The standard activity-based anorexia procedure provides rats with access to a running wheel while restricting their access to dry food. This can produce reduced food intake and progressive weight loss. Using this procedure, in the present study (Experiment 1) the authors found changes in drinking patterns both in the period of high activity preceding food access and during the feeding period. Varying the procedure by providing wet mash (Experiment 2) or by prior adaptation to a drinking schedule (Experiment 3) prevented the self-starvation effect. Access to a running wheel combined with restricted feeding produced body weight loss at an equivalent rate in male and female litter-mate rats (Experiment 1). Thus, despite weighing less and running more, females were not more vulnerable to this procedure. When factors influencing weight loss were varied, no sex difference was found in adaptation to a new feeding schedule or in the effect of single versus group housing (Experiment 2). The apparent critical difference was that body weight loss increased running in males but not in females (Experiment 3). In all rats, rapid recovery of body weight occurred when food access was no longer restricted (Experiment 1), suggesting that "activity-based anorexia" is a misnomer for weight loss by rats in a running wheel. In 3 experiments rats given 8 sessions of preexposure to wheel running acquired a preference for a flavor that was given immediately after each of 4 subsequent sessions of wheel running. Such flavor preference was less likely when rats were given the same conditioning procedure but without preexposure to wheels (Experiment 1) or when access to flavor was delayed by 30 min following a wheel session (Experiment 2). When rats were given a flavor before each wheel session, the resulting conditioned aversion was greater in rats that had no prior exposure to wheel running (Experiment 3). These results show that whether an aversion or preference for a flavor is produced by wheel running depends on an interaction between prior wheel experience and the sequence of events. Anorexia has been related to reduced activity of the paraventricular hypothalamic (PVN) noradrenergic-feeding system. This study attempted to determine whether clonidine (an α2-adrenergic agonist) infused into the PVN reduced susceptibility to activity-based anorexia (ABA) in the rat. In Exp 1, clonidine (6 doses) was chronically infused into the PVN of male Sprague-Dawley rats. All animals were exposed to ABA (1.5 hrs/day food access; 22.5 hrs/day running wheel access) until a 25% body weight loss was reached. Dose-related increases in susceptibility to ABA and decreases in food intake were observed. In Exp 2, for which heavier animals and 3 doses of clonidine were used, no difference was found in food intake and wheel activity, but there was increased susceptibility to ABA. Chronic clonidine infused into the PVN does not produce hyperphagia and exacerbates rather than attenuates susceptibility to ABA. Running activates the hypothalamic–pituitary–adrenal (HPA) axis, increasing the release of stress hormones known to exert anorexic effects. HPA axis reactivity is strongly influenced by early postnatal manipulations, including removal of pups from the dam for short (handling) or prolonged (maternal separation) durations during the preweaning period. The authors examined the effects of handling and maternal separation on food intake, body weight loss, and running rates of young adult male and female rats in the activity-based anorexia (ABA) paradigm. Postnatal treatment did not affect adaptation to a 1-hr restricted feeding schedule before the introduction of wheel running. During the ABA paradigm, maternally separated animals lost weight faster, ate less, ran more, and required fewer days to reach removal criterion compared with handled rats. The effects of 0-, 2-, and 24-hr wheel access on the pattern of running, feeding, and weight were explored over 24 days in 3 groups of 8 male rats. Both 2 and 24 hr of wheel access suppressed feeding by about 15% for about 8 days before feeding gradually returned to normal. Weight in these 2 groups was similar and was suppressed for the 24 days. Like the pattern seen with drug self-administration (S. H. Ahmed & G. F. Koob, 1998, 1999), running levels stayed low with short, 2-hr daytime wheel access, but with long, 24-hr access, rats' running escalated over days to chronically high levels. Two studies examined whether brief access to sucrose solutions engages food-entrainable rhythms when pitted against a light-dark cycle. In Experiment 1, 32% sucrose was given once daily for 5-6 min at Zeitgeber Time 4 (ZT 4) for 17-18 days to 8 food-deprived and 8 ad-lib-fed rats. Chow was provided at ZT 16. Temperature, wheel running, activity, and corticosterone (B) anticipated sucrose only in food-deprived rats. Ad-lib rats were similar to controls. In Experiment 2, 32% sucrose was presented at ZT 4 for 5 or 10 min for 17 days to food-deprived rats. Chow was given at ZT 12. Exogenous cues were minimized. Both groups showed anticipation of sucrose, whereas room controls did not. After a shift to 4% sucrose for 8 days, the 10-min group showed signs of damping of these rhythms, whereas the 5-min group did not. In Experiment 1 a reciprocal equilibrium schedule suppressed the running, drinking, and switching of 3 rats whose baselines were recovered. In Experiment 2 the 2 rats whose baselines were recovered deviated from their baseline distributions of responding on four equilibrium schedules that permitted unscheduled responding. In Experiment 3 four rats that had to lever press on four variable-requirement schedules to gain unconstrained access to water performed more lever pressing in some conditions than calculations of the maximum amount they could have drunk per bout suggested was necessary. In Experiment 4 the responding of 3 rats was found to decrease systematically on a schedule with a fixed running to drinking ratio, as the size of the terms was increased. These findings challenge two assumptions of many molar models of instrumental performance: that the paired baseline distribution of responding is a behavioral ideal that subjects attempt to approach when performing on schedules and that performance on a schedule depends on the ratio of instrumental to contingent responding that it requires, and not on the way that ratio is made up. In Experiment 1, we showed that active- and passive-avoidance responding in a running wheel was learned because of the avoidance contingency. In Experiment 2, strain differences among four commercially bred rats were assessed in an active-avoidance paradigm. Wistar, Donryu, and Fischer rats learned faster than Sprague-Dawleys. In Experiment 3, learning in a multiple active/passive avoidance schedule was examined, and both components of this task were learned. This multiple schedule was used to investigate strain differences in selectively bred rats in Experiments 4 and 5. Tsukuba low-emotional (TLE) rats responded more than Tsukuba high-emotional (THE) rats in both components. However, discrimination of passive components was better in THE than in TLE rats. Syracuse high-avoidance rats were superior in the active component, whereas Syracuse low-avoidance rats showed superior performance in the passive component. Four autoshaping experiments with pigeons investigated the associative decrement produced by nonreinforcement of stimuli with a history of partial or continuous reinforcement. In each experiment, one keylight was reinforced on a 25% schedule and one on a 100% schedule. Then nonreinforced presentations of each stimulus were accompanied by different diffuse stimuli. In each experiment, the diffuse stimulus nonreinforced in the presence of the 100% excitor developed inhibition more rapidly than the diffuse stimulus nonreinforced in the presence of the 25% excitor. This inhibition was measured by transfer to another excitor reinforced on 100% (Experiment 1) or 25% (Experiment 2) schedule. The same difference was observed when the 25% excitor underwent a period of 100% reinforcement (Experiment 3) or was associatively stronger than the 100% excitor (Experiment 4). These results suggest that partial reinforcement acts in part to reduce the subsequent effectiveness of a nonreinforcement in producing associative change. Previous findings indicate that wheel running can have either an aversive or an appetitive effect. That is, wheel running for 30 min induces conditioned taste aversion (CTA) in rats trained while hungry and thirsty but facilitates feeding in non-deprived rats. In Experiment 1, wheel running was also found to be effective in producing CTA in non-deprived rats. Therefore, Experiment 2 tested whether wheel running produces the aversive and appetitive effects simultaneously. During each of four training trials, two groups of non-deprived rats were given a flavored solution to drink for 10 min. Then those in the wheel group were put in running wheels for 30 min whereas those in the cage group spent 30 min in small cages. Finally, all rats were given a 60-min feeding test. After the first trial, the wheel group drank less flavored solution than the cage group during each of the remaining trials. The wheel group also ate more than the cage group on each feeding test. These results indicate that wheel running produces CTA and facilitates eating at the same time. A role for the mesolimbic dopamine reward system in these effects was considered.  相似文献

Methylphenidate (MPH) is one of the most widely prescribed drugs for treating attention-deficit hyperactivity disorder. Previous research suggested that MPH is a reinforcer for rats, but not all of the manipulations to show that lever pressing is controlled by the contingency to obtain MPH have been examined. In Experiment 1, responding for MPH on a progressive ratio (PR) schedule was assessed. Rats self-administered varying doses of MPH (0.056–1.0 mg/kg/infusion) on a PR schedule of reinforcement, and self-administered more MPH than saline, with maximal responding occurring at a unit dose of 0.56 mg/kg/infusion. Experiment 2 examined if there were differences in responding between contingent and noncontingent MPH (0.56 mg/kg/infusion) on a fixed ratio schedule of reinforcement. Results showed that rats responded for contingent MPH, and that responding was not maintained when MPH was delivered noncontingently. Experiment 3 examined self-administration of MPH (0.1 or 0.3 mg/kg/infusion) during long access (6 hr) compared to short access sessions (1 hr). Results showed that rats given long access to MPH showed an escalation of intake across sessions, with this escalation being more pronounced at the lower unit dose (0.1 mg/kg/infusion); in contrast, rats given short access to MPH did not show an increase in MPH self-administration across sessions at either MPH dose tested. Taken together, these results indicate that MPH is an effective intravenous reinforcer for rats and that, similar to other stimulants such as cocaine, amphetamine and methamphetamine, MPH is subject to abuse as reflected by dysregulated intake across repeated long access sessions. The metaphor of the behavior stream provides a framework for studying the effects of response-independent food presentations intruded into an environment in which operant responding of pigeons was maintained by variable-interval schedules. In the first two experiments, response rates were reduced when response-independent food was intruded during the variable-interval schedule according to a concomitantly present fixed-time schedule. These reductions were not always an orderly function of the percentage of response-dependent food. Negatively accelerated patterns of key pecking across the fixed-time period occurred in Experiment 1 under the concomitant fixed-time variable-interval schedules. In Experiment 2, positively and negatively accelerated and linear response patterns occurred even though the schedules were similar to those used in Experiment 1. The variable findings in the first two experiments led to three subsequent experiments that were designed to further illuminate the controlling variables of the effects of intruded response-independent events. When the fixed and variable schedules were correlated with distinct operanda by employing a concurrent fixed-interval variable-interval schedule (Experiment 3) or with distinct discriminative stimuli (Experiments 4 and 5), negatively accelerated response patterns were obtained. Even in these latter cases, however, the response patterns were a joint function of the physical separation of the two schedules and the ratio of fixed-time or fixed-interval to variable-interval schedule food presentations. The results of the five experiments are discussed in terms of the contributions of both reinforcement variables and discriminative stimuli in determining the effects of intruding response-independent food into a stream of operant behavior.  相似文献

Prolactin (Prl) increases food consumption in ring doves (Streptopelia risoria) and may promote the hyperphagia exhibited by parent doves when provisioning young. These experiments tested whether Prl also enhances appetitive aspects of feeding behavior. Prl elevated pecking rates in food-restricted doves on a variable-interval (VI) reinforcement schedule and supported continued responding when doves were returned to ad-lib feeding. Prl-treated doves learned the key-pecking response when food intake was clamped at ad-lib levels exhibited before Prl treatment but not when given free access to food. Median break points on a progressive ratio schedule were 2–3 times greater in food-restricted doves than Prl-treated, food-clamped doves even though response rates were similar on VI schedules. These results indicate that Prl enhances appetitive aspects of feeding, although food restriction at the level imposed in this study was more effective in this regard. We explored whether the generalization of rules based on simple structures depends on attention. Participants were exposed to a stream of artificial words that followed a simple syllabic structure (ABA or AAB), overlaid on a sequence of familiar noises. After passively listening, participants successfully recognized the individual words present in the stream among foils, and they were able to generalize the underlying word structure to new exemplars. Yet, when attention was diverted from the speech stream (by requiring participants to monitor the sequence of noises), recognition of the individual words fell dramatically irrespective of word structure, whereas generalization depended on stimulus structure. For structures based on vowel repetitions across nonadjacent syllables (ABA; Experiment 1), generalization was affected by attention. In contrast, for structures based on adjacent repetitions (AAB; Experiment 2), generalization capacity was unaffected by attention. This pattern of results was replicated under favorable conditions for generalization, such as increased token variability and the implementation of the rule over whole syllables (Experiments 3 and 4). We report data from 4 experiments using a recognition design with multiple probes to be matched to specific study positions. Items could be accessed rapidly, independent of set size, when the test order matched the study order (forward condition). When the order of testing was random, backward, or in a prelearned irregular sequence (reordered conditions), the classic Sternberg result was obtained: Response times were slow and increased linearly with set size. A number of explanations for forward-condition facilitation were ruled out, such as the predictability of the study order (Experiment 2), the predictability of the probe order (Experiment 1), the covariation of study and test orders (Experiments 1, 2, and 4), processes of probe encoding and perception that did not rely on STM access (Experiments 1, 2, and 4), specific support of the forward condition by articulatory processes (Experiment 3), or condition-dependent strategic differences (Experiment 4). More detailed analyses demonstrated that fast forward responses could not be accounted for by the effects of input position and output position that modulated random responses, effects that did account for the slower responses of the reordered conditions (Experiments 1, 3, and 4). A final analysis of probe-to-probe transitions as a function of encoding distance revealed a sizeable single-step benefit in the random condition. We concluded that STM representations were serial rather than spatial and that forward probes benefited from their serial adjacency. Eight exclusive cola drinkers in Experiment 1 (mean caffeine intake = 157 +/- 74 mg/day) and 16 drinkers of both cola and coffee in Experiment 2 (mean caffeine intake = 579 +/- 201 mg/day) underwent 6 independent, double-blind weekly trials. Each trial began with a randomized cross-over sampling period of 1 day of access to noncaffeinated cola and 1 day of access to caffeinated (33 mg/8 oz) cola. During the subsequent 1- or 2-day test period, participants had unlimited concurrent access to the 2 colas. Reliable caffeine self-administration occurred in 2 of 8 participants in Experiment 1 and in 4 of 16 participants in Experiment 2. Self-reported drowsiness, fatigue, and headache were higher when participants received only placebo colas in Experiment 2, but not Experiment 1. Caffeine self-administration via cola occurs both among people whose primary source of caffeine is cola and among those whose primary source of caffeine is coffee.  相似文献

Four related experiments studied operant performance of mice on differential reinforcement of low rates of responding (DRL) paradigms. Experiment 1 showed that excitotoxic hippocampal lesions impaired performance of a 10-s DRL schedule (DRL-10). Experiments 2 and 3 showed that GluR-A AMPA receptor subunit knockout mice, which are deficient in CA3-CA1 long-term potentiation (LTP), were markedly impaired at 15 s (DRL-15), but less impaired at DRL-10. Experiment 4 compared DRL-15 performance in mice from the 2 strains from which the GluR-A colony was derived and showed that they did not differ. The results show that GluR-A-containing AMPA receptors are required for normal performance on hippocampus-dependent, nonspatial working memory tasks, consistent with a role for GluR-A in the temporal encoding (what happened when) of nonspatial information. In 4 experiments, picture-word translation was studied in Spanish-English bilinguals and English monolinguals. In Experiment 1, bilinguals drew pictures or wrote English or Spanish words for picture or English or Spanish word stimuli. Equivalent increases in production onset latency for cross-language/modality translation were found. In Experiment 2, bilinguals and monolinguals drew pictures or wrote English words for picture or English word stimuli. Cross-modality translation equivalence was replicated, though bilinguals were slower than monolinguals overall. In Experiment 3, bilinguals and monolinguals were equivalent when they drew or wrote names from pictures as blocked tasks. In Experiment 4, bilinguals replicated Experiment 1 but were faster for blocked than mixed tasks, indicating that stimulus-processing uncertainty slows them. Food-deprived rats learned to avoid a flavor negatively correlated with access to a rich nutrient, 20% maltodextrin (20M) solution. This avoidance in two-bottle choice tests was produced by training consisting of either an unpaired condition where sessions of unflavored 20M were intermixed with sessions of 2 or 3% maltodextrin (2M or 3M) flavored with salt (Experiment 1) or almond (Experiments 3 and 4) or a differential conditioning procedure where one flavor was mixed with 20M and another with 2M (Experiment 2). Avoidance was counter-conditioned by mixing the target flavor with 20M (Experiment 1), generalized to a neutral context (Experiment 3), and displayed strong resistance to extinction (Experiment 4). The results demonstrated that food avoidance learning can occur in the absence of an aversive unconditioned stimulus and indicated that unpaired control groups and differential conditioning procedures may be misleading in flavor preference learning research when further control conditions are absent.
