Different doses of ketamine (10 mg/kg, 20 mg/kg, 30 mg/kg, 40 mg/kg, 50 mg/kg, and 60 mg/kg) were injected i.p. (I.P.), respectively, to male ICR mice to determine the optimal dosage for chronic administration. At and above 40 mg/kg I.P. injection, mice had almost no hindlimb movement during swimming test. Subsequently, 30 mg/kg was used as the dose for the study in the toxicity of long-term ketamine administration on urinary bladder and sperm motility. The treatment group were subdivided into two (n = 10 each group); one received daily ketamine treatment i.p. for 3 months and another group for 6 months. Corresponding number of mice in control groups (n = 5 each group) received saline injection instead of ketamine. Terminal dUTP nick and labeling (TUNEL) study and Sirius red staining were carried out on the sectioned slides of the urinary bladders to study the degree of apoptosis in both epithelium and muscular layers of the urinary bladder and the relative thickness of the muscular layers in this organ was also computed. Apoptosis in the bladder epithelium was observed initially in the 3-month ketamine treated mice and the number of apoptotic cells was significantly different (P < 0.05) between the 3-month and 6-month ketamine treated mice and the control. The relative thickness of muscular layers in the bladder wall also decreased significantly (P < 0.05) when the 6-month treated mice and the control were compared. Sirius red staining revealed increase of collagen in the urinary bladder of the treated mice, most evidently 6 months after ketamine treatment. In addition, the sperm motility was studied and there was a statistically significant difference between the control and ketamine treated groups in the percentages of sperms which were motile (P < 0.05). This suggested that the chronic administration of ketamine affected the genital system as well.  相似文献

The mucins of colonic murine mucus are highly O‐glycosilated sulfosialoglycoproteins. We have characterized the sialylation pattern of oligosaccharide chains of colonic murine mucins by conventional histochemical methods and by lectin histochemistry combined with chemical pretreatments and sialidase digestion. Oligosaccharide chains are strongly sulphated, with an increase of sulfation from the proximal toward the distal colon and a decrease of sialic acid expression and acetylation toward the distal colon. In the goblet cells of proximal colon, sialic acid bound α2,3 to Galβ1,3GalNAc subterminal dimers is diacetylated at C7,C8;C7,C9;C8,C9 or triacetylated at C7,8,9. In the distal colon, sialic acid‐linked α2,3 to Galβ1,3GalNAc subterminal dimers shows reduced O‐acetylation at C7 and/or C8, while acetyl substituents at C9 and at C4 are almost absent. Sialic acid is involved in different essential physiological functions; thus, alterations of its expression and acetylation in oligosaccharide chains of intestinal mucins are generally associated with diseases, such as ulcerative colitis and cancer. Mice may represent a suitable animal model to study alterations of oligosaccharidic chains in colonic mucins and lectin histochemistry combined with chemical pretreatments, and enzyme digestion may be a valuable tool for this study. Our present work may represent a landmark for further lectin histochemical studies to evaluate alterations of mouse colon mucins under different physiological, pathological, or experimental conditions, with possible translational value in humans. The gastric glands synthesize glycoproteins whose oligosaccharides are linked to the peptide core mainly by the O-glycosidic bond, specifically removed by beta-elimination procedure. Our aim was to research the possibility of the existence of two subtypes of O-linked oligosaccharides with a different behavior to the removal procedure. The lectins from peanut (PNA) and Maackia amurensis (MAA-I) were histochemically used as markers of the O-linked oligosaccharides. Sections were also pretreated with beta-elimination and/or peptide N-Glycosidase F (PNGase-F) for the specific removal of O- and N-linked oligosaccharides, respectively. The lectin GNA, which mainly labels to N-linked oligosaccharides, was used to test the correct working of PNGase-F. To test the possibility that the beta-elimination treatment could remove the terminal sialic acid residues, the lectin LFA was used. The surface epithelium was negative to PNA, while it became strongly positive when beta-elimination was performed for 1 day. This staining was resistant to PNGase-F, suggesting that PNA was labeling to O-linked oligosaccharides. However, after beta-elimination for 5 days this staining is not observed. A similar pattern appeared with MAA-I. We propose the existence of two subtypes of O-linked oligosaccharides: labile and resistant. The labile O-linked oligosaccharides are removed with beta-elimination for 1 day, unmasking the PNA-positive oligosaccharides. These oligosaccharides are resistant O-linked oligosaccharides because staining is abolished with longer treatment of beta-elimination. The results with MAA-I also support this suggestion. In summary, the labile O-linked oligosaccharides are removed with short treatment, while the resistant O-linked oligosaccharides need a stronger procedure (for 5 days).  相似文献

While Corriedale sheep survive in a wide range of climates, which prevents them to specialize for one climatic condition only, dromedary camels strictly adapted to desert areas. This demands more adaptive mechanisms to hot, dry conditions in camels than in sheep. Being the entrance of the nasal cavity, nasal vestibule is subjected to various environmental stressors. A protective way is the lining epithelium which is cornified in camel, but not in sheep. Mucus nasal secretions also play a key role in the protection of underlyings. Additionally, arterio‐venous anastomosis is present in the lamina propria of the nasal vestibule of camel. In the present paper, sugar residues in the nasal vestibule of camel were analyzed and compared with those of sheep using 14 types of lectins to explore the distribution of glycoconjugates that may help the function of camel nasal vestibule in desert environment. In camel, none of the lectins could label the basal cells of the vestibular epithelium, although the basal cells reacted with six lectins in sheep. In camel, LEL and RCA‐120 markedly labeled the luminal surface. WGA, DBA, SBA, and VVA produced marked intensities on the luminal surface in sheep. The mucous glands reacted with six lectins: WGA, s‐WGA, VVA, PNA, PHA‐E, and PHA‐L in camel, while all lectins used except s‐WGA and PHA‐E reacted in the sheep. In summary, great differences are observed in the glycoconjugate expression between camel and sheep. This suggests that these glycoconjugate are related to camel's tolerance for environmental stressors.  相似文献

This study describes the very first assessment of the expression and localization of translationally controlled tumor protein (TCTP) in adult rat urinary organs. TCTP expression levels in kidneys, urinary bladder, and urethra were evaluated by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS–PAGE) and Western blotting, and its cellular localization was examined immunohistochemically in paraffin sections of various urinary organs. TCTP was found in all urinary organs. Its expression was high in the urethra and low in the bladder. TCTP was localized in glomerular podocytes, epithelium of proximal and distal renal tubules, in the loop of Henle, and in the transitional epithelium of the bladder and urethra, mostly in basal cell layers). The subcellular localization of TCTP in these urinary organs was cytoplasmic. These findings suggest that TCTP may be involved in urine formation and excretion. Microsc. Res. In this study, the ultrastructure of living BIU-87 cells of human bladder cancer was mapped using atomic force microscopy to reveal the dynamic change of single cancerous cell division. Simultaneously, the feasibility and functional reliability of the atomic force microscope (AFM) were established and a laboratory model using AFM to study living cancerous cells was created. In this experiment, BIU-87 cells of human bladder cancer were cultured by conventional methods and grown in gelatin-treated dishes. A thermostat was used for preserving the cell's living temperature. Scanning of these cells using AFM was carried out in physiologic condition. The AFM images of the ultrastructure of living BIU-87 cells as well as those of the cell's membrane and cytoskeleton were very clear. The dynamic phenomenon of single cell division was observed. It was concluded that the AFM was able to observe and depict the ultrastructure of living cells of human bladder cancer directly and in real time. This experimental model is expected to play an important role in elucidating the cancerous mechanism of bladder normal cells at the atomic or nanometer level.  相似文献

Many pathogens require direct binding to mucosal cells to cause an infection. The mucosal epithelium of the digestive tract, which is covered by a mucin layer, fulfills several protective functions that are essential to maintaining the health of the digestive tract. Mucins are glycoproteins, which are found on membranes and in mucus gels and protect the underlying mucosal cells. Both membrane‐associated mucins and secreted mucins are critical components of mucosal defense. The aim of this study was to determine the localization and expression of mucin profile of the abomasum via histochemistry and immunohistochemistry. The abomasums of 20 bulls and 20 rams were evaluated. Histochemical examination showed that neutral and acidic mucins were present in the mucosa and the glands of the pars cardiaca, fundus, and pars pylorica of the abomasums of both bulls and rams. However, the expression of acidic mucins was weak in the superficial glands and strong in the deep glands of the abomasum of rams. In both bulls and rams, MUC1, MUC5AC, and MUC6 were expressed in the glandular epithelial cells in all regions of the abomasum. Interestingly, while MUC2 was not expressed in the pars cardiaca and fundus, it was weakly expressed in the parietal cells of the pars pylorica in both species. In conclusion, the presence of neutral and acidic mucins and MUC1, MUC2, MUC5AC, and MUC6 proteins in luminal epithelial and glandular cells of abomasum in the bulls and rams support the hypothesis that mucins play a key role in the protection of the abomasal mucosa against infectious agents.  相似文献

Utilizing electron microscopic observation, several contacts between small, granule-containing cells (SGC) and postganglionic neurons (PGN) in the celiac ganglion of the guinea pig have been observed. A SGC in very close association with a PGN was seen to receive a distinct synaptic contact that contained many vesicles with dense cores. This contact was morphologically unlike cholinergic synapses previously reported on chromaffin cells. Because the SGC and PGN were clearly separated by a thin rim of satellite cell cytoplasm mutual to both cells, it is not known how or if the SGC would possibly exert a synaptic or paracrine effect on the PGN. Also, intraganglion SGC existed as large well-vascularized islands within the celiac ganglion. These intraganlion clusters sometimes contained more than 50 cells and perhaps could be considered to function as localized neuroendocrine components within the ganglion by secreting granule products into the nearby blood vessels for local or distant effects, although this certainly is not known. This work reports a unique synaptic ending upon a single-occurring SGC, which, in turn, closely approximates a ganglion neuron in a soma-somatic relationship. In addition, a very close association (but no actual contact) was observed between granule-containing processes, presumably emanating from the intraganglion clusters, and PGN. Whatever the function of ganglionic SGC may be, the exact relationship between SGC and PGN presumably would be of great interest and potential importance. © 1994 Wiley-Liss, Inc.  相似文献

This report describes a method for the detection of metals in tissues that is suitable for a routine pathology laboratory. The method has been developed specifically for the examination of soft tissues taken at biopsy from the site of surgical implants. It involves the mineralization of organic matter in sections by sodium metasilicate and the conversion of the metals to sulphates by sulphuric acid, which are then stained by appropriate solutions. High-quality protective garments are essential in many occupational assignments, e.g., in the petrochemical industry and for firefighting. The problem addressed in this paper is how to objectively asses the performance of protective garments designed to resist fire. We present a system based on a flame mannequin equipped with an array of temperature sensors that provide information about the temperature on the mannequin’s surface during exposure to fire. Particular attention is devoted to reconstructing the heat flux reaching the mannequin’s surface on the basis of the available temperature records (the inverse heat-conduction problem). The estimated heat flux is used in a skin-simulation model to predict the level of injury that would occur in human exposed to the same heat. The paper includes a novel computational procedure based on a detailed model of the temperature sensor and a numerical solution of the accompanying heat equation. The effectiveness of the solution is demonstrated with experimental data.  相似文献

The problems of statistical tests of isotropy, degree of anisotropy and stereological formulae are solved. This paper presents an overview of the problems and possible solutions for automation information management and utilization in the process industries. The process plant life-cycle model is introduced and the cost structure of process automation investments is reviewed in this context. The general trends in process industry and developments in automation technologies are described and their effects are discussed. The advances in engineering CAD systems and their usability in solving the information management needs over the lifetime of process plants is assessed. As long as neodymium amplifiers will be used in high-energy lasers, preionization problems will be present in firing the flash lamps. It is difficult to find papers describing technical solutions using preionization concept; only a few internal reports and proceedings from Pulsed Power conferences describe the usage of such circuits in power laser facilities. In all of them, a special negative high voltage power supply is used. A new simple solution, without any additional power supply, using only a small part of the energy gathered in the main capacitor bank for generating a preionization pulse is used. The firing order is split into two signals; the first one is triggering preionization circuit and the second one, after delaying, is triggering the main discharging circuit.  相似文献

This paper introduces a generic decision-making framework for assigning resources of a manufacturing system to production tasks. Resources are broadly defined production units, such as machines, human operators, or material handling vehicles; and tasks are activities performed by resources. In the specific context of FMS, resources correspond to individual machines; tasks correspond to operations to be performed on parts. The framework assumes a hierarchical structure of the system and calls for the execution of four consecutive steps to make a decision for the assignment of a resource to a task. These steps are 1) establishment of decision-making criteria, 2) formation of alternative assignments, 3) estimation of the consequences of the assignments, and 4) selection of the best alternative assignment. This framework has been applied to an existing FMS as an operational policy that decides what task will be executed on which resource of this FMS. Simulation runs provide some initial results of the application of this policy. It is shown that the policy provides flexibility in terms of system performance and computational effort.  相似文献

Friction involving human skin plays a key role in human life. The availability of a portable tribometer improves the accessibility to large number of both subjects and anatomical sites. This is the first mobile device suitable to measure skin friction with a controlled and variable normal load (range 0.5–2 N) and velocity (range 1–10 mm/s) of a material of choice making a sliding rotational movement over the skin.The results of a pilot study, using stainless steel samples on the ventral forearm compare to results in the literature. The new device can be used for more extensive research to found these results and determine the effects of normal load or sliding velocity on the skin's frictional behaviour.  相似文献

A multi-field approach is developed for simulating the continuum-scale mechanical response of cellular materials. This approach departs from traditional methods used to model cellular materials, which focus almost exclusively on the mechanical response of the cellular solid, while essentially ignoring the fluids permeating these material systems. In the present work, conservation equations are derived in multi-field form, producing a coupled set of governing equations with source terms depending on gradients in the cellular solid stress, but also on gradients in the permeating fluid pressure and momentum exchange resulting from relative motion between the cellular solid and permeating fluid fields. The multi-field equations of motion are implemented in a standard finite-volume computational test bed and used to study the dynamic response of cellular material systems. The influence of various permeating fluids, along with the effects of aperture size, loading rate, and boundary conditions, also are examined. By incorporating an advanced constitutive model for cellular solids into a multi-field response formulation, a promising new approach for simulating the finite-strain dynamic response of cellular materials is offered. Results demonstrate that the permeating fluid can play a major role in the general response of cellular material systems, contributing to the overall load-carrying capacity of the materials and affecting rate dependence and signal propagation speeds. Furthermore, the results point to the usefulness of the multi-field formulation and provide evidence to suggest that any modeling approach developed for cellular materials gives a proper accounting of the pressure evolution and flow behavior of the fluids present in these material systems.  相似文献

This paper presents a matrix approach coupled to the notion of constraints for the representation of tolerance zones within CAD/CAM (computer-aided design and manufacture) systems. The proposed theory, reproduces themeasurable ornon-invariant displacements associated with various types of tolerance zone. This is done using the homogeneous transforms commonly associated with robotic modelling.This matrix representation is completed by a set of inequalities defining the bounds of the tolerance zones. Moreover, the generation of the model and its mathematical definition allows for its use in the representation of clearances and for the computation of tolerance transfer. Apoptotic micronuclei have been studied, in different cell types, from a morphologic and functional point of view. Conventional electron microscopy, in various staining conditions, selective cytochemistry for DNA, and freeze fracture for the analysis of chromatin fiber organization and size were performed. In situ TdT and Pol I immunofluorescent techniques were carried out to detect double- and single-strand DNA breaking points by confocal laser scanning microscopy. Apoptotic cell ultrathin cryosections were also performed and were analysed by field emission in lens scanning electron microscopy. Double/single strand massively cleaved DNA was detected in micronuclei, with a highly supercoiled, uniformly packed, very dense arrangement.  相似文献
