Constitutive phenolase activity of plants has a profound ability to modulate disease in insects caused by baculoviruses. We investigated the influence of damage-induced plant phenolic oxidases in cotton and tomato on mortality caused by two different baculoviruses in their respective hosts, Heliothis virescens (L.) and Helicoverpa zea (Boddie). For both plant species, peroxidase (POD) and phenolic levels were predictive of larval mortality caused by baculoviruses. The higher the POD activity, the lower the mortality in both hosts. Different classes of phenolics (e.g., monohydroxyphenolics vs. catecholic phenolics) in combination with POD activity had different effects on the severity of viral disease depending upon mixture, which implies that viral efficacy is predictable only if total chemical content of the plants is specified. Inhibition of baculoviral disease by plant phenolase activity has potential implications for the compatibility of baculoviruses with induced resistance in IPM programs.  相似文献

The effects of chlorogenic acid on larval survival and growth of the tobacco budworm Heliothis virescens and larval susceptibility to Helicoverpa zea nucleopolyhedrovirus (HzSNPV) were studied with an artificial diet and transgenic tobacco Nicotiana tabacum. Survival of neonates on over- or underexpressed phenylalanine ammonia-lyase (PAL) transgenic tobacco lines was positively correlated with the level of chlorogenic acid. Larval weight was not correlated with the level of chlorogenic acid in tobacco foliage. On the other hand, larval weights on artificial diet supplemented with chlorogenic acid were negatively correlated with chlorogenic acid concentration. Second instars treated on virus-treated (10 occlusion bodies/larva) tobacco leaf disks for 24 hr and then reared on tobacco leaves showed higher survival time and increased mortality on lines with increasing levels of chlorogenic acid. However, viral-related larval mortality on artificial diet was negatively correlated with dietary chlorogenic acid. These results illustrate the inadequacy of artificial diet studies in establishing causal relationships among plant chemistry, herbivores, and natural enemies.  相似文献

The infectivity of the nuclear polyhedrosis virus, HzSNPV toHeliothis zea was significantly reduced when viral occlusion bodies were exposed to the plant phenolic chlorogenic acid in the presence of polyphenol oxidase. Chlorogenic acid is rapidly oxidized to the ortho-quinone, chlorogenoquinone, by foliar polyphenol oxidases of the tomato plant, Lycopersicon esculentum, when foliage is damaged during feeding by larvalH. zea.Our results indicate that chlorogenoquinone, a powerful oxidizing agent, covalently binds to the occlusion bodies of HzSNPV and significantly reduces their digestibility and solubility under alkaline conditions. This binding is proposed to interfere with the infection process by impairing the release of infective virions in the midgut.  相似文献

Baculoviral efficacy against lepidopteran larvae is substantially impacted by the host plant. Here, we characterized how baculoviral pathogenicity to cotton-fed Heliothis virescens larvae is affected by induction of systemic acquired resistance (SAR). Numerous studies have shown that SAR induced by the plant elicitor benzo-(1,2,3)-thiadiazole-7-carbothioic acid S-methyl ester (BTH) can protect against plant pathogens, but reports on the impacts of SAR on chewing herbivores or on natural enemies of herbivores are few. We found that BTH application significantly increased foliar peroxidase activity, condensed tannin levels, and total phenolic levels but did not alter dihydroxyphenolic levels. Consumption of BTH-treated foliage did not influence H. virescens pupal weight or larval mortality by the microbial control agent Autographa californica multiple nucleopolyhedrovirus any more than did consumption of untreated foliage. Thus, activation of SAR, although it did not protect the plant against a chewing herbivore, also did not reduce the effect of a natural enemy on a herbivore, indicating that SAR and microbial control agents may be compatible components of integrated pest management.  相似文献

Partial electroantennograms (EAGs) and single cell recordings fromHeliothis virescens males have demonstrated the presence of pheromones receptor neurons in sensilla trichodea type 2 as well as in type 1. This is supported by cobalt tracing experiments, showing that primary axons from the distal flagellum, containing only s. trichodea type 2, project into the macrogiomerulus complex in the male antennal lobes. Four types of finely tuned pheromone receptor neurons were found in males, whereas in females the corresponding neurons responded mainly to host odors. In males the majority (75 and 18%, respectively) were tuned to the majorHeliothis virescens pheromone components (Z)-11-hexadecenal (Z11-16A1) and (Z)-9-tetradecenal (Z9-14A1). The others (5 and 2%, respectively) responded specifically to (Z)-1 1-hexadecen-1-ol (Z1 1-16OH) and (Z)-1 1-hexadecen-1-ol acetate (Z1 1-16Ac). No neurons responding selectively to the minor pheromone components were found. The Z11-16A1 neurons of both sensilla types possessed similar specificity. However, the sensitivity decreased toward the medial and distal part of the flagellum, where s. trichodea type 2 are located. This suggests that the pheromone concentrations can be detected peripherally by a spatial as well as a temporal mechanism. Differences in temporal response patterns (pronounced phasic vs. tonic component) were found within the same type of neurons, suggesting different ability to encode intermittency of the pheromone plume as well as to mediate maintenance of flight.  相似文献

(Z)-11 -Hexadecenal (77–91%), (Z)-7-hexadecenal (0.1–2%), (Z)-9-hexadecenal (0.3–2%), hexadecanal (3–19%), (Z)-11-hexadecen-1-ol (1–5%), tetradecanal (1–3%), and (Z)-9-tetradecenal (1–3%) were identified from the heptane washes of the ovipositor of femaleHeliothis virescens (F.) females. In field bioassays, a 152-g mixture of these seven compounds deployed in an insect trap exceeded the attractiveness of 4 virgin femaleH. virescens for males and was 5–6 times more attractive than a mixture of (Z)-1 1-hexadecenal and (Z)-9-tetradecenal (virelure) that was previously reported as the sex pheromone of the species. Four of the seven compounds produced byH. virescens females are also produced byH. zea (Boddie). Specificity of pheromonal signals among the two species is ostensibly dependent upon one or more of the three additional compounds [tetradecanal, (Z)-9-tetradecenal, and (Z)-1 1-hexadecen-1-ol] produced by femaleH. virescens.  相似文献

Young tomato plants (Lycopersicon esculentum) grown in sand in a greenhouse and subjected to different fertilization regimes were used to test the effects of nitrogen availability on constitutive levels of phenolics and on constitutive and inducible activities of polyphenol oxidase and proteinase inhibitors. Theories that emphasize physiological constraints on the expression of phytochemicals predict an increase in levels of carbon-based allelochemicals under moderate nitrogen stress but predict, under the same conditions, an attenuation of chemical responses involving nitrogen-containing compounds such as proteinase inhibitors and polyphenol oxidase. We found that nitrogen availability had a strong effect on constitutive levels of phenolics; weaker effects on constitutive polyphenol oxidase activity, constitutive proteinase inhibitor activity, and inducible polyphenol oxidase activity; and no effect on inducible proteinase inhibitor activity. These results point to a need for the integration of theories that emphasize physiological influences on secondary metabolism with those that emphasize ecological influences on secondary metabolism and suggest that current theories of plant defense do not adequately account for enzymatic and proteinaceous defenses against arthropods.  相似文献

Evaluations conducted by placingHeliothis virescens (F.) sex pheromone (virelure) dispensers at different distances in the predominant downwind and upwind directions fromHeliothis zea (Boddie) pheromone traps indicated that reductions inH. zea male captures were greatest relative to distance when theH. zea traps were located downwind from the virelure dispensers than when the traps were located upwind. In flight-tunnel assays, mated femaleHeliothis virescens (F.) moths responded by positive anemotaxis to volatiles from extracts of two host plants (cotton and tobacco), but they did not fly to an extract from elderberry (Sambucus simpsonii Rehd.), a nonhost that contains an oviposition deterrent forH. virescens. When the elderberry extract was mixed with extract from either cotton or tobacco, the flight response by moths to volatiles emanating from the extract blends was reduced significantly at most doses when compared to the positive response to extracts from either host alone. The number of landings (including brief contacts) and landings that resulted in oviposition on the substrates treated with extract blends also were reduced significantly in most tests.This article reports the results of research only. A large amount of 20-hydroxyecdysone was orally administered to larvae of the tobacco budworm,Heliothis virescens, in order to investigate its detoxification mechanisms. Four major relatively nonpolar metabolites were isolated from their frass. These compounds were identified as the 22-linoleate, 22-palmitate, 22-oleate, and 22-stearate of 20-hydroxyecdysone using various forms of spectroscopy, including NMR. Mated and virgin female and virgin male tobacco budworm moths,Heliothis virescens (F.), responded via directed upwind flight in wind-tunnel bioassays to volatiles from floral extracts of cotton,Gossypium hirsutum L. Significantly more male and female moths landed on cloth dispensers treated with a methylene chloride extract of debracted flowers than on control dispensers treated only with methylene chloride. Only mated females landed in significant numbers on the dispensers treated with extract of cotton squares, flowers (including bracts), or flower petals. After landing, most of the moths examined the dispensers by probing with their antennae, proboscis, and/or ovipositor.  相似文献

Forty-nine secondary metabolites were isolated from aqueous and alcoholic extracts of the aquatic plant Shoenoplectus lacustris. All compounds were characterized based on spectroscopic data. Eleven free and glycosylated low-molecular polyphenols, 17 cinnamic acid and dihydrocinnamic acid derivatives, 11 flavonoids, and 10 C13 nor-isoprenoids were identified. The structure of the new compound, 1-benzoyl-glycerol-2-α-l-arabinopyranoside, was elucidated by 2D NMR experiments (COSY, HSQC, HMBC, NOESY). To evaluate potential phytotoxic effects, all compounds were tested on the green alga Selenastrum capricornutum, a unicellular organism commonly used in tests of toxicity as a bioindicator of eutrophic sites. The most active compound was (−)-catechin, showing an inhibition similar to that of the algaecide CuSO4.  相似文献

Female moths of the tobacco budworm,Heliothis virescens (F.), oviposit in the terminals of the cotton plant,Gossypium hirsutum (L.). The hatched larvae migrate to the terminal area and then to small squares (buds), on which they feed, finally burrowing into the anthers where they grow and develop. They attempt to avoid gossypol glands as they feed. Chemically related evidence explains, in part, these observations. The calyx crown of resistant lines (which is avoided) is high in the terpenoid aldehydes (TAs) including gossypol. HPLC data showed that the gossypol content of both susceptible and resistant glanded lines is equal, while the hemigossypolone and heliocides H1 and H2 are greatly increased in resistant lines and presumably are more closely associated with resistance. Analysis for total amino acids in cotton square tissues showed that there was a gradation from the calyx and calyx crown, which were lowest, to the anthers, the site of final insect development, which were highest. Synthetic diets mimicking amino acid distribution in anthers were found to be successful for larval growth and development.  相似文献

None of the isolated degradation products of (Z)-11-hexadecenal [(Z)-11-HDAL] affected the catches of either tobacco budworm [Heliothis virescens (F.)] or bollworm [H. zea (Boddie)] moths when dispensed with pheromone from cotton dental rolls in cone traps. Also, none of the degradation products of (Z)-9-tetradecenal [(Z)-9-TDAL] had an effect on trap catches of tobacco budworm moths. Two of the three chemicals that have previously been identified in ovipositor washes of tobacco budworms but that are absent in those of bollworms caused a reduction in capture of bollworms: (Z)-9-TDAL (1.0 g/trap) caused a 96% reduction in trap catch and (Z)-11-hexadecen-1-ol (20.0 g/trap) caused a similar reduction. Two chemicals implicated in resistance of the tomato plant, chlorogenic acid and polyphenol oxidase, are known to form orthoquinones in damaged plant tissue. Orthoquinones have been reported to alkylate –NH2 and –SH groups of proteins and amino acids, altering solubility, digestibility, and, for some pathogenic viruses, infectivity. Here we explore effects of quinone alkylation on toxicity of an important microbial insecticide,Bacillus thuringiensis subsp.kurstaki (BTk), to larvalHeliothis zea. BTk incubated with these phytochemicals and fed to larvae was more toxic than untreated BTk. Similar but less dramatic results arose when BTk was incubated with polyphenol oxidase alone. Digestibility experiments suggest that alkylation enhanced the solubilization and/or proteolysis of crystal protein in vivo. Implications of our results for compatibility of BTk with host-plant resistance and biological control are discussed.  相似文献

Induced plant responses to attack by chewing insects have been intensively studied, but little is known about plant responses to nonchewing insects or to attack by multiple herbivores with different feeding habits. We examined volatile emissions by tobacco, Nicotiana tabacum, in response to feeding by the piercing–sucking insect western flower thrips (WFT), Frankliniella occidentalis, the chewing herbivore Heliothis virescens, and both herbivores simultaneously. In addition, we examined the effects of herbivore-induced plant defenses on host-plant selection by WFT. Plants responded to thrips feeding by consistently releasing five compounds. Simultaneous feeding by WFT and H. virescens elicited the same 11 compounds emitted in response to caterpillar feeding alone; however, two compounds, α-humulene and caryophyllene oxide, were produced in greater amounts in response to simultaneous herbivory. In choice tests, thrips consistently preferred uninduced plants over all other treatments and preferred plants damaged by caterpillars and those treated with caterpillar saliva over those treated with caterpillar regurgitant. The results are consistent with a previous finding that caterpillar regurgitant induces the release of significantly more volatile nicotine than plants damaged by caterpillars or plants treated with caterpillar saliva. A repellent effect of nicotine on WFT was confirmed by encircling unwounded plants with septa releasing volatile nicotine. Our results provide the first direct evidence that thrips feeding induces volatile responses and indicates that simultaneous herbivory by insects with different feeding habits can alter volatile emissions. In addition, the findings demonstrate that induced plant responses influence host-plant selection by WFT and suggest that the induction of volatile nicotine may play a role in this process.  相似文献

Field studies were conducted to determine the effect of (Z)-11-hexadecen-1-ol (Z11-16 OH) on capture of tobacco budworm (TBW),Heliothis virescens (F.), males when added to two-component (HV-2) and six-component (HV-6) pheromone blends dispensed from a polyvinyl chloride (PVC) substrate. Numbers of males caught in traps baited with PVC dispensers containing 0.25% Z11-16 OH, relative to total pheromone content, were significantly higher than numbers caught with dispensers containing noZ11-16 OH. Catches of TBW were also increased significantly in some tests whenZ11-16 OH was formulated in the bait at a 1% concentration. Catches were significantly reduced with dispensers containing 5.95%Z11-16 OH compared with those containing noZ11-16 OH. Sensitivity of caterpillars of Heliothis virescens, a generalist, and H. subflexa, a specialist, to eight different plant secondary compounds was examined behaviorally. The compounds were nicotine hydrogen tartrate, hordenine, caffeine, sinigrin, linamarin, arbutin, chlorogenic acid, and salicin. All compounds deterred feeding, at least at the higher concentrations, but the generalist was less affected than the specialist. Thus the hypothesis that specialists have greater sensitivity to deterrents than generalists was supported. In most cases deterrence occurred on first encounter, indicating that the response was sensory; in some cases short-term postingestive effects also appeared to play a role. The larger quantities of deterrent-containing food ingested by H. virescens sometimes resulted in measurable postingestive effects during the second control test. This did not occur in H. subflexa, which more commonly rejected the deterrent-containing food on first contact. The contrast between the species is discussed in relation to tradeoffs involved in different diet breadths.  相似文献

The foliage and fruit of the tomato plantLycopersicon esculentum contains polyphenol oxidases (PPO) and peroxidases (POD) that are compartmentally separated from orthodihydroxyphenolic substrates in situ. However, when leaf tissue is damaged by insect feeding, the enzyme and phenolic substrates come in contact, resulting in the rapid oxidation of phenolics to orthoquinones. When the tomato fruitwormHeliothis zea or the beet army-wormSpodoptera exigua feed on tomato foliage, a substantial amount of the ingested chlorogenic acid is oxidized to chlorogenoquinone by PPO in the insect gut. Additionally, the digestive enzymes of the fruitworm have the potential to further activate foliar oxidase activity in the gut. Chlorogenoquinone is a highly reactive electrophilic molecule that readily binds cova-lently to nucleophilic groups of amino acids and proteins. In particular, the —SH and —NH2 groups of amino acids are susceptible to binding or alkylation. In experiments with tomato foliage, the relative growth rate of the fruitworm was negatively correlated with PPO activity. As the tomato plant matures, foliar PPO activity may increase nearly 10-fold while the growth rate of the fruitworm is severely depressed. In tomato fruit, the levels of PPO are highest in small immature fruit but are essentially negligible in mature fruit. The growth rate of larvae on fruit was also negatively correlated with PPO activity, with the fastest larval growth rate occurring when larvae fed on mature fruit. The reduction in larval growth is proposed to result from the alkylation of amino acids/protein byo-quinones, and the subsequent reduction in the nutritive quality of foliage. This alkylation reduces the digestibility of dietary protein and the bioavailability of amino acids. We believe that this mechanism of digestibility reduction may be extrapolatable to other plant-insect systems because of the ubiquitous cooccurrence of PPO and phenolic substrates among vascular plant species.  相似文献
