to spring-planted lodgepole pine seedlings, pinosylvin is effective in significantly reducing feeding damage. Seedlings treated with pinosylvin suffered damage levels of 2.3% and 21.2% compared to the controls with damage levels of 24.5% and 41.0%. Weasel scent was found to reduce damage levels, but the reduction was not statistically significant in all blocks.  相似文献

In order to compare the metabolism of 1,8-cineole in the pyrgo beetle, Paropsisterna tigrina, three other herbivorous insect species, Faex nigroconspersa, Chrysophtharta bimaculata, and Oxyops vitiosa, were fed 1,8-cineole leaf diets. F. nigroconspersa adults excreted predominantly 9-hydroxy-1,8-cineole (36.2% of the volatile constituents) with some 2-hydroxy-1,8-cineole (11.4%). In contrast, larvae excreted predominantly 2-hydroxy-1,8-cineole (27.4%) and smaller proportions of 9-hydroxy-1,8-cineole (5.2%) and 3-hydroxy-1,8-cineole (4.3%). C. bimaculata adults excreted predominantly 3-hydroxy-1,8-cineole (16.5%). Oxyops vitiosa adults, on a lower 1,8-cineole diet, excreted predominantly 2,9-dihydroxy-1,8-cineole (4.2%) and 2-hydroxy-1,8-cineole (3.5%), with smaller proportions of 3-hydroxy-1,8-cineole (1.1%) and 9-hydroxy-1,8-cineole (0.5%). This is the first reported occurrence of a dihydroxycineole as an insect metabolite. Gas chromatographic and mass spectral data for hydroxycineoles are recorded and interspecific metabolite variation discussed.  相似文献
