Olfactory perception of pheromones and host odors byDendroctonus pseudotsugae males and females was investigated through single cell recordings. Responses of 71 cells (35 cells in males; 36 cells in females) were recorded to 1 g of 10 pheromones and three host terpenes. The olfactory cells were classified into four types based on sensitivity and specificity for the various odors: (1) 3,2-MCHone (3-methyl-2-cyclohexenone) cells; (2) 3,2-MCHol (3-methyl-2-cyclohexenol) cells; (3) frontalin cells; and (4) synergist cells. 3,2-MCHone and 3,2-MCHol cells in both males and females were the most sensitive and specific of all cells recorded with a threshold ca. 0.001–0.01 g on filter paper. Frontalin cells were less specialized than the two aforementioned cell types and had a somewhat higher threshold of ca. 0.01 g. A larger percentage of these three cell types were recorded in males than in females. The synergist cells had the highest threshold of all recorded cell types and were the least specific. Each of these cells was most responsive to eithercis-verbenol,trans-verbenol, 1,2-MCHol, verbenone, -pinene, or limonene; however, responses could be elicited by stimulation with other pheromones or host terpenes at higher dosages. Instances of inhibition of nerve impulse activity from spontaneously active cells were observed during 3,2-MCHol and 3,2-MCHone stimulation, thus suggesting multifunctional effects for these compounds. The results help explain the role of chemicals in host selection, aggregation, and colonization behavior of the beetle and further elucidate hypotheses put forth in an earlier electroantennogram study.Texas Agricultural Experiment Station Paper NumberTA-18689. When both sexes of the bark beetle,Dendroctonus brevicomis LeConte, were exposed to vapors of myrcene from ponderosa pine, only the male produced (+)-ipsdienol. In the field, racemic ipsdienol significantly reduced the attraction of both sexes in flight to a mixture of myrcene and the aggregation pheromones,exo-brevicomin and frontalin. This suggests that ipsdienol may be involved in regulating colonization density ofD. brevicomis. The implications of the biosynthesis of various enantiomers of ipsdienol byD. brevicomis and the cohabitating bark beetles,Ips paraconfusus andI. pini, in relation to their behavioral responses are discussed in regard to reducing interspecific competition.Coleoptera: Scolytidae.  相似文献

Quantities of attractive (exo-brevicomin and frontalin) and inhibitory (trans-verbenol, verbenone, and ipsdienol) pheromones were monitored in both sexes ofDendroctonus brevicomis during their colonization of a ponderosa pine. Verbenone was found in males in the greatest amounts at the time of landing, and it declined more rapidly than the other pheromones in either sex. The amounts of frontalin andexo-brevicomin in males and females, respectively, increased after initial boring within the host but began to decline after mating. The quantity oftrans-verbenol in both sexes (females had significantly more) declined more gradually thanexo-brevicomin, frontalin, and verbenone. Ipsdienol was found only in males during the initial stages of attack when encountering the resin. It is suggested that along with a general decline in all pheromonal components, a sufficient change in the ratio of the attractive pheromones to an inhibitory pheromone,trans-verbenol, may play a role in termination of aggregation.trans-Verbenol may also function along with verbenone and ipsdienol in limiting the density of attack and thus intraspecific competition. These inhibitory pheromones also appear to cause several competing species of bark beetle to avoid landing in areas infested withD. brevicomis, even when their own pheromone is present.Coleoptera:Scolytidae. In bioassays conducted with walking beetles in the laboratory (S)–(+)–, (R)-(–)-, and (±)-ipsdienol were attractive alone, but reduced the attraction of both sexes of the mountain pine beetle,Dendroctonus ponderosae Hopkins, to extracts of female frass. Field trapping studies indicated that attraction ofD. ponderosae to a stimulus composed of myrcene +trans-verbenol +exo-brevicomin was significantly reduced with the addition of (±)- and sometimes (S)-(±)-ipsdienol. Thus, (S)-(+)-ipsdienol produced by males of this species may act as an antiaggregation pheromone. (S)-(+)-Ips-dienol is thought to function as a repellent allomone against the pine engraver,Ips pini (Say), in regions whereI. pini utilizes (R)-(–)-ipsdienol as an aggregation pheromone. However, in southwestern British ColumbiaI. pini was attracted to the (±)-ipsdienol used in field bioassays ofD. ponderosae, a finding consistent with the production of both enantiomers byI. pini in this region. When presented with the ternary semiochemical bait forD. ponderosae, (±)-ipsdienol was not attractive toI. pini. Electroantennograms (EAGs) were obtained forD. ponderosae to the bark beetle pheromonestrans-verbenol,cis-verbenol,exo-brevicomin,endo-brevicomin, frontalin, verbenone; to the kairomones, -pinene, -pinene, camphene, -3-carene, limonene, myrcene; to a blend (111) oftrans-verbenol,exo-brevicomin, and myrcene; and to diacetone-alcohol. Male and female responses, in general, did not differ significantly over the whole EAG dose-response curves but differed at a few concentrations on many of the curves. There were more differences noted for pheromones than kairomones. The blend yielded among the largest EAGs in both sexes and appeared to show synergism. Responses of females were lower than those for males in most instances. Significant differences in responses by the two sexes were much fewer for the kairomones than the pheromones. EAG recovery rates tested at only one concentration showed significant differences between males and females for three pheromones,trans-verbenol,cis-verbenol, and verbenone, and two kairomones, camphene and -3-carene. Thresholds were quite low for most of the odorants exceptcis-verbenol, camphene, verbenone, and diacetone-alcohol in females, andcis-verbenol, verbenone, -pinene, and diacetone-alcohol in males. The results, using at least one EAG parameter, support behavioral and field studies involvingexo-brevicomin,trans-verbenol, frontalin, and the blend.  相似文献

Antennal olfactory (electroantennogram) and laboratory and field behavioral tests were carried out on the response ofDendroctonus frontalis to its aggregation pheromone frontalin and analogs. The analogs were compounds modified by altering the position and methyl groups and/or by their deletion. Any modification to the frontalin structure significantly reduced both the antennal olfactory and behavioral response byD. frontalis. Beetle response, although significantly reduced, was elicited at the receptor level and in a laboratory bioassay by all analogs. However, only one analog (endo-5,7-dimethyl-Frontalin) elicited significant response from field populations of the beetle.Texas Agricultural Experiment Station paper TA-22507. Volatiles were found to be distributed throughout adult Douglasfir beetles,Dendroctonus pseudotsugae Hopkins, suggesting that the hindgut may not be the sole site of production. The volatile content of individual beetles increased dramatically with feeding; qualitative changes reflected the different bark habitats of newly emerged and established beetles. All detected volatiles were also found in beetle frass. Myrcene was the predominant monoterpene of emergent beetles and -pinene of fed beetles. Linalool and bornyl acetate occurred in significant amounts after feeding, and diacetone alcohol and sulcatol were also detected for the first time. The presence of sulcatol explains the reported cross-attraction withGnathotrichus spp.cis-Verbenol was also found to occur in addition totrans-verbenol. The volatile content of male beetles was similar to that of females but amounts of individual compounds were less. Acetic acid was found in both emerged and fed beetles and in lesser amounts in frass, suggesting that microbial metabolism could be a potential source of volatile production.  相似文献

Electroantennograms (EAGs) from male and femaleIps avulsus, I. calligraphus, andI. grandicollis to their pheromones and selected host odorants or kairomones verified the presence of antennal olfactory receptors in both sexes of each species capable of detecting ipsdienol, ipsenol,cis- andtrans-verbenol,endo-brevicomin -pinene, frontalin, and verbenone. Each species possesses receptors with lower thresholds and in greater abundance for the compounds they produce and to which they are behaviorally most responsive. Detection of bothIps andDendroctonus pheromones by the three cohabiting species provides a sensory basis for olfactory interactions among the species. exo-brevicomin, a multifunctional pheromone of the mountain pine beetle,Dendroctonus ponderosae, was tested at release rates of 0.5 and 2.5 mg/day alone and in combination with the antiaggregation pheromone verbenone against unbaited controls. Significantly more lodgepole pinePinus contorta var.latifolia trees were attacked, and at higher densities, with both release rates ofexo-brevicomin than with all other treatments. Verbenone significantly reduced the response of mountain pine beetles toexo-brevicomin. Verbenone alone did not reduce the number of trees attacked by mountain pine beetle or the attack density when compared to the unbaited controls.  相似文献

Dendroctonus brevicomis was attracted to a mixture of theIps paraconfusus pheromones, ipsenol,cis-verbenol, and ipsdienol at 10–9 g each/l but was not attracted to these pheromones at higher and lower release rates.I. paraconfusus was not attracted to theD. brevicomis pheromonesexo-brevicomin, frontalin, and myrcene at any release rate tested. Increased release rates of a mixture of the three pheromones ofI. paraconfusus inhibited the attraction ofD. brevicomis to its synthetic pheromones. A mixture of ipsenol + ipsdienol orcis-verbenol alone failed to cause inhibition indicating that at least two of theI. paraconfusus pheromones are required to inhibit the response ofD. brevicomis. Olfactory perception of pheromonal enantiomers by male and female Douglas-fir beetles,Dendroctonus pseudotsugae Hopk. (Coleoptera: Scolytidae), was investigated by electrophysiological techniques and behavioral bioassays. Electroantennograms (EAGs) and single-cell responses indicated both sexes to be more responsive to racemic frontalin and the (–)-enantiomer at lower dosages. At higher dosages, little difference was noted in responses to either enantiomer. However, response to the racemic mixture at higher dosages was slightly greater than responses to either enantiomer alone. In laboratory behavioral bioassays, responses to low concentrations of (–)-frontalin and the racemic mixture exceeded response to the (+)-enantiomer alone. At a higher concentration, responses to the racemic mixture or either enantiomer alone did not differ. Release of the antiaggregation pheromone, verbenone, at 3.8 mg/ day from a concentrated source within a multiple-funnel trap completely inhibited response by the mountain pine beetle (MPB),Dendroctonus ponderosae Hopkins, to attractive semiochemical lures. When aerial applications were simulated and verbenone was released at the same rate from beads lying in a 2×2-m area on the forest floor 15–35 cm below a trap, the response of the MPB was inhibited by only 50%. This reduced inhibition may be explained in part by the photoisomerism of verbenone. When exposed to full sunlight on two occasions, the times required for 50% of verbenone vapors to be converted to chrysanthenone were 75 and 100 min, respectively. Trap and tree-baiting experiments indicated no biological activity of chrysanthenone. Rapid photoisomerization could reduce the concentration of verbenone below biologically active levels and would allow the MPB to colonize trees close to already occupied hosts, contributing to the characteristic clumped distribution of MPB attack. The rate of verbenone photoisomerization may vary according to geographic location, stand elevation and density, and should be considered before verbenone is applied to control the MPB and other bark beetles.  相似文献

The response of the European elm bark beetle,Scolytus multistriatus, to host bacterial isolates was studied qualitatively under field conditions. Initial experiments indicated that such isolates were attractive to in-flight beetles. These isolates, identified asBacillus subtilis (five strains),B. pumilus, andEnterobacter cloacae, were grown on nutrient agar in glass vials and attached to sticky traps in elm woods. Although beetles were caught on the bacterial isolate-baited traps, the catches were variable, inconsistent, and often contradictory from one experiment to another. High numbers ofS. multistriatus were caught on traps baited with three strains ofB. subtilis, but in addition to thesubtilis strains, there were also aerial contaminants in the treatments, namelyE. aerogenes, Corynebacterium sp., andFlavobacterium sp. Also, relatively high catches were recorded on nutrient agar controls. When elm wood-bark plugs, sterilized (by gamma irradiation) and unsterilized, were placed in vials with the host bacterial isolates, the presence or absence of fresh elm, gamma irradiated or not, had no noticeable effect on beetle attractancy.Coleoptera: Scolytidae.  相似文献

Females of a pinyon pine bark beetle,Ips hoppingi Lanier, were less attracted by the aggregation pheromone produced by conspecific males than by the pheromone produced by the neighboring sibling species,I. confusus (LeConte). Cross-attraction was elicited by males infesting the regional pinyon pine hosts (P. discolor andP. edulis) of eitherIps species in south-eastern Arizona. Electroantennograms in two populations ofIps pini (Idaho and New York) were very similar, even though previous work documented behavioral specificity and revealed that the populations produce different ratios of the enantiomers of the aggregating pheromone ipsdienol. The insect- and host-produced compounds tested evoked characteristic response curves in both populations. EAG amplitude was highest to ipsdienol and the aggregation inhibitor ipsenol. Among the other compounds tested, responses were generally higher to the beetle-produced odorants (linalool, verbenone,trans-verbenol) than to host terpenes (1-octanol, -pinene). Electroantennograms (EAGs) were recorded fromD. ponderosae to the enantiomers of the terpenoid bark-beetle pheromonestrans-verbenol,cis-verbenol, verbenone, and the bicyclic ketals frontalin,exo-brevicomin, andendo-brevicomin. Male and female responses to enantiomers of the terpenoids differed significantly only at the two highest concentrations. No sex differences were seen in response to the bicyclic ketals. Significantly different responses to the enantiomers of all the chemicals, except frontalin, were noted over at least part of the dosage-response ranges tested. The negative antipode for all of the terpenoids elicited higher responses, while for the bicyclic ketals, the positive antipode effected the largest responses except for the two highest concentrations ofexo-brevicomin.  相似文献

Electrophysiological recordings inIps pini were made from single olfactory cells stimulated by six concentrations of eleven compounds—either pheromones or host constituents. The receptor cells were grouped according to their differential responses to these biologically significant compounds. Cells specialized to either ipsdienol or ipsenol (pheromones for severalIps species) appeared to be relatively uniform in their sensitivity to all compounds tested. Another group of cells specialized tocis- andtrans-verbenol were more variable in their sensitivities to the substances tested. The activation of separate groups of cells by the different pheromones and host compounds shows (by exclusion) that behaviorally expressed syner-gistic as well as inhibitory action mainly is due to an interaction in the central nervous system rather than an interaction of the compounds on the receptors. All ipsdienol cells responded to both enantiomers of ipsdienol. Although some differences between responses to the enantiomers by individual cells were observed, these did not form a consistent pattern, and no net differences could be found between the eastern and western populations ofI. pint.Coleoptera: Scolytidae.  相似文献

Ponderosa pine logs infested withIps paraconfusus males inhibited the attraction ofDendroctonus brevicomis in the field to either attractive logs cut from a ponderosa pine tree under attack byD. brevicomis or to their synthetic pheromones,exo-brevicomin, frontalin, and myrcene. Logs cut from trees under attack byD. brevicomis inhibited the response ofI. paraconfusus to logs infested with maleI. paraconfusus.Exo-brevicomin, frontalin, and myrcene did not inhibit their response but verbenone did. Verbenone was found in maleD. brevicomis dissected from attractive logs under attack during the same time the response ofI. paraconfusus was inhibited by these logs.Trans-verbenol andexo-brevicomin were found in femaleD. brevicomis while verbenone,trans-verbenol, and frontalin were found in maleD. brevicomis in relatively large amounts near the beginning of the aggregation phase of host colonization. All of these compounds had decreased at a similar rate 5 days later. This gradual decrease inexo-brevicomin and frontalin probably caused the observed reduction in attraction. The ecological significance of these compounds in relation to termination of the aggregation phase ofD. brevicomis and reduction of interspecific competition is discussed.Coleoptera: Scolytidae. In laboratory and field bioassays, the response ofDendroctonus frontalis was significantly greater to the mixture of (1S, 5R)-(–)-frontalin andalpha-pinene than to (1R,5S)-(+)-frontalin andalpfa-pinene. Electro-physiological studies revealed that antennal olfactory receptor cells were significantly more responsive to (1S, 5R)-(–)-frontalin than to (1R, 5S)-(+)-frontalin. The antennal response ofDendroctonus valens to host monoterpenes from the resin of ponderosa pine was studied using the electroantennogram (EAG) technique. Male and female beetles were given a single dose of each of 11 different monoterpenes. Response amplitude to the different compounds did not vary between sexes and was generally well correlated with results from field attraction studies. Response to (S)-(–)--pinene was greatest. The relative amplitude of the responses to the (R)-(+) and (S)-(–) enantiomers of a-pinene, however, were reversed from their relative attractiveness in the field. A dose-response study was conducted for the (R)-(+) and (S) -(–) enantiomers of a-pinene, plus a reciprocal differential saturation test with successive doses of first one enantiomer of-pinene and then the other. Comparison of EAG traces suggests different receptors for the two stereoisomers of-pinene. Differential saturation curves suggest that while one set of receptors may respond to both enantiomers, some receptors respond only to the (S)-(–) enantiomer.  相似文献
