Determination of total phenolic (TPC), flavonoid and anthocyanin contents, and various antioxidant activities (2,2-diphenyl-1-picrylhydrazyl radical scavenging, ferric reducing power, ferrous ion chelating and lipid peroxidation inhibition) of leaves and flowers of Bauhinia kockiana, Caesalpinia pulcherrima and Cassia surattensis were performed in this study. The B. kockiana flower was found to possess the highest TPC (8280 ± 498 mg GAE/100 g), free radical scavenging activity (ascorbic acid equivalents 14,600 ± 2360 mg AA/100 g) and reducing ability (72.4 ± 8.7 mg GAE/g). Rutin and chlorogenic acid were detected in the plants, where the C. pulcherrima leaf contained the highest amount of rutin (669 ± 26 mg/100 g), while minute amounts of chlorogenic acid were detected in C. surattensis leaf (9.13 ± 0.44 mg/100 g). The C. pulcherrima leaf displayed the highest ferrous ion chelating and lipid peroxidation inhibition activities. Positive correlation was observed between TPC and various antioxidant activities.  相似文献

Methanolic extracts of six cultivars of Portulaca oleracea were analyzed for their total phenol content (TPC) using the Folin–Ciocalteu method. The antioxidant activity was measured using the 1,1-diphenyl-2-picrylhydrazyl, ferric-reducing antioxidant power (FRAP) and β-carotene bleaching (BCB) assays. The iodine titration method was used to determine the ascorbic acid content (AAC). The TPC of the cultivars of P. oleracea ranged from 127 ± 13 to 478 ± 45 mg GAE/100 g of fresh weight of plant. There was good correlation between the TPC value and its AEAC, IC50 and FRAP values (r2 > 0.9) for all the cultivars. The AAC for the cultivars ranged from 38.5 ± 0.6 to 73.0 ± 17.5 mg/100 g. The TPC value of the common variety PO1, was the lowest compared to the ornamental cultivars (PO2–PO6). The BCB assay showed that all cultivars were capable of inhibiting lipid peroxidation and the inhibition power did not correlate with TPC value.  相似文献

Methanolic extracts from fresh leaves of five Etlingera species were screened for total phenolic content (TPC), antioxidant activity (AOA), and antibacterial activity. Analysis of TPC was done using the Folin–Ciocalteu method. Evaluations of AOA included 1,1-diphenyl-2-picrylhydrazyl free radical-scavenging ability, ferric-reducing antioxidant power (FRAP), ferrous-ion chelating (FIC) ability, and β-carotene bleaching (BCB) activity. Antibacterial activity was screened using the disc-diffusion method. Highest TPC, ascorbic acid equivalent antioxidant capacity (AEAC), and FRAP were found in leaves of E. elatior and E. rubrostriata. Leaves of E. maingayi, with the lowest TPC, AEAC, and FRAP, had the highest FIC ability and BCB activity. Ranking of TPC and AOA of different plant parts of E. elatior was in the order: leaves > inflorescences > rhizomes. Leaves of highland populations of Etlingera species displayed higher values of TPC and AEAC than those of lowland counterparts. Leaves of Etlingera species exhibited antibacterial activity against Gram-positive but not Gram-negative bacteria.  相似文献

Four (red, violet, white and green) varieties of Allium cepa were studied for their total phenolic contents (TPC), antioxidant (AOA) and free radical scavenging activities (FRSA). The TPC varied from 4.6 to 74.1 mg/g GAE, AOA varied from 13.6% to 84.1% and FRSA showed wide range in terms of IC50 (inhibitory concentration) from 0.1 to 15.2 mg/ml, EC50 (efficient concentration) from 4.3 to 660.8 mg/mg and ARP (antiradical power) from 0.15 to 23.2. The outer dry layers of red and violet varieties showed better inhibition of lipid peroxidation assayed by ammonium thiocyanate than α-tocopherol. The non-site-specific inhibition of hydroxyl radical induced deoxyribose degradation was also higher in the outer dry layers of red and violet varieties than in their middle and inner layers. The outer layers were also potential inhibitors of nitroblue tetrazolium chloride (NBT) reduction caused by superoxide anions. On the other hand the ferrous ion chelating capacity of the red and violet varieties was highest in the inner layers. Specific phenolic composition performed through HPLC and LC–MS/MS showed the presence of gallic acid, ferulic acid, protocatechuic acid, quercetin, and kaempferol. The unutilised outer layers of the red variety were a rich source of quercetin (5110 μg/g) with high AOA, FRSA and also showed significant protection of DNA damage caused by free radicals.  相似文献

In order to find antioxidant potential, seeds of 30 varieties of Glycine max were studied for their total phenolic contents (TPC), flavonoids and antioxidant activity (AOA). The seed extracts showed wide variation of TPC from 6.4 to 81.7 mg GAE/g, flavonoids 3.5 to 44.6 mg QE/g and AOA 7.5% to 74.7%. Free radical-scavenging activity (FRSA), assayed by DPPH in terms of IC50 (inhibitory concentration), ranged from 0.14 to 0.80 mg/ml, EC50 (efficiency concentration) from 6.1 to 34.8 mg/mg DPPH, ARP (anti-radical power) 2.9 to 16.4 and reducing power from 1.9 to 4.7 ASE/ml. Variety Kalitur showed highest the FRSA followed by Alankar and Hara soya, as evident from their low IC50, EC50 and high ARP values. Alankar, Kalitur NRC-37, PK-472, VLS-47, Hara soya varieties were with comparatively higher TPC (52.7–81.7 mg GAE/g), AOA (50.5–74.7%) and showed better inhibition of peroxide formation assayed through ammonium thiocyanate and egg yolk, non-site-specific and site-specific inhibition of hydroxyl radical induced deoxyribose degradation and ferrous ion-chelating capacity than did the other varieties. Seed extracts of these varieties and leaves of Kalitur showed significant protection against DNA damage caused by free radicals. The agri-wastes of some promising varieties, e.g. Alankar, Kalitur, NRC-37 and PK-472, showed TPC ranging from 27.4 to 167 mg GAE/g, total flavanoids from 10.4 to 63.8 mg QE/g and AOA from 26.5% to 84.7% and their values were highest in the leaves, followed by pod pericarp and twigs. Out of all the varieties studied, leaves of Alankar and Kalitur varieties were more potent free radical-scavengers than were seeds, pod pericarp or twigs. The specific phenolic compositions and their quantifications were performed by HPLC and MS/MS, which showed that the seeds of Kalitur were higher in genistin (127 μg/g), seeds and leaves of Alankar in diadzin (113 μg/g) and gallic acid (87.2 μg/g), respectively. The present studies may be of importance in varietal improvement, nutraceuticals, bio-pharmaceuticals and utilization of agri-wastes as possible cost-effective natural antioxidants.  相似文献

The methanolic fresh leaf extracts of Macaranga gigantea, Macaranga pruinosa, Macaranga tanarius and Macaranga triloba were screened for their antioxidant properties (AOP), tyrosinase inhibition and antibacterial activities. Total phenolic content (TPC), 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging, ferric-ion reducing power (FRAP), ferrous-ion chelating (FIC) and lipid peroxidation inhibition (LPI) activities were used to evaluate the AOP. Modified 3,4-dihydroxy-L-phenylalanine (L-DOPA) method was used to determine tyrosinase inhibition activity, whereas antibacterial activity was determined using the disc-diffusion technique. TPC screening of the same species from different collection sites showed no significant difference between sites. M.triloba showed the highest ascorbic acid equivalent antioxidant activity (AEAC), FRAP and LPI values. M. tanarius, which showed the lowest TPC, AEAC, FRAP and LPI activities, exhibited the best FIC activity. M. pruinosa showed the best tyrosinase inhibition activity, whereas M. triloba showed the best antibacterial activity against Gram-positive bacteria species, with minimal inhibition dosage (MID) values as low as 10 μg/disc.  相似文献

Citrus sulcata was subjected to ultrasound, high-pressure, and Soxhlet extractions. The antioxidant and anti-inflammatory activities of the extracts were evaluated qualitatively and quantitatively. The antioxidant content of the peel extract was twice that of the fruit extract. The quantitative analysis showed that the narirutin and hesperidin contents in the peel extracts were 8.8 and 7.5 mg/100 g, respectively. These extracts had a total phenolic content of 112.22 ± 2.89 gallic acid equivalent (GAE) mg/100 g, a total flavonoid content of 54.09 ± 1.01 rutin equivalent (RE) mg/100 g, DPPH radical scavenging activity of 46%, and antioxidant activity of 213.25 ± 2.82 μM of Trolox equivalents (TEAC). C. sulcata extracts could prevent hydrogen peroxide (H2O2)-induced oxidative stress, reduce expression of the inflammatory markers nuclear Factor kappa B (NFκB) and phosphorylated IκBα (p-IκBα) in A549 human lung carcinoma cells, and inhibit Lipopolysaccharide (LPS)-induced THP-1 monocyte differentiation to an extent of 85%.  相似文献

The aim of this study was to determine the antioxidant potency and total phenolic and flavonoid contents of kiwifruit (Actinidia deliciosa) in vitro by analysing the radical scavenging activity of lyophilised water extract from kiwifruit (LEK) for 2,2′-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) (ABTS), 1,1-diphenyl-2-picryl-hydrazyl (DPPH), N,N-dimethyl-p-phenylenediamine (DMPD), and superoxide anion radical (O2) as well as the total reducing power by FRAP and CUPRAC assays and the metal chelating activities. LEK showed efficient radical scavenging activity with DPPH, ABTS, DMPD, and O2 radicals; ferric (Fe3+) and cupric (Cu2+) ion reducing power and metal chelating activities. Moreover, the amounts of phenolic compounds, such as caffeic acid, ferulic acid, syringic acid, ellagic acid, catechol, pyrogallol, p-hydroxy benzoic acid, vanillin, p-coumaric acid, gallic acid, quercetin, ??-tocopherol and ascorbic acid, in LEK were quantified by LC-MS-MS. The results show that pyrogallol (2070.0 mg/kg LEK) is the main phenolic compound responsible for the antioxidant and radical scavenging activities of LEK. Finally, total phenolic and flavonoid contents were determined as gallic acid (GAE) and quercetin equivalents (QE). The GAE and QE values in LEK were 16.67 ± 2.83 ??g GAE/mg and 12.95 ± 0.52 ??g QE/mg, respectively. The results suggest that consumption of kiwifruit (A. deliciosa) can be beneficial effects due to its antioxidant properties.  相似文献

Phenol content and antioxidant activity of two Spanish onion varieties, namely white onion and Calçot de Valls, have been studied. White onions contained higher phenol content than Calçot onions, with values which ranged from 2.57 ± 0.51 to 6.53 ± 0.16 mg gallic acid equivalents/g dry weight (GAE/g DW) and 0.51 ± 0.22 to 2.58 ± 0.16 mg GAE/g DW, respectively, depending on the solvent used. Higher phenol content was associated with higher antioxidant capacity. White onion extracts had the highest antioxidant activity at 86.6 ± 2.97 and 29.9 ± 2.49 μmol Trolox/g DW for TEAC and FRAP assays, respectively, while the values for the Calçot variety were 17.5 ± 0.46 and 16.1 ± 0.10 μmol Trolox/g DW.  相似文献

Wild greens are nutritionally well-balanced vegetables. Herein, nutritional and in vitro antioxidant properties of the sprouts of three commonly used species were determined. Wild asparagus revealed the highest levels of moisture (84.6 g/100 g fw), ash (12.3 g/100 g dw), proteins (22.4 g/100 g dw), total sugars (9.24 g/100 g dw), including sucrose (4.27 g/100 g dw), and of the essential n-6 fatty acid linoleic acid (44.5%); white bryony gave the highest contents of reducing sugars, including glucose (2.97 g/100 g dw), essential n-3 fatty acid α-linolenic acid (70.3%), and the best ratios of PUFA/SFA, and n-6/n-3 fatty acids (3.59 and 0.0907, respectively); black bryony showed the highest concentrations of carbohydrates (69.3 g/100 g dw), fructose and trehalose (3.83 and 1.34 g/100 g dw, respectively). Besides their culinary characteristics, their high content in vitamins (asparagus, 135 and 142 mg/100 g dw of total tocopherols and ascorbic acid, respectively), chlorophylls (white bryony, 50.9 mg/100 g dw), carotenoids (23.3 mg/100 g dw) and phenolics (black bryony, 759 mg GAE/g extract), together with the antioxidant properties (EC50 values lower than 640 μg/ml) and potential health benefits increase their importance in traditional as well as in contemporary diets.  相似文献

Total phenolic content, proanthocyanidins, gallotannins, flavonoids, and antioxidant activities of Sclerocarya birrea and Harpephyllum caffrum methanolic extracts were evaluated using in vitro assays. S. birrea young stem extract contained the highest levels of total phenolic content (14.15 ± 0.03 mg GAE/g), flavonoids (1.21 ± 0.01 mg CE/g) and gallotannins (0.24 ± 0.00 mg GAE/g). H. caffrum stem bark extract had the highest content of proanthocyanidins (1.47%). The EC50 values of the extracts in the DPPH free radical scavenging assay ranged from 4.26 to 6.92 μg/ml, compared to 6.86 μg/ml for ascorbic acid. A dose-dependent linear curve was obtained for all extracts in the ferric-reducing power assay. Dichloromethane and methanol extracts exhibited dose-dependent acetylcholinesterase inhibitory activity. Similarly, all extracts exhibited high antioxidant activity comparable to butylated hydroxytoluene based on the rate of β-carotene bleaching (84.1–93.9%). The two Anacardiaceae species provide a source of natural antioxidants and acetylcholinesterase inhibitors, and may be beneficial to the health of consumers.  相似文献

Methanol extracts of fresh tea leaves from a lowland plantation in Malaysia were screened for total phenolic content (TPC) and antioxidant activity (AOA). AOA evaluation included 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical-scavenging ability, ferric-reducing antioxidant power (FRAP), and ferrous-ion chelating (FIC) ability. Ranking, based on TPC and AOA, was as follows: shoots > young leaves > mature leaves. TPC and AOA of lowland leaves were comparable to those of highland plants. A green tea produced by drying young leaves in a household microwave oven for 4 min showed significantly higher TPC and AOA than did four commercial brands of green and black tea.  相似文献

Ergosterol peroxide, the steroidal derivative with cytotoxic activity, has been isolated for the first time from the mycelium of edible and medicinal mushroom Hericiumerinaceum (lion’s mane mushroom) together with erinacine A. The new densitometric method was applied for the quantitative determination of ergosterol peroxide in n-hexane extracts of H. erinaceum, Laetiporus sulfureus (chicken mushroom), and Morchella esculenta (common morel) mycelia, as well as in Boletus edulis (king bolete), Suillus bovinus (Jersey cow mushroom), and B. badius (bay bolete) fruiting bodies. The ergosterol peroxide content reached 15.98 ± 0.78, 10.07 ± 0.75, 13.37 ± 0.56, 29.32 ± 1.43, 17.27 ± 0.84, and 12.60 ± 0.59 mg per 100 g, respectively. What is significant was that ergosterol peroxide was identified for the first time, to the best of our knowledge, in edible mushrooms mentioned above.  相似文献

Arbutin is a hydroquinone derivative that has been found in species of several plant families. Within the genus Origanum the formation of arbutin is polymorphic, with arbutin present in considerable amounts (O. dubium 20.8 ± 15.3 mg/g; wild O. majorana 51.3 ± 15.4 mg/g, cultivated O. majorana 40.6 ± 11.2 mg/g), minor amounts (O. microphyllum 0.1 ± 0.1 mg/g, wild O. onites 0.3 ± 0.1 mg/g, cultivated O. onites 0.1 ± 0.1 mg/g, O. saccatum 0.1 ± 0.1 mg/g, O. solymicum 0.4 ± 1.0 mg/g) or completely absent (O. husnucan-baseri, O. syriacum, O. vulgare). Whereas the most important commercial oregano species (O. onites and O. vulgare) contain no or only minor amounts of arbutin, marjoram (O. majorana) has considerably high amounts. The high variability of arbutin in O. majorana would allow a selection into cultivars with high arbutin content and low arbutin varieties. In a segregating F2-generation of a species crossing between O. majorana (high content of arbutin) and O. vulgare ssp. vulgare (free of arbutin), the presence of arbutin followed a Mendelian segregation of 3:1, indicating that only one gene is responsible for the polymorphism of arbutin in the genus Origanum. The absence of arbutin in O. vulgare ssp. vulgare or O. syriacum would even enable the breeding of marjoram with no arbutin at all.  相似文献

An investigation into the effects of ethanol concentration (0–100%, v/v), extraction time (20–120 min) and extraction temperature (25–65 °C) on the extraction of phenolic antioxidants from mengkudu (Morinda citrifolia) was performed using a single-factor experiment. Total phenolic content (TPC) and total flavonoid content (TFC) assays were used for determination of phenolic compounds. Antioxidant capacity was evaluated by measuring the scavenging effect on 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) and 2,2′-diphenyl-1-picrylhydrazyl (DPPH) radicals. Experimental results showed that extraction conditions had significant effect on extraction of phenolic compounds and antioxidant capacities. The optimised conditions were 40% ethanol for 80 min at 65 °C, with values of 919.95 mg GAE/100 g DW for TPC, 472.73 mg CE/100 g DW for TFC, 791.71 μmol TEAC/100 g DW for ABTS and 1928.5 μmol TEAC/100 g DW for DPPH. TPC was significantly correlated with DPPH under the effects of ethanol concentration (r = 0.932) and extraction time (r = 0.938).  相似文献

The flora of Latin America attracts gaining interest as it provides a plethora of still unexplored or under-utilized fruits that can contribute to human well-being due to their nutritional value and their content of bioactive compounds. Clidemia rubra (Aubl.) Mart. is a shrub belonging to the family of the Melastomataceae that grows preferably in a tropical climate. This paper comprises a nutritional characterization of the berries from Clidemia rubra and provides data on the phenolic compounds as well as the antioxidant capacity of the fruit. Findings in macronutrients like protein, carbohydrates, and fat were comparable to that of common berry fruits. Clidemia rubra berries seemed to be a good source for dietary fibers and some minerals (Ca, Mn, and Zn). In contrast, contents of titratable acids and ascorbic acid were low. The polyphenolic profile was determined by using HPLC-MS/MS in comparison to standard compounds. Noteworthy amounts of cyanidin 3-O-rutinoside (39.43 ± 1.66 mg/100 g fresh weight (FW)), delphinidin 3-O-rutinoside (23.74 ± 1.18 mg/100 g FW), cyanidin 3-O-glucoside (11.68 ± 0.56 mg/100 g FW), and delphinidin 3-O-glucoside (6.08 ± 0.35 mg/100 g FW) were found. Non-anthocyanin phenolic constituents were phenolic acids (gallic, protocatechuic, p-hydroxy-benzoic, vanillic, and caffeic acid), flavan-3-ols (epigallocatechin, epigallocatechin gallate, and epicatechin gallate), and 11 different myricetin and quercetin derivatives of which quercetin 3-O-arabinoside (5.26 ± 0.16 mg/100 g FW) and quercetin 3-O-rhamnoside (5.06 ± 0.08 mg/100 g FW) were dominating. Anthocyanins and ascorbic acid were mainly responsible for the antioxidant capacity of Clidemia rubra berries assessed with the total oxidant scavenging capacity (TOSC) assay.  相似文献

The total phenolic content (TPC), total antioxidant status (TAS), free radical scavenging capacity, inhibition of low density lipoprotein (LDL) cholesterol and metal chelating capacity of extracts of whole black and whole white sesame seeds and their hull fractions in 80% aqueous ethanol were investigated. The TPC of whole black sesame seeds and hull extracts were 29.9 ± 0.6 and 146.6 ± 0.6 mg catechin equivalents/g crude ethanolic extract, respectively. The corresponding values for white sesame were 10.6 ± 1.6 and 29.7 ± 0.9 mg catechin equivalents/g crude ethanolic extract. The TAS as determined by Trolox equivalent antioxidant capacity assay and expressed as Trolox equivalents was highest for black sesame hulls (65.9 ± 1.7) while white seeds showed the lowest (4.4 ± 0.6). Free radical scavenging capacity of sesame extracts (5–40 μg/mL) was measured using 2,2-diphenyl-1-picrylhydrazyl radical. The highest scavenging capacity was obtained at 40 μg/mL and was 94.9 ± 0.8, 25.1 ± 0.4, 14.4 ± 0.9 and 2.5 ± 0.4 for black sesame hulls, black sesame seeds, white sesame hulls and white sesame seeds, respectively. Inhibition of LDL oxidation at 100 ppm level was highest for black sesame hulls (96.7%) followed by those for white sesame hulls (84.6%), black sesame (78.4%) and white sesame seeds (57.3%). Sesame products displayed good ferrous ion chelating capacities, which ranged from 12% to 46% and 17% to 62% at 50 and 100 ppm levels, respectively. Results demonstrated considerable antioxidant activity of sesame products tested especially black sesame hulls.  相似文献

Plants are a source of compounds that may be used as pharmacologically active products. Cytisus multiflorus, Filipendula ulmaria and Sambucus nigra have been used as important medicinal plants in the Iberian Peninsula for many years, and are claimed to have various health benefits. Herein, the phytochemical composition and antioxidant activity of the mentioned wild medicinal plants were evaluated in vitro, based on chemical, biochemical and electrochemical methods. F. ulmaria was found to be richest in antioxidant phytochemicals, such as phenolics (228 mg GAE/g DW), flavonoids (62 mg CE/g DW), ascorbic acid (2700 μg/g DW) and tocopherols (497 μg/g DW). The antioxidant activity was found to vary in the order: F. ulmaria S. nigra > C. multiflorus, irrespective of the analysis method. Electrochemical methods have proven to be rapid and inexpensive techniques to characterise the antioxidant activity of plant extracts.  相似文献

The antioxidant capacities and phenolic composition in six native, commercially grown, Australian herbs and spices were investigated. Tasmannia pepper leaf, followed by anise myrtle and lemon myrtle contained the highest levels of total phenolics (TP; 102.1; 55.9 and 31.4 mg gallic acid equivalents (GAE)/g dry weight (DW), respectively). Tasmannia pepper leaf exhibited the highest oxygen radical absorbance capacity (ORAC assay) followed by lemon myrtle and anise myrtle. Anise myrtle exhibited the highest total reducing capacity [TRC; Ferric Reducing Antioxidant Power (FRAP) assay], followed by Tasmannia pepper leaf and lemon myrtle. Australian bush tomato, with TP content of 12.4 ± 0.9 mg GAE/gDW and TRC of 206.2 μMol Fe+2/gDW, resembled the Chinese Barbary Wolfberry fruit. The TP content of Tasmannia pepper berry (16.86 mg GAE/gDW) was similar to that of black pepper, but it’s TRC was 25% lower. Cinnamic acids and flavonoids, tentatively identified by mass spectrometry, were identified as the main sources of antioxidant activities.  相似文献

We investigate in this work the chemical composition by GC–EIMS, the antibacterial and the cytotoxic activities of Tunisian Nigella sativa essential oil and its bioactive compound, thymoquinone, were tested against various clinical cariogenic bacteria (n = 30). Eighty-four compounds were identified in the essential oil. The major one was p-cymene (49.48%) whereas thymoquinone represented only 0.79%. The essential oil (2.43 mg/disc) containing only 3.35 μg of thymoquinone showed pronounced dose dependant antibacterial activity against Streptococcus mitis, Streptococcus mutans, Streptococcus constellatus and Gemella haemolysans (15.5 ± 0.707 mm). However, pure thymoquinone compound (150 μg/disk) was active against all the studied strains especially S. mutans and S. mitis (24.5 ± 0.71 and 22 ± 1.41 mm inhibition zones, respectively).  相似文献
