Milk fatty acid composition is a parameter of great interest for evaluation of nutritional quality of milk. Stearoyl-CoA desaturase (SCD) is a key enzyme in mammary lipid metabolism because it is able to add a double bond in the cis Δ9-position in a large spectrum of medium- and long-chain fatty acids. A polymorphism with 2 alleles (A and V) in the fifth exon of the SCD gene has been reported. The effect of SCD genotype on individual milk fatty acid composition and on cis-9 unsaturated/saturated fatty acid ratios of 297 Holstein Italian Friesian cows was investigated in this paper. The SCD genotypes were determined by using a single strand conformation polymorphism method. Relative frequencies of SCD genotypes were 27, 60, and 13% for AA, AV, and VV, respectively. Milk of AA cows had a greater content of cis-9 C18:1 and total monounsaturated fatty acids and a higher C14:1/C14 ratio than did milk of VV cows. The relative contribution of SCD genotype to variation of monounsaturated fatty acids, cis-9 C18:1, and cis-9 C14:1 was 5, 4, and 7.7%, respectively. No significant differences were detected between SCD genotypes in the milk content of cis-9, trans-11 C18:2. Results of the present work provide some indication of an association between SCD locus and the fatty acid profile in the examined sample of Italian Holsteins, thus suggesting a possible role of this gene in the genetic variation of milk nutritional properties. 相似文献

Effect of dose of calcium salts of conjugated linoleic acid (CLA) on percentage and fatty acid content of milk fat in midlactation holstein cows 总被引:2,自引:0,他引:2
Increasing conjugated linoleic acid (CLA) content of milk fat from lactating dairy cattle has become a research interest due to the possible health benefits afforded humans consuming CLA. Dietary supplementation of CLA to lactating dairy cows is one potential method by which CLA content of milk and dairy products may be enhanced. Feeding CLA in calcium salt form could potentially deliver CLA to the lower digestive tract through prevention of biohydrogenation by rumen microbes. Milk fat depression (MFD) occurs when cows receive CLA-60, a commercially available CLA source containing numerous CLA isomers, abomasally. Our objectives were to determine the quantity of CLA as calcium salts required to elicit maximal MFD and to evaluate the effects of CLA supplementation on fatty acid composition of milk fat. Five Holstein cows at approximately 93 DIM were utilized in a 5 x 5 balanced Latin square crossover design. Periods were 14-d in length with a 5-d treatment phase and 9-d rest phase. Treatments were 5-d supplementation of 0, 12.5, 25, 50, and 100 g of CLA-60 in calcium salt form. Milk samples were collected on d 5 of CLA supplementation and analyzed for composition and fatty acid profile. Regression analysis of milk fat data suggested that MFD was not maximized over the dose levels investigated, despite delivery of 34.5 g of trans-10, cis-12 CLA in the 100-g dose of CLA. Supplementation with 50 and 100 g of CLA per day resulted in a reduction of milk fat percent of 29 and 34%, respectively. Trend analysis indicated a linear decrease in the milk fat content of caprylic, capric, and lauric acids as the dose of CLA increased. Effectiveness of oils rich in linoleic and linolenic acids to enhance conjugated linoleic acid in milk from dairy cows 总被引:3,自引:0,他引:3
Forty Holstein dairy cows were used to determine the effectiveness of linoleic or linolenic-rich oils to enhance C18:2cis-9, trans-11 conjugated linoleic acid (CLA) and C18:1trans-11 (vaccenic acid; VA) in milk. The experimental design was a complete randomized design for 9 wk with measurements made during the last 6 wk. Cows were fed a basal diet containing 59% forage (control) or a basal diet supplemented with either 4% soybean oil (SO), 4% flaxseed oil (FO), or 2% soybean oil plus 2% flaxseed oil (SFO) on a dry matter basis. Total fatty acids in the diet were 3.27, 7.47, 7.61, and 7.50 g/100 g in control, SO, FO, and SFO diets, respectively. Feed intake, energy-corrected milk (ECM) yield, and ECM produced/kg of feed intake were similar among treatments. The proportions of VA were increased by 318, 105, and 206% in milk fat from cows in the SO, FO, and SFO groups compared with cows in the control group. Similar increases in C18:2cis-9, trans-11 CLA were 273, 150, and 183% in SO, FO, and SFO treatments, respectively. Under similar feeding conditions, oils rich in linoleic acid (soybean oil) were more effective in enhancing VA and C18:2cis-9, trans-11 CLA in milk fat than oils containing linolenic acid (flaxseed oil) in dairy cows fed high-forage diets (59% forage). The effects of mixing linoleic and linolenic acids (50:50) on enhancing VA and C18:2cis-9, trans-11 CLA were additive, but not greater than when fed separately. Increasing the proportion of healthy fatty acids (VA and CLA) by feeding soybean or flaxseed oil would result in milk with higher nutritive and therapeutic value. 相似文献

Milk conjugated linoleic acid response to fish oil supplementation of diets differing in fatty acid profiles 总被引:4,自引:0,他引:4
The objective of this experiment was to examine the effect of feeding fish oil (FO) along with fat sources that varied in their fatty acid compositions (high stearic, high oleic, high linoleic, or high linolenic acids) to determine which combination would lead to maximum conjugated linoleic acid (cis-9,trans-11 CLA) and transvaccenic acid (TVA) concentrations in milk fat. Twelve Holstein cows (eight multiparous and four primiparous cows) at 73 (+/- 32) DIM were used in a 4 x 4 Latin square with 4-wk periods. Treatment diets were 1) 1% FO plus 2% fat source high in stearic acid (HS), 2) 1% FO plus 2% fat from high oleic acid sunflower seeds (HO), 3) 1% FO plus 2% fat from high linoleic acid sunflower seeds (HLO), and 4) 1% FO plus 2% fat from flax seeds (high linolenic; HLN). Diets formulated to contain 18% crude protein were composed of 50% (dry basis) concentrate mix, 25% corn silage, 12.5% alfalfa haylage, and 12.5% alfalfa hay. Milk production (35.8, 36.3, 34.9, and 35.0 kg/d for diets 1 to 4) was similar for all diets. Milk fat percentages (3.14, 2.81, 2.66, and 3.08) and yields (1.13, 1.02, 0.93, and 1.08 kg/d) for diets 1 to 4 were lowest for HLO. Milk protein percentages (3.04, 3.03, 3.10, and 3.08) and dry matter intake (DMI) (25.8, 26.0, 26.2, and 26.2 kg/d) for diets 1 to 4 were similar for all diets. Milk cis-9,trans-11 CLA concentrations (0.70, 1.04, 1.70, and 1.06 g/100 g fatty acids) for diet 1 to 4 and yields (7.7, 10.7, 15.8, and 11.3 g/d) for diets 1 to 4 were greatest with HLO and were least with HS. Milk cis-9,trans-11 CLA concentrations and yields were similar for cows fed the HO and the HLN diets. Similar to milk cis-9,trans-11 CLA, milk TVA concentration (1.64, 2.49, 3.74, and 2.41 g/100 g fatty acids) for diets 1 to 4 was greatest with the HLO diet and least with the HS diet. Feeding a high linoleic acid fat source with fish oil most effectively increased concentrations and yields of milk cis-9,trans-11 CLA and TVA. 相似文献

Conjugated linoleic acid increases in milk when cows fed fish meal and extruded soybeans for an extended period of time 总被引:3,自引:0,他引:3
The objective of this study was to determine the effect of feeding a conjugated linoleic acid (CLA) stimulating diet for an extended period of time on milk cis-9, trans-11 CLA and vaccenic acid (VA) concentrations. Twenty cows (16 Holstein and 4 Brown Swiss) were divided into 2 groups (n = 10 per treatment) for a 10-wk study. Cows in group 1 were fed a traditional corn-soybean-basal diet (control), while those in group 2 were fed a blend of 0.5% fish oil from fish meal and 2% soybean oil from extruded soybeans (FMESB) to achieve higher milk fat cis-9, trans-11 CLA and VA. Diets were formulated to contain 18% CP and were composed (dry matter basis) of 50% concentrate mix, 25% corn silage, and 25% alfalfa hay. Dry matter intake was not affected by diet. Milk production increased in cows fed the FMESB diet. Milk fat and milk protein percentages decreased with the FMESB diet; however, milk fat and protein yields were not affected by treatments. Milk fat cis-9, trans-11 CLA and VA concentration (g/100 of fatty acids) and yield (g/d) were 2.5-fold greater for cows fed the FMESB diet over the 10 wk of fat supplementation. For cows fed the FMESB diet, contents of milk fat cis-9, trans-11 CLA and VA gradually increased from the first week of fat supplementation, reached the highest concentrations in wk 3, then gradually decreased during wk 4 and 5 and then remained relatively constant until wk 10. The concentration of cis-9, trans-11 CLA and VA from the control diet was relatively constant over the 10 wk of fat supplementation. Concentrations of cis-9, trans-11 CLA and VA in milk fat can be increased within a week by feeding a blend of fish meal and extruded soybeans, and that increase remains relatively constant after wk 5 of fat supplementation. 相似文献

Dairy products are the main source of conjugated linoleic acid (CLA), a functional food component with health benefits. The major source of cis-9, trans-11 CLA in milk fat is endogenous synthesis via delta9-desaturase from trans-11 18:1, with the remainder from incomplete rumen biohydrogenation of linoleic acid. Diet has a major influence on milk fat CLA; however, effects of physiological factors have received little attention. Our objectives were to examine milk fat content of CLA and the CLA-desaturase index with regard to: 1) effect of breed, parity, and stage of lactation, and 2) variation among individuals and the relationship to milk and milk fat. Holstein (n = 113) and Brown Swiss (n = 106) cows were fed a single diet and milk sampled on the same day to avoid confounding effects of diet and season. Frequency distributions demonstrated that milk fat content of CLA and CLA-desaturase index varied over threefold among individuals, and this needs to be considered in the design of experiments. Holsteins had a higher milk fat content of CLA and CLA-desaturase index, but breed differences were minor. Parity and days in milk also had little or no relationship to the individual variation for these two CLA variables. Breed, parity, and days in milk accounted for < 0.1, < 0.3, and < 2.0% of total variation in CLA concentration in milk fat, respectively. Milk fat content of CLA and CLA-desaturase index were essentially independent of milk yield, milk fat percent, and milk fat yield. We speculate that the basis for the genetic variation among individuals is related to rumen output of trans-11 18:1 and to a lesser extent cis-9, trans-11 CLA, and to the tissue amount and activity of delta9-desaturase. 相似文献

Trans-10, cis-12 conjugated linoleic acid (CLA) is a potent inhibitor of milk fat synthesis, and the magnitude of milk fat depression is often correlated with the fat content of this isomer. However, the trans-10, cis-12 CLA content does not always correspond to the extent of milk fat depression, and in some instances, an increase in the milk fat content of trans-10, trans-12 CLA has been observed. We synthesized trans-10, trans-12 CLA (>90% purity) and investigated its effect on milk fat synthesis and incorporation into plasma lipids. Three rumen-fistulated Holstein cows were randomly assigned in a 3 × 3 Latin square experiment. Treatments were a 4-d abomasal infusion of 1) ethanol (control), 2) a trans-10, cis-12 CLA supplement (positive control), and 3) a trans-10, trans-12 CLA supplement; 5 g/d of the CLA isomer of interest was provided. Milk yield, dry matter intake, and milk protein were unaffected by treatment. Treatment with trans-10, trans-12 CLA had no effect on milk fat yield, whereas treatment with trans-10, cis-12 CLA reduced milk fat yield by 28%. Incorporation of CLA was greatest for the plasma triglyceride fraction, and the milk fat content was subsequently elevated within the respective treatment groups. The milk fatty acid composition indicated that Δ9-desaturase was reduced significantly for both CLA treatments, but the reduction was greater for the treatment with trans-10, trans-12 CLA. Overall, abomasal infusion of trans-10, trans-12 CLA and trans-10, cis-12 CLA altered the desaturase ratios, but only trans-10, cis-12 CLA reduced milk fat synthesis. 相似文献

Stearoyl-CoA desaturase enzyme converts specific medium- and long-chain saturated fatty acids to their monounsaturated form. Transgenic goats expressing a bovine beta-lactoglobulin promoter-rat stearoyl-CoA desaturase cDNA construct in mammary gland epithelial cells were produced by pronuclear microinjection. The fatty acid composition of milk from 4 female transgenic founders was analyzed on d 7, 14, and 30 of their first lactation. In 2 animals, the expression of the transgene changed the overall fatty acid composition of the resulting milk fat to a less saturated and more monounsaturated fatty acid profile at d 7 of lactation; however, this effect diminished by d 30. In addition, one animal had an increased proportion of the rumen-derived monounsaturated fatty acid C18:1 trans11 converted by stearoyl-CoA desaturase to the conjugated linoleic acid isomer C18:2 cis9 trans11. Milk that has higher proportions of monounsaturated fatty acids and conjugated linoleic acid may have benefits for human cardiovascular health. 相似文献

The aim of this study was to estimate genetic parameters and identify genomic regions associated with selected individual and groups of milk fatty acids (FA) predicted by milk mid-infrared spectrometry in Dual-Purpose Belgian Blue cows. The used data were 69,349 test-day records of milk yield, fat percentage, and protein percentage along with selected individual and groups FA of milk (g/dL milk) collected from 2007 to 2020 on 7,392 first-parity (40,903 test-day records), and 5,185 second-parity (28,446 test-day records) cows distributed in 104 herds in the Walloon Region of Belgium. Data of 28,466 SNPs, located on 29 Bos taurus autosomes (BTA), of 1,699 animals (639 males and 1,060 females) were used. Random regression test-day models were used to estimate genetic parameters through the Bayesian Gibbs sampling method. The SNP solutions were estimated using a single-step genomic best linear unbiased prediction approach. The proportion of genetic variance explained by each 25-SNP sliding window (with an average size of ~2 Mb) was calculated, and regions accounting for at least 1.0% of the total additive genetic variance were used to search for candidate genes. Average daily heritability estimated for the included milk FA traits ranged from 0.01 (C4:0) to 0.48 (C12:0) and 0.01 (C4:0) to 0.42 (C12:0) in the first and second parities, respectively. Genetic correlations found between milk yield and the studied individual milk FA, except for C18:0, C18:1 trans, C18:1 cis-9, were positive. The results showed that fat percentage and protein percentage were positively genetically correlated with all studied individual milk FA. Genome-wide association analyses identified 11 genomic regions distributed over 8 chromosomes [BTA1, BTA4, BTA10, BTA14 (4 regions), BTA19, BTA22, BTA24, and BTA26] associated with the studied FA traits, though those found on BTA14 partly overlapped. The genomic regions identified differed between parities and lactation stages. Although these differences in genomic regions detected may be due to the power of quantitative trait locus detection, it also suggests that candidate genes underlie the phenotypic expression of the studied traits may vary between parities and lactation stages. These findings increase our understanding about the genetic background of milk FA and can be used for the future implementation of genomic evaluation to improve milk FA profile in Dual-Purpose Belgian Blue cows. 相似文献

The objective of this trial was to study the interaction between the supplementation of lipid-encapsulated conjugated linoleic acid (CLA; 4.5 g of cis-9,trans-11 C18:2 and 4.5 g of trans-10,cis-12 C18:2) and feeding level to test if milk performance or milk fatty acid (FA) profile are affected by the interaction between CLA and feeding level. Twenty-four dairy goats were used in an 8-wk trial with a 3-wk adaptation to the experimental ration that contained corn silage, beet pulp, barley, and a commercial concentrate. During the third week, goats were assigned into blocks of 2 goats according to their dry matter intake (DMI), raw milk yield, and fat yield. Each block was randomly allocated to control (45 g of Ca salt of palm oil/d) or CLA treatment. Within each block, one goat was fed to cover 100% (FL100) of the calculated energy requirements and the other was fed 85% of the DMI of the first goat (FL85). Individual milk production and composition were recorded weekly, and milk FA composition was analyzed in wk 3, 5, and 7. Conjugated linoleic acid supplementation reduced milk fat content and fat yield by 17 and 19%, respectively, independent of the feeding level. It reduced both the secretion of milk FA synthesized de novo, and those taken up from the blood. No interaction between CLA and feeding level was observed on milk secretion of any group of FA. The CLA supplementation had no effect on DMI, milk yield, protein, and lactose yields but it improved calculated net energy for lactation balance. Goats fed the FL100 × CLA diet tended to have the highest DMI and protein yield. The interaction between CLA and feeding level was not significant for any other variables. Compared with the goats fed FL100, those fed FL85 had lower DMI, lower net energy for lactation balance, and lower digestible protein in the intestine balance. The body weight; milk yield; milk fat, protein, and lactose yields; and fat, protein, lactose, and urea contents in milk were not affected by feeding level. In conclusion, reduction in energy spared via fat yield reduction after CLA supplementation was not partitioned toward milk lactose or protein in goats at a low feeding level, possibly because of a simultaneous shortage of energy and amino acids. In goats on the high feeding level, energy spared tended to be partitioned toward milk protein yield, and at the same time to the prevention of excessive lipid mobilization. 相似文献

Conjugated linoleic acid (c9, t11 CLA) is a dietary fatty acid produced mainly by ruminant animals and exhibits promising health‐promoting biological effects. For lipid fatty acid composition analyses, including CLA, lipids must be pre‐treated so that the free and esterified fatty acids (triacylglycerols, phospholipids, etc) are available for determination. The most common treatments involve fatty acid methyl ester derivatives from relatively simple chemical reactions, but this becomes complicated when esterification of CLA is involved because of potential changes in its positional and geometrical isomers by reaction with certain reagents. In this review we explain concisely the advantages and disadvantages of the most popular methods (acid‐ and base‐catalysed methods) generally employed for total fatty acid derivatization and their determination on a gas chromatograph. Based on our experiences we put forward the (trimethylsilyl)diazomethane method as an alternative and successful approach for ruminant tissue lipid determinations. 研究了牛乳中共轭亚油酸含量的紫外分光光度测定方法。利用紫外分光光度计,先建立标准曲线与回归公式,再将牛奶样品提取、甲酯化后,进行紫外检测。结果表明,共轭亚油酸甲酯的最大吸收波长是234nm;共轭亚油酸浓度的有效测定范围是0.00856-0.0428mg/mL;吸光度与共轭亚油酸浓度间的回归公式为Y=52.602X 0.0917(R2=0.9862);得出共轭亚油酸浓度与吸光度之间的经验公式为:共轭亚油酸浓度C(mg/mL)=0.019×吸光度(ABS);并且推出了利用紫外分光光度法测定牛奶中共轭亚油酸含量的计算公式。同时,检验了几种市售牛奶中共轭亚油酸,其平均含量为1.64mg/g。 相似文献

Sufficient maternal supply of essential fatty acids (EFA) to neonatal calves is critical for calf development. In the modern dairy cow, EFA supply has shifted from α-linolenic acid (ALA) to linoleic acid (LA) due to the replacement of pasture feeding by corn silage–based diets. As a consequence of reduced pasture feeding, conjugated linoleic acid (CLA) provision by rumen biohydrogenation was also reduced. The present study investigated the fatty acid (FA) status and performance of neonatal calves descended from dams receiving corn silage–based diets and random supplementation of either 76 g/d coconut oil (CTRL; n = 9), 78 g/d linseed oil and 4 g/d safflower oil (EFA; n-6/n-3 FA ratio = 1:3; n = 9), 38 g/d Lutalin (BASF SE, Ludwigshafen, Germany) providing 27% cis-9,trans-11 and trans-10,cis-12 CLA, respectively (CLA; n = 9), or a combination of EFA and CLA (EFA+CLA; n = 11) in the last 9 wk before parturition and following lactation. The experimental period comprised the first 5 d of life, during which calves received colostrum and transition milk from their own dam. The nutrient compositions of colostrum and transition milk were analyzed. Plasma samples were taken after birth and before first colostrum intake and on d 5 of life for FA analyses of the total plasma fat and lipid fractions. Maternal EFA and CLA supplementation partly affected colostrum and transition milk composition but did not change the body weights of calves. Most EFA in calves were found in the phospholipid (PL) and cholesterol ester (CE) fractions of the plasma fat. Maternal EFA supplementation increased the percentage of ALA in all lipid fractions of EFA and EFA+CLA compared with CTRL and CLA calves on d 1 and 5, and the increase was much greater on d 5 than on d 1. The LA concentration increased from d 1 to 5 in the plasma fat and lipid fractions of all groups. The concentrations of docosapentaenoic acid, docosahexaenoic acid, and arachidonic acid in plasma fat were higher on d 1 than on d 5, and the percentage of n-3 metabolites was mainly increased in PL if dams received EFA. The percentage of cis-9,trans-11 CLA was higher in the plasma fat of EFA+CLA than CTRL calves after birth. By d 5, the percentages of both CLA isomers increased, leading to higher proportions in plasma fat of CLA and EFA+CLA than in CTRL and EFA calves. Elevated cis-9,trans-11 CLA enrichment was observed on d 5 in PL, CE, and triglycerides of CLA-treated calves, whereas trans-10,cis-12 CLA could not be detected in individual plasma fractions. These results suggest that an altered maternal EFA and CLA supply can reach the calf via the placenta and particularly via the intake of colostrum and transition milk, whereas the n-3 and n-6 FA metabolites partly indicated a greater transfer via the placenta. Furthermore, the nutrient supply via colostrum and transition milk might be partly modulated by an altered maternal EFA and CLA supply but without consequences on calf performance during the first 5 d of life. 相似文献

Genetic parameters for major milk fatty acids and milk production traits of Dutch Holstein-Friesians

The objective of this study was to estimate genetic parameters for major milk fatty acids and milk production traits. One morning milk sample was collected from 1,918 Holstein-Friesian heifers located in 398 commercial herds in the Netherlands. Each sample was analyzed for total percentages of fat and protein, and for detailed fatty acid percentages (computed as fatty acid weight as a proportion of total fat weight). Intraherd heritabilities were high for C4:0 to C16:0, ranging from 0.42 for C4:0 to 0.71 for C10:0. Saturated and unsaturated C18 fatty acids had intraherd heritability estimates of approximately 0.25, except for C18:2 cis-9, trans-11, which was 0.42. Standard errors of the heritabilities were between 0.07 and 0.12. Genetic correlations were high and positive among C4:0 to C14:0, as well as among unsaturated C18, but correlations of C4:0 to C14:0 with unsaturated C18 were generally weak. The genetic correlation of C16:0 with fat percentage was positive (0.65), implying that selection for fat percentage should result in a correlated increase of C16:0, whereas unsaturated C18 fatty acids decreased with increasing fat percentage (−0.74). Thirty-six Holstein cows were blocked by parity and allotted by stage of lactation to 6 treatments to evaluate the effects of dietary soy oil, conjugated linoleic acid (CLA; free acid or calcium salt), or both, on CLA content of milk. Diets were fed for 4 wk and are as follows: (1) control, (2) control + 5% soy oil, (3) control + 1% CLA, (4) control + 1% Ca(CLA)2, (5) control + 1% CLA + 4% soy oil, and (6) control + 1% Ca(CLA)2 + 4% soy oil. Rumen volatile fatty acid -concentrations, blood fatty acid concentrations, milk yield, and milk composition were measured weekly or biweekly. Dry matter intake and milk yield were recorded daily. Dietary supplementation of soy oil or CLA had no effect on daily milk yield, milk protein concentration and production, or milk lactose concentration and production. Supplementation of unsaturated fatty acids as soy oil, CLA, or Ca(CLA)2 increased total fatty acid concentration in plasma, decreased milk fat concentration and production, and had no effect on rumen volatile fatty acid concentrations. The weight percentage of CLA in milk was increased from 0.4 to 0.7% with supplementation of 1% CLA, to 1.2% with supplementation of soy oil, and to 1.3% with supplementation of 1% CLA plus soy oil. Supplementation with Ca(CLA)2 or Ca(CLA)2 + soy oil increased the CLA content of milk fat to 0.9 and 1.4%, respectively. In summary, adding 5% soy oil was as effective as supplementing CLA, Ca(CLA)2, or a combination of 1% CLA (free acid or calcium salt) + 4% soy oil at increasing CLA concentrations in milk fat. Feeding CLA as the calcium salt resulted in greater concentrations of CLA in milk fat than did feeding CLA as the free acid. Dietary supplementation of 5% soy oil or 4% soy oil + 1% CLA as the free acid or the calcium salt increased the yield of CLA in milk. 相似文献

The objective of this work was to evaluate the effect of the supplementation of conjugated linoleic acid (CLA; 4.5 g of cis-9,trans-11 C18:2 and 4.5 g of trans-10,cis-12 C18:2) on milk performance, milk fatty acid (FA) composition, and adipose tissue reactivity in dairy goats fed a high-concentrate diet based on corn silage. Twenty-four multiparous dairy goats in early to mid lactation were used in a 10-wk trial, with a 3-wk adaptation to the experimental total mixed ration that contained corn silage (35%, dry matter basis), beet pulp (20%), barley (15%), and a commercial concentrate (30%). Goats were randomly allocated to 2 experimental groups and they were fed 45 g/d of a lipid supplement (either CLA or Ca salts of palm oil added on top of the total mixed ration). Individual milk production and composition were recorded weekly, and milk FA composition was analyzed in wk 2, 5, and 6. In the last week of the trial, an isoproterenol challenge was performed for 12 goats before morning feeding. The CLA supplementation had no effect on dry matter intake (DMI), body weight (BW), milk yield, milk protein content, and lactose yield and content, but it significantly decreased milk fat yield and content by 18 and 15%, respectively. The decrease in milk fat yield was related to a lower secretion of FA synthesized de novo, of the medium-chain FA, and to a lesser extent of the long-chain FA that are taken up from the peripheral circulation. The CLA supplementation decreased the proportion of the sum of C16:0 and C16:1 and the sum of total cis C18:1, and it increased the proportions of the sum of long-chain (C >16) and the sum of iso FA without modification of the total trans C18:1 and the sum of FA synthesized de novo (C <16). During the first 25 min relative to isoproterenol injection, the maximal concentrations, the increases above basal concentration, the changes in area under the curve, and the total area under the curve for glucose and nonesterified FA were not affected by CLA treatment. In conclusion, CLA supplementation associated with a high-concentrate diet based on corn silage resulted in decreased milk fat yield, increased net energy balance, and it did not affect the sensitivity of the adipose tissue to lipolytic challenge in lactating goats. 相似文献

Under certain dietary situations, rumen biohydrogenation results in the production of unique fatty acids that inhibit milk fat synthesis. The first of these to be identified was trans-10, cis-12 conjugated linoleic acid (CLA), but others are postulated to contribute to diet-induced milk fat depression (MFD). Our objective was to examine the potential role of trans-9, cis-11 CLA in the regulation of milk fat. In a preliminary study, we used gas-liquid and high-performance liquid chromatography techniques to examine milk fat samples from a diet-induced MFD study and found that an increase in trans-9, cis-11 CLA corresponded to the decrease in milk fat yield. We investigated this further using a CLA enrichment of 9, 11 isomers to examine the biological effect of trans-9, cis-11 CLA on milk fat synthesis. Four rumen-fistulated Holstein cows were randomly assigned in a 4 × 4 Latin square experiment involving 5-d treatment periods and abomasal infusion of 1) ethanol (control), 2) a 9, 11 CLA mix (containing 32% trans-9, cis-11, 29% cis-9, trans-11, and 17% trans-9, trans-11), 3) a trans-9, trans-11 CLA supplement, and 4) a trans-10, cis-12 CLA supplement (positive control). The trans-9, trans-11 CLA and trans-10, cis-12 CLA supplements were of high purity (>90%), and all supplements were infused at a rate to provide 5 g/d of the CLA isomer of interest. Milk yield and dry matter intake did not differ among treatments. Compared with the control treatment, milk fat yield was reduced by 15% for the 9, 11 CLA mixture and by 27% for the trans-10, cis-12 CLA treatment. We also found that trans-9, trans-11 CLA had no effect on milk fat yield, and previous research has shown that milk fat yield is unaltered when cows are infused with cis-9, trans-11 CLA. When all treatments were considered, results suggested that trans-9, cis-11 was the CLA isomer in the 9, 11 CLA mix responsible for the reduction in milk fat synthesis, although the magnitude was less than that observed for trans-10, cis-12 CLA. Interestingly, trans-9, trans-11 CLA altered the milk fat desaturase index, further demonstrating that alterations in desaturase can occur independently of effects on milk fat synthesis. Overall, our investigations identified that an increase in milk fat content of trans-9, cis-11 CLA was associated with diet-induced MFD and provided evidence of a role for this isomer in MFD based on the 15% reduction in milk fat yield with abomasal infusion of a CLA enrichment that supplied 5 g/d of trans-9, cis-11 CLA. 相似文献

In view of the potential of rumen-protected conjugated linoleic acid (CLA) as a means to increase the CLA content of bovine milk, a study was undertaken to evaluate the effect of synthetic CLA on milk production and composition. Four Holstein cows received abomasal infusion of: 1) control, no lipid infusion, 2) 150 g/d of synthetic CLA, 31.7% cis-9, trans-11; 30.4% trans-10, cis-12, 3) 150 g/d of safflower oil, and 4) 150 g/d of tallow. Infusion was carried out for 20 to 22 h/d for 11-d periods in a 4 x 4 Latin square design. The milk fat concentration of cis-9, trans-11 and trans-10, cis-12 isomers of CLA was significantly increased with infusion of CLA. However, CLA infusion had other unexpected effects on milk production and composition. Milk yield dropped significantly during the period of CLA infusion. Furthermore, as well as the typical depression in milk fat reported with trans-10 isomers of CLA, other negative effects specific to CLA infusion were observed including a drop in lactose concentration and yield, a drop in protein yield, and an elevated somatic cell count. The important difference between synthetically produced CLA and CLA produced naturally in the cow is the much higher proportion of trans-10 isomers of CLA in the former. The results of this study suggest that the extent of enrichment possible for trans-10 isomers of CLA, and hence the usefulness of synthetic CLA for this purpose, may be limited because of unacceptable effects on milk yield and composition. 相似文献

The aim of this study was to investigate the effects of conjugated linoleic acid supplementation on the synthesis of milk fat in pasture-fed Friesian cows. In four cows, a commercial mixture containing 62.3% (wt/vol) conjugated linoleic acid was infused intraabomasally to avoid rumen fermentation and biohydrogenation. The design was a 4 x 4 Latin square in which each cow received infusions of 0, 20, 40, and 80 g/d of conjugated linoleic acid mixture for 4 d. Cows were fed freshly cut ryegrass/white clover pasture ad libitum. Milk fat concentration was decreased by 36, 43, and 62% and milk fat yield was decreased by 32, 36, and 60% by the 20, 40, and 80 g of conjugated linoleic acid/d treatments. Dry matter intake, milk protein concentration, and protein yield were unaffected by treatments; however, milk yield was increased by 11% during the 40-g conjugated linoleic acid/d treatment. The effects of conjugated linoleic acid infusion were most pronounced in reducing de novo fatty acid synthesis and desaturation. Results show that the inhibitory effect of this conjugated linoleic acid mixture on milk fat synthesis occurs in pasture-fed cows, and demonstrate the potential to dramatically alter gross milk composition. Fatty acid profiles with emphasis on linoleic, linolenic, oleic, and conjugated linoleic acid (CLA) were compared in milk from dairy cows fed diets containing 3.25% supplemental fat and a control diet containing no supplemented fat. The fat was supplied by either whole ground solin, flax, or canola oilseed. Solin (linola) is a new cultivar of flax that contains 28% linoleic acid in the seed, Twelve multiparous Holstein cows were assigned to one of four dietary treatments. The experimental design was a 4 x 4 Latin square with each period consisting of 16 d for adjustment to the diet followed by a 5-d sampling period. Feed intake, milk yield, milk fat yield, and milk fat percentage were not affected by treatment. Adding solin, flax, or canola oilseed to lactation diets produced the highest proportions of linoleic (C18:2), linolenic (C18:3), and oleic (C18:1) acids, respectively, in the lipid fraction of the milk of the cows consuming these diets. The proportions of C6:0 to C16:1 were depressed in the milk fat of cows fed the oilseed diets, compared with the control diet. Increasing the lactation diet levels of C18:2, by using different oilseeds, increased CLA to 1.5% of milk fatty acids. The content of CLA in milk fatty acids, however, did not increase with the solin-supplemented diet compared with the canola-supplemented diet even though the C18:2 level was higher in the former diet. 相似文献
