There is an increasing global trend towards efficiently utilizing agricultural biomass. Tomato leaf waste is an unexploited source of bioactive molecules, and we aimed develop methods for extracting compounds of high commercial value. In particular, tomato leaf represents a rich source of phylloquinone (vitamin K1), which has a variety of agricultural and health applications. Phylloquinone was efficiently recovered from tomato leaf waste (29.17 ± 0.96 μg g−1) by a solvent-free process that employed high pressure CO2 at 180 bar pressure and room temperature. This recovery rate for vitamin K1 is higher than traditional extraction processes that involve using organic solvents. Tomato leaf also contained phenolics (240 mg (GAE) g−1) and flavonoids (184 mg Qe g−1). A high level of protein (24.47 ± 0.38%) could be recovered from tomato leaf, with aspartic acid, glutamic acid, and leucine being the dominant free amino acids (13 ± 0.1, 15.1 ± 0.2, 12.8 ± 0.1 mg g−1 dry weight). In summary, tomato biomass is a low cost source of high-value compounds that can be purified for the production of nutritional foods and nutraceuticals.  相似文献

BACKGROUND: Recently much attention has been paid to biologically active plants because of their low production cost and fewer adverse effects compared with chemical drugs. In the present investigation the bioactivity of Phyllanthus niruri ethanol and aqueous extracts was evaluated in vitro. RESULTS: The ethanol extract of P. niruri showed a high level of flavonoid content (123.9 ± 0.002 mg g?1), while the aqueous extract showed the highest 2,2‐diphenyl‐1‐picrylhydrazyl (DPPH; IC506.85 ± 1.80 µmol L?1) and 2,2′‐azino‐bis(3‐ethylbenzothiazoline‐6‐sulfonic acid) (ABTS; 46.44 ± 0.53 µmol L?1) free radical scavenging activities with high phenol content (376 ± 0.02 mg g?1) and elevated levels of ferric reducing antioxidant power (FRAP; 23 883 ± 0.019 mmol g?1) with excellent antibacterial activity against Staphylococcus aureus (20 mm inhibition zone) and Streptococcus agalactiae (12 mm inhibition zone), respectively, in addition to the best immune activation potential of human peripheral blood mononuclear cells (450.5%). CONCLUSIONS: It is clear from our results that both extracts of P. niruri has excellent bioactivity roles via elevated levels of antibacterial, antioxidant and percentage of peripheral blood mononuclear cell proliferation, which could lead to the development of medications for clinical use. The objective of this study was to optimise drying conditions for the extractions of β‐carotene, phenolic and ascorbic content from yellow‐fleshed sweet potato using response surface methodology. A face‐centred cubic was used to investigate the effects of three independent variables namely drying temperature 55–65 °C, citric acid concentration 1–3%w/v and soaking time 1–3 min. The optimal conditions for parameters were 55–62 °C, citric acid concentration 1.08–2.19% and soaking time 1.53–2.00 min. Under the above mentioned conditions, the experimental β‐carotene content was 14.61 mg g?1, total phenolic and ascorbic acid content were 4.0 and 17.53 mg g?1, respectively, which were close to the predicted values. Therefore, the results showed that optimise conditions could be used to enhance the antioxidant activities of functional foods.  相似文献

Decanoic acid reverse micelle-based coacervates were used to provide simple, rapid and almost solventless extraction of bisphenol A (BPA) from canned fatty foods. The procedure involved the extraction of 200–400 mg of homogenised food sample with an aqueous solution containing 20% THF and 200 mg of decanoic acid, conditions under which the coacervate (around 550 µl) formed in situ and instantaneously. The overall sample treatment took about 30 min and several samples could be simultaneously treated using conventional laboratory equipment. No clean-up or solvent evaporation were required before determination of BPA by liquid chromatography and fluorescence detection. Recoveries in samples were between 90 and 99%, with relative standard deviations in the range 2–7%. The limit of quantification ranged 29–15 ng g?1 for 200–400 mg of sample, being far below the current specific migration limit (SML) set by the European Commission (600 ng g?1). The method was successfully applied to the determination of BPA in the solid content of canned fish (from 20 to 129 ng g?1) and meat (from undetected to 37 ng g?1).  相似文献

BACKGROUND: Total phenolics content, antioxidant activity and cytotoxicity of the methanol extracts from leaf parts of 13 Korean traditional salad plants were investigated in order to determine their properties. RESULTS: The highest phenolics content (mg ferulic acid equivalents kg?1 dry weight (d.w.), omit one) was found in methanol extracts from Polygonum aviculare, at 293.7 ± 6.0, followed by Euonymus alatus, at 250.7 ± 3.3, Saxifraga stolonifera, at 125.0 ± 8.1 and Ligularia fischeri, at 122.5 ± 5.9. The methanol plant extracts dose‐dependently increased free radical scavenging activity. Methanol extracts of Polygonum aviculare, Euonymus alatus and Saxifraga stolonifera, at 31 mg kg?1, exhibited the highest 1,1‐diphenyl‐2‐picrylhydrazyl (DPPH) radical scavenging activity (%) by 90.8 ± 4.2, 85.7 ± 3.9 and 64.1 ± 3.2, respectively. According to 3‐(4,5‐dimethylthiazol‐2‐yl)‐2,5‐diphenyltetrazolium bromide (MTT) assay, the methanol extracts from Portulaca oleracea (IC50 < 25.0 µg mL?1) showed the highest cytotoxicity against Calu‐6, followed by Plantago asiatica (49.2 µg mL?1) and Osmunda japonica (89.6 µg mL?1). CONCLUSION: Total phenolics content of the tested plant extracts was correlated with the DPPH radical scavenging activity, suggesting the phenolics compounds are contributing to the antioxidant properties of Korean salad plants. The leaf parts of the 13 Korean traditional salad plants described here that are currently used as foods may also provide some benefit to human health, and research into their potential benefits as preventative and/or therapeutic agents is warranted. BACKGROUND: The conventional method of processing ginseng (Panax ginseng) roots into red ginseng involves mainly heating and drying processes. In the present study, this method was modified by using high hydrostatic pressure (HHP) to improve the physicochemical characteristics of red ginseng. RESULTS: The HHP process (600 MPa for 1 min) significantly improved the histological properties of red ginseng by increasing cellular disruption and release of cell contents. The total reducing sugar content was significantly (P < 0.05) higher (increased from 10.67 to 15.25 mg g?1) in red ginseng processed at 600 MPa for 1 min. Similarly, the total free amino acid content also increased significantly (from 2.81 to 7.77 mg g?1). The HHP process resulted in superior and more even colouration and gave an attractive visual appearance to red ginseng. The optical density at 420 nm and Hunter's colour a value (redness) of extracts prepared from red ginseng increased significantly (P < 0.05) with the application of HHP. CONCLUSION: HHP‐processed red ginseng has significantly higher reducing sugar and free amino acid contents together with a more compact cell structure and superior visual quality (brighter red colour). Hence the application of HHP in red ginseng processing can result in ginseng products of improved quality compared with those obtained by the conventional method. Taro leaves are an important food in the Pacific Islands but the overall oxalate composition and its nutritional effect has not been investigated. The oxalate contents of taro leaves were determined using chemical and in vitro extraction methods. Maori‐type taro leaves contained 524.2 ± 21.3 mg total oxalates 100 g?1 fresh weight (FW) and 241.1 ± 20.9 mg soluble oxalates 100 g?1 FW while the Japanese‐type leaves contained 525.6 ± 19.9 mg total oxalates 100 g?1 FW and 330.4 ± 28.3 mg soluble oxalates 100 g?1 FW. Maori‐type taro leaves contained 416.4 ± 1.5 mg gastric soluble oxalates 100 g?1 FW and 212.4 ± 34.8 mg intestinal soluble oxalates 100 g?1 FW while the Japanese‐type leaves contained 433.3 ± 9.7 mg gastric soluble oxalates 100 g?1 FW and 224.2 ± 38.7 mg intestinal soluble oxalates 100 g?1 FW). Human feeding experiments were conducted to determine the availability of the oxalates in the baked leaves following additions of cows milk and coconut milk. The consumption of a test meal of baked taro leaves resulted in a significant increase (P < 0.001) in the output of urinary oxalate in the following 6 h when compared to the output of oxalate during a reference collection. When the leaves were baked with cows milk or cows milk and coconut milk combined and consumed there was a small non‐significant reduction in urinary oxalate output which suggests that less soluble oxalate was absorbed from these mixtures.  相似文献

In the present study, betalains content, phenolic composition, and antioxidant activity of different parts of red beet (Beta vulgaris L. conditiva) (i.e., roots and stems) were compared. Crude extract of root showed the highest betalain content with a maximum of 53 ± 4 mg betanin eq and 46 ± 3 mg vulgaxantin I eq g?1 of extract stems showed higher total phenolic concentration than roots, ranging between 2.0 ± 0.4 and 14.6 ± 0.5 mg gallic acid eq?1 of extract. Chemical composition was analyzed using LC-MS. Betalains (vulgaxanthin I, betanin, and isobetanin) and phenolics (gallic acid, ferulic acid, chlorogenic acid, caffeic acid, vanillic acid, syringic acid, ellagic acid myricetin, quercetin, rutin, kampferol) were identified in roots and stems. Betalain extract obtained from roots showed higher antioxidant activity than extract obtained from stems.  相似文献

Aspergillus oryzae biomass grown on effluents produced during the extraction of palm oil was found to have a crude protein content of 39.6 g 100 g?1 and a ‘true protein’ content of 32.1 g 100 g?1. Amino acid analysis showed that the essential amino acid content was 58.2 g 16 g?1 N and the essential amino acid index was calculated to be 87.4. The sulphur-containing amino acids were present at a combined level of 2.8 g 16 g?1. In rat feeding trials the biomass had a biological value of 0.68 ± 0.03, a net protein utilisation of 0.65 ± 0.05 and a true digestibility of 0.96 ± 0.05 based on its crude protein content. No unusual fatty acids were detected in the biomass.  相似文献

In this study, response surface methodology was applied to determine the optimal conditions for the extraction of tea polyphenols, EGCG and theanine from summer green tea. As results, the optimal extraction conditions were determined to be temperature 96 °C, time 40 min and ratio of water to raw material 16 mL g?1. Under these optimal conditions, the experimental values for tea polyphenols, EGCG, theanine and IC50 for DPPH free radical scavenging activity were 133.41 ± 3.12 mg gallic acid g?1, 30.23 ± 1.22, 3.99 ± 1.13 mg g?1 and 724.95 ± 15.12 μg mL?1, respectively. There was no statistical difference at significant level of 0.05 between the experimental and predicted values. The results suggested that the regression models were accurate and adequate for the bioactives extraction from summer green tea.  相似文献

Research reports on the ethnomedical and pharmacological potential of Kigelia africana extracts. In this study, the nutritional potential of K. africana seed and seed oil was evaluated by chemical analyses. Organic matter and ash constituted 915.23 ± 7.98 g kg?1 DM and 49.05 ± 4.55 g kg?1 DM of the seed mass, respectively. Oil constituted 492.2 g kg?1 DM of the seed mass with oleic acid, linoleic acid and α‐linolenic acid constituting 17.6%, 12.9% and 54.3%, respectively, of the seed oil. Vitamin E concentration was 0.94 ± 0.25 μg g?1. Crude protein was 357.35 ± 3.39 g kg?1 DM. Arginine (6.14 ± 0.31 g per 100 g) as the most abundant amino acid and hydroxyproline (0.11 ± 0.06 g per 100 g) the least. Phosphorus (1123.2 mg per 100 g) and calcium (56.1 mg per 100 g) were, respectively, the most and least abundant minerals. Gross energy was 29.6 ± 0.06 MJ kg?1. Kigelia africana seeds could be exploited as nutrient‐dense dietary supplement rich in protein, oleic acid and essential fatty acids.  相似文献

Four lines of oca, No. 38 and 41, Inca Gold and Market were grown in four replicated plots and the soluble oxalate content was determined on the freshly harvested tubers and tubers that had been stored for 6 weeks at 16.4 ± 0.5 °C. The mean dry matter content of freshly harvested tubers was 14.3 ± 0.5 g 100 g?1 fresh weight (FW), and after storage was 14.6 ± 0.5 g 100 g?1 FW. The mean soluble oxalate content of freshly harvested tubers was 162.1 ± 8.8 mg 100 g?1 FW, and, after 6 weeks storage, 173.5 ± 0.9 mg 100 g?1 FW. The different cultivars behaved differently during storage, the soluble oxalate content of Inca Gold tubers fell 7.2% while the soluble oxalate content of the other three cultivars increased (mean 13%).  相似文献

Food-grade titanium dioxide (TiO2) is a common and widespread food additive in many processed foods, personal care products, and other industrial categories as it boosts the brightness and whiteness of colours. Although it is generally recognised as safe for humans, there is a growing interest in the health risks associated with its oral intake. This study quantified and identified TiO2 nanoparticles present in confectionery foods, which are children’s favourite foods, with inductively coupled plasma optical emission spectrometry (ICP-OES) and transmission electron microscopy (TEM). A reliable digestion method using hot sulphuric acid and a digestion catalyst (K2SO4:CuSO4 = 9:1) was suggested for titanium analysis. Validations of the experimental method were quite acceptable in terms of linearity, recoveries, detection limits, and quantification limits. Of all the 88 analysed foods, TiO2 was detected in 19 products, all except three declared TiO2 in their labelling. The mean TiO2 content of candies, chewing gums, and chocolates were 0.36 mg g?1, 0.04 mg g?1, and 0.81 mg g?1, respectively. Whitish particles isolated from the confectionery foods were confirmed as TiO2 nanoparticles via TEM and energy dispersive X-ray spectroscopy (EDX), in which nanosized particles (<100 nm) were identified.  相似文献

BACKGROUND: The seeds of musk lime (Citrus microcarpa) represent a substantial waste product of small‐scale citrus‐processing factories, as they constitute about 100.0 ± 3.2 g kg?1 of the whole fruit and contain a considerable amount of crude fat (338.0 ± 11.3 g kg?1). Thus the aim of the present study was to determine the physicochemical properties of this fat with a view to potential applications. RESULTS: The iodine and saponification values and unsaponifiable matter and free fatty acid contents of the freshly extracted oil were 118.0 g I2 per 100 g oil, 192.6 mg KOH g?1 oil, 22 mg g?1 oil and 18 mg oleic acid g?1 oil respectively. The oil had a Lovibond colour index of 33.1 Y + 1.1 B. Its fatty acid profile indicated that 73.6% of the fatty acids present were unsaturated. Linoleic (L, 31.8%), oleic (O, 29.6%) and palmitic (P, 21.4%) acids were the predominant fatty acids, existing mainly as the triacylglycerols POL (18.9%), PLL (13.7%) and OLL (11.9%). The melting and cooling points of the oil were 10.7 and ? 45.2 °C respectively. Electronic nose qualitative analysis of the oil showed the presence of volatile (aroma) compounds, although the concentrations of the more volatile compounds were lower than those present in the seeds. CONCLUSION: Musk lime seeds are a rich source of oil, which is unusual in having linoleic, oleic and palmitic acids dominating the fatty acid composition. This property should make the oil both relatively stable to thermal oxidation owing to the combined presence of oleic and palmitic acids (61.0%) and highly nutritive owing to its high concentration of unsaturated fatty acids (73.6%). Chlorophyll-free protein has been recovered at a yield in excess of 50 % from sugar beet (Beta vulgaris L) leaf juice after two successive pH treatments. This corresponds to about 60 kg of white protein per hectare. The main component of the final preparation is ribulose-1,5-bisphosphate carboxylase (EC 4.1.1.39). Compared with the unfractionated protein from leaf extracts, the white protein preparation has higher contents of tyrosine, threonine, glutamic acid and proline and lower ones of isoleucine, lysine (although still high: 76mg g?1) and glycine. The content of methionine is high (24mg g?1) probably as a result of the addition of sulphite during protein extraction. The white protein preparation has a low level of ether extractives (17mg g?1) and a moderate ash (52mg g?1) content. The white protein is rapidly degraded by pepsin, trypsin and pancreatic juice.  相似文献

BACKGROUND: Thymus argaeus Boiss. & Bal. (Lamiaceae), an endemic plant species of Turkey known as wild thyme, is traditionally used as a spice and a wild tea in the Inner Anatolia region of Turkey. In this study the composition of the essential oil and the antimicrobial and antioxidant effects of the methanolic extract and essential oil of T. argaeus were determined. RESULTS: The main components of the essential oil were linalool (499 g kg?1), α‐terpineol (150 g kg?1), linalyl acetate (97 g kg?1) and thymol (94 g kg?1). The total phenolic, flavanol and flavonol contents of the extract were 83.31 ± 0.59 mg gallic acid equivalent g?1, 6.26 ± 0.00 mg catechin equivalent g?1 and 28.81 ± 0.21 mg rutin equivalent g?1 respectively. The antioxidant activities of the extract and essential oil determined by the 2,2‐diphenyl‐1‐picrylhydrazyl radical‐scavenging method were 830.18 ± 0.42 and 20.47 ± 2.3 mg g?1 respectively. The antimicrobial activities of the extract and essential oil against 13 bacteria and two yeasts were studied by the agar diffusion method. The micro‐organisms most sensitive to the essential oil were Aeromonas hydrophila and Pseudomonas aeruginosa, while the micro‐organism most sensitive to the extract was P. aeruginosa. CONCLUSION: Only the extract of T. argaeus could be used as a natural antioxidant, while both the extract and the essential oil could be useful as natural antimicrobial agents in food preservation. Four species of edible tubers endemic to and domesticated in the Andes, native potato (Solanum sp.), mashua (Tropaeolum tuberosum Ruiz & Pavón), oca (Oxalis tuberosa Molina) and ulluco (Ullucus tuberosus Caldas), were studied for their antioxidant capacity and associated secondary metabolites. The antioxidant capacity was measured using ABTS? radicals and total phenolics, carotenoids, anthocyanin, betaxanthin and betacyanin content were also characterized. The antioxidant capacity found in the crops studied ranged from 483 to 9800 µg trolox equiv. g?1, phenolics ranged from 0.41 to 3.37 mg chlorogenic acid equiv. g?1, anthocyanins ranged from 0.08 to 2.05 mg cyanidin 3‐glucoside g?1 and carotenoids ranged from 1 to 25 µg β‐carotene g?1. The content of bioactive compounds was high and variable between crops and within the genotypes studied. In general, mashua tubers showed the highest antioxidant capacity and phenolic, anthocyanin and carotenoid content compared with other crops. Ulluco was the only crop that contained betalains in the acid form of betaxanthins (22–96 µg g?1) and betacyanins (64 µg g?1) with no presence of carotenoids or anthocyanins. This is the first publication regarding the antioxidant capacity of and associated secondary metabolites in Andean tubers. This information can be useful in the identification of Andean tubers species and genotypes with potential value as a novel dietary source of antioxidants for food, and also for medicinal use. This study investigates the drying kinetics of residue of acerola, in a fixed‐bed dryer, analysing the effect of the process variables on the antioxidant properties of the residue. A complete factorial design 32 has been performed, where the independent variables studied were as follows: air velocity (0.5, 1.0 and 1.5 m s?1) and air‐drying temperature (40, 50 and 60 °C). The bioactive compounds studied were L‐ascorbic acid, total phenolic, total flavonoids and antioxidant activity (expressed as IC50) was determined using free radical DPPH?. Both independent variables studied have shown to be statistically significant. The content of ascorbic acid was the highest at 60 °C and 1.0 m s?1 (126.2 ± 0.004 mg 100 g?1) while for the fresh residue 16.12 ± 0.003 mg 100 g?1, whereas the total phenolic showed the highest content at 50 °C and 1.5 m s?1 (46.2 ± 0.003 mg gallic acid.100 g?1) while for the fresh residue 12.59 ± 0.001 mg gallic acid.100 g?1. The drying conditions play an important role in determining the final quality of the product mainly in terms of antioxidant activity.  相似文献

BACKGROUND: Hydrolysates prepared from sea urchin (Strongylocentrotus nudus) gonad by enzymatic treatment showed strong 1,1‐diphenyl‐2‐picrylhydrazyl radical scavenging activity and reducing power. RESULTS: Hydrolysis of S. nudus gonad by the commercial protease papain was optimised for maximum degree of hydrolysis (DH) and trichloroacetic acid‐soluble peptide index (TCA‐SPI) using response surface methodology. Results showed that the optimal conditions were the following: temperature of 48.83 °C, pH of 6.92, enzyme‐to‐substrate ratio of 3143 U g?1, and substrate concentration of 83.5 g L?1. Under these conditions, a DH of 27.96 ± 0.54% and a TCA‐SPI of 57.32 ± 0.63% were obtained. The hydrolysate prepared in the optimal conditions was fractionated by an ultra‐filtration system and the resultant fraction below 10 kDa was found to effectively scavenge hydroxyl radical (EC50 = 13.29 ± 0.33 mg mL?1) and hydrogen peroxide (EC50 = 16.40 ± 0.37 mg mL?1), inhibit lipid peroxidation (EC50 = 11.05 ± 0.62 mg mL?1), chelate Fe2+ (EC50 = 7.26 ± 0.44 mg mL?1), and protect mice macrophages against death induced by tert‐butyl hydroperoxide. The flavour compound 2‐hydroxy‐4‐methoxy benzaldehyde from normal roots of swallow root (Decalepis hamiltonii) raised in vitro was extracted with dichloromethane, evaporated to dryness and dissolved in ethanol for qualitative (TLC) and quantitative (GC‐MS) analysis. Maximum root biomass and the maximum content of flavour compound (40 ± 2.1 µg g?1 dry weight) were recorded after 45 days of growth on Murashige and Skoog medium containing 1.0 mg l?1 α‐naphthaleneacetic acid.
