The present experiments were conducted to provide a more detailed behavioral analysis of the dissociable roles of the basolateral (BLA) and central nucleus (CeA) of the amygdala in mediating intra-accumbens (Acb) opioid-induced feeding of a high-fat diet. Confirming previous findings, temporary inactivation of the CeA with the GABAA agonist muscimol reduced DAMGO (D-Ala2-NMe-Phe4-Glyol5-enkephalin)-induced and baseline food intake, whereas intra-BLA muscimol selectively blocked only DAMGO-induced food intake, leaving baseline feeding intact. However, although inactivation of the BLA reduced DAMGO-induced food intake to control levels, this treatment led to exaggerated number and duration of food hopper entries after food intake had ended. A subsequent experiment under conditions of limited access to the diet found the identical pattern of behavior following intra-Acb administration of DAMGO, regardless of whether the BLA was inactivated. Last, BLA inactivation was shown to have no influence on feeding driven by a state of negative-energy balance (24-hr food deprivation), demonstrating a specific influence of the BLA on opioid-driven feeding. These findings suggest that BLA mediates palatability-driven feeding and that this influence is particular to the consummatory act of ingestion. Amounts consumed of 4 foods were recorded for each child in nursery school. 10 children were randomly selected for repeated sessions of verbal conditioning, which occurred shortly before snack time, and consisted of reinforcing the verbal behavior (food term) denoting the food selected for manipulation. The data give evidence that when positive reinforcement is associated with the verbal response denoting a food, then the consumption of that food increases. Conceptually, food can be considered a stimulus that sets up a class of responses, including verbal and eating responses. If the verbal response in that class is reinforced, hence strengthened, then the other responses of that class will be strengthened as well. Dopamine signaling has been implicated in the control of food intake and body weight. In particular, dopamine is important in the control of meal size and number and is thought to mediate the response to metabolic deprivation states. In the present experiments, the authors assessed the role of the dopamine-3 receptor (D?R) in the feeding responses to 2-deoxy-D-glucose, mercaptoacetate, and peripheral insulin. All 3 compounds increased food intake in wild-type mice, but the hyperphagic responses were blunted in D?R-/- mice. In other experiments, D?R-/- mice were hyperresponsive to the administration of amylin and leptin relative to wild-type mice. These results support the hypothesis that D?Rs chronically inhibit the effects of adiposity hormones, thereby contributing to a net anabolic state. Substantial evidence suggests that pharmacological manipulations of neural serotonin pathways influence ingestive behaviors. Despite the known role of the nucleus accumbens in directing appetitive and consummatory behavior, there has been little examination of the influences that serotonin receptors may play in modulating feeding within nucleus accumbens circuitry. In these experiments, the authors examined the effects of bilateral nucleus accumbens infusions of the 5-HT1/7 receptor agonist 5-CT (at 0.0, 0.5, 1.0, or 4.0 μg/0.5 μl/side), the 5-HT? receptor agonist EMD 386088 (at 0.0, 1.0, and 4.0 μg/0.5 μl/side), or the 5-HT2C preferential agonist RO 60–0175 (at 0.0, 2.0, or 5.0 μg/0.5 μl/side) on food intake and locomotor activity in the rat. Intra-accumbens infusions of 5-CT caused a dose-dependent reduction of food intake and rearing behavior, both in food-restricted animals given 2-hr free access to Purina Protab RMH 3000 Chow, as well as in nondeprived rats offered 2-hr access to a highly palatable fat/sucrose diet. In contrast, stimulation of 5-HT? receptors with EMD 386088 caused a dose-dependent increase of intake under both feeding conditions, without affecting measures of locomotion. Infusions of the moderately selective 5-HT2C receptor agonist RO 60–0175 had no effects on feeding or locomotor measures in food-restricted animals, but did reduce intake of the fat/sucrose in nonrestricted animals at the 2.0 μg, but not the 5.0 μg dose. Intra-accumbens infusions of selective antagonists for the 5-HT? (SB 269970), 5-HT? (SB 252585), and 5-HT2C (RS 102221) receptors did not affect locomotion, and demonstrated no lasting changes in feeding for any of the groups tested. These data are the first to suggest that the activation of different serotonin receptor subtypes within the feeding circuitry of the medial nucleus accumbens differentially influence consummatory behavior. The orexigenic neuropeptide ghrelin is an endogeneous ligand for the growth hormone secretagogue receptor (GHS-R). This orexigen is expressed in both the periphery and in the central system, including portions of mesolimbic dopaminergic circuitry that play a role in affective behaviors. Here we examined pharmacological antagonism of GHS-R in motivational incentive learning, as reflected in Pavlovian-to-instrumental transfer (PIT). Furthermore, it is currently unclear whether the previous effects of ghrelin on food intake are mediated by pre- and/or postingestive influences on ingestive behavior. Thus, the authors also conducted detailed analyses of the temporal dynamics of sucrose licking. Mice received low (50 nmol), moderate (100 nmol), and high (200 nmol) intraperitoneal injections of the GHS-R antagonist GHRP-6 [D-Lys3] prior to subsequent transfer and sucrose consumption tests. Low and moderate doses led to an augmentation of PIT, while high dose injections led to generalized performance deficits. In addition, moderate and high doses of the antagonist resulted in reductions in sucrose intake by reducing palatability of the sucrose. The current study investigated the nature of the flow state among older adults. Flow is a pleasurable experiential state that occurs during full-capacity engagement in which an individual is performing at a level that is matched with the demands of the task. Each participant completed a scale assessing dimensions of flow in a particular activity selected by the participant. More cognitively demanding activities elicited higher levels of flow for those with higher fluid ability, but lower levels of flow for those with lower fluid ability. This pattern was reversed for activities that were low in demand. Our data highlight the potential importance of considering motivational states such as flow in understanding cognitive optimization in adulthood. The standard activity-based anorexia procedure provides rats with access to a running wheel while restricting their access to dry food. This can produce reduced food intake and progressive weight loss. Using this procedure, in the present study (Experiment 1) the authors found changes in drinking patterns both in the period of high activity preceding food access and during the feeding period. Varying the procedure by providing wet mash (Experiment 2) or by prior adaptation to a drinking schedule (Experiment 3) prevented the self-starvation effect. This study examined the effect of anticipated food deprivation on intake in restrained and unrestrained eaters. Participants were randomly assigned to a diet condition, in which they expected to diet for a week, or to a control (no-diet) condition. Immediately after being assigned to a condition, participants completed a taste-rating task in which food consumption was measured. Restrained eaters in the diet condition consumed significantly more food than did restrained eaters in the no-diet condition or unrestrained eaters in either condition. Unrestrained eaters consumed the same amount regardless of condition. These results confirm that merely planning to go on a diet can trigger overeating in restrained eaters, reflecting the dynamic connection between dieting and overeating. The current studies were designed to evaluate whether incentive motivation for palatable food is altered after manipulations of opioid, GABAergic, and dopaminergic transmission within the nucleus accumbens. A progressive ratio schedule was used to measure lever-pressing for sugar pellets after microinfusion of drugs into the nucleus accumbens in non-food-deprived rats. The mu opioid agonist D-Ala2, NMe-Phe4, Glyol5-enkephalin and the indirect dopamine agonist amphetamine induced a marked increase in break point and correct lever-presses; the GABAA agonist muscimol did not affect break point or lever-presses. The data suggest that opioid, dopaminergic, and GABAergic systems within the accumbens differentially modulate food-seeking behavior through mechanisms related to hedonic evaluation of food, incentive salience, and control of motor feeding circuits, respectively. Affective priming of aggression was examined in groups low and high in trait negative emotionality (NEM) using a Buss aggression paradigm. Negative affect was induced by exposure to aversive air blasts during some intervals (threat) and not others (safe). Phasic negative affect was assessed using startle reflex potentiation, and tonic distress was indexed by startle sensitization. Participants delivered shocks faster during threat versus safe intervals, indicating that phasic distress primed aggression. Following initial exposure to air blasts, high NEM participants showed enhanced tonic distress and delivered persistently more intense shocks than low NEM participants. These findings indicate that sustained negative affect biases high stress-reactive individuals toward more intense acts of aggression, with phasic distress affecting the rapidity of aggressive response.
