A novel monolithic material was developed to obtain efficient and cost‐effective purification of IgG from human plasma. The porous monolith was obtained by bulk polymerization in a glass tube of 2‐hydroxyethyl methacrylate (HEMA) and N‐methacryloyl‐(L )‐histidine methyl ester (MAH). The poly(HEMA‐MAH) monolith had a specific surface area of 214.6 m2/g and was characterized by swelling studies, porosity measurement, FTIR, scanning electron microscopy, and elemental analysis. Then the monolith was loaded with Cu2+ ions to form the metal chelate. Poly(HEMA‐MAH) monolith with a swelling ratio of 74% and containing 20.9 μmol MAH/g was used in the adsorption/desorption of IgG from aqueous solutions and human plasma. The maximum adsorption of IgG from an aqueous solution in phosphate buffer was 10.8 mg/g at pH 7.0. Higher adsorption was obtained from human plasma (up to 104.2 mg/g), with a purity of 94.1%. It was observed that IgG could be repeatedly adsorbed and desorbed with the poly(HEMA‐MAH) monolith without significant loss of adsorption capacity. © 2006 Wiley Periodicals, Inc. Poly(acrylamide‐allyl glycidyl ether) [poly(AAm‐AGE)] cryogel was prepared by bulk polymerization which proceeds in an aqueous solution of monomers frozen inside a glass column (cryo‐polymerization). After thawing, the monolithic cryogel contains a continuous polymeric matrix having interconnected pores of 10–100 μm size. Cibacron Blue F3GA was immobilized by covalent binding onto poly(AAm‐AGE) cryogel via epoxy groups. Poly(AAm‐AGE) cryogel was characterized by swelling studies, FTIR, scanning electron microscopy, and elemental analysis. The equilibrium swelling degree of the poly(AAm‐AGE) monolithic cryogel was 6.84 g H2O/g cryogel. Poly(AAm‐AGE) cryogel containing 68.9 μmol Cibacron Blue F3GA/g was used in the adsorption/desorption of human serum albumin (HSA) from aqueous solutions and human plasma. The nonspecific adsorption of HSA was very low (0.2 mg/g). The maximum amount of HSA adsorption from aqueous solution in acetate buffer was 27 mg/g at pH 5.0. Higher HSA adsorption value was obtained from human plasma (up to 74.2 mg/g). Desorption of HSA with a purity of 92% from Cibacron Blue F3GA attached poly(AAm‐AGE) cryogel was achieved using 0.1M Tris/HCl buffer containing 0.5M NaCl. It was observed that HSA could be repeatedly adsorbed and desorbed with poly(AAm‐AGE) cryogel without significant loss in the adsorption capacity. © 2007 Wiley Periodicals, Inc. The aim of this study was to investigate in detail the performance for removal of heavy metal ions of beads composed of poly(2‐hydroxyethyl methacrylate) (pHEMA) to which N‐methacryloylhistidine (MAH) was copolymerized. The metal‐complexing ligand MAH was synthesized by using methacryloyl chloride and histidine. Spherical beads with an average size of 150–200 μm were obtained by the radical suspension polymerization of MAH and HEMA conducted in an aqueous dispersion medium. Owing to the reasonably rough character of the bead surface, p(HEMA‐MAH) beads had a specific surface area of 17.6 m2/g. The synthesized MAH monomer was characterized by NMR; p(HEMA‐MAH) beads were characterized by swelling studies, FTIR and elemental analysis. The p(HEMA‐MAH) beads with a swelling ratio of 65%, and containing 1.6 mmol MAH/g, were used in the adsorption/desorption experiments. Adsorption capacity of the beads for the selected metal ions, i. e., Cu(II), Cd(II), Cr(III), Hg(II) and Pb(II), were investigated in aqueous media containing different amounts of these ions (10–750 mg/L) and at different pH values (3.0–7.0). Adsorption equilibria were established in about 20 min. The maximum adsorption capacities of the p(HEMA‐MAH) beads were 122.7 mg/g for Cu(II), 468.8 mg/g for Cr(III), 639.4 mg/g for Cd(II), 714.1 mg/g for Pb(II) and 1 234.4 mg/g for Hg(II). pH significantly affected the adsorption capacity of MAH incorporated beads. The chelating beads can be easily regenerated by 0.1 M HNO3 with high effectiveness. These features make p(HEMA‐MAH) beads a potential candidate for heavy metal removal at high capacity.  相似文献

N‐Methacryloyl‐(L )‐histidine methyl ester (MAH) was synthesized from metharyloyl chloride and histidine. Spherical beads with an average size of 150–250 μm were obtained by the suspension polymerization of ethylene glycol dimethacrylate and MAH in an aqueous dispersion medium. Magnetic poly(ethylene glycol dimethacrylate‐coN‐Methacryloyl‐(L )‐histidine methyl ester) [m‐p(EGDMA‐co‐MAH)] microbeads were characterized with swelling tests, electron spin resonance, elemental analysis, and scanning electron microscopy. The specific surface area of the beads was 80.1 m2/g. m‐p(EGDMA‐co‐MAH) microbeads with a swelling ratio of 40.2% and 43.9 μmol of MAH/g were used for the adsorption of bovine serum albumin (BSA) in a batch system. The Cu(II) concentration was 4.1 μmol/g. The adsorption capacity of BSA on the Cu(II)‐incorporated beads was 19.2 mg of BSA/g. The BSA adsorption first increased with the BSA concentration and then reached a plateau, which was about 19.2 mg of BSA/g. The maximum adsorption was observed at pH 5.0, which was the isoelectric point of BSA. The BSA adsorption increased with decreasing temperature, and the maximum adsorption was achieved at 4°C. High desorption ratios (>90% of the adsorbed BSA) were achieved with 1.0M NaSCN (pH 8.0) in 30 min. The nonspecific adsorption of BSA onto the m‐p(EGDMA‐co‐MAH) beads was negligible. © 2004 Wiley Periodicals, Inc. J Appl Polym Sci 93: 2669–2677, 2004  相似文献

In this study, we developed a novel approach to obtain a high protein‐adsorption capacity utilizing 2‐methacryloylamidohistidine (MAH) as a biollgand. MAH was synthesized by reacting methacryloyl chloride and histidine. Spherical beads, with an average size of 150–200 μm, were obtained by the radical suspension polymerization of MAH, ethyleneglycol dimethacrylate (EGDMA), and 2‐hydroxyethyl methacrylate (HEMA) conducted in an aqueous dispersion medium. p(EGDMA–HEMA–MAH) beads had a specific surface area of 17.6 m2/g. The synthesized MAH monomer was characterized by NMR. p(EGDMA–HEMA–MAH) beads were characterized by a swelling test, FTIR, and elemental analysis. Then, Cu(II) ions were incorporated into the beads and Cu(II) loading was found to be 0.96 mmol/g. These beads, with a swelling ratio of 65% and containing 1.6 mmol MAH/g, were used in the adsorption/desorption of human serum albumin (HSA) from both aqueous solutions and human serum. The adsorption of HSA onto p(EGDMA–HEMA–MAH) was low (8.8 mg/g). Cu(II) chelation onto the beads significantly increased the HSA adsorption (56.3 mg/g). The maximum HSA adsorption was observed at pH 8.0 Higher HSA adsorption was observed from human serum (94.6 mg HSA/g). Adsorptions of other serum proteins were obtained as 3.7 mg/g for fibrinogen and 8.5 mg/g for γ‐globulin. The total protein adsorption was determined as 107.1 mg/g. Desorption of HSA was obtained using a 0.1M Tris/HCI buffer containing 0.5M NaSCN. High desorption ratios (to 98% of the adsorbed HSA) were observed. It was possible to reuse Cu(II)‐chelated p(EGDMA–HEMA–MAH) beads without significant decreases in the adsorption capacities. © 2003 Wiley Periodicals, Inc. Porous polymeric beads were obtained by the suspension polymerization of 2‐hydroxyethyl methacrylate (HEMA) and ethylene glycol dimethacrylate (EGDMA). Poly(HEMA–EGDMA) beads were characterized by surfacearea measurements, swelling studies, FTIR, scanning electron microscopy (SEM), and elemental analysis. Poly (HEMA–EGDMA) beads had a specific surface area of 56 m2/g. SEM observations showed that the poly(HEMA–EGDMA) beads abounded macropores. Poly(HEMA–EGDMA) beads with a swelling ratio of 55%, and containing different amounts of Reactive Red 120 (9.2–39.8 μmol/g) were used in the adsorption/desorption of human serum albumin (HSA) from aqueous solutions and human plasma. The nonspecific adsorption of HSA was very low (0.2 mg/g). The maximum HSA adsorption amount from aqueous solution in phosphate buffer was 60.1 mg/g at pH 5.0. Higher HSA adsorption value was obtained from human plasma (up to 95.7 mg/g) with a purity of 88%. The equilibrium monolayer adsorption amount, Qmax was determined as 172.4 mg/g. The dimensionless separation factor (RL) value shows that the adsorption behavior of HSA onto the Reactive Red 120 attached poly(HEMA–EGDMA) beads was favorable (0 < RL < 1). Desorption of HSA from Reactive Red 120 attached poly (HEMA–EGDMA) beads was performed using 0.1M Tris/HCl buffer containing 0.5M NaCl. It was observed that HSA could be repeatedly adsorbed and desorbed with Reactive Red 120‐attached poly(HEMA–EGDMA) beads without significant loss in the adsorption amount. © 2007 Wiley Periodicals, Inc. N‐methacryloyl‐(L )‐alanine (MALA) was synthesized by using methacryloyl chloride and alanine as a metal‐complexing ligand or comonomer. Spherical beads with an average diameter of 150–200 μm were obtained by suspension polymerization of MALA and 2‐hydroxyethyl methacrylate (HEMA) conducted in an aqueous dispersion medium. Poly(HEMA–MALA) beads were characterized by SEM, swelling studies, surface area measurement, and elemental analysis. Poly(HEMA–MALA) beads have a specific surface area of 68.5 m2/g. Poly(HEMA–MALA) beads with a swelling ratio of 63%, and containing 247 μmol MALA/g were used in the removal of Hg2+ from aqueous solutions. Adsorption equilibrium was achieved in about 60 min. The adsorption of Hg2+ ions onto PHEMA beads was negligible (0.3 mg/g). The MALA incorporation into the polymer structure significantly increased the mercury adsorption capacity (168 mg/g). Adsorption capacity of MALA containing beads increased significantly with pH. The adsorption of Hg2+ ions increased with increasing pH and reached a plateau value at around pH 5.0. Competitive heavy metal adsorption from aqueous solutions containing Cd2+, Cu2+, Pb2+, and Hg2+ was also investigated. The adsorption capacities are 44.5 mg/g for Hg2+, 6.4 mg/g for Cd2+, 2.9 mg/g for Pb2+, and 2.0 mg/g for Cu2+ ions. These results may be considered as an indication of higher specificity of the poly(HEMA–MALA) beads for the Hg2+ comparing to other ions. Consecutive adsorption and elution operations showed the feasibility of repeated use for poly(HEMA–MALA) chelating beads. © 2006 Wiley Periodicals, Inc. Metal‐chelating membranes have advantages as adsorbents in comparison with conventional beads because they are not compressible and they eliminate internal diffusion limitations. The aim of this study was to explore in detail the performance of poly(2‐hydroxyethyl methacrylate–methacryloylamidohistidine) [poly(HEMA–MAH)] membranes for the removal of three toxic heavy‐metal ions—Cd(II), Pb(II), and Hg(II)—from aquatic systems. The poly(HEMA–MAH) membranes were characterized with scanning electron microscopy and 1H‐NMR spectroscopy. The adsorption capacity of the poly(HEMA–MAH) membranes for the selected heavy‐metal ions from aqueous media containing different amounts of these ions (30–500 mg/L) and at different pH values (3.0–7.0) was investigated. The adsorption capacity of the membranes increased with time during the first 60 min and then leveled off toward the equilibrium adsorption. The maximum amounts of the heavy‐metal ions adsorbed were 8.2, 31.5, and 23.2 mg/g for Cd(II), Pb(II), and Hg(II), respectively. The competitive adsorption of the metal ions was also studied. When the metal ions competed, the adsorbed amounts were 2.9 mg of Cd(II)/g, 14.8 mg of Pb(II)/g, and 9.4 mg of Hg(II)/g. The poly(HEMA–MAH) membranes could be regenerated via washing with a solution of nitric acid (0.01M). The desorption ratio was as high as 97%. These membranes were suitable for repeated use for more than three adsorption/desorption cycles with negligible loss in the adsorption capacity. The stability constants for the metal‐ion/2‐methacryloylamidohistidine complexes were calculated to be 3.47 × 106, 7.75 × 107, and 2.01 × 107 L/mol for Cd(II), Pb(II), and Hg(II) ions, respectively, with the Ruzic method. © 2005 Wiley Periodicals, Inc. A supermacroporous cryogel was prepared to obtain an efficient and cost effective purification of IgG from human plasma. N-methacryloyl-(L)-histidine methyl ester (MAH) was chosen as the pseudospecific ligand and/or comonomer. Poly(hydroxyethyl methacrylate-N-methacryloyl-(L)-histidine methylester) [PHEMAH] cryogel was produced by free radical polymerization initiated by N,N,N′,N′-tetramethylene diamine (TEMED) and ammonium persulfate (APS) pair in an ice bath. PHEMAH cryogel had a specific surface area of 38.6 m2/g. PHEMAH cryogel was characterized by swelling studies, scanning electron microscopy and elemental analysis. PHEMAH cryogel containing 113.7 µmol MAH/g was used in the purification of IgG from human plasma. Compared with the poly(hydroxyethyl methacrylate) (PHEMA) cryogel (0.4 mg/g), the IgG adsorption capacity of the PHEMAH cryogel (24.7 mg/g) was improved significantly due to the MAH incorporation into the polymeric matrix. The maximum amount of IgG adsorption from aqueous solution in phosphate buffer was observed at pH 7.4. The IgG adsorption amount from human plasma was 97.3 mg/g with a purity of 94.6%. It was observed that IgG could be repeatedly adsorbed and eluted with PHEMAH cryogel without significant loss in the adsorption capacity.  相似文献

Monosize poly(2‐hydroxyethyl methacrylate‐coN‐methacryloly‐L ‐histidinemethylester) [mon‐poly(HEMA‐MAH)] nanospheres were prepared via surfactant‐free emulsion polymerization method. L ‐Histidine groups of the mon‐poly(HEMA‐MAH) nanospheres were chelated with Fe3+ ions. Mon‐poly(HEMA‐MAH) nanospheres were characterized by Fourier transform infrared spectroscopy, proton NMR, and scanning electron microscopy. Particle size of the mon‐poly(HEMA‐MAH) nanospheres was measured by Zeta Sizer. Elemental analysis of MAH for nitrogen was estimated as 0.94 mmol/g polymer. The catalase immobilized onto the mon‐poly(HEMA‐MAH)–Fe3+ nanospheres resulted in increasing the enzyme stability with time. Optimum operational temperature for both immobilized preparations was the same, and the temperature profiles of the immobilized preparations were significantly broader. It was observed that enzyme could be repeatedly adsorbed and desorbed on the mon‐poly(HEMA‐MAH)–Fe3+ nanospheres without loss of adsorption capacity or enzymic activity. © 2009 Wiley Periodicals, Inc. Metal chelating properties of Cibacron Blue F3GA‐derived poly(EGDMA‐HEMA) microbeads have been studied. Poly(EGDMA‐HEMA) microbeads were prepared by suspension copolymerization of ethylene glycol dimethacrylate (EGDMA) and hydroxy‐ethyl methacrylate (HEMA) by using poly(vinyl alcohol), benzoyl peroxide, and toluene as the stabilizer, the initiator, and the pore‐former, respectively. Cibacron Blue F3GA was covalently attached to the microbeads via the nucleophilic substitution reaction between the chloride of its triazine ring and the hydroxyl groups of the HEMA, under alkaline conditions. Microbeads (150–200 μm in diameter) with a swelling ratio of 55%, and carrying 16.5 μmol Cibacron Blue F3GA/g polymer were used in the adsorption/desorption studies. Adsorption capacity of the microbeads for the selected metal ions, i.e., Cu(II), Zn(II), Cd(II), Fe(III), and Pb(II) were investigated in aqueous media containing different amounts of these ions (5–200 ppm) and at different pH values (2.0–7.0). The maximum adsorptions of metal ions onto the Cibacron Blue F3GA‐derived microbeads were 0.19 mmol/g for Cu(II), 0.34 mmol/g for Zn(II), 0.40 mmol/g for Cd(II), 0.91 mmol/g for Fe(III), and 1.05 mmol/g for Pb(II). Desorption of metal ions were studied by using 0.1 M HNO3. High desorption ratios (up to 97%) were observed in all cases. Repeated adsorption/desorption operations showed the feasibility of repeated use of this novel sorbent system. © 1999 John Wiley & Sons, Inc. J Appl Polym Sci 71: 1397–1403, 1999  相似文献

Lysozyme adsorption onto dye‐attached nonporous monosize poly(2‐hydroxyethyl‐methacrylate‐methylmethacrylate) [poly(HEMA‐MMA)] microspheres was investigated. Poly(HEMA‐MMA) microspheres were prepared by dispersion polymerization. The monochloro‐triazine dye, Cibacron Blue F3GA, was immobilized covalently as dye–ligand. These dye‐affinity microspheres were used in the lysozyme adsorption–desorption studies. The effect of initial concentration of lysozyme and medium pH on the adsorption efficiency of dye‐attached and metal‐chelated microspheres were studied in a batch reactor. Effect of Cu(II) chelation on lysozyme adsorption was also studied. The nonspecific adsorption of lysozyme on the poly(HEMA‐MMA) microspheres was 3.6 mg/g. Cibacron Blue F3GA attachment significantly increased the lysozyme adsorption up to 247.8 mg/g. Lysozyme adsorption capacity of the Cu(II) incorporated microspheres (318.9 mg/g) was greater than that of the Cibacron Blue F3GA‐attached microspheres. Significant amount of the adsorbed lysozyme (up to 97%) was desorbed in 1 h in the desorption medium containing 1.0M NaSCN at pH 8.0 and 25 mM EDTA at pH 4.9. In order to examine the effects of separation conditions on possible conformational changes of lysozyme structure, fluorescence spectrophotometry was employed. We conclude that dye‐ and metal‐chelate affinity chromatography with poly(HEMA‐MMA) microspheres can be applied for lysozyme separation without causing any significant changes and denaturation. Repeated adsorption/desorption processes showed that these novel dye‐attached monosize microspheres are suitable for lysozyme adsorption. © 2000 John Wiley & Sons, Inc. The aim of this study was to investigate the heavy metal adsorption performance of supermacroporous poly(hydroxyethyl methacrylate) [PHEMA] cryogel. The PHEMA cryogel was produced by cryo‐polymerization. The PHEMA cryogel was characterized by scanning electron microscopy (SEM). The PHEMA cryogel containing 385 μmol Reactive Green HE‐4BD/g were used in the adsorption studies. Adsorption capacity of the PHEMA cryogel for the metal ions, i.e., Cu2+, Cd2+, and Pb2+ were investigated in aqueous media containing different amounts of the ions (5–600 mg/L) and at different pH values (3.2–6.9). The maximum adsorption capacities of the PHEMA cryogel were 11.6 mg/g (56 μmol/g) for Pb2+, 24.5 mg/g (385 μmol/g) for Cu2+ and 29.1 mg/g (256 μmol/g) for Cd2+. The competitive adsorption capacities were 10.9 mg/g (52 μmol/g) for Pb2+, 22.1 mg/g for Cd2+ (196 μmol/g) and 23.2 mg/g (365 μmol/g) for Cu2+. The PHEMA/Reactive Green HE‐4BD cryogel exhibited the following metal ion affinity sequence on molar basis: Cu2+ > Cd2+ > Pb2+. The PHEMA/Reactive Green HE‐4BD cryogel can be easily regenerated by 50 mM EDTA with higher effectiveness. These features make the PHEMA/Reactive Green HE‐4BD cryogel a potential adsorbent for heavy metal removal. © 2010 Wiley Periodicals, Inc. J Appl Polym Sci, 2010  相似文献

Novel hydrophobic nanospheres with an average size of 100 nm utilizing N‐methacryloyl‐(l)‐tryptophan methyl ester (MAT) as a hydrophobic monomer were prepared by surfactant free emulsion polymerization of 2‐hydroxyethyl methacrylate (HEMA) and MAT. MAT was synthesized using methacryloyl chloride and l‐tryptophan methyl ester. Specific surface area of the nonporous nanospheres was found to be 1914 m2/g. Poly(HEMA–MAT) nanospheres were characterized by Fourier transform infrared spectroscopy (FTIR) and scanning electron microscopy (SEM). Average particle size, size distribution, and surface charge measurements were also performed. Elemental analysis of MAT for nitrogen was estimated as 1.95 mmol/g polymer. Then, poly(HEMA–MAT) nanospheres were used in the adsorption of lysozyme in batch system. Using an optimized adsorption protocol, a very high loading of 1075 mg lysozyme/g nanosphere was obtained. The adsorption phenomena appeared to follow a typical Langmuir isotherm. It was observed that enzyme could be repeatedly adsorbed and desorbed without significant loss in adsorption amount or enzyme activity. © 2009 Wiley Periodicals, Inc. J Appl Polym Sci, 2010  相似文献

We prepared poly(ethylene glycol dimethacrylate–1‐vinyl‐1,2,4‐triazole) [poly(EGDMA–VTAZ)] beads (average diameter = 150–200 μm) by copolymerizing ethylene glycol dimethacrylate (EGDMA) with 1‐vinyl‐1,2,4‐triazole (VTAZ). The copolymer composition was characterized by elemental analysis and found to contain five EGDMA monomer units for each VTAZ monomer unit. The poly(EGDMA–VTAZ) beads had a specific surface area of 65.8 m2/g. Poly(EGDMA–VTAZ) beads were characterized by Fourier transform infrared spectroscopy, elemental analysis, surface area measurements, swelling studies, and scanning electron microscopy. Poly(EGDMA–VTAZ) beads with a swelling ratio of 84% were used for the heavy‐metal removal studies. The adsorption capacities of the beads for Cd(II), Hg(II), and Pb(II) were investigated in aqueous media containing different amounts of these ions (5–750 mg/L) and at different pH values (3.0–7.0). The maximum adsorption capacities of the poly(EGDMA–VTAZ) beads were 85.7 mg/g (0.76 mmol/g) for Cd(II), 134.9 mg/g (0.65 mmol/g) for Pb(II), and 186.5 mg/g (0.93 mmol/g) for Hg(II). The affinity order toward triazole groups on a molar basis was observed as follows: Hg(II) > Cd(II) > Pb(II). pH significantly affected the adsorption capacity of the VTAZ‐incorporated beads. The equilibrium data were well fitted to the Redlich–Peterson isotherm. Consideration of the kinetic data suggested that chemisorption processes could have been the rate‐limiting step in the adsorption process. Regeneration of the chelating‐beads was easily performed with 0.1M HNO3. © 2006 Wiley Periodicals, Inc. J Appl Polym Sci 102: 4276–4283, 2006  相似文献

We investigated a new adsorbent system, Reactive Red 120 attached poly(2‐hydroxyethyl methacrylate ethylene dimethacrylate) [poly(HEMA–EDMA)] beads, for the removal of Ni2+ ions from aqueous solutions. Poly(HEMA–EDMA) beads were prepared by the modified suspension copolymerization of 2‐hydroxyethyl methacrylate and ethylene dimethacrylate. Reactive Red 120 molecules were covalently attached to the beads. The beads (150–250 μm), having a swelling ratio of 55% and carrying 25.5 μmol of Reactive Red 120/g of polymer, were used in the removal of Ni2+ ions. The adsorption rate and capacity of the Reactive Red 120 attached poly(HEMA–EDMA) beads for Ni2+ ions was investigated in aqueous media containing different amounts of Ni2+ ions (5–35 mg/L) and having different pH values (2.0–7.0). Very high adsorption rates were observed at the beginning, and adsorption equilibria were then gradually achieved in about 60 min. The maximum adsorption of Ni2+ ions onto the Reactive Red 120 attached poly(HEMA–EDMA) beads was 2.83 mg/g at pH 6.0. The nonspecific adsorption of Ni2+ ions onto poly(HEMA–EDMA) beads was negligible (0.1 mg/g). The desorption of Ni2+ ions was studied with 0.1M HNO3. High desorption ratios (>90%) were achieved. The intraparticle diffusion rate constants at various temperatures were calculated as k20°C = 0.565 mg/g min0.5, k30°C = 0.560 mg/g min0.5, and k40°C = 0.385 mg/g min0.5. Adsorption–desorption cycles showed the feasibility of repeated use of this novel adsorbent system. The equilibrium data fitted very well both Langmuir and Freundlich adsorption models. The pseudo‐first‐order kinetic model was used to describe the kinetic data. © 2006 Wiley Periodicals, Inc. J Appl Polym Sci 100:5056–5065, 2006  相似文献

Magnetic poly(ethylene glycol dimethacrylate‐N‐methacryloyl‐(L )‐histidine methyl ester) [m‐poly(EGDMA‐MAH)] beads were prepared by suspension polymerization for the affinity depletion of immunoglobulin‐G (IgG) from human serum in a batch system. Elemental analysis of the magnetic beads for nitrogen was estimated as 70 μmol MAH/g polymer. IgG adsorption onto the m‐poly(EGDMA) was negligible. Higher adsorption value (up to 46.8 mg/g) was obtained in which the m‐poly(EGDMA‐MAH) beads were used. IgG adsorption capacity of the magnetic beads increased with an increase in the concentration of IgG. The maximum IgG adsorption was observed at pH 6.5 for MOPS buffer. IgG molecules could be repeatedly adsorbed and eluted with these adsorbents, without noticeable loss in their IgG adsorption capacity. Adsorption capacity decreased for both increasing salt concentration and temperature. In this study, we show that m‐poly(EGDMA‐MAH) beads (wherein IgG molecules bind directly with the matrix) can be used directly for affinity depletion without further modification. Higher adsorption value was obtained from human serum (up to 85.7 mg/g). The elution results demonstrated that the adsorption of IgG to the adsorbent was reversible. The depletion efficiencies for IgG were above 85% for all studied concentrations. Eluted portion was analyzed for testing the IgG removal efficiency by two dimensional gel electrophoresis. Eluted proteins include mainly IgG, and a small number of nonalbumin proteins such as apolipoprotein A1, serotransferrin, haptoglobulin, and α1‐antitrypsin. IgA was not identified in eluted fraction. © 2007 Wiley Periodicals, Inc. J Appl Polym Sci 2007  相似文献

Poly(N‐vinyl 2‐pyrrolidone‐g‐citric acid) (PVP‐g‐CA) hydrogels with varying compositions were prepared from ternary mixtures of N‐vinyl 2‐pyrrolidone–citric acid–water by using 60Co γ‐rays. The effect of gel composition on the uranyl ions adsorption capacity of PVP‐g‐CA hydrogels was investigated. Uranyl adsorption capacity of these hydrogels were found to be in the range of 18–144 mg [UO]/g dry gel from the aqueous solution of uranyl nitrate and 22–156 mg [UO]/g dry gel from the aqueous solution of uranyl acetate, depending on the content of citric acid in the hydrogel, while poly(N‐vinyl 2‐pyrrolidone) hydrogel did not sorb any uranyl ion. The swelling of PVP‐g‐CA hydrogel containing 2.7 mol % CA was observed in water (1620%), in uranyl acetate solution (1450%) and in uranyl nitrate solution (1360%), as compared to 700% swelling of pure PVP hydrogels. The diffusion coefficients were varied from 12.57 up to 4.04 • 10−8 m2 s−1. © 2000 John Wiley & Sons, Inc. J Appl Polym Sci 77: 1037–1043, 2000  相似文献

We used a molecular imprinting approach to achieve specific metal binding utilizing N‐methacryloyl‐(L )‐cysteine methyl ester (MAC) as a metal‐complexing ligand. MAC was synthesized using methacryloyl chloride and cysteine methyl ester. Then, Fe3+ was complexed with MAC monomer. Fe3+‐imprinted poly(hydroxyethyl methacrylate‐N‐methacryloyl‐(L )‐cysteine methyl ester) [poly(HEMA‐MAC)] beads with average size of 63–140 μm were produced by suspension polymerization. After that, the template ions (i.e. Fe3+ ions) were removed by 0.1M HCl. Fe3+‐imprinted beads were characterized by swelling studies, FTIR, and elemental analysis. The Fe3+‐imprinted beads with a swelling ratio of 72%, and containing 3.9 mmol MAC/g were used in the binding of Fe3+ ions from aqueous solutions, tap water, certified reference serum sample, and real serum sample. Maximum binding capacity, optimum pH, and equilibrium binding time were 107 μmol/g, pH 3.0, and 30 min, respectively. It was observed that even in the presence of other ions, Fe3+‐imprinted beads selectively bound Fe3+ ions with 97% efficiency. Removal of Fe3+ ions from certified reference serum sample was approximately found to be 33%. © 2006 Wiley Periodicals, Inc. J Appl Polym Sci 101: 3520–3528, 2006  相似文献
