Two experiments were conducted to study the effect of feeding raw or treated common vetch seeds (V) on BW, feed intake (FI), egg production (EP), feed conversion (FC), egg weight (EW), shell thickness (ST), yolk color score (YC), and Haugh unit score (HU) of Single Comb White Leghorn hens for 56 d. In Experiment 1, diets contained 0, 7.5, 15, and 22.5% raw V. Compared with the control, the 22.5% V diet decreased (P < .05) BW, FI, and EP. In Experiment 2, intact or ground (G) V were either soaked (S) in water (1:5) for 24 h or autoclaved (A) at 103.5 x 10(3) Pa for 8 h, and then dried (D) at 55 C for 24 h. Eight diets were used, a corn soybean (control) and seven others, each containing 25% V, previously subjected to the forementioned treatments as follows: untreated V (UV), SDV, GSDV, ADV, GADV, SADV, and GSADV. Compared with the control, the UV diet decreased (P < .05) FI (75 vs 98 g) and EP (47.3 vs 88.7%), increased FC (1.99 vs 1.4 kg feed per dozen eggs), and induced BW loss (-63 vs 49 g). Soaking intact V improved (P < .05) FI (85 g) and EP (69%). Remaining treatments resulted in further improvement. Hens fed all V diets produced eggs with HU score 13 points better than that of the control (P < .05). In both experiments, EW, YC, and ST were not different among treatments. Results indicated that autoclaved V at 25% level was not detrimental to layers' performance.  相似文献

Three studies were conducted to evaluate amino acid utilization by cattle. In Exp. 1, five steers (580 kg) were fed 86% rolled corn diets with mixtures of amino acids containing up to 6 g/d DL-Met, 24 g/d L-Lys, 6 g/d L-Thr, and 3 g/d L-Trp. Treatments had little effect on ruminal fermentation, diet digestibility, N flow to the duodenum, or microbial efficiency. Ruminal concentrations of Met and Lys increased linearly (P < .05) with amino acid supplementation, whereas Thr responded quadratically, and Trp was not altered. In Exp. 2, four steers (414 kg) were used to measure effects of dietary monensin or laidlomycin propionate in high-grain diets supplemented with amino acids. Ionophores had no significant effect on ruminal fermentation or outflows of amino acids from the rumen. In Exp. 3, 100 steers (287 kg initial BW) were fed diets containing 1% of a nonprotein N source. Treatments were 1) no supplemental N (UREA), 2) UREA plus soybean meal (SBM), 3) UREA plus 2 g/d DL-Met, 8 g/d L-Lys, 2 g/d L-Thr, and 1 g/d L-Trp, or 4) UREA plus 4 g/d DL-Met, 16 g/d L-Lys, 4 g/d L-Thr, and 2 g/d L-Trp. During the growing period (diets based on whole-plant milo silage), gains were higher for SBM-supplemented steers than for UREA steers and intermediate for steers supplemented with amino acids. Few significant differences in performance were observed among treatments during the finishing phase (diets based on dry-rolled corn) or for the entire experiment, but cattle fed SBM or amino acids tended to be fatter and have better marbling scores and quality grades. Three groups of intact hinds (n = 10-18) and one group of ovariectomized hinds were treated with progesterone by mean, of Controlled Internal Drug Releasing (CIDR) devices for 13 days (device removal = Day 0). Group 1 served as controls; group 2 received injections of 4 mg recombinant bovine interferon-alpha,1 twice daily from Days 13 to 21; group 3 was run with a stag from Days 0 to 3, and all hinds were subsequently diagnosed pregnant; group 4 (ovariectomized) was treated with CIDR devices and estradiol to mimic steroid secretion during the estrous cycle. Progesterone profiles were determined from thrice-weekly plasma samples from Days -13 to 28. Rectal temperature was measured in a subset of groups 1 and 2 from Days 9 to 21. Oxytocin-induced prostaglandin F2 alpha release was measured in a subset of groups 1, 2, and 4 on Days 2, 4, 10, 16, and 18. Data are presented as means +/- SEM. Exogenous interferon delayed luteolysis (> or = 28 vs. 21.2 +/- 0.55 days, P < 0.0005) and induced transient pyrexia after the first injection (39.89 +/- 0.11 vs. 38.88 +/- 0.19 degrees C, p < 0.0005). Incidence of oxytocin-induced PGF2 alpha release in control hinds was greater on Days 2 and 18 than on Days 4 and 10 (8/8 and 7/8 vs. 3/8 and 0/8, respectively; p < 0.05) and was greater in control than in interferon-treated hinds on Days 16 and 18 (5/8 and 7/8 vs. 1/8 and 1/8, respectively; p < 0.05). Profiles of plasma progesterone concentration and oxytocin sensitivity in steroid-treated ovariectomized hinds did not differ from those in control hinds. These results suggest that steroid-controlled uterine oxytocin sensitivity is important in luteolysis and is suppressed by the administration of interferon, the putative embryonic pregnancy recognition signal in red deer.  相似文献

This study evaluated the effects of recombinant bovine somatotropin (bST; one injection of 320 mg per ewe) on milk production and composition and on the grazing behavior of multiparous ewes in the third to fourth lactation. Forty Comisana lactating ewes were divided into four groups: 1) untreated, grazing on natural pasture (botanical composition: 35% of Graminaceae, 49% of Fabaceae, 6% of Cruciferae, 10% of other families) at a low stocking rate (16 m2/d); 2) untreated, grazing at a high stocking rate (8 m2/d); 3) treated with bST, grazing at a low stocking rate; and 4) treated with bST, grazing at a high stocking rate. The diets of the ewes were supplemented with vetch and oat hay (500 g/d) and with concentrate (500 g/d). Treatment increased milk production (923.8 vs. 669.5 g/d) but had little effect on fat and protein contents. Administration of bST significantly increased herbage intake; the effect on intake was more marked at the high stocking rate. Under these grazing conditions, the treated ewes reduced selective intake behavior and, thus, achieved good feed intake despite the low biomass availability.  相似文献

Engorged females and 1- to 3-d-old eggs of the lone star tick, Amblyomma americanum (L), were exposed to 9 treatments of pyriproxyfen (4, 8, and 16 micrograms/cm2) in glass vials at exposure periods of 7 d, 14 d, and continuous at each dosage level. Treatment of newly engorged females did not affect the number of females ovipositing, but the number of eggs oviposited decreased as dosage and exposure time increased. Complete inhibition of egg hatch occurred at all treatment levels except that of the lowest dosage and exposure time where 99.9% inhibition was observed. Egg masses from treated females contained eggs that turned a dark amber color and imploded, retained normal shape and color without visible evidence of embryogenesis, or had developed embryos that were unable to emerge. Treatments of 1-to 3-d-old eggs were effective in reducing hatch and larval survivorship to 25 d after emergence only at the continuous exposures of dosages of 4, 8, and 16 micrograms/cm2. Larvae emerging from eggs treated at the lower dosage rates of 0.2 and 0.02 microgram/cm2 were as successful in feeding on chickens and subsequently molting as acetone treated control ticks.  相似文献

In our laboratory, preliminary studies have indicated that recombinant bovine somatotropin (rbST) can stimulate protective immunity against coccidia infection. A floor pen trial on coccidia-seeded litter was run to further test its activity as an adjuvant during immunization of chicks with a live oocyst vaccine. Five hundred day-old male broiler strain chicks were randomly assigned to five experimental Treatments: 1, medicated controls; 2, unimmunized, not treated with rbST; 3, unimmunized, rbST-treated; 4, immunized, not treated with rbST; 5, immunized, rbST-treated. Each treatment consisted of four pens of 25 chicks each. At the end of the growout period (7 wk), the chicks in Treatment 1 (medicated controls) had the highest mean BW, but mean BW of chickens in Treatment 3 (rbST treatment only) were not significantly less. On the other hand, the mean weights of chicks in Treatments 4 (immunized only) and 5 (immunized plus rbST) were significantly reduced, and not different from those of the untreated chickens (Treatment 2). However, when challenged at 3 wk, the chicks in Treatment 5 had a mean combined total lesion score that was significantly lower than that from Treatment 3, indicating that they had developed a higher degree of specific immunity, but of the expense of weight gain. The results suggest that rbST has a potential for use as an adjuvant with live oocyst vaccination, but that the ratio between rbST dose and numbers of oocysts in the live vaccine needs to be carefully controlled.  相似文献

In vitro progesterone (P4) production by hamster luteal cells is stimulated throughout pregnancy by FSH and LH. Prolactin (PRL) by itself, however, increases P4 synthesis only on Day 12; on Day 4, FSH+LH+PRL induces optimal P4 secretion [Biol Reprod 1994; 51:43-49]. In light of these findings, in this study we investigated FSH, hCG, and PRL receptors in hamster CL or dispersed luteal cells on Days 4, 8, and 12 of pregnancy. Scatchard analysis of hamster CL on Days 4 and 8 showed considerably more unoccupied hCG receptors than FSH receptors: on Day 4, there was 9.5 fmol/mg protein for FSH binding sites vs. 1741 fmol/mg protein for hCG binding. Moreover, the binding affinity of hCG was greater than for FSH: the Day 4 Kd was 0.136 nM for hCG vs. 0.308 for FSH. Similar differences were observed on Day 8. Dispersed luteal cells (large+small cells) were incubated for 24 h with or without 10 ng of ovine FSH, LH, and PRL or human recombinant FSH (r-hFSH), alone or in different combinations. The cells were then washed and incubated for 4 h with iodinated hCG, FSH, or PRL with or without 100-fold excess of unlabeled hormones. The number of binding sites per 200,000 luteal cells did not change appreciably for FSH and hCG on Days 4 and 12 of pregnancy, whereas PRL binding sites significantly increased on Day 12.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Coturnix kept in a 14L:10D photoperiod from hatch began to lay their first eggs at a mean age of 42.8 days (range 38-55). Approximately 2/3 of Coturnix held from hatch in photoperiods of 6L:16D light were laying at 165 days of age. Mean age at first egg was 112.7 days (range 68-162 days) in 8L:16D and 130.8 days (range 117-158 days) in 6L:18D photoperiod. Coturnix transferred from a non-stimulatory (8L:16D) photoperiod to a stimulatory one (14L:10D or 24L) begun laying in 15-20 days if less than 140 days old, and in about 5 days if greater than 140 days old, when trasferred. Birds which has spontaneously begun to lay in an 8L:16D photoperiod did not stop laying when the photoperiod was reduced to 6L:18D. Those which began laying under 14L:10D photoperiod ceased laying in about 15 days if 89 or fewer days old when switched to 8L:16D, or in about 6 days if 140 or more days old. Those switched from 14L:10D to 6L:18D ceased laying in about 13 days when 76 days old, and 7 days when 89 days old.  相似文献

In a series of four experiments, the effects of differences in dietary protein intake on BW, skeletal growth and the weight of the Pectoralis major muscle was studied in Hubbard Standard and Hubbard Hi-Y broiler breeder pullets. In Experiment 1, pullets were fed diets with either 15, 17, or 19% CP from 1 to 21 d. There was a linear increase in BW gain and feed consumption with each increase in CP and Hubbard Standard pullets consumed significantly more feed than Hubbard Hi-Y pullets. From 21 to 34 d, pullets were subjected to feed restriction every day (ED) or every-other-day (EOD). On Day 35, pullets were given ad libitum access to feed and ED-restricted pullets consumed significantly more feed than EOD-restricted pullets through 3.5 h postfeeding. In Experiment (Exp.) 2, pullets were fed the 19% CP diet for approximately 14 d (300 g) or 20 d (600 g) before being switched to the 15% CP diet. In Experiment 3, the 19% CP diet was fed for exactly 7 or 21 d before switching to the 15% CP diet. In all experiments, Hubbard Standard pullets weighed significantly more than Hubbard Hi-Y pullets and had increased overall skeletal growth. When the pullets from both strains were restricted to only 380 g BW at 28 d, (Exp. 3) the longer period of protein intake significantly enhanced breast muscle weight but there were no significant strain differences. When BW at 28 d was increased to 480 g, (Exp. 2), dietary protein and strain differences were observed.  相似文献

Sixteen mature, ruminally cannulated wethers (average BW = 41 +/- 1 kg) were fed a low-quality hay diet with or without a cottonseed meal (CSM) supplement and the parasympathomimetic agonist slaframine (SF). Treatments were basal diet (Mitchell grass hay, 4.8% CP, 46.8% ADF) available on an ad libitum basis, basal diet plus SF (8 micrograms/kg BW, 2 x daily i.m. injection), basal diet plus CSM (41.0% CP; 100 g/d), or basal diet plus SF and CSM. Treatments were arranged as a 2 x 2 factorial within a replicated 4 x 4 Latin square with 20-d periods followed by a 10-d adjustment during which only the basal diet was fed. All measurements were performed within the final 10 d of each period. Slaframine increased salivary flow by 10 to 35% (P < .07), ruminal fluid dilution rate by 8 to 11% (P < .10), and pH by 3 to 4% (P < .001). A twofold increase (P < .05) in ruminal cellulolytic bacteria numbers occurred in SF-treated wethers. Despite these SF-induced changes in the ruminal environment, whole-tract apparent nutrient digestibility, N and mineral balance, and ruminal VFA concentrations were not changed. Cottonseed meal increased forage intake by 34 to 54% (P < .001) and DM digestibility by 30% (P < .001). Cottonseed meal supplementation of a Mitchell grass hay diet improved nutritional status and attenuated live weight loss.  相似文献

We have previously shown that changes in glutathione peroxidase-1 (GPX1; H2O2:oxidoreductase, EC 1.11.1.9), plasma thyroid hormone and glutathione-S-transferase were not associated with changes in growth observed in second-generation (F2) severely Se-deficient rats; we also found that liver phospholipid hydroperoxide glutathione peroxidase (GPX4; EC 1.11.1.12) activity falls in first-generation (F1) Se-deficient rats to 41% of levels in Se-adequate rats. The purposes of this study were to determine the effect of F2 Se deficiency on GPX4 and to detect early changes in Se parameters associated with growth after single, small Se injections. Se-deficient male F2 weanling rats were randomly divided into two groups and fed a Se-deficient crystalline amino acid (0.003 microg Se/g diet; -Se) diet or that diet supplemented for 14 d with 0.2 microg Se/g diet (+Se) as Na2SeO3. Growth of -Se rats was 55% of the rate of +Se rats. Liver Se, GPX1 activity, GPX4 activity and testis GPX4 activity in -Se rats at 14 d were 1, 2, 23 and 13%, respectively, of levels in +Se rats. In a series of experiments, additional F2 male weanling rats were fed the -Se diet for 14 d and then were given an intraperitoneal single saline injection of 0, 1 or 5 microg Se/100 g body weight (BW) as Na2SeO3 and killed 1 or 7 d later. Rats injected with 1 or 5 microg Se/100 g BW grew 36 or 48%, respectively, above the rate of saline-injected rats. Liver Se concentration increased 367% and testis GPX4 activity doubled in rats 1 d after injection of 1 microg Se/100 g BW compared with saline-injected rats; these parameters were further elevated with 5 microg Se/100 g BW injections. Increases in liver Se and testis GPX4 activity were the parameters best associated with improved growth after Se injection, but the molecular role for Se in growth remains unclear.  相似文献

Novel male house mice (Mus musculus) can disrupt early pregnancy in females. Previous research (e.g., A. E. Storey; see record 1986-16382-001) focused on pheromonal rather than behavioral mediation of this phenomenon. In Experiment 1, novel males were housed with females shortly after insemination. Litter production was negatively correlated with the males' sexual activity. Experiment 2 replicated this finding with a larger sample. In Experiment 3, females were exposed to castrated males. Testosterone-treated males completely blocked pregnancy, whereas untreated males did not. In Experiment 4, castrated testosterone-treated males were presented at intervals after insemination. Pregnancy was totally blocked at Days 3 and 4 and mostly blocked at Days 1 and 2 but was less affected at Days 5 and 6. In Experiment 5, females were exposed through a wire-mesh grid to castrated males. Pregnancies occurred in all conditions, even with testosterone-treated males. An experiment was conducted to quantify genetic differences in response to dietary protein level of male vs female broilers. Broilers (1 d old) from a "high-yield" strain cross (Ross x Ross 208) and a "fast-growing" strain cross (Peterson x Arbor Acres) were placed on fresh pine shavings in floor pens. From Day 0 to 18, all birds were fed a 23% CP starter diet. During Days 18 to 53 male birds were fed either a 16, 18, 20, 22, 24, or 26% CP diet (3,200 kcal ME/kg) and female birds were fed the 16, 20, or 24% CP diet. Significant differences (P < 0.05) were noted in the performance of the different strains. Ross x Ross 208 male birds had a higher body weight (3.37 vs 3.16 kg), higher feed intake (7.08 vs 6.78 kg), higher breast yield (31.76 vs 29.25%), higher carcass yield (73.90 vs 71.85%), and a lower adjusted feed conversion ratio (FCR; 2.10 vs 2.16 g:g) than Peterson x Arbor Acres males at 53 d of age. As compared to Peterson x Arbor Acres females, Ross x Ross 208 female broilers also had a higher body weight (2.68 vs 2.55 kg), higher breast meat yield (33.61 vs 30.80%), higher carcass yield (75.31 vs 73.91%), and lower adjusted FCR (1.97 vs 2.04 g:g). Qualitative differences in the response of these strains were confirmed and better qualitative data is presented that can be used to predict the important output parameters from the import inputs in broiler production.  相似文献

We used a new injury device that produces consistent spinal cord contusion injuries (T8) in rats to compare the behavioral and histologic effects of methylprednisolone sodium succinate (MPSS) administration, the clinical standard of therapy after acute spinal cord injury (ASCI), with the 21-aminosteroid, U-74006F (U74), and the TRH analogue, YM-14673 (YM), at different trauma doses. Three sequential experiments were conducted: Experiment 1. U74 (3.0/1.5/1.5 mg/kg; 10/5/5 mg/kg; 30/15/15 mg/kg), MPSS (30/15/15 mg/kg), or vehicle were administered intravenously (i.v.) at 5 min, 2 and 6 h after the injury (n = 8/group). U74 (10/5/5 mg/kg) and MPSS animals scored better than controls (Days 8-43) in open field walking (OFW); no other differences were seen between groups. Experiment 2. Dose-response evaluation of MPSS determined more effective doses. Groups (n = 16) receiving 30/30/30/30 mg/kg and 60/60/60/60 mg/kg i.v. at 5 min and 2, 4, and 6 h after the injury had better OFW scores than controls (Days 8-29; Day 29). Both groups performed better than controls (Days 8-29) on inclined plane (IP); 30 mg/kg animals scored higher on Day 29. Percentage tissue spared (%TS) at the lesion center was greater for 60 mg/kg animals (23.4%) than controls (17.3%). Experiment 3. Compounds were administered as in experiment 2 (n = 15/group); MPSS (60/30/30/30 mg/kg) and YM (1/1/1/1 mg/kg and 1 mg/kg/day ip) were most effective. YM and MPSS combination produced no additive effects. YM animals scored better than MPSS and control animals in OFW (Days 8-29) and better than controls on IP (Days 8-29; Day 29) and grid walking (Day 29). MPSS animals scored better than controls on IP (Days 8-29). YM and MPSS groups had greater %TS than controls. This series of experiments demonstrates the utility of this injury model and simple behavioral measures for preclinical assessment of pharmacologic agents. Under these experimental conditions, U74 demonstrated equivalent efficacy to MPSS, and YM demonstrated greater efficacy than MPSS in the treatment of ASCI.  相似文献

The effects of constant photoperiod on serum prolactin concentrations and sexual behavior of ovariectomized goats was investigated. Fourteen ovariectomized goats were divided into groups of seven goats each and placed in photoperiod-controlled rooms with either 8L:16D or 16L:8D. All goats received six estradiol (E2) treatments in a Latin square design. Following each treatment six 1-h observation periods were conducted. Blood was collected before E2 treatment and at each observation period for prolactin quantification. During the observation periods measures of attractivity, proceptivity, and receptivity were recorded. Goats exposed to 8L:16D displayed sexual behavior in response to doses of E2 lower than those required by goats exposed to 16L:8D. Prolactin concentrations were higher in goats exposed to 16L:8D than in those exposed to 8L:16D. It was concluded that photoperiod affects prolactin concentrations and behavioral sensitivity to E2 in goats. The effect of continued exposure to constant photoperiod on these variables was then investigated. The goats were held in constant photoperiod for 211 d and behavior tests as described for Exp. 1 were repeated. Scores for attractivity, proceptivity, and receptivity did not differ between the two groups after 211 d of exposure to constant photoperiod. Prolactin concentrations did not differ in this study between the two groups. It was concluded that after chronic exposure to 8L:16D ovariectomized goats become refractory to the stimulatory effects of short photoperiod.  相似文献

We conducted experiments to determine whether lambs fed grain prefer foods and solutions containing sodium bicarbonate (NaHCO3) and lasalocid, compounds capable of attenuating acidosis. In Exp. 1, we determined whether lambs fed barley preferred flavored rabbit pellets (RP) containing NaHCO3 and lasalocid. Lambs in two groups (n = 10/group) were fed increasing amounts of barley on d 1 to 12 (300 to 1,100 g) and again on d 23 to 34 (300 to 1,350 g). After ingesting barley on d 1 to 12, lambs were fed ground RP containing lasalocid and NaHCO3 (i.e., medicated) and flavored with either 2% onion (group 1) or 2% oregano (group 2). During d 23 to 34, lambs were fed unmedicated RP containing NaCl and flavored with either 2% oregano (group 1) or 2% onion (group 2). During preference tests on d 35 to 40, lambs fed grain preferred RP with NaHCO3 to RP with NaCl (151 vs. 96 g; P < .01). In the Exp. 2, we determined whether wheat ingestion affected consumption of aqueous solutions containing NaHCO3. In trial 1, 28 lambs were assigned to four treatments: 1) low-wheat + 2% NaHCO3, 2) high-wheat + 2% NaHCO3, 3) low-wheat + water, and 4) high-wheat + water. For 12 d from 0800 to 0830, lambs in treatments 1 and 3 were fed 300 g of wheat and lambs in treatments 2 and 4 were fed up to 1,300 g of wheat; fluids (NaHCO3 and water) were then offered from 0930 to 1230 daily. Lambs drank more NaHCO3 on the high- than on the low-wheat diet (1,332 vs 890 g; P = .03); water consumption was similar for lambs on the high- and low-wheat diets (1,675 vs 1,700 g; P > .10). In trial 2, lambs in treatments 3 and 4 were offered a solution containing 1.4% NaCl. For 13 d from 0800 to 0830, lambs in treatments 1 and 3 were fed 500 g of wheat and lambs in treatments 2 and 4 were fed up to 1,700 g of wheat. Lambs had access to fluids from 0800 to 1200 daily. Lambs drank nearly twice as much NaHCO3 solution on the high- than on the low-wheat diet (1,066 vs 572 g), whereas they drank only 1.4 times more NaCl solution on the high- than on the low-wheat diet (888 vs. 634 g; P < .001). Fewer lambs showed signs of acidosis in treatment 2 than in treatment 4 in trials 1 (2 vs 9) and 2 (7 vs 17). Collectively, these results are consistent with the hypothesis that lambs fed grain prefer substances that attenuate acidosis.  相似文献

Production of T3 from T4 in tissues is catalyzed by two 5'-deiodinases, type I (D1) and type II (D2), but the quantitative contribution of each pathway to whole body T3 production is not well established. In the presence of propylthiouracil (PTU), D1, but not D2, can be effectively blocked, providing an experimental probe for addressing this problem. Decades ago, this approach provided indirect estimates ranging from 23-44% contribution by D2, based on plasma T3 appearance rate comparisons (PAR3 = PCR3 [T3]p) in periodically T4-injected athyreotic rats vs. controls. Two, more recent studies, using constant infusions of T4 for replacement, achieved 22% and 65% estimates, respectively, from PAR3 comparisons. We have revisited this problem more directly and precisely, with two major differences in experiment design. We used direct whole body steady state measurements of T3 production, instead of indirect plasma-only data (PAR3). We also used (euthyroid) physiological doses of both T4 (0.9 microg/day x 100 g BW) and T3 (0.15 microg/day x 100 g BW) for replacement in two thyroidectomized rat groups, instead of T4 only, in a 7-day constant steady state, dual tracer infusion protocol. The first group also had chronically implanted 150-mg PTU pellets (TXR-PTU); the other had implanted 0.1 N NaOH placebo pellets (TXR-EU); each delivered their product at constant rates. A third euthyroid intact group was used as the controls. The completeness of D1 inhibition was ascertained in a fourth group, identically treated with 150-mg PTU pellets, in which negligible D1 activity was found in liver and kidney using labeled rT3 as substrate for the 5'-D assays and minimal (1 mM) dithiothreitol as cofactor. In the TXR-PTU group, the percentage of T4 converted to T3 was 11.8%, compared with 23.4% (P < 0.0005) in the TXR-EU group, and 22.7% (P = NS) in controls. Previous studies on crustacea have demonstrated significant diurnal rhythms in blood glucose. However, glucose concentration in the blood of food-deprived Penaeus monodon, held in indoor or outdoor tanks, did not exhibit a diurnal rhythm under photoperiods of 8 h light and 16 h darkness (8L: 16D) or under a 13.5L: 9.5D photoperiod, with simulated or natural full moon conditions. Prawns held on photoperiods of constant light, 20L : 4D, 16L : 8D, 12L : 12D, 8L : 16D, 4L : 20D, or continuous darkness did not have significantly different mean blood glucose levels. Mean blood glucose levels varied between 0.77 and 1.39 mmol/L, depending on conditions. Pronounced and significant increases in blood glucose levels occurred within 20 min of feeding, with peak levels after 100 min. The rise in blood glucose level observed after feeding was independent of the eyestalks, and hence putative crustacean hyperglycaemic hormone, and was not from endogenous carbohydrate stores. Under appropriately controlled conditions, blood glucose concentrations can be used as an index of nutritional status in penaeid prawns.  相似文献

High-dose cyclophosphamide (HD-CY) has been shown to decrease the tumor mass in multiple myeloma (MM) patients and to be effective in the mobilization of PBPC. By administering hematopoietic growth factor the quantity of progenitor cells in the peripheral blood increased and the hematological toxicity of CY could be reduced. Thirty-two patients with stage II and stage III MM were treated to mobilize and harvest a sufficient amount of PBPC for autologous transplantation. Sixteen patients received 4 g/m2 CY and 16 patients 7 g/m2 CY in divided doses of 1 g/m2 every 2 h. Both patient groups were comparable for disease stages as well as previous therapies. Twenty-four hours after chemotherapy 300 micrograms GCSF were administered subcutaneously once daily until the last day of leukapheresis. Administration of 7 g/m2 HD-CY resulted in statistically significantly higher peak values for CD34+ progenitor cells (47.86/microliters vs 18.75/microliters, P = 0.0198) in the peripheral blood. PBPC autografts containing > 2.5 x 10(6) CD34+ cells/kg BW could be obtained at the first attempt from 14 of 16 patients treated with 7 g/m2 CY as compared to 10 of 16 patients treated with 4 g/m2 CY (P = 0.11). The analysis of potentially malignant CD19+ B cells showed a highly significant lower mean CD19+ cell content/kg BW per leukapheresis in the 7 g/m2 compared to the 4 g/m2 CY group (0.75 vs 1.81 x 10(6), P = 0.001). WHO grade IV treatment-related non-hematologic toxicity was not observed. We prefer the 7 g/m2 CY dosage followed by cytokine administration for the mobilization of PBPC in advanced state MM patients pretreated with alkylating agents.  相似文献

An experiment was performed to determine the effect of elevated prostaglandin F2 alpha (PGF2 alpha) on pregnancy rates of progestogen-treated bred cows in the presence or absence of luteal tissue. Ninety-one beef cows were bred (Day 0) and assigned randomly to receive either 3 mL saline (CON), 15 mg PGF2 alpha, or 15 mg PGF2 alpha + lutectomy (P + L) administered intramuscularly (i.m.) at 8 h intervals on either Days 5-8, 10-13, or 15-18 postbreeding. Lutectomies were performed by transrectal digital pressure before initiation of treatment on Day 5, 10, or 15 for the respective treatment groups. All cows were fed 4 mg/day of melengesterol acetate from two days prior to initiation of treatment until Day 30 postbreeding. Mean concentrations of 13,14-dihydro-15-keto-PGF2 alpha (PGFM) were increased in cows administered PGF2 alpha and P + L treatments (398 +/- 23 and 413 +/- 22 pg/ml, respectively; p < 0.01) compared to the CON group (80 +/- 29 pg/ml) regardless of treatment group. Mean concentrations of oxytocin (OT) were increased in cows given PGF2 alpha on Day 10 and 15 (p < or = 0.0001) and tended to be increased on d 5 when compared to CON and P + L treatment groups on Day 5. Pregnancy rates were reduced (p < or = 0.03) in the PGF2 alpha treatment group (23%) and by Day 5-8 compared to CON (72%). Lutectomy tended to improve pregnancy rate in P + L (5-8; 55%) compared to PGF2 alpha (5-8; p = 0.1). Pregnancy rates tended (p < or = 0.07) to increase in the PGF2 alpha treatment groups on Days 5-8 treatment (23%, 50%, and 60% for Days 5-8, 10-13, and 15-18, respectively). The later the treatments were initiated pregnancy rates did not differ between treatments given on Days 10-13 and 15-18. In conclusion, the most susceptible period of embryonic growth to the negative effects of PGF2 alpha was during morula to blastocyst development. Removal of luteal tissue diminishes the negative effects of PGF2 alpha through interruption of the luteal oxytocin-uterine PGF2 alpha feedback loop.  相似文献
