A high degree of polymorphism was recently found at the kappa-casein (CSN3) locus in the domesticated goat (Capra hircus). In the present study, 2 new patterns previously identified by PCR-single-strand conformation polymorphism analysis (SSCP) were characterized. The allele provisionally named "X" (GenBank Accession no. AY350425) differs from CSN3*C (AF485341) by a (silent) A-->G substitution at position 509 of the goat CSN3 reference sequence (X60763). As this newly identified sequence changes the amino acid sequence, and the already known CSN3*C allele (AF485341) has an additional silent mutation, we proposed a change in nomenclature to reflect these changes, indicating the silent mutation with the prime symbol (i.e.,'). The CSN3*M allele (provisionally named "Y") results in a new protein variant, differing by 2 nonsynonymous mutations from the CSN3*F allele. The new variant is characterized by a G-->A transition at nucleotide position 384, resulting in the amino acid exchange Asp90-->Asn90, and a C-->T transition at position 550, resulting in a Val145-->Ala145 substitution. Thus, the number of alleles identified in the domesticated goat has increased to 16, of which 13 are protein variants and 3 are silent mutations, involving a total of 15 polymorphic sites in CSN3 exon 4. Data on the distribution of the main alleles in 7 goat breeds of Europe, West Africa, and the Near East show differences in the occurrence and frequency of the alleles between breeds and geographic origin with the highest number of alleles found in goat breeds from the Near East.  相似文献

We assessed polymorphisms in exon IV of the kappa-casein gene (CSN3) in ten different breeds of domestic goat (Capra hircus) from three continents and in three related wild caprine taxa (Capra ibex, Capra sibirica and Capra aegagrus). Thirty-five DNA samples were sequenced within a 558 bp fragment of exon IV. Nine polymorphic sites were identified in domestic goat, including four new polymorphisms. In addition to four previously described polymorphic positions, a total of 13 polymorphisms allowed the identification of 13 DNA variants, corresponding to 10 protein variants. Because of conflicting nomenclature of these variants, we propose a standardized allele designation. CSN3*A, CSN3*B, and CSN3*D were found as widely distributed alleles in European goat breeds. Within Capra ibex we identified three variants and showed that the sequence of Capra aegagrus is identical to the most common Capra hircus variant, consistent with Capra aegagrus being the wild progenitor of domestic goats. A dendrogram was drawn to represent the molecular network between the caprine CSN3 variants.  相似文献

Until now, a total of nine polymorphic sites corresponding to six different alleles have been described at the κ-casein (CSN3) locus in the domestic goat (Capra hircus). A protocol for the rapid and simultaneous genotyping of five goat CSN3 alleles by using polymerase chain reaction-single strand conformation polymorphism (PCR-SSCP) technique was developed. Moreover, the developed test was validated by screening the CSN3 variability in four Italian breeds, Garganica, Jonica, Maltese, and Camosciata. Seven different patterns were readily identifiable. These corresponded to five known alleles and two newly identified variants. The G/A substitution at nucleotide position 471, which is not identifiable at the protein level but was found to be very frequent in the typed breeds, is easily detectable by the protocol developed. The PCR-SSCP analysis is a powerful tool for the genetic study of CSN3 variability in domestic goats, allowing both the simultaneous identification of different alleles, and the detection of new variants.  相似文献

The variation in the casein genes has a major impact on the milk composition of goats. Even though many casein polymorphisms have been identified so far, we do not know yet whether they are evolutionarily ancient (i.e., they existed before domestication) or young (i.e., they emerged after domestication). Herewith, we identified casein polymorphisms in a data set of 106 caprine whole-genome sequences corresponding to bezoars (Capra aegagrus, the ancestor of domestic goats) and 4 domestic goat (Capra hircus) populations from Europe, Africa, the Far East, and the Near East. Domestic and wild goat populations shared a substantial number of casein SNP, from 36.1% (CSN2) to 55.1% (CSN1S2). The comparison of casein variation among bezoars and the 4 domestic goat populations demonstrated that more than 50% of the casein SNP are shared by 2 or more populations, and 18 to 44% are shared by all populations. Moreover, the majority of casein alleles reported in domestic goats also segregate in the bezoar, including several alleles displaying significant associations with milk composition (e.g., the A/B alleles of the CSN1S1 and CSN3 genes, the A allele of the CSN2 gene). We conclude that much of the current diversity of the caprine casein genes comes from ancient standing variation segregating in the ancestor of modern domestic goats.  相似文献

The main goal of the current work was to identify single nucleotide polymorphisms (SNP) that might create or disrupt microRNA (miRNA) target sites in the caprine casein genes. The 3′ untranslated regions of the goat αS1-, αS2-, β-, and κ-casein genes (CSN1S1, CSN1S2, CSN2, and CSN3, respectively) were resequenced in 25 individuals of the Murciano-Granadina, Cashmere, Canarian, Saanen, and Sahelian breeds. Five SNP were identified through this strategy: c.*175C > T at CSN1S1; c.*109T > C, c.*139G > C, and c.*160T > C at CSN1S2; and c.*216C > T at CSN2. Analysis with the Patrocles Finder tool predicted that all of these SNP are located within regions complementary to the seed of diverse miRNA sequences. These in silico results suggest that polymorphism at miRNA target sites might have some effect on casein expression. We explored this issue by genotyping the c.*175C > T SNP (CSN1S1) in 85 Murciano-Granadina goats with records for milk CSN1S1 concentrations. This substitution destroys a putative target site for miR-101, a miRNA known to be expressed in the bovine mammary gland. Although TT goats had higher levels (6.25 g/L) of CSN1S1 than their CT (6.05 g/L) and CC (6.04 g/L) counterparts, these differences were not significant. Experimental confirmation of the miRNA target sites predicted in the current work and performance of additional association analyses in other goat populations will be an essential step to find out if polymorphic miRNA target sites constitute an important source of variation in casein expression.  相似文献

Seven lactating goat mammary glands from 6 goats were perfused for several hours in the presence of [U-14C]L-leucine (4 experiments) or [2-3H; 1-14C]DL-leucine (3 experiments) and received adequate quantities of glucose, acetate and amino acids. Radioactivity in casein was mainly recovered in leucine and 90% of casein leucine was derived from free plasma leucine. About 64% of the leucine molecules were used for casein synthesis. Up to 12% of the molecules were channelled into lipid synthesis, while the remaining (up to 24%) were metabolized to CO2. From the 3H/14C ratio of casein and casein leucine, it was calculated that 70-80% of the leucine molecules were reversibly transaminated before their incorporation into milk protein. However, only 4-8% of the plasma leucine molecules were transaminated during passage through the udder. Different pools for oxidation and for protein synthesis may be present in the goat mammary gland.  相似文献

通过纳米粒度分析仪和扫描电子显微镜,分别对水牛乳、牛乳及羊乳中的酪蛋白颗粒直径大小分布情况及酪蛋白胶束结构进行研究。结果表明:水牛乳、牛乳及羊乳中酪蛋白的粒径分布及胶束结构方面存在明显的差异。水牛乳酪蛋白平均颗粒直径为182.3nm,酪蛋白颗粒互相连接成较细长的胶束,胶束之间交联成网络状;牛乳酪蛋白平均颗粒直径为207.4nm,酪蛋白颗粒聚集成直径较大的胶束;羊乳酪蛋白平均颗粒直径为173.8nm,酪蛋白颗粒仅能够形成较短的胶束,也不能交联成网络状。  相似文献

Acid gels and yoghurts were made from goat milk that was heated at 72°C/30 s, 85°C/5 min, and 95°C/5 min, followed by acidification with starter culture at 43C until pH 4.6. The rheological and textural properties of acid gels and yoghurts were analyzed using dynamic low amplitude oscillatory rheology and back extrusion texture analysis, respectively. The effect of goat milk heat treatment on the mean casein micelle diameter and protein profile was also determined by dynamic light scattering and SDS PAGE electrophoresis, respectively. The shortest gelation and fermentation time was recorded for yoghurt prepared from milk heated at 85°C/5 min. Also, the pH of gelation, the storage moduli (G′) and yield stress were higher for this yoghurt, compared with the other two. Textural properties of goat milk yoghurts such as firmness and consistency were strongly affected by milk heat treatment, and the highest values were recorded for yoghurt produced from milk preheated at 85°C/5 min, as well. The largest casein micelles were measured after 85°C/5 min treatment and their size decreased at higher temperature, despite higher denaturation of whey proteins at the most intense heat regime, indicating the structure changes that influence on the acid gelation.  相似文献

The gastric digestion behaviours of infant formulae made with goat milk and cow milk, with different ratios of casein to whey protein, were investigated using an in vitro dynamic infant human gastric simulator. The goat milk infant formulae formed smaller flocs of aggregated protein and oil droplets under gastric conditions, leading to faster protein digestion in goat milk infant formulae than in cow milk infant formulae. The extent of coagulation of protein and the size of flocculated oil droplets were dependent on the protein composition of the formulae. The casein-dominated cow milk infant formula had greater aggregation initially during gastric digestion, but a lower rate of casein digestion. The results suggest that the different composition of the casein micelles in goat milk may play an important role in the lower extent of coagulation and the faster protein gastric digestion in goat milk compared with cow milk.  相似文献

采采用新鲜和复原的牛羊乳为原料,等电点沉淀,通过洗涤、干燥等步骤分别制得牛乳和羊乳酪蛋白,用凯氏定氮法对其进行定量检测,应用十二烷基硫酸钠聚丙烯酰胺凝胶电泳(SDS—PAGE)分析比较牛羊乳酪蛋白组分差异。结果表明:牛乳酪蛋白得率为86.75%,羊乳酪蛋白得率为90.55%,高含量的酪蛋白主要集中在电泳图谱的中分子量组,可分为ds,CN、as2-CN、β-CN和K—CN,牛羊乳as2-CN分子量羊乳大于牛乳,牛乳a-CN含量比较多,羊乳β—CN含量比较多,鲜乳与复原乳全蛋白主要组分在电泳图谱中除酪蛋白差别外,上端的高分子量组清蛋白区羊乳IgG重链的分子量比牛乳小。应用蛋白质电泳分析技术可以区分羊乳和牛乳的蛋白质组分,等电点沉淀法制备酪蛋白的方法简单,易于操作。  相似文献

建立一种快速、高效的分析酪蛋白组分氨基酸序列的LTQ-Orbitrap质谱方法。为了研究水牛奶酪蛋白、乳牛奶酪蛋白和山羊奶酪蛋白的氨基酸序列组成及氨基酸的替换现象,采用LTQ-Orbitrap液-质联用技术分别对乳源酪蛋白的4种主要组分进行分析,搜索数据库获得4种组分的氨基酸全序列。与水牛奶4种酪蛋白组分进行比对。结果表明,乳牛奶的4种酪蛋白发生氨基酸替换的部位和比率明显小于山羊奶。这意味着与水牛奶酪蛋白相比,乳牛奶酪蛋白氨基酸的稳定性优于山羊奶酪蛋白。  相似文献

The 4 casein loci were evaluated as haplotypes in 2 dairy goat breeds kept in the Czech Republic. Analysis of 41 White Shorthaired (WSH) trio families and 44 Brown Shorthaired (BSH) trio families revealed 14 and 20 haplotypes, respectively. Various genomic techniques were used to type the casein loci. Twenty-two different combinations of these alleles (casein haplotypes, in the order CSN1S1-CSN2-CSN1S2-CSN3) were found. Only 5 haplotypes in the WSH breed and 6 haplotypes in the BSH breed occurred at frequencies >0.05. For the WSH breed, the most common haplotype was FCFB (0.260), whereas for the BSH breed, the most common haplotype was FCFA (0.217). The information on the haplotype variability in both breeds could be used in breeding programs aimed at preserving biodiversity or selecting animals for specific protein production and cheesemaking.  相似文献

Non-specific binding (NSB) of compounds other than the capture antigen or antibody to the solid support is one of the most important error sources occurring in ELISA. Undesirable NSB may be minimized by saturating the remaining binding sites of the plastic surface with different protein compounds (blocking agents) that must not interfere with the immunoassay. This work had as main objective the evaluation of different blocking agents for their ability to avoid unwanted NSB in ELISA using bovine muscle crude extracts and a polyclonal antibody raised against purified cathepsin L. Results obtained showed that skimmed milk, casein, BSA, goat and rabbit serum proved to be efficient in suppressing NSB. In the case of skimmed milk and casein, an effective blocking effect was observed even in absence of the surcoating step. However, these two compounds and BSA interfered considerably the immunochemical reaction, making their use inadequate. Of the two assayed serums, rabbit serum would be a more suitable blocking agent than goat serum when working with bovine crude extracts because it did not interfere in great manner with the antigen–antibody reaction and, at the same time, did not increase background.  相似文献

In the Mediterranean region, goat milk production is an important economic activity. In the present study, 4 casein genes were genotyped in 5 Sicilian goat breeds to 1) identify casein haplotypes present in the Argentata dell’Etna, Girgentana, Messinese, Derivata di Siria, and Maltese goat breeds; and 2) describe the structure of the Sicilian goat breeds based on casein haplotypes and allele frequencies. In a sample of 540 dairy goats, 67 different haplotypes with frequency ≥0.01 and 27 with frequency ≥0.03 were observed. The most common CSN1S1-CSN2-CSN1S2-CSN3 haplotype for Derivata di Siria and Maltese was FCFB (0.17 and 0.22, respectively), whereas for Argentata dell’Etna, Girgentana and Messinese was ACAB (0.06, 0.23, and 0.10, respectively). According to the haplotype reconstruction, Argentata dell’Etna, Girgentana, and Messinese breeds presented the most favorable haplotype for cheese production, because the casein concentration in milk of these breeds might be greater than that in Derivata di Siria and Maltese breeds. Based on a cluster analysis, the breeds formed 2 main groups: Derivata di Siria, and Maltese in one group, and Argentata dell’Etna and Messinese in the other; the Girgentana breed was between these groups but closer to the latter.  相似文献

BACKGROUND: There are two main proteins in milk; whey and casein. Casein contains casein phosphopeptides (CPP), which are released on digestion of the milk. These may increase calcium solubility by binding calcium in the small intestine. Thus increasing casein in the diet may help to stimulate bioavailability of calcium and increase bone density. The present study tested this hypothesis in growing male rats fed diets containing three different concentrations of casein from goat milk. RESULTS: Rats fed the diet containing no casein had significantly lower calcium absorption when compared to rats fed the diets that contained 80% and 57% of goat milk protein as casein; however, no significant difference was observed between rats fed diets with 80% and 57% casein. The varying amounts of casein had no effect on mineral uptake or retention in the femur. Biomechanical testing and mineral analysis of the femurs showed no differences between diet groups. The mechanism to explain this lack of retention remains unclear. CONCLUSION: The diets containing 80% and 57% of goat milk protein as casein delivered increased calcium absorption compared to the diet containing no casein, suggesting a minimum level of casein is needed to optimize calcium absorption from goat milk. This study aimed to evaluate the effects of added jujube polysaccharide (JP) and Lycium barbarum polysaccharide (LBP) on the texture, rheological properties, and microstructure of goat milk cheese. Seven groups of fresh goat milk cheese were produced with 4 levels (0, 0.2, 0.6, and 1%, wt/wt) of JP and LBP. The goat milk cheese containing 1% JP showed the highest water-holding capacity, hardness, and the strongest rheological properties by creating a denser and more stable casein network structure. In addition, the yield of goat milk cheese was substantially improved as a result of JP incorporation. Cheeses containing LBP expressed lower fat content, higher moisture, and softer texture compared with the control cheese. Fourier-transform infrared spectroscopy and low-field nuclear magnetic resonance analysis demonstrated that the addition of JP improved the stability of the secondary protein structure in cheese and significantly enhanced the binding capacity of the casein matrix to water molecules due to strengthened intermolecular interactions. The current research demonstrated the potential feasibility of modifying the texture of goat milk cheese by JP or LBP, available for developing tunable goat milk cheese to satisfy consumer preferences and production needs.  相似文献

以自由基清除能力、金属离子螯合能力和抗脂质过氧化能力为指标,采用均匀试验研究山羊乳酪蛋白酶解物的制备及其体外抗氧化活性,并且比较自由基清除能力的方法。结果表明:山羊乳酪蛋白适宜的酶解工艺为在60g/kg的底物浓度下,中性蛋白酶和碱性蛋白酶的添加量分别为4000U/g和250U/g,45℃、pH7.5条件下酶解24h。山羊乳酪蛋白经酶解后其体外抗氧化活性显著增强。山羊乳酪蛋白酶解物 ·OH清除率的EC50值是其蛋白的3.59倍,ABTS+ ·的清除能力是其蛋白的158.72倍,螯合Fe2+的能力是其蛋白的5.44倍;在亚油酸体系中抗脂质过氧化能力强于TBHQ,与VE相当。清除 ·OH、DPPH自由基、O2- ·和ABTS+ ·的方法不能相互替代。  相似文献

The effect of heat, transglutaminase and combination of heat and transglutaminase treatments on the solubility of films prepared from goat milk casein, goat milk whey proteins and whole goat milk proteins was investigated. Goat milk casein films were less soluble when treated with transglutaminase and combination of heat with transglutaminase compare with heat‐treated caseins alone. Heat treatment was more effective at decreasing the solubility of whey protein films. SDS‐PAGE patterns demonstrated that goat milk caseins were better cross‐linked by transglutaminase, whereas whey proteins were better cross‐linked by heat. The extent of cross‐linking was further enhanced when a combination of heat and transglutaminase was used.  相似文献
