Nimbin and salannin, major triterpenoids accompanying azadirachtin in extracts of neem (Azadirachta indica) seeds, were photooxidized by UV light in the presence of oxygen to more polar, unstable intermediates that rearranged on silica gel to two final products in which the furan ring had been oxidized to isomeric hydroxybutenolides. The isomeric hydroxybutenolides were also readily formed when a crude extract of triterpenoids from neem seeds was irradiated, and both isomers of salannin have been isolated from seeds. Photooxidation of nimbin and salannin proceeded much faster than that of azadirachtin. All photoproducts showed some biological activity againstSpodoptera littoralis, Locusta migratoria, andSchistocerca gregaria. Isonimbinolide was as potent as azadirachtin at inhibiting feeding in all three species, and it also inhibited the growth ofS. littoralis. Isosalanninolide showed potent antifeedant and growth inhibitory activity againstS. littoralis.  相似文献

Eleven polyhydroxy alkaloids of plant origin were tested for anti-feedant effects against larvae of the lepidopteransSpodoptera littoralis, Spodoptera frugiperda, Heliothis virescens, andHelicoverpa armigera. Data from behavioral and electrophysiological investigations were correlated to reveal information on the mode of action of the antifeedants. The pyrrolidine DMDP was an effective antifeedant for all four species, whereas the piperidines fagomine and XZ-1 and the pyrrolizidine alexine were all ineffective as antifeedants. The activity of the pyrrolidines CYB-3 and DAB-1, the piperidines DNJ, DMJ, and BR1, and the bicyclic octahydroindolizine castanospermine varied among species. The investigation focuses on the structural similarities between some of the alkaloids and some common phagostimulatory sugars and illustrates a neural interaction involving the neurons that are differentially responsive to alkaloids and sugars. InS. littoralis, the neurons responding specifically to the alkaloids DMDP, DAB-1, and castanospermine and to the sugars fructose, sucrose, and glucose are more active when the compounds are applied singly than when an alkaloid and a sugar are applied together. The implications for the occurrence and functioning of different sugar receptor sites are discussed.  相似文献

Bioassay-guided isolation of bioactive natural substances requires monitoring of all fractionation and purification steps using a bioassay system. This is often a long and tedious process, especially with insect feeding bioassays. We report here a new method, based on the principle of bioautography, for a quick isolation of insect antifeedant compounds. TLC plates, after development, are coated with a thin layer of artificial diet and fed toSpodoptera litura larvae. The location of uneaten areas is then compared with theR fvalues of the TLC spots, in order to determine rapidly the active fractions. This methods allows for a very fast determination of the most active antifeedant compounds in a complex mixture and considerably speeds up the isolation process. This new method was successfully applied in the study of antifeedant activity of several plant samples, and results are presented here for a model plant,Skimmia japonica (Rutaceae). Using this new method, the compounds responsible for the feeding-deterrent activity, three furanocoumarins (bergapten, xanthotoxin, and oxypeucedanin), were quickly and efficiently identified.  相似文献

The tetranortriterpene, meliatoxin A2, induced a significant level of antifeedant activity inSpodoptera litura F. larvae, while meliatoxin B1, which lacks the ring D epoxide, was much less active. Nothofagus species from Chile and New Zealand were surveyed in the field for invertebrate abundance and leaf feeding damage and in the laboratory for antifeedant activity against leafrollers (Ctenopsteustis obliquana, Epiphyas postvittana), deterrent activity against pea aphid (Acyrthosiphon pisum), insect growth regulatory activity (Oncopeltus fasciatus), nematicidal activity (Caenorhabditis elegans), antibiotic activity (Pseudomonas solanaciarium), and general toxicity. A data matrix indicated that N. alessandri and N. pumilio most likely have a chemical barrier to insect attack as leaves showed low faunal abundance, low herbivory, and activity in the leafroller antifeedant, aphid deterrent, and nematicidal assays. A chemical examination of N. alessandri that used the leafroller antifeedant test to guide the separation yielded an active fraction containing the flavonoid, galangin, and the stilbene, pinosylvin, which appear to act in concert to deter leafroller feeding. The discovery of the phytoalexin, pinosylvin, in the leaves, raises the possibility that Nothofagus in general, and N. alessandri in particular, may have induced chemical defense mechanisms.  相似文献

Linden (Tilia cordata) bark was shown to contain an antifeedant effective against the large pine weevil, Hylobius abietis. Soxhlet extraction of inner and outer bark resulted in an extract that showed antifeedant activity in a microfeeding assay. The extract was fractionated by chromatography on silica gel using gradient elution with solvents of increasing polarity. The content of the fractions obtained was monitored by thin layer- and gas chromatography. Fractions of similar chemical composition were merged. Two of the 17 fractions showed antifeedant activity in the microfeeding assay. Nonanoic acid was identified in both of these fractions. Subsequent testing in the microfeeding assay showed that nonanoic acid possessed strong antifeedant activity against H. abietis adults.  相似文献

Bitter gourd, Momordica charantia, was less palatable to two species of armyworms, Spodoptera litura and Pseudaletia separata, than two other cucurbitaceous plants. A methanol extract of M. charantia leaves inhibited feeding of the armyworm larvae. The two most active fractions obtained by silica gel chromatography were purified by HPLC. Momordicine II, a triterpene monoglucoside, was identified as an antifeedant compound from the more active of these fractions. The second active fraction led to the isolation of a new triterpene diglucoside. Fresh leaves of M. charantia contained ca. 0.3% of momordicine II. Momordicine II showed significant antifeedant effects on P. separata at concentrations of 0.02, 0.1, and 0.5% in artificial diets. Momordicine II caused a significant feeding reduction in S. litura only at the highest concentration (0.5%) tested. The difference in feeding response of the two armyworms to momordicine II may be related to the diversity in their host range.  相似文献

S. latifascia andS. descoinsi are closely related species that occur sympatrically over limited areas in French Guiana. We examined allopatric populations,S. latifascia originating from Barbados andS. descoinsi from French Guiana. Studies on nocturnal activity cycles showed temporal partitioning of female calling behavior, male sexual activity, and mating behavior.S. descoinsi were sexually active in the first half of the scotophase whereasS. latifascia were sexually active in the second half. Seven compounds (Z9–14: Ac,Z9,E12–14: Ac,Z11–16: Ac,E9,E12–14: Ac,Z9–14: Ald,Z9,E11–14: Ac andZ11–14: Ac) were identified in females of bothS. latifascia andS. descoinsi extracts.Z9–14: Ac was a main pheromone component for the two species. The major difference between the pheromones ofS. latifascia andS. descoinsi was the proportion ofZ9,E12–14: Ac in the extracts: 7% forS. latifascia and 42% forS. descoinsi. The proportion ofZ9,E12–14: Ac relative to the sum ofZ9–14: Ac andZ9,E12–14: Ac in individual gland extracts was 4±1% (mean ± standard deviation) forS. latifascia and 44.8±6% forS. descoinsi. Electrophysiological studies showed no major differences between species in the morphology and physiology of the pheromone receptors of males. Receptors were identified forZ9–14: Ac andZ9,E12–14: Ac, but no receptor was found for the other compounds. In the wind tunnel, synthetic blends withZ9–14: Ac andZ9,E12–14: Ac gave the same behavioral responses as conspecific female extracts for the males of the two species. Some cross-attraction was observed with synthetic blends and female extracts. Nethertheless, previous field trapping experiments in French Guiana were species-specific and suggested differences in the attractivity of males. In the laboratory,S. latifascia andS. descoinsi could hybridize in both reciprocal crosses. FemaleS. descoinsi × maleS. latifascia mating rate was significantly lower than for the reciprocal cross, and 26.7% of femaleS. descoinsi could not separate from maleS. latifascia after mating. These copulatory problems may involve genital incompatibilities between males and females. Several barriers against interbreeding betweenS. latifascia andS. descoinsi seem to combine including differences in nocturnal activity cycles, pheromone differences, and genital barriers. The study of sympatric populations will be necessary to define the role of sex pheromones in the reproductive isolation ofS. latifascia andS. descoinsi.  相似文献

Green ash is one of the few tree species rejected as food by larvae of the generalist gypsy moth,Lymantria dispar L. (Lepidoptera: Lymantriidae). Such rejection is based especially on chemicals present in green ash foliage. The gypsy moth larval feeding-inhibitory activity is contained in the ethyl acetate extractables of green ash foliage. Three representative columnchromatographed fractions of the extractables contained antifeedant activity. Individual fractions showed weaker antifeedant activity compared to the total ethyl acetate extractables. Acid hydrolysis of the extractables destroyed antifeedant activity and yielded feeding stimulant activity at higher concentrations. The aqueous extractables were not feeding-inhibitory. Compounds in the green ash extractables were separated by TLC, HPLC, CC, and GC.  相似文献

A bioassay-guided fractionation of the aerial parts ofSenecio palmensis resulted in the isolation of two sesquiterpenes, 2,10-bisaboladien-1-one and 11-acetoxy-5-angeloyloxy-silphinen-3-one. The bisabolene and the silphinene represented 0.012% and 0.024% of the plant dry weight, respectively. Both compounds showed antifeedant activity againstLeptinotarsa decemlineata larvae and adults in short-term choice and no-choice bioassays. Both compounds were also tested against different species of phytopathogenic fungi. The beetles were more sensitive to these compounds in choice than in no-choice assays, with a gradient of increasing sensitivity from second instars to adults. Bisabolene was 45 times less active as an antifeedant than juglone, which was tested as a positive control. The silphinen was more active than the bisabolene, with a range of activity similar to juglone. Furthermore, exposure of fourth instars to these compounds over a 24-hr period resulted in reduced feeding and growth rates. To distinguish between antifeedant and toxic effects, growth efficiencies were calculated as the slope of the regression of relative growth rate on relative consumption rate. The comparison of these results with those of antifeedant simulation and contact toxicity bioassays indicates that feeding inhibition is the primary mode of action of the bisabolene, while the silphinene shows both antifeedant and toxic effects.  相似文献

The antifeedant activity of 13 quassinoids of different structural types has been studied against the Mexican bean beetle (Epilachna varivestis Mulsant) 4th instar larvae and the southern armyworm (Spodoptera eridania Crawer) 5th instar larvae. All quassinoids tested displayed significant activity against the Mexican bean beetle and, thus, do not reveal a simple structure-activity relationship. Five quassinoids were active against the southern armyworm. Interestingly, four of these-bruceantin (I), glaucarubinone (VI), isobruceine A (VIII), and simalikalactone D (XI)-possess the required structural features for antineoplastic activity. Treatment of potato leaf disks with fourneo-clerodane diterpenes fromTeucrium (eriocephalin, teucrin-A, teucvin, and teuscorolide) significantly reduced feeding by larvae ofLeptinotarsa decemlineata (Colorado potato beetle). Choice and no-choice tests suggest that teuscorolide acts as a feeding deterrent, whereas the antifeedant activity of teucrin-A, teucvin, and eriocephalin is likely associated with a toxic mode of action. Nutritional tests, antifeedant simulation assays, and posttreatment studies confirmed that teucrin-A can be categorized by its mode of action as a toxin, rather than as a feeding deterrent.  相似文献

Ethanolic extract of aerial parts of Artemisia annua L. and artemisinin were evaluated as anti-insect products. In a feeding deterrence assay on Epilachna paenulata Germ (Coleoptera: Coccinellidae) larvae, complete feeding rejection was observed at an extract concentration of 1.5 mg/cm2 on pumpkin leaf tissue. The same concentration produced a feeding inhibition of 87% in Spodoptera eridania (Cramer) (Lepidoptera: Noctuidae). In a no-choice assay, both species ate less and gained less weight when fed on leaves treated with the extract. Complete mortality in E. paenulata and 50% mortality in S. eridania were observed with extract at 1.5 mg/cm2. Artemisinin exhibited a moderate antifeedant effect on E. paenulata and S. eridania at 0.03–0.375 mg/cm2. However, a strong effect on survival and body weight was observed when E. paenulata larvae were forced to feed on leaves treated at 0.03 and 0.075 mg/cm2. Artemisia annua ethanolic extract of aerial parts at 1.5 mg/cm2 showed no phytotoxic effect on pumpkin seedlings.  相似文献

The leaf extract fromS. berthaultii Hawkes (PI 473340) contains exudate from trichomes (type B). Consumption ofS. tuberosum var. Norchip foliage by the Colorado potato beetleLeptinotarsa decemlineata (Say) was reduced when treated with the leaf extract. The leaf extract from a resistantS. berthaultii clone without type B trichome had no antifeedant activity. It suggests that more than one mechanism of resistance to the Colorado potato beetle exists inS. berthaultii.  相似文献

A systematic examination was made of the plant family Apiaceae (Umbelliferae) in which extracts of 33 species, representing 32 genera, were screened for antifeedant activity against the field slug Deroceras reticulatum by using an electrophysiological recording assay. In this assay, the olfactory sensory epithelium of the posterior tentacle of the slug was exposed to volatile components of the plant extracts presented in an airstream, and any subsequent activity of the olfactory nerve was recorded. Extracts of 22 species elicited a range of nervous activity in the preparation. A feeding bioassay was used to measure any change in consumption when extracts were added to a standard food. Statistical analysis of data obtained from both electrophysiological traces and the feeding bioassays identified extracts of Petroselinum crispum, Conium maculatum, and Coriandrum sativum as being the most neuroactive as well as the most antifeedant.  相似文献

The phagodepression activity of five furanocoumarins (FC), bergapten, xanthotoxin, psoralen, imperatorin, and angelicin, has been studied against larvae of Egyptian cotton leafworm Spodoptera littoralis (Boisduval) (Lepidoptera: Noctuidae) using a leaf-disk choice bioassay. The dose range used was 0–10 g/cm2 for linear furocoumarins and 0–30 g/cm2 for angelicin, the angular furanocoumarin. Dose–feeding deterrency activity correlations were governed by various sigmoidal functions, except in the case of imperatorin and bergapten, which had dose–response curves showing irregular traces with two maxima. All five FCs induced various degrees of phagodepression in S. littoralis; at 3 g/cm2, the relative feeding deterrence was bergapten = xanthotoxin > psoralen = imperatorin = angelicin. Structure–activity correlations indicated that a methoxy group on C-5 or C-8 enhanced the activity. Comparison of experimental feeding deterrence of binary mixtures of imperatorin with xanthotoxin, bergapten, or psoralen at various relative concentrations with the calculated additive activity of each combination indicated that a proportion of 40% or more of imperatorin may exert a greater antifeedant effect on S. littoralis than the sum of individual compounds. The effect of angelicin also was examined in combination with psoralen, both of which are present in Psoralea plants. Their mixtures yielded a clear synergistic effect in the 20–80% angelicin range (coactivity coefficient = 124–133). By contrast, the effect of angelicin on xanthotoxin was only additive. In the dose–response curves of imperatorin/bergapten combinations, synergism was found at >60% imperatorin relative concentration and 1 g/cm2, whereas lower proportions led to antagonism. The threshold between the two opposing effects was found to depend on the total FC concentration employed. Some natural systems contain FCs in the range of synergistic proportions recorded here and, thus, may have been produced by the host to increase its defensive effect at a lower metabolic cost.  相似文献

Hexane and ethanol extracts of seeds from 22 species of plants of the family Meliaceae from a number of countries were prepared. The extracts were submitted to antifeedant and toxicity bioassays utilizing fall armyworm [Spodoptera frugiperda (J.E. Smith)] (Lepidoptera: Noctuidae) larvae and striped cucumber beetle [Acalymma vittatum (F.)] (Coleoptera: Chrysomelidae) adults. Toxicity tests were also performed with brine shrimp,Anemia salina Leach. Feeding inhibition and mortality produced by some of these extracts were comparable to and, in certain cases, slightly greater than the effects produced by comparable neem (Azadiracta indica A. Juss.) seed preparations. Essential oils were extracted fromMajorana syriaca, Satureja thymbra, Micromeria fruticosa, andSalvia triloba, and their volatile fractions were tested for their antifungal activity against the soil-borne pathogensFusarium oxysporum andMacrophomina phaseolina and the foliar pathogensBotrytis cinerea andExserohilum turcicum. Results showed a fungistatic effect of 1, 2.5, and 5l of the various essential oils on fungal mycelium growth. Females of severalAttagenus species demonstrated calling behaviors similar to those previously reported forA. elongatulus. Attagenus rufipennis females did not call until they were 12–14 days old. OtherAttagenus species called at approx. 4 days of age. All species exhibited diurnal periods of calling activity. Interspecific pheromone responses were tested, withA. megatoma megatoma, A. megatoma canadensis, A. megatoma japonicus, andA. schaefferi spurcus showing equal cross-responses. Structure–dose–feeding deterrency relationships were compared between the Colorado potato beetle, Leptinotarsa decemlineata (Say), and the western corn rootworm, Diabrotica virgifera virgifera LeConte, using 15 alkaloids, terpenoids, and phenolic derivatives. The former species, a specialist herbivore on selected alkaloid-rich Solanaceae species, was on average 100-times less sensitive to the antifeedant effects of alkaloids, but more similarly sensitive to the terpenoids and phenolics than the latter species, a generalist flower herbivore predominantly on Graminae, Cucurbitaceae, and Compositae species. Antifeedant ED50 values for the potato beetle and corn rootworm, each from closely related subfamilies of Chrysomelidae, ranged over four orders of dose magnitude among the 15 compounds with major species differences in stereosensitivity to -hydrastines and analog sensitivity with the silphinenes. Extremes in sensitivity ranged from silphinene, a rare tricyclic sesquiterpene that is 53 times more active on the potato beetle to aconitine, which is 430 times more antifeedant to the corn rootworm. Among silphinene and its two hydrolysis derivatives, there was not a strong correlation between antifeedant potency and injected toxicity for the two beetle species, but there was correlation between behavioral activity and galeal taste cell electrophysiological threshold and frequency responses. That all of the established GABA- and glycinergic compounds tested were antifeedant for both species suggests a shared molecular mechanism for antifeedant taste chemoreception in these divergent Chrysomelidae species. Moreover, the wide differences in antifeedant sensitivities among these and other chrysomelids to a suite of ligand-gated ion channel antagonists implicate a common protein neuroreceptor type with extraordinary heterogeneity in beetle taste.  相似文献
